A decontamination trial on the effectiveness of hot water or acidified sodium chlorite (SANOVA) treatment on Salmonella spp., Escherichia coli and Total Viable Count (TVC) was undertaken on pork carcases prior to primary chilling in two large pork abattoirs in Australia using belly-strip excision sampling. A total of 123 samples from Abattoir A and 400 samples from Abattoir B were cultured and analysed. Test pigs were selected from herds with a known high level of on-farm Salmonella infection. At Abattoir A, Salmonella spp. were not isolated from carcases. The prevalence of E. coli on control carcases was 92.9% compared with 9.8% for hot water and 12.5% for SANOVA treated carcases. The mean log(10) E. coli concentration for control carcases was 0.89 cfu/gram, compared with -0.83 cfu/gram from hot water and -0.75 cfu/gram from SANOVA treated carcases. The mean log(10) TVC for control carcases was 4.06 compared with 1.81 cfu/gram for hot water and 2.76 cfu/gram for SANOVA treated carcases. At Abattoir B, the prevalence of Salmonella on control carcases was 16% compared with 2.7% for hot water and 7.0% for SANOVA treated carcases. The prevalence of E. coli on control carcases was 69.3% compared with 22% for hot water and 30% for SANOVA treated carcases. The mean log(10) E. coli concentration for control carcases was 0.45 cfu/gram, compared with -0.65 cfu/gram from hot water and -0.60 cfu/gram from SANOVA treated carcases. The mean log(10) TVC for control carcases was 3.00 cfu/gram compared with 2.10 cfu/gram for hot water and 2.53 cfu/gram for SANOVA treated carcases. The reductions in prevalence and mean log(10) concentrations in the present trial were all found to be statistically significant and indicate that carcases decontamination with either hot water or SANOVA are effective risk management options immediately available to the pork industry.  相似文献

Eighty-eight rabbits weaned at the age of 35 days were divided into four groups. Rabbits of the first two groups were fed a granulated feed, free of antimicrobials. Rabbits of the 3rd and the 4th groups were fed the same diet, supplemented with caprylic acid at 5 g/kg, and with triacylglycerols (TAG) of caprylic and capric acid at 10 g/kg, respectively. Rabbits of the 2nd, 3rd and 4th groups were challenged orally with 10(9) cells of Escherichia coli of the O103 serogroup. Numbers of coliform bacteria in faeces of non-infected rabbits averaged 4.66 log(10)cfu/g. Six days after inoculation, caprylic acid and TAG of caprylic and capric acid decreased faecal output of coliforms from 10.18+/-0.62 to 7.79+/-0.48 log(10)cfu/g and 8.04+/-0.50 log(10)cfu/g, respectively. In the 2nd, 3rd and 4th groups 15, 11 and 9 infected rabbits died, respectively. However, the differences in mortality rate were not statistically significant. Surviving rabbits were slaughtered at 53 days of age. In caecal contents of infected rabbits, numbers of coliform bacteria were significantly reduced from 8.71 log(10)cfu/g to 5.55-5.83 log(10)cfu/g in treated groups. It can be concluded that both antimicrobial lipids are active against coliform bacteria, and may improve the resistance of weaned rabbits to enterocolitis.  相似文献

This study was done to evaluate the microbial contamination by Salmonella spp., Escherichia coli, fecal coliforms, and total aerobic count of the stick wound of swine carcasses. The effectiveness of trimming the stick wound in the 2 Québec slaughterhouses visited was evaluated. A bacteriological analysis was done on 276 stick wounds. Results indicated that, before trimming, 0.9% stick wounds were contaminated by Salmonella spp. Contamination by coliforms was observed in 40.6% of samples, and 27.7% were positive for E. coli. After trimming the stick wounds, 1.1% were contaminated by Salmonella spp., 34.1% were contaminated by coliforms, and 26.2% were positive for E. coli. The results showed that trimming contributes to reducing significantly the bacterial total count at the site and that the bacterial load at this site was less important than that found on the brisket.  相似文献

