[Objective] Study on the genetic diversity in wild populations of Poacynum hendersonii.[Method] Random amplified polymorphic DNA(RAPD)technique was employed to analyze the genetic diversity in five wild populations of P.hendersonii sampled from Xinjiang,Gansu and Qinghai provinces.[Result] Totally 165 clear and repeatable bands were generated in RAPD reaction by using 20 primers screened from 80 primers,of which 110 were polymorphic,accounting for 66.67%.At species level,Nei’s gene diversity index(H),Shannon’s information index(I)and genetic differentiation coefficient(Gst)were 0.220 5,0.304 7 and 0.908 2,respectively.P.hendersonii germplasm resources share a high level of genetic diversity,and genetic differentiation mainly exists among the populations.Results from genetic distances and cluster analysis showed that relationships among P.hendersonii populations were to some extent related with their geographical and climatic characters.[Conclusion] This study suggests that the conservation of P.hendersonii should focus on the protection of many populations,particularly the Qinghai population.  相似文献

To investigate the genetic diversity of an edible fungus Pleurotus ferulae, a total of 89 wild samples collected from six geographical locations in the Xinjiang Uygur Autonomous Region of China and two geographical locations in Italy, were analyzed using three DNA fragments including the translation elongation factor(EF1α), the second largest subunit of t he RNA polymerase II(RPB2) and the largest subunit of the RNA polymerase II(RPB1). The results indicated relatively abundant genetic variability in the wild resources of P. ferulae. The analysis of molecular variance(AMOVA) showed that the vast majority of the genetic variation was found within geographical populations. Both the Chinese populations and the Italian populations of P. ferulae displayed a limited genetic differentiation. The degree of differentiation between the Chinese populations and the Italian populations was obviously higher than that between the populations from the same region, and moreover the genetic differentiation among all the tested populations was correlated to the geographical distance. T he phylogeny analyses confirmed that samples from China and Italy belonged to another genetic group separated from Pleurotus eryngii. They were closely related to each other but were clustered according to their geographical origins, which implied the Chinese populations were highly differentiated from the Italian populations because of distance isolation, and the two populations from different regions might be still in the process of allopatric divergence.  相似文献

Genetic Variation of Host Populations of Liriomyza sativae Blanchard   总被引:1,自引:0,他引:1  
In this study, partial sequences of the mitochondrial cytochrome oxidase subunit I (mtDNA-COI) gene and the ribosomal internal transcribed spacer 1 (rDNA-ITS 1) gene, isolated from five artificial populations of Liriomyza sativae (Diptera: Agromyzidae), were sequenced and compared, to analyze their genetic variation. Analysis of the mtDNA-CO1 gene showed that a low genetic variation was detected among the five populations and only five variable sites were found in the nucleotide sequences. Most of the observed variations that occurred within the populations were because of nucleotide transitions, whereas, the interpopulation variation was because of the differences in haplotype frequencies occurring among the host populations. Analysis of the rDNA-ITS1 gene revealed a small diversity in the five host populations. The trend of genetic differentiation in the host populations was consistent with the preference of L. sativae to the plant hosts.  相似文献

It was helpful for the wheat improvement to evaluate the genetic resources of Triticum turgidum L. ssp. turgidum landraces. In this study, 68 turgidum landraces accessions, belonging to four geographic populations in China, were investigated by using EST-SSR markers. A total of 63 alleles were detected on 22 EST-SSR loci, and the number of alleles on each locus ranged from 1 to 5, with an average of 2.9. The results of the analysis of molecular variance (AMOVA) indicated that 92.5% of the total variations was attributed to the genetic variations within population, whereas only 7.5% variations among populations. Although the four populations had similar genetic diversity parameters, Sichuan population was yet distinguished from other populations when comparing the population samples in pairs. Significant correlations were detected by the statistic analysis among six genetic diversity parameters among each other. The selection difference between heterozygosty and homozygosty was also observed among different EST-SSR locus. The genetic similarity (GS) ranged from 0.18 to 0.98, with the mean of 0.72, and all accessions could be clustered into 7 groups. The dendrogram suggested that the genetic relationships among turgidum accessions evaluated by EST-SSR markers were unrelated to their geographic distributions. These results implied that turgidum landraces from China had the unique characters of genetic diversity.  相似文献

