Background

Since transport evokes physiological adjustments that include endocrine responses, the objective of this study was to examine the responses of circulating β-endorphin, adrenocorticotrophic hormone (ACTH) and cortisol levels to transport stress in stallions.

Methods

Forty-two healthy Thoroughbred and crossbred stallions were studied before and after road transport over distances of 100, 200 and 300 km. Blood samples were collected from the jugular vein: first in a single box immediately before loading (pre-samples), then immediately after transport and unloading on arrival at the breeding stations (post-samples).

Results

An increase in circulating β-endorphin levels after transport of 100 km (P < 0.01), compared to basal values was observed. Circulating ACTH levels showed significant increases after transport of 100 km (P < 0.001) and 200 km (P < 0.001). Circulating cortisol levels showed significant increases after road transport over distances of 100, 200 and 300 km (P < 0.001). An effect of transport on β-endorphin, ACTH and cortisol variations was therefore evident for the different distances studied. No significant differences (P > 0.05) between horses of different ages and different breeds were observed for β-endorphin, ACTH and cortisol levels.

Conclusion

The results obtained for short term transportation of stallions showed a very strong reaction of the adrenocortical system. The lack of response of β-endorphin after transport of 200–300 km and of ACTH after transport of 300 km seems to suggest a soothing effect of negative feedback of ACTH and cortisol levels.  相似文献

Background

Artificial insemination is not as widely used in horses as in other domestic species, such as dairy cattle and pigs, partly because of the wide variation in sperm quality between stallion ejaculates and partly due to decreased fertility following the use of cooled transported spermatozoa. Furthermore, predictive tests for sperm fertilising ability are lacking. The objective of the present study was to assess sperm morphology and chromatin integrity in ejaculates obtained from 11 warmblood breeding stallions in Sweden, and to evaluate the relationship of these parameters to pregnancy rates to investigate the possibility of using these tests predictively.

Methods

Aliquots from fortyone ejaculates, obtained as part of the normal semen collection schedule at the Swedish National Stud, were used for morphological analysis by light microscopy, whereas thirtyseven were used for chromatin analysis (SCSA) by flow cytometry. The outcome of inseminations using these ejaculates was made available later in the same year.

Results

Ranges for the different parameters were as follows; normal morphology, 27–79.5%; DNA-fragmentation index (DFI), 4.8–19.0%; standard deviation of DNA fragmentation index (SD_DFI) 41.5–98.9, and mean of DNA fragmentation index (mean_DFI), 267.7–319.5. There was considerable variation among stallions, which was statistically significant for all these parameters except for mean_DFI (P < 0.001, P < 0.01, P < 0.001 and P < 0.2 respectively). There was a negative relationship between normal morphology and DFI (P < 0.05), between normal morphology and SD_DFI (P < 0.001), and between normal morphology and mean_DFI (P < 0.05). For specific defects, there was a direct relationship between the incidence of pear-shaped sperm heads and DFI (P < 0.05), and also nuclear pouches and DFI (P < 0.001), indicating that either morphological analysis or chromatin analysis was able to identify abnormalities in spermiogenesis that could compromise DNA-integrity. A positive relationship was found between normal morphology and pregnancy rate following insemination (r = 0.789; P < 0.01) and a negative relationship existed between DFI and pregnancy rate (r = -0.63; P < 0.05). Sperm motility, assessed subjectively, was not related to conception rate.

Conclusion

Either or both of the parameters, sperm morphology and sperm chromatin integrity, seem to be useful in predicting the fertilising ability of stallion ejaculates, particularly in determining cases of sub-fertility.  相似文献

Background

Variability among stallions in terms of semen cryopreservation quality renders it difficult to arrive at a standardized cryopreservation method. Different extenders and processing techniques (such us colloidal centrifugation) are used in order to optimize post-thaw sperm quality. Sperm chromatin integrity analysis is an effective tool for assessing such quality. The aim of the present study was to compare the effect of two single layer colloidal centrifugation protocols (prior to cryopreservation) in combination with three commercial freezing extenders on the post-thaw chromatin integrity of equine sperm samples at different post-thaw incubation (37°C) times (i.e., their DNA fragmentation dynamics).

