ObjectiveTo characterise the effects of alfaxalone by intramuscular (IM) injection in red-eared slider turtles and the influence of body temperature on anaesthetic duration and depth.Study designProspective, randomised part-blinded experimental trial.AnimalsTen healthy adult female red-eared sliders.MethodsEach turtle was anaesthetized four times with 10 and 20 mg kg?1 alfaxalone at 20 and 35 °C respectively. Time to maximal effect and plateau and recovery periods were recorded. Skeletal muscle tone, presence of various reflexes, response to noxious stimuli, and heart rate were assessed.ResultsResults are given for protocols 10 mg kg?1 20 °C; 20 mg kg?1 20 °C; 10 mg kg?1 35 °C and 20 mg kg?1 35 °C, respectively: mean time (±SD) to maximal effect was 16 ± 8, 19 ± 6, 5 ± 2 and 7 ± 5 minutes; duration of the plateau phase was 13 ± 12, 28 ± 13, 8 ± 5 and 8 ± 5 minutes and recovery time was 76 ± 20, 126 ± 17, 28 ± 9 and 41 ± 20 minutes. Endotracheal intubation was successful in 80%, 100%, 0% and 30% of turtles, respectively. At 35 °C, all animals retained nociceptive sensation in the front limbs, hind limbs and vent, whereas at 20 °C a few turtles lost peripheral nociceptive sensation. Corneal and tap reflexes were retained in all trials. Mean heart rates were 30 ± 2 and 66 ± 4 beats minute?1 at 20 and 35 °C, respectively.Conclusions and clinical relevanceAlfaxalone administered IM in red-eared sliders provided smooth, rapid induction and uneventful recovery. At 35 °C either dosage provided only short (5–10 minutes) and light sedation. At 20 °C, 10 mg kg?1 provided sedation suitable for short non-invasive procedures. About 20 mg kg?1 provided anaesthesia of approximately 20 minutes duration, appropriate for induction of inhalational anaesthesia or for brief surgical procedures with supplemental analgesia.  相似文献

Veiled chameleons (Chamaeleo calyptratus) and panther chameleons (Furcifer pardalis) are the most popular chameleons over the world, and consequently, two of the most frequent species presenting to veterinary practices. However, published studies on normal ultrasonographic anatomy for these lizards are currently lacking. The objectives of this prospective anatomic study were to develop an ultrasound protocol for evaluation of the coelomic cavity in these species and describe the normal ultrasonographic anatomy of the coelomic organs. Seventeen healthy veiled chameleons and 15 healthy panther chameleons were included. A linear 18 MHz transducer was used. Chameleons were sedated and restrained in right lateral recumbency by an assistant. Longitudinal and transverse images were acquired, and authors recorded qualitative and quantitative ultrasonographic characteristics of the coelomic structures. The kidneys, liver, caudal vena cava, hepatic veins, portal vein, gallbladder, wall of the stomach and intestine, gonads and, when distended, urinary bladder could be visualized during ultrasonography of the coelomic cavity in both species. The spleen, pancreas, and adrenal glands could not be identified. Findings from the current study supported the use of ultrasonography for veiled chameleons (Chamaeleo calyptratus) and panther chameleons (Furcifer pardalis) with suspected intracoelomic diseases and provided normal reference information for future studies of these chameleon species.  相似文献

