Present work was undertaken to screen some drought tolerant indigenous aromatic rice cultivars (IARCs), commonly cultivated in West Bengal, India, based on their capacity to produce osmolytes, redox-sensitive phenolic acids and flavonoids, as contrivances for redox-regulation under drought stress. Polyethylene glycol induced post imbibitional dehydration stress mediated changes in redox regulatory properties of the germinating seeds of the four IARCs (Jamainadu, Tulaipanji, Sitabhog, Badshabhog), which were assessed in terms of changes in prooxidant accumulation (in-situ localization of reactive oxygen species (ROS) by confocal microscopy, DCFDA (2′,7′-dichlorofluorescin diacetate) oxidation, O2 and H2O2 accumulation), cumulative antioxidative defense (radical scavenging property and total thiol content), ROS scavenging phenolic acids (gallic acid, protocatechuic acid, gentisic acid, para-hydroxy benzoic acid, chlorogenic acid, caffeic acid, syringic acid, salicylic acid, sinapic acid and p-coumaric acid) and flavonoids (catechin, naringin, rutin, quercetin, kaempferol, myricetin and apigenin). The capability of germinating seeds to accumulate osmolytes (like glycinebetaine, proline, soluble carbohydrates and K+ ion) and polyphenolic compounds was also correlated with their corresponding redox status and redox biomarkers (conjugated diene, hydroperoxide, thiobarbituric acid reactive substances and free carbonyl content) produced under the same conditions. The results in general showed that accumulation of osmolytes along with the redox-sensitive phenolics and flavonoids conferred the ability to maintain the redox homeostasis under drought stress for the tolerant IARCs (Badshabhog and Tulaipanji).  相似文献

The aim of this work was to evaluate the antioxidant potential of teas prepared from twenty-four commercially available berries and flowers of Sambucus nigra L. in relation to their phenolic profile, as reflected by the most representative phenolic acids (caffeic, chlorogenic, p-coumaric, ferulic, gallic and syringic acids); flavonols (quercetin, kaempferol, myricetin and rutin); and total phenolic (TPC), phenolic acid (TAC) and flavonoid (TFC) contents. The infusions prepared from elderflowers contained more abundant phenolic compounds than the elderberry infusions. The TPC of these infusions ranged from 19.81 to 23.90 mg of gallic acid equivalents/g dry weight of sample (GAE/g DW) for elderberries and from 15.23 to 35.57 mg GAE/g DW for elderflowers, whereas the TFC ranged from 2.60 to 4.49 mg of rutin equivalents/g dry weight of sample (RUTE/g DW) in elderberry infusions and from 5.27 to 13.19 mg RUTE/g DW in elderflower infusions. Among the phenolic compounds quantified in this study, quercetin (2.07–9.48 mg/g DW) and myricetin (1.17–9.62 mg/g DW) had the highest concentrations in the teas prepared from berries and flowers, respectively. Moreover, the antioxidant potential of elder infusions assessed by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical and ferric reducing antioxidant power (FRAP) assays revealed that the teas prepared from flowers had higher mean DPPH and FRAP activities than the teas prepared from berries. Therefore, elder beverages could be important dietary sources of natural antioxidants that contribute to the prevention of diseases caused by oxidative stress.  相似文献

The compositional characteristics of grape stalk in terms of lignin, sugars, ash, extractives and tannins contents were established. The biomass composition of grape stalk is 34% lignin, 36% cellulose, 24% hemicellulose and 6% tannins. The tannins, analysed by solution 13C NMR, were essentially of procyanidin type and the dichloromethane extractives fraction, characterized by GC-MS, was mainly composed of fatty acids. Different processes for the pretreatment of grape stalk were compared. Due to the high content of lignin and tannins, dilute sulfuric acid and ethanol organosolv pretreatments resulted in a low degree of delignification. However, a wet oxidation process permitted a significant reduction of the Klason lignin content of the pulp with a good carbohydrate recovery and improved enzymatic digestibility.  相似文献

