Primiparous sows (n = 36) were used to evaluate the effects of dietary lysine intake in lactation on follicular development and oocyte maturation after weaning. Sows were assigned randomly to one of three diets containing .4% (low lysine, LL), 1.0% (medium lysine, ML), or 1.6% (high lysine, HL) total lysine. All diets contained 2.1 Mcal NE/kg and exceeded NRC (1988) requirements for all other nutrients. Actual lysine intakes over an 18-d lactation were 16, 36, and 56 g/d for sows consuming LL, ML, and HL, respectively. Ovarian data were analyzed for sows determined to have been slaughtered during the first proestrus period after weaning, using previously established criteria. Compared with sows fed ML and HL, sows fed LL tended to have lower uterine weight, follicular fluid volume, and follicular fluid (FF) estradiol (E2) content (P < .15), but similar ovarian weight and follicular fluid IGF-I concentration. Within the largest 15 preovulatory follicles, sows fed LL had a lower percentage of large (> or = 7.0 mm) follicles (33 vs 50 and 58%; P < .01) and a higher percentage of medium (5.5 to 7.0 mm) follicles (62 vs 44 and 39%; P < .01) but a similar percentage of small (< or = 5.5 mm) follicles (4.4 vs 5.9 and 3.7%; P > .15), respectively, compared with sows fed ML or HL. Standardized pools of oocytes aspirated from follicles of prepubertal gilts were incubated for 44 h with pooled FF recovered from the largest 15 follicles of each experimental sow. Fewer oocyte nuclei matured to metaphase II of meiosis when cultured with FF recovered from sows fed LL, than from sows fed ML or HL (47.1 vs 59.8 and 63.8%, respectively; P < .01). Our results suggest that low lysine (protein) intake in primiparous lactating sows impaired follicular development and reduced the ability of follicles to support oocyte maturation. However, high compared with medium lysine (protein) intake had no further positive effects on ovarian function.  相似文献

Large multiple cystic ovarian follicles were induced in three sows and small multiple cystic ovarian follicles were induced in three other sows by injections of adrenocorticotrophic hormone (ACTH) during the follicular phase of the oestrous cycle. Plasma progesterone levels in sows with large cysts were relatively high, while those of sows with small cystic follicles were low. Removal of the ovaries with large cysts from one sow resulted in a precipitous drop in progesterone levels indicating that the large cysts were primarily the source of progesterone. The method by which ACTH may induced cystic ovaries was also investigated. Plasma progesterone levels in two ovariectomised sows during ACTH treatment fluctuated markedly during a 24 h period achieving peak values of 4 or 5 ng/ml. The results suggest that progesterone of adrenal cortical origin may be a factor in the development of the cystic ovarian condition.  相似文献

The weaning to estrus and weaning to ovulation intervals in sows are controlled by ovarian follicular growth after weaning. Longer intervals could be caused by smaller diameter follicles at weaning that take more time to reach a preovulatory size. We addressed this hypothesis by decreasing the diameter of follicular populations before weaning and then measuring follicular development and interval to estrus and ovulation after weaning. The posterior vena cava, cranial to the entry of the ovarian vein, was cathetered for blood sampling and infusion in 20 sows at 12 +/- 1 d after farrowing. Sows were assigned randomly to receive either 30 mL of charcoal-treated follicular fluid (FF, n = 9; a treatment known to decrease serum FSH and follicular diameter) or 30 mL of saline (n = 11) by venous infusion thrice daily (0700, 1500, and 2300 h) for 96 h beginning at 14 +/- 1 d after farrowing. Sows were weaned 48 h after the last infusion. Blood samples were collected for FSH analysis thrice daily beginning on the day of catheterization and continuing until ovulation. Follicular diameter was determined once daily by transrectal ultrasonography. A treatment x time interaction was detected for serum FSH (P < 0.001) and follicular diameter (P < 0.001) because serum FSH and the diameter of follicular populations decreased in FF sows during the infusion period. After the infusion period, serum FSH rebounded in FF sows, and follicles resumed growth but grew at the same rate as those of saline-treated sows, thus failing to achieve equivalent diameters relative to saline-treated sows on a given day after weaning. As a result, sows treated with FF had longer (P < 0.05) weaning to estrus (6.1 +/- 0.4 d) and weaning to ovulation (8.6 +/- 0.5 d) intervals compared with saline-treated sows (4.7 +/- 0.4 d and 7.2 +/- 0.4 d, respectively). We conclude that the diameter of the follicular population at weaning is one factor that controls interval to estrus and ovulation in sows. Small follicles at weaning cannot undergo compensatory growth and require additional time to reach a preovulatory size.  相似文献

