Three experiments evaluated the effects of dietary Se and vitamin E on the ultrastructure of spermatozoa, ATP concentration of spermatozoa, and the effects of adding sodium selenite to semen extenders on subsequent sperm motility. The experiment was a 2 x 2 arrangement of treatments in a randomized complete block design. A total of 10 mature boars were fed from weaning to 18 mo of age diets fortified with two levels of supplemental Se (0 or .5 ppm) or vitamin E (0 or 220 IU/kg diet). The nonfortified diets contained .06 ppm Se and 4.4 IU vitamin E/kg. In Exp. 1, the spermatozoa from all boars were examined by electron microscopy. Vitamin E had no effect on structural abnormalities in the spermatozoa. When the low-Se diet was fed the acrosome or nuclei of the spermatozoa was unaffected, but the mitochondria in the tail midpiece were more oval with wider gaps between organelles. The plasma membrane connection to the tail midpiece was not tightly bound as when boars were fed Se. Immature spermatozoa with cytoplasmic droplets were more numerous when boars were fed the low-Se diet, but the occurrence of midpiece abnormalities occurred in boars fed diets with or without Se or vitamin E. Our results suggest that Se may enhance spermatozoa maturation in the epididymis and may reduce the number of sperm with cytoplasmic droplets. In Exp. 2, the concentration of ATP in the spermatozoa was evaluated in the semen of all treatment boars. When the low-Se diet was fed, ATP concentration was lower (P < .01), whereas vitamin E had no effect on ATP concentration. Experiment 3 investigated the effect of diluting boar semen with a semen extender with sodium selenite added at 0, .3, .6, or .9 ppm Se. Three ejaculates from each boar were used to evaluate these effects on sperm motility to 48 h after dilution. Sperm motility declined (P < .01) when Se was added to the extender, and this decline was exacerbated as the concentration of added Se increased (P < .01). The added Se was demonstrated to be tightly adhered to the spermatozoa. Overall, these results suggest that low Se-diets fed to boars resulted in abnormal spermatozoal mitochondria, a lower ATP concentration in the spermatozoa, and a loose apposition of the plasma membrane to the helical coil of the tail midpiece, but no effect from inadequate vitamin E was demonstrated. The objective was to compare growth and physiological responses in boars fed diets supplemented with organic or inorganic sources of Se. At weaning, crossbred boars (n = 117; 8.3 kg of BW) were placed in nursery pens (3 boars/pen) and assigned within BW blocks to receive on an ad libitum basis 1 of 3 dietary treatments: I) basal diets with no supplemental Se (controls), II) basal diets supplemented with 0.3 mg/kg of organic Se, and, III) basal diets supplemented with 0.3 mg/kg of sodium selenite (13 pens/dietary treatment). Average daily gain (470 g/d), ADFI (896 g/d), and G:F (0.54) were similar among groups. Blood Se concentrations were greater (P < 0.01) for boars consuming organic Se (107.5 ± 4.8 μg/L) or sodium selenite (114.7 ± 4.8 μg/L) compared with controls (28.4 ± 4.8 μg/L). Intact pens of boars (11 pens/dietary treatment) were moved to a grow-finish barn and continued to receive appropriate diets on an ad libitum basis. Average daily gain (1,045 g/d) and ADFI (2,716 g/d) were similar among groups. Gain:feed was affected by treatment (P = 0.02) and was greater (P < 0.06) for boars fed organic Se (0.378 ± 0.004) compared with boars fed sodium selenite (0.368 ± 0.004) or controls (0.363 ± 0.004). Blood Se concentrations were greater (P < 0.01) in grow-finish boars consuming organic Se (198.9 ± 5.5 μg/L) than boars consuming sodium selenite (171.4 ± 5.4 μg/L) or controls (26.7 ± 5.4 μg/L). Treatment did not affect (P > 0.15) HCW, dressing percent, carcass length, LM area, standardized fat-free lean, lean percentage, backfat thickness, visual color, firmness, marbling, or Minolta loin color scores. Selenium supplementation did not affect (P > 0.17) testis or accessory sex gland sizes. Concentrations of Se in loin, liver, kidney, testis, cauda epididymis, and accessory sex glands were greatest (P < 0.01) in boars receiving organic Se, intermediate in boars receiving sodum selenite, and least in control boars. Microarray analysis of testis gene expression did not detect differences (P > 0.05) due to dietary treatment. Testis gene expression of glutathione peroxidase 4, as determined using quantitative PCR, was increased (P < 0.01) in boars fed organic Se compared with those fed sodium selenite. In summary, dietary supplementation of boars with organic Se failed to alter ADG or ADFI but enhanced G:F during grow-finish. More research is needed to discern the mechanism by which organic Se improves feed efficiency in boars.  相似文献

