为研究戈戈健和本草劲方对猪肠上皮细胞因子表达的影响,本实验分别以200μg/m L的戈戈健和500μg/m L的本草劲方溶液处理猪肠上皮细胞,培养后收集细胞,提取总RNA,反转录成c DNA,通过荧光定量PCR测定各种相关细胞因子m RNA表达水平。结果显示,戈戈健能显著上调IFN-β(P0.05)、IL-6(P0.05)、IL-8(P0.01)、TNF-α(P0.05)和TGF-β(P0.05)m RNA表达水平;本草劲方能显著上调IFN-α(P0.05)、IFN-β(P0.01)和TGF-β(P0.05)m RNA表达水平,显著下调IL-6(P0.05)和TNF-α(P0.01)m RNA表达水平。表明这两种中药制剂均能通过诱导I型干扰素产生而发挥抗病毒作用;戈戈健能通过促进抗炎因子IL-6、IL-8和TNF-α分泌来增强细胞免疫,同时上调抑炎因子TGF-β表达来防止过度炎症;本草劲方能抑制促炎因子IL-6和TNF-α的产生并促进抑炎因子TGF-β的分泌,从而增强细胞的抗炎能力。本研究为戈戈健和本草劲方用于预防和治疗猪场病毒病提供了理论依据。  相似文献

紫锥菊(echinacea purpurea)是一种安全、高效的多功能药用植物,具有免疫调节、抗炎、抗氧化、杀菌、抗病毒等多种功能。本研究旨在研究紫锥菊提取物在体外对巨噬细胞免疫功能的影响。结果表明:紫锥菊提取物可以激活巨噬细胞,提高其吞噬能力;显著提高促炎细胞因子IL-1β、IL-6、IL-12/P40、TNF-α和抑炎细胞因子IL-10、TGF-β1以及抗病毒因子IFN-β、IFN-γ的基因表达和分泌,同时促进巨噬细胞合成NO进而提高其杀菌功能。以上结果提示,紫锥菊提取物可激活巨噬细胞的吞噬和杀菌功能,并提高其细胞因子的分泌发挥其免疫增强活性。  相似文献

为了研究副干酪乳杆菌类坚韧亚种(Lactobacillus paracasei subsp.Tolerans)对致病性大肠杆菌腹泻小鼠免疫机能及免疫相关激素水平的影响,试验将小鼠随机分为7组,分别为副干酪乳杆菌高、中、低剂量组,Nisin组,抗生素组,阴性对照组和正常对照组。采用碳廓血清法检测巨噬细胞的吞噬能力;ELISA法检测小鼠血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6 (interleukin-6,IL-6)、转化生长因子-β(transforming growth factor-β,TGF-β)、IL-17、IL-23和IL-1β以及内分泌激素糖皮质激素(glucocorticoid,GC)、三碘甲状腺原氨酸(triiodothyronine,T3)、甲状腺素(thyroxine,T4)、促肾上腺皮质激素(adrenocorticotropic hormone,ACTH)和生长激素(growth hormone,GH)水平。结果表明:与阴性对照组相比,灌胃高剂量副干酪乳杆菌可显著提高腹泻小鼠单核巨噬细胞的吞噬能力(P0.05);可极显著提高TGF-β水平(P0.01),极显著降低IL-6和IL-1β水平(P0.01),显著降低TNF-α、IL-17和IL-23水平(P0.05);可显著降低GC含量(P0.05),极显著降低ACTH含量(P0.01),显著提高T3、T4和GH含量(P0.05)。说明灌胃高剂量副干酪乳杆菌可以提高小鼠免疫力,调节内分泌激素水平,从而维持内环境的相对稳定,使机体处于健康状态。  相似文献

