Haemocytes from penaeid shrimp (Farfantepenaeus californiensis, Litopenaeus vannamei and L. stylirostris) were separated using a discontinuous Percoll gradient. Shrimp haemocytes were spontaneously adhered to glass, allowing slide preparations for staining and microscopic differential counting. Like other crustaceans, shrimp has three main populations differing in presence and size of cytoplasmic granules and each population seems to be biochemical or functionally compromised. Prophenoloxidase (proPO )activity was mainly located in large granules haemocytes (75%) while the small granules cells participate with 25%, but seem to be responsible for encapsulation. Haemocyte discrimination ability was tested using Sephadex? (Seph?), DEAE‐Seph? and CM‐Seph?. Only DEAE‐Seph? was encapsulated by shrimp haemocytes and provoked the release of proPO activating system, indicating the role of particle charge in the activation of shrimp immune response.  相似文献

The bioflocs technology (BFT) for shrimp production has been proposed as a sustainable practice capable of reducing environmental impacts and preventing pathogen introduction. The microbial community associated with BFT not only detoxifies nutrients, but also can improve feed utilization and animal growth. Biofloc system contains abundant number of bacteria of which cell wall consists of various components such as bacterial lipopolysaccharide, peptidoglycan and β‐1, 3‐glucans, and is known as stimulating nonspecific immune activity of shrimp. Bioflocs, therefore, are assumed to enhance shrimp immunity because they consume the bioflocs as additional food source. Although there are benefits for having an in situ microbial community in BFT systems, better understanding on these microorganisms, in particular molecular level, is needed. A fourteen‐day culture trial was conducted with postlarvae of Litopenaeus vannamei in the presence and absence of bioflocs. To determine mRNA expression levels of shrimp, we selected six genes (prophenoloxidase1, prophenoloxidase2, prophenoloxidase activation enzyme, serine proteinase1, masquerade‐like proteinase, and ras‐related nuclear protein) which are involved in a series of responses known as the prophenoloxidase (proPO) cascade, one of the major innate immune responses in crustaceans. Significant differences in shrimp survival and final body weights were found between the clear water and in the biofloc treatments. mRNA expression levels were significantly higher in the biofloc treatment than the clear water control. These results suggest that the presence of bioflocs in the culture medium gives positive effect on growth and immune‐related genes expression in L.vannamei postlarvae.  相似文献

Sixteen candidate disease‐resistant parameters were selected through which to evaluate the acute hepatopancreatic necrosis disease (AHPND)‐resistant capability of Exopalaemon carinicauda after three generations of selection for AHPND‐causing strain of Vibrio parahaemolyticus (VPAHPND) resistance in our previous study. However, these parameters required further verification. In this study, another AHPND‐resistant E. carinicauda series was obtained through a short‐term selection procedure, consisting of three virulent challenge rounds of selection (about three‐week interval for each challenge) with VPAHPND infection. After this selection, the survival rate at 144 hr post infection (hpi) increased from 23.33% to 37.78% and the observed 48‐hr LD50 of VPAHPND to shrimp increased from 105.5 cfu/ml to 106.5 cfu/ml. Then, the immune response of this AHPND‐resistant E. carinicauda was studied using the 16 candidate AHPND‐resistant parameters selected for in our previous study. The improved VPAHPND clearance rate in hpi, increased total haemocyte counts, haemocyanin concentration, alkaline phosphatase activity and expressions of six immune‐related genes (Tollip and ALF in haemocytes and hepatopancreas; lysozyme, crustin and cathepsin B in hepatopancreas; and LGBP in haemocytes) at 24 hpi after the three‐round challenge selection suggest that these immune parameters may be reliable markers for the evaluation of the physiological status and potential AHPND‐resistant phenotypes in E. carinicauda.  相似文献