An experiment involving a total of 61 crossbred boars evaluated the effects of dietary Se and vitamin E on spermatogenic development at various stages of sexual development and the prostaglandin F2alpha (PGF2alpha) content in the seminal vesicle and prostate glands at 18 mo of age. The experiment from 5.4 to 9 mo of age was conducted as a 2 x 2 factorial in a randomized complete block design. Dietary Se at 0 or .5 ppm was the first factor and vitamin E at 0 or 220 IU/kg diet was the second. From 9 to 18 mo of age, a group of sexually active and inactive boars was a third factor. Treatment diets were fed from weaning (28 d of age) to the end of the experiment. Three boars per treatment group at 5.4 (105 kg BW), 6.2 (130 kg BW), and 9.0 (150 kg BW) mo of age were killed and the testes collected. From 9 to 18 mo of age, three boars from each dietary treatment group were used for semen collection, and another set of three to four boars from each treatment group remained sexually inactive. At 18 mo, both sets of boars were killed and their testes, prostates, and seminal vesicles were collected. The testis at each age was evaluated for sperm reserve numbers and germ and Sertoli cell populations. At 5.4 or 6.2 mo of age, testicular sperm reserves were not affected by dietary Se (P > .15), at 9.0 mo of age there was a trend for a higher (P < .10) number of sperm reserves, and by 18 mo of age the Se-fed boars had higher (P < .01) numbers of sperm reserves. Vitamin E had no effect (P > .15) on testicular sperm reserves at any age period. Boars fed dietary Se had a greater number of Sertoli cells (P < .01) and round spermatids (P < .01) at 6.2 mo of age, but by 18 mo of age the boars fed Se had more Sertoli cells (P < .05), more secondary spermatocytes (P < .01), and more round spermatids (P < .05). Vitamin E did not affect Sertoli or germ cell populations at the various ages. Boars at 18 mo of age had lower PGF2alpha concentrations in the prostate (P < .05) and seminal vesicles (P < .01) when vitamin E was fed, whereas Se had no effect. Sexually active boars had lower PGF2alpha concentrations in the seminal vesicles (P < .01) than sexually inactive boars, but there was no effect (P > .15) of sexual activity on the number of Sertoli cells, primary or secondary spermatocytes, or round spermatids. Our results indicate that Se has a role in establishing the number of boar spermatozoal reserves and Sertoli cells, whereas supplemental vitamin E did not affect these criteria.  相似文献

Sugarcane molasses is a widely used animal feed by-product, but is concentrated in S (approximately 1%, DM basis) and has been shown to reduce the Cu status of cattle. Dietary S may also antagonize Se; therefore, two 90-d studies were conducted with forage-fed, yearling steers (12 pens; 2 steers/pen for each study) to investigate the impact of molasses supplementation on measures of Se status. In Exp. 1, steers were assigned isonitrogenous supplements with equivalent amounts of TDN from 2 sources (molasses or corn). Supplemental Se was provided (3.0 mg of Se/d; Na selenite) to both treatments. After 90 d of supplementation, steers provided corn diets had greater (P = 0.02) liver Se concentrations and tended (P = 0.07) to have greater ADG compared with steers supplemented with molasses. Irrespective of treatment (P >/= 0.54), plasma Se concentrations decreased (P < 0.001) and plasma glutathione peroxidase activity increased (P < 0.001) from d 0 to 90. In Exp. 2, sources of supplemental Se (2.5 mg/ d), fed within molasses supplements, were compared. Thirty-two boars were fed diets containing 0, 3, 6 or 9 ppm zearalenone from 32 d of age up to 145 or 312 d of age. Rate of gain, feed intake and gain:feed were not significantly different among treatments. Feeding of diets containing 9 ppm zearalenone did not affect the libido in seven boars. There was a tendency for boars fed 9 ppm dietary zearalenone to produce lower total and gel free volumes of semen, with lower total motile sperm; however, only the percent motility of the sperm was reduced (P less than .05).  相似文献