Matrix metalloproteinases (MMPs) play very important roles in extracellular matrix (ECM) remodeling during ovarian follicular development, ovulation and atresia. The aim of the present study was to determine the content of gelatinases in follicular fluid in various sized bovine follicles. Bovine ovaries were collected from local slaughterhouse and follicular fluid from follicles of 2 to over 25 mm in diameter was collected. Gelatinase activity within the follicular fluid was analyzed by gelatin zymography. The concentration of inhibin in the follicular fluid was also measured by immunoblot analysis. The proMMP-2 and alpha-subunit (alphaN) inhibin was detected in all follicles regardless of their size. The abundance of proMMP-2 varied with follicular size, while alphaN inhibin increased significantly (P<0.01) in follicles of 10-14 and 15-20 mm in size. There was a positive and negative correlation between estradiol (E(2)) and progesterone (P(4)) concentrations with abundance of proMMP-2, respectively. Follicles of diameter over 25 mm had greater proMMP-9 activity than other follicles. These same follicles had significantly (P<0.01) lower inhibin levels than follicles of 10-14 and 15-20 mm in size. In conclusion, these results suggest a significant role of these proteases in growth and development of bovine follicle, particularly proMMP-2 and active MMP-2 activities in the follicular fluid could serve as markers of follicular health while abundance of proMMP-9 may possibly denote a follicular cyst.  相似文献

Twenty primiparous sows were allocated to two treatments to evaluate the effects of maternal 25‐hydroxycholecalciferol (25OHD3) supplementation during gestation and lactation on milk quality and serum bone status markers of sows and bone quality of piglets. Immediately after mating, sows were randomly allotted to one of two diets supplemented with 50 µg/kg 25OHD3 or basal diets without 25OHD3. Blood and milk samples were obtained. At birth and weaning, 10 piglets from each treatment were killed for bone quality analysis. 25OHD3‐fed sows provided one more piglet at farrowing and 1.17 more piglets at weaning than sows fed basal diets. The contents of solids not‐fat, protein, fat or lactose were increased in milk from days 7 and 14 of lactation in 25OHD3‐supplemented sows and 25OHD3 concentrations in milk were increased by dietary 25OHD3 supplementation. Dietary 25OHD3 supplementation increased serum alkaline phosphatase activity but had no effect on serum tartrate‐resistant acid phosphatase activity of sows. Maternal 25OHD3 supplementation improved bone strength, density and ash content of newborn piglets rather than those of weaning piglets. In conclusion, 25OHD3 supplementation in maternal diets improved reproductive performance, milk quality and bone status of sows as well as bone quality of newborn piglets.  相似文献

Three experiments investigated ovarian follicular development in sows whose litters were weaned at 28 to 31 d of lactation. Unilateral ovariectomy near the time of weaning was used to assess early follicular characteristics and to identify those sows that would not return to estrus within 10 d after weaning. This allowed segregation of and exclusion from the study those sows that had a prolonged interval from weaning to first estrus. In Exp. 1, 82 and 72% of the large follicles that were marked at 48 or 72 h after weaning (10 sows per time point) were subsequently identified as corpora lutea. In Exp. 2, sows (seven to nine per time point) were unilaterally ovariectomized at 0, 6, 12, 18, 24, or 48 h after weaning, and follicular fluid was evaluated for changes in steroid concentrations. Progesterone concentrations in fluid from medium-sized (4 to 6 mm) follicles increased by 6 h after weaning and then declined through 24 h concomitant with increases in testosterone and estradiol. For Exp. 3, follicular fluid and granulosa cells from individual follicles were obtained from sows (seven to nine per time point) at 0, 6, and 24 h after weaning. In follicular fluid, insulin-like growth factor I (IGF-I) concentrations were not correlated (P greater than .05) with concentrations of progesterone, testosterone, or estradiol, or with granulosa cell production of estradiol during culture in androstenedione-supplemented medium.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The aim of the present study was to clarify the ovarian and hormonal dynamics after the aspiration of follicular fluid in cows with follicular cysts. Follicular fluid was aspirated from the follicular cysts and follicles that were fated to become cystic follicles and other coexisting normal follicles, respectively, in lactating cows (n = 3). After the aspiration procedure, new follicles developed and reached a diameter of 25 mm without ovulation within 13–19 days. The plasma concentrations of inhibin decreased and follicle-stimulating hormone increased rapidly after the aspiration procedure, and subsequently increased and decreased, respectively, as a new follicle grew. No luteal structures developed after the aspiration procedure, and the animals’ plasma progesterone levels remained low. The present study indicates that the cystic follicles are never luteinized by the aspiration of follicular fluid, and consequently, new follicular cysts are observed to repeatedly develop.  相似文献

