The establishment and development of Haemonchus contortus in goats   总被引:1,自引:0,他引:1  
Twelve goats were inoculated with 40,000 third-stage Haemonchus contortus larvae and two were killed on each of Days 4, 7, 11, 14, 18 and 21 after inoculation (DAI). The number of worms that established, and the site of development were recorded. More worms established in the fundic, than in the middle or pyloric thirds of the abomasum. Early development occurred within the mucosa; emergence into the lumen started between 7 and 11 days after infection. By 4 DAI, all worms had completed the third moult to the L4 stage. At 11 DAI the majority of the worms were adults. A mean of 13.2% of the female worms had eggs in their uteri at 18 DAI; by 21 DAI more than half of the female worms had eggs in their uteri. The development of H. contortus was essentially similar to that described in sheep.  相似文献

Twelve goats were inoculated with 20,000 infective larvae of Trichostrongylus colubriformis and two were killed on each of Days 4, 7, 11, 14, 18 and 21 after inoculation (DAI). The number of worms that established, and the site of development were recorded. Most of the worms established within the first 3 m of the small intestine. There was little relocation or loss of nematodes after establishment. The worms started to migrate from the mucosa to the lumen 11 days after infection. At 4 DAI, 63% of the worms were still at the late L3 stage; the remainder of the worm population had completed the third moult to the L4 stage. The population at 11 DAI comprised largely young adults. When 21 DAI was reached, about 57% of the female worms had eggs in their uteri.  相似文献

Pairs of sheep infected with 120 000 larvae of Trichostrongylus rugatus were killed at intervals from 2 to 56 days after infection (DAI). Worms were located in tunnels in the epithelium of villi or upper intestinal crypts at all stages of development. Villus atrophy developed progressively until 16 DAI, when surface microtopography, characterised by subtotal villus atrophy, stabilised. Most severe lesions were in the first 3 m of small intestine where the density of nematodes was highest. Discontinuities in the epithelium and effusion of inflammatory cells and tissue fluids into the lumen were rare. No effects of infection on body-weight gain, appetite or serum total protein and albumin were evident in the first 20 days of infection. However, it was concluded that T. rugatus fundamentally resembled T. colubriformis and T. vitrinus in the response it elicited in the intestine of sheep, and must be considered potentially pathogenic.  相似文献

Fifteen lambs were each inoculated orally with 150 000 infective Camelostrongylus mentalutus larvae. Lambs were killed 3, 4, 8, 12, 16, 21 and 28 days after infection (DAI). Feed consumption and plasma total protein levels of the lambs killed 21 and 28 DAI did not differ significantly from those of uninfected controls. Three and 8 DAI most larvae were recovered from the fundic mocusa, but the intensity of establishment of larvae per gram of tissue was greatest at the cardia and diminished gradually toward the pylorus. Emergence of worms from the cardia and fundus occurred between 8 and 12 DAI, and a proportion of the lume-dwelling population shifted into the antral area, while the number of worms in the antral mucosa remained stable. Nodularity of the mucosa, mucous metaplasia of fundic epithelium, and vascular permeability as indicated by colloidal carbon label, were evident 3–4 DAI, and were most severe 8–12 DAI, when erosion of and inflammatory cell effusion from heavily infected mucosa was marked. pH and Na+ concentration in abomasal content were elevated at all times after infection, with peak values 12 DAI. Plasma pepsinogen levels in infected lambs were elevated beginning 3–12 DAI.  相似文献

Immunological resistance to Trichostrongylus colubriformis was developed in sheep by challenging them with 2500 larvae per day for 34 weeks. They were then rested for 24 weeks before being re-challenged with the same dose rate of the same larvae for 10 weeks. Nutritional, haematological and parasitological parameters were measured during the first 8 weeks of re-challenge infection. There were no faecal worm eggs excreted during the re-challenge infection. There was a small but significant increase in plasma-N leakage from Days 4 to 9 of dosing but this then declined to initial levels. There was no other detectable nutritional disturbance associated with re-infection. Re-challenge caused a rapid development of eosinophilia which peaked during Week 6 before declining to levels of the control animals by Week 8. Sheep resistant to T. colubriformis appear to be able to re-activate their immunological mechanisms when re-challenged with the parasite with little associated nutritional penalty.  相似文献