The purpose of this study was to investigate the effects of inbreeding on the immune responses and antioxidant status of Portunus trituberculatus juveniles. Results showed that inbreeding affected the total haemocyte count, and phagocytic, pro‐phenoloxidase (propo), phenoloxidase and antibacterial activities decreased after the seventh generation. Antioxidant status showed a similar pattern: total antioxidant capacity, superoxide dismutase (SOD) activity and GSH/GSSG in the cell‐free haemolymph and hepatopancreas decreased, while catalase activity in the cell‐free haemolymph increased (P < 0.05). There were no significant differences in α2‐macroglobulin and bacteriolytic activities in the cell‐free haemolymph and glutathione peroxidase activity in the cell‐free haemolymph and hepatopancreas among nine inbreeding generations. Gene expression levels of proPO and crustin in haemocytes and SOD in haemocytes and hepatopancreas also decreased significantly as the inbreeding generations increased. The results suggest that a high level of inbreeding could severely reduce the physiological health of P. trituberculatus. Our obtained data would be particularly useful for P. trituberculatus breeding programmes.  相似文献

The effects of fulvic acid (FA) on survival and immune‐related gene expression were investigated in Litopenaeus vannamei challenged with Vibrio parahaemolyticus by immersion. Shrimp were fed with different dietary FA concentrations (1, 2, 4 and 6 g/kg feed) for 20 days (first bioassay) or 8 days (second bioassay, 2 g/kg feed of FA added every 2 days) and then challenged with V. parahaemolyticus. In a third bioassay, the expression of three immune‐related genes (translationally controlled tumour protein [TCTP], superoxide dismutase [SOD] and heat‐shock protein 70 [HSP70]) in haemocytes or hepatopancreas of experimental shrimp was measured by real‐time quantitative PCR at 0, 6, 12, 24, 48, 72 and 96 hr after FA (2 g/kg feed) administration. Fulvic acid increased survival at a concentration of 2 g/kg feed supplied every two days. Interestingly, TCTP gene expression was upregulated, whereas gene expression of SOD and HSP70 was downregulated. In conclusion, dietary fulvic acid improves survival in white shrimp challenged with V. parahaemolyticus and modulates the immune response. Therefore, FA merits further evaluation as prophylactic treatment in commercial shrimp farms.  相似文献

Pacific white shrimp Litopenaeus vannamei (1050 individuals with initial weight of 1.01 ± 0.001 g) were fed either control diet or one of six dietary astaxanthin (AX) concentration (25, 50, 75, 100, 125 and 150 mg kg−1) diets for 56 days in 35 tanks (30 shrimp per tank). After 56 days of culture, shrimp‐fed AX125 and AX150 diets had higher (< 0.05) weight gain, specific growth rate, total antioxidant status and lower (< 0.05) superoxide dismutase (SOD), catalase (CAT) than shrimp fed control diet. After low dissolved oxygen stress for 1 h, survival rate of shrimp fed AX75, AX100, AX125 and AX150 diets was higher (< 0.05) than that of shrimp fed control diet. Hypoxia inducible factor‐1α (HIF‐1α), cytosolic manganese superoxide dismutase (cMnSOD) and CAT mRNA expression levels of shrimp fed seven diets were significantly down‐regulated under hypoxia than under normoxia, but their expression levels were higher under hypoxia in shrimp fed AX‐supplemented diets than in shrimp fed control diet. About 70‐kDa heat‐shock protein (Hsp70) mRNA expression level of shrimp fed seven diets was significantly up‐regulated under hypoxia than under normoxia, but its expression level was lower under hypoxia in shrimp fed AX‐supplemented diets than in shrimp fed control diet.  相似文献

Penaeid shrimps are among the most studied crustaceans, mainly regarding their immune system, and several proteins involved in the defense against pathogens have been described. Haemocytes are very dynamic cells responsible of recognition, phagocytosis, degranulation and nodule formation, but still little is known about their gene expression. Using differential display, we found modification in haemocyte gene expression after inoculation with an abiotic particle (DEAE‐Sephadex) or potential pathogenic bacteria (Vibrio alginolyticus). We also noticed that some of the newly expressed genes are exclusive from a specific treatment. Here we report that haemocytes from Litopenaeus vannamei are capable of recognizing and distinguishing different foreign materials, and respond specifically to each treatment, indicating some specificity in shrimp immune response.  相似文献

