中文摘要:初步探究刺五加内生放线菌对马铃薯疮痂病菌的拮抗作用,采用平板纸片法检测265株刺五加内生放线菌发酵产物对马铃薯疮痂病菌的拮抗活性;运用PCR技术检测活性菌株次级代谢产物合成相关基因PKSⅠ、PKSⅡ、NRPS、Halo、CYP;利用BOX-PCR检测活性菌株,选取谱带差异较大菌株进行16SrRNA基因序列同源性分析。结果表明,56株(21.13%)内生放线菌具有不同程度的抗性,抑菌活性较好的菌株大部分都能检测到至少一种功能基因,活性菌株分布于3个亚目、4个科、4个属。可见,刺五加内生来源放线菌是筛选马铃薯疮痂病拮抗菌的重要来源,筛选所获得活性菌株为马铃薯疮痂病生防菌剂研究奠定良好基础。 |

Antagonism of Endophytic Actinomycetes Isolated from Acanthopanax senticosus to a Potato Scab Pathogen |

Abstract:To explore the antagonism of endophytic actinomycetes isolated from Acanthopanax senticosus to a potato scab pathogen. The paper disc diffusion method was used to test the antagonistic activity against a potato scab pathogen. 56 strains(21.13%) showed that the activity of 265 endophytic actinomycetes strains was from Acanthopanax senticosus.Secondary metabolite biosynthesis genes (PKSⅠ, PKSⅡ, NRPS, Halo, CYP) were detected by PCR of the tested strains,and at least one functional gene was found in most of strains which had good inhibiting activities. The 16S rRNA gene sequences analysis were conducted to the different strains after classified with BOX-PCR, and the activity strains belong to 3 suborder, 4 families, 4 genera. Endophytic actinomycetes strains from Acanthopanax senticosus is an important source of antagonistic potato scab pathogen, and has laid a good foundation for the study of biocontrol agents against potato scab pathogen. |
