为了研究新疆驴盲肠、腹结肠、背结肠、小结肠中食糜细菌组成的多样性,试验以3头成年、健康的新疆驴为研究对象,自由采食含90%稻草和10%精料的混合日粮,舍饲30 d后屠宰,分段取样备用,通过PCR-DGGE技术分析不同肠段食糜细菌的多样性。结果表明:新疆驴盲肠段消化道细菌的Shannon-Wiener指数和Simpson指数最大,与腹结肠、背结肠比较差异不显著(P0.05),但都显著高于小结肠(P0.05),相同消化道不同个体间的细菌条带组成具有很高的相似性。说明驴盲肠中食糜细菌的多样性最丰富,腹结肠和背结肠次之,小结肠最弱,相同肠道的细菌组成相似。  相似文献

生长育肥猪后肠纤维分解菌的数量及微生物发酵作用   总被引:3,自引:0,他引:3  
本试验采用严格厌氧微生物学方法 ,对生长肥育猪的盲肠、结肠和直肠内容物中总厌氧菌和纤维分解菌的数量、纤维素酶活性、氨态氮和VFA含量进行了测定。结果表明 ,盲肠和直肠内容物中纤维分解菌的数量显著高于结肠(P<0.01) ;盲肠内容物中羧甲基纤维素酶活性最高 ,其次是结肠 ,直肠最低(P<0.05) ;直肠和盲肠的氨态氮浓度显著高于结肠(P<0.05) ;盲肠、结肠和直肠的总VFA含量分别为82.79、154.47和143.80mmol/L ;乙酸的摩尔百分比为盲肠>结肠>直肠 ;丙酸的摩尔百分比无明显改变 ;丁酸的摩尔百分比为盲肠<结肠<直肠(P>0.05)。本试验证明 ,饲喂玉米—豆粕型日粮的生长肥育猪后肠中存在相当数量的纤维分解菌 ,盲肠、结肠和直肠内容物中纤维分解菌的数量分别占可培养微生物区系的8.6 %、3.1 %和22.1 %。后肠微生物发酵类型为乙酸发酵型。  相似文献

试验采用体外产气量法研究日粮中添加不同水平的棉籽油对绵羊瘤胃发酵功能的影响。棉籽油的添加水平分别设0(对照组)、1%、2%和3%。结果表明:日粮添加不同水平的棉籽油使人工瘤胃的最大产气量依次降低,但各处理组差异不显著(P>0.05)。与对照组相比,添加棉籽油并未显著改变发酵液pH值和NH3-N浓度。与对照组相比,添加棉籽油可以降低羧甲基纤维素酶(CMCase)和木聚糖酶(XYLase)活性,仅在发酵48h后,各处理组的CMCase活性达到显著水平(P<0.05),但各处理组差异不显著(P>0.05)。与对照组相比,随棉籽油添加水平的升高可以依次降低干物质(DM)和中性洗涤纤维(NDF)的降解率,发酵48h后,2%和3%组DM显著低于对照组(P<0.05),但两者间差异不显著(P>0.05)。与对照组相比,添加棉籽油并未显著影响总挥发酸浓度(TVFA)、乙酸摩尔百分比、丙酸摩尔百分比和乙丙酸比。  相似文献

为研究小黑麦(Triticale Wittmack)干草在德州生长驴上应用的可行性,本试验选取11月龄(150±10)kg的德州公驴25头,随机分为5组,按粗饲料日采食量一致设置小黑麦干草和羊草(Leymus chinensis)饲喂比例(其中小黑麦干草饲喂比例依次为:0%,25%,50%,75%,100%),测定了德州生长驴的生产性能和血液指标,并结合线性回归模型确定小黑麦干草的适宜饲喂量和饲喂比例。结果表明:小黑麦干草替代羊草对德州生长驴生产性能和血液抗氧化指标具有显著影响;75%组平均日增重显著高于0%,25%组(P<0.05),料重比显著低于0%,25%组(P<0.05);0%组超氧化物歧化酶(Superoxide dismutase, SOD)活性显著高于75%,100%组(P<0.05);25%,75%,100%组谷胱甘肽过氧化物酶(Glutathione peroxidase, GPX)活性显著高于0%组(P<0.05)。综上所述,小黑麦干草可以替代羊草在德州生长驴上应用,以干物质计算,11月龄(150±10)kg的德州生长驴小黑麦干草适宜饲喂量为...  相似文献

