Weaning of piglets complicated with an exposure to pathogenic strains of Escherichia coli was scrutinized in two sets. The first set comprised 20 animals representing two litters and the second set included 30 animals from five litters. The piglets were either left as controls or exposed to one or three pathogenic strains of E. coli. Aiming to simulate a natural exposure the challenge strains were spread on the floor of the pens at weaning. In addition the pigs experienced several non‐infectious stress factors commonly occurring at that occasion. Some groups were given adrenocorticotropic hormone (ACTH), aiming to simulate a stressful weaning. The balance and the composition of the faecal coliform populations, measured by a metabolic fingerprinting method, was disturbed among all animals following weaning. This disturbance was more pronounced and lasted longer among piglets exposed to pathogenic strains of E. coli. All piglets exposed to pathogenic E. coli shed these strains in faeces. Diarrhoea was induced in the groups exposed to E. coli, but not among the control animals. Pigs not treated with ACTH and subjected to a single pathogenic strain of E. coli became infected but did not develop diarrhoea unless if coinciding with shed of rotavirus. Control pigs excreting rotavirus had no diarrhoea. Diarrhoea was most frequent in the groups exposed to three pathogenic strains of E. coli, and in these groups diarrhoea was seen in the absence of rotavirus. ACTH administration amplified the clinical signs. The litter of origin influenced the development of post‐weaning diarrhoea.  相似文献

The objective of the study was to investigate the effect of offering supplementary creep feed to piglets during the suckling period on the faecal shedding of haemolytic Escherichia coli bacteria and occurrence of spontaneous post-weaning diarrhoea 0-5 days after weaning. Supplementary creep feed was offered to half of the piglets in 12 litters, from 2 weeks of age until weaning at 4 weeks, and the individual feed contact was recorded by direct observations. It was found that diarrhoea occurrence was associated with faecal shedding of haemolytic E. coli bacteria (p=0.003), specifically E. coli O149 (p=0.004). Occurrence of diarrhoea and faecal shedding of haemolytic E. coli was not associated with creep feeding per se. However, the faecal E. coli O149 shedding occurred significantly less often in piglets that were offered creep feed in the suckling period but only showed limited interest in the feed (i.e. contacted the feed less than or equal to the median level of contact) compared to piglets that had frequent creep feed contact or piglets that had not had access to creep feed at all (p=0.015). Correspondingly, the diarrhoea occurrence tended to be lower in these piglets (p=0.081). Piglets with low creep feed contact during the suckling period ate the same total amount of feed during the 5 days after weaning, however, they ate less feed on days 0-2 after weaning compared to the piglets with frequent creep feed contact and non-creep fed piglets. It is suggested that intestinal function associated with a voluntary low creep feed contact during the suckling period leads to decreased feed intake just after weaning, and thus reduces the intestinal proliferation of E. coli O149 in these piglets.  相似文献

An evaluation of the efficacy of a commercial crystalline cordial concentrate for preventing post-weaning diarrhoea (PWD) and improving weight gains in pigs following weaning, was carried out in 3 herds using 861 pigs. The incidence of PWD in pigs drinking the cordial was reduced in 2 herds and its severity was reduced in one of these. There was no effect on either incidence or severity in the other herd. The pigs given cordial consumed considerably more water per day and less feed per day than pigs drinking plain water during the 10-day supplementation period. There was no improvement in average daily gain or feed conversion ratio attributable to adding cordial to drinking water in any of the herds. In one herd the growth rate of pigs receiving cordial was actually depressed during the supplementation period. There was no reduction in mortality or in number of treatments given for diarrhoea in pigs receiving cordial.  相似文献