Results

Post-thaw DNA fragmentation levels in semen samples subjected to either of the colloidal centrifugation protocols were significantly lower (p<0.05) immediately after thawing and after 4 h of incubation at 37°C compared to samples that underwent standard (control) centrifugation. The use of InraFreeze® extender was associated with significantly less DNA fragmentation than the use of Botu-Crio® extender at 6 h of incubation, and than the use of either Botu-Crio® or Gent® extender at 24 h of incubation (p<0.05).

Conclusions

These results suggest that single layer colloidal centrifugation performed with extended or raw semen prior to cryopreservation reduces DNA fragmentation during the first four hours after thawing. Further studies are needed to determine the influence of freezing extenders on equine sperm DNA fragmentation dynamics.  相似文献

Transition proteins (TNPs) are essential in chromatin condensation during spermiogenesis, and hence, they are the candidate genes for identifying sperm motility markers. Coding and in silico predicted promoter regions of these genes were investigated in crossbred and purebred cattle, and also, their mRNA quantification was done to explore its use as a diagnostic tool of infertility. PCR‐SSCP analysis revealed two band patterns in fragment III of TNP1 and fragment II of TNP2 gene. Sequence analysis revealed a deletion of “G” nucleotide in 3′UTR region of TNP1 and C>T SNP in intronic region of TNP2 gene. Least square analysis of variance did not reveal any significant influence of nucleotide deletion on any sperm motility parameters in both crossbred and purebred cattle. However, C>T SNP had a significant effect on initial progressive motility (p < 0.05) in purebred cattle and post‐thaw motility in overall cattle population. RT‐qPCR analysis did not reveal any significant variation in TNP1 and TNP2 gene expression among poorly motile and good quality spermatozoa of Vrindavani bulls.  相似文献

Although computer‐assisted systems for sperm morphometry and morphological analysis are important tools in the study of male fertility, their use in extensive systems in alpacas is limited by factors such as the expense of equipment and the high altitudes of the Andean region. The objectives of this study were to evaluate alpaca sperm head morphometry using a nonautomated digital method and determine the frequency of sperm abnormalities based on strict criteria for sperm morphology in fertile male alpacas. Ejaculates (n = 15) from seven alpacas were collected, and sperm smears stained with modified Papanicolaou were processed. For morphometric analysis, 3,000 sperm (200 cells/sample) images were captured at 400× magnification and Quick Photo MICRO 3.0 software was used for manual measurement of basic (sperm head length, width, perimeter and area) and derived variables (ellipticity, shape factor, elongation and regularity). For morphology assessment, smears were observed at 1000× magnification according to WHO and strict criteria. Average morphometric parameters were length 5.48 μm, width 2.99 μm, perimeter 13.62 μm, area 12.43 μm2, ellipticity 1.86, shape factor 1.20, elongation 0.29 and regularity 1.05. Significant between‐individual and within‐individual differences were found in morphometric parameters. Based on morphometric study, sperm heads were classified as elliptical or normal (49%), long (18%), short (2%), pyriform (12%), round (9%), large (6%) and small (4%). Morphological analysis found no additional sperm head defects in 49% of normal sperm obtained by morphometry, although a 4% incidence of neck/mid‐piece defects and a 16% incidence of principal‐piece defects were found. We conclude that sperm head morphometry assessment in fertile alpacas using a nonautomated digital method is feasible, and that defects in sperm heads constitute the main morphological alteration (>50% of the sperm population), based on WHO and strict criteria.  相似文献

Background

Proximal cytoplasmic droplets (PCDs), a remnant of germ cell cytoplasm, are common non-specific morphological defects in bovine semen. This study evaluated the effect of higher percentages of PCDs on the quality of frozen-thawed bovine semen, embryo production and early embryo development.

Methods

Three ejaculates from each of five (group 1: PCD ≤ 1%, control) and eight adult Bos indicus bulls (group 2: PCD ≥ 24%) were analysed. Semen samples were examined for: post-thaw motility, vigour of movement, concentration, sperm morphology, slow thermoresistance test (STT), membrane integrity, acrosome status, mitochondrial function using fluorescent probes association (FITC-PSA, PI and JC-1) and sperm chromatin integrity using acridine orange assay. Two bulls from group 2, with 28.5% and 48.5% PCD, respectively, and three bulls from the control group, each with 0% PCD, were selected for IVF (in vitro fertilisation).