本试验旨在探讨饲粮蛋白质水平与纤维来源对生长猪盲肠液消化酶活性及水解能力的影响,为生长猪大肠消化液的模拟提供参考。试验1考察饲粮的蛋白质水平(饲粮1和饲粮2粗蛋白质含量分别为19.03%和13.96%)对生长猪盲肠液组成的影响。采用完全随机设计,将12头在盲肠安装瘘管的生长猪按体重随机分为2组,每组6个重复,每个重复1头猪,预试期5 d,正试期10 d。试验2在试验1结束并经过10 d恢复期后考察饲粮的纤维来源(饲粮3:玉米-大豆皮饲粮;饲粮4:玉米-小麦麸饲粮)对生长猪盲肠液组成的影响,试验设计同试验1。试验3在试验1和试验2获得猪盲肠食糜的基础上考察盲肠液的来源与底物对还原糖释放量的影响。采用4×8两因素完全随机设计,盲肠液来源设4个处理,分别为试验猪饲喂饲粮1、饲粮2、饲粮3和饲粮4后收集的盲肠液;底物为8个处理,包括饲粮1~4、2个玉米-豆粕饲粮(饲粮5和6)和饲喂饲粮5和6后获得的回肠冻干食糜。每个处理5个重复,每个重复1根消化管。结果表明:1)高蛋白质饲粮(饲粮1)与低蛋白质饲粮(饲粮2)相比显著降低猪盲肠食糜干物质含量(P≤0.05),饲粮蛋白质水平对盲肠液p H以及纤维素酶、木聚糖酶、葡聚糖酶、淀粉酶、中性蛋白酶、酸性蛋白酶活性无显著影响(P>0.05);2)饲粮纤维来源对猪盲肠食糜干物质含量、p H以及纤维素酶、木聚糖酶、葡聚糖酶、淀粉酶、中性蛋白酶、酸性蛋白酶活性均无显著影响(P>0.05);3)盲肠液来源和底物对还原糖释放量均有显著的影响(P<0.05),且两者对还原糖释放量有显著的互作效应(P<0.05),盲肠液中水解酶活性高,饲粮纤维含量低,则盲肠液体外消化后还原糖释放量多。盲肠液中木聚糖酶、葡聚糖酶、中性蛋白酶及淀粉酶活性影响了对饲粮的消化能力,而木聚糖酶与淀粉酶活性影响了对回肠末端食糜的消化能力。由此表明,饲粮营养水平虽然对生长猪盲肠液中6种水解酶活性影响未达到统计学显著性,但多种水解酶微小的差异叠加后引起了水解能力的差异。  相似文献

80只断奶新西兰兔分成5组,饲喂5种中性洗涤纤维(NDF)水平的日粮(分别为24%、27%、30%33%和36%),研究了日粮NDF水平对断奶至2月龄的肉兔生产性能、营养物质消化利用、免疫指标、盲肠纤维酶活性和盲肠内环境的影响。结果表明:日粮NDF水平对试兔日增重、饲料转化率影响显著(P<0.05),随NDF水平增加,日增重先增加后降低,料重比先降低后增加,30%NDF水平对日增重最高为31.52g/d,饲料转化率最低为3.23:1。随日粮NDF水平的增加,粗纤维、能量、粗蛋白和粗脂肪消化率显著或极显著降低(P<0.05,P<0.01),NDF消化率显著提高(P<0.05),氮沉积先升高后下降,以30%NDF组最高为1.24g/d。不同NDF间胸腺指数和脾脏指数差异不显著(P>0.05)。日粮NDF能极显著提高盲肠纤维素酶的活性(P<0.01),纤维素酶活性由13.79IU/g提高到58.68IU/g。NDF水平对断奶至2月龄试兔盲肠发育影响显著(P<0.05),盲肠占活重的比例随NDF水平的增加呈上升趋势。盲肠氨氮浓度呈下降趋势,24%、27%NDF组与36%NDF组差异显著(P<0.05),总VFA含量呈上升趋势,24%NDF和36%NDF组差异显著(P<0.05),VFA中乙酸比例上升,丙酸、丁酸比例下降。综合以上结果,断奶至2月龄肉兔NDF水平以30%~33%为宜。  相似文献

