Pterocarpus santalinoides is a multipurpose tree species, native to Africa and South America. However, excessive pruning for fodder is jeopardising its seed production leading to poor regeneration in the Sahel region. Efficient vegetative propagation techniques may overcome this concern. To address this issue, four experiments were conducted to determine the effects of substrate (Experiment 1), cutting type (Experiments 2 and 3), genotype (Experiment 4) and auxins (Experiments 1, 2, and 4) on rooting ability in a non-mist propagating unit in Mali. Among the four tested substrates, no significant differences were found in rooting parameters, including the percentage of rooted cuttings, the number of roots and secondary roots per rooted cutting, and the length of the longest root per rooted cutting. Semi-hardwood cuttings rooted significantly better than softwood cuttings with a higher percentage of rooted cuttings (31.7?±?6.5% vs. 9.2?±?2.7%), greater number of roots (8.2?±?4.8 vs 1.5?±?0.7), greater number of secondary roots (16.7?±?4.2 vs 4.6?±?1.8) and longer root length (17.8?±?12.0?cm vs 2.3?±?1.0?cm) per rooted cutting. In addition, one-node cuttings cut below the second and third nodes, respectively, had the highest rooting success (88% and 90%, respectively) followed by cuttings cut below the fourth node (68%) and the first node (62%). Significant differences in the percentage of rooted cuttings were found among 12 randomly selected genotypes with rooting varying from 2% to 75%.The results also showed that auxin application, especially IBA, had a significant effect on rooting success, the number of roots and secondary roots per rooted cutting. Single-node cuttings, and cuttings of 10?cm length with multiple softwood and semi-hardwood nodes, can be used with or without auxin treatment for successful clonal propagation of P. santalinoides. The results of the present study indicated that P. santalinoides is amenable to clonal propagation using low-cost propagators and sand-based rooting media.  相似文献

Applied auxin, node position, leaf area and cutting length were examined to investigate the requirements for rooting stem cuttings of Khaya ivorensis. All these variables were shown to be important factors affecting rooting, confirming the hypothesis that successful rooting can be achieved if these primary variables are optimised.The best concentration of the auxin IBA was found to be 200 g per cutting, which hastened rooting, increased the percentage of cuttings rooted and increased the number of roots per cutting. One clone (8013) was unresponsive to auxins in terms of the percentage of cuttings rooted, but was the most responsive in terms of the numbers of roots per cutting. A greater percentage of cuttings from basal nodes were rooted than from apical nodes. Cuttings cut squarely at the base produced a radially-arranged root system, whereas an oblique cut resulted in a one-sided root system.Trimming the leaf area of cuttings to 10 cm2 gave greater rooting percentages than trimming to 100 cm2. In general, long cuttings (39 mm) rooted better than short cuttings (19 mm), however, there was an interaction between leaf area and cutting length, in which cuttings with short stems and large leaves had the lowest rooting percentage.  相似文献

Softwood shoots were produced from 40 cm long stem segments placed horizontally in flat trays containing sterilized sand under natural light or shade conditions for subsequent rooting and micropropagation studies in teak (Tectona grandis L.). Higher number of shoots (6.17) per log was produced under natural light as compared to shade conditions. Forcing was also better in natural light as compared to shade in terms of shoot length, number of nodes or leaves. For rooting, 2–4 cm long softwood shoots were excised and treated with either indole-3-butyric acid (IBA) or α-naphthyl acetic acid (NAA) at 0, 1000, 2000 or 3000 μmol·L–1 each or with combinations (1000 + 1000, 2000 + 2000 or 3000 + 3000 μmol·L–1) and then placed in flat trays containing autoclaved sand at 25 ± 2ºC in 16 h photoperiod at 35 µmol·m–2·s–1. After 28 days, softwood cuttings treated with IBA + NAA (3000 + 3000 μmol·L–1) had highest rooting percentage (89.3%) with 5.5 mean roots. Shoot apex and nodal explants of softwood cuttings were pretreated with 0.1% (w/v) ascorbic acid, boric acid, activated charcoal, citric acid, glutamine or polyvinylpolypyrollidone (PVP) for 24 h to remove phenolic compounds before surface disinfestation. Glutamine (Gl) and PVP were equally effective resulting in 60% establishment of shoot apices on MS medium supplemented with 10 μmol·L–1 6-benzylaminopurine (BAP) + 5 μmol·L–1 NAA. Using shoot apices, highest (42.80) number of multiple shoots with 54.33 mm shoot length were obtained on MS + BAP (8.8 μmol·L–1) + IBA (2 μmol·L–1) after 45 days. Leaf area, IBA concentration and age of stockplants were all found to be important factors for successful rooting for vegetative propagation using single-node cuttings of the two main African mahogany species: Khaya anthotheca and K. ivorensis. Cuttings with leaf area of 30–50 cm2 had the best rooting percentage and cuttings with about 30 cm2 had the most number of roots per cutting in K. anthotheca. Khaya ivorensis, cuttings with 10–30 cm2 leaf area had the highest rooting percentage. Cuttings collected from 1-year-old stockplants recorded the highest rooting percentage and largest number of roots per cutting. Cuttings from 3-year-old stockplant of the same seeds sources had the lowest rooting suggesting aging negatively impacted rooting ability. The effect of auxin concentration, on rootability was examined with cuttings of K. anthotheca. An IBA concentration of 0.8% was the best exogenous auxin concentration for percentage rooting, number of roots per cutting and the length of the longest root per cutting.  相似文献