Results

Semen analyses revealed a significant correlation (P < 0.01) between increased rates of PCD and sperm quality traits. Nevertheless, no differences were observed in sperm motility and vigour either before or after the STT or in the percentage of intact acrosomes (analysed by differential interference contrast microscopy (DIC) after STT), but membrane integrity, acrosome status (evaluated with FITC-PSA staining method after thawing) and mitochondrial function were reduced, when compared with group 1 (P < 0.05). The higher incidence of PCD was positively correlated to chromatin damage, especially after three hours of incubation at 37°C. IVF showed similar results for bull C2 (group 1, control) and bull P2 (group 2, group with higher PCDs).

Conclusion

Higher PCD levels influenced spermatozoa quality traits. IVF and embryo development data showed that cleavage, blastocyst formation and blastocyst hatching may have been influenced by the interaction of morphology traits and individual bull effects.  相似文献

Background

Unlike other domestic mammals, in which metaphase-II oocytes are ovulated, canine ovulation is characterized by the release of primary oocytes, which may take 12 to up to 36 hours. Further 60 hours are needed for maturation to secondary oocytes which then remain fertile for about 48 hours. Oestrus takes 7 to 10 days on average and may start as early as a week before ovulation. This together with the prolonged process of post-ovulatory oocyte maturation requires an according longevity of spermatozoa in the female genital tract in order to provide a population of fertile sperm when oocytes have matured to fertilizability. Therefore the distribution and viability of spermatozoa in the bitch genital tract was examined during post-ovulatory oocyte maturation.

Methods

Thirteen beagle bitches were inseminated on the day of sonographically verified ovulation with pooled semen of two beagle dogs containing one billion progressively motile spermatozoa. Ovariohysterectomy was performed two days later (group 1, n = 6) and four days later (group 2, n = 7). The oviduct and uterine horn of one side were flushed separately and the flushing’s were checked for the presence of gametes. The oviducts including the utero-tubal junction and the uterine horns, both the flushed and unflushed, were histologically examined for sperm distribution.

Results

The total number of spermatozoa recovered by flushing was low and evaluation of viability was limited. Prophase-I oocytes were collected from oviduct flushing in group 1, whereas unfertilized metaphase-II oocytes were detected in group 2. From day 2 to day 4 after ovulation a significant decrease in the percentage of glands containing sperm (P<0.05) and a marked reduction of the mean sperm number in uterine horn glands were observed. A concomitant diminution of spermatozoa was indicated in the utero-tubal junction accompanied by a slight increase in sperm numbers in the mid oviduct.

Conclusions

Oocyte maturation to metaphase-II stage is accompanied by a continuous sperm detachment and elimination in the uterine horns. Entrance of spermatozoa into the caudal oviduct seems to be steadily controlled by the utero-tubal junction thus providing a selected sperm population to be shifted towards the site of fertilization when oocyte maturation is completed.  相似文献

