贵州省烟草黑胫病菌遗传多样性分析   总被引:2,自引:0,他引:2  
运用RAPD分子标记技术分析了采自贵州各个地区的38个烟草黑胫病菌株的遗传多样性,探索了贵州省主要烟草种植区间的烟草黑胫病病菌的遗传分化关系.结果表明,受试38个菌株共产生65条谱带.其中多态性为59条,占90.7%.说明贵州省主要烟草种植区的烟草黑胫病病菌具有丰富的遗传多样性,不同地点烟草黑胫病病菌的遗传结构之间都有一定的差异.根据引物扩增的指纹图谱,运用UPGMA分析法,以遗传相似系数0.74,将供试38个菌株划分为9个基因型,发现除少数地区的菌株被划分为同一基因型外,其他不同地理区域的菌株基因型的划分与地理来源没有直接的相关性.  相似文献

本研究对来自黑龙江、辽宁、河北、山东、江苏5个省12个黄瓜主产区的77个黄瓜霜霉菌菌株,采用SRAP分子标记进行了遗传多样性分析。从35对SRAP引物中筛选出10对引物,共产生9 554条扩增条带,其中9 132条表现多态性,占95.6%。基于SRAP分子标记,77个黄瓜霜霉菌菌株的遗传距离为0.60~1.00,表明黄瓜霜霉菌具有丰富的遗传多样性。通过聚类分析,77个黄瓜霜霉菌菌株聚类为8个类群,并且来自相同地区的菌株大多数聚集于同一类群中,表明黄瓜霜霉菌群体的遗传多样性与其地理来源密切相关。  相似文献

辣椒疫霉(Phytophthora capsici)对辣椒的致病力分化研究   总被引:2,自引:0,他引:2  
 辣椒疫霉(Phytophthora capsici Leonian)是具有重大危害性的病原卵菌,其寄主范围较广,可引致辣椒、番茄、茄子、黄瓜、南瓜等多种重要蔬菜作物的疫病。由辣椒疫霉引起的辣椒疫病是一种毁灭性病害,在世界各地的辣椒种植区普遍发生,我国尤以江苏、浙江、安徽、上海等长江中下游地区发生严重。迄今国内关于辣椒疫霉致病力分化的研究较少,且存在分歧。有报道指出辣椒疫霉菌株对辣椒的致病力差异与菌株地区来源直接相关[1];也有研究认为辣椒疫霉菌株对辣椒的致病力与地理来源不直接相关[2]。此外,辣椒疫霉菌株致病力分化和菌丝生长速率是否相关尚属未知。本文对采自安徽合肥、淮南、和县、潜山、岳西、江苏南京和四川邛崃7个县市的23个辣椒疫霉菌株对辣椒的致病力和菌丝生长速率进行了测定,旨在明确辣椒疫霉菌株对辣椒的致病力是否存在分化现象及致病力与菌株地区来源及菌丝生长速率之间的关系,为辣椒疫霉所致辣椒疫病的抗病育种和综合治理提供依据。  相似文献

为了明确福建省辣椒疫霉菌群体遗传结构的表型特征,对分离自福建省15个市(县)的300株辣椒疫霉菌的交配型、甲霜灵敏感性及生理小种进行了测定。结果显示,288株为A2交配型,12株为A1交配型,出现频率分别为96%和4%,以A2占优势,在各采集点均有分布。263株对甲霜灵表现敏感,28株为中间型,9株为抗性,分别占总体的87.67%、9.33%和3.00%,甲霜灵敏感菌株为主要菌系。根据菌株对辣椒疫霉菌生理小种鉴别寄主的致病性测定结果,可将辣椒疫霉菌划分为3个生理小种,其中168株为小种3,126株为小种2,6株为小种1,分别占56%、42%和2%,小种3为福建辣椒疫霉菌优势小种。  相似文献

中国不同地区致病疫霉遗传多样性的RAPD分析   总被引:4,自引:0,他引:4  
 本文应用RAPD技术检测了我国主要马铃薯产区致病疫霉的遗传分化情况及不同地区菌株间的亲缘关系。用筛选出的10个随机引物对1997-2001年间采自我国9省市的82株及3株来自日本的致病疫霉DNA进行了PCR扩增,获得了79条谱带,其中多态性标记75条,占95%。根据扩增结果,运用UPGMA分析,获得了表现菌株间亲缘关系的树状图。菌株间的最大遗传距离为0.5,以距离0.3为阈值,可将供试菌株划分为10个组(RG1-10)。结果发现:A1交配型菌株群体内的差异大于A1和A2菌株群体之间的;RAPD分组与菌株的地理来源、交配型及对甲霜灵的敏感性无明显相关性。研究结果显示,来自中国北方甘肃、内蒙、吉林、黑龙江地区的菌株与一些来自云南、四川等西南地区的菌株亲缘关系相近。病原菌随种薯的迁移可能是导致这种现象的原因之一。  相似文献

