The alcoholic extract of Trewia polycarpa roots, when administered orally to rats at doses of 50-400 mg/kg, exhibited a dose-dependent anti-inflammatory activity in both acute and chronic models. It also showed a significant antinociceptive action mice in the dose range of 25-200 mg/kg. The extract did not reveal any toxicity in rats up to a dose of 3.2 g/kg (p.o.). It showed the presence of terpenoids, alkaloids, flavonoids, quinones and glycosides on phytochemical screening.  相似文献

The ethanolic extract of Leucas aspera root was subjected to acetic acid induced writhing inhibition, 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging assay and brine shrimp lethality bioassay for screening of antinociceptive, antioxidant and cytotoxic activity, respectively. The extract produced significant writhing inhibition in acetic acid induced writhing in mice at the doses of 250 and 500 mg/kg. The extract showed a significant free radical scavenging activity with an IC(50) of 8 microg/ml. The extract showed significant lethality to brine shrimp with an LC(50) value.  相似文献

The antinociceptive effect of the aqueous extract of Zizyphus spina-christi root bark was investigated in mice and rats. Acetic acid-induced writhing, formalin and thermal (hot plate) tests were used. The extract (50 and 100 mg/kg, i.p.) showed a dose-dependent analgesic effect in all the tests used. Its i.p. Leandra lacunosa, popularly known as "erva-do-jabuti", is used in Brazilian folkloric medicine for the treatment of diabetes mellitus. Based on this traditional indication, the aim of this work was to evaluate the hypoglycemic activity of the hydroalcoholic extract of L. lacunosa aerial parts (LLH) in normal and alloxan-induced diabetic rats. Chromatographic fractionation of LLH was also carried out by several techniques, affording isolation of the following major compounds: ursolic acid (1), kaempferol (2), luteolin (3), and quercetin (4). The oral administration of LLH (500 mg/kg) in normal rats caused a significant reduction of 24.7% (P<0.05) in the blood glucose levels after 2 h of treatment, while the administration of chlorpropamide (20 mg/kg, p.o.) led to a reduction of 40.2% (P<0.01). After oral administration of glucose (10 g/kg, p.o.), LLH (500 mg/kg, p.o.) significantly inhibited the increase in blood glucose levels compared with the negative control group. The oral treatment with LLH (500 mg/kg) in alloxan-induced diabetic rats significantly reduced the blood glucose levels in 47.8% after 4 h of treatment, while chlorpropamide resulted in a significant reduction of 71.7% in the 4th hour. Our results showed that LLH, displays hypoglycemic activity, which may be related to the effect of the major compounds identified in the crude extract. This study seems to provide biological evidence for the folkloric use of L. lacunosa in the treatment of diabetes mellitus.  相似文献

The present study reports the potential antinociceptive, anti-inflammatory and hepatoprotective activities of lycorine from Sternbergia fischeriana (Herbert) Rupr. (Amaryllidaceae). Lycorine was evaluated on mice by using acetic-acid induced writhing and tail-flick tests. Lycorine exhibited stronger inhibition than aspirin in acetic-acid induced abdominal stretching at 1.0 mg/kg dose. Lycorine also showed antinociceptive activity at 1.0 mg/kg dose in tail-flick test. The anti-inflammatory activity of lycorine was not found to be significant at dose of 0.5 mg/kg. However, at doses of 1.0 mg/kg and 1.5 mg/kg, i.p. showed a significant reduction with 53.45% and 36.42%, respectively in rat paw oedema induced by carrageenan against the reference anti-inflammatory drug indomethacin (3 mg/kg, i.p.) (95.70%). The ED50 of lycorine was determined as 0.514 mg/kg. Hepatoprotective activity of lycorine on carbon tetrachloride (CCl4) induced acute liver toxicity following biochemical parameters were also evaluated. Rats were treated with lycorine at doses of 1.0 mg/kg and 2.0 mg/kg, i.p. Results of biochemical tests were confirmed by histopathological examination. Lycorine exhibited significant hepatoprotective effect at dose of 2.0 mg/kg i.p. dose.  相似文献

The gastroprotective effects of some extracts of the dried leaves of Ficus sur in animals were investigated in order to validate the use of the plant in traditional medicine. The water and methanol extracts showed antiulcer activity, the hot water extract being the most effective. These extracts had also spasmolytic effects on the rabbit jejunum. LD50 values ranged from 900 to 1200 mg/kg in mice. These results show that F. sur has potential in the treatment of ulcers.  相似文献

Anti-nociceptive and anti-oedematogenic effects of the aqueous extract from the leaves of Erythrina velutina were tested through experimental models of nociception in mice and paw oedema induced by carrageenin in rats. The extract (300 and 600 mg/kg) did not change the carrageenin-induced paw oedema. In the hot plate test the extract also did not alter the latency time for mice liking the rear paws. Moreover, the extract (600 mg/kg) decreased by 96.5% the paw liking time in the second phase of the formalin test. This effect was antagonized by naloxone (5 mg/kg). In the acetic acid-induced writhing test, the extract (300 and 600 mg/kg) reduced the number of writhing by 88.8% and 96.4%, respectively. Our present results demonstrated that the crude extract from the leaves of E. velutina has anti-nociceptive but not anti-oedematogenic properties.  相似文献

Antihyperglycemic activity of the aqueous extract of Urtica dioica   总被引:5,自引:0,他引:5  
When administered 30 min before glucose loading, the aqueous extract of Urtica dioica (nettle) (250 mg/kg) showed a strong glucose lowering effect. The decrease of glycemia has reached to 33+/-3.4% of the control value 1 h after glucose loading. This effect was persistent during 3 h. In contrast, nettle did not show hypoglycemic effect in alloxan-induced diabetic rats. The amount of glucose absorbed in a segment jejunum in situ was 8.05+/-0.68 mg in presence of nettle extract vs. 11.11+/-0.75 mg in control rats during 2 h (P<0.05). The results indicate that nettle has a significant antihyperglycemic effect in OGTT model. This effect may be caused in part by the reduction of intestinal glucose absorption. LD(50) is 3.5 g/kg (i.p.).  相似文献

