Thrips are damaging pests in pepper worldwide. They can cause damage directly by feeding on leaves, fruits or flowers, and also indirectly by transferring viruses, especially tomato spotted wilt virus (TSWV). Although thrips are among the most damaging pests in pepper, until now there is no commercial variety with a useful level of resistance to thrips. This is at least partly due to the lack of knowledge on resistance levels in pepper germplasm of QTLs and/or genes for resistance, and of information about resistance mechanisms to thrips in pepper. This paper describes our research aimed at developing practical and reliable screening methods for thrips resistance in pepper and at identifying pepper accessions showing a strong resistance to thrips. Thirty-two pepper accessions from four species of pepper (Capsicum annuum, C. baccatum, C. chinense and C. frutescens) and two species of thrips (Frankliniella occidentalis and Thrips parvispinus) were used in this study. Our results indicate that the laboratory based leaf disc test and the detached leaf test can be used as reliable screening methods for thrips resistance in pepper. We observed a large variation for resistance to thrips in Capsicum that can be exploited in breeding programs.  相似文献

采用反复干旱存活法研究了吸湿-回干处理对干制辣椒幼苗抗旱性的影响,筛选可有效提高干制辣椒幼苗抗旱性的最佳吸湿-回干处理方法。结果表明:吸湿-回干处理可有效提高干制辣椒幼苗的抗旱性,处理组的反复干旱幼苗存活率显著高于未处理对照,处理次数之间的差异不明显,吸湿时间对反复干旱幼苗存活率有显著影响,其中吸湿6 h处理的反复干旱幼苗存活率最高,显著优于吸湿24 h的处理;此外,6 h吸湿-回干处理对种子发芽率、发芽势的提高也具有显著的促进作用。综合分析认为,6 h吸湿-回干处理可有效提高干制辣椒幼苗抗旱性。  相似文献

Summary The genes for resistance to pepper strains of TMV in five C. chinense Jacq. accessions, the PI numbers 152225, 159236, 315008, 315023 and 315024, were investigated for allelism. The resistance of these five C. chinense accessions was found to be inherited monogenically and to be partially dominant. The resistance genes in these accessions appeared to be allelic and were also found to be allelic with the known alleles L1 and L. Over 540 accessions of wild Lycopersicon species or their crosses with L. esculentum were screened for resistance in a series of trials. Forty-six accessions were selected for the final screening trial based on lower disease ratings in previous trials. Of these, L. hirsutum had the greatest number of resistant accessions, followed by L. esculentum and L. peruvianum. Twenty accessions were quantified for their levels of resistance based on leaf area infected, area under disease progress curve (AUDPC), and the degree of sporulation. There was a significant positive correlation between the AUDPC calculated from 20 accessions evaluated under growth room and field conditions. Five L. hirsutum accessions had no sporulation associated with leaf lesions, whereas L. esculentum accessions had an average of 1.6×104 conidia/cm2 of leaf tissue. There was significant positive correlation between the AUDPC values and the number of conidia per cm2 of leaf tissue.  相似文献

Mist-chamber, field, and detached leaf inoculation procedures identified plants resistant to bacterial soft rot [Erwinia carotovora subsp. carotovora, (Ecc)] in Brassica rapa and related species. The mist-chamber seedling inoculation provided the best correlation of mean disease severity ratings with the field plant inoculation (r = 0.67**) and was used to identify resistant materials. The optimum mist-chamber incubation conditions to distinguish the resistance of accessions were 23 °C and 100% RH and were used for primary screening. A total of 752 accessions of B. rapa and related Cruciferae were screened. In general, accessions of B. oleracea were more resistant than accessions of B. rapa. Within B. rapa, subspecies pekinensis and chinensis were more susceptible than other subspecies. No completely resistant material was found. In species of B. rapa, only 7% of accessions showed some degree of resistance with plant-to-plant variation within the accessions. G30444, G30449, and AVRDC2837 were identified as the most resistant materials in B. rapa by both mist-chamber and field inoculations. The resistance was correlated between an USA isolate (Geneva-1) and two Chinese isolates (RL4-1 and RL-19) of Ecc. A collection of Cannabis accessions was evaluated for resistance to the root-knot nematode Meloidogyne hapla Chitwood in a seedling test. After inoculation with a larval suspension, significant variation was found for the number of galls and egg masses on the seedling roots. These parameters were considered as estimates for nematode infection and larval multiplication, respectively. A subset of the tested accessions was grown on a naturally infested arable field to study the relation between the test results and host characteristics in the field. The ranking order of accessions for the number of galls in the seedling test agreed well with that for the number of root galls on juvenile plants in the field. Therefore the test provides a useful indication for nematode infection. The number of egg masses in the seedling test and the population density of Meloidogyne measured in post-treatment soil samples of the field trial, showed a comparable ranking of accessions. Differences between accessions in the field were however smaller and not statistically significant.  相似文献

