Sainfoin (Onobrychis viciifolia Scop.) is a perennial forage legume that possesses beneficial properties in the context of sustainable agriculture. In order to initiate a pre-breeding programme, we have assembled a germplasm collection of O. viciifolia from many geographic regions, as well as a collection of Onobrychis species that might be crossed with O. viciifolia to improve its biological and agronomic properties. The objective of this study was to obtain DNA sequences from a representative sample of these accessions in order to characterise their genetic diversity and to assess the robustness of the Onobrychis taxonomy. Because of potential problems with copurification of tannins and polyphenols from Onobrychis leaf tissue, three methods for DNA extraction were assessed and the most appropriate one identified. DNA sequences were obtained for the nuclear internal transcribed spacer region and the trnH-psbA and trnT-trnL intergenic spacers of the chloroplast genome. Neighbour joining trees were constructed and accessions were assigned to operational taxonomic units. The results indicated that there is substantial genetic diversity among Onobrychis species. Redundancies in species nomenclature were identified, as well as possible overlap between some of the sections into which the Onobrychis genus has been divided. A genetic distinction was apparent between O. viciifolia accessions from western Europe and those from eastern Europe and Asia, reflecting a similar division based on agronomic properties. Recognition of these different though related germplasms will be valuable in the design of breeding programmes for the rational improvement of sainfoin as a forage crop for sustainable agriculture.  相似文献

The high cost and supply shortage of seed yam propagules for planting are major constraints in yam production. In the water yam (Dioscorea alata L.), aerial tubers have potential as alternative sources of planting material. In this study, we investigated morphological, molecular and ploidy variation across multiple aerial tuber producing accessions of Dioscorea alata. Initial screening of over 800 accessions from the International Institute of Tropical Agriculture germplasm collection for aerial tuber production identified a subset (15 %) of accessions, which produced aerial tubers. The aerial tuber producing accessions (along with 18 non-aerial tuber accessions) were further characterized for phenotypic and ploidy variation. In addition, using SSR markers we characterized the genetic diversity amongst all of the aerial tuber producing accessions, along with six non-aerial tuber producing accessions. Multiple Correspondence Analysis (MCA) using morphological data grouped the accessions according to their aerial tuber production. The aerial tuber production characteristics of accessions were associated with phenotypic variables and ploidy levels. The MCA analysis revealed three main groups consisting of; Group 1) all non aerial tuber producing accessions (n = 15), hastate leaf shape, less or no anthocyanin pigmentation and diploid (2n = 2x = 40), Group 2) group with some aerial tuber producing accessions, different extent of anthocyanin pigmentation, sagittate leaf shape, mainly diploid (n = 44) and three triploid (2n = 3x = 60) and 3) a group where all individuals bear aerial tuber, cordate leaf shape, intermediate anthocyanin pigmentation and majority (n = 74) tetraploid (2n = 4x = 80) and three triploid individuals. Aerial tuber production may be subject to a genome dosage effect as an increase in aerial tuber production was associated with increased ploidy level. For instance, tetraploid plants produce more aerial tubers per sprout than either triploids or diploids. Principal coordinate analysis based on SSR markers using Jaccard’s coefficient also revealed distinct groups associated with the pattern of aerial tuber formation, leaf shape and anthocyanin pigmentation. Overall our study indicates the usefulness of combining SSR markers, ploidy level and phenotypic data for identification and classification of Dioscorea alata accessions according to their extent of aerial tuber production.  相似文献

