The toxicity of four waste water samples from reserve pit upon the root-tip cells of Allium ascalonicum L. was researched by applying modified Allium test. The samples were a mixture of substances and their chemical composition was only partly known. The first task was to find out the optimal duration for root-tips treatment. The treatment lasting 48 h has been accepted because it enables delivery of results only four days after the sample has been received. All undiluted four waste drilling fluid samples showed cytotoxicity as well as toxicity, which was proved by cytogenetic parameters such as high inhibition of mitotic activity or low mitotic index and high percentage of chromosomal and genomic aberrations. The toxicity was significantly reduced by increasing the degree of sample dilutions. The results of this investigation demonstrated that the modified Allium test could be easily used for detection of waste drilling fluids toxicity degree.  相似文献

Allium test has already been used to determine cytotoxicity of waste drilling fluids. In the present work the cytotoxicity of four pure chemicals (Defoamex, Idthin 400, Magco Thin and Slick Pipe) was investigated. Those chemicals are components of drilling fluids, therefore, they are usually constituents of oil and gas industry waste waters. The tested chemicals were prepared in 1:9 dilution and cytotoxic effects on root-tip meristem ofAllium ascalonicum were analysed after 24-, 48- and 72-hour-treatments. All samples showed cytotoxicity which was proved by cytogenetic parameters such as inhibition of mitotic activity and increase of mitotic abnormalities and chromosomal aberrations in comparison with the control. Almost all mitotic abnormalities induced by four chemicals tested were the result of disturbed spindle mechanisms accompanied with stickiness. Slick Pipe revealed the most prominent mitodepressive effect and induced a high number of abnormalities. It caused a significant decrease of mitotic activity and increase of mitotic abnormalities after all three treatment durations, while chemicals Defoamex and Magco Thin showed a significant decrease of mitotic activity only after 72-hour-treatment. Chemicals Defoamex, Idthin 400 and Magco Thin revealed a significant increase of mitotic abnormalities after 48- and 72-hour-treatments.  相似文献

For taxonomic purposes and because many wild relatives of Allium crops are endangered in their native habitat (Sancir et al., 1989), a collection of Allium species was built up in the IPK at Gatersleben. At present it consists of more than 3000 accessions representing about 330 species. In this context it was necessary to study the seed germination behaviour and dormancy of certain wild Allium species. Therefore, seeds of 91 wild and three cultivated Allium species were tested at germination temperatures of 5°, 11°, 16° and 26°C. The purpose of this study was to develop routine germination tests for the genebank work with wild alliums.  相似文献

Abstract

The objective of the present study was to determine the content of polycyclic aromatic hydrocarbons (PAH) in the rhizospheres of some selected plant species (Cucumis dativus, Daucus carota, Allium cepa, Cueurbita, Petroselinum sativum, Cucurbitaceae, Phaseolus vulgaris, Beta vulgaris, Apium graveolens, Allium porrum, Vicia faba). Various anthropogenic influences on the PAH content in the rhizospheres of the three species selected were also studied. The PAH content was determined using the high performance liquid chromatography (HPLC) method with ultraviolet (UV) detection after a preliminary sample treatment with ultrasonic techniques. On the basis of the PAH content, the organic carbon content and the value of log Koc, a potentially bioavailable PAH fraction present in the rhizospheres of the plants studied, were calculated.

The lowest content of the PAH sum (<1100 µg/kg) was found in the Petroselinum sativum, Cucurbitaceae, and Allium porrum rhizospheres. The content of the compounds studied was more than 67% lower than in the control soil (3307 µg/kg). Moreover, in the rhizosphere of the Allium porrum, the lowest content of all the five‐ and six‐ring PAH samples studied was found. The contribution of the 16 PAHs' sum, determined on the basis of an equilibrium partition in all rhizospheres studied, was higher than in the control soil. The evaluation of the PAH content in the rhizospheres of the three plants as related to the influence of anthropogenic sources showed a higher PAH content in them than in the control soil. The evaluation of the relation between the PAH content and the soil properties studied showed only sporadically statistically significant correlations.  相似文献