The aim of the present study was to determine the effects of dietary supplements of vitamins on vitamin status, libido, and semen characteristics in young boars under normal and intensive semen collection. Sixty Landrace, Yorkshire, and Duroc boars were allocated randomly from 6 to 10 mo of age to one of the following diets: 1) basal diet (industry level) for minerals and vitamins (Control, n = 15); 2) basal diet supplemented with vitamin C (ASC, n = 15); 3) basal diet supplemented with fat-soluble vitamins (FSV, n = 15); and 4) basal diet supplemented with water-soluble vitamins (WSV, n = 15). After puberty (approximately 12 mo of age), semen was collected at a regular frequency (three times every 2 wk) for 5 wk. Thereafter, all boars were intensively collected (daily during 2 wk). A recovery period (semen collection three times every 2 wk) followed and lasted for 10 wk. Sperm quality (percentage of motile cells and percentage of morphologically normal cells) and quantity (sperm concentration, semen volume, and total sperm number) were recorded as well as direct and hormone related measurements of boar libido. Blood and seminal plasma samples were taken to monitor vitamin status. High concentrations of B6 (P < 0.05) and folic acid (P < 0.05) were observed in the blood plasma of WSV boars, whereas greater concentrations of vitamin E (P < 0.01) were obtained in FSV boars. In the seminal plasma, folic acid concentrations tended to be greater in WSV boars (P < 0.08). During the intensive collection period, there was a tendency (P < 0.06) for semen production to be greater in WSV boars, the effect being less pronounced (P < 0.10) in FSV boars. During the recovery period, the percentage of motile sperm cells was greater in WSV boars (P < 0.03) and, to a lesser extent, in FSV boars (P < 0.10) compared with Control boars. Sperm morphology and libido were not affected by treatments. These results indicate that the transfer of vitamins from blood to seminal plasma is limited and the dietary supplements of water-soluble and fat-soluble vitamins may increase semen production during intensive semen collection.  相似文献

To investigate the effects of supplemental Se on the transfer of Se to nursing pigs when sows are fed diets containing a Se level above the NRC recommendation (0.15 ppm), sows were fed diets containing no supplemental Se or supplemental (0.3 ppm) Se from sodium selenite or Se yeast. A nonSe-fortified corn-soybean meal basal diet with a high endogenous Se content served as the negative control (0.20 to 0.23 ppm Se). Fifty-two sows were fed diets from 60 d prepartum until 14 d of lactation. Six sows per treatment were bled at 60 and 30 d prepartum, at farrowing, and at 14 d postpartum to measure serum Se concentrations. Colostrum was collected within 12 h postpartum, and milk was collected at 14 d of lactation. Blood was obtained from 3 pigs each from 12 litters per treatment at birth and at weaning (d 14), and pooled serum was analyzed for Se and immunoglobulin G concentrations and glutathione peroxidase activity. Regardless of treatment, serum Se in sows declined throughout gestation and gradually increased during lactation. Sows fed Se yeast tended (P < 0.06) to have greater serum Se at farrowing than sows fed unsupplemented diets. Colostrum and milk (d 14) Se concentrations increased (P < 0.01) when sows were fed Se from yeast but not from sodium selenite. At birth, serum Se was increased (P < 0.01) for pigs whose dams were fed Se yeast compared with pigs from sows fed the basal diet. At 14 d of age, there was no difference in serum Se concentration of pigs from dams fed any of the treatments. Pig serum immunoglobulin G concentrations and glutathione peroxidase-1 activity were unaffected by dietary Se source. Supplementation of gestating and lactating sow diets with Se (0.3 ppm) from an organic or inorganic source reduced the number of stillbirths per litter. However, only pigs born to sows fed organic Se (Se yeast) had greater serum Se at birth. A total of 96 crossbred pigs received various levels of sodium selenite to determine the effect of dietary selenium (Se) on growing swine fed corn-soybean meal diets. Levels of supplemental Se were 0, 4, 8, 12, 16 and 20 micrograms/g. There were linear decreases (P less than .01) in both gain and feed intake with increasing levels of dietary Se. Feed/gain increased numerically as dietary Se increased. Hair Se increased quadratically (P less than .01) and blood Se increased linearly (P less than .01) with increasing level of dietary Se. Cell volume and hemoglobin were not affected by dietary treatment. Increasing dietary Se significantly increased glutathione peroxidase (GSH-Px), glutamic-oxalacetic transaminase (GOT). and glutamic-pyruvic transaminase (GPT). External signs of selenosis were noted in some pigs fed 12 or 20 micrograms/g of Se. The toxic level of Se in a corn-soybean meal diet for crossbred pigs appears to be between 4 and 8 micrograms/g. Of variables studied, growth rate was the most sensitive indicator of chronic selenosis in swine.  相似文献