为了解促炎、抗炎细胞因子在小鼠LPS致炎模型中的动态变化,探讨中药方剂清温消热饮的抗炎治疗作用,采用酶联免疫法测定112例用LPS致小鼠急性炎症模型治疗过程中血清中促炎因子(TNF-α、IL-1β、PGE2)、抗炎因子(IL-10)含量。与LPS致小鼠急性炎症模型组比较:清温消热饮组在18、24 h血清中IL-1β含量明显降低,差异显著(P0.05);清温消热饮组在6、12、18 h血清中TNF-α含量明显降低,6、18 h差异显著(P0.05),12 h差异极显著(P0.01);清温消热饮组血清中的PGE2含量均低于模型组,在6、18、24、36 h时血清中的PGE2含量明显降低,差异极显著(P0.01);清温消热饮组在18、48 h血清中IL-10含量明显升高,差异极显著(P0.01)。同时,清温消热饮组与地塞米松组在各时间段血清中TNF-α、IL-1β、IL-10、PGE2含量无显著差异(P0.05)。结果表明,清温消热饮在急性炎症模型中能有效降低血清促炎因子TNF-α、IL-1β、PGE2含量水平,提升抗炎因子(IL-10)含量水平以表现出明显的抗炎作用。  相似文献

为探讨辣蓼黄酮正丁醇部分(FNB)对内毒素血症小鼠炎性细胞因子的影响,采用酶解-超声偶联法提取辣蓼全草中的总黄酮,结合大孔树脂纯化富集获得辣蓼正丁醇部分黄酮,通过不同剂量的FNB作用于脂多糖(LPS)刺激所诱导的小鼠内毒素血症模型,测定丙二醛(MDA)、MPO、T-AOC、T-SOD、GSHPX、GSH、LZM、ACP及炎性细胞因子肿瘤坏死因子α(TNF-α)、干扰素(IFN,IFN-α、IFN-γ)、白介素-2(IL-2)水平。结果表明,FNB能通过提升小鼠肝脏的抗氧化防御酶促体系的活性,减少LPS刺激所诱导的促炎因子TNF-α、IL-1β、IL-6、IL-8的释放,降低肺部TNF-α、IFN-α、IFN-γ以及IL-2mRNA的表达水平。说明一定浓度的FNB能减轻LPS诱导的内毒素血症对小鼠的损伤,改善生存率。  相似文献

为探讨辣椒碱的免疫调节作用。试验利用体外分离培养小鼠脾淋巴细胞和腹腔巨噬细胞,辣椒碱单药和辣椒碱与脂多糖(LPS)复合刺激脾淋巴细胞,MTT法检测辣椒碱和复合物对脾淋巴细胞增殖的影响;用辣椒碱单药刺激脾淋巴细胞,酶联免疫吸附(ELISA)法检测辣椒碱对淋巴细胞分泌干扰素-γ(IFN-γ)和白细胞介素-6(IL-6)的影响;用辣椒碱单药刺激腹腔巨噬细胞,MTT法检测辣椒碱腹腔巨噬细胞吞噬功能的影响;用ELISA法检测辣椒碱对巨噬细胞分泌肿瘤坏死因子-α(TNF-α)和IL-6的影响。结果表明,辣椒碱单药和辣椒碱与脂多糖复合均能促进脾淋巴细胞增殖,辣椒碱单药能促进淋巴细胞IFN-γ和IL-6的分泌。辣椒碱单药能增强巨噬细胞的吞噬功能,促进巨噬细胞TNF-α和IL-6的分泌。辣椒碱具有免疫调节作用。  相似文献

丝氨酸蛋白酶抑制剂在寄生虫入侵宿主的过程中发挥着关键作用,能够参与到入侵宿主、免疫逃避、炎症反应、凝血系统和细胞迁徙等过程。为了探讨两种丝氨酸蛋白酶抑制剂(Ts Ad SPI和Ts Ka SPI)对巨噬细胞所分泌炎性细胞因子的调节作用,应用实时荧光定量PCR技术对两种SPIs分别处理的小鼠巨噬细胞进行TNF-α、IL-1β、IL-6、IL-12、IL-10、TGF-βmRNA表达水平的检测。结果显示,Ts Ad SPI和Ts Ka SPI均可不同程度的抑制LPS活化的小鼠巨噬细胞促炎性细胞因子TNF-α、IL-1β、IL-6和IL-12 mRNA的表达水平。并且两种SPI均可不同程度的促进巨噬细胞抗炎性细胞因子IL-10和TGF-β的表达。表明这两种旋毛虫SPI可通过调节宿主巨噬细胞影响入侵时宿主的免疫应答。  相似文献