In this paper, three dissolution methods using pressure digestion vessels (low-, medium-, and high-pressure vessels) for the determination of metals in plant samples are described. The Plackett-Burman saturated factorial design was used to identify the significant factors influencing wet ashing and to select optimized dissolution conditions. The three methods were statistically compared (on-way ANOVA) on the same sample; no significant differences were obtained. In all cases the relative standard deviation values were <3%. The digestion method based on the use of low-pressure vessels and a microwave oven was validated against CRM GBW07605 tea leaves. This method was applied to the determination of Cu, Zn, Mn, Fe, Mg, and Ca in 22 different medicinal, aromatic, and seasoning plants by flame-atomic absorption spectrometry. The concentration intervals of metal in the plants analyzed were the following: Cu, 4 (Allium sativum)-35 (Thea sinensis) microg g(-1); Zn, 7 (Piper nigrum)-90 (Betula alba) microg g(-1); Mn, 9 (Allium sativum)-939 (Caryophylus aromaticus) microg g(-1); Fe, 33 (Allium sativum)-2486 (Anethum graveolens) microg g(-1); Mg, 495 (Allium sativum)-7458 (Ocimum basilicum) microg g(-1); Ca, 386 (Allium sativum)-21500 (Ocimum basilicum) microg g(-1).  相似文献

为给新型生物农药的研发提出依据,用黄连、黄芩、大蒜、金银花、柴胡和连翘6种植物的浸提液单剂及其复合剂对尖孢镰刀菌进行处理,探索浸提液单剂及复合剂对尖孢镰刀菌的影响。综合对尖孢镰刀菌菌落生长和产孢量两方面的影响,植物浸提液单剂的抑菌效果相对较好的处理分别是50%黄连、30%黄芩、20%金银花、60%柴胡、50%连翘、60%连翘以及10%大蒜、30%大蒜、60%大蒜。6种植物浸提液对尖孢镰刀菌均有抑菌作用,以黄连的抑菌效果最好,平均抑菌率为52.95%,平均抑产孢率为59.46%。  相似文献

针对藠头(Allium chinense)种植劳动强度大、人工成本高等问题,该研究设计了一种由排种盘、取种勺、取投种凸轮、种箱、安装板等组成的勺夹式藠头排种器。对该排种器工作原理和取种、携种、投种过程进行理论分析,建立离散元仿真模型,对取种过程进行仿真分析,确定了最优种勺结构。以大叶藠为对象,以取种合格率和漏取率为试验指标,进行种勺直径、种勺深度、取种起始角、种面高度与种箱高度之比和取种速度5因素显著性筛选试验,并以种勺直径、取种起始角、种面高度与种箱高度之比为试验因素进行回归正交试验;采用Plackett-Burman试验法和Box-Behnken中心试验法建立取种合格率和漏取率的回归模型,并进行参数优化,获得最优参数组合为种勺直径27mm、取种起始角30°和种面高度与种箱高度之比92.5%。在最优参数组合下进行台架试验,得到取种合格率91.17%,漏取率6.17%。田间试验结果表明在前进速度0.1~0.2 m/s的条件下,该排种器平均播种合格率、平均重播率和漏播率分别为63.10%、12.76%和24.14%。研究结果可为藠头播种机械的研究与设计提供参考。  相似文献

Leaching of heavy metals from coal fly ash dumpsites is an environmental threat often polluting both groundwater and surface water. This led to a detailed investigation on the cyto-genotoxicity and oxidative stress associated with the fly ash leachate (FL). Allium cepa L. bulbs were exposed to three concentrations of FL. Response to FL resulted in a reduction of root growth, inhibition of cell viability by up to 50%, and decrease in mitotic indices (control: 10.27; FA:water 1:4–4.52) with increased frequency of chromosome aberrations (5.7 fold) and bi-nucleate cells. Results of comet assay indicated that approximately sixfold increase in genotoxicity of FL was congruent with the endpoints of oxidative stress evaluated. The malondialdehyde content, superoxide dismutase, guaiacol peroxidase and glutathione-S-transferase activities were elevated in the treated plants in a dose dependent manner. The elemental analysis of FL showed the presence of heavy metals like cobalt (Co), chromium (Cr), copper (Cu), manganese (Mn), cadmium (Cd), nickel (Ni), lead (Pb) and zinc (Zn). The study demonstrated that FL can cause adverse effects to the plants. Therefore, the disposal of coal fly ash should be handled with caution and prevented from contaminating the surroundings.  相似文献