A 2 x 3 factorial experiment conducted in three replicates of a randomized complete block design compared the effects of calcium selenite and sodium selenite at three different levels of Se (.3, 5, or 15 ppm) in the diets of growing swine on performance and tissue Se concentrations. Ninety pigs averaging 12.5 kg of BW were given ad libitum access to corn-soybean meal diets fortified with one of the treatment Se sources and dietary levels for a 35-d experimental period. Growth and feed intake were similar in pigs fed .3 and 5 ppm of Se but were lower (P less than .01) in those fed 15 ppm from either Se source. Serum Se increased (P less than .01) as dietary Se level increased with no difference between Se sources at each dietary Se level. Liver, kidney, and longissimus muscle Se concentrations increased (P less than .01) as the dietary level of Se increased and were similar when either Se sources was provided. These results indicate that calcium selenite was as effective as sodium selenite using the measurement criteria of growth, serum, and tissue Se concentrations and glutathione peroxidase activities of growing swine when fed at approved, marginally toxic, and toxic dietary Se levels.  相似文献

旨在评价酵母培养物(YC)对公猪生产性能的影响。将90头体重约为80kg的公猪随机分为3组,即对照组、试验Ⅰ组和试验Ⅱ组;对照组饲喂基础日粮,试验Ⅰ组和试验Ⅱ组基础日粮中分别添加5kg/t和7kg/t的酵母培养物,预试期5d,正试期45d,试验期间,3组饲养管理条件一致;试验结束时,评定各组公猪精液的颜色、气味,测定并比较各组公猪的精液量、精液pH值、精子活力、精子密度、精子畸形率、抗力指数。结果表明,试验Ⅰ组和试验Ⅱ组公猪的精液颜色和精液气味与对照组相比没有明显差异;3组公猪的精子抗力指数一致;试验Ⅰ组和试验Ⅱ组的精液量、精子活力、精子密度均显著大于对照组(P<0.05),精子畸形率均显著小于对照组(P<0.05),但这4项指标在2个试验组之间差异均不显著(P>0.05);3组公猪的精液pH值差异均不显著(P>0.05)。综上提示,基础日粮中添加一定量的酵母培养物能够提高公猪部分生产性能指标。  相似文献

The objective of this study was to investigate the effects of a preparation from herbal extracts (PHE) on libido and semen quality in breeding artificial insemination boars. Ten fertile boars were divided into control and experimental groups according to significant difference of libido. There were no differences in semen quality between groups. Animals were fed a commercial feeding mixture for boars. The feeding mixture for the experimental group was enriched with PHE, which was prepared from Eurycoma longifolia, Tribulus terrestris and Leuzea carthamoides. Duration of the experiment was 10 weeks. Samples of ejaculate were collected weekly. Libido was evaluated according to a scale of 0-5 points. Semen volume, sperm motility, percentage of viable spermatozoa, sperm concentration, morphologically abnormal spermatozoa, daily sperm production and sperm survival were assessed. Amounts of mineral components and free amino acids were analysed in seminal plasma. Significant differences were found in these parameters: libido (4.05 ± 0.22 vs 3.48 ± 0.78; p < 0.001), semen volume (331.75 ± 61.91 vs 263.13 ± 87.17 g; p < 0.001), sperm concentration (386.25 ± 107.95 vs 487.25 ± 165.50 × 10(3) /mm(3); p < 0.01), morphologically abnormal spermatozoa (15.94 ± 11.08 vs 20.88 ± 9.19%; p < 0.001) and Mg concentration (28.36 ± 11.59 vs 20.27 ± 13.93 mm; p < 0.05). The experimental group's libido was increased by 20% in comparison with the beginning of the experiment. Results of this study showed positive effect of PHE on libido and some parameters of boar semen quality.  相似文献

