Fifteen worm-free lambs (two-and-a-half to three months or four to four-and-a-half months old) were infected with 3500 or 4000 Ostertagia ostertagi larvae on five days each week for six weeks, and their performance compared to that of controls. Eleven lambs were killed after eight weeks and four were challenged with O circumcincta to determine whether any cross resistance had developed. A feature of the O ostertagi infection was the considerable variation in response. Overall liveweight gain was lowered by 24 per cent in the two-and-a-half to three-month-old infected lambs, mainly due to reductions of 27 to 40 per cent in four of the seven lambs. There was no consistent effect in the older lambs. The worm populations consisted mainly of early fourth stage larvae and developing worms, but a small percentage reached sexual maturity and these produced a low faecal egg count (1 to 63 eggs per gram). Numerous intraluminal refractive crystals were present in the gut of both adult worms and developing stages, possibly reflecting degenerative changes. Hypertrophy of the abomasal mucosa with patchy loss of differentiation was a feature of the infection, and in four lambs serum pepsinogen concentrations were elevated. Exposure to O ostertagi did confer some protection against challenge with O circumcincta in that worm counts were reduced to about 60 per cent of those in controls, although no increase was observed in the numbers of arrested larvae. The successful passage of O ostertagi through young lambs could be important in mixed or alternate grazing systems by providing a reservoir of infection for the alternate host.  相似文献

In an experiment lasting 4 years, changes in the Teladorsagia circumcincta and Trichostrongylus colubriformis populations were compared in lambs and adult sheep with differing resistance statuses. Two flocks of 30 rams (resistant R and susceptible S) grazed separate pastures and 8 rams were slaughtered in the middle and at the end of each grazing season. Five groups of tracer lambs were added each year to estimate the pasture infectivity and were killed for worm counts. The availability of animals with differing resistance statuses (rams and tracer lambs) and differing levels of infection made it possible to investigate the number, size and fecundity of worms of these two species. The inflammatory response was measured in the rams by counting the globule leukocytes, mast cells and eosinophils in the fundic, pyloric and intestinal mucosa. In the tracer lambs, the daily egg production by the female worms of both species was negatively correlated with the worm burden. Worm length accounted for 60 and 70% of the variation in the number of eggs in utero for T. circumcincta and T. colubriformis respectively. Worm length was closely associated with the resistance status of the host; there were greater differences between lambs, and S and R rams for T. colubriformis. T. circumcincta worm lengths were not affected by the worm number. Globule leukocyte counts were related to the worm burdens, and mast cell counts to worm length in the R and S rams. The number, size and fecundity of the worms may well be regulated by similar mechanisms in both species, but T. colubriformis seemed to be more intensively regulated than T. circumcincta. This finding could be useful in devising more effective methods of parasite control.  相似文献

The dynamics of Trichostrongylus axei and intestinal Trichostrongylus spp. were studied in grazing lambs over a 2-year period. Monthly comparisons were made between worm counts of experimental lambs continuously exposed to infection from 3 to 7 months of age, and tracer lambs which were allowed to graze on the experimental plots for successive 4-week periods.It was found that irrespective of the level of larval intake, experimental lambs were unable to regulate their worm burdens until at least 5 months of age. When larval intake was high, resistance to establishment of larvae apparently occurred at approximately 6 months of age followed by rejection and resistance to reinfection. In conditions of low larval intake, worms accumulated with no apparent control operating in lambs up to 7 months of age.These results suggest that a rapid turnover of worm populations does not occur in Trichostrongylus spp., and that in young lambs, worms accumulate until sufficient exposure to infection stimulates resistance to larval establishment and ultimately rejection of adults.  相似文献

Six-month-old lambs that had been naturally infected with predominantly Teladorsagia (Ostertagia) circumcincta were tested for plasma pepsinogen concentrations because pepsinogen concentrations may reflect the extent of damage to the abomasum. The distribution of pepsinogen concentrations among these lambs was positively skewed with most individuals having relatively low concentrations. Pepsinogen concentrations were more strongly associated with variation in the mean length of the adult female worms than with variation in the number of nematodes present. Previous trials have suggested that genetic variation in the growth of lambs is strongly influenced by genetic variation in worm length. Together these results imply that variation among lambs in the pathogenic effects of T. circumcincta depends upon the mean size of the worms as well as the number of worms present.  相似文献