Hexane extracts of aerial parts (flowered browes), leaves and roots of Sambucus ebulus were investigated for their antiinflammatory activity in rats. Aerial parts and roots extracts produced statistically significant and dose dependent inhibition of edema induced by carrageenan at all doses when compared to the control groups. On the contrary, hexane extract of leaves did not show any activity up to 600 mg/kg i.p. The highest activity was showed in the aerial parts and roots that at 600 mg/kg i.p. inhibited inflammation by ca. 80% (78% for diclofenac at 100 mg /kg i.p.). No extracts exhibit toxicity when injected up to 2 g/ kg intraperitoneally in mice.  相似文献

The antiinflammatory activity of the aqueous extract of Polygala japonica (AEPJ) was investigated in mice and rats to find the pharmacological basis for its ethnomedical use. The extract produced a significant inhibition of peritoneal and cutaneous vascular permeability induced by acetic acid and histamine, respectively and ear swelling induced by picryl chloride in mice at the dose of 25.0 mg/kg. Moreover, the extract markedly inhibited footpad edema induced by histamine in rats, and decreased prostaglandin E(2) (PGE(2)) content in carrageenan-induced air-pouch at doses of 12.5 and 6.25 mg/kg respectively.  相似文献

The protective effects of Ginkgoselect Phytosome((R)) (GBP) on Rifampicin (RMP) induced hepatotoxicity and the probable mechanism(s) involved in this protection were investigated in rats. Liver damage was induced in Wistar rats by administering rifampicin (500 mg/kg, p.o.) daily for 30 days. Simultaneously, GBP at 25 mg/kg and 50 mg/kg, and the reference drug silymarin (100 mg/kg) were administered orally for 30 days/daily to RMP treated rats. Levels of marker enzymes (SGOT, SGPT and SALP), albaumin (Alb) and total proteins (TP) were assessed in serum. The effects of GBP on lipid peroxidation (LPO), reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX) and glutathione reductase (GR) were assayed in liver homogenates to evaluate antioxidant activity. GBP (25 and 50 mg/kg) and silymarin elicited a significant hepatoprotective activity by lowering the levels of serum marker enzymes and lipid peroxidation and elevated the levels of GSH, SOD, CAT, GPX, GR, Alb and TP in a dose dependant manner. The present findings suggest that the hepatoprotective effect of GBP in RMP induced oxidative damage may be related to its antioxidant and free radical scavenging activity.  相似文献

Ethanol extract of Maytenus rigida stem bark and its fractions were assessed for antinociceptive activity in tail-flick test in rats. The activity was located in the chloroform, ethyl acetate and aq.methanol fractions. Phytochemical screening revealed that catechin was the only common class of compounds present on the ethanol extract as well as on the active fractions. 4'-Methylepigallocatechin, isolated from the ethyl acetate and aq.methanol fractions, showed antinociceptive effect in the tail-flick test (75 mg/kg; p.o.), which was reversed by the opiate antagonist naloxone (3 mg/kg; i.p.).  相似文献

The anti-diarrheal activity of methanol extract of dried bark and aerial parts of Litsea polyantha (MELP) has been evaluated in mice using different models (castor oil-induced diarrhea and propulsive gut motility in mice). MELP (50, 75, 100 mg/kg, p.o.) significantly (P<0.01) reduced the onset of diarrhea, fecal excretion and also showed a significant (P<0.001) reduction in gastrointestinal motility on charcoal meal test in mice. The results of the study support the folklore use of the plant for diarrheal remedies.  相似文献

The present study was carried out in order to examine the anticancer properties of two sesquiterpene lactones, millerenolide and thieleanin, isolated from Viguiera sylvatica and Decachaeta thieleana, against cell lines in vitro, and on the growth B16/BL6 melanoma tumors in C57BL/6 mice. Millerenolide and thieleanin showed a similar pattern of cytotoxicity with the greatest effect on viability being evident with A549 human lung cancer cells (IC(50) - 40 and 32 microM respectively), and with the 3T3/HER2 cell line which are 3T3 mouse fibroblasts transfected with the HER2 oncogene (IC(50) - 16 and 28 microM respectively). The parent 3T3 cells and the B16/BL6 mouse melanoma cells were less sensitive to these compounds, with thieleanin showing an IC(50) with B16/BL6 greater than the highest dose tested (203 microM). Treatment with millerenolide (8 mg/kg, i.p. on days 0, 2 and 4 post-inoculation) significantly inhibited the growth of subcutaneous B16/BL6 tumors in C57BL/6 mice, (50% inhibition at day 25, P=0.015), as well as retarding the appearance of detectable tumor (millerenolide - day 15.2+/-0.4 vs control - day 12.8+/-0.5, mean+/-SEM, P=0.011). In contrast, treatment with thieleanin (8 mg/kg every other day up to the day of kill) neither retarded the appearance of the tumor nor its growth.  相似文献

The defatted methanolic extract of Entada abyssinica was evaluated for anti-inflammatory activity in acute and chronic models of inflammation. The extract (50--200 mg/kg, p.o.) exhibited dose-dependent and significant inhibition of both the carrageenan-induced rat paw oedema and granuloma tissue formation in rats. The extract (50--200 mg/kg, p.o.) was also found to inhibit the acetic acid-induced vascular permeability in a dose-dependent fashion in mice.