Lemon balm (Melissa officinalis L.) is a well-known aromatic and medicinal plant. Because of its proven sedative, spasmolytic and antiviral effects, it is often used in watery or alcoholic extracts for self-medication or pharmaceutical purposes. Its therapeutic effect is due to the content of essential oil and rosmarinic acid. A set of 28 balm accessions was evaluated for the variability of essential oil content and composition and the content of rosmarinic acid. For analysis of secondary metabolites distillation, gas chromatography and high performance liquid chromatography was used. The content of essential oil in the first cut of the years 2009 and 2010 varied in this study in a range between 0.02 and 0.48% and in the second cut between 0.01 and 0.72%. The rosmarinic acid content of the collection ranged in the year 2009 for the first cut from 3.99 to 8.78% and in the second cut from 2.45 to 6.78%. Ploidy was determined for all accessions and two cytotypes were found: diploid 2n = 2x = 32 (15 accessions), tetraploid 2n = 4x = 64 (13 accessions). Via statistical analyses two chemotypes of essential oil were found: chemotype citral and chemotype germacrene D.  相似文献

A new synthetic species of Momordica (2n = 56) has been developed by crossing natural tetraploid Momordica subangulata subsp. renigera (2n = 56) with induced tetraploid Momordica dioica (2n = 4x = 56). The hybrid produces adventitious root tubers through which it perpetuates and propagates like its female parent and maintains its morphological characteristics in the progeny. The hybrid is naturally fertile and has the superior agronomic traits of both parents making it a good choice as a new vegetable crop. The new species is named Momordica × suboica Bharathi and described.  相似文献

A perennial version of grain sorghum [S. bicolor (L.) Moench] would create opportunities for greatly reducing tillage and preventing soil degradation. Efforts to select for perenniality and grain production among progeny of hybrids between S. bicolor (2n = 20) and the weedy tetraploid perennial S. halepense (L.) Pers. (2n = 40) are complicated in that F1 hybrids produced by diploid × tetraploid sorghum crosses are usually tetraploid. In 2013, a set of random pollinations between 19 diploid cytoplasmic male-sterile inbred lines and 43 tetraploid perennial plants produced 165 F1 hybrid plants, more than 75% of which had highly atypical plant, panicle, and seed phenotypes. Phenotypic segregation in F2 populations derived from atypical hybrids was also anomalous. Examination of mitotic metaphase cells in F1 or F2 root tips revealed that 129 of the 165 hybrids were diploid. Parentage of the diploid progenies was confirmed using simple-sequence repeat analysis. The mechanism by which diploid hybrids arise from diploid × tetraploid crosses is unknown, but it may involve either production of monohaploid (n = 10) pollen by the tetraploid parent or chromosome elimination during early cell divisions following formation of the triploid zygote. The ability to produce diploid germplasm segregating for S. bicolor and S. halepense alleles could have great utility, both for the development of perennial sorghum and for the improvement of conventional grain sorghum.  相似文献

Diospyros kaki Thunb. originated in Eastern Asia, as evidenced of its culture in China as early as several centuries B.C. In the seventh century, persimmon was introduced to Japan and later, in the fourteenth century to Korea. There is no information about persimmon culture in Europe until the seventeenth century, with the spread over the world occurring in the eighteenth century. The genus Diospyros contains more than 400 species, with levels of ploidy ranging from diploid (2n = 2x = 30) up to nonaploid (2n = 9x = 135). The primary economic crop species is Diospyros kaki Thunb., which is mainly hexaploid (2n = 6x = 90) and includes hundreds of cultivars. Although a relatively recent introduction in Europe, the species has adapted well, and the genetic diversity have been expanded with culture and selection for the past 200 years in the Mediterranean basin. These locally adapted cultivars were evaluated with cultivars from Asian origin in a germplasm collection established at IVIA in Valencia, Spain. In this paper 27 cultivars from the IVIA collection were studied by multivariate analysis, and 37 variables were analyzed using a Principal Components Analysis and cluster analysis following the method UPGMA. Studies on correlations and significance among variables identified the most relevant ones, and thus provided information for a future core collection.  相似文献