Nuclear RFLP between pepper cultivars (Capsicum annuum L.)   总被引:6,自引:0,他引:6  
Summary Forty one nuclear probes, distributed across the different linkage groups of the previously published map, were used to examine restrition fragment length polymorphism between cultivated peppers. Total DNA from thirteen accessions of Capsicum annuum var. annuum and one accession of C. baccatum var. pendulum was separately cut with ten restriction enzymes. The analyses were restricted to only one enzyme per probe to reduce the polymorphism redundancy. Nei & Li's genetic distances between accessions were calculated from the 141 resultant nuclear DNA restriction fragments. The genetic variation was larger between C. annuum and C. baccatum than between C. annuum cultivars. Large fruited related accessions closely clustered together. Distances between the small fruited cultivars were larger than within the bell pepper group. A correspondence analysis performed on differences between the global RFLP patterns of each accessions of C. annuum revealed the particular genomic structure of four small fruited cultivars: Criollo de Morelos 334, H3, Perennial and Doux Long des Landes. The percentage of probes revealing at least one RFLP with at least one enzyme ranged from zero to fifty percent for all the pairwise comparisons of C. annuum accessions. 82 presumptive loci were detected with a mean number of 1.46 alleles per locus within C. annuum and 1.83 within all the accessions. This result indicates that molecular markers will be more usable in intraspecific study of C. annuum than isozyme markers.  相似文献

The morphological and genetic variations in somaclones of chili pepper (Capsicumannuum L.) derived from tissue culture were evaluated. Cotyledonary node explants of cultivars, Shishitou and Takanotsume, were cultured on Murashige and Skoog (MS) medium supplemented with benzyladenine (BA)5 mg/l for shoots regeneration and regenerated shoots were rooted on MS medium supplemented with naphthalene acetic acid(NAA) 0.1 mg/l and indol-3-butyric acid(IBA) 0.05 mg/l. The regenerated plants(R0) were selfed to obtain seeds for next generation (R1 lines). Qualitative characters were studied in R0generation and both qualitative and quantitative characters were studied in R1 generation. In R0 generation, variations were noticed in plant growth habit, stem color, flower color and color of unripe fruits, and expression of anthocyanin in unripe fruits. Comparison among the R1 lines and their parents were made for morphological and agronomic characters. Significant variation among R1 lines and differences between R1 lines and their parents were observed. Genetic variations among three somaclones were revealed by random amplified polymorphic DNA (RAPD) analysis. Variation, such as early flowering and increase of yield components, is an indication of the response to selection for any specific character. Occurrence of productive variants among somaclones of established cultivars, like Shishitou and Takanotsume, indicates the possibility of their improvement through somaclonal variation. Summary Twenty three accessions of nine Portuguese cabbage and kale land races from different geographic origins were tested at the seedling stage for resistance to several important brassica diseases. Resistance to downy mildew (Peronospora parasitica), expressed as necrosis of the cotyledon mesophyll, was found in all the accessions. Type A resistance to cabbage yellows (Fusarium oxysporum f. sp. conglutinans race 1) was present in most of the landraces. Resistance to clubroot (Plasmodiophora brassicae race 6) was found in one accession of the Portuguese tree kale. High resistance to blackleg (Leptosphaeria maculans) and white rust (Albuco candida) was not detected, although several accessions showed 20 to 30% of plants with intermediate expression of resistance. All Portuguese cole accessions were susceptible to blackrot (Xanthomonas campestris pv. campestris).  相似文献