1. The efficacy of the AvGard Trisodium Phosphate (TSP) immersion carcase wash process was evaluated during 5 industrial trials against Salmonella , Enterobacteriaceae , thermotolerant coliforms and total aerobic count. The effect against Pseudomonas was also studied in the first 3 trials. 2. Dramatic reductions in Salmonella incidence were seen using a whole carcase rinse method. In 4 of the 5 trial sites, only one positive sample was found after AvGard of an average control incidence of 57.7%. In the 5th site, a water-chilled broiler plant, an average control incidence of 74.0% was reduced to 9.4% after AvGard 3. In the latter case, Most Probable Number (MPN) analyses were performed on some of the Salmonella positive samples taken from the control and post-treatment series; the average MPN count per carcase on controls was 115, whereas for AvGard treated birds the figure was only 0.6 per carcase, a greater than 2 log reduction. 4. In addition, AvGard treatment gave average log reductions for all trials of: Enterobacteriaceae ; 2.5 log; Coliforms; 2.7 log, and Total Aerobic Count; 1.1 log, leading to carcases substantially free of Gram negative pathogens. 5. Pseudomonas was reduced by an average of 1.7 log in the first 3 trials, dramatically reducing the carcase loading of this important spoilage organism. treatment (average 0.5% incidence), in spite treatment.  相似文献

1. Neck skin samples were taken from chickens and turkeys at all the main stages of processing to monitor changes in total viable count (TVC) and counts of coliforms and pseudomonads.

2. Processing reduced TVC by up to 100‐fold. Geometric mean counts after packaging were log10 4.4 to 5.3 CFU/g whilst corresponding counts of coliforms were 2.7 to 3.8 CFU/g.

3. Increases in mean TVC or coliforms as a result of either defeathering or evisceration did not exceed 0.6 log.

4. Pseudomonads represented only a minor fraction of the initial microflora of the bird and were often reduced by scalding to a figure which could not be detected by direct plating of samples; however, subsequent contamination resulted in means between log10 2.9 and 4.0 CFU/g for packaged carcases.

5. Although Staphylococcus aureus was readily isolated from defeathering equipment, mean counts from defeathered carcases were always below log10 3.0 CFU/g.  相似文献

Samples of 51 mink feeds and 101 raw material batches were analysed for pathogenic staphylococci. Of the feed samples 37 % were contaminated with pathogenic staphylococci, the mean count being 3.02 log CFU (colony forming units)/g and the highest count being 4.48 log CFU/g of feed. Of the raw materials investigated, slaughter-house offals were most frequently contaminated and contained highest counts of S. aureus, 60 % of the samples being positive with a mean log count of 3.86/g and a maximum of 5.70/g. Forty-eight staphylococcal isolates were tested for eniterotoxin production. Toxin A, B or G were produced by eight isolates, A being the most common. Rangelands worldwide have been subject to broadscale modification, such as widespread predator control, introduction of permanent livestock water and altered vegetation to improve grazing. In Australia, these landscape changes have resulted in kangaroos (i.e. large macropods) populations increasing over the past 200 years. Kangaroos are a key contributor to total grazing pressure and in conjunction with livestock and feral herbivores have been linked to land degradation. We used 22 years of aerial survey data to investigate whether the density of 3 macropod species in the southern rangelands of Western Australia was associated with: (i) land use, including type of livestock, total livestock, density of feral goats, type of land tenure, and kangaroo commercial harvest effort; (ii) predator management, including permitted dingo control effort, estimated dingo abundance, and presence of the State Barrier Fence (a dingo exclusion fence); and (iii) environmental variables: ruggedness, rainfall, fractional cover, and total standing dry matter. Red kangaroos (Osphranter rufus) were most abundant in flat, open vegetation, on pastoral land, where area permitted for dingo control was high, and numbers were positively associated with antecedent rainfall with a 12-month delay. Western grey kangaroos (Macropus fuliginosus) were most abundant on flat, agricultural land, but less abundant in areas with high permitted dingo control. Euros (Osphranter robustus) were most abundant in rugged pastoral land with open vegetation, where permitted dingo control was high. While environmental variables are key drivers of landscape productivity and kangaroo populations, anthropogenic factors such as land use and permitted dingo control are strongly associated with kangaroo abundance.  相似文献