ObjectiveTo characterise the anaesthetic effects of alfaxalone administered intramuscularly (IM) at 10, 20, and 30 mg kg?1.Study designProspective, randomized cross-over study.AnimalsTen juvenile green iguanas (Iguana iguana) of mean body weight (±SD) 480 ± 134 g.MethodsAlfaxalone was administered IM in the triceps of both thoracic limbs. Times for anaesthetic induction, plateau and recovery periods were recorded. Skeletal muscle tone of the jaw, neck, thoracic limbs, pelvic limbs, and tail was scored. The palpebral, corneal and righting reflexes, and the response to painful stimuli were also assessed. Pulse rate and respiratory rate were recorded. Comparisons between different dosages and over time were made using anova.ResultsTimes are given for 10, 20 and 30 mg kg?1 dosages respectively: mean time to maximal effect was 7.7 ± 2.2, 5.4 ± 1.7 and 3.9 ± 1.2 minutes; duration of the plateau phase was 11.3 ± 3.8, 22.1 ± 6.5 and 39.1 ± 11.5 minutes; recovery time was 10 ± 2.4, 17.5 ± 8.6 and 25 ± 7.1 minutes; and total anaesthetic duration was 29 ± 35.7, 45 ± 8.2 and 68 ± 9.8 minutes. Endotracheal intubation was possible in 40% of the subjects given 10 mg kg?1 and in 100% subjects given both 20 and 30 mg kg?1. Loss of response to a painful stimulus was seen in 0/10, 8/10 and 9/10 animals at 10, 20, and 30 mg kg?1 respectively. There was an initial dose-dependent depression of respiration followed by a significant increase in frequency over time. In contrast, pulse rates decreased by 20% over the duration of the anaesthetic events.Conclusions and clinical relevanceIntramuscular administration of alfaxalone is a simple, rapid and reliable means of achieving relatively brief sedation or anaesthesia in healthy green iguanas. A dosage of 10 mg kg?1 provides light sedation, appropriate for examination and venipuncture; 20 mg kg?1 provides a level suitable for minor procedures or for endotracheal intubation and supplementation with inhalational anaesthesia; 30 mg kg?1 produces an anaesthetic plane suitable for surgical procedures of limited duration (up to 40 minutes).  相似文献

The distribution on serovars of 60 Salmonella isolates from reptiles kept in captivity in Denmark during the period 1995–2006 was investigated. The isolates were all recovered from clinical specimens submitted to the National Veterinary Institute. A majority of the samples were from reptiles in zoological gardens or similar, while a minor number was from reptiles kept in private homes. A total of 43 serovars were detected, most of them being what is usually called exotic serotypes, and many not having a trivial name, while a few isolates belonged to well‐known human pathogenic serovars, such as S. Enteritidis, S. Typhimurium, S. Bovismorbificans. One isolate was rough and two were non‐typeable. Isolates from turtles belonged to the subspecies enterica, while many isolates from both sauria and snakes belonged to other subspecies. The findings underline the potential zoonotic risk by handling reptiles in zoological garden or other public settings, or keeping pet reptiles in private homes.  相似文献

Two-dimensional echocardiography was performed on Burmese pythons (Python molurus bivittatus) to determine an optimal echocardiographic imaging technique for snakes and to describe the echocardiographic anatomy of the snake heart. Five snakes immobilized with tiletamine/zolazepam and maintained on isoflurane in oxygen were imaged in dorsal recumbency. The portion of the snake's body containing the heart was submerged in warm water to reduce the artifact created by air trapped between and under the scales. Imaging in sagittal planes demonstrated the caudal vena cava, sinus venous valve, right atrium, various portions of the ventricle, horizontal septum, the left aortic arch, and pulmonary artery. Transverse imaging depicted the spatial relationship of the left and right aortic arches and pulmonary artery and the horizontal septum. Basic knowledge of cardiac blood flow in the reptile was necessary to understand the echocardiographic anatomy.  相似文献

Among ectotherms, individuals raised in cooler temperatures often have larger body size and/or larger cell size. The current study tested whether geographic variation in cell size and plasticity for cell size exist in a terrestrial, ectothermic vertebrate, Anolis carolinensis Voigt, 1832. We demonstrated temperature-induced plasticity in erythrocytes and epithelial cells of hatchlings lizards derived from the eggs of females sampled from four populations and incubated at multiple temperatures. Larger cells were produced in hatchlings from cooler treatments; however, hatchling body size was unaffected by temperature. Therefore, temperature-induced plasticity applies at the cellular, but not organismal, level in A. carolinensis. In addition, reaction norms for cell size differed among populations. There was a latitudinal trend in cell size and in plasticity of cell size among our study populations. The two southernmost populations showed plasticity in cell size, whereas the two northernmost ones did not. We suggest that selection pressure for larger cell size in northern, cooler environments has restricted plasticity in A. carolinensis applied at the cellular level in response to variable incubation environments.  相似文献

