Different conservation measures including domestication approaches are needed to preserve rare and extensively used plant species and to satisfy future market demands. Snow lotus (a common name used for a number of high altitude species in central Asia) species in Mongolia are important medicinal plants, mostly endangered at the regional level due to the extensive use for medicinal purposes and naturally limited distribution ranges. In order to develop effective conservation and cultivation approaches we studied the morphological and genetic diversity as well as the seed germination behavior of three populations of Saussurea involucrata in the Altay Mountains of Western Mongolia. Plant height and leaf length were measured on 41–53 randomly selected individuals in each population. Enhancement of seed germination was studied using either mechanical or chemical scarification with potassium nitrate and gibberellic acid. Genetic diversity within and among three populations of the species was analyzed using Amplified Fragment Length Polymorphisms. Plants in three populations differed in height and leaf length (P < 0.001). Plant size tended to be negatively correlated with habitat altitude. Seed germination rate was low and increased with seed soaking in gibberellic acid. However, the response of seeds to different treatments was rather population-specific. Most treatments did not significantly affect mean days to germination and the ratio of shoot to root dry weight of 2-weeks old seedlings. The level of genetic diversity in the studied populations was lower than the average value for other long-lived perennial herbs. Analysis of molecular variance revealed a high differentiation within populations (92 % of total variation) indicating sufficient gene flow among populations.  相似文献

Purpose

Eucalyptus forest plantations are normally devoid of understory vegetation that is often assumed to be associated with Eucalyptus allelopathic effects. The objective of this study was to determine the influence of high soil compaction and low soil moisture content on inhibition of the germination of understory seeds in Eucalyptus forests and thus would result in the scarcity of understory vegetation.

Materials and methods

The soil water content above the depth of 1 m of six major understory vegetation types was analyzed to determine if there was a correlation between soil water content and understory vegetation. The effects of soil treatment (soil-loosening vs. no soil-loosening) and water supply amount (2500, 2000, 1500, 1000, 500, 250, or 0 ml of water per day) on the seed germination rate of Stylosanthes sp. were explored using an artificial climate chamber experiment. Influence of soil source (five Eucalyptus forest soils vs. two non-Eucalyptus forest soils) and water supply (0, 50, 150, 200, or 400 ml of water every day) on the germination rate of five types of seed were assessed using a three-factor analysis of variance (ANOVA).

Results and discussion

Soil-loosening and water supply significantly (P?<?0.05) increased seed germination rate with the contribution rates of 26.14 and 42.93 %, respectively. Analysis of variance for three-factor experiments revealed a significant (P?<?0.05) effect of water supply and vegetation seed type on the germination rate of plant seeds. No significant effect of soil type was observed on germination rate, indicating that germination rate was not affected by soils in Eucalyptus forest.

Conclusions

The conservation of soil characteristics, such as water content and compaction, during the development of a Eucalyptus forest plantation may be an effective strategy for encouraging the growth of understory vegetation. This study highlights the importance that in dry areas or areas prone to long-term drought, it would be preferable to retain native vegetation.

The efficient storage and germination of seeds underpin the effective use of plants for livelihoods and sustainable development. A total of 204 wild species useful for local communities of the Tehuacán–Cuicatlán Valley were collected and stored in seed banks in country for long term conservation, and 66 % (i.e., 134) duplicated in the U.K., as an effective means of ex situ conservation. Of the 204 species, 147 (122 of which also duplicated in the U.K.) were previously listed as useful plants in the ethnofloristic inventory of the Valley. Based on literature surveys, we found that one of the major impediments to the use of stored seeds of wild species is the lack of knowledge of how to germinate the seed. In detailed studies, we found that seeds of 18 useful plant species from 10 different families germinated readily and could be propagated. In contrast, four species (Actinocheita filicina, Bursera submoniliformis, Karwinskia mollis and Lippia graveolens) produced dormant seeds and therefore further studies are needed before their use can be maximised in large scale propagation programmes in support of conservation and livelihoods. Overall, this large-scale study on useful wild plant species in Mexico confirms that conventional seed banking can effectively support sustainable development and livelihood programmes.  相似文献