Several studies have clarified that the follicular cysts degenerate and are replaced by newly growing follicles that develop into new follicular cysts without ovulation, i.e., turnover of ovarian follicular cysts in cows. However, the relativity of endocrinological changes, including the inhibin profile during turnover of spontaneous follicular cysts in dairy cows, is still unclear. In the present study, the relationship between turnover of follicular cysts and changes in the peripheral blood concentrations of progesterone (P), estradiol-17beta (E(2)), luteinizing hormone (LH), follicle stimulating hormone (FSH) and inhibin were examined in lactating dairy cows. Five cows diagnosed with follicular cysts (follicles of more than 25 mm in diameter in the absence of a corpus luteum) were investigated. Their ovarian dynamics were monitored using ultrasonography, and blood samples were collected at 2- or 3- day intervals throughout the experiment. The day when a follicle fated to become a follicular cyst reached more than 8 mm in diameter was defined as the start of a cystic follicular wave. Four of the 5 cows exhibited a similar patterns of cystic follicular changes and hormone profiles. The data from the 4 cows was used for analysis of the relationships between turnover of cystic follicles and the hormone profiles. Two or three new cystic follicular waves occurred in each cow during the experimental period. The mean diameter of the cystic follicles was more than 25 mm 13 to 15 days after the start of the cystic follicular wave, and it began to decrease 1 to 6 days before the start of the subsequent cystic follicular wave. The levels of E(2) and inhibin tended to decrease for 7 to 9 days before the start of a new cystic follicular wave and to increase concomitantly with new follicular cyst growth. The levels of FSH rose for 1 to 3 days before the start of a new cystic follicular wave. The present study clarified the relationship between FSH and inhibin during turnover of spontaneous follicular cysts in dairy cows and found that it was very similar to previous results for cows. The present results suggest that an increase in FSH secretion following a reduction in inhibin secretion triggers turnover of cystic follicles in cows with spontaneous follicular cysts.  相似文献

Ovarian follicular cysts are a major reproductive problem in lactating dairy cows. The primary physiological defect leading to the formation of ovarian follicular cysts is a failure of the hypothalamus to trigger the preovulatory surge of luteinizing hormone (LH) in response to estradiol. The factor responsible for this hypothalamic defect may be progesterone. Intermediate levels of progesterone have been shown to prevent ovulation and promote persistence of dominant follicles in normal cycling cows. Recently, we found that 66% of cows with ovarian follicular cysts had progesterone concentrations in an unusual, intermediate range (0.1-1.0 ng/mL) at the time of their detection. A majority of new follicles (76%) that develop in the presence of these intermediate progesterone concentrations became cysts. Only 10% ovulated. Based on these observations, a novel model for the formation and turnover of ovarian follicular cysts is proposed.  相似文献

One hundred Yorkshire × Landrace sows were randomly assigned to one of two dietary treatments (diet ND: 6,000 IU vitamin D3/d feed; diet 25‐D: 200 μg/day 25OHD3 feed). The experiment began on d 90 of gestation and continued until weaning on day 21 of lactation. In sows that received 25OHD3, the growth rate of the piglets before weaning was significantly accelerated (0.266 kg/day, p < .05). Sow serum was collected after weaning, and those in the 25OHD3 group were found to have significantly higher serum calcium (CA) and phosphorus (PI) levels (p < .05). Interestingly, the oestrus cycle of sows fed 25OHD3 was significantly shortened (p < .05), the oestrus time was concentrated on the fifth day after weaning, and the piglets were born with a higher degree of uniformity (p < .05). Colostrum was collected on the day of delivery, and the colostrum of sows fed 25OHD3 contained higher milk fat content than the control group (p < .05). 25OHD3 supplementation increased the mRNA and protein expression of INSIG1 and SREBP1, which regulate milk fat synthesis, in the mammary gland of lactating sows (p < .05). In conclusion, 25OHD3 supplementation in maternal diets improved reproductive performance, milk fat content and the mRNA and protein levels of genes regulating milk fat synthesis in lactating sows.  相似文献