本试验旨在探明绵羊感染16型蓝舌病病毒(Bluetongue virus type 16,BTV16)后细胞因子IFN-γ、IL-2、IL-4和IL-10的消长特点。用实时荧光定量PCR检测方法对感染BTV16后3只绵羊上述4种因子mRNA进行检测,同时设立阴性对照绵羊,并以0 d mRNA为基准,计算mRNA的相对表达量,同时检测病毒抗体效价、测量绵羊体温。结果显示,接种BTV16的3只绵羊均不同程度产生抗体和体温症状,4种细胞因子的mRNA在接种病毒2~4 d内均出现显著上升,其中IFN-γ峰值在2.58~27.84倍之间,IL-2峰值在5.24~17.19倍之间,IL-4峰值在2.16~3.43倍之间,IL-10峰值在15.78~48.77倍之间,个体上升幅度存在显著差异,4种细胞因子均在高水平持续6 d左右后逐渐下降。对照绵羊上述参数在正常范围内波动。本研究阐明了接种BTV16后绵羊细胞因子IFN-γ、IL-2、IL-4、IL-10在转录水平上的消长特点,为进一步深入开展BTV感染特征、宿主机体免疫机制研究提供参考。  相似文献

The lysosomal activity of enterocytes of the small intestine mucosa was investigated in gnotobiotic and conventional piglets experimentally infected on the first day after birth (DAB) by the oocysts of the coccidia Isospora suis. A method of the proof of beta-D-glucuronidase (EC.3.2.1.31.) activity was used to demonstrate lysosomes. The piglets were infected by different infection doses of oocysts (100,000 oocysts in gnotobiotic piglets and 200,000 oocysts in conventional piglets). In the gnotobiotic infected piglets the activity of beta-D-glucuronidase in enterocyte lysosomes was investigated in the period from day 3 to day 11 after infection (DAI) and in the infected conventional piglets in the period from day 2 to day 10 after infection. Comparing the control piglets, the group of gnotobiotic piglets at the age of 2-5 days and the group of conventional piglets at the age of 4-7 days, the higher activity of beta-D-glucuronidase was demonstrated in the lysosomes of intestinal mucosa enterocytes in the gnotobiotic control piglets (+5.30 of the average density value, Dx). In the infected gnotobiotic and conventional piglets the pattern of beta-D-glucuronidase activity was found to have three stages in the course of this infection. Two stages can be characterized by a great increase in the enzyme activity (DAI 3-9 in gnotobiotic piglets, DAI 2-3 and 7-9 in conventional piglets. The third stage, which is manifest mainly in the conventional infected piglets, is characterized by a marked decrease in the activity of beta-D-glucuronidase, reaching the level of control findings (DAI 10 and mainly 11 in gnotobiotic piglets. DAI 4-6 and 10 in conventional piglets). A topographical picture shows that the two stages of increase and the stage of beta-D-glucuronidase activity decrease occur in the whole small intestine without any predisposition defect of the enzyme in the different sections of the small intestine.  相似文献

Eleven rabbits were infected with 10 embryonated eggs of Toxocara vitulorum per g body weight on Days 0, 35 and 72. Embryonated eggs and larvae were enumerated in feces on Days 1-3 after each infection. Two rabbits were killed and larvae were enumerated in small intestine, liver, lungs, skeletal muscles, heart, kidney, brain, eye, uterus, and mammary glands on Days 5, 15, 30, 65 and 101. Serum was obtained on Days 0, 5, 15, 30, 42, 50, 65, 78, 86 and 101 to perform enzyme-linked immunosorbent assays (ELISAs) and Western blots against an extract of embryonated eggs. Between 4 and 10% of the administered parasites, almost all embryonated eggs, were found in the feces after the first or second infection, but 32% (27% of them larvae) after the third. Yields of tissue parasites were 4.1% of the administered dose on Day 5, 2% on Day 15, and 0.8% on Day 30 of the first infection, 0.1% on Day 30 of the second infection, and 0.06% on Day 30 of the third. Larvae were found only in liver, lungs and muscle, including heart. Larva content declined steadily in liver and lungs from Day 5 to 30 of the first infection, was absent in the liver at Day 30 of the second, and in both organs at Day 30 of the third. Muscle larva content increased from Day 15 to 30 of the first infection, and persisted throughout the third infection. Production of IgM antibody was minimal, IgG and the sum of IgMGA antibodies increased slightly or moderately after the first and second infections, but dramatically after the third. Western blots revealed the first antigens (12) by Day 15 of the first infection. Their total number increased with time and number of infections, but some antigens disappeared, whereas new antigens appeared in the course of the observations. Four antigens (32,500-41,000 mol.wt.) may be related to protection. Comparison of the Western blot patterns of two rabbits showed differences in the antigens, recognizable for each rabbit.  相似文献