A set of 68 M. officinalis genotypes was evaluated for content and composition of essential oil and the content of rosmarinic acid. For all genotypes the level of ploidy was determined. The 68 genotypes were clone plants grown and evaluated for two years at Quedlinburg. For analysis of secondary metabolites distillation, gas chromatography and high performance liquid chromatography was used. The content of essential oil varied in this study in ranges from 0.03 to 0.33% for the second cut 2010 and 0.01–0.35% for the second cut 2011. The rosmarinic acid content ranged in the year 2010 from 3.67 to 7.55% and in the year 2011 from 4.92 to 8.07%. Via statistical analyses two chemotypes of essential oil were found: chemotype citral and chemotype β-caryophyllene oxide. Ploidy was determined for all genotypes and two cytotypes were found: diploid 2n = 2x = 32 (62 of 68 genotypes) and triploid 2n = 3x = 48 (6 of 68 genotypes).  相似文献

This study aims to explore the potential of Dioscorea nummularia Lam. and to gather information as a prelude to its improvement by plant breeding. After a taxonomic clarification and a review of the botanical data, its geographical areas of origin and distribution are determined. A germplasm survey conducted on nine islands of Vanuatu enabled the collection of 110 accessions corresponding to 84 cultivars. These were grouped into: (1) wild forms under domestication, (2) annual types, (3) perennial-type cultivars of D. nummularia and (4) natural interspecific hybrids with D. alata L. Within each group, remarkable morphological variation exists for stem spininess, leaf shape, emerging leaf colour, tuber shape and tuber flesh colour. Selected accessions examined for ploidy had chromosome numbers ranging from 2n = 3x = 60 to 2n = 6x = 120. Chloroplast counts conducted on stomatal guard cells confirmed the variation in ploidy levels. Eight different male cultivars were studied for pollen fertility, but <5% of the pollen grains appeared to be fertile when stained with acetocarmine. However, profuse flowering and synchrony favour pollination and result in fruit set. All accessions are resistant to anthracnose. Viruses were detected but are morphologically not discernible, with no incidence on yield. Physicochemical analyses of tubers from 16 accessions revealed that D. nummularia and interspecific hybrids have high percentages of dry matter (mean of 33.11%) and starch (82.81%). The Pacific yam could be improved through conventional hybridisation or used for interspecific crosses with related economically important species. Microsatellite (SSR) polymorphism was assessed across 90 diploid Avena strigosa Schreb. and tetraploid Avena barbata Pott ex Link accessions obtained from the USDA-ARS National Small Grains Collection using 105 genomic SSRs. Eleven polymorphic SSRs that detected 69 different alleles were identified and used to genotype the 90 accessions, which were chosen from a larger set of 385 accessions based on geographical source-diversity and variable reaction responses to five Australian pathotypes of the crown rust pathogen Puccinia coronata Corda f. sp. avenae Eriks. Eight diploid and eight tetraploid clades were identified among the 90 accessions. Diploid accessions displayed the lowest genetic diversity, with all accessions being at least 86 % similar, and included accessions from countries in the Americas such as Canada, USA, Argentina, Uruguay and Brazil, and European accessions from France, Romania and Poland. Although both species formed distinct clusters in the dendrogram, a few instances of diploids showing high similarity with tetraploids and vice versa were observed. An AMOVA analysis revealed 86 % of the total genetic variation to be distributed within the two oat species, while between-species differences accounted for only 14 %. Heterozygosity (H) index values of 0.32 and 0.40 were obtained for diploids and tetraploids respectively. Our study effectively differentiated A. strigosa and A. barbata, and identified 11 SSRs suitable for future characterisation of accessions of the two species.  相似文献

Wild Rosa resources in Yunnan are very abundant and hold many important and useful traits for improving modern rose. However, many of them have no cytological data and few were karyologically analysed. In this article, karyological study of 24 wild Rosa taxa belonging to different sections was carried out by means of traditional squashing and pressing. The chromosome number of 8 taxa and karyotypes of most studied ones were reported for the first time. 22 resources were diploids with 14 chromosomes (2n = 2x = 14), while Rosa moyesii Hemsl. et Wils. and Rosa macrophylla Lindl. from Sect. Cinnamomeae DC. were hexaploids with 42 chromosomes (2n = 6x = 42). Only type 1A and type 2A been found, karyotypes of genus Rosa in Yunnan were symmetric. Also, the studied taxa differed much from each other in chromosome length, position of centromere, satellite position and satellite number, etc. The results implied that wild Rosa germplasm in Yunnan were rich in karyologcal diversity, and polyploidization might have played and were still playing very important roles in the evolution and speciation in genus Rosa, mainly in Sect. Cinnamomeae in alpine areas of northwestern Yunnan. The cytological data would supply chromosomal proofs not only for the taxonomy, phylogeny and evolution of genus Rosa in this special geological area, but also for the exploitation of useful traits of wild Rosa into the breeding of new cultivars.  相似文献