The wild Brassica juncea (L.) Czern. et Coss., systematically belonging to the genus Brassica L. in the Cruciferae family, has become a noxious weed for cropping systems nowadays. Here, simple sequence repeat (SSR) markers were applied to investigate the genetic diversity, genetic structure and migration routes of the wild B. juncea populations in China. The results showed that a total of 90 alleles, with extensive allelic diversity, were observed at the 11 SSR loci of the wild B. juncea. The STRUCTURE analysis indicated that all the 25 wild populations were best described as belonging to two lineages. High Fst value (0.568), together with the partitioning, provided significant evidence for lineage differentiation in wild B. juncea. The high differentiation between the two lineages was, perhaps, due to limited gene flow (Nm?=?0.301) of this species. The analysis of molecular variance with distances among individuals corrected for the dominant nature of SSRs showed that most of the variation (59%) occurred within populations, and the remaining 41% variance was attributed to differences among populations. The distribution of diversity across China was significantly geographically dependent. NJ cluster analysis, based on genetic distance, grouped populations geographically, which further corroborated spatial pattern of two lineages. Based on these results, two routes were proposed for the migration of wild B. juncea in China after its origin from northwest China, heading east along the Yellow River or Yangtze River, respectively. We concluded that China, especially the northwest, is one of the primary origins of B. juncea.  相似文献

Jatropha spp. from Mexico includes high species richness and endemism; five species inhabit in the Tehuacan-Cuicatlan Biosphere Reserve (TCBR), and they are important resources as food, medicine and biofuel. The assessment of morphological and agronomic characteristics is essential to identify, use and maintain plant genetic resources. Given the lack of information on the morphological variability of Jatropha species in relation to environment, the objective was to analyze the influence of physiographic, climatic, and anthropogenic factors in the morphological variability of the species: Jatropha neopauciflora and J. rzedowskii, both not-endangered; J. oaxacana, special protection; J. ciliata and J. rufescens, both endangered in the Reserve. Twelve quantitative morphological variables were measured in 24 populations of these species; 14 environmental variables were registered, and the disturbance index in the sites was estimated. The information was analyzed with Principal Components Analysis (PCA), Cluster Analysis and Canonical Correspondence Analysis (CCA). PCA detected interspecific variation: J. ciliata and J. rufescens have longer and broader leaves and longer flowers, while the other three species have smaller leaves and flowers. J. oaxacana population has intermediate size of leaves, fruits and seeds, compared with those of J. neopauciflora and J. rzedowskii. CCA detected intra-specific variation among the populations of J. neopauciflora and J. rzedowskii, which were separated in two groups due to fruit and seed size. Axis 1 of CCA correlated positively with altitude and annual temperature range, and negatively with mean annual temperature; at the intra-specific level, both species are adapted to variations of temperature and altitude.  相似文献

The content of raffinose family oligosaccharides (RFOs) in pea seeds constrains their usage in feeding humans and animals. In our research, the content of soluble carbohydrates—particularly α-d-galactosides of sucrose (RFOs)—was analyzed. The materials were as follows: 248 accessions from the Polish Pisum Genebank including representatives of taxa (from species to convarietas), type lines for genes controlling seed characters, and breeding materials and cultivars. Accessions were divided into homogeneous groups considering content of total soluble carbohydrates, total RFOs and individual RFOs: raffinose, stachyose and verbascose. The highest content of total soluble carbohydrates and total RFOs were stated for accessions with wrinkled seeds (r and rb genes) and the lowest content for seeds of the wild species P. fulvum Sibth. et Sm. Accessions valuable for breeding (for further decreasing of anti-nutritional compounds) were found among domesticated taxa (P. sativum subsp. sativum convar. vulgare Alef. and speciosum (Dierb.) Alef.), breeding lines, and some wild taxa. Accessions with decreased content of a total RFOs and verbascose are particularly valuable. It was found that the content of total RFOs was the most highly, frequently, and positively correlated with a stachyose and verbascose content. However, in P. fulvum seeds with the lowest content of RFOs and verbascose, a high correlation between the content of total RFOs and stachyose was revealed. Contents of all oligosaccharides were substantially lower in lines with dominant alleles of main pea seed genes (R, A, and I). It can be assumed that wild, primitive peas were characterized by not-all-to-high (rather not high) content of oligosaccharides; then recessive mutations in mentioned genes resulted in an increased content of RFOs. A collection of yeasts isolated from soils of different types (soddy-podzolic, gray forest, and chernozems) in European Russia was analyzed. Soil yeasts were found to accelerate the development of seeds of common crops (wheat, barley, and rye). Cultures of yeasts from different taxa (representatives of the Rhodotorula, Cystofilobasidium, Sporobolomyces, Metschnikowia, Saccharomyces, Aureobasidium, Debaryomyces, Cryptococcus genera) are shown to stimulate the seed germination. The impact of soil yeasts (and preparations made on their basis) on seed germination is determined by their penetration to seed endosperm (endophytic microflora) followed by the isolation of amylolytic enzymes and physiologically active substances stimulating seed germination. The method of seed treatment is shown to affect greatly the direction of the preparations’ action. When soaking seeds in solutions with the preparation, microorganisms multiply on the seed surface, and when treating seeds using a semi-dry method, seeds themselves and endophytic microorganisms inside them begin to develop. This study can be considered one of the new directions of soil microbiology related to studying the participation of soil microorganisms in the formation of endophytic communities and their role in the germination of vascular plant seeds.  相似文献