对30个西瓜枯萎病菌Fusarium oxysporum f.sp.niveum菌株基因组DNA进行相关序列扩增多态性(SRAP)分子标记分析,以探究其遗传多样性与地理来源的关系。采用尖孢镰刀菌西瓜专化型Fusarium oxysporumf.sp.niveum0、1、2号生理小种的基因组DNA为模板,对225对SRAP引物进行筛选,筛选出20对多态性、重复性较好且条带清晰的引物,对30个菌株进行PCR扩增,共扩增出386条带,其中多态性条带有371条,多态性比率为96.11%,平均每对引物扩增出19.3个位点和18.55个多态性位点。UPGMA法聚类分析结果显示,供试菌株两两之间的遗传相似系数范围为0.69~0.90,平均为0.79,说明尖孢镰刀菌西瓜专化型的遗传多样性较为丰富。基于SRAP标记聚类分析表明,30个菌株在遗传相似系数为0.70处被划分为3个类群,I类群包含24个菌株,其中18个来自湖南省,Ⅱ类群只包含1个来自黑龙江省哈尔滨市的菌株,它和另一个来自黑龙江地区的菌株被划分到不同的类群,且遗传距离相对较远;Ⅲ类群包含了5个菌株,其中3个来自海南三亚,其余两个来自湖南省。根据菌株的分布情况来看,菌株的聚集与地理来源没有明显的相关性。  相似文献

稻曲病菌遗传多样性与群体结构的初步分析   总被引:5,自引:0,他引:5  
 利用随机扩增多态性DNA (random amplified polymorphic DNA,RAPD)初步分析了稻曲病菌(Ustilaginoidea virens)的群体遗传结构。从1 60个随机引物中筛选32个扩增带型清晰、重复性好的引物,对不同年份采自辽宁、云南、湖北和浙江等水稻种植区的5 6个菌株进行扩增。32个引物扩增出2 2 3条带,绝大多数引物对不同年度采自不同稻区的菌株扩增的DNA谱型相同,大多数菌株间相似性系数达0.80以上。根据扩增DNA片段的多态性,从空间分布来看,来源于北方、长江流域和南方的菌株难以划分出明显的地理宗谱;不同年度的菌株DNA多态性也无明显的差异。上述结果初步表明稻曲病菌遗传稳定,寄主选择作用(寄主的基因型及其时空分布)对稻曲病菌变异的影响较小。但是尚需采用其它的分子技术测试更多的菌系,才能较系统地分析我国稻曲病菌系的遗传变异及群体结构特点。  相似文献

我国植物青枯菌菌株的遗传多样性和组群划分   总被引:14,自引:0,他引:14  
 采用15条随机引物对我国11个省(市、区)6种不同寄主植物的43个青枯菌代表性菌株和4个国外青枯菌菌株,进行了PCR扩增.引物OPB11、OPA15、OPE1和OPZ10对上述所有菌株扩增获得了相似的产物电泳图谱,分别具1~5条谱带不等;引物OPB7、OPA10和OPF1对马铃薯菌株获得了相同的产物图谱,但对其它寄主菌株的产物间有明显差别;引物OPA14、OPC,6、OPG14、OPF5、OPK14、OPK20和OPK17对于不同菌株的扩增产物多态性很强.供试菌株被聚类为2个组群,即组群A和组群B.组群A中又可分为7个亚组(A1、A2、A3、A4、A5、A6、A7),其中A1含有2个类型(A1-1、A1-2);组群B中也可分为2个亚组(B1、B2),其中B1含有3个类型(B1-1、B1-2、B1-3),B2也含有3个类型(B2-1、B2-2、B2-3).RAPD组群A中包含了27个来自我国不同地区的马铃薯菌株,主要是3号小种、生化变种2;组群B中含有20个来自不同地区、不同寄主的菌株,分属于其它不同的小种和生化变种.研究结果表明,我国青枯菌菌株RAPD组群的划分与菌株的地理来源关系不大,而与寄主来源有明显相关性.此外,通过对我国青枯菌菌株组群进行同源性PCR分析表明,来源自马铃薯的3号小种菌株属于美洲分支"Americanmm",而来自其它寄主的青枯菌1号、5号小种菌株属于亚洲分支"Asiaticum",与本研究RAPD组群A和组群B的划分是一致的.  相似文献