One-day-old Taiwan native male chicks were fed with maize-soybean rearing diets without supplemental vitamin E to 23 weeks of age. From 23 to 52 weeks of age, the cockerels (n = 90) were assigned at random to 5 dietary treatments and fed with maize-soybean diets supplemented with 0, 20, 40, 80 and 160 mg/kg of vitamin E (dl-alpha-tocopherol acetate). Pullets (225) of the same age were fed with standard diets throughout. They were artificially inseminated with one dose of 0.04 ml/bird intact and 5-fold diluted pooled semen at 31 to 43 weeks of age and at 49 weeks of age, respectively. The criteria evaluated included: semen quality, fertility and maximum and effective duration of fertility, blood characteristics, body and testes weight. Supplemental vitamin E did not affect cockerels' effective duration of fertility and percentage of fertility. However, when pullets were inseminated with diluted semen, supplementing 160 mg/kg vitamin E increased the maximum duration of fertility at 49 weeks of age. Cockerels receiving 40 to 160mg/kg supplements had higher sperm viability and motility after 39 weeks of age and those fed 80 mg/kg had higher sperm concentration at 39 weeks of age. Cockerels receiving supplements of more than 40 mg/kg vitamin E had higher body weight gain. Plasma cholesterol and testosterone were not affected by supplemental vitamin E. However, plasma luteinising hormone (LH) concentration was lower in cockerels fed 160 mg/kg. Lack of supplemental vitamin E over 39 weeks was associated with lower semen quality but did not reduce the proportion of fertile eggs laid by inseminated hens, perhaps because the insemination dose compensated for low sperm quality. We found that the maximum duration of fertility might be improved by supplementing 160 mg/kg vitamin E at 49 weeks of age.  相似文献

High dilution rates have been documented as detrimental for boar spermatozoa, shortening their lifespan (Centurion et al. 2003, Biol Reprod 69: 640–646). Addition of seminal plasma (SP) to semen extenders, or selenium (Se) and vitamin E (VE) in diet of boars could increase motility of highly diluted spermatozoa (HDS). The aim of this work was to evaluate the effect of seminal plasma on sperm motility of HDS from boars feed with Se and VE. Sixteen 12 month-old boars were designed to one of four dietary treatments: (i) control, Se 0 ppm–VE 0 IU/kg; (ii) Se 0–VE 250; (iii) Se 0.5–VE 250 and (iv) Se 0.5–VE 0. Boars were treated for 8 weeks before semen collection. Sperm rich fractions from each boar were diluted to 5 × 106 sperm/ml in PBS medium and incubated at 37°C with or without 10% SP. The measurements were done at 0, 2 or 5 h. Data were analyzed as a mixed model for a factorial design [2 (Se) × 2 (VE) × 2 (SP) × 3 (h)]. Percentage of sperm motility (PSM) increases significantly (p < 0.001) with addition of Se (81.3 ± 1.52), VE (81.0 ± 1.62) and SP (81.5 ± 1.57) vs control (73.4 ± 1.61). There was significant interaction Se × VE (p < 0.001) and Se × VE × SP (p < 0.05) in PSM. However, PSM was affected significantly by time (0 h 83.4 ± 1.92; 2 h 80.7 ± 1.92 and 5 h 67.9 ± 1.92; p < 0.001). There was significant interaction SP × Time (p < 0.05) in PSM. These results indicate that Se, VE and SP improve seminal viability. Addition of 10% of SP maintains PSM at least during 5 hours.  相似文献