We examined germination and seedling survival of Petrocoptis grandiflora and Petrocoptis viscosa (Caryophyllaceae), two narrow endemic species from the northwest Iberian Peninsula. The experiments were carried out with seeds of three of eight populations of P. grandiflora and one of the three sole populations of P. viscosa. Under natural conditions, both P. grandiflora and P. viscosa produce very large numbers of seeds. However, the specific microhabitat of these species (cracks and crevices of limestone rockfaces) has very marked effects on seed germination, and subsequent seedling survival. In the present study, we examined the effects of light, cold treatment and seed weight on germination capacity. In the case of P. grandiflora, we also compared the germination of seeds of the different populations. In addition, we assessed seedling survival over a 1-year period. Seeds maintained in darkness showed higher germination percentages than seeds maintained with a 12:12 h photoperiod. The application of a short period of cold prior to germination had no significant effect in either species. In the case of P. grandiflora, population of origin had a significant effect on germination percentage. Of all the factors considered, seed weight was the most important source of variability, both in seed germination and subsequent seedling survival. In both species in the natural habitat, less than 10% of germinated seeds survived by the end of the year. Seedling survival was affected by microhabitat. Seedlings in non-rockface soil microhabitats were more likely to suffer herbivory or interspecific competition than seedlings in crevices in the rockface.  相似文献

Cowpea cultivation in many countries around the Mediterranean Basin depends on a number of locally adapted populations conserved on-farm at a small scale, rather than on the use of modern varieties. Documentation, characterization and exploitation of traditional local populations could contribute to their conservation and utilization as sources of desirable characteristics. Therefore, a study was conducted to (a) characterize, (b) assess diversity and (c) classify 23 on-farm conserved local cowpea populations based on 32 agro-morphological traits. Investigations on diversity of characteristics related to seed yield, mineral and seed crude protein content as well as on correlations among them were carried out. A relatively high phenotypic diversity was observed. In particular, a high level of within population diversity was found (\( \bar{H}s \) = 0.34) exceeding that among populations’ diversity (Gst = 0.27). Principal component analysis classified the majority of local populations into two groups (mainly according to populations’ seed coat color and eye color), further divided into six subgroups regardless of the populations’ geographical origin. Significant differences were also observed among the populations studied for potassium and calcium, as well as for their seed crude protein content which ranged from 22.14 to 28.37 %. The results show appreciable levels of intra- and inter-phenotypic diversity in on-farm conserved cowpea populations, which indicates the existence of a valuable gene pool for future exploitation in breeding programs.  相似文献