Fusarium wilt incited by Fusarium oxysporum f. sp. melonis (F.o.m) is one of the most widespread and devastating melon diseases. While resistance to physiological races 0, 1, and 2 is relatively frequent in different botanical varieties, sources of resistance to race 1,2 are restricted to a few Far-Eastern accessions. In this work, the results of a screening for resistance to F.o.m. race 1,2 among 32 accessions are presented. Three Japanese accessions (‘Kogane Nashi Makuwa’, ‘C-211’, and ‘C-40’) showed the highest resistance levels, but useful levels of resistance were also detected in one Russian ‘C-160’ and two Spanish (‘C-300’ and ‘Mollerusa-7’) accessions. These resistant materials, together with other accessions previously described as resistant to F.o.m. races 0, 1, and/or 2 have been morphologically and molecularly characterized. Based on cluster analysis, these accessions have been grouped according to the botanical subspecies they belong to. Assessment of genetic diversity indicated that the resistant accessions to races 0, 1 and 2, are scattered along the established clusters. On the other hand, high levels of resistance to the race 1,2 could be found only among accessions belonging to Cucumis melo subsp. agrestis, nevertheless, a certain degree of resistance to this race could also be found within some accessions belonging to subsp. melo. As far as we know, this is the first report of resistance to F.o.m race 1,2 found out from the Far-Eastern melon material. Based on fruits characteristics, it appears that several inodurus and cantalupensis accessions could be exploited in breeding programs as resistance sources to F.o.m races 0, 1 and/or 2 for the improvement of these melon types. The accessions with the highest levels of resistance to the race 1,2 appeared to be very distant both molecularly and morphologically from the commercial types. Nevertheless ‘C-160’, ‘C-300’, and ‘Mollerusa-7’ classified as var. inodorus are morphologically very similar to the Spanish commercial types and might be used as resistant sources in breeding these melon types.  相似文献

Summary A total of 125 novel accessions of Capsicum annuum L. and Capsicum baccatum L. were screened for sources of resistance to Verticillium wilt (Verticillium dahliae Kleb). A soil infestation method with 2000 microsclerotia of V. dahliae per gram of planting medium, and a soil temperature of 25±3°C, identified 27 Capsicum accessions with resistance to Verticillium wilt. P.I. 215699 (a mixture of Capsicum baccatum var. microcarpum and Capsicum annuum), P.I. 535616 (Capsicum annuum), and P.I. 555614 (Capsicum annuum), had the lowest disease severity and the highest percentage of resistant plants.  相似文献

Summary The current powdery mildew (Sphaerotheca fuligninea) resistant cucumber varieties suffer from leaf chlorosis during autumn, winter and early spring cultivation in the Netherlands. Therefore screening was carried out for novel powdery mildew resistance genes. From 177 accessions, derived from different sources, 108 accessions proved to be partially resistant to S. fuliginea. Crosses were made with 53 resistant accessions to distinguish the presence of novel genes. It is likely that the accessions C. sativus 2145, C. sativus LV 41, PI 188807, Vladivostokij, White and Yellow 1 have one or more recessive powdery mildew resistance genes, different from powdery mildew resistance genes of the line NPI, which was used for variety breeding. As one of the genic male sterility (GMS) materials in chili pepper ( Capsicum annuum L.), GMS1 has been used for commercial F1 hybrid seed production. The male sterility of GMS1 is controlled by a recessive nuclear gene, named ms 1 . In this study, we developed DNA markers linked to the ms 1 locus using a combination of bulked segregant analysis and amplified fragment length polymorphism (AFLP) in a segregating sibling population. From the screening of 1024 AFLP primer combinations, the AFLP marker E-AGC/M-GTG (514 bp) was identified as being linked to the ms 1 locus at a distance of about 3 cM. Based on internal sequencing analysis of the E-AGC/M-GTG marker between male fertile and sterile plants, we identified three small deletions with a size of altogether 42 bp in the male-fertile plant and developed a codominant sequence characterized amplified region (SCAR) marker. This SCAR marker may be valuable for marker-assisted breeding in the hybrid seed production system of chili pepper using the GMS1 line.  相似文献