Milk from sows whose progeny developed post weaning E. coli diarrhoea (PWD milk) and from sows which were immunized by intramuscular vaccination using a homologous strain of E. coli (immune milk) were tested in ligated segments of pig intestine. The results showed that PWD milk neutralized the enterotoxigenic, fluid accumulating capacity of the lysate of the disease-causing E. coli pathogen. A similar effect was seen by using immune milk (Table I). Neither PWD milk nor immune milk contained sufficient antibacterial activity to neutralize the fluid accumulating capacity of live cultures of E. coli O149:K91, while such activity was contained in immune serum. It is concluded that milk from sows whose progeny developed PWD contains antibodies capable of neutralizing the enterotoxigenic effects of the homologous E. coli organisms. It is suggested that the presence in milk from these sows of antibody-mediated activity against enteropathogenic E. coli organisms may be instrumental in preventing the disease in the progeny during the suckling period and consequently, when this protective milk supply stops at weaning, the disease may develop in susceptible animals, mainly because their own production of specific E. coli antibodies is insufficient to prevent PWD.  相似文献

Background

Post-weaning diarrhoea (PWD) is a significant gastrointestinal disease in pigs. It is considered a multifactorial disease associated with proliferation of enterotoxigenic Escherichia coli in the intestinal tract of affected pigs. The aim of this study was to analyse risk factors related to the occurrence of PWD on Finnish piglet producing farms.

Methods

The data of a follow-up study of 73 conventional piglet producing farms was used in the case-control study. The selection of the 41 PWD case and 28 control farms was based on the use of antimicrobials for treating diarrhoea in weaned pigs and the answers related to the occurrence of diarrhoea after weaning in the questionnaire. Four intermediate farms were excluded from the statistical analysis.Altogether 39 factors related to herd characteristics, weaner pig management and pig health were studied. The median number of sows was 59.0 (IQR = 44.0; 74.5) and 52.5 (IQR = 36.8; 61.5) on the case and the control farms, respectively.The significances of the univariable associations between the explanatory variables and the outcome variable were tested, and in the multivariate analysis quasibinomial generalized linear models were applied.

Results

An increased risk of PWD was associated with the regimen of twice a day feeding and feed restriction after weaning (P = 0.02; compared to feeding three or more meals a day or the use of ad libitum feeding) and with a higher number of sows on the farm (P = 0.02; risk increasing with increasing number of sows). Automatic temperature control was associated with a decreased risk of PWD (P = 0.03; compared to manual temperature control).

Conclusion

Twice a day feeding of newly-weaned pigs should be avoided if the amount of feed given is restricted. Variation in ambient temperature should be minimized in housing of newly-weaned pigs and this can be achieved by using automatic temperature control. With increasing number of sows in the herds the risk of PWD increases and more attention should be paid to prevention of post-weaning diarrhoea.  相似文献

Immunoprophylaxis of porcine oedema disease and post-weaning diarrhoea caused by strains of Escherichia coli expressing fimbriae F18 is an unsolved problem. The study was designed to examine whether vaccination with a live F18ac vaccine of unweaned pigs born to sows with F18ac antibody in the colostrum requires preformed fimbriae in the vaccine, and whether protection against the heterologous fimbrial variant F18ab is induced as well. Genetically susceptible pigs were vaccinated orally on three consecutive days, beginning 10 days before weaning with 10(11) CFU of an F18ac culture. Challenge with a dose of 10(7) CFU of E. coli F18 on three consecutive days was initiated 9 or 11 days after weaning. Eighteen pigs given the fimbriated F18ac vaccine and challenged with a strain of the homologous fimbrial variant were protected against colonization; mean faecal viable counts of the challenge strain were >3 log10 lower than those from the 17 non-vaccinated control pigs. The vaccinated pigs developed a significant rise of F18ac IgA serum antibodies. The 23 pigs which had received the non-fimbriated vaccine showed no significant protection and exhibited much lower serum F18ac IgA ELISA reactivities. Eighteen pigs vaccinated with the fimbriated F18ac and challenged with an F18ab strain had faecal viable counts nearly as high as those from 16 non-vaccinated control pigs. It is concluded that only oral vaccines having preformed fimbriae induce protection limited to the homologous fimbrial variant.  相似文献