Among 371 oat accessions from the world collection of the Vavilov Institute of Plant Genetic Resources (VIR) mostly represented by the landraces from Asia (Maritime Territory of Russian Federation, Mongolia, China, Japan, North Korea, India, Iran, Afghanistan, and Bhutan) ninety five were heterogeneous for resistance to the North Caucasus populations of the greenbug (Schizaphis graminum Rondani). Damage ratings of resistance in 47 accessions were high, low level of resistance was identified in 48 genotypes. Higher frequency of greenbug resistant accessions (60.5%) was found among landraces from Mongolia. Lines with high expression of the resistance were selected from seven landraces with wide range of damage ratings. North Caucasus populations of S. graminum from Krasnodar area and Dagestan significantly differ by the frequencies of virulence to host plant. Seven virulence phenotypes differentially interacting with oat genotypes were found in the Krasnodar population and 10 phenotypes were revealed in the Dagestan population. Results of the experiments with the greenbug test clones revealed that all the lines selected from the accessions VIR-2490, VIR-2539, VIR-4074, VIR-12213, VIR-12214 (Mongolia), VIR-6688 (India) and VIR-13624 (North Korea) are protected by diverse alleles of resistance genes which differ from the earlier identified gene Grb3. These lines are supposed to have aphid resistance genes nonidentical to the Grb1 and Grb2. The accessions VIR-6688, VIR-4074 and VIR-12214 possess high resistance to Krasnodar and Dagestan greenbug populations and the accession VIR-13624 is protected by the most effective gene (genes) of resistance to the both populations from North Caucasus.  相似文献

Secondary metabolites (SMs), such as alkaloids and raffinose oligosaccharides (RFOs), play important roles in plant physiology. Although alkaloid and RFO phenotypic variation has been reported for yellow lupin (Lupinus luteus L.), most evaluations have used a reduced number of accessions; thus, limiting the understanding of accumulation patterns and variation ranges. The main goal of this research was to assess alkaloid and RFO content in a diverse L. luteus sample to understand possible SM accumulation patterns across this legume species. Eighteen yellow lupin accessions were analyzed using high performance thin layer chromatography to provide insights on seed and leaf RFO and alkaloid phenotypic variation. Co-dominant markers (170) were used to examine genetic relationships among L. luteus accessions and possible accumulation patterns across closely related genotypes. Significant differences were observed for seed and leaf RFOs. Total seed RFO accumulation ranged from 79.738 to 131.079 mg g?1. Raffinose, stachyose, and verbascose were observed in all genotypes’ seeds, but at different RFO concentrations. Raffinose was the only RFO detected in leaves (2.793–0.4224 mg g?1). Total alkaloid accumulation ranged from 0.22 to 15.12 and 0.00 to 8.007 mg g?1 for seeds and leaves, respectively. Lupinine, sparteine, and gramine were observed in seeds and leaves, and showed a wide range of variation. Neighbor-Joining (NJ) analysis showed an apparent pattern of seed alkaloid accumulation, most likely due to domestication events. However, high RFO accumulating accessions were scattered across the NJ tree. Alkaloid and RFO significant phenotypic variation will not only help to understand the roles of these SMs in L. luteus, but also to uncover the genetic basis behind their accumulation.  相似文献

Germplasm collections of C. reticulatum and C. echinospermum are limited, while both species face threats from over-grazing and habitat change in their natural environments. Recently many new accessions of C. reticulatum and C. echinospermum were collected in east and south-east Anatolia (Turkey) but they have not yet been evaluated for agro-morphological traits. Therefore, the current study investigated agro-morphological traits of new germplasm sources of C. reticulatum and C. echinospermum and evaluated resistance to biotic and abiotic stresses for chickpea improvement. The most attractive agro-morphological traits were canopy width, number of stems and pods per plant and biological yield. The most productive accessions of C. reticulatum and C. echinospermum had 712 and 625 pods per plant, respectively. Two distinct seed, flower and leaf shapes were found in accessions of C. echinospermum. Path analyses indicated that biological yield and harvest index had the most direct influence on seed yield in both species. Factor analyses showed that high seed yield in C. reticulatum depended on high biological yield and number of pods per plant, whereas high seed yield in C. echinospermum depended on harvest index. It was concluded that most accessions of C. reticulatum and C. echinospermum were not only resistant to some biotic and abiotic stresses but also had hidden alleles that could produce transgressive segregation in crosses to cultivated material.  相似文献