玉米大斑病菌ISSR反应体系的优化和遗传多样性分析   总被引:6,自引:3,他引:3  
以玉米大斑病菌基因组DNA为模板,采用单因素水平优化的方法对DNA聚合酶的来源及浓度、引物浓度、dNTPs浓度、DNA模板浓度、Tm(退火温度)、PCR反应循环数等重要参数进行摸索和优化,建立了玉米大斑病菌ISSR-PCR优化反应体系,并从40条ISSR引物中筛选出9条多态性较好的ISSR引物。对来自河北、河南、辽宁等玉米主产区的44个菌株进行ISSR分析表明,ISSR标记在我国玉米大斑病菌中存在较高的多态性,多态性条带占40.3%。聚类分析显示,在阈值为0.8时菌株被分为7个类群。对ISSR揭示的玉米大斑病菌的遗传多样性与菌株交配型、地理来源之间的关系进行分析,结果显示菌株的遗传多样性与交配型间的关系密切,而与其地理来源无明显相关性。  相似文献

采用RAPD-PCR分子标记技术分析了51株不同地理来源、寄主来源的绿僵菌Metarhizium anisopliae菌株的遗传多态性。从94条RAPD引物中筛选出18条引物,对所有试验菌株进行RAPD-PCR扩增,共获得96条扩增片段,其中81条片段表现多态性,占84.1%。聚类分析表明,供试的51株菌株间的相似性系数范围为0.52~0.98,表明菌株间存在丰富的遗传多态性。供试菌株在相似性系数0.7的水平可分为4个组群。按菌株DNA多态性与地理及寄主来源的聚类分析表明,大多数菌株的DNA多态性与地理或寄主有一定的相关性,即长期的地理环境和寄主适应性可能形成了种群的分化。  相似文献

ABSTRACT Verticillium dahliae isolates from artichoke, bell pepper, cabbage, cauliflower, chili pepper, cotton, eggplant, lettuce, mint, potato, strawberry, tomato, and watermelon and V. albo-atrum from alfalfa were evaluated for their pathogenicity on all 14 hosts. One-month-old seedlings were inoculated with a spore suspension of about 10(7) conidia per ml using a root-dip technique and incubated in the greenhouse. Disease incidence and severity, plant height, and root and shoot dry weights were recorded 6 weeks after inoculation. Bell pepper, cabbage, cauliflower, cotton, eggplant, and mint isolates exhibited host specificity and differential pathogenicity on other hosts, whereas isolates from artichoke, lettuce, potato, strawberry, tomato, and watermelon did not. Bell pepper was resistant to all Verticillium isolates except isolates from bell pepper and eggplant. Thus, host specificity exists in some isolates of V. dahliae. The same isolates were characterized for vegetative compatibility groups (VCGs) through complementation of nitrate nonutilizing (nit) mutants. Cabbage and cauliflower isolates did not produce nit mutants. The isolate from cotton belonged to VCG 1; isolates from bell pepper, eggplant, potato, and tomato, to VCG 4; and the remaining isolates, to VCG 2. These isolates were also analyzed using the random amplified polymorphic DNA (RAPD) method. Forty random primers were screened, and eighteen of them amplified DNA from Verticillium. Based on RAPD banding patterns, cabbage and cauliflower isolates formed a unique group, distinct from other V. dahliae and V. albo-atrum groups. Minor genetic variations were observed among V. dahliae isolates from other hosts, regardless of whether they were host specific or not. There was no correlation among pathogenicity, VCGs, and RAPD banding patterns. Even though the isolates belonged to different VCGs, they shared similar RAPD profiles. These results suggest that management of Verticillium wilt in some crops through crop rotation is a distinct possibility.  相似文献

The genetic and virulence variability of 112 isolates of Phaeoisariopsis griseola , collected from various locations in Central America, were studied using seven random amplified polymorphic DNA (RAPD) primers and 12 common-bean differential genotypes. Broad molecular diversity ( H  = 0·92) among isolates was found using RAPD markers. Fifty pathotypes were identified on 12 differential bean genotypes, 29 of which were represented by only one isolate. Only 18 pathotypes were found in two or more countries. Pathotype 63-63 was the most virulent and caused leaf spots on all 12 common-bean differential genotypes. Comparison of virulence phenotypes and RAPD profiles to known Andean P. griseola isolates confirmed that all isolates belonged to the Mesoamerican group. Pairwise comparison between individual RAPD loci showed that the majority were in gametic phase linkage disequilibrium, revealing that P. griseola maintains a genetic structure that is consistent with asexual reproduction. The molecular and virulence diversities of P. griseola isolates from Central America imply that using single resistance genes to manage angular leaf spot is inadequate and stacking resistance genes may be necessary to manage the disease effectively.  相似文献