Five-day-old pigs challenged with 10(3) pathogenic Escherichia coli (nalidixic acid resistant) showed no clinical signs of disease until subsequently weaned at three weeks. Dietary manipulation was shown to influence xylose malabsorption, diarrhoea and bacterial proliferation after weaning. Brief, but not continuous, contact with the diet before weaning markedly increased the severity of subsequent disease after weaning. Immunogenicity of the weaning diet was critical for the development of the disease. Two diets, identical except that in one the protein source (casein) had previously been enzymatically hydrolysed, were compared. Pigs fed the predigested diet showed no clinical signs of post weaning diarrhoea whereas those fed the untreated casein all developed diarrhoea.  相似文献

The effect of creep feeding on the response of pigs weaned at two or three weeks old to infection with an enteropathogenic strain of Escherichia coli (O149; K91[B], K88 a, c [L]) was studied in two separate experiments. Gastric intubation was adopted to regulate the intake of creep feed during the sucking period. Animals were given the enteropathogenic strain either before weaning or at weaning and the course and outcome of the infection followed bacteriologically and clinically. The response of the animals to the infection varied considerably in accord with recent reports of multiple phenotypes (to K88 +ve organisms) among pigs but consumption of creep feed before weaning did not significantly affect the prevalence, duration or severity of the diarrhoea induced experimentally by the organism. Hence reputed immunological responses mounted against dietary antigens did not predispose to or protect against this infection.  相似文献

Threonine (Thr) is important for mucin and immunoglobulin production. We studied the effect of added dietary Thr on growth performance, health, immunity and gastrointestinal function of weaning pigs with differing genetic susceptibility to E. coli K88ac (ETEC) infection and challenged with ETEC. Forty‐eight 24‐day‐old weaned pigs were divided into two groups by their ETEC susceptibility using mucin 4 (MUC4) gene as a marker (2 MUC4?/?, not‐susceptible, and 2 MUC4+/+, susceptible, pigs per litter). Within genotype, pigs were fed two different diets: 8.5 (LThr) or 9.0 (HThr) g Thr/kg. Pigs were orally challenged on day 7 after weaning and slaughtered on day 12 or 13 after weaning. Before ETEC challenge, HThr pigs ate more (p < 0.05). The diet did not affect post‐challenge growth, but HThr tended to increase post‐challenge feed efficiency (p = 0.087) and overall growth (p = 0.087) and feed efficiency (p = 0.055). Before challenge, HThr pigs excreted less E. coli (p < 0.05), while after challenge, diet did not affect the number of days with diarrhoea and ETEC excretion. MUC4+/+ pigs responded to the challenge with more diarrhoea, ETEC excretion and anti‐K88 IgA in blood and jejunal secretion (p < 0.001). HThr pigs had a higher increase of anti‐K88 IgA values in jejunal secretion (p = 0.089) and in blood (p = 0.089, in MUC4+/+ pigs only). Thr did not affect total IgA and IgM values, morphometry of jejunum, goblet cells count in colon, total mucin from jejunum and colon, but varied jejunal goblet cells counts (p < 0.05). In the first two post‐weaning weeks, 8.5 g Thr/kg diet may be not sufficient to optimize initial feed intake, overall feed efficiency and intestinal IgA secretion and to control the gut microbiota in the first post‐weaning week, irrespective of the pig genetic susceptibility to ETEC infection.  相似文献

FACTORS CONTRIBUTING TO POSTWEANING DIARRHOEA IN A LARGE INTENSIVE PIGGERY   总被引:4,自引:0,他引:4  
Some aspects of postweaning diarrhoea (PWD) in a piggery during the first week after early weaning were investigated. A haemolytic enterotoxigenic strain of E. coli (0149: K88: H10) was regularly recovered from piglets with PWD while rotavirus was demonstrated on a number of occasions. Prior to weaning piglets were either free of, or shed very few, haemolytic E. coli in their faeces. However, all piglets were excreting haemolytic E. coli between 5 and 7 days after weaning. The role of rotavirus in PWD was unclear. There appeared to be a direct relationship between serum antibodies to rotavirus in the sow at farrowing and those of the piglets soon after birth. The decline of maternal neutralising antibody to rotavirus coincided with the immediate postweaning period (3 to 5 weeks after birth). This was followed by an increase in antibody levels, 5 to 8 weeks after birth. There was no significant difference in the growth rate between affected and unaffected piglets over a period of 120 days. Medication of water during the first week after weaning had no significant effect on the incidence of PWD in the herd. A change in both the weaner diet and the weaning procedure reduced piglet mortality associated with PWD by more than half.  相似文献

