The composition of non-infected and Colletotrichum nymphaeae infected strawberry fruits (cultivar ‘Asia') in three different stages of ripeness were evaluated. Additionally, the effect of infection on strawberry runners was monitored. C. nymphaeae infection caused a significant increase in total sugar content in mature fruits (1.1 fold) and a decrease in total sugar content in semi-mature fruits (1.4 fold) and organic acid content in all stages of ripeness (1.7 fold). Ellagic acid derivatives (1.9 fold), flavanols (1.5 fold) and flavonols (5.1 fold) significantly increased during all stages of ripeness after infection. The pattern of phenolic accumulation in strawberry runners was altered by C. nymphaeae infection in contrast to strawberry fruits as most of identified phenols decreased as a response to the infection.  相似文献

The phenolic profile of healthy and infected raspberry canes was investigated in three raspberry cultivars: Autumn Bliss, Himbo Top and Polka. The content of total phenols and tannins was determined using spectrophotometric methods, whilst individual phenolic acids, flavan‐3‐ols, ellagic acid derivatives and glycosides of quercetin were analysed using HPLC/MS analysis. The content of secondary metabolites varied considerably among the analysed raspberry cultivars. Moreover, Didymella applanata and Leptosphaeria coniothyrium infection significantly altered the metabolism of phenolic compounds. Flavanols represented the greatest share of all identified phenolics in raspberry canes (90%), followed by glycosides of quercetin (6%), derivatives of ellagic acid (3%) and traces of hydroxycinnamic acid derivatives. Spur and cane blight diseases caused an increase of flavan‐3‐ols and tannins but the levels of hydroxycinnamic acid derivatives, conjugates of ellagic acid and quercetin glycosides were significantly reduced. Cultivars Himbo Top and Polka contained higher levels of hydroxycinnamic acid and ellagic acid derivatives in healthy and infected canes compared to cv. Autumn Bliss. Cultivar Polka also contained the highest level of flavanols and tannins. However, despite high levels of flavanols and total phenols measured in cv. Polka, the canes were highly diseased following infection with D. applanata and L. coniothyrium. The results of the study provide evidence that the level of phenolic compounds in the canes could be causally linked to the differences in disease susceptibility.  相似文献

The study was carried out on phytoplasma susceptible grapevine variety ‘Chardonnay’ (Vitis vinifera L.). The changes in total and individual phenolics, with a focus on hydroxycinnamic acids, flavanols and flavonol contents, were studied in phytoplasma-symptomatic and non-symptomatic berries of Bois noir (BN) infected and uninfected vines. Evident responses to BN infection at veraison have been monitored in a decreased accumulation of caftaric and coutaric acids, p-coumaroyl hexose, procyanidin B1, procyanidin trimer as well as of quercetin-3-O-glucoside, quercetin-3-O-glucuronide and quercetin-3-O-xyloside. At berry softening BN infection statistically increased the content of total phenolics, hydroxycinnamic acids and flavanols, but decreased the flavonol contents, especially at phytoplasma-symptomatic berry skins. Later, at harvest, the BN infection caused an additional significant decrease of coutaric acid and p-coumaroyl pentose contents, moreover of procyanidin B1 and procyanidin dimmers (1, 2, and 3), trimer, kaempferol-3-O-glucoside and of most identified quercetins, except of quercetin-3-O-xyloside. At harvest, non-symptomatic berries from infected plants showed similar dynamics in the total phenolic content compared to berry skins from uninfected plants, but in total flavanols and flavonols content similarity to those symptomatic was observed. The latter decreases grape quality and its antioxidant potential. The Bois noir disease showed specific, local and growth-phase-induced responses regarding the content of phenolics in berry skins, where in particular the differences between phytoplasma-symptomatic and non-symptomatic grapes have to be underlined.  相似文献