Objective To determine intraocular pressure (IOP) in adult yellow‐footed tortoises using applanation tonometry. Animals Fifteen healthy adult captive yellow‐footed tortoises (eight males and seven females). Procedures Intraocular pressures were estimated for tortoises by using an applanation tonometer after topical anesthesia. Body length, measured from nuchal to anal scutes, ranged from 27.5 to 57.2 cm. Five measurements from each eye were obtained by a single observer in an ambient temperature of approximately 30 °C. Results Mean ± SEM IOP of 30 eyes of 15 yellow‐footed tortoises was 14.2 ± 1.2 mmHg. Range of IOP was 6–30 mmHg for tortoises. Significant differences were detected neither between right and left eyes (P = 0.357) of individual tortoises, nor between males and females (P = 0.524). Observer's readability was good (intraclass coefficient = 0.65), and IOP did not change over the ordered five measurements. Conclusions There was no significant difference in IOP between males and females in this specie. Tonometry values for normal eyes may represent a useful diagnostic methodology for recognition and treatment of ocular diseases in reptiles.  相似文献

Two male spiny-tailed monitor lizard (Varanus acanthurus) broodmates were diagnosed with chondroblastic osteosarcomas associated with the pelvic girdle. One lizard was 6 months old and the other 5 months old at the time of diagnosis. Grossly, the tumors appeared as large, firm, multilobulated masses that were white on cut surface and firmly adhered to the underlying bone. Histologically, both tumors were characterized by proliferation of neoplastic spindloid to stellate cells that encompassed the vertebrae and invaded into the vertebral and pelvic bones. Both osteoid production and multifocal chondrous differentiation were observed within neoplastic masses. Retinoblastoma 1 was absent in neoplastic cells. Even though osseous neoplasms are rare amongst reptiles, Varanus spp. may have a genetic predilection for developing chondroblastic osteosarcoma.  相似文献

Objective

To describe the anaesthetic, physiological and side effects of intramuscular (IM) medetomidine and ketamine, followed by inhalational anaesthesia with sevoflurane, in Nile crocodiles (Crocodylus niloticus).

Study design

Observational trial.

Animals

Ten juvenile captive-bred Nile crocodiles undergoing surgical implantation of skeletal beads and muscular electrodes.

Methods

During preanaesthetic examination, the following variables were assessed: heart (HR) and respiratory (fR) rates, and response to palpebral, corneal and toe- and tail-pinch withdrawal reflexes. The crocodiles were injected IM with an initial combination of medetomidine and ketamine and re-evaluated at 5 minute intervals for 20 minutes, or until they appeared unresponsive. If that did not occur, the drugs were redosed according to a decision tree based on the observed effects. The righting, biting and palatal valve reflexes were assessed in the unresponsive crocodiles, and used to confirm anaesthetic induction. Anaesthesia was maintained with sevoflurane in oxygen. At the end of surgery, medetomidine was antagonized with IM atipamezole.

Results

The decision tree identified 0.3 mg kg?1 medetomidine and 15 mg kg?1 ketamine as a useful drug combination, which resulted in anaesthetic induction and surgical anaesthesia 16 ± 8 and 16 (25–20) minutes after injection, respectively. Compared to baseline, HR and fR significantly decreased after anaesthetic induction (p < 0.001), but then remained stable throughout surgery. Intraoperatively, cloacal temperature [27 (26–30) °C] did not change over time (p = 0.48). The total dose of atipamezole was 2 (1–3) mg kg?1 and time to recovery was 36 (20–60) minutes. Perioperative complications were not observed.