Weaned piglets were used to determine the effect of dietary spray-dried porcine plasma (SDPP) on the clinical response to an infection with a pathogenic Escherichia coli (E. coli) O139:K82 LT(-). The piglets were divided into two groups of 10 animals each. One group was fed the control diet containing soybean(meal) plus whey powder. The test piglets were fed a diet with 8% SDPP. Piglets were orally infected with the challenge strain on days 6 and 7 after weaning. The experimental period lasted 14 days after which the piglets were euthanised and necropsied. Faecal samples were collected daily for bacteriological analysis. Segments of jejunum, caecum and rectum were removed for bacteriological analysis post mortem. Feed intake and weight gain, faecal and condition scores and body temperature were measured daily. In the control and SDPP groups, 6 and 7 piglets died from diarrhoea. The average daily feed intake (ADFI) and average daily gain (ADG) were substantially higher in the SDPP group than in the control group. SDPP-fed piglets generally had a more favourable faecal score and a healthier appearance than did the control piglets. The faecal excretion of E. coli O139:K82 was similar for control and test piglets. There were no diet effects on the E. coli O139:K82 counts at different sites of the intestine. In this experiment, the inclusion of SDPP at an economically acceptable percentage in the diet could not prevent piglet losses due to challenge with a pathogenic E. coli, but improvements of ADG, ADFI and faecal and condition scores were achieved.  相似文献

This study comprised 48,931 litters in 89 sow herds. During the study (1976-82) weaning age decreased from approx. 42 days to approx. 30 days. The mean incidence of post-weaning diarrhoea was 6.0% of litters weaned, with little variation by year but with considerable variation among herds. Within the individual herd increased incidence occurred over limited periods, probably associated with specific infections. Litters with diarrhoea during the suckling period had increased risk of post-weaning diarrhoea. The incidence of post-weaning diarrhoea increased with litter size at weaning. Thus, a litter of 11-12 piglets at weaning had 1.2 times higher risk than litters with 8-10 piglets. In contrast to pre-weaning diarrhoea, there was no association between parity of the sow and diarrhoea in the litter after weaning. Litters weaned below 2 weeks of age had a 2-fold risk of developing diarrhoea after weaning and a 2.4-fold higher mortality rate than did litters weaned at 6-7 weeks. Similarly, litters weaned at an individual piglet weight below 3 kg bodyweight had a 3-fold higher risk of developing diarrhoea after weaning and a 5-fold higher mortality rate than did pigs from litters weaned at a bodyweight of 7-8 kg. The incidence of post-weaning diarrhoea decreased with increasing herd size. Piglets from litters with post-weaning diarrhoea had reduced weight gains after weaning and were 2.3 days older at 25 kg bodyweight than piglets from non-diarrhoeic litters. Likewise, diarrhoea after weaning was associated with an increased incidence of diseases of the skin and respiratory tract. Thus the risk of contracting respiratory disease was 4 times greater in diarrhoeic litters.  相似文献

When pigs are weaned at two days of age large numbers of Excherichia coli appear in the anterior gut and the incidence of diarrhoea rises. The two phenomena do not appear to be directly related because the strains of E coli isolated are not serotypes previously found to be associated with neonatal pig scouring. Representative strains of the non-enteropathogenic serotypes did not produce enterotoxin and did not adhere to small intestine brush borders. Moreover when antibiotics were fed to eliminate E coli from the gut, the pigs still scoured. Rotavirus was detected in the gut contents and gut epithelium of scouring pigs and a bacteria-free filtrate of gut contents produced diarrhoea when administered to germ-free pigs. It is suggested that rotavirus may be one of the causes of the scouring seen shortly after weaning pigs at two days of age.  相似文献

