Fluorescence in situ hybridization (FISH) and comparative genomic hybridization (CGH) were applied to somatic chromosomes preparations of Oryza sativa, O. officinalis, and O. meyeriana with labeled probes of Cot-1 DNA and genomic DNA'from the cultivated rice. The coverage percentage (%) and size (Mb) of Cot-1 DNA in O. sativa, O. officinalis, and O. meyeriana were 47.1 ±0.16, 38.61 ±0.13, 44.38+_0.13, and 212.33 ± 1.21,269.42 ± 0.89, 532.56± 1.68 Mb, respectively. The coverage percentage and size of genomic DNA from O. sativa in O. officinalis and O. meyeriana were 91.0, 93.6% and 634, 1 123 Mb, respectively, in which 365 and 591 Mb in O. officinalis and O. meyeriana were from O. sativa genomic DNA, but not from repetitive sequences of O. sativa, and the uncoverage genome size in O. officinalis and O. meyeriana were 64 and 78 Mb, respectively. In addition, karyotype analysis was conducted based on the signal bands of Cot-1 DNA in O. sativa, O. officinalis, and O. meyeriana. The results showed that highly and moderately repetitive sequences in Oryza genus were conserved as the functional genes during evolution. The repetitive sequences reduplication may be one of the important causes of the genome enlargement of O. officinalis and O. meyeriana, and O. officinalis genome enlarged more slowly when compared with O. meyeriana. Based on the above results, it is concluded that O. officinalis and O. meyeriana were formed by reduplication, rearrangement, and gene selective loss during the evolution process.  相似文献

Crop performance is determined by the combined effects of the genotype of the crop and the environmental conditions of the production system. This study was undertaken to develop a dynamic model for simulating environmental (temperature and solar radiation) and N supply effects on fiber fineness, maturity and micronaire. Three different experiments involving genotypes, sowing dates, and N fertilization rates were conducted to support model development and model evaluation. The growth and development duration of fiber fineness, maturity, and micronaire were scaled by using physiological development time of secondary wall synthesis (PDT SWSP ), which was determined based on the constant ratio of SWSP/ BMP. PTP (product of relative thermal effectiveness (RTE) and photosynthetically active radiation (PAR), MJ m-2) and subtending leaf N content per unit area (N A , g m-2) and critical subtending leaf N content per unit area (CN A , g m-2) of cotton boll were calculated or simulated to evaluate effects of temperature and radiation, and N supply. Besides, the interactions among temperature, radiation and N supply were also explained by piecewise function. The overall performance of the model was calibrated and validated with independent data sets from three field experiments with two sowing dates, three or five flowering dates and three or four N fertilization rates for three subsequent years (2005, 2007, and 2009) at three ecological locations. The average RMSE and RE for fiber fineness, maturity, and micronaire predictions were 372 m g-1 and 5.0%, 0.11 m g-1 and 11.4%, 0.3 m g-1 and 12.3%, respectively, indicating a good fit between the simulated and observed data. It appears that the model can give a reliable prediction for fiber fineness, maturity and micronaire formation under various growing conditions.  相似文献

Soybean (Glycine max L. Merr.) is the world's foremost source of edible plant oil and proteins, meantime, the biologically active secondary metabolites such as saponins and isoflavones are benefit to human health. The objective of this study was to identify quantitative trait loci (QTL) and epistatic interactions associated with isoflavone, protein, and oil contents in soybean seeds. An F13 recombinant inbred line (RIL) comprising 474 lines was derived from a cross between Jindou 23 and Huibuzhi cultivars. SSR technique was employed for mapping of the QTLs. The QTLs for isoflavone, protein, and oil contents were analyzed and 23 QTLs were detected based on the constructed linkage map. Six QTLs for isoflavone content were localized in linkage groups J, N, D2, and G, eleven QTLs for oil content were localized in the linkage groups A1, A2, B2, C2, and D2, and six QTLs for protein content were localized in linkage groups B2, C2, G, and H1. The correlative analysis demonstrated that the isoflavone content had significant correlation with protein content, while significantly negative correlations was existed between oil and protein content, and significantly positive correlations was existed between protein and oil content. All these findings have laid an important basis for the marker assisted breeding in soybean. The phenotypic correlations of quantitative traits may be resulted from the correlation of the QTL controlling those traits.  相似文献