Separate groups of 3 oestrid-free lambs were exposed to infestation on irrigated pasture for periods of approximalely 33 days each over30 months, and on dry-land pasture for approxomately 42 days over a period of 18 months. With some exceptions, the lambs slaughtered from October-June were found to be infested with Oestrus ovis while, with one exception, those slaughtered from July-September were free. A minimum of 4 sheeps' heads, obtained weekly over 24 months from the Pretoria Municipal Abattoir, was examined for infestation. Of a total of 542 heads examined, 73,4% were infested, having a mean burden of 15,2 larvae. Mean larval burdens were slightly greater in hornless than in horned sheep in Dorper-type than in Merino-type sheep, and in lambs than in sheep with 2 or more permanent incisors. The largest larval burdens were recovered from sheep slaughtered during May and June and the smallest during September and October. The greatest number of 1st instar larvae were recovered during May and June and the smallest during September, but those recovered during the latter month were the largest. With one exception, mature larvae which pupated after 21 March or before 16 August failed to hatch as viable flies. Those which pupated after 16 August hatched as flies after a pupal stage of approximately 50 days and the first flies to hatch were invariably recovered during the first 2 weeks of October. The pupal stage decreased to approximately 25 days during December and January and increased again to approximately 50 days for flies hatching during May. No flies hatched between 18 May and 1 Cctober. The following life cycle ofr Oestrus ovis is suggested: sheep are repeatedly infested from October-June; thereafter infestation survives in the sheeps' heads until August, mainly as 1st instar larvae, then as pupae and larvae until fresh infestation takes place during October.  相似文献

Rhipicephalus evertsi evertsi larvae were fed on the ears of rabbits. Seven days after larval infestation, unfed, newly moulted nymphae were manually removed to infest a splenectomized donkey showing a patent Babesia caballi infection. Engorged nymphae were collected from the donkey and the ensuing adult ticks were placed on a susceptible horse. The horse contracted a B. caballi infection showing a prepatent period of 19 days after tick infestation. A very low parasitaemia, (highest score 2), which was patent for only 10 days, was recorded. Alkaline phosphatase activity (EC. 3.1.3.1.) in goblet cells was investigated in the small intestine of 16 gnotobiotic piglets infected one day after delivery (DAD) by different rates of oocysts of Isospora suis coccidia. At a high infection rate of I. suis (750,000) the goblet cells were found to be highly positive to alkaline phosphatase on day 3 to day 4 after infection (DAI). In piglets infected by a low infection rate of I. suis oocysts (100,000) the activity of alkaline phosphatase activity in goblet cells was proved on days 4 to 10 after infection. In the first group of piglets, the positive goblet cells prevailed in the middle region of jejunum, with the peak on 4th DAI. It the second group of piglets a marked increase in the alkaline phosphatase activity was recorded in the goblet cells in the posterior part of jejunum on days 4 to 5 after infection and on 10th DAI. No alkaline phosphatase activity in the goblet cells was demonstrated in the control gnotobiotic piglets at the age of two to seven days.  相似文献

Radioimmunoassay of plasma gastrin has shown that a hypergastrinaemia occurs in yearling sheep, after infection with 10(6) infective third stage larvae of Haemonchus contortus. Plasma gastrin concentrations began to rise two to four days after infection, that is, at about the same time as, or slightly before, the abomasal pH was observed to increase.  相似文献

Erythrocytes, packed cell volume and monocytes decreased, but eosinophils and basophils increased, after each infection. Serum enzyme levels indicated that liver damage occured only after the first infection, but muscle injury occurred after each infection and was increasingly more precocious after each infection.  相似文献

A comparison was made between two worm counting procedures in sheep. One involved the washing of the abomasa and first 10m of the small intestine using a 250microm aperture sieve. The other the processing of the abomasa and total length of the small intestine using a 38microm aperture sieve. In the former case, the collection of worms and contents from the small intestine was performed by introducing water into the posterior end and then drawing it between finger and thumb, whereas in the latter instance the intestine was cut open before rinsing and washing. The data demonstrated that processing only the first 10m of the small intestine was likely to result in a serious underestimation of the total numbers of worms present. They also showed that cutting open and rinsing of this organ was unnecessary if it had already been washed out by introducing water into it. The proportions of the total numbers of both 5th and late 4th stage worms recovered using a 250microm aperture sieve were generally high. These ranged from 93 to 100% for all 5th stage worms and between 85 and 100% for late 4th stage larvae of worm genera other than Trichostrongylus where 45.8-61.5% of such larvae were recovered. However, only small proportions of early 4th stage larvae (range 7.7-33.3%) were retained by the use of a 250microm aperture sieve regardless of the parasite genus involved. It is suggested that judicious application of these findings may enable considerable time savings to be made when performing worm counts in sheep.  相似文献