Enterotoxigenic Escherichia coli (ETEC)-associated post-weaning diarrhea (PWD) is economically one of the most important diseases for the swine industry. Porcine ETEC strains typically express K88 or F18 fimbria and heat-labile (LT) and/or heat-stable (STa, STb) enterotoxins. However, recent studies indicate that EAST1 toxin, adhesin involved in diffuse adherence (AIDA-I) and porcine attaching and effacing-associated factor (paa) may also be expressed by ETEC strains associated with diarrhea. To better understand the virulence factors of E. coli strains that cause PWD, we applied PCR to screen for K88, F18, F41, 987P and K99 fimbrial genes; LT, STa, STb, Stx2e and EAST1 toxic genes; and AIDA-I, paa and EAE adhesin genes in E. coli strains recently isolated from young pigs with PWD in the US. Of 304 E. coli isolates from diarrheic pigs submitted for testing, 175 (57.6%) strains possessed fimbrial genes: K88 (64.6%), F18 (34.3%), F41 (0.57%), K99 (0.57%), 987P (0); toxin genes: LT (57.7%), STb (72.6%), STa (27.4%), STx2e (17.4%), EAST1 (35%); and adhesin genes: AIDA-I (26.9%), paa (60%), EAE (1.1%). All toxin genes except the EAST1 toxin gene, were almost exclusively associated with K88+ or F18+ isolates, and most of these isolates carried multiple toxin genes. The non-fimbrial adhesin paa was found present in over half of the K88+ isolates. A total of 129 (42%) isolates carried no fimbrial genes, including 66 (21.7%) isolates that did not have any of the above virulence genes. These results suggest a broad array of virulence genes associated with PWD in pigs.  相似文献

Four dietary factors (ad libitum versus feed restriction, control versus protein restriction at ad libitum feeding, control versus inclusion of lupin as a protein source at ad libitum feeding, and control versus extra vitamin E at ad libitum feeding) were tested in four separate experiments for the effect on diarrhoea. To introduce a diarrhoea-like condition, half of the piglets were challenged with an E. coli O 149 dose of 1 × 108 colony forming units on days one and two after weaning (day of weaning = day zero). All piglets were susceptible since the dams were tested mono-zygotic susceptible to the attachment site of E. coli O 149 in the intestines. Each of the four experiments included 32 piglets from 4 sows. The design was a 2 × 2 factorial with dietary factor and E. coli O 149 challenge as the two factors, each at two levels. The piglets were housed individually during the experiment which lasted for 10 days from weaning at 7 weeks of age. The daily recordings included feed intake, weight and faecal score (from 1 = solid and cloddy to 6 = watery and yellow). Faeces from days 1 to 4 were tested for E. coli strains. In addition, blood was sampled and serum was analysed for antibodies to E. coli, IgG and IgM. Generally the E. coli challenge had no effect on growth and feed intake whereas faecal score and number of faecal haemolytic bacteria increased and faecal dry matter decreased. Feed restriction decreased the weight gain while faecal characteristics were unaffected. An analysis including all four experiments revealed that a feed intake of less than 200 g during the first day after weaning seems to be associated with a relatively high incidence of a post-weaning diarrhoea-like condition. Protein restriction decreased faecal score and increased faecal dry matter while weight gain tended to decrease. Inclusion of lupin affected neither weight gain nor faecal characteristics. Extra vitamin E did not affect weight gain while faecal dry matter decreased, and faecal score and number of faecal haemolytic bacteria increased. The dietary treatments had no effect on the measured immunoglobulins. In conclusion, the studied dietary factors could not alleviate a diarrhoea-like condition and at the same time maintain the growth rate. Furthermore, the results indicate that performance can be improved if piglets achieve a daily feed intake of at least 200 g during the first day after weaning.  相似文献