Secondary metabolites and host defense compounds were shown to occur in xylem sap, and leaves of Vitis vinifera cv. Italia and cv. Matilde naturally infected by the esca-associated fungi Phaeomoniella chlamydospora (Pch), Togninia minima (Tmi) and Fomitiporia mediterranea (Fme). Samples of xylem sap and leaves were collected from healthy vines and from vines showing severe symptoms of brown wood-streaking caused by Pch and Tmi, or from vines with symptoms of both brown wood-streaking and white rot caused by Fme. Xylem sap collection was carried out during the early spring of 2003 and 2004, corresponding to the phenological phases: (A) cotton bud; (B) green tip; (C) leaves out; (D) stretched out leaves; and (E) visible clusters. In the present work we have studied the accumulation of biomolecules (pentaketides and α-glucans), host defense compounds (benzaldehydes, benzoic acid and cinnamic acid derivatives, flavonols, flavanols, flavan-3-ol derivatives and stilbenes) at different stages of grapevine development. Accumulation and changes in total phenolics and recurring phenolics, and of three phytotoxic secondary metabolites (scytalone, isosclerone and pullulan) were analyzed by HPLC. On comparing results for cv. Italia and cv. Matilde, it can be seen that phenolic concentrations are strongly related to the cv.  相似文献

Early anthracnose caused by Colletotrichum acutatum has become an increasingly serious disease on green, unripe bell pepper fruit in Florida. This contrasts with earlier reports of anthracnose occurring on bell pepper primarily as a ripe-rot disease of mature, colored pepper fruit caused by Colletotrichum gloeosporioides. Management of anthracnose on green bell pepper fruit using fungicides and a commercial inducer of systemic acquired resistance, acibenzolar-S-methyl (ASM), was evaluated during three seasons. In two of the three trials, all the fungicides tested including azoxystrobin, fludioxonil + cyprodinil, mancozeb, famoxadone + cymoxanil, copper hydroxide, and ASM significantly increased the number of marketable fruit compared with control plants. These trials identified fungicides that could contribute to a successful pest management program on pepper for controlling anthracnose caused by C. acutatum. The cross-infectivity potential of C. acutatum was investigated on tomato and strawberry by in vitro and field inoculation. Anthracnose lesions formed readily on wound-inoculated detached fruits of all hosts in in vitro assays. Under field conditions, after inoculation, anthracnose lesions occurred on pepper fruit but no lesions of anthracnose were found on either ripe or unripe tomato or strawberry fruit in adjacent plots.  相似文献

The research dealt with phenolics in healthy versus scab infected apple leaves and fruits. The leaf samples were picked in the period from May to September and the fruit samples at technological maturity. Infection with the Venturia inaequalis fungus enhanced the metabolism of phenolics at the infected sites, especially in the boundary tissue. Tissue infected with Venturia inaequalis showed in comparison to the healthy tissue up to 7.6 times more hydroxycinnamic acids, up to 2.6 times more flavan-3-ols and up to 2.9 times higher values of flavanols. The content level of total phenolics in the infected tissue was 1.3–2.4 times higher than in the healthy leaves and fruit.  相似文献

ABSTRACT Splash dispersal of Colletotrichum acutatum conidia from infected strawberry fruit was assessed using a rain simulator to determine the properties of rain (e.g., intensity [millimeters/hour] or drop size distribution) most related to dissemination. Dispersal with a simulated rain corresponding to a natural rain of about 11 mm/h was compared with dispersal of three other simulated rains that had larger and smaller drop sizes, on average, than idealized natural rains. Splash droplets were collected in sheltered petri plates with a selective medium for Colletotrichum, and colonies formed from conidia entrained in the droplets were counted and used as the measure of dispersal. Colonies were mostly confined to a 27-cm radius from the source, and density of colonies decreased exponentially with the distance squared, as indicated by the fit of a diffusion-type model to the data. Splash dispersal was more affected by drop size distribution than rain intensity or other properties of the generated rains. That is, there was a direct positive relationship between total colonies over 61 min of rain for a circular area with a 72-cm radius (Sigma) and the mass (volume) median diameter of impacting drops (D(0)') for four rain-simulation treatments. In a separate study, strawberry fruit were exposed to the same four simulated rains at two distances from a point source and for two rain durations. Although the proportion of infected fruit (y) increased with time and decreased with distance, rain treatment did not significantly affect y, as predicted based on past work with a wide range of intensities of simulated rains.  相似文献