Hybanthus parviflorus is a perennial shrub, widely distributed in the tropical and subtropical regions of America, and is known as ‘violetilla’ in Argentina. Previous phytochemical studies of this species led us to the isolation and determination of the primary structure of a novel macrocyclic polypeptide, the cyclotide hypa A. Here the insecticidal activities of extracts of H. parviflorus were determined against Ceratitis capitata Wied., the Mediterranean fruit fly or ‘Medfly’. Mortality in the different life stages of Medfly, total mortality and modifications of the insect’s physiology caused by 50% EtOH and CH2Cl2 extracts of H. parviflorus were evaluated. In addition, we determined the occurrence of ursolic acid, β-sitosterol and the polyphenols quercetin, quercetin-3-methyl ether, apigenin, luteolin, kaempferol, rutin, caffeic acid and chlorogenic acid. The promising insecticidal activity of 50% EtOH extracts of H. parviflorus and its purified fractions was related to the presence of cyclotides. The insecticidal activity of CH2Cl2 extracts could be related to the presence of polyphenols, ursolic acid and β-sitosterol.  相似文献

Traditional healthy beverages in several African counties were home-made by water extraction of flowers of Delonix regia (Boj.) Raf. This tree belongs to the local biodiversity of plants used in the local medicine. It is also part of the traditional bioproducts marketed within African village communities. Scaling-up at pilot plant level, by mimicking this local recipe, lead to concentrated bioproducts containing various natural phenolic compounds, most of them have never been yet published. A three-steps process was used to prepare concentrated of water-extracted soluble phenolics. This process included: phenol extraction of dried plant by overnight diffusion in acidified water, clarification then concentration of the macerate, using membrane technology. Total phenol contents were determined as gallic acid equivalents using the Folin-Ciocalteu method. Total flavonoid contents, as delphinidin equivalents, were determined using the UV-vis colorimetric method. Comparison of concentration factors of both phenol families (phenolic acids and flavonols) contained in the 3 co-products obtained during the process were made between crude extracts, cross-flow microfiltration extracts and reverse osmosis concentrated extracts. Individual phenol compounds found in the products obtained were characterized and quantified by HPLC-DAD and by HPLC-MS2. Among the 11 phenol compounds identified in the water-extracts, 3 have been already identified in the literature (2 phenolic acids and 1 flavonol), and 8 new compounds, in noticeable amounts, have never been cited. We have confirmed the presence of phenolic acids such as 3,4,5-trihydroxybenzoic (gallic acid), 3,4-dihydroxybenzoic (protocatechuic acid) and of quercetin. We have characterized for the first time the presence of 2-hydroxy 5-[(3,4,5 trihydroxyphenyl) carbonyl oxy] benzoic acid and of 7 other flavonols belonging to the quercetin, kaempferol and isorhamnetol compound families. Using both LCMS and NMR analyses, they have been identified and quantified as: rutin, quercetin 3-O-glucoside, quercetin 3-O-galactoside, quercetin trihexoside, quercetin 3-O-robinobioside, kaempferol rhamnosylhexoside and isorhamnetol rhamnosyl-hexoside.  相似文献

Chiltepin, a wild chili mostly used in different traditional foods and traditional medicine in Northwest Mexico, represents a source of polyphenols. However, studies about the bioaccessibility of polyphenols as a parameter to measure the nutritional quality and bioefficacy of them in the fruit after consumption are scarce. Chiltepin showed phenolic acids and flavonoids contents between 387 and 65 μg/g, respectively. Nevertheless, these values decreased after the digestion process. Before digestion, gallic acid, 4-hydroxibenzoinc acid, chlorogenic acid, caffeic acid, p-coumaric acid, quercetin and luteolin were the main polyphenols found in chiltepin by HPLC-DAD and confirmed by FIA-ESI-IT-MS/MS. Gallic and chlorogenic acids were non-detected in the gastric phase, while only p-coumaric acid (5.35?±?3.89 μg/g), quercetin (5.91?±?0.92 μg/g) and luteolin (2.86?±?0.62 μg/g) were found in the intestinal phase. The bioaccessibility of phenolic acids, flavonoids, and total polyphenols after the intestinal phase was around 24, 17 and 23%, respectively. Overall, results indicated that release of polyphenols from chiltepin fruit might be affected by the food matrix and gastrointestinal conditions due to the low bioaccessibility values observed.  相似文献