The effect of weaning pigs in pens with slatted versus solid floor dung alleys, and of moving at weaning, was studied on pigs from 201 litters. Weaning was at 5 weeks and no litters were mixed. Four treatments were performed: slatted + moved, slatted + not moved, solid + moved and solid + not moved. Pen hygiene was significantly better in the slatted floor pens and morbidity significantly lower. There were no significant differences in mortality or in daily weight gain. Escherichia coli-associated diarrhoea was the major cause of disease and death, and joint inflammation was also of importance. It was concluded that slatted flooring improved pen hygiene and reduced morbidity due to gastrointestinal diseases. In addition, moving the pigs at weaning to cleaned, slatted floor pens may further reduce disease problems.  相似文献

The use of monensin sodium against naturally transmitted swine dysentery was evaluated in 4-week-old piglets, with an average weight of 8 kg, over a period of 112 days. Three treatments were compared using between two and four pens per treatment and 12 pigs per pen. Monensin was administered via the feed, either immediately post weaning to four pens of pigs (T1), or after 12 days (T2, two pens). The T1 group received monensin at the rate of 100 ppm (days 0-56), 50 ppm (days 57-84) and 25 ppm until the end of the trial. In the other group monensin was given at 100 ppm (days 12-84) and at 50 ppm (days 85-112). Unmedicated feed was given to two pens (T3). The continuous administration of monensin from weaning was effective in the control or prevention of swine dysentery. A significant (P less than 0.05) improvement, in comparison with the other two groups, was observed in terms of mortality, diarrhoea score, average daily gain (ADG) and feed conversion ratio (FCR). There was a reduction in mortality, diarrhoea score/days and an improvement in growth performance parameters in pigs treated with monensin after the disease had been established, with ADG and FCR values significantly (P less than 0.05) different compared with the untreated controls.  相似文献

OBJECTIVE: To determine whether CHEMYDERTM polymer has potential for use in the control of porcine postweaning colibacillosis (PWC). PROCEDURE: Two experiments were conducted in which 50 young pigs, either receiving CHEMYDERTM polymer in their food or not, were challenged orally with cultures of beta-haemolytic Escherichia coli immediately after weaning. Their response in terms of development of diarrhoea, and the extent of colonisation of the intestinal tract by the bacteria was monitored. In a third experiment CHEMYDERTM polymer was added to the water supply of a group of 15 pigs on a piggery where PWC was an ongoing clinical problem. The response of these pigs was compared with that of pigs vaccinated against PWC or left unmedicated. RESULTS: In both experimental infection trials the pigs receiving CHEMYDERTM polymer showed significantly reduced intestinal colonisation with the challenge strain of E coli, and, in trial 2, significantly less diarrhoea after weaning compared to pigs not receiving CHEMYDERTM polymer. In the field trial the pigs receiving CHEMYDERTM polymer had significantly less diarrhoea and required significantly less antibiotic treatment than the other two groups of pigs. CONCLUSION: CHEMYDERTM polymer has potential for use in the control of PWC.  相似文献

Attempts were made to induce an intestinal hypersensitivity response to weaner diet by feeding pigs with small quantities of this material before weaning. In two trials using different weaner diets piglets subjected to this regimen showed no significant differences in small intestinal structure, in ability to absorb xylose, in bodyweight gain, in incidence of diarrhoea or excretion of enteropathogens after weaning compared with pigs not given any of the diet before weaning, or fed with a different diet before weaning. When post-weaning diarrhoea occurred it was associated with an earlier, more prolonged and greater proliferation of enterotoxigenic Escherichia coli in the small intestines than occurred in healthy pigs after weaning. The greater proliferation in pigs which developed diarrhoea could not be attributed either to an excessive dietary intake after weaning, or to a specific proliferation of rotaviruses.  相似文献