To study the phenomenon of host adaptation in Salmonella, a mathematical model has been developed which permits a definition and experimental investigation of the specific interaction between the adapted serovar and the adequate host. After experimental infection using a mixture of equal parts of two Salmonella strains, A and B, the bacterial concentrations CA and CB were determined in the organs of the animals infected. If an animal of species a and an animal of species b are infected with the same mixture of strain A adapted to a and strain B adapted to b, an expression: log10Qab = log10CaA + log10CbB - log10CaB - log10CbA may be calculated which describes the influence of the specific serovar-host interaction on the dynamics of the bacterial count. The variable Qab was determined using four Salmonella dublin, four Salmonella choleraesuis and five Salmonella gallinarum/pullorum strains in a total of 63 pairs of different hosts (calf and pig, calf and chicken, or pig and chicken). On this basis, the following statements can be made. The epidemiologically defined host adaptation of Salmonellas is accompanied by a specific agent-host interaction between the adapted serovar and adequate host. It promotes adherence and spreading of the agent in the adequate host. The effect was particularly expressed on day 3 post-infection and could be detected both in the lumen of the anterior sections of the intestine and in the intestinal lymph nodes and the liver. In part, the host-independent strain characteristics had a greater influence than the specific serovar-host interaction on the dynamics of the bacterial count. Strains of non-adapted serovars may thus result in a more intense colonization and invasion of the host than simultaneously administered bacteria of a serovar which is adapted to the respective host. The effects of a specific serovar-host interaction on colonization of and spreading in the host should be considered only as a component which contributes to the phenomenon of host adaptation.  相似文献

Beef and pig carcases, meat products, frozen chickens, and sewage were examined in 3 separate surveys for antibiotic resistant coliforms. Escherichia coli was isolated from 18 of 50 beef carcases; the numbers were low and resistance was found only to tetracycline. E. coli was isolated from 45 of 50 pig carcases; the numbers were high and showed a range of patterns of multiple antibiotic resistance. In meat products, the proportion of E. coli in contaminating organisms was low, and most resistance found was to tetracycline and streptomycin. E. coli was isolated from 66 of 75 chickens and these gave 23 patterns of antibiotic resistance, often multiple. Sewage from hospital or domestic origin and abattoir effluent yielded approximately 106 coliforms/ml, most of which were resistant to one or more antibiotics; few of those from hospital or domestic origin however, were classified as E. coli of faecal origin. Twenty-four patterns of resistance were found in coliforms from domestic sewage, 19 from hospital sewage and 11 from abattoir effluent. Transfer of resistance, often multiple, was achieved from 55% of 447 resistant strains to an E. coli K12 recipient. Much more information is required on the prevalence of R-factors in bacteria associated with food producing animals and their products.  相似文献

Background

Feed-borne spread of Salmonella spp. to pigs has been documented several times in recent years in Sweden. Experiences from the field suggest that feed-associated serotypes might be less transmittable and subsequently easier to eradicate from pig herds than other serotypes more commonly associated to pigs. Four Salmonella serotypes were selected for experimental studies in pigs in order to study transmissibility and compare possible differences between feed-assoociated (S Cubana and S Yoruba) and pig-associated serotypes (S Derby and S Typhimurium).

Methods

Direct contact transmission was studied in four groups of pigs formed by six 10-week-old salmonella negative pigs commingled with two fatteners excreting one of the four salmonella serotypes. Indirect transmission was studied by putting six 10-week-old salmonella negative pigs in each of four salmonella contaminated rooms. Each room had previously housed a group of pigs, excreting one of the four selected serotypes.All pigs were monitored for two weeks with respect to the faecal excretion of salmonella and the presence of serum antibodies. At the end of the trial, eight samples from inner tissues and organs were collected from each pig at necropsy.

Results

In the four direct transmission groups, one pig shed Salmonella (Cubana) at one occasion. At necropsy, S Typhimurium was isolated from one pig.In the indirect transmission groups, two pigs in the Yoruba room and one pig in each of the other rooms were excreting detectable levels of Salmonella once during the study period of two weeks. At necropsy, S Derby was isolated from one of six pigs in the Derby room and S Typhimurium was isolated from four of the six pigs in the Typhimurium room.No significant serological response could be detected in any of the 48 pigs.

