目的研究北五味子木脂素(Schisandra lignans,SCL)和北五味子粗多糖(Schisandra chinensis polysaccharides,SCP)对异丙肾上腺素(Isoproterenol,ISO)诱导小鼠心室重构的保护作用,并探讨其初步机制.方法采用连续10 d皮下注射ISO的方法诱导小鼠心室重构模型.小鼠随机分为对照组、ISO组、SCL低、中、高3个剂量组和SCP组,采用灌胃给药方法.实验结束后计算小鼠心脏重量指数;观察心肌病理形态学改变情况;HE染色后测定心肌纤维直径(MD);分光光度法测定心肌组织中羟脯氨酸(HYP)含量、一氧化氮(NO)含量、一氧化氮合酶(NOS)活性、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量.结果与对照组相比,ISO组小鼠心脏重量指数、HYP含量和MDA水平明显升高(P〈0.05或P〈0.01),NOS和SOD活性及NO水平明显下降(P〈0.05或P〈0.01);与异丙肾上腺素组相比,北五味子木脂素和北五味子多糖组能明显降低心脏重量指数(P〈0.05或P〈0.01),改善心肌病理学变化,抑制心肌组织中羟脯氨酸含量,提高SOD活性,降低MDA含量,提高心肌中NOS活性和NO水平(P〈0.05,P〈0.01或P〈0.001).结论北五味子木脂素和北五味子多糖对ISO诱导的小鼠心室重构具有抑制作用,其机制可能与提高NOS活性和NO水平、清除氧自由基、提高机体的抗氧化能力有关.  相似文献

[目的]介绍一种简便的建立大鼠心肌肥厚模型的方法。[方法]健康成年SD大鼠20只,试验组(10只)皮下注射异丙肾上腺素(ISO),对照组(10只)皮下注射生理盐水。计算2组大鼠HW/BW、LVW/BW;观察2组大鼠心脏的组织病理变化;检测血清乳酸脱氢酶、总抗氧化能力、碱性磷酸酶的活力及丙二醛的含量。[结果]与对照组相比,试验组大鼠的HW/BW、LVW/BW均明显升高;心肌细胞直径明显变大,细胞间质增多,细胞间距变大;血清中乳酸脱氢酶、碱性磷酸酶的活力及丙二醛的含量明显升高,而血清总抗氧化能力明显降低。[结论]大鼠心肌肥厚模型造模成功,该方法技术简便,容易掌握。  相似文献

[目的]利用FKBP结合蛋白12.6心脏特异性转基因(TG)小鼠,探讨FKBP12.6过表达在ISO所致心脏肥大纤维化病理过程中可能的保护作用。[方法]TG和对照非转基因(NTG)小鼠,连续皮下注射小剂量异丙肾上腺素(ISO),诱导扩张性心肌肥大和纤维化,通过心脏超声、PV-Loop等生理学和病理组织学等手段检测心脏功能和组织结构的变化。[结果]ISO未处理的TG和NTG小鼠相比较,心脏超声和PV-Loop指标以及纤维化和心肌细胞横截面积等无统计学差异;ISO处理后动物心脏呈扩张性肥大和纤维化表现,但TG和NTG小鼠之间功能学及病理组织学指标无统计学差异。[结论]未观察到心脏特异性过表达FKBP12.6对ISO诱导心肌损伤有明显的保护作用,FKBP12.6可能并不直接影响心肌细胞内质网RyR2的功能。  相似文献