Invasion of the oriental fruit lfy, Bactrocera dorsalis, into new niches containing different food sources (a process referred to as host shift), may cause population genetic differentiation and sympatric speciation. To attempt to infer that experimentally, test populations were established by transferring a subset of the original populations, which had been grown on banana for many generations, onto navel orange, and then subculturing the navel orange population and banana population for at least 20 generations. Four pairs of SSR primers with high polymorphism on laboratory strains were used to detect population genetic differentiation. All six tested populations (the 5th, 10th and 15th generations of B. dorsalis fed on banana and navel orange, respectively) were found to have low genetic diversity. Furthermore, the genetic diversity of the navel orange populations was found to decline after being crossed for several generations. Populations initially were deviated from Hardy-Weinberg equilibrium, however, equilibrium was achieved with increasing numbers of generations in both of the host populations. Limited gene lfows were found among the six populations. The Nei’s standard genetic distances between the two host populations of the same generation were initially low, but increased with generation number. Genetic distances between banana and navel orange populations of the same generation were lower than genetic distances between different generations grown on the same host plant. Analysis of molecular distance (AMOVA) results based on generation groups and host groups demonstrated that genetic variation among generations was greater than that between the two host populations. The results indicated that population genetic differentiation occurred after the host shift, albeit at low level. Biogeography and taxonomy of the B. dorsalis complex revealed that speciation of B. dorsalis might be tightly associated with host shift or host specialization of B. dorsalis following dispersal.  相似文献

Genetic diversity of three cultured populations of jade perch (Scortum bacoo) are studied using amplified fragment length polymorphism (AFLP) markers, which is Guangzhou(GZ) population, Foshan (FS) population and Qingdao(QD) population. Nine primer combinations were used and 385 fragments were detected. Among the 385 fragments, 80 bands (20.78%) were polymorphic. And it can be speculated that the genetic diversity of the three cultured populations of jade perch was very poor according to the gene genetic diversity among populations (Ht), the gene genetic diversity within populations (Hs) and Shannon-Weiner index (I). From gene differentiation (Gst), genetic distance (D), genetic similarity (5), and UPGMA analysis, it is found that the relationship among the three populations is very closed, and the difference in genetic diversity among the three populations is not significant. Further clustering relationships of the jade perch cultured populations also are correlated to historical-breeding observations and genetic relationships. The GZ population clustered together with the QD population first, then with FS population.  相似文献

Genetic relationship among Oxya chinensis populations and Oxya japonica populations collected from Xuzhou City, Jiangsu Province and Pingshan County, Hebei Province, China were analyzed by random amplified polymorphic DNA (RAPD) markers. A total of 125 DNA bands ranging from 200 to 2 200 bp were amplified by 10 random primers in DNA samples from 43 grasshopper individuals. One hundred and twenty-three (99%) of these bands were polymorphic. Shannon's index showed that the genetic, diversity within O. chinensis (0.3432) was higher than that of O. japonica (0.2781). Nei's genetic distance between O. chinensis population and O. japonica population from the same area was less than that between populations from two different areas. The dendrogram based on Nei's genetic distance of RAPD markers was constructed using the unweighted pair group method with the arithmetic average (UPGMA) and Neighbor-Joining (NJ) methods. Cluster analysis indicated that all the individuals were grouped into two main clusters. O. chinensis populations from Xuzhou and Pingshan formed one cluster, and O. japonica populations from the two regions belonged to another cluster. The results demonstrated that RAPD can detect within species, and among closely related species. polymorphisms to distinguish minor difference among individuals  相似文献

Aphis glycines(Hemiptera: Aphididae) is considered as a cosmopolitan pest of cultivated soybean, major diffi culties in its control measures may be due to its higher genetic diversity; however, the knowledge about population genetic diversity of this species is limited. This study aimed to represent the genetic differentiation among different geographic populations of soybean aphid in Northeast China. In order to investigate and assess the genetic diversity, genetic differentiation, molecular variance, population structure, ecological importance and evolutionary history of A. glycines, we sequenced a fragment of one protein-coding gene, the cytochrome c oxidaseⅡof mitochondrial DNA gene. The results showed that four haplotypes were defi ned among COⅡ gene of 180 sequences of soybean aphid in Northeast China including H1 shared by all the populations. Lower haplotype diversity(Hd=0.3590±0.0420) and nucleotide diversity(Pi=0.0012±0.0002) were observed and high gene flow was detected in every two populations, while most of the variation(80.81%) arose from variability within A. glycines from individuals. Low genetic differentiation and high gene fl ow(Nm=2.106) indicated a high migration rate between the populations, which might reveal that gene flow in different geographic populations did not affect by geographical distance. The phylogenetic tree and the haplotype network of A. glycines were obtained based on sequences of COⅡ gene, there were no signifi cant genealogical branches or clusters recognized in NJ tree, and no clear distribution, delineation of haplotypes were demonstrated in the haplotype network according to geographical location. This study rejected the vicariance hypothesis: geographic isolation could be a barrier and it restricted A. glycines gene fl ow among 10 populations.  相似文献