Qualitative and quantitative changes of individual and total phenolics induced by Colletotrichum coccodes fungal infection have been studied in two susceptible sweet pepper cultivars ‘Soroksari’ and ‘Bagoly’, and the role of soluble phenolic compounds in plant's defence mechanism has been evaluated. Three distinct parts were analysed on pepper fruit: healthy tissue, anthracnose lesion, and bordering tissue, and individual phenolic compounds have been identified with the use of HPLC-MS system. In pepper fruit pericarp 21 phenolic compounds have been determined; the prevalent apigenin, quercetin and luteolin glycosides, chlorogenic acid and one chrysoeriol glucoside. C. coccodes infection increased the accumulation of chlorogenic acid, chrysoeriol glucoside, quercetin and luteolin glycosides in infected bordering tissue of both analysed pepper cultivars compared to healthy pepper tissue or symptomatic spot. Total apigenin derivatives did not show a significant increase in bordering tissue compared to the healthy pepper fruit in contrast to other groups of phenolics. This suggests a lesser role of apigenin glycosides in pepper plant defence against the Colletotrichum fungus. Intense phenolic synthesis was characteristic for the bordering zone between the healthy and infected plant tissue resulting in higher total phenolic content which might hinder the fungus to spread from the infected cells into the healthy tissue.  相似文献

This study was carried out on the leaves of phytoplasma susceptible grapevine variety ??Chardonnay?? (Vitis vinifera L.), and included research of the alterations in chlorophyll and carotenoid contents, contents of phenol compounds and in related enzymes activity in the phenylpropanoid pathway during the Bois noir (BN) infection. Phytoplasma-infected leaves showed reduced contents of chlorophylls and carotenoids, which promoted their susceptibility to oxidative reactions. Furthermore, modulation of flavonoid biosynthesis occurred in infected leaves, leading to an increased activity of phenylalanine ammonia lyase, chalcone synthase/chalcone isomerase, flavanone 3-hydroxylase and polyphenoloxidase, but to a decreased peroxidase activity. Phytoplasma infection led to an increase of the contents of hydroxycinnamic acids (caftaric acid, sinapic acid glucose derivate and coutaric acid), flavanols (procyanidin B1, procyanidin dimer 3, catechin, epicatechin) and flavonols (quercetin 3-O-glucuronide, quercetin 3-O-glucoside) especially in the period up to vérasion. The study demonstrated that at certain phenological key-stages infection with phytoplasma (BN) induced different alterations in enzyme activities and in the contents of biochemical compounds from primary and secondary metabolism.  相似文献

Authentic herbarium specimens of Gloeosporium carthami and G. chrysanthemi as well as live Colletotrichum strains from Carthamus tinctorius, Chrysanthemum coronarium var. spatiosum, and Calendula officinalis were judged to be conspecific based on morphological and molecular phylogenetic analyses. Representative strains from three asteraceous plants were demonstrated by inoculation tests to possess specific pathogenicity to the host plants. We propose a new combination, Colletotrichum carthami, for the anthracnose pathogen of the asteraceous plants, designating our specimen of G. carthami as its epitype. We characterize the species, which differs from the closely related Colletotrichum simmondsii in its pathogenicity and molecular characteristics.  相似文献

This present study investigated the effects of heat shock treatments in strawberry seedlings against crown rot caused by Colletotrichum gloeosporioides. Heat shock treatment at 50 °C for 20 s reduced the disease index of strawberry crown rot and increased chitinase 2-1 gene expression as well as free salycilic acid accumulation. Heat shock treatment did not reduce mycelial growth of C. gloeosporioides. BIT(2-benzisothiazol-3(2H)-one,1,1-dioxide) treatment could not protect strawberry seedlings against crown rot disease. This finding suggests that heat shock induces strawberry resistance against Colletotrichum crown rot and assumes that mechanisms other than SAR probably mediated the protective effect of heat shock-induced resistance.  相似文献