本试验旨在研究饲粮中添加不同比例白三叶对肉兔肠黏膜形态、肠道消化酶活性和盲肠发酵的影响。试验选取体重相近的(35±2)日龄新西兰兔200只,随机分成5组,每组4个重复,每个重复10只。对照组饲喂不添加白三叶的基础饲粮,试验组分别饲喂添加10%、20%、30%、40%白三叶的试验饲粮,试验期35 d。结果表明:1)白三叶能够显著影响空肠的绒毛高度、隐窝深度、黏膜厚度、绒毛高度/隐窝深度以及盲肠隐窝深度(P<0.05),以20%白三叶组的空肠绒毛长度、黏膜厚度和绒毛高度/隐窝深度最大,但对盲肠黏膜厚度的影响不显著(P>0.05)。2)白三叶能够显著影响淀粉酶和纤维素酶的活性(P<0.05),以40%白三叶组的淀粉酶活性和纤维素酶活性最高,但是对脂肪酶活性的影响不显著(P>0.05)。3)随着白三叶添加量的增加,盲肠pH、乙酸比例和乙酸/(丙酸+丁酸)显著上升(P<0.05),丙酸比例和丁酸比例则显著下降(P<0.05),白三叶对盲肠氨态氮、总挥发性脂肪酸、盲肠重和盲肠比的影响不显著(P>0.05)。由此可见,白三叶能够影响肉兔的肠黏膜形态、主要消化酶的活性以及盲肠发酵。  相似文献

选择28日龄断奶仔猪54头 ,随机分为9组 ,分别饲喂3种不同日粮处理 ,每种日粮处理组为3个重复。3种日粮分别为添加0.05 %异麦芽低聚糖组(IMO组)、添加110mg/kg阿散酸组(抗生素组)和对照组。各组基础日粮相同。分别于35和42日龄从各重复中选1头屠宰 ,测定盲肠、结肠内容物中挥发性脂肪酸(VFA)浓度、pH值及盲肠、结肠粘膜上皮绒毛高度和隐窝深度。结果表明 ,35日龄时 ,IMO组盲肠乙酸、丙酸 ,结肠乙酸、丙酸及总VFA浓度均显著高于对照组(P<0.05) ;42日龄时 ,IMO组盲肠乙酸、丁酸及总VFA浓度 ,结肠乙酸浓度均显著或极显著(P<0.01)高于对照组 ;结肠绒毛高度受处理影响显著 ,在两次测定中 ,IMO组结肠绒毛高度均显著大于(P<0.05)对照组。35日龄时 ,抗生素组结肠绒毛高度也显著高于对照组(P<0.05)。  相似文献

The effects of mild chronic dehydration on fermentation rate and mucosal surface area in the cecum, dorsa and ventral colon, and descending colon of the hindgut were investigated in South African donkeys (n = 11) in agricultural work. Dehydration representing a 6% drop in BW (n = 6) was associated with increased fermentation activity in the cecum (252 +/- 22.9 vs. 161 +/- 13.5 micromol/g of DM.h(-1), P < 0.01) and enhanced fluid retention in the ventral colon (0.81 +/- 0.026 vs. 0.73 +/- 0.034 mL/g gut, P < 0.05). Fermentation activity in the next segment of the hindgut, the ventral colon, of dehydrated donkeys was also greater numerically (92.5 +/- 22.60 vs. 77.9 +/- 10.40 micromol/g of DM.h(-1)), but this difference was not significant. Total mucosal and crypt surface area per unit volume of gut (Sv, microm2/microm3) was greater in dehydrated donkeys for the cecum (253 +/- 23.0 vs. 161 +/- 13.5, P < 0.01), the ventral colon (286 +/- 6.2 vs. 171 +/- 9.8, P < 0.01), the dorsal colon (276 +/- 18.2 vs. 256 +/- 11.0, P < 0.05), and the descending colon (260 +/- 20.3 vs. 191 +/- 15.2, P < 0.05). Enhanced fermentation activity and enhanced mucosal absorptive or secretory capacity within the hindgut during chronic dehydration was associated with an observed maintenance of appetite. These adaptations in the hindgut are valuable physiological attributes for working donkeys in semi-arid regions where they are frequently exposed to chronic dehydration.  相似文献