为了探究酶制剂对苎麻混合青贮品质的影响,本试验在苎麻与麦麸(80%苎麻+18%麦麸+2%蔗糖)混合青贮中分别添加10 mL蒸馏水(对照组)、纤维素酶2 g/kg、纤维素酶2 g/kg+半纤维素酶0.4 g/kg、纤维素酶2 g/kg+半纤维素酶0.4 g/kg+果胶酶0.7 g/kg,30天后开包取样分析各项指标。结果表明,苎麻与麦麸混合青贮发酵品质和营养品质均较好。与对照相比,添加酶制剂显著提高了碳水化合物、干物质和粗蛋白含量(P<0.05),显著降低了中性洗涤纤维、酸性洗涤纤维、粗纤维的含量和氨态氮/总氮比值(P<0.05),但是对pH值、乳酸、乙酸和丙酸含量无显著影响(P<0.05)。本试验条件下,添加酶制剂能够有效提高苎麻混合青贮的品质,单独添加纤维素酶与复合酶制剂无显著性差异,从节约成本角度考虑,单独添加纤维素酶组为最佳选择。  相似文献

建立同时测定槟榔壳中6种酚类物质的高效毛细管电泳方法,研究分析不同浓度和不同pH值缓冲液对10种标准品(即:表儿茶素、儿茶素、柚皮素、芦丁、山奈素、阿魏酸、绿原酸、香豆酸、咖啡酸和没食子酸)的分离效果。结果显示,最佳电解缓冲液为0.1 mol/L、pH 9.0硼酸缓冲液,紫外检测波长为280 nm,分离电压为20 kV。该方法简便快速,能在20 min之内将10种酚类物质完全分离开,具有良好的精密度、回收率和线性关系。检测限范围为0.587 5~4.415 2μg/mL。  相似文献

We investigated the change in seed shape and weight as well as the accumulation and distribution of anthocyanins, melanin, and flavonoids during tartary buckwheat seed development. Pericarp dry weight increased slowly while the increase in flour dry weight was much greater. Increases in the dry weight of the embryo and endosperm were associated with change in seed shape. The highest anthocyanin content was observed in stage two while the highest melanin content occurred in stage five. There was a considerable decrease in total flavonoids, rutin, quercetin and kaempferol in mature seeds relative to seeds that were still developing. Additionally, there were significant correlations (correlation coefficient > .900, p < .05) among the changes in rutin, quercetin and kaempferol between flour and the pericarp. In mature seeds, 95% of the flavonoids were present in flour.  相似文献

Growth characteristics, dry‐matter (DM) yield, chemical components and in vitro dry‐matter digestibility (IVDMD) of Napier grass were studied in a randomized complete block design with three replications at Haramaya University, Ethiopia. The treatments were three defoliation frequencies (60, 90 and 120 d after Napier grass establishment) and five cutting heights (5, 10, 15, 20 and 25 cm above ground level). Except for leaf‐to‐stem ratio, all growth characteristics increased with decreasing frequency of defoliation. Similarly, DM yield of Napier grass increased as the frequency of defoliation decreased. Total ash, crude protein (CP), hemicellulose and IVDMD contents of Napier grass increased with increasing frequency of defoliation, whereas DM, acid detergent fibre, acid detergent lignin and cellulose contents increased with a decrease in defoliation frequency. In contrast, cutting heights had no significant effect on most chemical components and IVDMD contents, although total ash and CP decreased as the height of cutting increased. The CP yield per cut and per day was highest at an intermediate defoliation frequency. The results of this study indicated that Napier grass could be categorized under medium‐ to high‐quality herbage if defoliated at 90 d after establishment in the mid and highlands of Ethiopia.  相似文献