Real-time PCR (TaqMan®) assays were developed for the specific detection and discrimination of Colletotrichum spp., C. acutatum and C. gloeosporioides causing anthracnose in strawberry using the most divergent area of the internal transcribed spacers (ITS1 and ITS2) and 5·8S ribosomal RNA (rRNA) gene region. The specificity of the new assays was tested using DNA from six species of Colletotrichum and nine fungal species commonly found associated with strawberry material, and additionally by comparing the sequences with those from databases using a blast search. The sequences only showed identity with homologous sequences from the desired target organisms. The new assays were 10–100 times more sensitive than conventional PCR methods previously published for the diagnosis of strawberry anthracnose. When real-time PCR was compared with ELISA methods, PCR improved the sensitivity of the identification by obtaining positive results for samples of strawberry plant material that tested negative with ELISA. The development of C. acutatum was monitored using artificially infected strawberry crowns from two strawberry cultivars (Camarosa and Ventana) and a real-time PCR assay specific for this species between January and June 2006. The amount of C. acutatum detected using real-time PCR varied significantly by month ( P  < 0·001), but not by cultivar ( P  = 0·394). The new assays were shown to be useful tools for rapid detection and identification of these pathogens and to allow rapid and accurate assessment of the casual agents of anthracnose in strawberry.  相似文献

Tracing Latent Infection of Colletotrichum acutatum on Strawberry by PCR   总被引:1,自引:0,他引:1  
Colletotrichum acutatum, a quarantine organism on strawberries in the EU, was found in Finland for the first time in 2000. Concern about rapid, unnoticeable spread of this pathogen has necessitated studies to find methods with which the quiescent fungus infection can be detected in imported, cold-stored strawberry plant material. Successful detection of C. acutatum in strawberry tissues by polymerase chain reaction (PCR) is dependent on the method of DNA extraction used. Good-quality nucleic acid, free of PCR inhibitors, was successfully prepared by slightly modifying the DNA extraction method of a commercially available kit. Species-specific primers, previously described in the literature, were successfully used in the PCR reaction. C. acutatum was detected by PCR both on symptomatic and asymptomatic plant parts and in artificially and naturally infected strawberry tissues. Positive PCR results were obtained from ripe and unripe berries, runners, petioles and different parts of crowns. The data demonstrate that the PCR technique can be used to detect C. acutatum in strawberry tissue even in plant parts that do not show visible symptoms.  相似文献

A new disease was found in Japan, on celery (Apium graveolens var. dulce) having severe chlorotic leaf spot, stunt, and dwarf with leaf curl. A spore suspension from the fungus isolated from affected plants induced identical symptoms 14 days after plants were sprayed. Identification and molecular characterization showed that the causal agent is Colletotrichum simmondsii. This report is the first of stunt anthracnose on celery caused by C. simmondsii. We propose the name “stunt anthracnose” for the new disease. Colletotrichum acutatum sensu lato, as reference pathogen of celery anthracnose, should be changed to C. fioriniae based on morphological and molecular characteristics.  相似文献

ABSTRACT Isolates of Colletotrichum spp. from diseased strawberry fruit and crowns were evaluated to determine their genetic diversity and the etiology of the diseases. Isolates were identified to species using polymerase chain reaction primers for a ribosomal internal transcribed spacer region and their pathogenicity was evaluated in bioassays. Isolates were scored for variation at 40 putative genetic loci with random amplified polymorphic DNA and microsatellite markers. Only C. acutatum was recovered from diseased fruit. Nearly all isolates from crowns were C. gloeosporioides. In crown bioassays, only isolates of C. gloeosporioides from strawberry caused collapse and death of plants. A dendrogram generated from the genetic analysis identified several primary lineages. One lineage included isolates of C. acutatum from fruit and was characterized by low diversity. Another lineage included isolates of C. gloeosporioides from crowns and was highly polymorphic. The isolates from strawberry formed distinctive clusters separate from citrus isolates. Evaluation of linkage disequilibrium among polymorphic loci in isolates of C. gloeosporioides from crowns revealed a low level of disequilibrium as would be expected in sexually recombining populations. These results suggest that epidemics of crown rot are caused by Glomerella cingulata (anamorph C. gloeosporioides) and that epidemics of fruit rot are caused by C. acutatum.  相似文献