EST-SSR from Medicago truncatula Gaertn. and Glycine max (L.) Merr. were tested for transferability in various species of Onobrychis (O. pyrenaica Sennen, O. argentea Boiss. and O. viciifolia Scop.). Repeatable amplification was obtained for 81% of the microsatellites and 52% were polymorphic. Six selected SSRs from M. truncatula were used to fingerprint and estimate the genetic similarity of a set of 23 accessions of O. viciifolia. PCA analysis discriminated among the different Onobrychis species and the sainfoin accessions were clustered in a single major group. This grouping is discussed in terms of the history of cultivation of sainfoin in Spain. The selected SSRs will allow fingerprinting and genetic studies in Onobrychis species, solving the lack of available SSR markers in this species.  相似文献

Naturally occurring/spontaneously produced polyploids with six/more genomes are rarely found in Alliums. A hexaploid form of Allium tuberosum with 2n = 48 chromosomes has been isolated for the first time amongst the open-pollinated seedlings of a hypotetraploid plant (2n = 4x = 31); latter being the seed-derived product of a normal tetraploid stock (2n = 4x = 32) growing in Jammu University Botanical Garden. Except for the guard cells and pollen grains that are of increased size, this form compared to its progenitor is dwarf, has smaller leaves and bears inflorescences with few flowers. This plant is also different from its progenitor in having nearly one-fourth (27.8 %) of its pollen mother cells (PMCs) with varying chromosome number viz. 27–64, with the remaining cells having somatic or double the somatic number of chromosomes. To assess the nature of hexaploid form, its chromosomes were studied for morphological details, putative grouping and pairing properties during reduction division. Morphological similarity in the chromosomes of the present cytotype and its progenitor, arrangement of 48 chromosomes in eight groups of six chromosomes each and presence of 21.88 % euploid cells with eight hexavalents pointed towards the autopolyploid nature of the present strain. Regarding the origin of this strain, observation made on the meiosis in the two sex mother cells of the progenitor provides some clues. In the later plant, presence of most of embryo-sac mother cells with 62 chromosomes that showed 31:31 segregations and existence of majority of the PMCs with 31 chromosomes exhibiting erratic segregations indicate that the hexaploid strain has probably originated as a result of the fusion of reduced male (n = 17) and unreduced female gamete (2n = 31).  相似文献

Meiosis in the tetraploid (2n = 80) males of Dioscorea alata L. was investigated for the first time. During metaphase I, the chromosomes were associated mostly in 6–8 quadrivalents and the remaining ones as bivalents with no trivalents or univalents. Anaphase I and subsequent stages of meiosis were normal. The observation of quadrivalents in the tetraploids provides cytological evidence for autotetraploidy. The autotetraploid males and females were highly fertile and they produced viable seeds on artificial pollination. Pollination between diploids (2n = 40) and tetraploids (2n = 80) were also successful via embryo rescue, producing triploid (2n = 60) progenies. The discovery of fertile autotetraploids could initiate polyploidy breeding in D. alata by conventional hybridization for the first time. The reduced number of quadrivalents and the high fertility of the autotetraploids are thought to be the result of partial diploidization of meiosis. The findings also refute the assumption of the allopolyploid origin of D. alata from two putative progenitors.  相似文献