The present study was conducted to assess the molecular characterization and genetic diversity amongst natural populations of Myrica rubra in Guangxi Zhuang Autonomous Region, China, thus to provide scientific evidence for germplasm conservation and exploitation. Using ISSR (inter-simple sequence repeats) markers, the level of genetic variation and the molecular characterization of 10 natural populations of M. rubra, originated from Guangxi Zhuang Autonomous Region in China, were performed. Based on 11 primers, 123 clear and reproducible DNA fragments were generated, of which 95 (77.24%) were polymorphic. The average value of Nei's gene diversity (He) was 0.268. The coefficient of genetic differentiation (Gst) was 0.341, revealing that 34.1% of the total molecular variance existed among populations. The Mantel statistical testing showed that the genetic distance was correlated to the geographic distance, but the correlation was not significant. Ten populations were divided into two big clusters according to unweighted pair group method with arithmetic mean (UPGMA) analysis. One consisted of populations of Rongxian (RX), Hepu (HP), Liangqing (LQ), Marshan (MS), Lingshan (LS) and Shansi (SS), which originated from the southern Guangxi, while the other was composed of Guanyang (GY) and Lingui (LG) populations of northern Guangxi, Huanjiang (HJ) populations of northwestern Guangxi and Shanglin (SL) populations of southern Guangxi. The level of genetic variation in wild M. rubra population distributed in Guangxi is high. Gene drift within the population was responsible for genetic variation in wild M. rubra in Guangxi, and the effect of the genetic flow among inter-populations was not significant. Classification of wild M. rubra populations was correlated to climate and environment. The molecular characterization and diversity assessment of M. rubra is of immense value for planning conservation of its genetic resources and their exploitation for further studies.  相似文献

Ten temperate and adapted exotic breeding populations of maize were studied with electrophoretic techniques. Three isozyme systems coded by nine allozyme loci were used for evaluating the genetic variability within and among populations. The results revealed that 78.57% of allozyme loci were polymorphic. Low allelic variation with a mean number of 1.84 alleles per locus per population was detected. But, these populations still maintained higher level of heterozygosity; moreover, the exotic populations had greater gene diversity than the temperate populations. All the populations were non-panmictic with negative Wright's fixation indexes (-0.091— -0.424). The tropical BS 16 was typified by maximum allelic richness, percent of polymorphic loci and heterozygosity. More than 93% of the gene diversity maintained within populations, and the genetic differentiation among populations was low (0.002—0.191). Multivariate analysis demonstrated that the tropical BS29 diverged from other populations in the reverse direction. The temperate BS9 and tropical BS16 were divergent each other, and highly differentiated from other temperate and tropical populations, consequently, these two populations would be analogically postulated as potential germplasms to establish new heterotic groups for temperate maize breeding programs.  相似文献

Two cycles of biparental mass selection (MS) and one cycle of half-sib-S3 family combining selection (HS-S3) for yield were carried out in 2 synthetic maize populations P4C0 and P5C0 synchronously. The genetic diversity of 8 maize populations, including both the basic populations and their developed populations, were evaluated by 30 SSR primers. On the 30 SSR loci, a total of 184 alleles had been detected in these populations. At each locus, the number of alleles varied from 2 to 14, with an average of 6.13. The number and ratio of polymorphic loci in both the basic populations were higher than those of their developed populations, respectively. There was nearly no difference after MS but decreased after HS-S3 in both the basic populations in the mean gene heterozygosity. The mean genetic distance changed slightly after MS but decreased in a bigger degree after HS-S3 in both the basic populations. Analyses on the distribution of genetic distances showed that the ranges of the genetic distance were wider after MS and most of the genetic distances in populations developed by HS-S3 were smaller than those in both the basic populations. The number of genotypes increased after MS but decreased after HS-S3 in both the basic populations. The genetic diversity of intra-population was much more than genetic diversity of inter-population in both the basic populations. All these indexes demonstrated that the genetic diversity of populations after MS was similar to their basic populations, and the genetic diversity was maintained during MS, whereas the genetic diversity of populations decreased after HS-S3. This result indicated that heterogeneity between some of the individuals in the developed populations increased after MS, whereas the populations become more homozygotic after HS-S3.  相似文献

