OBJECTIVE: To identify the major allergenic proteins from the 3 main species of dust mites to which dogs react (Dermatophagoides farinae, D. pteronyssinus, and Euroglyphus maynei) and evaluate the potential cross-reactivity of dust mite allergens with antigens from the ectoparasitic mite Sarcoptes scabiei var canis. SAMPLE POPULATION: Sera from 83 dogs with atopic dermatitis. PROCEDURE: Sodium dodecylsulfate-polyacrylamide gel electrophoresis and immunoblotting using serum from atopic dogs was used to identify IgE-binding proteins in extracts of the 4 mite species. RESULTS: Sera of atopic dogs contained IgE against 23, 17, 25, and 17 allergens from D. farinae, D. pteronyssinus, E. maynei, and S. scabiei, respectively. Unlike the situation for humans, the major allergens for dogs are mostly proteins that are larger than 90 kd molecular weight. Dermatophagoides farinae and E. maynei appear to be more allergenic for dogs than is D. pteronyssinus. Some dogs with serum IgE against dust mites also had IgE against antigens of S. scabiei var canis. CONCLUSIONS AND CLINICAL RELEVANCE: Multiple dust mite allergens induce an IgE response in dogs. These allergens are mostly greater than 90 kd molecular weight.  相似文献

White leghorn hens were experimentally infested with northern fowl mites (Ornithonyssus sylviarum) and antibody responses to mite immunogens were monitored over 12 weeks. Mite burdens increased during the early phase of infestation and declined over the latter weeks of the study. Antigen was prepared from homogenized whole mites, which were then sonicated and extracted with non-ionic detergent. Antigen extract was fractionated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and antibody-reactive polypeptides were identified by immunoblotting. At the start of infestation, hens had natural, pre-existing antibodies that reacted with several mite-extract components. Individual hens had different natural antibody reactivities; however, all birds had immunoglobulins reactive with extract polypeptides of 117,000, 77,000 and 36,000 molecular weight. A variety of mite extract components reacted with hen antibodies generated in response to experimental infestation. The number of antibody-reactive mite polypeptides increased through week 8 of infestation and then decreased by week 12. Fifteen polypeptides of northern fowl mite extract were reactive with antibodies developed by the majority of infested birds. These commonly reactive polypeptides had molecular weights ranging from 40,000 to 160,000. Glycoconjugates of fractionated mite extract were identified by blotting with lectins that have different carbohydrate binding specificities. Also identified were lectins that bound extract components with the same molecular weights as those moieties complexed by immunoglobulins of infested birds.  相似文献

Sarcoptic mite hypersensitivity and skin lesions in slaughtered pigs   总被引:1,自引:0,他引:1  
The association between papular skin lesions in slaughtered pigs and hypersensitivity to Sarcoptes scabiei var suis was examined in experimental and field studies, and by the retrospective analysis of monitoring records obtained at slaughter. A causal role for S scabiei was indicated by the production of lesions in experimentally infested pigs, by increased clinical mange in herds having high lesion scores, and by a reduction in the severity of lesions after the implementation of more intensive mange control measures. The specificity of the lesions as an indicator of sarcoptic mite hypersensitivity was estimated to be 0.92 from experimental data and 0.79 from field data. Generalised lesions were rare in mange free pigs, indicating that such lesions were highly specific (1.0 from experimental data, greater than 0.98 from field data) for mange. The assessment of the severity of the lesions in samples of slaughtered pigs appears to be a useful aid to assessing the severity of sarcoptic mange in pig herds.  相似文献

Using light microscopy, polarizable amorphorous bodies with tinctorial characteristics of lipochromes and flocculent pigmental materials were found within the alveolar macrophages and peribronchiolar region of the lungs in a pig-tailed macaque (Macaca nemistrina) infected with simian lung mite (Pneumonyssus simicola). Examination of these pigmental bodies, using a high-voltage (1.2 meV) electron microscope and an energy-dispersive X-ray analysis system, indicated that the pigmental bodies contained a high concentration of silica. One female adult P simicola also was examined, and its digestive tract had visible, polarizable pigment and a high concentration of silica. Thus, lesions associated with lung mite infection in Old World monkeys may be superimposed by this silicotic condition, which may be associated with their arid, dusty, environment. Therefore, until the link between lung mite infection and silicosis is clarified, experimental inhalation toxicologic findings in mite-infected Old World monkeys should be interpreted cautiously.  相似文献

Large-bodied Demodex mites were detected in 4 dogs. The mites were readily detected in material obtained via deep skin scrapings and were most commonly found on the trunk. The mites were distinguishable from D. canis, because adult males were approximately 100% longer and adult females were approximately 50% longer than adult male and female D. canis mites, respectively. The large-bodied mites were found in the hair follicles, sebaceous ducts, and sebaceous glands in histologic sections of skin from 2 dogs. All dogs had adult-onset generalized demodicosis. Two dogs had coexistent iatrogenic hypercortisolism, 1 dog had hypothyroidism, and 1 dog did not have coexistent disease. Infestations responded to miticidal therapy, control of the coexistent disease, or both.  相似文献

Records of domestic animals brought to the Veterinary Entomology Laboratory for diagnosis of suspected lice and mite infestation over a 10 year period were analysed. From a total of 794 suspected cases, 137 (17.3%) and 247 (31.1%) were positive for lice and mange mites respectively. The most common lice species recorded were Linognathus vituli (66.7%) on cattle, L. ovillus (83.3%) on sheep, Haematopinus suis (100%) on pigs and Menacanthus stramineus (54.5%) on poultry. Other lice species recorded included Haematopinus bovis and Solenopotes capillatus on cattle, Damalinia ovis on sheep, Linognathus stenopsis and Menacanthus stramineus on goats, Goniocotes sp. on a horse, Linognathus setosus and Menacanthus stramineus on dogs, Goniodes gigas, Lipeurus caponis, Menopon gallinae and Chelopistes meleagrides on poultry. The most common mite species were Demodex folliculorum on cattle (96.9%) and on dogs (80.8%), Sarcoptes scabiei on pigs (100%) and Notoedres cati (80.3%) on rabbits. Other mite species included Psoroptes communis, Cheyletiella parasitivorax, Ornithonyssus gallinae and Dermanyssus gallinae.  相似文献