The aim of this study was to characterize growth and sperm production parameters in Ogaden bucks fed a basal diet of hay and supplemented with agro-industrial by-products and Khat leftovers in Ethiopia. Thirty-five bucks with a mean (+/-SD) initial live body weight (BW) of 15.5 +/- 1.5 kg were randomly assigned to one of four dietary treatments for a period of 13 weeks. Treatments consisted of native hay fed ad libitum (control; C), native hay supplemented with a 1% of BW agro-industrial by-products (treatment 1; T1), native hay supplemented with Khat (Catha edulis) leftovers at a rate of 1% of BW (treatment 2; T2) and Khat leftovers fed ad libitum (treatment 3; T3). Bucks fed on T1-T3 had higher BW, body condition score, scrotal circumference (SC), testicular width and testicular length, compared to controls (P < 0.05). Also, bucks in T1-T3 had higher sperm progressive motility, sperm concentration per ml and total number of spermatozoa per ejaculate compared to controls (P < 0.05). Between treatments, bucks in T3 recorded the highest BW (17.2 +/- 0.16) and testicular size (21.1 +/- 0.17 cm). Both testicular and epididymal weight and dimensions were significantly affected (P < 0.05) by supplementation compared to controls. Testicular size was positively correlated to live BW (r = 0.53, P < 0.001). SC was positively correlated with ejaculate volume (r = 0.37, P < 0.001), sperm mass activity (r = 0.65, P < 0.001) and individual sperm progressive motility (r = 0.40; P < 0.001). Supplementation with Khat leftovers induced the highest improvement in live BW, testicular size, semen production and sperm motility in Ogaden bucks and can possibly be considered as a feed supplement to enhance goat production under smallholder livestock farming system in Ethiopia.  相似文献

On December 2, 1999, 120 pregnant cows were weighed, their body condition scored, and then sorted into six groups of 20 stratified by BCS, BW, breed, and age. Groups were assigned randomly to six, 5.1-ha dormant common bermudagrass (Cynodon dactylon [L.] Pers.) pastures for 2 yr to determine the effects of supplemental Se and its source on performance and blood measurements. During the winter, each group of cows had ad libitum access to bermudagrass/dallisgrass (Paspalum dilatatum Poir.) hay plus they were allowed limited access (1 to 4 d/wk) to a 2.4-ha winter-annual paddock planted in half the pasture. Treatments were assigned randomly to pastures (two pastures per treatment), and cows had ad libitum access to one of three free-choice minerals: 1) no supplemental Se, 2) 26 mg of supplemental Se from sodium selenite/kg, and 3) 26 mg of supplemental Se from seleno-yeast/kg (designed intake = 113 g/cow daily). Data were analyzed using a mixed model; year was the random effect and treatment was the fixed effect. Selenium supplementation or its source had no effect (P > or = 0.19) on cow BW, BCS, conception rate, postpartum interval, or hay DMI. Birth date, birth weight, BW, total BW gain, mortality, and ADG of calves were not affected (P > 0.20) by Se or its source. Whole blood Se concentrations and glutathione peroxidase (GSH-Px) activity at the beginning of the trial did not differ (P > or = 0.17) between cows receiving no Se and cows supplemented with Se or between Se sources. At the beginning of the calving and breeding seasons, cows supplemented with Se had greater (P < 0.01) whole blood Se concentrations and GSH-Px activities than cows receiving no supplemental Se; cows fed selenoyeast had greater (P < or = 0.05) whole blood Se concentrations than cows fed sodium selenite, but GSH-Px did not differ (P > or = 0.60) between the two sources. At birth and on May 24 (near peak lactation), calves from cows supplemented with Se had greater (P < or = 0.06) whole blood Se concentrations than calves from cows fed no Se. At birth, calves from cows fed seleno-yeast had greater (P < or = 0.05) whole blood Se concentrations and GSH-Px activities than calves from cows fed sodium selenite. Although no differences were noted in cow and calf performance, significant increases were noted in whole blood Se concentrations and GSH-Px activities in calves at birth as a result of feeding of seleno-yeast compared to no Se or sodium selenite.  相似文献

1. The effects of supplemental dietary vitamin E and organic selenium (Se), and their combination, on improving semen quality characteristics and antioxidative status were investigated in cockerels exposed to high ambient temperature.

