水稻白叶枯病菌dsp基因的克隆,同源性和功能分析   总被引:1,自引:0,他引:1  
以经转座子Tn5诱变的水稻白叶枯病菌dsp基因突变体XooM3104为材料,构建了dsp基因探针质粒pVIR3。采用分子杂交法,从基因文库克隆中筛选出1个dsp基因克隆pNAX3111,含22kb的外源DNA插入片段,具有4个KpnI酶切位点,其中3.0,2.0kb两个片段与探针质粒中Tn5侧翼的dsp基因序列同源。以pNAX3111为探针与Xoo日本系统、中国系统及菲律宾系统的菌株,细菌性条斑病  相似文献

利用转座子Tn5gusA5上的抗性基因,通过“鸟枪法”克隆到用这种转座子诱变甘蓝黑腐病黄单胞菌所获得的三个胞外多糖突变体菌株T106、T113和T117中的转座子及其相邻序列的DNA片段,并利用含有这些片段的重组质粒,通过标记置换构建了突变体。通过检测标记置换突变体的胞外多糖产量及Southern杂交,证实了胞外多糖突变株T106、T113和T117不仅确系转座子插入所致,而且转座子插入的位点分布在染色体的不同区域。从而证实了我们所报道的诱变体系是可行的  相似文献

变铅青链霉菌DNA存在一种异常的位点特异性修饰,使其在含有Fe^2+的电泳缓冲液中电泳时遭到双链切割。用NTG诱变变铅青链霉菌JT46,获得了一株修饰缺陷突变株,命名为ZX1。该突变菌株除了丧失修饰其DNA的能力以外(亦即其DNA在含有Fe^2+的电泳缓冲液中电泳时不被降解),在某些基因[如黑色素基因(mel)和某些抗生素抗性基因]的表达方面也发生了变化,同时还丧失了产孢能力和对唾噬体ΦHAU3的  相似文献

利用高盐法从鸡血中提取高分子量DNA,以λEMBL3为载体构建了鸡染色体文库,用鸡生长激素受体cDNA基因的部分序列作探针,从文库中筛选得到3个阳性克隆。通过对阳性克隆I插入片段的酶切和Southern杂交分析,表明它含有全长的鸡生长激素受体基因,建立了该基因的限制性酶切图谱。将插入片段亚克隆到pGEM-7Zf(+)质粒上,对含有基因5’端序列的2.5kbEcoRI酶切片段进行了序列分析。  相似文献

根据E.coli基因组rDNA保守序列设计的两对引物,分别用于扩增30株来源于我国不同地区的快生型大豆根瘤菌菌株。扩增产物经几种较少识别序列的内切酶消化后,得出特征的限制性内切酶酶切图谱。该图谱可用于供试菌株种水平的鉴定及菌株rDNA分子的遗传多样性研究。选用来源于E.coli基因组16SrDNA保守序列的一对引物将所有供试菌株都扩增出一条1.5kb的带,经四种内切酶消化后发现:新疆地区来源的菌株  相似文献

应用广谱寄主载体pLAFR1,在大肠杆菌中建立了青枯假单胞菌Pseu-domonas solanacearum两个不同生理小种T2003(在马铃薯上高发病,在花生上不致病)和T2005(在花生上高发病)的基因文库。把菌株T2005基因文库的克隆转移至T2003受体中,结果得到两个能引起花生枯死的结合子,这表明通过导入菌株T2005文库中的两个克隆,使原来在花生上不致病的菌株T2003的寄主范围扩大到在花生上能致病。  相似文献

由革兰氏阴性细菌水稻白叶枯病菌引起的水稻白叶枯病是亚洲、北美以及非洲部分地区最严重的水稻病害之一,水稻白叶枯病可使水稻减产高达50%以上。研究表明水稻白叶枯病菌的毒力主要依靠三型分泌系统所分泌的效应物。为了解水稻白叶枯病菌广西菌株GX1329中含有avrBs3/pthA家族基因的情况,本研究应用Alu I部分酶切其基因组DNA,构建了含有736个克隆的菌株GX1329的基因组文库。BamHI酶切分析随机挑取的15个文库克隆表明,克隆的外源DNA随机性良好,克隆的最小片段为27.7kb,最大为58.5kb,平均大小为39.9kb,文库克隆容量约为2.8×10^3Mb,该文库中包含基因组中任一个基因的概率为99.4%。利用来自水稻白叶枯病菌菲律宾菌株PX086的无毒基因avrXa10的第252位~第486位核苷酸序列作为探针,通过菌落原位杂交从GX1329基因组文库中筛选到37个含avrBs3/pthA家族基因的克隆。再通过Southern杂交分析,得到了17个独立克隆。这17个克隆中至少含有13个不同的avrBs3/pthA家族基因。这些基因在GX1329基因组中有的单独存在,有的两个或两个以上串联存在。本工作基本上明确了菌株GX1329基因组中avrBs3/pthA家族基因的数量,为进一步研究菌株GX1329中avrBs3/pthA家族基因的功能奠定了基础。  相似文献