Impaired reproductive performance exhibited by the domestic sow during the late summer and early autumn months is referred to as seasonal infertility. This study was carried out to determine whether there are changes in ovarian morphology and follicular steroidogenesis associated with season, which may be associated with seasonal infertility. Ovaries were collected in pairs from sows sourced from two farms and slaughtered 4 days after weaning during winter and summer. The mean progesterone concentration in follicular fluid (FF) collected from small follicles was lower in summer (701.3 ± 115.54 nm) compared with winter (1235.55 ± 164.47 nm; p<0.001). The mean progesterone concentration in the FF of large follicles was also lower in summer (1469.2 ± 156.51 nm) compared with winter (2470.9 ± 169.13 nm; p<0.001). The number of large surface antral follicles (5-8 mm in diameter) on the ovaries recovered from Farm A sows was higher during summer (17.76 ± 0.56) than in winter (15.38 ± 0.54; p<0.05). Similarly, the number of small follicles (3-4 mm in diameter) on Farm A sow ovaries was higher in summer (8.46 ± 0.66) than in winter (4.63 ± 0.53; p<0.001). In contrast, the number of small follicles on the surface of ovaries recovered from Farm B sows was higher during winter (10.17 ± 1.50) than in summer (6.45 ± 1.00; p<0.01). The number of pre-ovulatory follicles (>8 mm in diameter) was also higher in winter (1.23 ± 1.68) when compared to summer (0.51 ± 0.3; p<0.001) on the ovaries of sows from Farm B. The results suggest that there are seasonal differences in follicular steroidogenesis and ovarian dynamics. These findings add support to the theory that altered follicular steroidogenesis and ovarian morphology may possibly be the mechanism behind reduced reproductive performance during the period of seasonal infertility in sows.  相似文献

The aims of this study were to assess the accuracy of different common methods of differentiating between follicular and luteal ovarian cysts, and to monitor the response of the cysts to 12 days treatment with a progesterone-releasing intravaginal device (PRID). On the basis of agreement between the different methods, 25 of the 46 cases examined were diagnosed as follicular and 14 as luteal cysts; for the other seven cases the methods disagreed. The use of ultrasound was more accurate in diagnosing follicular cysts than luteal cysts, and combined with plasma progesterone concentrations gave the most accurate assessment of cyst type (92 per cent for follicular cysts and 82 per cent for luteal cysts). The mean (se) plasma progesterone concentration was lower in the cows with follicular cysts than in those with luteal cysts (0.29 [0.05] v 3.90 [0.63] ng/ml; P<0.05). Luteal cysts had thicker walls (5.3 [0.04] v 2.5 [0.2] mm; P<0.0001), and the wall thickness of all the cysts was positively correlated with plasma progesterone concentration (r=0.52, P<0.0004). Cows with luteal cysts had more additional follicles greater than 5 mm in diameter (P<0.01). In cows with follicular cysts and other follicles greater than 5 mm in diameter, the mean oestradiol concentration was 7.9 (1.8) pg/ml compared with 24.2 (3.1) pg/ml (P=0.002) in cows without other follicles greater than 5 mm in diameter on either ovary. At the time of PRID removal, plasma progesterone concentration had increased in the cows with follicular cysts to 1.59 (0.06) ng/ml (P<0.05) and decreased in the cows with luteal cysts to 0.87 (0.01) ng/ml (P<0.05), although there was no change in original cyst structure in 45 per cent of the cases. However, new ovarian structures were frequently observed during the treatment. The overall pregnancy rate for cows with both types of cyst after treatment was 50 per cent after three inseminations, but the first service pregnancy rate was only 18 per cent for cows with follicular cysts and 28 per cent for cows with luteal cysts. After treatment, the fertility of cows with follicular cysts was similar to that of paired herdmates, whereas cows with luteal cysts took 40 days longer to calve again than healthy herdmates. However, the culling rate was higher for cows with follicular cysts (41 v 11 per cent).  相似文献