Quantitative, qualitative and phenotypic body measurements were determined on Longling Yellow goats in Longling of Yunnan Province in China. Body weight was determined at birth and then at 3, 6, 9, 12, 18, 24, 36, and 48 mon of age. Body measurements traits including height at wither (HW), body length (BL), heart girth (HG), chest depth (CD), and circumference of cannon bone (CCB) were determined at 3, 6, 9, 12, 18, 24, 36, and 48 mon of age. Phenotypic traits including coat colour, horns type, tip horn torsion, nose type, hair on forehead, wattles, ear lobes, black back line, hair on legs, and black hair on belly were determined at 12, 18, 24, 36, and 48 mon of age. Males were heavier (P〈0.01 or P〈0.05) than females; and except for chest depth at 24 mon of age (P 〉 0.05), all other body measurements were also higher (P 〈 0.01 or P 〈 0.05) in males. HW, BL, CD, HG, and BW increased (P 〈 0.01) with age, except for 48 mon of females (P 〉 0.05); while CCB did not always increase with age. HG had the highest correlation (P〈0.01) with body weights of both sexes. No differences were observed between the two sexes in the colour of the coat, horn type, tip horn torsion, nose type, ear lobes, black back line, and black hair on belly. However, differences exist between the two sexes in the presence of hair on forehead, wattles and long hair on legs. The female goats absence of tip horn torsion had positive effect on all variables measured at 36 and 48 mort of age (P〈0.01 or P〈0.05). The presence of wattles was associated with heavier body weights from 18 mort (P〈0.01) of age onward, and also with longer HW, BL and CD (P〈0.01 or P〈0.05) in female goats. Female goats with ear lobes had heavier body weights at 12, 36 and 48 mon (P〈0.01) and longer CCB at 12 and 24 mon of ages (P〈0.05). Hair on legs and black hair on belly only had partly effect body measurements on 12 mon of Longling Yellow female goats. Results of this study provide valuable information that will assist breeders and genetic improvement specialists when conducting selection and preservation programs in Longling Yellow goats in China.  相似文献

以10只(雌雄各半)280 d海兰鸡为材料,比较雌雄个体体重和14个主要脏器重量及系数的差异。结果表明,雄性海兰鸡体重及腺胃、胸腺重量显著高于雌性(P〈0.05),心脏重量极显著高于雌性(P〈0.01) 雄性海兰鸡心脏系数极显著高于雌性(P〈0.01),脾脏和胸腺系数显著高于雌性(P〈0.05),肝脏系数显著低于雌性(P〈0.05) 其他脏器重量及系数差异均不显著。  相似文献

目的:探讨三七总皂甙(PNS)对转化生长因子β1(TGF-β1)诱导的人肾小管上皮细胞(HK-2)总胶原分泌及细胞生长的影响。方法:采用消化法检测细胞培养上清液中羟脯氨酸的含量,换算成总胶原分泌量;采用流式细胞仪(FCM)检测PNS对TGF-β1诱导的HK-2细胞周期、细胞增殖指数(PI)的影响。结果:消化法示:TGF-β1组羟脯氨酸的含量及总胶原分泌量均高于正常对照组,PNS 200~800mg/L可降低细胞培养上清中羟脯氨酸的含量及总胶原分泌置,且随着PNS浓度的升高而降低(P〈0.01);流武细胞仪(FCM)示:TGF-β1组G1期细胞低于正常对照组,PI高于正常对照组(P〈0.01),PNS 400~800mg/L使G1期细胞显著升高,PI显著降低,且呈剂量依赖性。结论:PNS可通过抑制TGF-β1诱导的肾小管上皮细胞总胶原分泌及细胞增殖,从而参与延缓肾间质纤维化的进程,改善肾脏功能。  相似文献

目的 研究养心通脉方对异丙肾上腺素诱导的大鼠心肌缺血损伤模型心肌细胞线粒体呼吸功能的影响。方法 实验大鼠被随机分为3组(每组15只):对照组、模型组、养心通脉方组,实验结束后所有动物采用脱颈椎法处死,通过测定呼吸控制率(RCR)、二磷酸腺苷磷/氧(ADP/O)比值及ATP的浓度来评价线粒体的呼吸功能,并对线粒体内胆固醇、磷脂(PL)、甘油三酯(TG)和游离脂肪酸(FFAs)的含量进行测定。结果 模型组中大鼠心肌细胞线粒体内RCR、ADP/O比值及ATP浓度均明显低于对照组(P<0.001),而在养心通脉方组中心肌细胞线粒体内RCR、ADP/O比值及ATP浓度均接近于对照组(P>0.05)。与对照组相比,模型组中胆固醇、TG和FFAs的水平均明显升高(P<0.001),同时磷脂(PL)水平明显降低(P<0.001)。但在养心通脉方组中心肌细胞线粒体内胆固醇、TG、FFAs以及PL的水平均与对照组无明显差异(P>0.05)。结论 养心通脉方对异丙肾上腺素诱导的大鼠心肌缺血损伤模型心肌细胞线粒体的呼吸功能及其膜完整性具有保护作用。  相似文献