Marasmin, which is especially known from the two South African species Tulbaghia alliacea and Tulbaghia violacea , but was also described for the garlic mushroom Marasmius alliaceus , is the precursor of the thiosulfinate marasmicin. Marasmicin has attracted considerable attention because of its antifungal and tuberculostatic activities. However, many Allium species of the subgenus Melanocrommyum, especially Allium suworowii , are also very rich in marasmin. A. suworowii revealed concentrations of marasmin up to 1.6%, related to the fresh weight of bulbs, and up to 3.0%, related to air-dried fruiting bodies, of the corresponding γ-glutamylmarsmin was found in M. alliaceus. Both species show much higher amounts of marasmin as Tulbaghia and could be considered as natural sources for the isolation of this compound. Further promising Allium species with considerable amounts of marasmin besides other cysteine sulfoxides are Allium stipitatum and Allium altissimum . (R(S),R(C))-Marasmin is typical for the investigated species of the subgenus Melanocrommyum, whereas γ-glutamyl-(S(S),R(C))-marasmin is the only cysteine sulfoxide for the genus Marasmius known until now. Both cysteine sulfoxides were isolated and described as o-phthaldialdehyde (OPA) derivatives. Furthermore, the cysteine sulfoxides methiin, propiin, S-(2-pyrrolyl)-cysteine sulfoxide, eventually S-(2-pyridyl)-cysteine sulfoxide and S-(2-pyridyl)-L-cysteine N-oxide were found.  相似文献

The long-term study objective was to determine the suitability of selected species of genus Pinus (Pinus sylvestris, Pinus nigra and Pinus mugo) for use as bioindicators of environmental pollution. In these experiments a new methodology of evaluation of mitotic abnormalities was established and its potential for common use tested. Various Allium hybrids, obtained by the crossbreeding of Allium cepa (onion) as the mother plant and six taxonomically distant wild species obtained by embryo rescue, were investigated with special respect to their individual profiles of cysteine sulfoxides as well as enzymically and nonenzymically formed aroma substances. Alliinase (EC 4.4.1.4) catalyzes the conversion of odorless (+)-S-alk(en)yl-L-cysteine sulfoxides into volatile thiosulfinates. These thiosulfinates were converted to a variety of sulfides by steam distillation. SPME-gas chromatography (GC) and high-performance liquid chromatography (HPLC) used for the analysis of aroma components and their precursors permitted a high sample throughput, so that numerous gene bank accessions and Allium breeding materials were analyzed within a comparatively short time. Cysteine sulfoxides as well as alliinase activity were found in all investigated samples at different levels, but (+)-S-methyl-L-cysteine sulfoxide (methiin) was the most abundant sulfoxide present. (+)-S-(trans-1-Propenyl)-L-cysteine sulfoxide (isoalliin) is typical for onion and was found in all investigated hybrids. The pattern of the other cysteine sulfoxides depended strongly on the parent plants used. The profile of aroma components corresponded with the related pattern of aroma precursors (cysteine sulfoxides). Successful hybridization was proven by randomly amplified polymorphic DNA analysis. Together with these established marker techniques, HPLC and SPME-GC analysis provide support to breeding projects designed to improve the health and aroma properties of Allium hybrids.  相似文献

High-performance liquid chromatography-diode array detector (HPLC-DAD) coupled with electron spray mass spectrometry (ESI-MS-MS) was used to determine the flavonol profile in southern Italian red onions (Allium cepa L.). This on-line technique allowed the identification of seven flavonols in southern Italian red onion, quercetin 4'-glucoside and quercetin 3,4'-diglucoside being the most abundant components. Five minor flavonols have been recognized, offering a characteristic profile of such compounds in red onions under study. Quercetin 3-glucoside, quercetin 7,4'-diglucoside, quercetin 3,7,4'-triglucoside, and isorhamnetin 4'-glucoside have been previously reported as minor flavonoid components in Allium cepa, while isorhamnetin 3,4'-diglucoside was previously found in Allium ascalonicum. Traces of isorhamnetin 3-glucoside and free quercetin were also detected.  相似文献