Genetic Diversity of Japanese Strains of Ralstonia solanacearum   总被引:2,自引:0,他引:2  
ABSTRACT The genetic diversity of 74 Japanese strains of Ralstonia solanacearum was assessed by pathogenicity tests and the repetitive sequencebased polymerase chain reaction (rep-PCR) fingerprint method. Based on their genomic fingerprints, biovar N2 strains were divided into two distinct groups, one consisting of potato isolates belonging to race 3, and the other consisting of tomato, eggplant, pepper, and tobacco isolates belonging to race 1. Biovar 3 strains had low average similarity and were divided into five groups that differed in original host or pathogenicity. Biovar 4 strains consisted of only one group at the 80% similarity level. Comparative analysis of the rep-PCR fingerprints of 78 strains, including six biovars from Japan and various countries, revealed two main clusters. Cluster 1 comprised all biovar 3, 4, and 5 strains, biovar 1 strains from Reunion, and some biovar N2 strains from Japan. Cluster 2 included most of the biovar 1, 2, and N2 strains. The fingerprints showed low average similarity with biovar N2 strains from Japan and Brazil.  相似文献

Phenotypic variants of Clavibacter michiganensis subsp. michiganensis (Cmm) were isolated from pepper fields and from pepper seeds during quarantine inspections. All strains isolated from pepper (pepper isolates) produced orange-coloured colonies with lower mucoidy than typical Cmm strains isolated from tomato (tomato isolates). However, the results of ELISA, fatty acid analysis, 16S rDNA sequencing, and PCR analysis showed that all pepper isolates were similar enough to be identified as Cmm. In addition to phenotypic variations, the pepper isolates showed different pathogenic and genetic characteristics from tomato isolates from the USA, Europe, or other countries. They could be clearly distinguished in terms of pathogenicity, as they showed increased pathogenicity to pepper but reduced pathogenicity to tomato. Tomato isolates caused strong wilting and canker in tomato, but caused only canker and no wilting in pepper and bell pepper. However, pepper isolates caused no wilting, even in tomato, and only caused canker in the three host plants. In addition, compared to tomato isolates, pepper isolates showed increased colonization efficiency and caused a greater reduction in shoot dry weight in pepper. Pepper and tomato isolates could be separated into two groups according to host origin on the basis of 16S rDNA and ITS sequence analysis. They also showed different rep-PCR genomic fingerprints. All pepper isolates showed higher cellulase activity than tomato isolates on M9CMC plates. However, two plasmid-borne virulence genes of Cmm, pat-1, and celA, were not detected in any pepper isolates by PCR. Furthermore, PCR for pathogenicity-related genes located on a pathogenicity island (PAI) revealed that all tomato isolates were positive for these genes, whereas the pepper isolates did not show any PCR products for the chpC, chpG, ppaA, or tomA genes. Therefore, we suggest that the pepper isolates may represent a separate Cmm population that has evolved within the limits of this host.  相似文献

 采自新疆24个不同植棉县(市或团场)的37株棉花枯萎病菌代表菌株,经人工接种于国际通用鉴别寄主,致病性反应均表现为典型的7号生理小种特征。RAPD分析结果也显示出这37个供试菌株与7号小种各对照菌株间基因组DNA的指纹图谱高度相似,属同一遗传相似组,而与3号和8号小种的对照菌株间遗传差异较大,亲缘关系较远,即7号生理小种是组成目前新疆棉花枯萎病菌群体的优势小种,而原分布于新疆吐鲁番等地的3号小种在本研究中未被发现。结合部分自选辅助鉴别寄主对其中18个菌株进行的致病力分化研究表明,在7号小种内部还存在着侵染力的分化,显示出棉花枯萎病菌较强的变异性和适应性。  相似文献

 对分离自玉米弯孢菌叶斑病标样中的77株新月弯孢菌和1株来自水稻的新月弯孢菌进行RAPD分析表明,菌株间具有丰富的遗传多样性,在相似系数约0.60处,所有菌株被聚为3个组,但88.0%的菌株聚入第Ⅰ组内,其余菌株被聚入另外2个组内。第Ⅰ组内共有69个菌株,包含来自不同区域的致病性较强的菌株,是玉米弯孢菌叶斑病的优势类群,其余2个类群主要是一些致病能力较弱或不致病的菌株。结果表明,新月弯孢菌种内菌株的遗传多样性与致病性相关,但与菌株地理来源无明显的直接关系。  相似文献

四川省不同寄主立枯丝核菌的遗传分化和致病力研究   总被引:4,自引:1,他引:4  
 在四川省生态条件下,从不同水稻和玉米植株上分别分离到来源不同的立枯丝核致病菌15株和7株。致病力、菌丝融合实验结果表明,菌株均属于AG-11A群,各菌株间致病力差异显著。对分离菌株进行RAPD分析,结果显示,相似系数为0.92处菌株可聚合为5类,聚类分组和寄主来源有一定的相关性,来自相同寄主菌株的亲缘关系较近,不同寄主对立枯丝核菌的遗传分化有一定的影响,与病原菌的致病力差异没有直接的相关性。  相似文献