With an annual production of about 10 Gt concrete is by far the most used solid man-made material. In order to adjust the workability of fresh concrete most often so-called superplasticisers, essentially water-soluble organic polymers, are utilised. As concrete is commonly in direct contact with soil or water, the leaching of organic and inorganic compounds and their environmental impact need to be assessed. The present study contributes to this purpose by investigating leachates from hardened cement pastes gained using the horizontal dynamic surface leaching test. Pastes were prepared with and without superplasticiser. The root growths as well as the germination behaviour of cress (Lepidium sativum) and white mustard (Sinapis alba) were examined with respect to phytotoxicity. Different proportions (100, 50, 25 and 12.5%) of the leachates were considered in the test scheme. The results indicate a positive effect of most of the leachates on seed germination and root growth, although statistical significance was only found in some cases. Both test species showed no or only slight harmful effects as relative root growth exceeded always 66% for S. alba and 74% for L. sativum. Seed germination was not negatively influenced by the leachates. Slight beneficial effects on both test species could be observed for leachates containing superplasticiser compared to samples in absence of superplasticiser.  相似文献

The influence of moisture and salinity of quartz sand on activation of the CO2 emission and water consumption by seeds of spring wheat (Triticum), winter rye (Secále cereále), triticale (Triticosecale), and spring barley (Hórdeum) treated by a stimulant was studied under conditions of a model experiment. It was found that the seeds treated with a stimulant get advantages in the development upon a decrease in the water content of quartz sand. However, in the case of sand salinization, stimulant application did not lead to acceleration of seed development. The deficiency of moisture in saline sand also resulted in the increase of stimulant application efficiency. It was supposed that optimum moisture contents of quartz sand for seed germination and for the development of endophytic microorganisms do not coincide: active development of endophytic microorganisms stimulating seed development began under a lower moisture in comparison with that of the active seed development. Thus, active development of endophytic microorganisms upon the low moisture content mitigates the deceleration of seed development in the case of water deficiency. Hence, application of the microbial stimulant for seeds should be efficient in the cases, when germination of seeds proceeds under conditions of possible moisture deficiency, i.e., in the regions, where early spring droughts are frequent.  相似文献

Germination of wild and managed populations of columnar cacti was compared to analyze differences associated to management intensity. The species studied: Polaskia chende, Escontria chiotilla, Myrtillocactus schenckii, Polaskia chichipe, and Stenocereus pruinosus are in a gradient from lower to higher management intensity, respectively. Within each species wild, silvicultural, and cultivated populations also represent a gradient from lower to higher management intensity. We hypothesized that seeds of plants under higher management intensity would require more water to germinate than others since managed environments are relatively wetter than unmanaged environments. Such pattern could be visualized within and among species according to management intensity. We tested germination of seeds from different populations of the different species to the water potentials: 0.0, ?0.2, ?0.4, ?0.6, ?0.8, and ?1.0 MPa. In all species studied seeds of managed populations had significantly more mass than seeds from wild populations. Each species had different susceptibility to water availability decrease, which could be related to particular adaptations to drought resistance. Stenocereus pruinosus and Polaskia chende were the most and least susceptible species, respectively. In control treatments of all species, seeds of managed populations had higher germination rate than seeds from wild populations, but when water availability decreased the pattern was the contrary. Our results suggest that artificial selection for larger fruits and heavier seeds in plants established in wetter environments have affected germination patterns. Although species have particular critical points of water availability for germination, differences in germination patterns are more pronounced according to management and artificial selection intensity.  相似文献

Spider plant (Gynandropsis gynandra) is a traditional leafy vegetable widespread in sub-Saharan Africa and tropical Asia that is also valued for its medicinal properties. Developing a breeding program for the species requires detailed knowledge of its phenology, floral morphology and pollination system. This study investigates the effects of floral morphology and pollination mechanisms on the reproductive success in G. gynandra. The experiments were conducted in two locations in Benin. A split-plot design was used with four randomized complete blocks. Three accessions were randomly assigned to the whole plots and five treatments including natural self-pollination, hand self-pollination, geitonogamy, open pollination and hand cross-pollination were randomized over the sub-plots. We observed that individual plant exhibited 70% of staminate (male) flowers and 30% of hermaphrodite flowers. G. gynandra was andromonoecious. Open pollination and hand cross-pollination led to higher fruit and seed set. Natural self-pollination and hand self-pollination resulted in lower fruit and seed production. G. gynandra is a self-compatible and predominantly out-crossing species. Cross-pollination resulted in a significant increase in fruit set. This study set the ground for the development of improved cultivars in G. gynandra.  相似文献