Sweetpotato, Ipomoea batatas(L.) Lam., is an important food crop widely cultivated in the world. Evaluation of genetic relationships among diverse cultivars and landraces is necessary for efficient exploitation of genetic diversity in the existing germplasm resources. In the present study, a collection of 380 sweetpotato accessions assembled from different agro-climatic zones of China and other countries were genotyped using 30 SSR primer pairs. Model-based structure analysis separated the germplasm into three populations, P1, P2 and P3, containing 228, 133 and 19 accessions, respectively, which was consistent with the results of phylogenic and principal component analysis(PCA). Analysis of molecular variance(AMOVA) revealed significant genetic differentiation among inferred populations, accounting for 16.47% of the total molecular variance, however, the differences between the regions were not significant, the total variation were due to the differences between the genotypes within the population. Pairwise fixation index(F ST) suggested that populations P1 and P3 had the highest differentiation, while populations P1 and P2 had the lowest differentiation. The diversity among populations was wide, which confirmed the genetic distinction of populations. Through comparing model-based structure and domestication-based classification, it was found that the accessions of population P1 mainly belonged to modern cultivars, and the accessions of populations P2 and P3 basically corresponded to landraces, by which we suggest that modern cultivars maybe had experienced a two-step domestication history. Our results illustrated clear genetic relationships among 380 sweetpotato accessions, exhibiting the potential of accelerating the process of future sweetpotato breeding program by molecular marker based parental selection.  相似文献

Psathyrostachys huashanica Keng is endemic to China and only distributed in Huashan Mountain in Shaanxi Province, China. In this study, 15 P. huashanica populations consisting of 450 individuals sampled across their main distribution were investigated by using the simple sequence repeats (SSRs) markers. A total of 184 alleles were detected on 24 SSR loci, and the number of alleles on each locus ranged from 2 to15, with an average of 7.667. The total gene diversity (HT= 0.683) and the coefficient of population differentiation (GST = 0.125) showed that P. huashanica had a relatively high level of genetic variation, and the genetic variation was mainly distributed within the populations. The gene flow among the populations of P. huashanica (Nm = 1.750) was much less than that of the common anemophytes (Nm = 5.24). Correlation analysis demonstrated that the number of alleles as well as genetic diversity of the five populations of Huangpu valley decreased along with the increase of altitudes, but the correlation was not significant. Implications of these results for future P. huashanica collection, evaluation and conservation were discussed.  相似文献

Bemisia tabaci (Gennadius) biotypes B and Q are two invasive biotypes in the species complex. The comparison of the population genetic structure of the two biotypes is of significance to show their invasive mechanism and to their control. The intersimple sequence repeats (ISSR) marker was used to analyze the 16 B-biotype populations and 4 Q-biotype populations worldwide with a Trialeurodes vaporariorum population in Shanxi Province, China, and a B. tabaci non-B/Qbiotype population in Zhejiang Province, China, was used as control populations. The analysis of genetic diversity showed that the diversity indexes of biotype Q including Nei's gene diversity index, Shannon informative index, and the percentage of polymorphic loci were higher than those of biotype B. The high genetic diversity of biotype Q might provide the genetic basis for the excellent ecological adaptation. Cluster analysis suggested that the ISSR could not be used in the phylogenetic analysis though it could easily distinguish the biotypes of B. tabaci. The difference of the population genetic structure between the biotype B and the biotype Q exists based on the ISSR marker. Meanwhile, the results suggested that the molecular marker has its limitation in the phylogenetic analysis among the biotypes of B. tabaci.  相似文献