The precursor of the orange-red pigment formed upon wounding the bulbs of Allium giganteum (Allium subg. Melanocrommyum) was isolated and shown to be S-(2-pyrrolyl)cysteine S-oxide. In addition, two other pyrrolylsulfinyl derivatives were found in an extract from the bulbs, namely, 3-(2-pyrrolylsulfinyl)lactic acid and S-(3-pyrrolyl)cysteine S-oxide. Contrary to a previous report, the latter compound was shown not to serve as the precursor of the pigment, being in fact only an artifact formed during isolation. The formation of pyrrolyl-containing compounds following disruption of A. giganteum bulbs was studied by a combination of LC-MS, LC-NMR and DART-MS. It was found that S-(2-pyrrolyl)cysteine S-oxide is cleaved by a C-S lyase (alliinase) to yield 2-pyrrolesulfenic acid. Two molecules of the latter compound give rise to highly reactive S-(2-pyrrolyl) 2-pyrrolethiosulfinate which in turn converts into red 2,2'-epidithio-3,3'-dipyrrole (dipyrrolo[2,3-d:2',3'-e]-1,2-dithiin). Several other pyrrolyl-containing compounds were detected in A. giganteum for the first time, including S-methyl 2-pyrrolethiosulfinate, S-(2-pyrrolyl) methanethiosulfinate, di(2-pyrrolyl) disulfide, and S-(2-pyrrolyl) 2-pyrrolethiosulfonate. It can be concluded that the formation of the orange-red pigment in Allium subg. Melanocrommyum species, despite sharing several analogous features, is of a different nature than the pink discoloration of onion (A. cepa).  相似文献

Abstract

Odour-mediated effects of leek Allium porum and chives Allium schoenoprasum (Alliaceae) on the host searching behaviour of the aphid Myzus persicae (Sulzer) (Homoptera: Aphididae) were studied. In an olfactometer, odour of the host plant sweet pepper Capsicum annuum L. (Solanaceae) was significantly attractive, whereas odour of chives was significantly repellent. Combined odour of sweet pepper and chives was neither attractive nor repellent. When sweet pepper plants were exposed to volatiles from chives for five days, their odour subsequently became repellent to M. persicae. An extract of leek plants was significantly repellent to aphids in the olfactometer, as were sweet pepper plants sprayed with this extract. Because both leek and chives can disrupt host finding by the aphid, both plants have potential for intercropping with sweet pepper. If successful in the field, such a system could bring economic benefits for farmers in the wet zone of Sri Lanka and other parts of the world.  相似文献

Chemotype analyses and random amplified polymorphic DNA (RAPD) genomic analyses have been applied to the characterization of Allium sativum variety from Voghiera (Ferrara, Italy), a typical Italian product actually demanding the Protected Designation of Origin (PDO). The garlic from Voghiera is characterized by peculiar morphological and composition characteristics. The proximate composition and atomic absorbance spectrometry elemental pattern of this garlic suggested as the chemical composition did not depend on the intrinsic pedologic soil features only, but it was probably connected to some peculiar genetic characters. Amplification of genomic DNA using random primers highlighted a good clustering differentiating of Voghiera Allium sativum from five commercial reference samples used in this study (Piacentino, Serena, France, China, and Adriano varieties), confirming the existence of intervarietal genetic difference. The intravarietal polymorphisms of Voghiera samples were low.  相似文献

The content of S-alk(en)ylcysteine sulfoxides, important nonvolatile flavor precursors, was determined in 15 different Allium species by means of gas chromatography. The method employed is based on derivatization of S-alk(en)ylcysteine sulfoxides with ethyl chloroformate followed by their reduction with sodium iodide. The total content of S-alk(en)ylcysteine sulfoxides varied considerably in the wide range between 0.02 and 1.3% fresh weight. Not only the total content but also relative proportions of individual derivatives varied to a great extent. A novel S-alkylcysteine derivative, S-ethylcysteine sulfoxide (ethiin), not previously reported to occur in Allium species, was found in most of the samples examined in trace amounts. None of the other S-alk(en)ylcysteine sulfoxides, for example, isopropyl, (Z)-1-propenyl, butyl, or pentyl, were detected in any of the samples analyzed, limiting possible levels of each of these components to 相似文献