Conclusions

These results show that all four selected serotypes were able to be transmitted in at least one of these field-like trials, but the transmission rate was low in all groups and no obvious differences between feed-associated and pig-associated serotypes in the transmission to naïve pigs and their subsequent faecal shedding were revealed. However, the post mortem results indicated a higher detection of S Typhimurium in the ileocecal lymph nodes of pigs introduced into a contaminated environment in comparison with the other three serotypes.  相似文献

Salmonella and Campylobacter are common bacterial pathogens associated with human gastro-enteritis; and raw poultry is considered to be an important source of these bacteria. To evaluate whether the Salmonella serovars and Campylobacter spp. bacteria could be monitored for the purpose of microbial presence, enumeration and antimicrobial resistance in raw poultry, 152 poultry carcasses were randomly selected from 10 markets in retail outlets of Phnom Penh during March 2006 to February 2007. The majority of poultry samples was contaminated by Salmonella serovars (88.2%) and Campylobacter spp. (80.9%). A very high contamination of Salmonella was found at 3-4 log?? CFU/g for 22.4% of samples and of Campylobacter at 7-8 log?? CFU/g for 1.3% of samples. Fifty nine different Salmonella serovars contaminated 134 poultry carcasses; five most prevalent serovars covered 29.1% of serovars isolates (Anatum, Typhimurium, Corvallis, Stanley and Enteritidis). Three Campylobacter species contaminating 123 raw poultry were Campylobacter jejuni (50.0%), Campylobacter coli (29.0%) and Campylobacter lari (21.0%). High antibiotic resistance percentages were found among Salmonella serovars and Campylobacter spp. isolates. This study revealed that raw poultry at the retail outlets in Phnom Penh markets are contaminated with high prevalences of food-borne pathogens, and communicating the importance of minimizing this risk in reducing human infections.  相似文献

Confined animal feeding operations can be a source of odor emissions, global warming gases, water pollution, and food contamination. Laboratory studies have indicated that plant oils with antimicrobial activity can be used to control pathogens and odor emissions from cattle and swine wastes. However, these oils are aromatic and may volatilize when applied topically. Our objectives were to evaluate the volatility of thymol from a feedlot surface and the effectiveness of topically applying thyme oil (2.5% thymol), incorporated into corncob granules and added once per week, to control odor emissions and total coliforms in feedlot manure. In the first study, thymol either volatilized or was degraded within 28 d after topical application. In a second study, thyme oil (2.5% thymol) was incorporated into corncobs and applied to pen surfaces weekly. Manure samples from 6 locations in each pen were collected from 3 untreated and 3 thymol-corncob-treated pens (15 x 150 m; fifty 400-kg cattle/pen), 3 times per week for 8 wk. Samples were analyzed for thymol concentration, total VFA, branched-chain VFA, aromatic compounds, and the number of Escherichia coli and total coliform bacteria. Over the 8 wk, with the exception of wk 7, the desired thymol concentration of 15 to 20 micromol/g DM was maintained in the manure. Concentrations of VFA and branched chain-VFA increased over time in untreated and treated pens. However, the rate of VFA accumulation in treated pens (7.5 +/- 1.3 micromol.g DM(-1).wk(-1)) was less (P < 0.01) than the rate of accumulation in untreated pens (18.0 +/- 2.1 micromol.g DM(-1).wk(-1)). Likewise, the rate of branched-chain VFA accumulation in treated pens (0.31 +/- 0.04 micromol.g DM(-1).wk(-1)) was less (P < 0.01) than in untreated pens (0.55 +/- 0.06 micromol.g DM(-1).wk(1)). The concentrations of E. coli in treated pens (2.9 +/- 1.2 x 10(5) cfu.g DM(-1)) were 91% less (P < 0.04) than in untreated pens (31.1 +/- 4.0 x 10(5) cfu.g DM(-1)). Similarly, concentrations of coliforms in treated pens (3.7 +/- 1.3 x 10(5) cfu.g DM(-1)) were 89% less (P < 0.04) than those of untreated pens (35.3 +/- 4.2 x 10(5) cfu.g DM(-1)). These results indicate that odor emissions and total coliforms can be reduced in feedlot manure with a once per week application of thymol incorporated in a granular form. However, corncobs are bulky, and other granular carriers with a greater carrying capacity for thyme oil should be explored.  相似文献