To estimate genetic variation in rhizome lotus (Nelumbo nucifera Gaertn. ssp. nucifera) germplasms in China, a total of 94 rhizome lotus germplasms collected from 18 provinces in China were assessed. The RAPD (randomly amplified polymorphic DNA) marker was employed. The selected 17 random primers detected 139 polymorphic alleles out of a total 207 (67.15%). Nei's gene diversity statistics and region differentiation parameters indicated that all germplasms had a relatively high level of genetic diversity with ne = 1.3202, h = 0.1937, I= 0.2982 and the gene flow among all regions was Nrn = 5.5742. The UPGMA dendrogram clustered all 94 germplasms into two clusters: One contained eight commercial cultivars and major landraces, and the other included the wild and some special landraces from five regions, and the PCA analysis exhibited the similar result. Those germplasms from southwestern and eastern China had higher genetic diversity than those from the southern, northern and central China. Predominant proportion of genetic variation (95.61%) was found significant within rather than among (4.39%) regions, as revealed by AMOVA analysis. The data analysis also revealed that the genetic diversity of rhizome lotus germplasms among different regions is positively related to their geographic distances, though it is ambiguous to find the trend from the UPGMA dendrogram and the PCA analysis. A relatively high genetic diversity and gene flow resided in the root lotus germplasms; about 96% of the variation was found within region; accessions from southwest and eastern China have higher genetic diversity than those from the southern, northern and central China.  相似文献

Bulk-SSR method was used to analyze the genetic diversity of 44 open-pollinated varieties collected from Henan, Shandong, Shanxi, and Jilin provinces and Guangxi Zhuang Autonomous Region, China using 70 pairs of SSR primers. The purposes of this study were to (1) compare the genetic diversity among 44 Chinese maize open-pollinated varieties; (2) estimate the minimum number of alleles for construction of a stable dendrogram; and (3) trace the genetic relationships among local germplasm from different regions of China. In total, these 70 SSR primers yielded 292 alleles in 176 samples (4×44) analyzed. The number of alleles per locus was 4.17 on average and ranged from 2 to 8. The highest number of alleles per open-pollinated variety (55.25) was detected in Shanxi germplasm, which indicated that open-pollinated varieties from Shanxi possessed the largest genetic diversity among those from the five locations. The correlation coefficients between different genetic similarity matrices suggested that 200 alleles were sufficient for analysis of the genetic diversity of these 44 open-pollinated varieties. The cluster analysis showed that 44 open-pollinated varieties collected from three growing regions in China were accurately classified into three groups that were highly consistent with their geographic origins, and there is no correlation between GS and geographic distance in this study.  相似文献

Chloroplast inter-simple sequence repeat markers in mango were developed and used to analyze the genetic relationship and diversity of mango and its relatives. Thirty-six mango cultivars (Mangifera indica L.) and its relative species collected from the fruit germplasm collection in the Guangxi Academy of Agricultural Sciences, China, were examined by ISSR-PCR with chloroplast DNA (cpDNA). Eight better primers for chloroplast DNA that provided reproducible, polymorphic DNA amplification patterns were screened from 50 ISSR primers and used for UPGMA analysis. According to the band patterns with 8 primers for chloroplast DNA, all cultivars tested were distinguished from each other and these showed ample genetic diversity; the average percentage of polymorphism was 77.2%. The 36 samples could be clustered into four groups by UPGMA analysis at the coefficient 0.74. The results indicated that the cplSSR marker was a new powerful tool for the identification of mango cultivars or its relative species, and their genetic relationship analysis and diversity evaluation.  相似文献

Variations of structural loci among 4 sheep populations in China were examined by the method of multiloci electrophoresis, and similar data from 11 sheep populations were taken as basic references to analyze the genetic structure of the native sheep populations in East and South Asia. The results showed that the average heterozygosity and effective number of alleles among 15 populations were 0.2746 and 1.559, respectively. Mongolian sheep possessed the largest average heterozygosity and effective number of alleles. Genetic diversity of sheep populations in Mongolia, China, Vietnam, Bangladesh and Nepal was reduced in this order. The coefficients of genetic differentiation were between 0.0126 and 0.3083, with the average of 0.148, demonstrating that genetic variations lay mainly in populations with 85.2% of the total variations. There exists no correlation between geographical distances and genetic distances. Gene flow was smooth among most populations, which led to inconsistency between geographical distances and genetic distances. The 15 native sheep populations in East and South Asia could be divided into two groups: One group included part populations of China and Mongolia, and the other included Yunnan populations of China, and part populations of Nepal and Bangladesh. Other populations did not cluster together and divide into the above-mentioned two groups. Tills study indicated genetic differentiation of the 15 native sheep populations in East and South Asia was relatively low, geographical isolation was not the main reason affecting genetic differentiation, and the fifteen sheep populations could be divided into two groups according to phylogenetic relationships.  相似文献