The soybean aphid, Aphis glycines Matsumura, is a pest of soybean [Glycine max L. (Merrill)] in Asia, and its recent establishment in North America has led to large, recurring outbreaks that have challenged pest management practitioners there to seek environmentally responsible means for its control. Growth-chamber experiments were conducted to determine and characterize host-plant resistance among several soybean accessions. Soybean plants were first screened for resistance by rating the population growth of A. glycines in two tests. All plants of PI 230977 and 25% of PI 71506 plants were resistant (≤100 aphids per plant) in the first screening test. All ‘Dowling’, PI 71506 and PI 230977 were resistant (≤150 aphids per plant), and 50% of plants of line ‘G93-9223’ were resistant in the second test. Follow-up experiments showed that antixenosis was a modality of resistance based on reduced nymphiposition by A. glycines on Dowling, PI 230977 and PI 71506 in no-choice tests and on fewer numbers of A. glycines on Dowling, PI 230977, PI 71506 and G93-5223 in distribution tests. Antixenosis in Dowling and PI 230977 was stronger in the unifoliolate leaves than in other shoot structures, whereas distribution of A. glycines within plants of PI 71506 and G93-5223 suggested comparable suitability between unifoliolate leaves and other shoot structures of these accessions. Antibiosis to A. glycines was evident as a lower proportion of aphids that reproduced on PI 230977 and from fewer progeny on PI 230977 and Dowling than on 91B91. The number of days from birth to reproduction by A. glycines did not differ among accessions. Results confirmed Dowling and PI 71506 as strong sources of resistance to A. glycines. The levels of antixenosis and antibiosis to A. glycines in PI 230977 and antixenosis to A. glycines in G93-9223 suggest that these accessions may also be valuable to soybean breeding programs as sources of resistance.  相似文献

Summary Seedling responses to one Australian isolate of each of the stripe rust, stem rust and leaf rust pathogens were determined for 541 accessions of T. dicoccoides collected from 23 locations in Israel. Resistance to stripe rust was more frequent than resistance to stem rust. Stripe rust responses showed a wide range of variability indicative of a number of genes for resistance. Comparison of the present stem rust data and that reported for the same accessions tested in Israel indicated that different genes were operating in each country. Only moderately resistant responses to stem rust were obtained. This level of resistance is probably inadequate for transfer to commercial wheat cultivars. We found no potentially useful seedling resistance to leaf rust.  相似文献

Seventeen tea accessions belonging to Chinese (Camellia sinensis), Assamic (C. sinensis var. assamica), and Shan tea (C. sinensis var. pubilimba) groups, which are either commercially planted or new promising tea germplasm, were morphologically described at Phu Tho province (Viet Nam) and assessed for their diversity. Fourteen phyllometric parameters were qualitatively and quantitatively investigated using digital image analysis. The accessions were then discriminated by a dedicated artificial neural network for univocal plant identification and a hierarchical cluster analysis was performed in order to build a dendrogram reporting the relationships among them. Results proved the diversity of investigated tea morphotypes from Phu Tho province based on a morphological screening. More, the artificial neural network was able to perform a correct identification for almost all the accessions using simple dedicated instruments.  相似文献