The aim of this study was to investigate the effect of gonadotrophin-releasing hormone (GnRH) immunisation on mature stallions that had been used for breeding. Four Standardbred stallions were used in the study: 3 experimental animals and 1 control animal. Semen was collected regularly, i.e. twice/week, during the 4 months prior to the experimental period. The stallions were immunised against GnRH with a GnRH-BSA conjugate. Equimune was used as the adjuvant. The stallions were immunised on 5 occasions, 4 at 2 week intervals, and the fifth 4 weeks after the fourth. Blood samples were taken once a week for analysis of GnRH antibody titre and every third week for testosterone and oestrone sulphate analyses. Semen was collected once a week, and libido and sexual behaviour were observed. Ejaculate volume, sperm concentration, total number of sperm in the ejaculate, sperm motility and sperm morphology were evaluated. Testicular size was measured once a week. At the end of the study, the stallions were castrated, and a histological examination of the testes performed. All immunised stallions produced antibodies against GnRH, and plasma testosterone concentration decreased. However, the effect of immunisation varied between stallions. In 2 of the stallions, high levels of antibodies were found, while in the third, the level was moderate. Four weeks after the first immunisation, a decrease in libido was observed. Two months after the first immunisation, marked changes in semen quality were observed in the 2 stallions with high antibody titres. Fourteen weeks after the first immunisation, the total number of sperm/ejaculate had decreased from >8.6 x 10(9) to <2.7 x 10(9), sperm motility from >59 to <10% and the frequency of morphological normal spermatozoa had decreased from >60 to <14%. The dominating abnormalities were abnormal head shapes, proximal cytoplasmic droplets and detached heads. In the third stallion, only slight changes in semen quality were found. No changes were observed in the control stallion. Decreases in testicular size were noted in all of the experimental stallions. Pronounced histological alterations in the testes were observed in 2 of the stallions. It is concluded that the vaccine was effective in stimulating production of GnRH antibodies and in suppressing testicular function and androgen secretion. However, there was an individual variation in the responses among the stallions and, further, libido was not totally suppressed.  相似文献

The South African Dorper is an important terminal meat sire breed in Africa that was recently imported into the United States. The objective of this study was to evaluate the Dorper as a terminal meat sire breed for U.S. production. Semen from purebred Dorper sires was used to artificially inseminate Columbia ewes to produce F1 crossbred lambs. Growth and carcass characteristics of F1 Dorper-Columbia lambs (n = 165), F1 Suffolk-Columbia (n = 89), and purebred Columbia lambs (n = 207) were compared based on subsets of the total number of animals. The F1 Dorper lambs grew significantly slower (313 g x d(-1)) and weighed less (29.8 kg) than F1 Suffolk- (357 g x d(-1), 33.5 kg) and Columbia-sired lambs (328 g x d(-1), 31 kg) at 77 d of age. However, at a weaning age of 118 d weaning weight and ADG did not differ (P > 0.20) among sire breed groups. Postweaning growth of F1 Dorper(239 g xd(-1)) wether lambs did not differ from that of purebred Columbia wethers (230 g x d(-1)) but was less than that of F1 Suffolk lambs (259 g x d(-1); P= 0.09). Feed efficiency did not differ among breed types. Breed types had similar dressing percentages (53%), shoulder fat depth (2.8 mm), body wall thickness (3 cm), leg conformation score (Choice), Yield grade (2.4), and Quality grade (Choice). Weight of wholesale shoulders and racks made up approximately 38% of the carcass weight in the Columbia and F1 Suffolk-Columbia type but only 33% in the F1 Dorper-Columbia lambs. However, the more expensive wholesale loins from F1 Dorpers were heavier (P < 0.01) than the other breed types. Total weights of wholesale legs were similar among F1 Dorpers and F1 Suffolks but were heavier than those for the purebred Columbia (P < 0.05). Percentages of total wholesale primal cuts were similar among breed types (P > 0.10). Chemical composition of the carcass did not differ significantly between breed types with a mean composition of 52% moisture, 30% lipid (ether extract), 17% protein, and 0.76% ash. Warner-Bratzler shear force values were less (P < 0.05) and sensory panel ratings for tenderness were significantly more favorable for lamb chops from Dorper sired lambs. Dorper rams can be used as terminal meat sires to produce lambs whose growth rate to 118 d of weaning age, postweaning ADG and feed efficiency, and carcass characteristics are similar to that of Suffolk crossbred lambs and purebred Columbia lambs but with a slight improvement in tenderness.  相似文献

Background

The aim of the present study was to investigate the efficacy of the Improvac on testosterone concentration in blood serum, sexual behavior and sperm quality in matured AI boars. A total of nine Danish Landrace AI boars were included in the analysis.

Methods

The trial period lasted for 15 weeks and was divided into four periods: Control period: three weeks before vaccination; Period I – four weeks after first vaccination; Period II – four weeks after second vaccination, Period III – four weeks after third vaccination. Blood and sperm samples were collected at weekly intervals. Freshly collected sperm samples were analyzed.