2. A total of 36 Egyptian local cross males, 40 weeks old, were housed individually in cages in an open-sided building (average daily temperature ranged from 33 to 36°C and relative humidity from 60 to 70%). Birds were divided randomly into 4 experimental treatments (n?=?9 each): (1) control (basal diet without any supplementation with vitamin E or Se); (2) vitamin E (basal diet +200?mg α-tocopherol acetate/kg diet); (3) Se (basal diet +0·3?mg organic Se/kg diet); and (4) vitamin E?+?Se (basal diet +200?mg α-tocopherol acetate/kg diet +0·3?mg organic Se/kg diet).

3. Under heat stress conditions, inclusion of vitamin E and/or organic Se in the diets enhanced the semen quality traits, including the spermatozoa count and motility, and reduced the percentage of dead spermatozoa.

4. A combination of 200?mg/kg vitamin E with 0·3?mg/kg organic Se reduced the thiobarbituric acid reactive substance concentration in seminal plasma samples to about 28% of the controls; and also enhanced the seminal plasma glutathione peroxidase activity by two-fold compared with controls.

5. It was concluded that dietary vitamin E in combination with organic Se has a synergistic effect in minimising lipid peroxidation and improving the antioxidative status in seminal plasma of the domestic fowl, which probably translated into enhanced spermatozoa count, motility and reduced percentage of dead spermatozoa under heat stress conditions.  相似文献

本试验旨在研究牛磺酸(Tau)对种公猪性欲、精液品质、血清激素含量及精浆抗氧化能力的影响。选用24头年龄、体重相近的健康成年大约克夏种公猪,随机分为4个组,每组6个重复,每个重复1头猪。各组分别饲喂牛磺酸添加水平为0(对照组)、2、4、6 g/kg的饲粮,试验期90 d,分为1~45 d和46~90 d 2个阶段。结果表明:1)试验46~90 d时,添加6 g/kg牛磺酸显著提高了种公猪的性欲(P0.05)、采精量(P0.01)、精子活力(P0.05),4 g/kg牛磺酸改善了精子密度和精子畸形率(P0.05);2)与对照组相比,6 g/kg牛磺酸水平组极显著提高了种公猪血清促黄体素(LH)含量(P0.01),显著提高了睾酮(T)含量(P0.05);3)试验46~90 d时,6 g/kg牛磺酸显著降低了种公猪精浆丙二醛(M DA)含量(P0.05),显著提高了超氧化物歧化酶(SOD)活力(P0.05),4 g/kg牛磺酸显著提高了谷胱甘肽过氧化物酶(GSH-Px)活力(P0.05)。由此可见,饲粮中长期添加牛磺酸可以调控种公猪血清激素含量,增加精浆抗氧化能力,进而增强种公猪性欲,提高种公猪精液品质,本试验条件下牛磺酸适宜添加水平为6 g/kg。  相似文献

本试验旨在研究不同精液品质特征长白公猪血清和精浆中元素含量的差异,分析血清和精浆中元素含量对精液品质的影响。基于107头长白公猪的1402次精液品质记录,将公猪群按照精液可利用率划分为3组:低利用率组(利用率<60%,n=21)、中等利用率组(60%≤利用率≤80%,n=27)和高利用率组(利用率>80%,n=59)。采集每头长白公猪的血清和精浆样品,采用电感耦合等离子体质谱法检测血清和精浆中营养元素和毒性元素含量。结果表明:1)低利用率组长白公猪的有效精子数和精子活力显著低于中等利用率和高利用率组(P<0.01),精子畸形率显著高于中等利用率和高利用率组(P<0.01)。2)不同利用率组间长白公猪的血清和精浆元素含量无显著差异(P>0.05);但在血清和精浆元素含量与精液品质参数相关性分析中发现,精浆铅元素含量与精子活力呈显著负相关(P<0.05),与精子畸形率呈显著正相关(P<0.05)。3)对精浆铅含量分组进一步分析发现,精浆铅含量为0μg/L时长白公猪的精子畸形率显著低于精浆铅含量>10.0μg/L时(P<0.05),精子畸形率降低约6.11%。总的来说,长白公猪精浆中毒性元素铅的存在会通过损害精子活力和形态,影响公猪精液品质。  相似文献