Conclusions

and clinical relevance Medetomidine and ketamine, injected IM and followed by sevoflurane anaesthesia, may be regarded as a useful anaesthetic technique for juvenile Nile crocodiles undergoing minimally invasive experimental surgery.  相似文献

We examined the distribution of the orexin‐like peptides in the pituitary and median eminence of the flat‐tailed house gecko (Hemidactylus platyurus) using immunohistochemistry. Orexin‐B‐like, but not orexin‐A‐like, immunoreactivity was detected in the pituitary, specifically in the pars intermedia, and these cells corresponded to alpha‐melanocyte‐stimulating hormone (αMSH)‐producing cells. Orexin‐B and αMSH secreted from pars intermedia may modulate secretion of adenohypophyseal cells in the pars distalis. In the median eminence, orexin‐B‐immunoreactive puncta and fibres were observed, and these structures corresponded to gonadotropin‐releasing hormone (GnRH)‐immunoreactive puncta and fibres. Orexin‐B secreted from GnRH‐containing neurons in the hypothalamus may affect thyrotropin‐releasing hormone‐containing neurons resulting in modulation of αMSH secretion of melanotrophs in the pars intermedia.  相似文献

Chrysosporium‐related infections have been increasingly reported in reptiles over the last 2 decades. In this report, we describe clinical, cytologic, histopathologic, and ultrastructural aspects of Chrysosporium‐related infection in 2 Inland Bearded Dragons (Pogona vitticeps). Case 1 was presented for an enlarging raised lesion over the left eye and multiple additional masses over the dorsum. Case 2 was submitted to necropsy by the referring veterinarian for suspected yellow fungus disease. Impression smears of the nodules in case 1 revealed granulomatous to pyogranulomatous inflammation and many septate, variably long, 4–10 μm wide, often undulated hyphae, and very rare conidia. Postmortem impression smears of the superficial lesions of case 2 contained large numbers of solitary conidia and arthroconidia and low numbers of hyphae with similar morphology to case 1. Histopathology of the 2 cases revealed severe, multifocal, chronic, ulcerative, nodular pyogranulomatous dermatitis, with myriad intralesional septate hyphae, and arthroconidia. Fungal culture and molecular sequencing in both cases indicated infection with Nannizziopsis guarroi.  相似文献

Continued research efforts into diagnostic techniques, in particular imaging (e.g., contrast radiography and ultrasonography), of gastrointestinal tract have resulted in clinical advancements for practicing veterinarians. An antibacterial compound from crocodile blood was partially purified and functionally characterised. The freshwater crocodile (Crocodylus siamensis) plasma with antibacterial activity was partially purified by using a centrifugal concentrator and reverse phase high powered liquid chromatography, and designated as crocosin. Crocosin exhibits antibacterial activity toward Salmonella typhi and Staphylococcus aureus. Crocosin is thermostable and resistant to pronase digestion. The structure of crocosin analyzed by mass spectrometry contains repeating units of 94 and 136 m/z. Scanning electron microscopy indicates that crocosin probably penetrates progressively into cytoplasm space, perturbing and damaging bacterial membranes. Crocosin may provide an early defense mechanism toward bacterial infection in freshwater.  相似文献

Objective

To characterise the minimum dose of intramuscular alfaxalone required to facilitate intubation for mechanical ventilation, and to investigate the impact of cranial versus caudal injection on anaesthetic depth.

Study design

Randomised crossover study.

Animals

Six healthy juvenile ball pythons (Python regius).

Methods

Three dosages (10, 20 and 30 mg kg–1) of alfaxalone were administered to each python in a caudal location with a minimum 2 weeks washout. Induction and recovery were monitored by assessing muscle tone, righting reflex, response to a noxious stimulus and the ability to intubate. A subsequent experiment assessed the influence of injection site by comparing administration of 20 mg kg–1 alfaxalone in a cranial location (1 cm cranial to the heart) with the caudal site. Respiration rate was monitored throughout, and when intubation was possible, snakes were mechanically ventilated.