Four worm-free lambs were grazed from May to October 1972 on a paddock carrying a low residual infection of O. circumcincta larvae whilst another group grazed a similar paddock carrying a substantial residual larval infection. On the first paddock the lambs acquired low worm burdens of mature worms passing large numbers of eggs, but in spite of this, pasture larval infection remained low. On the second paddock the lambs acquired high worm burdens, a large proportion of which were immature. On this paddock a rise in pasture larval count was observed and this reached a peak in July and August. In spite of the differences in worm burden of the two groups there was no significant difference in weight gain. The mechanism by which the differences in worm burdens may have developed as well as their implications in worm control are discussed.  相似文献

Infective larvae of Ostertagia circumcincta were radiolabelled with 75selenium by a method which did not affect their viability. Three groups of five-month-old lambs were infected daily with 1000 unlabelled infective larvae for four, eight and 12 weeks, respectively. After each period one of these groups and a group of worm-free controls were challenged with three consecutive daily doses of 1000 radiolabelled third stage larvae. The lambs were killed 13 days after the first dose of challenge larvae and their worm burdens examined. The first indication of immunity was retardation of developing worms observed at four weeks. Resistance to the establishment of incoming worms developed between four and eight weeks and a brief period of population turnover probably took place at this time. Simultaneously a greater inhibition of worm development occurred resulting in an increase in the number of parasites recovered as early fourth stage larvae. By 12 weeks the animals were almost completely immune to incoming worms. The development of resistance to incoming worms correlated with a rise in serum antibody titre and an increase in the number of intraepithelial globule leucocytes in the gastric mucosa.  相似文献

Several necropsy techniques are available for estimating the abundance of gastro-intestinal nematodes in abomasum of ruminants. Standardization of techniques is needed to allow accurate comparisons between laboratories. Here we propose a standardized technique for estimating the abundance of worms. We intend to compare the worms' number estimations in lambs and ewes based on contents and washings, to determine the uniformity of worm counts in aliquots, and to estimate the total worm number from washings. The digesta (or "contents") and the washings of the abomasum are treated separately. The worms of each subsample are diluted with water and the total number of worms is estimated on a small volume (aliquots) of these subsamples. The use of aliquots assumes that the worms are uniformly distributed in the whole volume of each subsample. We first confirmed that the use of aliquots is appropriate in most cases. We then show that the use of the washings alone allows a faster and a suitable estimation of the total worm burden for all strongyle species of the abomasum in both ewes and lambs. The evaluation of our necropsy procedure is a first step to a standardized technique which should be improved by validation in other laboratories.  相似文献

Lambs premunized with a relatively less pathogenic (RLP) isolate of Haemonchus contortus were challenge exposed each day with 500 3rd-stage larvae (L3) of a normally pathogenic isolate of the same worm, starting on day 9 of the premunition-induced infection. In 6-month-old lambs, there was a fourfold decrease in egg production by the premunizing RLP isolate with challenge exposure compared with egg production in lambs given only the premunizing RLP isolate and not challenge exposed. Premunized lambs were able to carry larger worm burdens with less hemoglobin loss than were nonpremunized challenge-exposed control lambs. Although challenge exposure had affected parasitic egg production by the 9-day-old premunizing RLP worms, the same young infective isolates were able to survive and protect lambs against the additional parasitism.  相似文献

As part of a trial to test the profitability of finishing weaner lambs over winter and spring on the Highveld of Gauteng province, an investigation was carried out over 4 consecutive years as to whether or not worm control was necessary while the lambs were on irrigated, improved pastures. Pastures that had not been grazed by sheep or other livestock for at least 5 years were planted to annual ryegrass (Lolium multiflora) that was ploughed under and replanted annually. Weaner lambs were randomly divided into either 2 (A and B) or 3 (A, B and C) groups and finished from April/May to November/December on a separate pasture per group. While Group A lambs were dewormed before being placed on pasture, those in Groups B and C were not. Faecal egg counts were performed at irregular intervals, and when the lambs were sold at the conclusion of each year's trial, the gastrointestinal tracts of 4-5 lambs per trial group were processed for worm recovery. Haemonchus contortus was the dominant worm species, while Trichostrongylus colubriformis, Ostertagia circumcincta and Trichuris spp. were encountered sporadically. From the results obtained it seems unlikely that there is appreciable 'carry-over' of the various worm species from year to year on pastures that are ploughed and replanted annually; thus it is improbable that there will be a build-up of anthelmintic-resistant worms. While on pasture, no drenching was required for the Group A lambs (dewormed before placing on the pasture), nor in 2 of the 4 years in Groups B or B+C that were not drenched and were lightly infected at the start. In the remaining 2 years Groups B or B+C were drenched once only during the course of each trial. Thus, under the conditions as in this study, little worm control is necessary, provided faecal worm egg counts are done to gauge the levels of infection before the lambs are placed on pasture, and to guard against the possibility of an increase in worm burdens thereafter in some years. Also, because of the apparent lack of carry-over between years, anthelmintic treatment at the time of introduction of the lambs or during the period of finishing should be safe as regards progressive selection for anthelmintic resistance. It is also likely to be cost-effective, considering the low cost of anthelmintics in relation to the price of lambs, to counter the possibility of a loss in production if lambs were to harbour relatively heavy worm burdens when introduced.  相似文献