Dietary supplementation with 25-hydroxyvitamin D3 (25OHD3), as an alternative source of vitamin D, is becoming increasingly popular due to its commercialization and more efficient absorbability. The addition of 25OHD3 rather than its precursor vitamin D3 can circumvent the 25-hydroxylation reaction in the liver, indicating that supplementation of 25OHD3 can rapidly improve the circulating vitamin D status of animals. Emerging experiments have reported that maternal 25OHD3 supplementation could increase sow performances and birth outcomes and promote circulating vitamin D status of sows and their offspring. Increased milk fat content was observed in many experiments; however, others demonstrated that adding 25OHD3 to lactating sow diets increased the contents of milk protein and lactose. Although an inconsistency between the results of different experiments exists, these studies suggested that maternal 25OHD3 supplementation could alter milk composition via its effects on the mammary gland. Previous studies have demonstrated that adding 25OHD3 to sow diets could improve the mRNA expressions of insulin-induced gene 1 (INSIG1) and sterol regulatory element-binding protein 1 (SREBP1) in the mammary gland cells from milk and increase the mRNA expressions of acetyl-CoA carboxylase α (ACCα) and fatty acid synthase (FAS) in the mammary gland tissue. Maternal 25OHD3 supplementation promotes skeletal muscle development of piglets before and after parturition, and improves bone properties including bone density and bone breaking force in lactating sows and their piglets. Interestingly, 25OHD3 supplementation in sow diets could improve neonatal bone development via regulation of milk fatty acid composition related to bone metabolism and mineralization. In this review, we also discuss the effects of adding 25OHD3 to sow diets on the gut bacterial metabolites of suckling piglets, and propose that butyrate production may be associated with bone health. Therefore, to better understand the nutritional functions of maternal 25OHD3 supplementation, this paper reviews advances in the studies of 25OHD3 for sow nutrition and provides references for practical application.  相似文献

Comparisons of numbers of antral ovarian follicles and corpora lutea (CL), of blood hormone concentrations, and of follicular fluid steroid concentrations and IGFBP activity were conducted between cows selected (twinner) and unselected (control) for twin births to elucidate genetic differences in the regulation of ovarian follicular development. Ovarian follicular development was synchronized among cows by a single i.m. injection of PGF2alpha on d 18 of the estrous cycle; six cows per population were slaughtered at 0, 24, 48, and 72 h after PGF2alpha. Jugular vein blood was collected from each animal at PGF2alpha injection and at 24-h intervals until slaughter. Ovaries of twinner cows contained more small (< or = 5 mm in diameter, P < 0.05), medium (5.1 to 9.9 mm, P < 0.05), and large (> or = 10.0 mm, P < 0.01) follicles and more (P < 0.01) CL than ovaries of controls. Follicular fluid concentrations of estradiol, androstenedione, testosterone, and progesterone reflected the stage of follicular development and were similar for twinner and control follicles at the same stage. Earlier initiation of follicular development and/or selection of twin-dominant follicles in some twinner cows resulted in greater concentrations of estradiol in plasma at 0, 24, and 48 h and of estradiol, androstenedione, and testosterone in follicular fluid of large follicles at 0 h after PGF2alpha for twinner vs. control cows (follicular status x time x population, P < 0.01). Binding activities of IGFBP-5 and -4 were absent or reduced (P < 0.01) in follicular fluid of developing medium and large estro-gen-active (estradiol:progesterone ratio > 1) follicles but increased with atresia. Only preovulatory Graafian follicles lacked IGFBP-2 binding, suggesting a possible role for IGFBP-2 in selection of the dominant follicle. Concentrations of IGF-I were twofold greater (P < 0.01), but GH (P = 0.10) and cholesterol (P < 0.05) were less in blood of twinners. Three generations of selection of cattle for twin ovulations and births enhanced ovarian follicular development as manifested by increased numbers of follicles within a follicular wave and subsequent selection of twin dominant follicles. Because gonadotropin secretion and ovarian steroidogenesis were similar for control and twinner cattle, enhanced follicular development in twinners may result from decreased inhibition by the dominant follicle(s), increased ovarian sensitivity to gonadotropins, and/or increased intragonadal stimulation, possibly by increased IGF-I.  相似文献