We investigated the effects of warming the basal ends of Carolina poplar (Populus × canadensis Moench.) softwood cuttings at controlled low-air-temperature on their root growth and leaf damage after planting. The warming treatment was applied to the cuttings by soaking 10 mm beyond the cut end in warmed water maintained at 30 °C in a cold chamber maintained at an air temperature of 10 °C and a photosynthetic photon flux density (PPFD) of 10 μmol m?2 s?1 (near the light compensation point at 10 °C) until rooting was observed. The warmed cuttings were then grown in a growth chamber at an air temperature of 30 °C, relative humidity 85–90 %, and a PPFD of 100 μmol m?2 s?1. Control cuttings were grown in the growth chamber throughout the experiment. Rooting occurred simultaneously for both warmed and control cuttings, irrespective of air temperature. Root development was greater and leaf damage, evaluated on the basis of extent of necrosis, was less for warmed cuttings than for control cuttings. The reduction of leaf damage for warmed cuttings probably resulted from reduced post-planting water stress and leaf senescence, because of improved root development as a result of the pre-planting warming treatment. This technique could improve the propagation of cuttings of woody plants, because it would ensure that the cuttings are ready to develop roots with minimum loss of carbohydrates, irrespective of weather conditions.  相似文献

A study to determine the best vegetative propagation protocol was carried out for Dalbergia melanoxylon. Natural regeneration of D. melanoxylon is limited due to poor seed viability and germination. The effects of age of donor plant, indole-3-butyric acid (IBA) treatment and cutting position on rooting of stem cuttings were investigated. The effect of age of donor plant was significant (P < 0.001) only for percent rooting, position effect was significant for all studied parameters, and IBA treatment at 300 ppm was insignificant only for percent callusing. Interactive effects of cutting position × age of donor and cutting position × IBA treatment were significant for percent rooting and root number per cuttings while interactive effect of age × IBA was significant only for percent rooting. Observed difference in rooting performance could be due to uneven distribution of root promoting and inhibiting substances with respect to age of donor plant and IBA which intensified root formation. Cuttings from juvenile donor plant performed better in all rooting parameters than mature donor plant. The highest percentages in rooting were (71.11%) and (24.42%) for cuttings from juvenile and mature donor plants, respectively. IBA treated cuttings produced higher percentage rooting, number of roots and root length than untreated cuttings which revealed a strong IBA influence on rooting ability of stem cuttings in D. melanoxylon. It was concluded that stem cuttings should be taken from juvenile donor plants and basal cutting position should be used to produce high quality planting stock material for the afforestation and conservation programme of D. melanoxylon.  相似文献

Regeneration to ensure sustainable production and conservation of mahogany species globally is considered very important to foresters, investors in plantation, conservationists and timber industries worldwide. Natural regeneration of the mahoganies has been found to be far less than the rate of exploitation hence strong efforts are being made to develop other methods to regenerate mahogany seedlings to augment the low natural replacement and to ensure sustainability. This study was commissioned to investigate the effect of rooting media (river-sand, top soil and a mix of the two (50:50 v/v) on the rooting ability of four mahogany species (Khaya grandifoliola, K. ivorensis, Entandrophragma angolense and E. utile) using non-mist propagators. We investigated the effects of the source of stem cutting within a shoot (top, middle and basal position) on the rooting ability of stem cuttings from the four species in river-sand. Roots per cutting, longest root length and rooting percentage differed significantly among species in all three rooting media. Rooting ability was greatest in K. grandifoliola, which exhibited the highest average rooting percentage (73 %), longest average single root (9.9 mm) and the most roots per cutting (4.9). Rooting percentage was higher in the Khaya species than in the Entandrophragma species in all three rooting media. The mixed medium had the highest rooting percentage and the highest number of roots per rooted cutting in the two Khaya species, while the topsoil media had the best rooting percentage in the Entandrophragma species. Rooting of the two Khaya species can be maximized using a mixture of river-sand and topsoil. Cutting position within a shoot had significant impact on rooting in all four species with rooting percentage being best from middle positions in all four species. However, length of longest root and the number of roots per rooted cutting varied between the middle and basal portions. The study provides useful information for vegetative propagation of leafy stem cutting of the four African mahogany species, which could contribute to regeneration and conservation of these important timber species in the tropics.  相似文献