In order to determine whether the efficacy of moxidectin against Ostertagia circumcincta is enhanced by its persistency, therapeutic efficacy was compared at intervals after treatment and with that of ivermectin, a closely related but more transient endectocide. Groups of 7-month-old New Zealand Romney lambs were infected with a strain of O. circumcincta known to be resistant to moxidectin. At patency of the infections, groups of lambs were treated with either moxidectin or ivermectin at the manufacturer's recommended dosages, or left untreated. At 3, 6 and 10 days post-treatment, faecal egg count was measured and groups of lambs were slaughtered for estimation of adult worm burden. Drug-resistant worm burdens were significantly reduced in those animals treated with moxidectin but not in those treated with ivermectin. No effect of time of slaughter on worm burden was observed with either drug, demonstrating that the higher therapeutic efficacy of moxidectin against this parasite was not due to an increased period of drug exposure. Faecal egg counts in the moxidectin treated animals increased with time after treatment indicating a temporary suppression of egg output by surviving worms. The implications of these findings on selection for anthelmintic resistance are discussed.  相似文献

The addition of molybdenum (0.05 mmol kg-1 dry-matter) to the diet of lambs given a trickle infection of Haemonchus contortus larvae (500 third stage larvae d-1 over six weeks) reduced mean faecal egg counts (epg) from 3952 to 2312 +/- 402 by 32 days (P less than 0.02) and greatly reduced the mean number of worms recovered from the abomasum 14 days after infection ceased (907 compared with 4167: P less than 0.01). Infection reduced haemoglobin concentrations less in lambs given molybdenum although the difference was small relative to the reduction in worm burden. Lambs not given molybdenum had low intraepithelial mast cell counts in the abomasal mucosa and less abomasal hypertrophy than expected from abomasal parasitism. Molybdenum did not consistently reduce the copper status of the host or the parasite. Previous exposure to molybdenum greatly reduced protein but not proteinase activity in, or secreted by, adult worms cultured for eight hours. It is suggested that molybdenum either increased the inflammatory response which preceded worm rejection or that it indirectly enhanced that reaction by reducing the effectiveness of copper-dependent, anti-inflammatory enzymes in the gastrointestinal mucosa.  相似文献

Responses to a single or repeated infection with 7000 infective larvae of Haemonchus contortus were studied in an experiment using a total of 106 3-month-old lambs with AA, AB or BB haemoglobin (Hb) genotypes. Results were assessed by faecal egg counts, adult worm counts, haematocrit values, haemoglobin concentrations, total serum protein and serum antibody IgG1 and IgA ELISA titres. None of these parameters showed a strong relationship to the Hb type. The prevalence of low responder (greater than 500 worms) and of high responder (less than 50 worms) animals in groups AA, AB and BB Hb types was 3.8 and 34.6, 20.6 and 35.2, 28.1 and 43.7%, respectively, suggesting that the responsiveness to nematode infection is under the control of gene(s) not closely linked with those determining the Hb genotype. Worm counts of a primary infection are more subject to variation than those of a secondary infection. There is a strong relationship between adult worm counts and faecal egg counts taken close to the time of slaughter. In living animals low and high responder discrimination can be based on individual faecal egg counts around 50 days after a secondary infection. Haematocrit values proved to be of little value in the low and high responder selection. In this regard neither Hb concentration nor total serum protein values are of practical significance. In 3-month-old lambs primary infection induced partial immunity which could prevent the establishment of a part of the secondary infection, irrespective of the presence or absence of the primary worm population. The development of immunity was not associated with an increase of serum IgG1 and IgA antibody levels. Specific antibody production was not influenced by Hb types. Mean antibody levels of low responder lambs showed no difference from those of high responders. Thus, serum IgG1 and IgA levels are of no predictive value in identifying lambs which are genetically resistant to Haemonchus infection.  相似文献