Results

Testosterone concentration correlated with libido (r = 0.531; p < 0.001), volume of ejaculate (r = 0.324; p < 0.001) and the percentage of morphologically normal spermatozoa (r = 0.207; p < 0.05). Testosterone concentration rised significantly (p < 0.05) in 5–6 week of trial, e. i. after the first dose of Improvac and after this peak the level of testosterone further progressively decreased (p < 0.05).

Conclusions

Results from this study indicate that active immunization of sexually matured boars against GnRH has negative impact on testosterone concentration, sexual behavior, volume of ejaculate and total number of normal spermatozoa in ejaculate.  相似文献

In order to study the beef traits and beef quality of Jiaxian Red bull with crossbred beef cattle, 5 purebred Jiaxian Red bull, 5 Red Angus cattle×Jiaxian Red bull F1 generation, 5 Simmental cattle×Jiaxian Red bull F1 generation and 5 Charolais cattle×Jiaxian Red bull F1 generation were chosen and assigned into groups A,B,C and D for continual finishing for 3 months of age under the same feeding and management conditions and slaughtered at the end of the trial. The results showed that there were no significant difference in body height, body length, chest girth and cannon circumference between Red Angus cattle×Jiaxian Red bull F1 generation and purebred Jiaxian Red bull, the same as in Charolais cattle×Jiaxian Red bull F1 generation (P>0.05). The weight of head, skin, fore hooves and tail, the skin thick, esophagus, trachea, liver, spleen, lung, kidney, and carcass meat percentage, loin eye area and the meat bone ratio in crossbred beef cattle groups were not significant differences compared with purebred Jiaxian Red bull (P>0.05), but its hind legs wide in carcass was significantly higher than that of purebred Jiaxian Red bull (P<0.05). The high-grade meat weight and high quality meat weight in Red Angus cattle×Jiaxian Red bull F1 generation were 70.22 and 105.22 kg, respectively, and was the highest in four groups,but the high-grade beef percentage among four groups did not show significant differences (P>0.05). The high quality meat percentage in Red Angus cattle×Jiaxian Red bull F1 reached 34.43%, was significantly higher than that of Jiaxian Red bull and Simmental cattle×Jiaxian Red bull F1 generation (P<0.05), and was higher than that of Charolais cattle×Jiaxian Red bull F1 generation (P>0.05). In terms of meat quality, the content of inosinic acid that related with the meat flavor in Red Angus cattle×Jiaxian Red bull F1 was the highest in four groups, while the content of unsaturated fatty acid and the flavor related amino acid, such as aspartic acid, glutamic acid, glycine and alanine, in Charolais cattle×Jiaxian Red bull F1 were the highest in four groups,and their values were 53.85% and 8.04%, respectively. To sum up, the results suggested that there was no significant difference in body development and beef traits using Red Angus cattle, Simmental cattle and Charolais cattle to improve Jiaxian Red bull, while Red Angus cattle crossbred with Jiaxian Red bull could moderately improve the weight of high-grade beef and high-quality beef, and Charolais cattle could moderately improve the beef quality of Jiaxian Red bull.  相似文献

To clarify the physiological changes of sperm morphology in active Thoroughbred stallions during the breeding season, we examined the dismount semen collected from the penile urethra immediately after service. The spermatozoa were analyzed for relationships between the morphology and the stallion’s age or the number of services. Seasonal variation was apparent in the rate of the sperm tail abnormalities, spermatozoa with cytoplasmic droplets, appearance of medusa cells, and sperm head length. Area and width of the sperm head correlated negatively with age (P<0.05). The rate of appearance of medusa cells and the length of the sperm head were positively related to the number of services (P<0.05), and the aspect ratio was negatively related (P<0.01).  相似文献