Larix olgensis is a major reforestation species in areas of the northeast part of China. Rooted cutting for vegetative propagation on this Larch has been carried out since 1990, and later the bulk method of hardwood cutting propagation also has been studied. The result is as follows: (1) Cuttings from physiologically juvenile trees are easy to root, and develop better roots. In addition, there are many methods to enhance the cuttings rooting. (2) The good time for seedling transplanting is in the middle of August. Treatments with plant hormones and rare-earth compounds can improve the seedling survival rate, and the survival rate of the seedlings soaked in chelate rare-earth molybdenum compound (300×10−6) for 3 hours reached 94.00%, 19.49% higher than that of the control. (3) Methods of maintaining juvenility such as hedging, is essential to further developments for operational planting of vegetative propagates. The region of West and Central Africa is endowed with high-value fruit trees and medicinal plants, which are currently traded locally as well as on regional and international markets. Unfortunately, they are all exploited from the wild and there has been little or no focussed effort to domesticate and cultivate them. Prunus africana is one of these important medicinal plant under domestication. A series of nursery experiments were conducted to assess the effects of rooting medium (sawdust, sand and a 50:50 mixture of sand and sawdust), auxin concentration (0, 50, 100, 150, and 200 μg IBA), and leaf area (0, 5, 10, 20, and 25 cm2) on rooting success of juvenile cuttings of P. africana. The percentage of cuttings rooted was significantly greater (P < 0.05) in sawdust (80%), than in sand alone (72%) or in mixture with sawdust (71%). Leaf area also significantly affected the percentage of rooting. Leafless cuttings did not root and were all dead by week 6, but in leafy cuttings rooting ability increased proportionally with leaf area up to 20 cm2 (79%). Larger leaf cuttings (25 cm2) rooted at the same level as those of 20 cm2. The cuttings with the largest leaves also had the greatest mean number of roots per cutting (14 roots cutting−1), while those with the smallest (5 cm2) leaf area produced the fewest roots (5 roots cutting−1). The application of auxin (IBA) promoted rooting (P < 0.05) up to an optimum application of 100–200 μg IBA per cutting, but 300 μg was supraoptimal. 本文以广东梅州野生及人工种植的多年生鸡血藤Spatholobus suberectus为材料,以提高鸡血藤扦插育苗繁殖系数为目的,对影响鸡血藤扦插生根的主要因素开展扦插试验。结果表明:人工植株和野生植株的穗条扦插生根率无显著差异;扦插基质类型对生根率有显著影响,采用黄心土作为基质生根效果最好;1~3a生枝条中,鸡血藤的木质化枝条生根率要高于半木质化嫩枝,粗度>0.8cm的木质化枝条生根率最高;外源激素种类及其质量体积浓度均对扦插生根率产生显著影响,采用质量体积浓度为500mg·L^-1的GGR浸泡插穗基部1h扦插生根效果最好;不同季节扦插对比试验显示春季扦插的生根率要显著高于其他季节;扦插方式试验表明营养袋扦插生根率与插床扦插无显著差异。  相似文献

在重庆市城口县咸宜乡苗圃和中国林科院温室开展崖柏扦插繁殖试验,研究采穗母树年龄、不同激素种类及其组合、激素浓度、扦插基质、物理促根等不同处理方式下崖柏扦插生根效应.结果表明:(1)在咸宜乡苗圃,采用自动喷雾设施,以蛭石-泥炭土-珍珠岩(1∶1∶1)为扦插基质,用3种激素和清水处理崖柏幼树1年生枝条,扦插后160d的生根率为74.76%~96.70%,不同处理组合生根效应具有显著差异,2000 mg· L-1 IBA>清水对照(CK)>1 000mg·L-1 IBA>1 000 mg·L-1IBA+ 500 mg· L-1NAA;(2)在林科院温室用2种激素和清水处理崖柏幼树1年生枝条,扦插于蛭石-泥炭土-珍珠岩(1∶1∶1)的基质,搭塑料拱棚,扦插后120 d的生根率远低于咸宜乡苗圃,为25.93%~45.37%,激素处理后的枝条与对照差异显著,1 000 mg·L-1 IBA+ 50 mg·L-1 NAA=2 000 mg·L-1 IBA>清水对照(CK);(3)在咸宜乡苗圃用不同激素和物理促生根方法组成的27种组合处理的成年崖柏枝条,最高生根率仅为20.83%.生根率超过10%的处理组合是:清水处理,泥炭土∶蛭石∶珍珠岩(1∶1∶1);1 000 mg·L-1IBA+500 mg·L-NAA,泥炭土∶珍珠岩(2∶1);1 000 mg·L-1IBA,泥炭土∶蛭石∶珍珠岩(1∶1∶1),流水冲洗.  相似文献