A group of three ewes and six lambs running on worm free pasture exposed to infection arising from a stimulated postparturient rise of faecal egg count was compared with a similar group not exposed to a postparturient rise. The second group remained almost worm free and gained weight steadily whilst the first acquired a high worm burden and ceased to gain weight from the time they began to acquire worms. Six F(1) Dorper (D) x Red Maasai (R) rams were mated to both D and R ewes to produce backcross lambs. These six double backcross resource families are being analysed to identify quantitative trait loci that may be controlling resistance to gastro-intestinal (GI) nematode parasites, mainly Haemonchus contortus. After assessing the phenotypic performance of the lambs following exposure to natural infections, the surviving lambs were drenched and moved indoors for an artificial challenge with H. contortus along with straightbred D and R lambs. A total of 1317 lambs were included in the analyses consisting of 523 3/4D, 580 3/4R, 87 D and 127 R. The D lambs were significantly and consistently heavier than R lambs and 3/4D were significantly heavier than the 3/4R lambs. The difference between the backcrosses was about half of that between the straightbreds. Resistance was assessed in terms of faecal egg counts (FEC) and total worm counts (TWC) at necropsy while packed cell volume (PCV) was used to assess resilience to weekly oral doses of 2500 infective larvae of H. contortus. No significant breed differences were observed for log transformed FEC (LFEC). A significant breed difference in PCV was recorded. The backcrosses had the higher values and while no differences were observed between the straightbreds, 3/4D had significantly higher PCV than the 3/4R. Despite the absence of breed differences in FEC the R and the 3/4R had significantly fewer worms than the D and the 3/4D. The D had significantly longer worms than the R and the 3/4D had significantly longer worms than the 3/4R. Worms recovered from D had more eggs than those recovered from R. Similarly worms from 3/4D contained more eggs than those from 3/4R. Thus, on a breed basis the breed with more worms had longer worms. In contrast, when, in a small part of the experiment two doses of larvae were used to check for any breed by dose interactions, worms from the low dose (and hence fewer worms) animals were longer. We postulate that in fast growing hosts like the D, worms also have a better potential for growth and reproduction than in hosts that have less potential for growth. The overall correlation coefficient between PCV and LFEC was -0.67 while that between LFEC and LTWC was 0.72. From these results it is clear that the R do not respond to the artificial challenge the way they do to natural infection suggesting that phenotyping of R and R cross lambs for purposes of selecting those that are resistant or susceptible is best done under natural challenge.  相似文献

Parasitic infection is one of the leading economic constraints in small ruminant production. This problem is exacerbated as the resistance of nematode populations to chemical treatment (anthelmintics) becomes increasingly more common. Condensed tannin (CT) containing plants are being investigated as alternative solutions to address these problems. This investigation was conducted to evaluate the effect of the CT containing forage, Lespedeza cuneata (sericea lespedeza, SL) fed as hay, on Haemonchus contortus infection in sheep. Naturally and experimentally infected lambs were fed either SL or bermudagrass (BG, control) hay for 49 days. All lambs were fed BG hay for an additional 14 days. SL hay effectively reduced (67-98%) fecal egg count (FEC) during the time of feeding for both infection groups. FEC increased in both infection groups after SL feeding was stopped which indicated an effect on fecundity. SL hay feeding also reduced worm numbers, with more of an effect on reducing naturally infected worm burdens (67.2%) than on establishment of incoming larvae (26.1%). SL fed as hay may be more useful to remove existing worms than establishing worms. The decrease in FEC would have the benefit of reduced pasture contamination.  相似文献

The epidemiology of H. contortus was studied for 21 months in 80 set-stocked Merino ewes and their grazing in an endemic area in Kenya. Observations included faecal egg counts (weekly), worm burdens (monthly), haematological indices and clinical signs. The levels of infective larvae on the pasture were estimated using tracer sheep.The classical acute form of haemonchosis was seen with equal intensity in both ewes and lambs during periods of high rainfall and self-cure was confirmed as a flock phenomenon which occured simultaneously on one occasion in sheep of all ages.Not previously recognised were the uniformity of the faecal egg counts in both ewes and lambs over long periods when the worm burdens fluctuated greatly, the marked decrease in worm burdens without the occurence of classical self-cure during periods when the intake of infective larvae was low, and the inverse relationship between the individual worm-weight and the total worm population. It was also concluded that one of the most significant features of haemonchosis is not the acute syndrome so characteristically associated with the disease, but the fact that a moderate infection of a few hundred worms persisting over a period of several months will produce chronic anaemia and, ultimately, severe loss of bodily condition and deathes in ewes and lambs grazing on poor quality pasture.  相似文献