We studied in vivo fiber digestibility and fermentation parameters such as volatile fatty acid concentration and in vitro degradation in each segment of hindgut of horses fed timothy hay or silage made from the same sward. Six Thoroughbred horses were fed timothy hay or silage in equal amounts (1.6% of bodyweight per day, bodyweight mean 572 kg) every 3 h per day, then slaughtered. There were no differences between hay and silage in the concentration of the total volatile fatty acids, the apparent digestibility of dry matter, organic matter, and fiber, and in vitro neutral detergent fiber (NDF) disappearance rate in each segment. The total volatile fatty acid concentration and the apparent digestibility of dry and organic matter and fiber differed ( P  < 0.01) depending on the digestive segments. Both for hay and silage, the concentration of total volatile fatty acids in the digesta liquid phase largely increased ( P  < 0.01) from the cecum to the right ventral colon, and kept a constant value from the right ventral colon to the right dorsal colon, and then decreased from the right dorsal colon to the small colon. For dry and organic matter and fiber components, the apparent digestibility changed in the same manner from former to hinder segments of the hindgut. Regardless of diet, dry and organic matter and fiber components showed lower values in the cecum and the right ventral colon and increased ( P  < 0.01) largely from the right ventral colon to the left dorsal colon, then kept constant values in segments to the rear of left dorsal colon. There were no differences in in vitro NDF disappearance among cecum, ventral colon and dorsal colon.  相似文献

Six healthy horses were examined by using transabdominal ultrasonography, as described (1-3), to evaluate activity and size of the large colon and cecum at various locations. Using size and number of sacculations, activity patterns and contractile frequency; significant differences that would allow ultrasonographic identification of dorsal versus ventral colons, if they were displaced, were not found. The cecum had significantly greater activity than the colon, and a trend was seen towards smaller sacculations in the cecum than in the large colon.  相似文献

As part of digestibility and energy metabolism studies in intact and ileorectostomized growing pigs model experiments were carried out on sampling techniques and the influence of storage conditions on CO2 and volatile fatty acid production in chyme. Results on the formation of CO2 and VFA in chyme and faeces are given. They indicate that chyme from the small intestine can be subjected to bacterial and enzymatic changes outside the animals and therefore must be stored in a cool place. In the chyme of IRA animals with closed colon descendens (end-to-end anastomosis) significantly reduced VFA contents were found in comparison to animals with open colon descendens (end-to-side anastomosis).  相似文献

The steady-state response characteristics of a pulse oximeter were evaluated on intestinal segments of seven clinically normal halothane-anesthetized horses. Arterial oxygen tension greater than 200 mm of Hg, end tidal carbon dioxide from 30 to 35 mm of Hg, and systemic mean arterial pressure greater than 70 mm of Hg were maintained throughout the recording periods. Values for percentage of pulse oximeter oxygen saturation, pulsatile blood flow, and percentage of signal strength were recorded from jejunum, ileum, cecum, left ventral colon, left dorsal colon, and descending colon. Probe placement on intestinal segments was recorded as over or not over visible subserosal or transmural vessels. There was no significant difference between median values on the basis of vessel codes for pulse oximeter oxygen saturations, pulsatile flow, and signal strength. Median values recorded for pulse oximeter oxygen saturation were 93% from jejunum and ileum and 95% from cecum, left ventral colon, left dorsal colon, and descending colon; median values for pulsatile flow were 576 from jejunum, 560 from ileum, 560 from cecum, 574 from left ventral colon, 578 from left dorsal colon, and 560 from descending colon; median values for signal strength were 50% from jejunum, 67.5% from ileum, 60% from cecum, 75% from left ventral colon, 50% from left dorsal colon, and 52.5% from descending colon. Median values obtained from each anatomic location were not significantly different for pulsatile flow or signal strength. Six Thoroughbred horses were used to determine the mean retention time (MRT) of digesta in the different segments of the hindgut. The horses were fed timothy hay or silage in equal amounts (1.6% bodyweight [BW]/day) every 3 h/day. Hay or silage labeled with the rare earth elements cerium, dysprosium, neodymium, ytterbium, lanthanum, samarium and praseodymium, were fed to the horses 36, 30, 24, 18, 12, 6 and 3 h before slaughter, respectively. Just after slaughter, digesta samples in the different segments were collected and determined for the contents of the rare earth elements. Using these data, a method to calculate the MRT in the different segments was proposed. There were no significant differences between diets in the MRT of each segment. The averaged MRT of hay and silage in the cecum, right ventral colon, left ventral colon, left dorsal colon, right dorsal colon and small colon was 2.9, 3.1, 5.9, 1.0, 4.0, and 4.0 h, respectively. The dry matter weight of the digesta was related to the MRT in the right ventral colon (r = 0.94, P = 0.005), left ventral colon (r = 0.87, P = 0.03), left dorsal colon (r = 0.98, P < 0.001) and right dorsal colon (r = 0.95, P = 0.004), but it was not related to the MRT in the cecum and small colon.  相似文献