This study was to identify the risk factors for Salmonella spp. contamination of Senegalese chicken carcases during slaughtering. One hundred and twenty traditional slaughterhouses were studied from January 2000 to December 2002 in and around Dakar. A questionnaire was answered by the slaughterers, and samples of breast skin were taken to assess the Salmonella status of chicken carcases. Results showed that 43.3% of the batches were contaminated with Salmonella, indicating Salmonella Hadar and Salmonella Brancaster as the two main serovars. Salmonella contamination of the carcases after slaughtering was related to contamination of the live birds. Feed withdrawal before slaughtering and thorough cleaning and disinfection procedures decreased the risk of contamination. One individual worker for each slaughtering stage was also associated with a decreasing risk of contamination. Using scalding water for plucking the chicken carcases increased contamination risk. These results will help slaughters to produce safer products for local consumers.  相似文献

Twenty-seven 9-12 months old healthy male dromedarian camels were used to determine total nucleated leucocyte count (TNCC), absolute and percentages of polymorphonuclear (PMN) and mononuclear leucocytes, and total protein (TP) concentration in synovial fluid from grossly and radiographically normal fetlock joints. Arthrocentesis was performed bilaterally from the fetlock joints of the forelimbs and hindlimbs. Blood contaminated samples and samples obtained from grossly or radiographically abnormal joints were excluded. The mean +/- SD of TNCC in 108 samples of fetlock joint synovial fluids was 500 +/- 400 cells/microl. Monocytes/macrophages were the predominant cell type. There were no significant differences in mean TNCC, absolute numbers and percentages of various leucocytes and TP concentrations between the right and left fetlock joints of the forelimbs and hindlimbs or between the fetlock joints of the forelimbs and hindlimbs. The mean +/- SD of absolute numbers and percentages of various cell types were: PMN leucocytes 1 +/- 2 cells/microl (2%), lymphocytes 116 +/- 167 cells/microl (26%), and monocytes/macrophages 383 +/- 323 cells/microl (72%). The mean +/- SD of TP concentration was 2 +/- 1 g/dl.  相似文献

Objective This study aimed to determine the presence and concentration of Escherichia coli O157 and Salmonella spp. on fleece, faeces and carcases of sheep during slaughter. Procedure Faeces, fleece and pre-chill carcase samples were collected from 164 sheep slaughtered at two Australian abattoirs. The presence of E. coli O157 and Salmonella spp. were determined by use of automated immunomagnetic separation (AIMS) with enumeration by use of the ‘most probable number’ (MPN) method. Results Escherichia coli O157 was isolated from 5% of faeces, 3% of fleeces and 0.6% of pre-chill carcases. The mean log10 count of E. coli O157 positive faecal samples was 2.32 MPN/g, but counts on fleeces and carcases were below the countable limit (−1 log10 MPN/cm2). Salmonella spp. were isolated from 20% of faeces, 13% of fleeces and 1.3% of pre-chill carcases. The mean log10 count of Salmonella spp. in faeces was 1.43 MPN/g and on fleece was −0.24 MPN/cm2, but counts on carcases were below the countable limit (−1 log10 MPN/cm2). Conclusion The prevalence and concentration of pathogens were low in the sheep tested in this study, indicating a low risk of human infection from products derived from these animals.  相似文献

The fecal flora of 56 clinically healthy and 23 sick horses were examined bacteriologically for counts of Clostridium perfringens, molds, coliforms, alpha- and beta-hemolytic streptococci, and microbes belonging to genus Bacillus, as well as for the presence of Salmonella spp. Of the healthy horses, 85.7% had a C perfringens count less than 10(1) colony-forming units/g of feces. Of the healthy horses, lowest counts were found in race-horses. Of the sick horses, equine intestinal clostridiosis was diagnosed in 2 horses with large C perfringens counts (10(4) to 10(7) colony-forming units/g) and with acute diarrhea. The 7 isolates of C perfringens were identified as serotype A. Salmonella spp were not detected from any of the horses. The study indicated that diagnosing equine intestinal clostridiosis based on the determination of the fecal C perfringens count was suitable.  相似文献