本文探讨了捻转血矛线虫的谷胱甘肽过氧化物酶(HC29)体外对山羊外周血单核细胞(PBMCs)功能的影响。颈静脉采取无捻转血矛线虫感染的山羊血液,分离单核细胞后加入终浓度为5μg/mL的HC29重组蛋白进行体外培养,通过免疫荧光抗体技术观察重组蛋白与单核细胞的结合情况;HC29刺激山羊外周血单核细胞,用qPCR技术测定各试验组单核细胞中IL-2、IL-4、IL-10、IL-17、IFN-γ和TGF-βmRNA的转录水平;重组蛋白刺激单核细胞,采用CCK-8法测定细胞的增值情况;将重组蛋白与巨噬细胞共培养,测定巨噬细胞吞噬和分泌NO的情况。结果表明,重组HC29能够与外周血单核细胞结合并刺激细胞因子IL-10和IFN-γ的表达水平显著提高(P<0.01),能够引起单核细胞增值(P<0.01),巨噬细胞吞噬作用增强(P<0.01)和NO分泌增加(P<0.01)。结果表明HC29是捻转血矛线虫的一个重要抗原,主要诱导Th1类免疫反应。  相似文献

探讨了经分子修饰后的人参皂苷-Rh2(衍生物)对小鼠脾T淋巴细胞分泌IL-2与IFNγ-、腹腔巨噬细胞分泌TNF-α与NO水平的影响。应用双抗体夹心ELISA法测定3种细胞因子的含量,硝酸还原酶法测定NO的含量。结果表明,衍生物的3个浓度(100,200和300μg/mL)均抑制了IL-2(P<0.01)和NO(P>0.05)的分泌,明显促进了IFN-γ和TNF-α的分泌(P<0.01)。提示衍生物可能是通过细胞因子途径来调节机体的免疫功能。  相似文献

为明确PYY对巨噬细胞炎性细胞因子分泌的调节作用,本试验分离培养健康小鼠腹腔巨噬细胞,不同浓度PYY预处理后,以LPS刺激。ELISA方法检测细胞培养上清中TNF-α、IL-6含量,半定量PCR方法检测细胞中TNF-α、IL-6mRNA表达变化。结果显示:高浓度的PYY1-36(10-910-7 mol/L)和PYY3-36(10-810-7 mol/L)和PYY3-36(10-810-7 mol/L)对LPS诱导小鼠腹腔巨噬细胞TNF-α分泌具有显著抑制作用(P<0.05);PYY1-36对LPS诱导小鼠腹腔巨噬细胞IL-6分泌无明显作用(P>0.05);不同浓度PYY3-36(10-1110-7 mol/L)对LPS诱导小鼠腹腔巨噬细胞TNF-α分泌具有显著抑制作用(P<0.05);PYY1-36对LPS诱导小鼠腹腔巨噬细胞IL-6分泌无明显作用(P>0.05);不同浓度PYY3-36(10-1110-7 mol/L)对LPS诱导小鼠腹腔巨噬细胞IL-6分泌均具有显著抑制作用(P<0.05)。表明PYY对LPS诱导小鼠腹腔巨噬细胞炎性细胞因子TNF-α及IL-6的分泌具有一定的抑制作用,提示PYY可能通过抑制炎性细胞因子的分泌而抑制炎症性疾病的发生发展。  相似文献

In late December 1985, 1485 institutional veterinarians and small and mixed animal clinics across Canada were sent a questionnaire in order to assess the status of Dirofilaria immitis in Canada in 1985 and 44% of them responded. Veterinarians reported that 137,300 dogs were blood-tested to check for microfilariae and 1210 dogs were found with heartworm. Another 36 dogs were amicrofilaremic but diagnosed with heartworm disease to give the total number diagnosed in 1985 as 1247 (0.91%).

Heartworm was reported from all provinces except Prince Edward Island, Newfoundland and Saskatchewan but most (1126) of the cases were in Ontario. Southwestern Ontario continued to be the primary focus of the infection in Canada. From Quebec, 91 cases were reported mostly from and around Montreal. From Manitoba, 19 cases were reported from Winnipeg and surrounding areas. Heartworm was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 28% of the cases were observed with clinical signs of heartworm disease and 78% had a history of not having left Canada.

