| 中文摘要:研究不同动物来源大肠埃希菌iss 基因分布及其致病性。分别将分离于发病猪、鸡和犊牛的44株大肠埃希菌计数后腹腔注射小白鼠，观察临床症状，记录发病及死亡情况；采用PCR方法分别扩增不同动物来源的44株大肠埃希菌的iss 基因存在情况，同时对部分PCR扩增产物测序并进行同源性分析。感染后小白鼠出现不同程度的临床症状。其中有14株对小白鼠的致病性最强，感染小鼠的死亡率达100%；有25株致病性较弱，死亡率为20%～80%；而另有5株接种感染后小鼠均无死亡；iss 基因在致病性大肠埃希菌中的PCR扩增总频率为79.5%；在毒力较强大肠埃希菌中的PCR扩增频率为92.8%；在无致病力（或低毒力）大肠埃希菌中的PCR扩增频率为16.7%；部分基因片段核苷酸序列与参考序列之间同源性在91.6%～99.0%。iss 基因致病性强的菌株扩增频率明显高于其他菌株，且不同来源菌株iss 基因片段变异程度不大。 |

| Abstract:The aim of the research is on the distribution of iss gene and pathogenicity of E.coli from different animal. Mice were infected with 44 E.coli strains from pigs, chickens and calves in intraperitoneal injection respectively and clinical symptoms, illness and death of mice were observed; iss gene of 44 strains from different animals were detected respectively with PCR and part of the PCR products were sequenced and analyzed. Mice appeared some symptoms in different degrees after infection, 14/44 strains caused 100% mortality of mice, 25/44 strains caused 20%-80% mortality, 5/44 strains did not cause the death of mice; the amplification frequency of iss gene was 92.8% in virulent strains, and was 16.7% in hypovirulent strains. The total amplification frequency of iss gene in 44 E.coli was 79.5%;the homology of nucleotide sequence of PCR products was 91.6%-99.0%.Amplification frequency of iss gene in virulent strain was significantly higher than that of hypovirulent strains, there were no significant variation of iss gene fragment in E.coli from different sources. |