为了研究郏县红牛与不同品种肉牛杂交后代的肉用性能和肉品质情况,试验选取红安格斯牛、西门塔尔牛、夏洛莱牛与郏县红牛的杂交1代公牛各5头,纯种郏县红牛5头,分为试验A组(郏县红牛公牛)、B组(红郏F1代公牛(红安格斯牛×郏县红牛))、C组(西郏F1代公牛(西门塔尔牛×郏县红牛))和D组(夏郏F1代公牛(夏洛莱牛×郏县红牛))4个试验组,在同样的饲养管理条件下,持续育肥3个月后屠宰,利用屠宰测定和检验分析的方法对郏县红牛及其不同杂交品种的体尺发育、肉用性能和肉品质进行分析测定。结果表明:采用红安格斯牛、夏洛莱牛改良郏县红牛,在体高、体斜长、胸围和管围方面与郏县红牛纯种牛相比均无显著性差异(P>0.05);屠宰测定方面,杂交后代组的头重、皮厚、皮重、前二蹄重和尾重,屠体器官食道、气管、肝脏、脾脏、肺脏、肾脏的重量,以及胴体产肉率、眼肌面积、肉骨比与郏县红牛纯种牛相比均无显著性差异(P>0.05),但其胴体后腿宽显著高于郏县红牛纯种牛(P<0.05);红郏F1代公牛高档肉块重和优质肉块重最高,分别为70.22和105.22 kg,但高档牛肉率4组之间差异不显著(P>0.05),红郏F1代公牛优质牛肉率达到34.43%,显著高于郏县红牛和西郏F1代公牛(P<0.05),高于夏郏F1代公牛(P>0.05)。在肉品质方面,红郏F1代公牛与肉品风味有关的肌苷酸含量最高,而不饱和脂肪酸含量和与鲜味有关的氨基酸(天门冬氨酸、谷氨酸、甘氨酸和丙氨酸)含量均为夏郏F1代公牛含量最高,分别为53.85%和8.04%。结果提示,采用红安格斯牛、西门塔尔牛和夏洛莱牛改良郏县红牛,没有在体尺发育和肉用性能方面表现出显著优势,但采用红安格斯牛改良郏县红牛可提高郏县红牛肉用性能的潜力,可适度提高高档牛肉和优质牛肉肉块重量,而夏洛莱牛改良郏县红牛可适度提高牛肉品质。  相似文献

Background

Soundness is important for welfare and utility of the riding horse. Musculoskeletal disorders are the most common causes of interruption in training and of culling. Despite great importance, heritability of a majority of health traits in horses has previously not been estimated. The objective was to perform genetic analyses of medical and orthopaedic health traits in young riding horses, including estimates of heritability and genetic correlations between health traits, and to reveal possibilities for genetic evaluation of stallions for progeny health.

Results

The heritability of health traits was estimated using records from 8,238 Swedish warmblood riding horses examined as 4–5 year olds at the Riding Horse Quality Test in 1983–2005. The analyses were performed using multi-trait linear mixed animal models. The heritabilities of palpatory orthopaedic health (PALP), including effusion, swelling, heat, soreness and stiffness/atrophy, and hoof examination results (HOOF), of hoof shape and hoof wall quality, were 0.12 and 0.10, respectively. The genetic variation in these traits resulted in distinct health differences between progeny groups of stallions. The highest heritability among clinical signs of PALP was found for synovial effusions at 0.14. For systemic locations, joint related findings had the highest heritability; 0.13. The heritabilities of medical health and locomotion examination results were low, 0.02 and 0.04, respectively. A genetic improvement of health status has occurred over time but accounts only partly for the decrease in clinical findings of health during the studied period.

Conclusions

The genetic variation found in PALP and HOOF implies distinct differences between progeny groups. Thus, there are possibilities for improvement of these traits in the population through selection. The weak and non-significant correlation between PALP and HOOF suggests that both traits need to be selected for in practical breeding to improve both traits. Some genetic improvements over time have already been achieved, possibly due to regular stallion health inspections and an indirect selection for lifetime performance. For further improvements stallion breeding values for health may be introduced, based on RHQT examinations, complementary to present breeding values for performance.  相似文献

Background

One of the challenges faced by equine breeders is ensuring delivery of good quality semen doses for artificial insemination when the mare is due to ovulate. Single Layer Centrifugation (SLC) has been shown to select morphologically normal spermatozoa with intact chromatin and good progressive motility from the rest of the ejaculate, and to prolong the life of these selected spermatozoa in vitro. The objective of the present study was a proof of concept, to determine whether fertilizing ability was retained in SLC-selected spermatozoa during prolonged storage.