The germination ecology of the endangered biennial grassland forb Gentianella campestris was studied. The seeds had a primary dormancy which was overcome by cold-stratification. Seeds buried in the soil became germinable in early spring (February), and autumn-sown seeds germinated during a four-week period in late spring (May). Exhumed seeds germinated in both light and darkness and in situ germination occurred even at 15 cm depth in the soil. No persistent soil seed bank could be detected in populations of G. campestris, and very few experimentally buried seeds were viable after 21 months in the soil. Together, these results indicate that the species has a transient seed bank, in which case yearly seed production is important for maintaining populations and preventing local extinction.  相似文献

Pinus gerardiana Wall. is an important nut-producing pine having restricted distribution in the world. It has been observed that natural regeneration in the species is extremely poor or entirely lacking. The species has erratic and infrequent seed years, dormancy-related problems, and slow growth of seedling, which reduces its regeneration process in natural habitats. Therefore, we investigated the effect of growing medium and seed size on germination and seedling growth of the Pinus gerardiana. The seeds were categorized into two sizes, viz, small (<2.35 cm) and large (>2.35 cm) and five growing medium treatments were used, viz, Soil:Sand:FYM; Soil:Sand:Moss:FYM; Soil:Moss:Vermicompost; Soil:Sand:Vermicompost; and Soil:Sand:Moss:Vermicompost, for assessing their impact on germination and seedling growth. It was observed that, among different growing medium treatments, higher germination and seedling growth parameters were recorded, when Soil:Sand:Moss:Vermicompost was used for the study. Between different seed sizes, higher germination and seedling growth were recorded when seed size was large.  相似文献

A collection of seeds from five Acacia species was made in Egypt in 1856. Since then, the seeds have been stored at room temperature in different Swedish museums. Due to the extreme longevity within the seeds of Acacia and related species, germination tests were performed on the now 151-year old seed. Seeds of two of the five species tested germinated. The first, Acacia farnesiana (L.) Willd., had two seeds germinate, and Acacia melanoxylon R. Br. ex Ait. f. had one seed germinate. In addition, DNA was extracted from the aged seed and DNA preservation was analyzed. Four of the tested species displayed well preserved DNA, whereas DNA from Albizia lebbeck (L.) Benth. showed signs of degradation. The 151-year longevity of the Acacia seeds is among the longest of dry-stored seeds reported. Several independent studies now report on extreme survival capacity for Acacia and related genera suggesting that these genera are suitable for studies on the characteristics of seeds with long storage performance. The results also demonstrate that herbaria and seed collections stored in museums and institutional depositories can be alternate sources of plants genetic material and should be given conservation attention.  相似文献

Little is known regarding the effect of fragmentation and human agricultural management on the genetic variation and gene flow of Cucurbita pepo L., 1753 in moderate fragmented areas in central Guatemala. We hypothesize that the genetic variation of C. pepo is affected by forest fragmentation and by traditional agricultural management. Therefore, we aim to determine: (1) the genetic diversity and genetic structure of C. pepo in the Cloud Forest Corridor (CFC) (2) the extent of genetic admixture between commercial variety (CV) and traditional landraces (TL) of C. pepo, (3) the effect of habitat fragmentation in the population genetics of C. pepo with a landscape approach, and (4) the potential relationship between traditional management practices and genetic diversity of C. pepo in the CFC. We detected the existence of high level of genetic diversity (AR = 3.43; He = 0.50), inbreeding (Fis = 0.25) and moderate population structure of C. pepo in the CFC (Fst = 0.16). No correlation between landscape and genetics was found. Also, we found high genetic admixture between CV and TL. It seems that human practices, mainly related with seed exchange patterns, could affect genetic diversity of C. pepo in the CFC. C. pepo populations in the CFC are structured, with inbreeding, and show admixture with the CV, an aspect that could affect its genetic diversity. The agricultural management influenced the population genetics of C. pepo in the CFC, but the landscape did not. We suggest that special efforts should be made to preserve the diversity of this important indigenous food source for Guatemalan people as well as their management practices.  相似文献