A 25‐day experiment was conducted to evaluate the optimal lipid level for postlarval Litopenaeus vannamei. Shrimp (1.7 mg) were fed five isonitrogenous diets containing grade levels of lipid (96.6, 114.3, 128.5, 136.5 and 154.5 g/kg diet, respectively). Each diet was assigned to four tanks (500 shrimp), and shrimp were fed six times a day. Weight gain was increased with the increasing dietary lipid levels, and the highest weight gain was observed in shrimp fed diet with 154.5 g/kg lipid (p < 0.05). On the contrary, the survival was lowest in shrimp fed the L15.45 and highest in shrimp fed the L11.43. Triglyceride in hepatopancreas was increased, and cholesterol was decreased with the increasing dietary lipid. Pyruvate kinase and AMPK mRNA expression were highest in shrimp fed the L12.85. Malondialdehyde in whole body was positively correlated with the dietary lipid levels. The mRNA expression of SOD and Caspase 3 was highest in shrimp fed the L12.85. After hypoxia stress, shrimp fed the L12.85 showed highest survival. The mRNA expression of superoxide dismutase and Akirin was highest in shrimp fed the L11.43 and L15.45, respectively. Based on the survival after 25‐day feeding trail and after the hypoxia stress, the optimal dietary lipid for postlarval L. vannamei should be 118–124 g/kg.  相似文献

为了评估饲料中添加酵母提取物对凡纳滨对虾免疫灵敏性和平衡性的相关基因表达的影响,以鱼粉含量为24.5%的基础饲料(对照组)及在基础饲料中添加2.5%酵母提取物的实验饲料,分别饲喂凡纳滨对虾56 d,检测两种饲料投喂的对虾在急性感染溶藻弧菌前后鳃组织Toll受体、IM D和溶菌酶mRNA表达量变化及感染后的对虾死亡情况。结果表明:摄食添加酵母提取物饲料的凡纳滨对虾鳃组织中Toll受体mRNA和溶菌酶mRNA表达量均显著高于对照组对虾(P0.05),IMD mRNA表达量与对照组无显著性差异(P0.05)。急性感染溶藻弧菌后,两组对虾鳃组织Toll受体、IMD和溶菌酶mRNA表达量随感染进程均出现显著变化,Toll受体、IMD和溶菌酶mRNA表达量峰值出现的时间分别为24、42和36 h,且摄食添加酵母提取物组对虾各基因的表达量峰值分别为对照组峰值的201.06%、481.46%和276.77%,均显著高于对照组对虾(P0.05)。摄食添加酵母提取物饲料组对虾鳃组织中Toll受体和IM D mRNA表达量在感染溶藻弧菌后12和24 h分别出现显著上调,而对照组对虾鳃组织中Toll受体和IMD mRNA表达量分别在感染弧菌后24和42 h才出现显著上调。两组对虾经溶藻弧菌人工急性感染后72 h内的累积死亡率无显著差异(P0.05)。实验表明,饲料中添加酵母提取物上调了溶藻弧菌感染前凡纳滨对虾Toll受体和溶菌酶mRNA的表达量,提早了溶藻弧菌感染后对虾Toll受体和IMD mRNA表达量上调时间,且增加了对虾Toll受体、IM D和溶菌酶mRNA表达量的峰值,在一定程度上提高了凡纳滨对虾免疫相关基因表达的灵敏性。  相似文献