The genus Aegilops L. comprises 22 annual wild species that are closely related to wheat (Hammer in Kulturpflanze 28: 33–180, 1980). Aegilops neglecta Req. ex Bertol. is a member of the Aegilops section of this genus and is distributed from Morocco and Spain in the west to Transcaucasia and western Iran in the east. This species includes tetraploid (2n = 28, genome UUMM) and hexaploid forms (2n = 42, UUMMNN). However, the geographical distributions of the two cytological forms remain unclear. Clarifying the distribution of the two cytological forms is essential for a better understanding of the diffusion of Ae. neglecta and its tetraploid and hexaploid forms. In the present study, chromosome numbers were determined for accessions of Ae. neglecta from a total of 137 populations, located in the western area of the species distribution from the Aegean Islands to Morocco. Taken together with previous studies, the present data reveal a difference in the geographical distribution of tetraploid and hexaploid forms: tetraploid form is distributed in the eastern part of the species area and hexaploid form predominantly occurs in the western part with their border on the western margin of the Aegean Sea. Near the border, tetraploid and mixed populations are sporadically found among hexaploid populations in the Balkan and Peloponnesus Peninsulas, while a few hexaploid and mixed populations are found among tetraploid populations in the East Aegean Islands and West Anatolia.  相似文献

The loci for expression of prolonged spikelet rachilla named ‘sham ramification’ in three tetraploid wheat accessions, two accessions of Triticum jakubzineri Udacz. et Schachm. (2n = 4× = 28, genome BBAA) ‘PI 585014’ and ‘R-101-03’ and one sample of Triticum turgidum L. (2n = 4× = 28, genome BBAA) ‘PI 67339’, were mapped by genotyping F2 populations using microsatellite markers. The segregation analysis confirmed that sham ramification was controlled by recessive, epistatic genes, shr1 (sham ramification 1) and shr2 (sham ramification 2). The genotypes of ‘sham ramification’ were shr1shr1Shr2Shr2 for T. jakubzineri and Shr1Shr1shr2shr2 for T. turgidum (PI 67339). Normal rachilla was determined by Shr1Shr1Shr2Shr2. The shr1 gene and the gene for extra glume (exg) were completely linked; and, the shr1/exg gene complex was bracketed by the markers Xbarc319 and Xbarc232 on the long arm of chromosome 5A. In the F2 of PI 67339/LD222 the gene shr2 was bracketed by Xwmc819 and Xwmc794 on the long arm of chromosome 2A.  相似文献

Chloroplast simple sequence repeats (cpSSRs) are widely distributed in the chloroplast genomes of all plant species, and are frequently employed for genotypic and phylogenetic analysis. However, information on intra- and interspecies variation in cpSSRs is lacking. In this study, we sequenced four intergenic (non-coding) chloroplast DNA regions in 57 accessions of 12 tetraploid, and 16 accessions of 4 hexaploid species of Triticum and Aegilops. These sequence data added to our previous data for diploid species in the same chloroplast regions. Intra- and interspecific genetic variation was analyzed for a total of 189 accessions of 13 diploid, 12 tetraploid, and 4 hexaploid species of Triticum and Aegilops, such that all species were represented by multiple accessions. The data were used to infer phylogenetic relationships within and among Triticum and Aegilops species. Based on this robust phylogenetic tree, seven of eight cpSSR loci clearly exhibited “size homoplasy,” referring to the fact that cpSSRs of identical size and DNA sequence can arise even if the alleles are not descended from a common ancestor. These data indicate that cpSSRs should be used with caution in phylogenetic analyzes. Interestingly, as observed from several previous studies, our data also suggest that observed mutation rates may increase significantly when mononucleotide (homopolymer) repeat numbers reach or exceed 9 bp. In the present report, using this sequence data set involving cpSSRs, 81 unique haplotypes among 189 accessions were detected, and five tetraploid Triticum and Aegilops species were successfully identified and genotyped. Our results indicate that combinations of nucleotide substitutions, indels and SSRs of chloroplast nucleotide sequences are available for genotyping at the species accession level.  相似文献