Findings

Sixteen mares were inseminated with SLC-selected sperm doses that had been cooled and stored at 6°C for 48 h, 72 h or 96 h. Embryos were identified in 11 mares by ultrasound examination 16–18 days after presumed ovulation.

Conclusion

SLC-selected stallion spermatozoa stored for up to 96 h are capable of fertilization.  相似文献

In the pig industry, purebred animals are raised in nucleus herds and selected to produce crossbred progeny to perform in commercial environments. Crossbred and purebred performances are different, correlated traits. All purebreds in a pen have their performance assessed together at the end of a performance test. However, only selected crossbreds are removed (based on visual inspection) and measured at different times creating many small contemporary groups (CGs). This may reduce estimated breeding value (EBV) prediction accuracies. Considering this sequential recording of crossbreds, the objective was to investigate the impact of different CG definitions on genetic parameters and EBV prediction accuracy for crossbred traits. Growth rate (GP) and ultrasound backfat (BFP) records were available for purebreds. Lifetime growth (GX) and backfat (BFX) were recorded on crossbreds. Different CGs were tested: CG_all included farm, sex, birth year, and birth week; CG_week added slaughter week; and CG_day used slaughter day instead of week. Data of 124,709 crossbreds were used. The purebred phenotypes (62,274 animals) included three generations of purebred ancestors of these crossbreds and their CG mates. Variance components for four-trait models with different CG definitions were estimated with average information restricted maximum likelihood. Purebred traits’ variance components remained stable across CG definitions and varied slightly for BFX. Additive genetic variances (and heritabilities) for GX fluctuated more: 812 ± 36 (0.28 ± 0.01), 257 ± 15 (0.17 ± 0.01), and 204 ± 13 (0.15 ± 0.01) for CG_all, CG_week, and CG_day, respectively. Age at slaughter (AAS) and hot carcass weight (HCW) adjusted for age were investigated as alternatives for GX. Both have potential for selection but lower heritabilities compared with GX: 0.21 ± 0.01 (0.18 ± 0.01), 0.16 ± 0.02 (0.16 + 0.01), and 0.10 ± 0.01 (0.14 ± 0.01) for AAS (HCW) using CG_all, CG_week, and CG_day, respectively. The predictive ability, linear regression (LR) accuracy, bias, and dispersion of crossbred traits in crossbreds favored CG_day, but correlations with unadjusted phenotypes favored CG_all. In purebreds, CG_all showed the best LR accuracy, while showing small relative differences in bias and dispersion. Different CG scenarios showed no relevant impact on BFX EBV. This study shows that different CG definitions may affect evaluation stability and animal ranking. Results suggest that ignoring slaughter dates in CG is more appropriate for estimating crossbred trait EBV for purebred animals.  相似文献

Genetic evaluations using purebred data alone and combined purebred and crossbred information were performed for lean meat percentage in a pig breeding scheme. One purebred (PB) model and 2 crossbred models (CCPS1 and CCPS2) were used in the analyses. Data were obtained from the Selección Batallé S.A. Company (Riudarenes, Spain) and spanned a period of 4 yr (2006 to 2009). The data corresponded to 3 nuclei of purebred populations, Landrace (LD), Duroc (DU), and Pietrain (PI); 1 multiplying farm with animals from a 2-way cross (TB1; DU × LD); and commercial farms with animals from a 3-way cross (TB2; TB1 × PI). Genetic parameters were similar across the models, with the exception of purebred PI. The DU and LD purebreds presented large heritabilities (0.5 to 0.6) for lean meat percentage, whereas the PI purebred showed a lower heritability (approximately 0.1) for the PB model and moderate heritability for the CCPS1 and CCPS2 models (0.2 to 0.3). The mean reliability of the predicted purebred breeding values was clearly increased when the CCPS1 and CCPS2 models were used. Moreover, a reranking of the animals with important changes in the selection decisions was observed in the PI purebred. In a simulation study, the CCPS1 model achieved a greater response to selection than the PB model for the PI purebred. On another hand, between the CCPS1 and CCPS2 models, CCPS1 was slightly superior in terms of predictive ability, exhibiting a greater robustness. These results illustrate the usefulness of using crossbred models to evaluate lean meat percentage in this pig breeding scheme.  相似文献