White spot syndrome virus (WSSV) occurs worldwide and causes high mortality and considerable economic damage to the shrimp farming industry. Considering the global environmental, the economic and sociological importance of shrimp farming, and the constraints of high intensity cultivation, development of novel control measures against the outbreak of WSSV become inevitable. In this study, we have explored the protective efficacy of DNA vaccination and tissue distribution of the recombinant plasmid in immunized Litopenaeus vannamei. The VP28 gene was cloned in the eukaryotic expression vector pVAX1, and the construct vector was named as lpv28. The protective effect of lpv28 against WSSV was evaluated in L. vannamei by injecting lpv28 construct and later challenging with WSSV. Expression of these proteins from the recombinant plasmids was confirmed in vitro by RT-PCR and Western blot analysis. The result of vaccination trials showed that a survival rate in shrimp vaccinated with lpv28 was 52.5% at most compared to control groups (100% mortality). The immunological parameters analyzed in the vaccinated and control groups showed that the vaccinated groups owned a high level of lysozyme, alkaline phosphatase, and total superoxide dismutase when compared to the control group. Furthermore, protein expression analysis indicated that VP28 can be detected in gill, muscle and head soft tissue of the shrimps in the immunized group after 14th day injection. Thus, the result indicated that DNA vaccination strategy has a potential utility against WSSV.  相似文献

用添加CpG寡聚核苷酸(CpG ODN)和表面展示VP28的解脂耶罗维亚酵母(VP28-yl)的饵料投喂凡纳滨对虾,进行田间中试实验。投喂30 d后进行WSSV感染实验,评估其对凡纳滨对虾的免疫保护作用。投喂实验结束后,CpG ODN投喂组对虾的相对增重率达到(65.8±7.8)% (P<0.05),这暗示CpG ODN可能具有促生长作用。WSSV攻毒后,CpG ODN和VP28-yl投喂组对虾中WSSV拷贝数与对照组相比均显著降低(P<0.05),相对免疫保护率分别可达到26.7%和36.7%。在投喂结束和WSSV刺激后,CpG ODN组对虾中的呼吸爆发水平均显著升高(P<0.05)。而在VP28-yl投喂组,WSSV引起的细胞凋亡则显著受到抑制(P<0.05)。此外,WSSV刺激后,STAT基因在CpG ODN组和VP28-yl组对虾中的表达水平均显著上调(P<0.05),分别在第5天和第3天达到最大值,而对照组中则显著下调。研究结果表明,CpG ODN和VP28-yl增强了凡纳滨对虾抗病毒免疫力,对养殖对虾病毒性疫病的防控具有显著作用,可以作为免疫增强剂添加在饵料中,具有在养殖生产中推广使用的前景。  相似文献

为探讨转vp28蓝藻(Anabaena sp.PCC7120)口服剂对凡纳滨对虾抗白斑综合征病毒能力及其相应的免疫反应,本研究将此口服剂免疫幼虾7 d,再分别通过投喂攻毒和浸泡攻毒,测定其存活率及相应的免疫指标。投喂攻毒和浸泡攻毒的实验组存活率分别为78.8%和83.19%,表明该口服剂能显著增强对虾抗白斑综合征病毒的能力。蓝藻口服剂免疫对虾的酶活性检测结果显示,超氧化物歧化酶(SOD)、酚氧化酶(PO)、过氧化氢酶(CAT)和碱性磷酸酶(AKP)活性在免疫后2 h均有上升趋势,且在48或96 h达到最高值,这表明该口服剂能引起对虾体内酶活性变化。投喂攻毒的对虾酶活性检测结果显示,实验组攻毒后的对虾肝胰腺SOD活性分别比阳性对照组、野生型组、空载体组显著提高42.10%、32.26%和16.04%,且攻毒后的肌肉SOD活性分别比阴性对照组、阳性对照组、野生型组和空载体组略微提高17.70%、11.50%、15.00%以及10.00%。实验组攻毒后的对虾肝胰腺PO、CAT和AKP活性比阳性对照组分别提高12.17%、88.80%和240.07%,比野生型组分别提高21.49%、30.90%和100%;酸性磷酸酶(ACP)活性比阴性对照组略微提高,而在肌肉中各组ACP活性无显著性差异。同时浸泡攻毒组结果与投喂攻毒组具有类似的趋势。浸泡攻毒的实验组CAT和AKP活性显著高于其余处理组,且CAT活性比投喂攻毒更为显著。浸泡攻毒的实验组肝胰腺PO活性显著高于阳性对照组、野生型组和空载体组,而各组肌肉ACP活性无显著性差异。研究表明,转vp28蓝藻口服剂能够增强凡纳滨对虾抗病能力并延缓对虾死亡。转vp28蓝藻PCC7120本身可作为幼虾饵料直接投喂,无需提取纯化,有望大规模应用于对虾养殖产业。  相似文献