[目的]观察獐牙菜苦苷对氧化应激引起的大鼠心肌细胞凋亡的保护作用,初步探讨獐牙菜苦苷抗心肌细胞氧化应激的保护作用机制。[方法]用差速贴壁法培养SD乳鼠心肌细胞,检测不同给药组中SD乳鼠细胞心肌细胞存活率以及培养液中超氧化物岐化酶(SOD)、丙二醛(MDA)含量;用免疫细胞化学检测心肌细胞的相关凋亡蛋白bcl-2、Bax表达;用吖啶橙荧光染色检测细胞的凋亡情况。[结果]3种不同浓度獐牙菜苦苷均能提高心肌细胞存活率和细胞上清液中SOD含量,且能降低MDA含量,与H2O2刺激组比较有显著性差异(P〈0.05;P〈0.01)。[结论]獐牙菜苦苷对H2O2诱导损伤的心肌细胞有保护作用,其作用机制可能是通过影响氧化系统以及调控特异性蛋白来抑制心肌细胞凋亡。  相似文献

This study was carried out to investigate the growth performance, carcass yield and meat quality of large-type Korean native ducks (KND). A total of 420 day-old ducklings were divided into four treatments (seven replicates/treatment, 15 birds/replicate) by strains (A and B) and gender (male and female) in a 2 × 2 factorial arrangement under complete randomized design. The body weight (BW) of female KND was higher at week 2 and for male it was higher in week 8 (P 〈 0.05). BW gain, feed intake and feed conversion ratio of male KND were higher than those of female KND (P 〈 0.05) during week 0 to week 8. The carcass yield of both strains increased from 6 weeks to 8 weeks (P 〈 0.05). The breast meat ratio was the highest at week 8 whereas the leg meat ratio was the highest at 6 weeks of age in both strains (P 〈 0.01). Lightness (L*), yellowness (b*) and juiciness of meat were the highest at 6 weeks (P 〈 0.05). Moisture, ash and fat contents of meat were the highest at 7 weeks (P 〈 0.05) and protein content was the highest at 8 weeks (P 〈 0.01). These results may provide the basic data on growth performance and meat quality of large-type KND, which can be used for genetic improvements and rearing at commercial scale.  相似文献

Selenium protects cells from oxidative damage and reduces lipopolysaccharide (LPS)-induced expression of inflammatory cytokines. Because inflammatory cytokines induce growth depression, we hypothesized that selenium-rich yeast (SeY) would inhibit growth depression caused by LPS. Twenty Meishan weanling pigs [28 (s.d. 2 d), 7.6 (s.d. 0.3 kg) body weight (BW)] were used in a 2×2 factorial design (n=5). The main factors were dietary Se sources [selenium-rich yeast (SeY) vs. sodium selenite (SSe)] with supplement level at 0.3 mg Se/kg, and immunological stress (LPS vs. saline). The experiment lasted for 28 d. On d 14 and d 21, half of pigs in each dietary treatment were intramuscularly injected with either 50 μg/kg BW LPS or an equivalent amount of sterile saline. Blood samples were collected at 3 h post-injection. There were no interactions between dietary Se sources and immunological stress on growth performance. LPS depressed average daily gain (ADG) (d 14-21, P〈0.05; d 21-28, P〈 0.01), and elevated feed to gain ratio (F/G) (d 21-28, P〈0.05), while SeY elevated ADG (d 0-14, P〈0.05; d 21-28 and d 0-28, P〈0.01), and decreased F/G (P〈0.01). Interactions were observed on interleukin-1β(IL-1β) and interleukin-6 (IL-6) from serum samples of d 14 and d 21 (P〈0.01), as well as superoxide dismutase (SOD) and insulin like growth factor (IGF-1) of d 21 (P〈0.05). In conclusion, SeY relieved the growth depression of piglets immunologically stressed by LPS, which maybe resulted from the decreased externalization of inflammatory cytokines and improved antioxidant status.  相似文献