Molybdenum(Mo) is an essential trace element in plant nutrition and physiology. It affects photosynthesis and photosynthate accumulation, therefore also affecting fruit quality and nutritional content. This study assessed the effects of different sodium molybdate(Na_2MoO_4) concentrations on strawberry. Five different Mo concentrations were applied in this experiment, including 0, 67.5, 135, 168.75, 202.5 g ha~(-1), respectively. The mineral concentration, including nitrogen(N), Mo, iron(Fe), copper(Cu), and selenium(Se) was assessed in strawberry fruit, as well as chlorophyll content, nutrition quality, taste and aroma. Results showed that net photosynthetic rate(P_n) and chlorophyll content for the strawberry plants increased with an increase in Mo concentration; and the contents of N, Mo, Fe, Cu, total soluble solids(TSS), titratable acidity(TA), sweetness, some sugars, organic acids, and some volatile compounds in the fruit all increased, as well. However, the Mo concentration did not significantly affect the concentrations of Se, sucrose, lactic acid, acetic acid, and some aroma compounds. Fruit sprayed with 135 g ha~(-1) Mo exhibited the highest TSS and sweetness values, as well as the highest N and Fe concentrations among all the treatments. P_n value and chlorophyll content, fructose, glucose, sorbitol and total sugar contents in fruit supplied with 135 g ha~(-1) Mo were also higher than that in other treatments. Fruit sprayed with a Mo concentration of 67.5 g ha~(-1) exhibited significantly higher ascorbic acid(AsA) values than that of control. Ninety-seven volatile compounds were identified in fruit extracted by head-space solid-phase microextraction(HS-SPME) coupled with gas chromatography-mass spectrometry(GC-MS). Fruits sprayed with 135 g ha~(-1) Mo had the highest concentrations of six characteristic aroma compounds, including methyl butanoate, γ-decalactone, ethyl butanoate, methyl hexanoate, γ-dodecalactone, and ethyl caproate.  相似文献

Focusing on the notion of competencies, the address explores important dimensions of human infrastructure for negotiating alternative agrifood systems. The analytical competencies emphasized are those of making connections and evaluating contradictions. Farm structure and food system connections with human health and consumer culture are chosen as examples. Examined in the context of social change strategies, relational competencies focus on new forms of food citizenship involving alternative organizational relationships between farmers, retailers, and customers. Ethical competencies are framed in relationship to the entire food system, to the valuing of non-market goods, and to linkages between ethics and emotions. A solution culture experiment was performed to investigate the effects of oxytetracycline (OTC) on wheat (Triticum aestivum L.) growth,chlorophyll contents,and photosynthesis at five levels of 0,10,20,40,and 80 mmol L-1 OTC.OTC is toxic to wheat.The wheat growth,especially wheat root was significantly decreased.Further OTC also significantly decreased root activity,chlorophyll contents,and photosynthetic parameters except for intercellular CO2 concentrations.The different responses of indicators such as root number,root activity and so on to OTC were also observed.The IC50 values for the tested indicators to OTC ranged from 7.1 to 113.4 mmol L-1 OTC.The order of indicator sensitivity to OTC was root number > stomatal conductance > chlorophyll a > total chlorophyll > photosynthetic rates > total surface area >transpiration rate > chlorophyll b > fresh weight of root > dry weight of root > total length > dry weight of shoot =fresh weight of shoot> total volume.The root number was more sensitive than other indicators with the ICs0 value of 7.1 mmol L-1 OTC,and could be taken as the sensitive indicator to predict the hazards of OTC to wheat.  相似文献