The standard procedure for assessing the breeding potential of a stallion includes the parameter total number of spermatozoa classified as morphologically normal. This study investigated sperm morphology of fresh semen in randomly chosen Estonian (E, n = 8) and Tori (T, n = 7) breed stallions with proven fertility. Two ejaculates were examined from each stallion. An aliquot from each ejaculate was fixed in 1 mL formol-saline immediately after collection and examined with phase-contrast microscope at a magnification 1000x for all types of morphological abnormalities. Furthermore smears were prepared and stained according to Williams (carbolfuchsin-eosin) for a more detailed examination of the sperm heads with light microscope at a magnification 1000x. Analysis of variance was applied to the data, and results are presented as LSmeans (+/- SE). One T stallion that had a disturbance in the spermatogenesis and one 22-year-old E stallion were not included in the analyses. The T stallions had on average 57.5 +/- 4.1% and the E-stallions 74.4 +/- 3.8% morphologically normal spermatozoa (p = 0.012). In 4 of 7 T stallions and 7 of 8 E stallions both ejaculates had > 50% morphologically normal spermatozoa. There was a significant difference between breeds in mean percentage of proximal droplets (17.3 +/- 2.7% and 2.9 +/- 2.5% for T and E stallions, respectively; p = 0.003).  相似文献

Cooling of equine semen obtained from some stallions results in lower seminal quality and viability when the seminal plasma (SP) is present. The objective of this study was to evaluate the effect of the removal of SP using a Sperm Filter on the viability of cooled stallion semen. For this purpose, 31 stallions were used. Their ejaculates were divided into three groups: CN, semen was diluted with an extender; FLT, SP was removed by filtration; and CT, SP was removed by centrifugation and cooled to 15°C for 24 hours. Sperm kinetics and plasma membrane integrity were evaluated immediately after collection (T0) and after 24 hours of refrigeration (T1). No difference (P > .05) was noted at T1 for total sperm motility (TM), progressive sperm motility, or plasma membrane integrity when semen samples from all the stallions were analyzed. However, when samples from stallions termed “bad coolers” were analyzed (TM = <30% at T1), a difference was observed in TM and progressive sperm motility for CN compared with FLT and CT at T1. Sperm recovery was greater when SP was removed using the filter (FLT) to that when the SP was removed by centrifugation (CN) (89% vs. 81%). Thus, we concluded that filtering with a Sperm Filter is an efficient and practical method for removal of SP from stallion ejaculates, with lower sperm loss than centrifugation. We also found that the presence of SP reduces the quality and viability of cooled semen from stallions whose semen is sensitive to the process of refrigeration.  相似文献

To investigate the influence of daylength on the seasonal reproductive cycle of stallions, 21 stallions were assigned to one of three treatments: control, ambient (natural) photoperiod; S-L, 8 h light and 16 h dark (8:16) for 20 wk beginning July 16, 1982 then 16:8 from December 2, 1982 until March 5, 1984; S-S, 8:16 from July 16, 1982 until March 1984. Temperature was not controlled and was similar for all groups. Total scrotal width (TSW) was measured every 4 wk throughout the experiment. During 10 periods, semen was collected and evaluated every other day for 3 wk and sexual behavior was assessed. The S-L stallions exposed to 16 h light in December had twice as much sperm output in February than in November. Within the February collection period, the sperm output for S-L stallions was greater (P less than .05) than that for either control of S-S stallions. The stimulatory effect of the S-L photoperiod also resulted in larger (P less than .05) testes and decreased (P less than .05) time to ejaculation for S-L stallions in February as compared with either controls or S-S stallions. Despite continued exposure to a 16:8 photoperiod, TSW and sperm output for S-L stallions eventually declined; presumably a consequence of photorefractoriness. The S-S stallions had seasonal cycles coincident with those for control stallions. Based on a sine wave model for TSW and sperm output, stallions in all three groups displayed significant seasonal cycles.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