观察糖康抗氧化应激对实验性糖尿病大鼠视网膜病变的疗效并探讨其作用机制。采用腹腔注射链脲佐菌素(STZ)复制糖尿病模型并随机分为正常对照组、模型组、糖康高、中和低剂量组和达纳康组,每日灌胃,定期检测大鼠体重和非空腹血糖;第12周末,消化铺片法观察视网膜微血管病理和形态学改变;化学比色法检测各组大鼠血清及视网膜组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。与正常组比较,模型组血清及视网膜SOD活性明显降低(P0.01),MDA含量明显升高(P0.01),无细胞毛细血管条索数明显升高,约为正常组的5.33倍(P0.01);与模型组比较,各治疗组均可提高血清及视网膜SOD活性,降低MDA含量,且以糖康高剂量组最为显著(P0.01),无细胞毛细血管条索数明显降低(P0.01)。糖康可显著降低糖尿病大鼠的氧化应激水平,提高糖尿病大鼠血清及视网膜的抗氧化能力。  相似文献

目的观察山葡萄多酚(PVAR)对大鼠缺血再灌注心肌线粒体损伤的保护作用.方法结扎Wistar大鼠冠状动脉30min再灌注20min,测定心肌线粒体中琥珀酸脱氢酶(SDH)、细胞色素C氧化酶(CCO)和SOD活性,膜磷脂及MDA含量.结果与模型组比较,PVAR(200,400mg/kg)组SDH,CCO,SOD活性明显升高(P〈0.05),膜磷脂含量增加(P〈0.05),MDA含量降低(P〈0.05).结论PVAR对缺血再灌注损伤的心肌线粒体功能具有保护作用.  相似文献

目的探讨粒细胞集落刺激因子(G-CSF)对急性心肌梗死(AMI)大鼠血清及心肌组织中一氧化氮(NO)、丙二醛(MDA)和超氧化物歧化酶(SOD)含量的影响.方法通过结扎冠状动脉前降支建立急性心肌梗死(AMI)大鼠模型,将AMI大鼠随机分为GCSF治疗组(GAMI组)、心肌梗死对照组(AMI组)和假手术组(SO组),GAMI组模型制备后3h给予生理盐水稀释的rhG-CSF(浓度为2rag/L)皮下注射10μg/kg/d,共5d.AMI组和SO组3h后给予等量的生理盐水皮下注射,共5d.最后一次给药后24h取各组大鼠血清及心肌组织检测NO、MDA、SOD的含量.结果AMI组大鼠血清和心肌组织中MDA水平显著高于SO组(P〈0.01),NO、SOD值显著低于SO组(P〈0.01).GAMI组大鼠血清和心肌组织中MDA水平显著低于AMI组(P〈0.05),NO、SOD值显著高于AMI组(P〈0.05).结论G-CSF能提高NO含量,增强SOD的活性.降低MDA的含量,对急性心肌梗死具有明显的保护作用.  相似文献

目的探讨胡桃楸叶乙醇提取物(AELJ)对X射线辐射损伤小鼠的保护作用。方法加只小鼠随机分为正常对照组、照射对照组、AELI低剂量(300mg/kg)组和AEU高剂量(600mg/kg)组。经2.0Gy X射线照射后,测定小鼠脾脏和胸腺指数、脾脏T淋巴细胞转化率、肝组织匀浆MDA含量和SOD的活性。结果与照射对照组比较,AEU高、低剂量组小鼠脾指数、胸腺指数、T淋巴细胞转化率、肝组织匀浆中SOD活力均显著增高(P〈0.05);MDA含量显著降低(P〈0.05)。结论AELI对X射线辐射损伤小鼠具有一定的保护作用。  相似文献

目的探讨粒细胞集落刺激因子(G-CSF)对急性心肌梗死(AMI)大鼠血浆及心肌组织中血管紧张素Ⅱ(AngⅡ)和内皮素(ET)含量的影响。方法通过结扎冠状动脉前降支建立急性心肌梗死(AMI)大鼠模型,将大鼠随机分为G-CSF治疗组(GAMI组)、心肌梗死对照组(AMI组)和假手术组(SO组),GAMI组模型制备后3h给予生理盐水稀释的rhG-CSF(浓度为2mg/L)皮下注射10μg/kg/d,共5d。AMI组和SO组3h后给予等量的生理盐水皮下注射,共5d。三组大鼠于急性心肌梗死模型制造成功后24h、1周和2周后检测各组血浆及心肌组织中AngⅡ和ET的含量。结果 24hAMI组与SO组血浆及心肌组织中AngⅡ和ET含量相比均有提高但只有心肌中ET含量有显著的差异(P〈0。05)。1周后AMI组与SO组血浆及心肌组织中AngⅡ和ET含量相比两者含量有明显增高(P〈0。05)。2周后AMI组与SO组血浆及心肌组织中AngⅡ和ET含量相比两者含量增加的更加明显(P〈0。01)。GAMI组血浆及心肌组织中AngⅡ和ET含量与相应的AMI组比较有明显的降低。结论 G-CSF能降低急性心肌梗死模型大鼠血浆和心肌组织中AngⅡ和ET的含量,G-CSF对AMI大鼠损伤心肌有保护作用。  相似文献