Naive and immune sheep, treated with anthelmintic, challenged directly into the abomasum and killed 48 hours later were used to examine the steroid sensitivity, persistence and dose dependency of immune mechanisms which result in the failure of incoming Haemonchus contortus larvae to establish (immune exclusion). Immune exclusion was almost totally abolished by corticosteroid treatment, the mean percentage of larvae establishing in the mucosa of immune sheep was 1.4 per cent of the challenge infection whereas in those treated with dexamethasone it was 24.4 per cent and in naive sheep 27.3 per cent. Immune animals challenged after seven and 42 days without antigenic experience excluded more than 90 per cent of the challenge larvae from their mucosae, whereas those challenged after 84 days without antigenic experience were as susceptible as naive animals. Immune exclusion was dose dependent, animals challenged with 10(6) and 10(5) larvae excluded 93 per cent and 82.5 per cent of the challenge dose whereas those challenged with 10(4) larvae failed to exclude larvae.  相似文献

Oestrosis, the nasal myiasis of sheep and goats, is caused by the larvae of Oestrus ovis L. 1758 (Diptera, Oestridae) that develop from the first to the third stage larva in the nasal cavities and frontal sinuses of affected animals. The authors report the results of an epidemiological study of oestrosis of sheep in Sardinia, Italy. Heads of 6-month to 10-year-old Sardinian sheep (n=566) from 124 free-ranging flocks were examined for the presence and location O. ovis larvae from December 1996 to November 1997. Larvae were collected, counted, and larval stages were identified. O. ovis larvae were found in 100% of examined flocks and in 91% (514/566) of examined sheep. The monthly prevalence ranged from 69% in May to 100% in July. First stage larvae were found in 82% (463) of all heads examined, second stage larvae in 65% (367) and third stage larvae in 10% (56). The majority of sheep harboured first stage larvae, with prevalences of over 80% throughout most of the study period. The prevalence of O. ovis found in this study of Sardinian sheep is the highest reported in the Mediterranean area. The high percentage of first stage larvae found throughout the entire study period may be due to a brief period of decreased rate of larval maturation, in particular in December 1996 (96%) and January-October 1997 (94%). Third stage larvae were consistently present, often however, with extremely low prevalences compared to total larval burden.  相似文献

OBJECTIVE: To examine the effect on wool growth of physiological changes associated with experimental flystrike in superfine Merino wethers. DESIGN: An animal house study comparing experimentally fly struck sheep with non-infected control groups that were pair fed or fed ad libitum. PROCEDURE: Sheep in the blow fly larval challenge group were each artificially infected with 500 first instar larvae per day for 8 days. Infections were terminated with insecticide on day 10. For all sheep, live weight change, feed intake and body temperature were monitored and blood collected for analysis of haematology, cortisol, serum haptoglobin, serum amyloid A and the pro-inflammatory cytokines, TNFa, IL-1 b, IL-6 and IL-8. Wool growth and staple strength were examined 4 months after infection. RESULTS: Experimental fly strike included moderate fever, depressed feed intake, and elevated cortisol, IL-6, serum amyloid A and haptoglobin. Staple strength was significantly decreased in struck sheep and did not differ between wool from sites adjacent to and remote from strike lesions. CONCLUSIONS: The results suggest that depression in feed intake alone is not sufficient to account for the decline in staple strength seen during fly strike. Cortisol, IL-6 and metabolic consequences of the acute phase response are likely to be major contributors to the systemic effects of blowfly strike on wool.  相似文献

Two similar experiments were conducted with groups of yearling sheep which had been trickle infected with Teladorsagia circumcincta for 6 weeks, treated with anthelmintic then challenged with a single dose of 50,000 larvae. Ten days after challenge these sheep contained significantly fewer worms and a significantly higher proportion of arrested early fourth stage larvae than controls which had not received the trickle infection. However, by 19 or 23 days after challenge most of the arrested worms had resumed development, a process which was accelerated in a group treated with corticosteroids. It was concluded that under these conditions arrested development of Teladorsagia was a short-lived, immunologically mediated phenomenon which could be reversed by immuno-suppressing the sheep.  相似文献