不同石油污染程度土壤细菌群落多样性及优势菌群分析   总被引:1,自引:0,他引:1  
  目的  探究辽河油田不同石油污染程度土壤中理化性质及细菌群落多样性和组成的变化规律,并对石油污染土壤中的石油降解菌进行分离培养和鉴定。  方法  采集了辽河油田不同石油污染程度土壤,采用高通量测序技术和化学分析法对土壤细菌群落组成和土壤理化性质进行测定,并进一步筛选出石油降解菌株。  结果  在出油口(A)、距离出油口50 m(B)和距离出油口150 m(C)采集的三个土壤样品,其土壤总石油烃含量分别为2467.44 mg kg?1、884.99 mg kg?1和141.63 mg kg?1,三个土壤样品具有不同的石油污染程度。石油污染显著提高了土壤总有机碳含量,土壤总石油烃含量与总有机碳含量呈现正相关(P < 0.001)。土壤细菌群落多样性和丰富度指数与土壤石油烃的浓度呈显著负相关(P < 0.01)。不同石油污染程度土壤具有不同的细菌群落组成和结构,土壤石油烃含量是影响细菌群落变化的主要因素。出油口石油污染土壤样品(A)中,变形菌门(Proteobacteria)为优势菌门,假单胞菌属(Pseudomonas)、假黄单胞菌属(Pseudoxanthomonas)、博代氏杆菌属(Bordetella)和伯克氏菌属(Burkholderia)为优势菌属。从出油口石油污染土壤(A)中分离出3株石油降解菌株,通过16S rRNA基因测序分别被鉴定为Pseudomonas baetica、黄褐假单胞菌(Pseudomonas fulva)和施氏假单胞菌(Pseudomonas stutzeri),其石油降解率分别为37.2%、46.9%和57.8%。此结果与A样品高通量测序属水平组成分析相吻合,表明石油污染能够选择性富集土壤中具有石油降解能力的假单胞菌属。  结论  石油污染提高了土壤总有机碳含量,降低了土壤细菌群落多样性,富集了具有烃类降解能力的优势菌属,是造成土壤细菌群落组成和结构改变的主要因素,并筛选出具有潜在开发应用价值的石油降解假单胞菌株。  相似文献

为了构建一套高效率的污水处理体系,从污水样品中分离、筛选出一株对动植物油脂均具有较强降解能力的菌株jzz2。在生理生化特性和16srDNA序列系统发育分析的基础上,评价了该菌株对不同类型动植物油脂的降解能力,同时对其脂肪酶和生物表面活性剂活性进行了分析。研究发现,该菌株属于Pseudomonas aeruginosa,在含1%油脂的MS培养基中,30℃培养24h的Pseudomonas aerugilzosaJZZ2对橄榄油、色拉油、芝麻油和牛油的降解率分别为72.4%、90.2%、82.3%和84.5%,同样条件下培养48h的Pseudomonas cleruginosaJZZ2脂肪酶活性为2200U·L^-1,研究进一步发现油脂的添加能够促使其生物表面活性物质的产生。由此可见,Pseudornonas tieruginosaJZZ2对动植物油脂均具有较强的降解能力,能生成脂肪酶和生物表面活性物质是其具有油脂降解特性的主要原因之一。  相似文献

Bacteriophages (phages) are the most abundant biological entities on the planet and are important as the greatest genomic reservoirs in both marine and terrestrial environments. In this study, we analysed T4-type phage communities in an upland black soil by monitoring g23 clones in DNA extracted from seasonal soil samples with no fertilizer, chemical fertilizers, chemical fertilizers plus manure, and natural restoration treatments. PCR products with degenerate primers MZIA1bis and MZIA6 were subjected to denaturing gradient gel electrophoresis. In total, 46 clones with different g23 sequences were obtained. Phylogenetic analyses indicated that T4-type phage communities in the upland black soil were distinctly different from those in marine environments and in an Antarctic lake, which strongly suggested that T4-type phage communities in soil differed from those in aquatic environments. Among 46 clones, 18 clones formed clusters with the clones from rice field soils, 14 clones formed three new clusters, and 13 clones were left as ungrouped, which indicated that T4-type phage communities in the upland black soil were relatively similar to those in rice field soils but that specific communities also inhabit in the upland black soil exclusively.  相似文献