This study was carried out to investigate the effect of water‐soluble polysaccharides extract of algae Ulva rigida (WPU) as dietary supplement on growth performance, antioxidant enzyme activity, lysozyme and phenoloxidase activity, and resistance of shrimp (Litopenaeus vannamei) subjected to bacterial infection with Photobacterium damselae. Three replicate groups of shrimp (1.0 g) were fed four diets containing four levels, 0 or control, 0.5, 1 and 1.5 g/kg of WPU for 8 weeks over the growth trial. Thereafter, 30 shrimps from each dietary treatment were infected with bacteria P. damselae to evaluate disease resistance of infected shrimp. The results of this study showed that WPU was effective as a growth promoter for L. vannamei. The best growth rate was observed in shrimp fed 1.5 g/kg of WPU diet. Regarding antioxidant defences, the diets supplemented with three levels of WPU stimulated glutathione peroxidase and catalase activates in experimental shrimps. MDA content of L. vannamei‐fed diet containing WPU 1.5 and WPU 1.0 was lower than WPU 0 and WPU 0. 5 diets. Also, lysozyme and phenoloxidase activities of shrimp receiving WPU at 1.0 and 1.5 level were significantly higher than those fed WPU 0 and WPU 0.5 diets. In addition, using WPU extract in all diets decreased mortality in L. vannamei in a dose‐dependent manner after challenge with P. damselae. These results suggest that incorporation of water‐soluble polysaccharides from green algae U. rigida at 1.5 g/kg doses improves growth and antioxidant activity and enhances the immune responses in shrimp L. vannamei.  相似文献

A portion of the VP26 gene (VP26F109) encoding a structural protein of white spot syndrome virus was expressed, purified by SDS‐PAGE and used for immunization of Swiss mice for monoclonal antibody (MAb) production. Three groups of MAbs specific to different epitopes on VP26 were selected; these MAbs can be used to detect natural WSSV infection in Penaeus vannamei using dot blotting, Western blotting or immunohistochemistry without cross‐reaction with other shrimp tissues or other common shrimp viruses. The detection sensitivity of the MAbs was ranged 7–14 fmole per spot of the rVP26F109 as determined using dot blotting. A combination of three MAbs specific to VP26 with MAbs specific to VP28, VP19 and ICP11 increased the detection sensitivity of WSSV during early infection. Therefore, the MAbs specific to VP26 could be used to confirm and to enhance the detection sensitivity for WSSV infection in shrimp with various types of antibody‐based assays.  相似文献

The Pacific white shrimp (Litopenaeus vannamei), as an economically important species, has been reared in low‐salinity water during the last decade. To investigate how juvenile L. vannamei shrimp fed with fructose‐enriched Artemia respond to acute low‐salinity stress, the shrimp were randomly divided into four treatment groups, three groups were fed with Artemia enriched with either 100, 200 and 300 mg L?1 of fructose and a control group fed with Artemia with no enrichment for 10 days. The results showed that the 300 mg L?1 fructose group demonstrated the maximum survival rate and glycogen content. Additionally, the 300 mg L?1 fructose group showed significantly higher Na+/K+‐ATPase activity, total antioxidant capacity, and expression levels of Na+/K+‐ATPase α‐subunit, V‐H ATPase α‐subunit, hexokinase, phosphofructokinase, antioxidants (SOD, CAT, GPX) and Hsp70 mRNA when compared with the control group. Furthermore, after exposure to low salinity, the mRNA levels of phosphofructokinase, V‐H ATPase α‐subunit, GPX, p38, JNK and Rac1 stayed constant in shrimp fed with fructose‐enriched Artemia but changed significantly in the control group. Thus, a diet of fructose‐enriched Artemia can improve the osmoregulation and survival of juvenile L. vannamei shrimp exposed to low salinities.  相似文献