Cystic ovarian disease (COD) is an important cause of infertility in cattle, and ACTH has been involved in regulatory mechanisms related to ovarian function associated with ovulation, steroidogenesis, and luteal function. Here, we examined the localization of 11β-hydroxysteroid dehydrogenase type 1 (11βHSD1) and 11βHSD2 proteins in the ovary of healthy cows and animals with spontaneous and ACTH-induced COD and the in vitro response of the follicular wall exposed to ACTH. After stimulation by ACTH, we documented changes in 11βHSD expression and cortisol secretion by the follicular wall of large antral and follicular cysts. Follicular cysts showed a higher constitutive expression of both enzymes, whereas ACTH induced an increase in 11βHSD1 in tertiary follicles and follicular cysts and a decrease in 11βHSD2 in follicular cysts. Moderate expression of 11βHSD1 was observed by immunohistochemistry in granulosa of control animals, with an increase (P < 0.05) from primary to secondary, tertiary, and atretic follicles. The level of immunostaining in theca interna was lower than that in granulosa. The expression of 11βHSD2 was lower in the granulosa of primary follicles than in that of secondary, tertiary, and atretic follicles and was lower in the theca interna than in the granulosa. In ACTH-induced and spontaneously occurring follicular cysts, differences from controls were observed only in the expression of 11βHSD1 in the granulosa, being higher (P < 0.05) than in tertiary follicles. These findings indicate that follicular cysts may be exposed to high local concentrations of active glucocorticoids and indicate a local role for cortisol in COD pathogenesis and in regulatory mechanisms of ovarian function.  相似文献

The expression of growth factors was evaluated immunohistochemically in normal and cystic ovaries of sows. The immunohistochemically stained area (IHCSA) was quantified by image analysis to analyse the expression of these proteins in the follicular wall of secondary, tertiary and cystic follicles. IGF‐I immunoreactivity was strong in the granulosa cell layer (GC), moderate in the theca interna (TI) and mild in the theca externa (TE) of the normal follicles. There was severe reduction of the labelling to IGF‐I in the GC of the follicular and luteinized cysts. In the normal follicles, the reactivity for IGF‐II was very similar to pattern noted in IGF‐I. There was reduction of the IHCSAs in the GC of the follicular and luteinized cysts, but the decrease was not significant. The staining of the IGF‐II in the TI and TE of the cysts was increased, in comparison with normal follicles. The IHCSAs for VEGF were higher in the GC and TE of the normal follicles in contrast to TI, but this difference was noted only in the tertiary follicle. The VEGF reactivity increased in the GC of the cysts, in relation to normal follicles. The results of the current study show that the formation of ovarian cysts in sows is associated with alterations in the immunohistochemical expression of some growth factors.  相似文献

Cystic ovarian disease (COD) is one of the main causes of infertility in dairy cattle. It has been shown that intra‐ovarian factors, such as members of the insulin‐like growth factor (IGF) system, may contribute to follicular persistence. The bioavailability of IGF to initiate its response by binding to specific receptors (IGFRs) depends on interactions with related compounds, such as pregnancy‐associated plasma protein A (PAPP‐A). The aim of this study was to determine IGFR1 and PAPP‐A expression both in follicles at different stages of development and in cysts, to evaluate the roles in the etiopathogenesis of COD in cattle. The mRNA expression of PAPP‐A was higher in granulosa cells of large tertiary follicles than in cysts, whereas the protein PAPP‐A present in the follicular fluid from these follicles showed no differences. Although no PAPP‐A mRNA expression was detected in smaller tertiary follicles, in their follicular fluid, this protease was detected in lesser concentration than in cysts. The mRNA expression of IGFR1 was lower in granulosa cells from cystic follicles than in those from tertiary ones. However, the protein expression of this receptor presented the highest levels in cystic structures, probably to increase the possibility of IGF response. The data obtained would indicate that animals with COD have an altered regulation of the IGF system in the ovary, which could be involved in the pathogenesis of this disease in cattle.  相似文献