Results

Regardless of dose and injection site, maximum effect was reached within 10.0 ± 2.7 minutes. When administered at the caudal injection site, intubation was only successful after a dosage of 30 mg kg-1, which is higher than in previous reports for other reptiles. However, intubation was possible in all cases after 7.2 ± 1.6 minutes upon cranial administration of 20 mg kg–1, and anaesthetic duration was significantly lengthened (p < 0.001). Both 30 mg kg–1 at the caudal site and 20 mg kg–1 at the cranial site led to apnoea approximately 10 minutes post-injection, at which time the snakes were intubated and mechanically ventilated.

Conclusions and clinical relevance

Alfaxalone provided rapid, smooth induction when administered intramuscularly to pythons, and may serve as a useful induction agent prior to provision of volatile anaesthetics. The same dosage injected in the cranial site led to deeper anaesthesia than when injected caudally, suggesting that shunting to the liver and first-pass metabolism of alfaxalone occur when injected caudally, via the renal portal system.  相似文献

A 13‐year‐old male spur‐thighed tortoise (Testudo graeca) was presented with anorexia, apathy, and prolapse of penile tissue. Ultrasonography revealed a large heterogeneous mass in the coelomic cavity, and fine‐needle aspiration demonstrated sperm. Magnetic resonance imaging showed a sharply defined mass originating from the left testis. Appearance and signal intensities were similar to those reported in testicular neoplasms in humans, in particular sharing similarities with seminomas. Necropsy results and histopathological findings were consistent with a seminoma. To the authors’ knowledge this is the first report of the diagnosis of testicular neoplasia in a reptile using imaging techniques.  相似文献

Midazolam is a benzodiazepine with sedative, muscle relaxant, anxiolytic, and anticonvulsant effects. Twelve ball pythons (Python regius) were used in a parallel study evaluating the pharmacokinetics of 1 mg/kg midazolam following a single intracardiac (IC) or intramuscular (IM) administration. Blood was collected from a central venous catheter placed 7 days prior, or by cardiocentesis, at 15 time points starting just prior to and up to 72 hr after drug administration. Plasma concentrations of midazolam and 1‐hydroxymidazolam were determined by the use of high‐performance liquid chromatography tandem‐mass spectrometry and pharmacokinetic parameters were estimated using noncompartmental analysis. The mean ± SD terminal half‐lives of IC and IM midazolam were 12.04 ± 3.25 hr and 16.54 ± 7.10 hr, respectively. The area under the concentration‐time curve extrapolated to infinity, clearance, and apparent volume of distribution in steady‐state of IC midazolam were 19,112.3 ± 3,095.9 ng*hr/ml, 0.053 ± 0.008 L hr?1 kg?1, and 0.865 ± 0.289 L/kg, respectively. The bioavailability of IM midazolam was estimated at 89%. Maximum plasma concentrations following an IM administration were reached 2.33 ± 0.98 hr and 24.00 ± 14.12 hr postinjection for midazolam and 1‐hydroxymidazolam, respectively, and 22.33 ± 20.26 hr postinjection for 1‐hydroxymidazolam following IC administration.  相似文献

A 7-year-old Thoroughbred racehorse developed loud exercise-related respiratory noises and exercise intolerance over a short period. Examination showed congenital cricopharyngeal-laryngeal dysplasia (rostral displacement of the palatopharyngeal arch) and also left sided idiopathic laryngeal hemiparesis, the latter appearing to be of recent clinical onset. The animal developed colic some months after laryngeal surgery and a nephrosplenic entrapment was surgically corrected. Aerophagia and regurgitation worsened after surgery and, after developing inhalation pneumonia, the animal was destroyed. Post-mortem examination confirmed the presence of idiopathic laryngeal hemiparesis and of cricopharyngeal-laryngeal dysplasia which had some previously undescribed laryngeal muscle abnormalities.  相似文献