Thirty-one farrow-to-finish pig units with a Zoonoses National Control Programme (ZNCP) for Salmonella above the UK target of 10 per cent during the previous 12 months were selected for the study. Pooled faecal samples were collected from different groups of pigs. Furthermore, mice, rat and bird faeces and carcases were collected, if found on the unit. In total, 937 samples were collected on-farm and analysed for Salmonella. The four carcases selected monthly per producer by the slaughterhouse for the British Pig Executive ZNCP Salmonella Programme were tested for antibodies to groups B and C with a mix-ELISA test. The same four carcases were swabbed externally and internally, mesenteric lymph nodes (MLNs) were collected and colonic contents were swabbed. A wide variety of Salmonella serovars was isolated from the samples. Most of the isolates were detected in the rearing herd (on-farm) and in the MLNs (slaughterhouse). There was no correlation between ELISA results from the meat juice and bacteriological isolations using Spearman correlation analyses. Furthermore, no significant association was found between positive results of ELISA and positive results from the bacteriological samples taken.  相似文献

The mean (+/- sd) batch prevalence of pleura stripping of finished pigs going for slaughter in four abattoirs in eastern England was 0.15 +/- 0.04, and the average batch proportion of lungs totally condemned was 0.23 +/- 0.05. There was a significant positive correlation between the number of carcases requiring total lung condemnation and the number of carcases requiring pleura stripping. There was no significant differences between the type of producer, the herd size or the mean batch live weight and the prevalence of pleurisy, either within individual abattoirs or combining the data from the four abattoirs. There was considerable variation in the batch prevalence of pleurisy from the same producer, indicating the need to monitor a number of batches in order to determine the prevalence of pleurisy on a farm.  相似文献

Chlortetracycline (CTC) resistance in anaerobic and coliform bacteria was examined in the large intestines of two groups of growing pigs that had received antibiotic-free diets since weaning. One group of pigs was from a low resistance herd (LR) that had not received antibiotics for 8 yr, while the other group of pigs was from a high resistance herd (HR) that routinely received antibiotics. After a 20-d adjustment period in a common production facility, LR pigs on an antibiotic-free diet had lower proportions of anaerobes (27%) and coliforms (22%) that were resistant to 25 micrograms CTC/ml than did similarly fed HR pigs (81 and 48%, respectively). Continued maintenance of LR and HR pigs on the antibiotic-free diet in a common production facility tended to increase resistance in anaerobes and coliforms from LR pigs to levels comparable with those in HR pigs at the end of an 85-d feeding trial, but not after 14 d. Administration of CTC in the feed at therapeutic (220 micrograms/g for 14 d) and growth-promoting (27.5 micrograms/g for 85 d) levels markedly increased percentages of resistant anaerobes and coliforms in LR pigs, but not in HR pigs after 14 d. Similar antibiotic effects were not seen at the end of the 85-d trial. These data demonstrate that, while exposure to antibiotics in feed may increase resistance in intestinal populations, other factors such as environment, cross-contamination and herd history can also influence the observed level of antibiotic resistance in swine.  相似文献

The aim of this experiment was to study the effect of the application of pig slurry on potential pathogen (total and faecal coliform bacteria) presence in a Typic Xerofluvent soil. The experiment was conducted in field conditions, in the Segura River valley in the South East of Spain. Four treatments, in a fully-randomised design with three replicates per treatment, were established in experimental plots of 16 m2 each. Fertiliser treatments were: MF, mineral fertiliser (with a complex 16N–16P2O5–16K2O) equivalent to 150 kg N ha− 1; PSF, organic fertilisation with swine manure slurry, at two different rates (supplying 150 and 210 kg N ha− 1), and C, a controlled treatment without fertilisation. A high variability of the concentrations of total and faecal coliforms was observed during the experiment, mainly depends on the time and the treatment. The pig slurry amendment induced the highest initial and also persistent presence of total and faecal coliform bacteria. The higher application rate seemed to induce a long-lasting persistence of the total, but especially the faecal coliform, population in the amended soils compared to the lower rate. In most cases, an increase in the total coliforms content was observed in the soils amended with mineral fertiliser compared to controlled soils, probably due to an increase in soil microorganisms populations. In general, the total coliform bacteria was 2–3 log10 units higher than the faecal ones. Mostly, a tendency to decrease with time was observed in both indicator microorganisms, in all the treatments.  相似文献