Optimal sperm cryopreservation is a prerequisite for the sustainable commercial application of frozen-thawed boar semen for AI. Three experiments were performed to identify factors influencing variability of postthaw sperm survival among 464 boar ejaculates. Sperm-rich ejaculate fractions were cryopre-served using a standard freezing-thawing procedure for 0.5-mL plastic straws and computer-controlled freezing equipment. Postthaw sperm motility (assessed with a computer-assisted semen analysis system) and viability (simultaneously probed by flow cytometry analysis after triple-fluorescent stain), evaluated 30 and 150 min postthaw, were used to estimate the success of cryopreservation. In the first experiment, 168 unselected ejaculates (1 ejaculate/boar), from boars of 6 breeds with a wide age range (8 to 48 mo), were cryopreserved over a 12-mo period to evaluate the predictive value of boar (breed and age), semen collection, transport variables (season of ejaculate collection, interval between collections, and ejaculate temperature exposure), initial semen traits, and sperm quality before freezing on sperm survival after freezing-thawing. In Exp. 2, 4 ejaculates from each of 29 boars, preselected according to their initial semen traits and sperm quality before freezing, were collected and frozen over a 6-mo period to evaluate the influence of interboar and intraboar ejaculate variability in the survival of sperm after cryopreservation. In Exp. 3, 12 ejaculates preselected as for Exp. 2, from each of 15 boars with known good sperm cryosurvival, were collected and frozen over a 12-mo period to estimate the sustainability of sperm cryosurvival between ejaculates over time. Boar and semen collection and transport variables were not predictive of sperm cryosurvival among ejaculates. Initial semen traits and sperm quality variables observed before freezing explained 23.2 and 10.9%, respectively, of the variation in postthaw sperm motility and viability. However, more that 70% of total variance observed in postthaw sperm quality variables among ejaculates was explained by boar. This indicates that boar is the most important (P < 0.001) factor explaining the variability among ejaculates in sperm cryosurvival, with most (14 of the 15 boars in Exp. 3) showing consistent (P > 0.05) sperm cryosurvival over time.  相似文献

The objective of this study was to evaluate the reproductive toxicity of gossypol and the possible counteracting effect of selenium supplementation in rams. Twenty-five mature crossbred (Barki × Rahamni) rams were randomly divided into 5 equal groups. The first group served as a control (no gossypol in diet). The second and third groups received diets containing free gossypol of 9 and 14 mg·kg(-1) BW·d(-1)(low level and high level of gossypol), respectively. The fourth and fifth groups received the same diets given to the second and third groups, respectively, with an oral administration of 1 mg selenium (as sodium selenite) daily for each animal. Results showed reductions in ejaculate volume (P = 0.028), percentage of dead sperm (P = 0.003), total functional sperm fraction (P < 0.001), and blood serum concentration of testosterone (P < 0.001) in the presence of both levels of gossypol inclusion. Furthermore, high level of gossypol reduced forward motility (P < 0.001) and semen initial fructose concentration (P = 0.002) and increased abnormal-head sperm (P = 0.003) and blood serum concentration of triiodothyronine (P = 0.006). Regardless of selenium supplementation, increasing level of free gossypol in diet resulted in a significant decrease in the percentage of forward motility (P = 0.037) and significant increases in the mean values of sperm concentration (P < 0.001), total sperm output (P = 0.002), percentage of total abnormal sperm (P = 0.058), and abnormal-head sperm (P = 0.016). On the other hand, regardless of levels of gossypol inclusion, selenium supplementation resulted in significant increases in libido (P < 0.001), mean values of ejaculate volume (P < 0.001), percentage of forward motility (P = 0.019), total sperm output (P < 0.001), total functional sperm fraction (P < 0.001), semen initial fructose concentration (P = 0.031), and blood serum concentrations of both testosterone and triiodothyronine (P < 0.001). In conclusion, free gossypol in diet induced adverse effects on semen quality in rams, but selenium supplementation successfully counteracts most of the hazardous effects of gossypol on semen characteristics.  相似文献