Lucerne populations that had previously been divergently selected for acid detergent lignin (ADL) concentrations of whole herbage were additionally divergently selected for in vitro dry-matter disappearance (IVDMD) of basal stems. Parental plants of the four selected populations were intercrossed and the half-sib progeny evaluated in a replicated field trial in 1988 over two harvests. Cell wall composition of basal stems was determined for detergent fibre components and neutral sugars, uronic acids, Klason lignin and esterified and etherified phenolic acids of the total fibre fraction. Lignin polymer composition was measured by nitrobenzene oxidation. Degradability of cell wall polysaccharide components was determined by a 48-h in vitro ruminal fermentation. Cell wall composition was changed by both selection criteria. Klason lignin proportion of the total fibre did not differ as a result of selection, whereas the ADL concentration of neutral detergent fibre was different among selection groups. The cell wall polysaccharides of lucerne basal stems shifted towards more cellulose (glucose residues) and less hemicellulose (xylose residues) with selection for either low ADL or high IVDMD. While degradability of the cell wall polysaccharides was correlated with various measures of lignin composition and concentration, the results were variable and inconclusive. Surprisingly, Kiason lignin and ADL were positively correlated with cell wall polysaccharide degradability, and esterified ferulic acid was negatively correlated with degradation of the cell wall. Selection for herbage ADL and basal stem IVDMD of lucerne did alter cell wall degradation, but the associated changes in cell wall lignification were not consistently correlated with cell wall degradability.  相似文献

Digestibility of fiber components namely neutral detergent fiber (total content of cellwall) cellulose, hemicellulose and lignin are estimated in 14 healthy vegetarian men during adlibitum feeding and at 3 energy levels namely 2526, 2868 and 3290 kcals/day. Values of digestibility for adlibitum experiments were 34.17±2.3 for neutral detergent fiber (NDF), 30.1±3.9 for cellulose and 53.4±3.0 for hemicellulose and 8.1±2.6 for lignin. Neutral detergent fibre (NDF), acid detergent fibre and water-soluble carbohydrates (WSC) were assayed in wet and dry samples of maize silage, either treated with NaOH or untreated (control), before and after ensiling.
Statistically significant effects of drying at 60°C were observed in WSC, NDF and cellulose estimations expressed on the basis of 105°C dry matter.
It could therefore be preferable to analyse wet samples, as these are not altered by the degradations induced by oven-drying and are more representative of the original condition of the forage.  相似文献

There is no data on the lipid profile of Fagopyrum tataricum grain and its milling products. Therefore, we aimed to assess the phytosterol and tocopherol content and fatty acid profile of the samples of F. tataricum. Grain was milled, then the milling product separated to hull, bran, and two flour fractions, differing in particle size. The highest level of lipids (total fatty acids – 83%, with dominant oleic acid – 40%) was observed in the bran fraction (380 μm > GA > 180 μm). All samples contained some natural cis fatty acids, such as vaccenic acid (C18:1 n-7; ~2.8%) and tocopherol (α-, β-, δ- and γ-tocopherol). The highest content of total tocopherol was also detected in the bran fraction (0.1% of lipids). The content of lipid soluble bioactive substances was different and depended on the size of obtained fraction, therefore properly designed grain processing technology can be used to produce food with high nutritional value.  相似文献

There is limited information on the effects of the increase in the density of shrubs on herbage production and nutritive value of natural grasslands in the Mediterranean region, currently facing major land use changes. Herbage production of drymatter (herbaceous fractions, of plant functional groups and by species), crude protein (CP), neutral‐detergent fibre (NDF), acid‐detergent fibre (ADF), acid‐detergent lignin (ADL) and hemicellulose concentrations and in vitro organic matter digestibility were determined at the time of peak of annual growth across four types of grassland vegetation each characterized by different shrub cover regimes. A sharp reduction in herbage production and a reduction in nutritive value were found as a result of the increase in shrub cover. These changes appeared to be closely related to the shift in plant functional groups detected as shrub density increased. Herbage production from grasses and legumes was found to be more sensitive to shrub cover changes than herbage production from forbs, whereas, as grassland types became denser, annual species were gradually replaced by perennials and C4 grasses by C3 ones. The impact of shrub encroachment on Mediterranean grasslands is discussed in relation to their use by livestock.  相似文献