The cutting propagation technique ofPicea koraiensis was carried under automatic spraying of total sunlight by adopting the same year epicormic branches from different trees with different age treated by different hormones in different concentration and time and the orthogonal and randomized block was designed. The results were as follows: (l)The rooting rate of the cutting reached at least 87 percent, when the cuttings were from the seed tree with the age below 25 years old and the cuts of the cuttings base were in vertical angle and on the fine sand medium. (2) The sequence of effective factors affecting the rooting rate were the style of cut, the age of seed tree and the type of medium. Mesquite (Prosopis alba Grisebach), an important species in arid and semiarid regions, is currently commercially propagated by seeds and so there is great interest in developing techniques for species propagation, domestication and improvement. Cloning through mini-cuttings is a new and promising technique for the production of clonal seedlings of forest species. The main objective of this work was to evaluate vegetative propagation using the mini-cutting technique and indolebutyric acid (IBA) at different concentrations (0, 3,000, 4,500, 6,000 and 7,500 mg L?1) on rooting of P. alba clones. Rooting was achieved in 98–100 % of the mini-cuttings at all concentrations tested. Increasing IBA concentration resulted in an increase in the number of leaves and leaflets, as well as in fresh matter weight and number and length of roots of clone seedlings, until an optimum point (between 3480 and 4800 mg L?1) was reached. Plants were also vegetatively propagated in the field via conventional clonal garden cuttings under the same rooting conditions to compare the efficiency of the two propagation techniques. Mini-cuttings showed higher rooting and survival percentages than cuttings. A correlation analysis conducted between characteristics of stock plants (height, diameter, number of shoots) and shoots (height, diameter) showed a positive correlation between rooting and height of shoots with total root length, leaflet number, and height of clonal seedlings. We propose the mini-cutting technique as a tool for P. alba cloning.  相似文献

Tea tree oil is extracted from the leaves and twigs of Melaleuca alternifolia (Maiden & Betche) Cheel, and it is widely used in medicines, food preservatives, cosmetics and health care products. Traditional propagation of M. alternifolia from seeds does not necessarily transfer the desired characteristics from their mother trees, the seedlings are not uniform, and the multiplication rate from cuttings is relatively low. For these reasons, it is necessary to develop tissue culture techniques for this species. This study showed that an efficient explant initiation medium for M. alternifolia was MS 1/2 + BA 0.6 mg L?1 + NAA 0.1 mg L?1 + sucrose 30 g L?1, which yielded a 75.9 % initiation rate. An efficient multiplication medium was MS + BA 0.3 mg L?1 + NAA 0.15 mg L?1 + sucrose 30 g L?1, which yielded a 4.3 multiplication rate and 3.2 cm shoot length. The rooting medium was MS 1/2 + IBA 0.1–0.25 mg L?1 + sucrose 15 g L?1, which yielded a 100 % rooting rate, 2.94–3.32 roots per individual and 1.36–1.44 cm root length. Local red-core soil was suitable as a transplant medium, and yielded 98 % survival. This study improved the tissue culture technique for mass-propagation of M. alternifolia, enabling the production of high quality plants for market.  相似文献

This study investigates the optimal conditions for the vegetative propagation of the multipurpose leguminous shrub Colutea istria from leafy stem cuttings. The conditions examined included 1) the origin of the cutting (apical, medial or basal), 2) indole-3-butyric acid (IBA) treatment (0, 50, 200 or 400 mg/l), 3) the relative humidity of the propagation environment (high or low), 4) wounding base of cutting, and 5) the season in which the cutting was taken. The rooting ability of the cuttings was significantly influenced by all these factors. Winter cuttings obtained from the middle and base of the branch showed a greater rooting capacity than those harvested from the apical area, while autumn cuttings harvested from the middle and apical areas of the branch showed a greater rooting capacity than those harvested from the base. Winter cuttings showed the best rooting capacity than autumn cuttings. IBA treatment (200 mg/l), high humidity (98-100%), and wounding increased the rooting capacity of the cuttings. The sprout of new leaves was higher in basal cuttings, while IBA and wounding also significantly enhanced the root number of rooted cuttings. A method for vegetatively propagating C. istria plants is given.  相似文献