Veterinarians reported that 97,794 dogs were blood-tested to check for microfilariae and 1417 dogs (1.45% of those tested) were found with heartworm. Another 34 dogs were amicrofilaremic, but were diagnosed as having heartworm disease, to give the total number diagnosed in 1984 as 1451 (1.48%). Heartworm was reported from all provinces except Prince Edward Island and Newfoundland but most (1310) of the cases were in Ontario. In Quebec, 126 cases were reported mostly from west of Montreal.

Heartworm was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 27% of the cases were observed with clinical signs of heartworm disease and 72% had a history of not having left Canada. Southwestern Ontario continued to be the primary focus of the infection.

In late November 1991, 1883 clinics in Canada were sent a questionnaire to assess the status of Dirofilaria immitis in dogs in 1991 and there was a 60.0% response. There were 344,031 dogs tested for heart-worm (HW), 627 were found infected and the prevalence of HW infection was 0.18%. There were 417 dogs with HW in Ontario, 116 in Manitoba, 38 in Quebec, 53 in British Columbia, three in Alberta, and one in Nova Scotia. In British Columbia, all of the infected dogs but one were from the Okanagan valley which, as from 1991, is a new focus of infection in Canada. Most dogs with HW had not been on preventive medication in 1990, and the prevalence among dogs tested and unprotected was 0.59%. That prevalence was considerably higher in endemic areas. Companion dogs, over three years of age and maintained primarily outdoors in rural areas, were most frequently infected. One cat was diagnosed with D. immitis and 33 dogs had Dipetalonema reconditium.  相似文献

In late November 1989, 1732 clinics and institutional veterinarians were sent a questionnaire to assess the status of Dirofilaria immitis, and 51.7% responded. Of 247,716 dogs tested, 394 had D. immitis microfilariae and 51 were amicrofilaremic for a total of 445 cases and heartworm prevalence of 0.17%. Most (408) of these dogs had no preventive medication and the prevalence among dogs tested and unprotected was 1.01%. That prevalence was considerably higher in endemic areas. Thirty-seven dogs with heartworm had preventive medication. Heartworm was most frequent in companion dogs over three years of age maintained outdoors in rural areas. About 75% of the cases had never left Canada, 26% had clinical signs and 125 were not treated.

Heartworm was reported from British Columbia, Manitoba, Ontario, Quebec, Nova Scotia and Newfoundland, but 383 cases were in Ontario. South-western Ontario was the primary focus of infection. There were 33 cases in Quebec and 24 in Manitoba, mainly found in and around Metropolitan Montreal and Winnipeg respectively.

Veterinarians reported that 150,989 dogs were blood-tested for microfilariae and 869 dogs were found with heartworm. Another 65 dogs were amicrofilaremic but diagnosed with heartworm disease and one was found with heartworm at necropsy to give the total number diagnosed in 1986 as 935 (0.62%).

Heartworm was reported from Manitoba, New Brunswick, Ontario and Quebec, but most (810) of the cases were from Ontario. South-western Ontario continued to be the primary focus of the infection in Canada. There were 103 cases reported from Quebec, mostly from and around Montreal, and 21 cases from Manitoba, from Winnipeg and surrounding areas. Heartworm was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 33% of the cases were observed with clinical signs of heartworm disease and 81% had a history of not having left Canada.

In late December 1983, 2 800 veterinarians across Canada were sent a questionnaire in order to assess the status of heartworm disease in Canada in 1983 and 26% of them responded. Veterinarians reported that 59 504 dogs were blood-tested to check for microfilariae and 771 dogs (1.30% of those tested) were found with Dirofilaria immitis. Heartworm disease was diagnosed in all provinces except New Brunswick and Newfoundland but most (733) of the cases were in Ontario.

Heartworm disease was found most frequently in companion dogs over three years of age maintained mainly outdoors in rural areas. About 31% of the cases were observed with clinical signs of heartworm disease and 64% had a history of not having left Canada. Southwestern Ontario continues to be the focus of the infection and most of the dogs there had not left the province previously.