Summary We investigated the reliability of antibiotic resistance and colony morphology of clones of a Bradyrhizobium sp. (Lotus) strain for strain identification in nodulation competitiveness experiments in soil. There was no difference in nodulation competitiveness between the wild type strain and each of five mutants resistant to streptomycin and spectinomycin at the time of their isolation from antibiotic-containing media. However, these mutants were significantly less competitive when tested 4 months later. The apparent instability of the newly isolated mutants and their subsequently decreased nodulation competitiveness show that mutants must be examined carefully after being allowed time to stabilize. Two clones of the Bradyrhizobium sp. (Lotus) strain that differed in colony morphology on yeast mannitol medium did not differ in antigenic properties, whole cell protein electrophoresis profiles, mean cell generation times in yeast mannitol medium, N2-fixing ability, nodulation of Lotus pedunculatus in growth pouches, or in nodulation competitiveness. Both clones retained their colony morphology after numerous transfers on yeast mannitol agar over 3 years and after at least 6 months in soil. A limiting factor, which may restrict the use of colony morphology as a marker for strain identification in competition experiments, is the problem of detecting double-infected nodules when the small colony type comprises a relatively small portion of the total nodule population.  相似文献

从西瓜根围分离得到的一株有效抑制西瓜枯萎病菌的枯草芽孢杆菌B 11菌株(B acillus subtilisstra in B 11)。为获得高产拮抗物质的诱变菌株,采用紫外线诱变方法对B 11菌株进行诱变,结果获得抑菌活性比野生菌B 11菌株强的6株诱变菌株,其中G 17菌株的抑菌活性及粗拮抗物质含量均高于B 11菌株,且遗传性较稳定;再以G 17菌株为供试菌株,用亚硝基胍(NTG)对其进行化学诱变,结果获得抑菌活性较强的4株诱变菌株,其中BV 22菌株对西瓜枯萎病菌的抑菌活性最高,其抑菌活性比G 17和B 11菌株分别提高了36.88%和80.00%;其粗拮抗物质的含量分别提高了30.15%和62.53%。研究结果说明,用物理诱变B 11菌株后获得的诱变菌株再用化学方法进行诱变,可选育出高产拮抗物质的诱变菌株。  相似文献

采用紫外线和氯化锂诱变 ,从甲基对硫磷降解菌假单胞菌 (Pseudomonassp.)DLL -1中获取高效突变株DLL -E1 ,DLL -E2 ,DLL -E3 ,DLL -E4。并对其降解甲基对硫磷性能、降解对硝基苯酚性能、底物广谱性及胞外酶及粗酶液活性等生物学功能进行了研究。与出发菌株相比 ,上述性能均有所提高。选择DLL -E4作为进一步研究的对象 ,详细研究了其对甲基对硫磷及对硝基苯酚的耐受和降解情况。高效菌株的选育为农药残留降解的酶学研究打下了基础。  相似文献

采用PCR方法从绿脓杆菌ATCC27853菌株基因组扩增其外毒素A(PEA)全长编码基因,并将PEA基因插入到pcDNA3.1A真核表达载体中,构建成pcDNA3.1/ PEA质粒表达载体。经磷酸钙介导将重组质粒转染 HEK293T 细胞进行表达。结果表明本文克隆的PEA基因与标准菌株PA103碱基序列基本相似,同源性达99%。表达产物经蛋白免疫印迹法(Western-blot)检测,表明本文构建的含有原基因信号肽的PEA基因能够在真核细胞进行分泌性表达,这为基于PEA的免疫毒素、疫苗佐剂和疫苗载体的研究奠定了基础。  相似文献

Wheat lines with reduced amylose content were recently produced by single and double mutation from a low‐amylose line, Kanto 107. They are appropriate for clarifying the influence of amylose content on starch gel properties because of their similar genetic background. When measured using the concanavalin A method (ConA), the total amylose content of isolated starches from Kanto 107 and three mutants (K107Afpp4, Tanikei A6599‐4, K107Wx2) was 24.8, 18.5, 7.1, and 1.7%, respectively. Results of differential scanning calorimetry (DSC) showed that the difference in amylose content strongly affected gelatinization conclusion temperature and enthalpy. We prepared 30 and 40% starch gels and measured their dynamic shear viscoelasticity using a rheometer with parallel plate geometry. Compressive and creep‐recovery tests were conducted under uniaxial compression. The storage shear modulus correlated highly with the amylose content of starch in 30 and 40% starch gels. The creep‐recovery test showed a clear distinction in creep curves among starch samples. When the compressive force required for 50, 80, and 95% strains was compared, starch gels with lower amylose content showed lower compressive force at 50% strain. Waxy starch gel (K107Wx2) showed higher compressive force at strain >80% than other samples due to its sticky property.  相似文献