The development of gastro-intestinal helminth diversity was monitored in lambs grazing alone or grazing with heifers in the ratio one heifer to four lambs. Five successive cohorts of lambs were studied from January 1994 to May 1996. Each cohort of lambs grazed irrigated pastures of Pangola grass for 4 months (from weaning to 6 months of age). A total of 50 lambs was necropsied and their worms counted and identified at the end of each grazing period. Four heifers were also necropsied on one occasion. Special attention was dedicated to the identification of the most pathogenic worm, i.e. Haemonchus spp. Malate dehydrogenase polymorphism in H. contortus was studied in order to evaluate changes between cohorts and between grazing managements. The species diversity was estimated by Shannon diversity indices (main species or all species). It was higher in the mixed grazing group than in the lambs that grazed alone. Diversity increased in successive cohorts. This was due in part to the acquisition of Cooperia spp. of cattle origin. The increase in diversity in the mixed grazing lambs corresponded to the lower faecal egg excretion and better weight gains recorded previously in that group. There seemed to be no cross-transmission of H. similis found in heifers and H. contortus harboured by lambs. The latter species was not morphologically or genetically different in the lambs grazed alone or with heifers, indicating that the presence of cattle did not modify qualitatively the transmission of H. contortus.  相似文献

Challenge with an equal mix of drug-resistant and drug-susceptible larvae of Teladorsagia circumcincta resulted in infections in groups of lambs (n = 6) either untreated or given controlled-release capsules, containing either albendazole or ivermectin. Lambs treated with albendazole capsules contained similar numbers of adult worms at necropsy to the other groups but had no detectable faecal egg count. Animals treated with ivermectin capsules had similar worm burdens and faecal egg counts to the control group but the worms had significantly higher numbers of eggs in utero. These results provide evidence for suppression of egg production by both anthelmintic treatments. The observation that albendazole caused a significant reduction in the developmental success of parasite eggs also has implications for the use of faecal egg count as an indicator for pasture contamination with resistant parasites. In two further groups of lambs, either untreated or given albendazole capsules, treatment caused a significant reduction in egg count and adult worm burden of Trichostrongylus colubriformis. No significant effects were observed on in utero egg counts or egg viability and the apparent effect on the number of eggs produced in faeces per adult female was not significant (p = 0.077). There was, therefore, no evidence that albendazole controlled-release capsules caused suppression of egg output in this species.  相似文献

Tracer lambs were used to study the pasture contamination with infective stages of helminth parasites during one annual cycle in a subtropical climate. Post-mortem worm counts indicated that low infections with Haemonchus contortus occurred throughout the year except in June. However, twenty five or more H. contortus per lamb were recorded in January, April, May and August. Trichostrongylus colubriformis infection was detected throughout the year and 150 or more worms per lamb were recorded during January to May and in August. Anoplocephalids were recorded from the lambs throughout the year but had no seasonal pattern. Low infections with Oesophagostomum columbianum and Trichuris ovis were observed. The faecal egg counts from the permanent flock with whom the tracer lambs were grazed revealed heavy to mild worm burdens throughout the year. Coproculture indicated that H. contortus predominated from the second fortnight of May to December except in the second fortnight of July. Infection with T. colubriformis was more severe from January to the first fortnight of May and in the second fortnight of July. Negligible infections with O. columbianum, Bunostomum trigonocephalum, Gaigeria pachyscelis and Dictyocaulus filaria were also observed. Biohythergraphs prepared for H. contortus and T. colubriformis showed differences between observed and expected results. It is suggested that for realistic biohythergraphs related parameters in addition to rainfall and temperature should also be considered.  相似文献

The seasonal changes in nematode populations of a flock of sheep in the Montreal area were determined using serial fecal egg counts, fecal culture of larvae and necropsy worm counts. It was found that Ostertagia spp.,Nematodirus spp., Trichostrongylus agei, Trichostrongylus spp. and Chabertia ovina over-wintered on pasture and could initiate patent infections the following spring. The development of populations of H. contortus was typical of that seen with most of the other species and was characterized by the following series of events. In early winter when the study was started with stabled pregnant ewes, most of the populations were immature and the egg counts were low and remained so throughout the entire winter. However, in the spring, following lambing, large numbers of adult worms were seen with a consequent decrease in immatures and a sudden increase in egg counts. When the ewes and lambs were pastured together, the egg counts in ewes dropped consequent to “self-cure”, the “spring-rise” providing the major source of overwhelming infections for lambs with deaths by the end of July. As the season progressed larvae taken in by both ewes and lambs did not mature, and by early fall, most of the worm population consisted of immature forms. It appeared that H. contortus could not have more than two generations in ewes or lambs in a single grazing season.  相似文献