In the Sahel region, many woody species are used as fodder because of their high nitrogen content, especially during the dry season, in contrast to grasses and crop residues. Unfortunately, this resource is being threatened by regular pruning, increasing livestock browsing and impeding their natural regeneration. Therefore, there is a need to find appropriate regeneration options to sustain fodder production. Thus, a series of vegetative propagation experiments were conducted to identify the most appropriate multiplication methods of some of the species used as fodder. This series included testing the effects of naphthalene acetic acid (NAA) and indole butyric acid (IBA) on rooting ability of stem cuttings of 12 species: Afzelia africana, Balanites aegyptiaca, Bauhinia rufescens, Commiphora africana, Faidherbia albida, Ficus gnaphalocarpa, Guiera senegalensis, Kigelia africana, Pterocarpus erinaceus, Pterocarpus lucens, Pterocarpus santalinoides and Terminalia avicennioides. The series also evaluated the impacts of the size (C. africana) and the age (P. erinaceus) of the cuttings on their rooting success. Pterocarpus santalinoides was revealed to be an easy-to-root species, showing a high percentage of rooted cuttings (up to 88±3%), followed by P. erinaceus, P. lucens and B. aegyptiaca (up to 37±4%, 31±5% and 26±6%, respectively), in contrast to A. africana, cuttings of which did not root at all. The application of NAA and IBA enhanced the number of roots and secondary roots per rooted cutting. Large-diameter cuttings (15–20?mm) of deeply planted C. africana gave the highest percentage of rooted cuttings (78.3±4.4%). Tree pruning improved the rooting capacity of cuttings collected from P. erinaceus with one-month resprouts giving the highest percentage of rooted cuttings (50.0±6.7%). These results indicate that P. santalinoides, C. africana, P. erinaceus, P. lucens and B. aegyptiaca are amenable to vegetative propagation as an alternative to sexual propagation.  相似文献

Khaya anthotheca is a hardwood species from Africa and recently introduced to Brazil. This species yields high-quality wood for diverse applications, but little has been done toward an effective propagation method for large-scale production in nurseries. We evaluated the effects of different concentrations of macro and micronutrients (i.e., 100, 50 and 25% of the concentration of a referenced solution) combined with indole-3-butyric acid (IBA) at 0 and 2 g L?1 on the survival rate of mini-stumps and the survival and adventitious rooting of K. anthotheca mini-cuttings. The mini-stumps were grown from a source of seeds imported from Ghana, West Africa. The mini-stumps survived at a high rate (97%). Consistently high shoot yields were obtained from the mini-stumps (average of 1.8 per mini-stump). High survival percentage of mini-cuttings and adventitious rooting were observed when solutions with 100 and 50% of the nutrients concentration were combined with 2 g L?1 IBA. Histological sections analyzed through optical microscopy indicated the tissues from mini-cutting stems were juvenile, which might have stimulated adventitious rooting. These results have important implications for further work aimed at establishing propagation strategies for K. anthotheca, which are of prime importance for assisting breeding programs of this species.  相似文献

Chinquapin(Castanea henryi) is a dual-purpose tree species in China valued for as a source of timber and starch.We investigated the effect of four cutting mediums(pure vermiculite;peat:river sand at 3:1 v/v;peat:krasnozem at 1:1 v/v;and pure krasnozem) and three stem cutting periods(March,May,and July) on rooting performance of C.henryi cuttings.Different cutting periods and cutting mediums greatly influenced the rooting rate of C.henryi,ranging from 3.35 to 77.31%.Principal component analysis indicated that the best combination of cutting period and cutting medium was semi-hardwood cuttings(May cuttings)+krasnozem.Histological evidence indicated that adventitious root initials were present by week 5-6,and that the site of root primordia initiation was observed in the vascular cambium.Stem anatomical structures observed at different periods indicated that a xylem/radius ratio of29.90-37.42% and a fractured phloem fiber ring are indicative of rooting success.The relational model between rooting index and medium properties indicated that nutrient content and porosity significantly influenced callus production.However,pH strongly affected C.henryi root formation,with the Pearson correlation coefficients for May and July cuttings of-0.856 and-0.947,respectively.Our protocol is helpful to achieve mass clone propagation of improved C.henryi genotypes,thus overcoming a common hurdle in chinquapin breeding programs.  相似文献