Oryza officinalis complex is the largest and the most complicated group in the genus Oryza L., consisting of about ten species with the B, C, BC, CD, and E genomes. Taxonomy and identification of the species, particularly those with the B, C and BC genomes, are difficult due to the similar morphology and overlapping distribution of some species. The difference in ploidy levels of some species adds more complexity. In the present study, we surveyed 64 accessions of rice germplasm in the O. officinalis complex using RFLP analysis of PCR-amplified Adh genes in addition to chromosome counting. The results confirmed that all O. rhizomatis accessions are diploids with the C genome, whereas all O. minuta accessions are tetraploids having the BC genome. However, both diploid and tetraploid forms were found for the accessions identified in the genebank as O. officinalis, O. punctata and O. eichingeri. The tetraploid form of O. officinalis with the BC genome is exclusively distributed in India and has been described as O. malampuzhaensis. The tetraploid form of O. punctata which has been called O. schweinfurthiana by some workers was found to be as widely distributed as its diploid form in Africa. It is noteworthy that two accessions that had been determined as tetraploid O. officinalis were actually belonging to a species with the CD genome (O. latifolia). Our results promote a better understanding of the genomic constitutions of many accessions in the O. officinalis complex and correct determination of the genebank material, which serves as an important basis of germplasm cataloguing for further research and utilization.  相似文献

The supposedly first plant of the coffee cultivar “Híbrido de Timor” (HT) was found in 1927, being denoted as HT CIFC 4106. According to different researchers, this plant originated from a natural interspecific hybridation between Coffea arabica (4x = 44) and Coffea canephora (2x = 22). From HT CIFC 4106, other HT accessions were obtained and employed to establish germplasm banks in some countries. As HT has been widely used in Coffea breeding programs, this study aimed to characterize different HT accessions with regard to ploidy, nuclear DNA content and base composition. Based on these data, the ploidy of HT CIFC 4106 was determined, suggesting that this accession is an allotriploid formed from reduced reproductive cell of C. canephora and of C. arabica. All HT CIFC 4106 plants exhibited the same 2C-value, AT% and chromosome number, showing that vegetative propagation has enabled the multiplication and germplasm conservation of this cytotype since 1927. Further five analyzed HT accessions showed distinct nuclear 2C-value and AT%. Since HT CIFC 4106 has been considered the first HT, it is suggested that aneuploid reproductive cells of this HT originated the other plants. Considering that HT accessions are used in the development of C. arabica cultivars, the findings of this study are important for the design of strategies to obtain new cultivars for breeding programs. Moreover, these data represent the first step to understand the origin and genome evolution of the HT.  相似文献

The genetic diversity was studied of 115 Agropyron cristatum accessions from 17 countries. Tetraploids were the most common (74.8%), followed by diploid (16.3%) and hexaploid (6.9%). We observed a relation between geographic distribution and ploidy level. The tetraploids, the most widespread, were found from Europe through Russia to East Asia. The diploids appeared over the same general range, except in Turkey, Iran and Georgia where no diploid accessions were found. Hexaploid accessions mainly came from a region comprising the east of Turkey, the north of Iran and Georgia. A selection of 71 accessions, including all three ploidy levels, were analyzed by capillary electrophoresis using six wheat simple sequence repeat (SSR) markers. All markers presented high levels of polymorphism, generating 166 different alleles ranging in size between 84 and 256 bp. Based on polymorphic information content values obtained (0.579–0.968), all the SSRs were classified as informative markers (values?>?0.5). According to the dendrogram generated, all the A. cristatum accessions were distinctly classified. Diploid, tetraploid and hexaploid accessions are not clearly differentiated from each other on the basis of SSR markers. A field experiment was conducted to morphologically characterize 18 accessions including the three ploidy levels. Significant differences were found between the accessions in spike length, spike width and number of spikelets per spike. All the cytological, molecular, and morphological data demonstrate the high genetic diversity present in A. cristatum, making it a valuable resource for future breeding programs.  相似文献