The interaction between the sequence of feeding of hay and concentrate and the hydrothermal processing of barley in alleviating concentrate effects on intake, and hindgut fermentation in horses was tested. Six Arabian mares (4–10 years of age, 410 ± 35 kg body weight) were used to evaluate the effects of feeding sequence (FS) and type of barley (TB) on intake, and faecal volatile fatty acids (VFA), activities of α‐amylase (AA: EC 3.2.1.1), carboxymethyl cellulase (CMCase: EC 3.2.1.4), microcrystalline cellulase (MCCase: EC 3.2.1.91) and general filter paper degrading activity (FPD). Mares were offered a ration of air‐dried alfalfa and concentrate (70:30 as‐fed) in four subsequent periods of 14 days including 8 days of adaptation and 6 days of sampling. In each period and each meal, mares received concentrate either 30 min after (HC) or 30 min before (CH) alfalfa hay. Barley was either milled or boiled in water. Rectal samples were grabbed directly from rectum once per period. Mares subjected to CH had higher dry matter intakes than mares under HC regime. The acetate:propionate ratio (A:P ratio) in rectal content was higher with CH than HC. The AA activity was higher under CH than under HC. Mares fed boiled barley had lower rectal concentrations of VFA and propionate and a higher A:P ratio than mares fed milled barley. Furthermore, the rectal content showed a higher MCCase activity but a lower AA activity when mares were fed boiled compared with milled barley. Interactions between FS and TB were observed with respect to CMCase activity, and concentrations of propionate and valerate. In conclusion, the present results suggest that both, feeding concentrate before hay and boiling the barley, might improve the hindgut environment in Arabian mares, and that the two measures were mostly additive and sometimes even synergistic.  相似文献

试验旨在研究泌乳驴的日粮蛋白水平对驴驹生长发育的影响。选取健康、体况良好的5岁龄、胎次(1.9±0.4)胎、体重(240±30) kg、泌乳日期(29±5) d的带驹德州三粉驴24头,分为3个处理组(H、M、L组),饲喂蛋白水平分别为13.18%、12.22%和11.34%的日粮。每个处理组4个重复,每个重复2头驴。正式试验期8周,分为试验前期(1~4周)和试验后期(5~8周)。同时,将24头驴驹跟随母驴相应分为3个处理组,驴驹的公、母比例一致,日龄(29±5) d与体重(48±8) kg相近。所有驴驹除摄取母乳外,补饲数量相同的精补料与粗饲料。试验表明,试验前期泌乳驴的日粮蛋白水平对驴驹日增重无显著的影响(P>0.10);试验后期,随着泌乳驴日粮蛋白水平的增加,驴驹日增重显著增加(P<0.05),饲料转化效率呈一定增加趋势,但对体尺无显著影响(P>0.10)。饲喂泌乳驴高蛋白水平日粮对驴驹血浆中天冬氨酸(Asp)、苏氨酸(Thr)、丝氨酸(Ser)和组氨酸(His)的含量有明显的促进作用。提高泌乳驴的日粮CP水平,对驴驹日增重具有一定促进作用,以12.22%和13.18%的效果较好。  相似文献

This study was conducted to compare physiological characteristics between Meishan and Yorkshire piglets in their early lives. Six healthy purebred Meishan sows and Yorkshire sows with close farrowing dates were used in this research. The piglets sucked their respective sow's milk for 14 days, then they were slaughtered to collect samples of blood, pancreas, contents of stomach, jejunum, cecum, colon as well as feces for analysis of blood biochemical parameters, digestive enzymes, and volatile fatty acid(VFA). The results showed that Yorkshire piglets had higher concentrations of high-density lipoprotein cholesterol(HDL-C) and total cholesterol(TC)(P 0.05). Gastric lipase activity was higher in Meishan piglets but Yorkshire piglets had higher lactase activity(P 0.05). The total VFA together with acetate and propionate in cecum and colon were higher in Meishan piglets than in Yorkshire piglets(P 0.05),but acetate in jejunum and ratio of acetate to propionate in colon were lower in Meishan piglets than in Yorkshire piglets(P 0.05). In conclusion, in early suckling period, significant differences exist in host metabolism and intestinal microbial metabolism between Meishan and Yorkshire piglets.  相似文献