??Pichi?? or ??pichi romero?? (Fabiana imbricata R. et. P., Solanaceae) is a Chilean plant used as a tea in the Andean regions of Chile and Argentina. A very simple and direct method was developed for the qualitative analysis of polyphenols in the tea by high-performance liquid chromatography (HPLC) with diode array detection and electrospray ionization tandem mass spectrometry. The phenolic constituents identified in the teas were chlorogenic acid (3-O-caffeoylquinic acid), p-hydroxyacetophenone, scopoletin and quercetin derivatives. The glycosides were mainly glucosides from p-hydroxyacetophenone and scopoletin while di- and tri-glycosides from quercetin were the main flavonoids. The content of the main phenolic compounds in the teas (g/100?g lyophilized infusion) was 0.8?C1.9?% for scopoletin, 0.4?C6.2?% for p-hydroxyacetophenone and 2.1?C4.3?% for rutin, respectively. The health-promoting properties reported for this herbal tea can be associated with the presence of several phenolics with known antioxidant, diuretic and antiinflammatory activity.  相似文献

Summary The phenolic acids benzoic, caffeic, chlorogenic, ferulic, p-coumaric, protocatechuic, salicylic, sinapic, syringic and vanillic together with vanillin, were tested for their ability to inhibit polygalacturonic acid lyase (PL) and polygalacturonase (PG) in culture filtrates ofErwinia carotovora subsp.carotovora. None of the compounds inhibited PL at 200 μg/ml, although syringic and sinapic acids caused a 54% and 43% reduction respectively at 400 μg/ml. PG was strongly inhibited by caffeic acid (57% inhibition at 100 μg/ml) and chlorogenic acid caused slight inhibition. Little if any inhibition of PG was caused by benzoic, ferulic, p-coumaric, protocatechuic, salicylic, sinapic, syringic, and vanillic acids or vanillin. The percentage inhibition of PG by chlorogenic acid was not affected by pH values over the range 4–7.  相似文献

The contents of total phenolic compounds, flavonoids, and phenolic acids were determined in selected garlic cultivars grown at four locations. The total phenolic content varied from 3.4 mg gallic acid equivalents (GAE)/g of dry matter (dm) to 10.8 mg GAE/g of dm with a mean value of 6.5 mg GAE/g of dm. The myricetin, quercetin, kaempferol, and apigenin flavonoids were not detected in any of the samples. Caffeic acid and ferulic acid were the major phenolic acids found with mean values of 2.9 mg/kg of dm and 2.6 mg/kg of dm, respectively. The mean contents of vanillic, p-hydroxybenzoic, and p-coumaric acids were comparable (0.4–0.8 mg/kg of dm), and the level of sinapic acid was negligible (< 0.1 mg/kg of dm). There was a significant effect of location but an insignificant effect of genotype on contents of caffeic, vanillic, p-hydroxybenzoic, and p-coumaric acids. However, genotype but not location affected the contents of total phenolics and ferulic acid. On average, the white garlic cultivars and Chinese garlic cultivars contained higher contents of total phenolics and ferulic acid than the purple garlic cultivars. However, the differences in the total phenolic content between the purple and white garlic cultivars were not significant.  相似文献

The impact of tropical seasons (dry and wet) and growth stages (8, 10 and 12 weeks) of Cosmos caudatus on the antioxidant activity (AA), total phenolic content (TPC) as well as the level of bioactive compounds were evaluated using high performance liquid chromatography (HPLC). The plant morphology (plant height) also showed variation between the two seasons. Samples planted from June to August (during the dry season) exhibited a remarkably higher bioactivity and height than those planted from October to December (during the wet season). The samples that were harvested at eight weeks of age during the dry season showed the highest bioactivity with values of 26.04 g GAE/100 g and 22.1 μg/ml for TPC and IC50, respectively. Identification of phytochemical constituents in the C. caudatus extract was carried out by liquid chromatography coupled with diode array detection and electrospray tandem mass (LC-DAD-ESIMS/MS) technique and the confirmation of constituents was achieved by comparison with literature data and/or co-chromatography with authentic standards. Six compounds were indentified including quercetin 3-O-rhamnoside, quercetin 3-O-glucoside, rutin, quercetin 3-O-arabinofuranoside, quercetin 3-O-galactoside and chlorogenic acid.