Anthracnose, caused by Colletotrichum acutatum (species complex), has become a troublesome problem in strawberry production worldwide. This paper reports (i) an optimized sampling method combined with a real‐time PCR technique to detect the latent presence of C. acutatum in cold‐stored strawberry plants used as planting material in several European countries, and (ii) a study of the spread of C. acutatum following a point inoculation under field conditions. Screening of different parts of planting material suggested that C. acutatum is most likely to be present on runners and old petioles. In addition, in seven out of nine batches of planting material from different nurseries, latent infection by C. acutatum was detected in at least one of five replicate samples. Field experiments in 2009 and 2010 showed extensive latent within‐field spread of the pathogen on strawberry leaves, with a within‐row dispersal distance up to at least 1·75 m in 1 week. A straw ground cover between the rows did not decrease C. acutatum spread, probably because introduced (and/or subsequent) inoculum was confined to the plant bed (within the row) and was not present between the beds. Moreover, the number of C. acutatum spores on the symptomless leaves, as estimated using a real‐time PCR method, was significantly (< 0·05) correlated with the incidence of fruit rot at harvest and post‐harvest (= 0·56–0·66). These results illustrate the importance of detecting latent infections in planting material and strawberry leaves in the field.  相似文献

Some important physicochemical and bioactive characteristics of disease free 10 wild grown bilberry (Vaccinium myrtillus) accessions were evaluated. External and internal fruit quality was assessed by standard parameters (fruit weight, fruit color, fruit firmness, soluble solids, pH and total acidity) and bioactive contents (total phenolics, total anthocyanins, total antioxidant capacity and, vitamin C) in fruit were also determined. The commercial grown northern higbush blueberry, Vaccinium corymbosum cv. Bluecrop also included in the study to make comparision with bilberry samples. The highbush blueberry cv. Bluecrop had distinctive external fruit characteristics, such as bigger and more attractive fruits. However, the wild grown bilberry accessions showed interesting characters in mesocarp, such as high total phenolic content, total anthocyanin and total antioxidant capacity. Total phenolic and total anthocyanin content was 327?mg gallic acid equivalent and 142?mg of cyanidin-3-glucoside equivalent in 100?g fresh fruit in cv. Bluecrop while it was between 576–624?mg gallic acid equaivalent and 296–324?mg of cyanidin-3-glucoside equivalent in 100?g fresh fruits of bilberry accessions. Moreover, wild accessions approximately had 2 folds higher antioxidant capacity than cv. Bluecrop. Results suggested that bilberry accessions may serve as a source of desirable genes to develop improved varieties that respond to the new needs of the market.  相似文献

Polyamine and phenolic acid levels as well as activities of some enzymes of their biosynthetic metabolism were examined in two sugarcane (Saccharum officinarum) cultivars differing in susceptibility to leaf scald, a disease caused by the bacterium, Xanthomonas albilineans. Juice obtained from both infected cultivars showed significantly increased levels of free putrescine and ornithine decarboxylase activity. However, the pathogen induced different changes in the two cultivars in subsequent metabolic steps. Whereas acid insoluble conjugated spermidine completely disappeared from the highly susceptible cv. C 439-52, an increase in acid insoluble conjugated polyamines was observed in the moderately susceptible cv. L 55-5. Phenolic acid metabolism also differed in the two cultivars. Since total phenolic acid content and phenylalanine ammonium lyase activity was greater in both cultivars after infection, distribution of phenolic acids between free or conjugated forms diverted into different pathways. The level of susceptibility of the two cultivars is discussed in terms of changes in these compounds.  相似文献