为了探讨大豆异黄酮(SI)对去卵巢大鼠脾脏中神经生长因子(NGF)、白细胞介素2(IL-2)蛋白表达的影响,本研究在成功建立去卵巢大鼠模型的基础上,对去卵巢大鼠分别隔天皮下注射高(1.5 mg·kg-1)、中(1.0 mg·kg-1)、低(0.5 mg·kg-1 )剂量的SI,假手术组(仅手术而不去卵巢)和溶媒组(去卵巢)大鼠隔天皮下注射溶媒剂(乙醇)。补充SI至14 d和42 d时,每组随机剖杀5只大鼠,采用免疫组织化学方法,对大鼠脾脏NGF、IL-2蛋白的表达变化进行研究。结果表明,脾脏中广泛分布着NGF、IL-2蛋白,主要分布于被膜下方的红髓区,在白髓分布量极少。去卵巢后,大鼠脾脏中NGF、IL-2蛋白的表达强度和阳性细胞数目均显著下降,而且随着去除卵巢时间的延长下降加剧。体外补充SI后14 d,各剂量组和42 d低、中剂量组仅有部分恢复,变化不显著;补充高剂量SI 42 d后,脾脏NGF、IL-2蛋白的表达强度和阳性细胞数目基本恢复至假手术组的水平。大豆异黄酮能上调NGF、IL-2蛋白在脾脏内的表达,并呈现时间和剂量的依赖性。  相似文献

目的:研究黄连素对软脂酸诱导人脐静脉内皮细胞( HUVECs)损伤的保护作用,并初步探讨其作用机制.方法采用500μmol/L软脂酸培养HUVECs 24 h,建立内皮细胞损伤模型,MTT法观察黄连素对细胞存活率的影响;检测细胞培养上清液中一氧化氮( NO)含量;RT-PCR法检测内皮型一氧化氮合酶( eNOS) mRNA水平;Western blot方法检测eNOS和磷酸化eNOS蛋白的表达.结果与对照组比较,软脂酸组细胞存活率降低( P〈0.05),培养液中NO含量下降(P〈0.05),细胞内eNOS mRNA水平、eNOS及磷酸化eNOS蛋白表达水平均显著下降(P〈0.01,P〈0.05).与软脂酸组比较,黄连素组细胞存活率增加,培养液中NO含量明显提高(P〈0.05),细胞内eNOS mRNA水平和磷酸化蛋白表达水平均显著提高(P〈0.01,P〈0.05).结论黄连素对软脂酸引起的血管内皮细胞损伤有显著的保护作用,并可能与上调eNOS、促进NO生成有关.  相似文献

以己二酸为相变材料,二氧化硅溶胶为基体,采用水热法制备己二酸/二氧化硅复合相变储热材料。应用X 射线衍射仪(XRD)、外光谱分析(FT-IR)、场发射扫描电子显微镜(FE-SEM)、同步热分析仪(TG)和差示扫描量热仪(DSC)测试所制备复合材料的显微结构、相变温度和相变焓。结果表明:当反应条件为pH=5、反应温度为150℃、反应时间为4 h时,己二酸与二氧化硅之间是简单的物理嵌合关系,复合材料粒径较均匀,平均粒径在1~2μm;当己二酸的质量分数为30%时,复合材料相变温度峰值为139.0℃,相变焓为48.82 J/g;当己二酸的质量分数为50%时,复合材料相变温度峰值为140.5℃,相变焓为71.89 J/g。  相似文献