Between 580 and 700 accessions of related cultivated and wild species of the genus Beta were assessed for resistance to four soil-borne diseases of sugar beet: two seedling damping-off diseases caused by the fungi Aphanomyces cochlioides and Pythium ultimum and two diseases of more mature plants, Rhizoctonia root and crown rot, caused by the fungus R. solani, and Rhizomania, caused by Beet necrotic yellow vein virus (BNYVV), a furovirus transmitted by the plasmodiophorid Polymyxa betae. Analysis of resistance data (assessed on an international standardised 1–9 scale of Resistance Scores) indicated that the highest levels of resistance ({RS} 2) to A. cochlioides and P. ultimum were to be found amongst accessions of the more distantly related sections Corollinae (93% of accessions tested) and Procumbentes (10%), respectively; although useful levels could also be found in the more closely related, and sexually compatible, section Beta (1–6%). Resistance to Rhizoctonia was also found in section Beta (5–7%), depending on whether field or glasshouse tests were used, but there was little evidence of generally high levels of resistance to Rhizomania among accessions of this section. None of the accessions of sections Corollinae and Procumbentes exhibited any notable resistance to Rhizoctonia. However, all sections Procumbentes and some sections Corollinae (4%) accessions were highly resistant to Rhizomania. Individuals with high levels of resistance to Rhizomania were identified from within some section Beta and Corollinae accessions, in which there was evidence of segregation.  相似文献

Nineteen accessions of the tuber-bearing species Solanum berthaultii, S. chacoense, S. leptophyes, S. microdontum, S. sparsipilum, S. sucrense, S. venturii, S. vernei and S. verrucosum were tested for their resistance to late blight in two years of field experiments. Plants were artifically inoculated with zoospores of race 1.2.3.4.5.7.10.11 and the development of the disease was followed. Resistance ratings, calculated as the areas under the disease progress curves (ADPC), demonstrated a high resistance in all accessions except in S. sparsipilum, S. leptophyes and their interspecific hybrid. Segregations suggest that major genes for resistance are present in S. sucrense and S. venturii, and may also play a role in S. verrucosum. It is not yet certain wether the resistance of the other accessions is comparable to the partial and durable resistance of S. tuberosum cultivars like Pimpernel, as inheritance and mechanism have yet to be established. However, segregations suggesting the presence of single major genes with complete dominance were not found in these other accessions. Tuber initiation in the field occurred in only one accession, S. tuberosum ssp. andigena, and maturity of the clones was not related to their resistance. In the other accessions maturity types could not be assessed, as the clones require short day conditions for tuber initiation.  相似文献

Summary Fruit rot in bell pepper caused by Phytopthora capsici Leon. is a limiting factor in its commercial production. Crosses were made between Waxy Globe, a small fruited hot type, resistant to fruit rot and four highly susceptible bell pepper commercial cultivars (Chinese Giant, California Wonder, Osh, Kosh and Yolo Wonder). Studies of the F1's, F2's and backcrosses indicated that Waxy Globe carries a dominant gene imparting resistance to fruit rot. After three backcrosses with California Wonder, lines were obtained combining the resistance gene of Waxy Globe with the large fruit size and low capsaicin of California Wonder.  相似文献

Summary The reactions to infection with two cultures of Puccinia hordei were determined for 292 Hordeum spontaneum (syn. H. vulgare ssp. spontaneum) accessions, collected at 16 sites which encompassed the ecological range of H. spontaneum in Israel. Culture Tel-Aviv was from Israel and culture 57.19 was from the United States. Fifty-two percent of the accessions were resistant to culture Tel-Aviv and 67% were resistant to culture 57.19. Forty-three percent of the accessions were resistant to both cultures. The average infection type (IT) of accessions within sites ranged from 2.7 to 7.5 on a 0–9 rating scale. The results showed that the presence of Ornithogalum species, the alternate hosts of P. hordei, may increase the percentage of H. spontaneoum accessions resistant to P. hordei. More accessions were resistant at sites where humidity at 1400 was higher, the annual evaporation was lower, and where the glumes were shorter. Kernel weight and annual rainfall was not correlated with resistance. A lower percentage of H. spontaneum accessions were resistant to P. hordei culture Tel-Aviv from Israel than to culture 57.19 from the United States. In a previous study a lower percentage of H. spontaneum accessions also was found to be resistant to a culture of Erysiphe graminis hordei from Israel than to cultures from other countries. Previous studies also have shown that cultures of P. hordei and E. graminis hordei from Israel have many genes for virulence on barley, and that H. spontaneum accessions from Israel have many genes for resistance to these two pathogens. Previous results and the results reported in this paper support the hypothesis of coevolution of resistant host genes and virulent pathogen genes where hosts and pathogens have coexisted for many thousand years.  相似文献
