It is well known that cattle ingesting aflatoxin B1 contaminated feed commodities excrete aflatoxin M1 into their milk. As aflatoxin M1 originates from hepatic metabolism, measures to prevent aflatoxin M1 formation need to be directed to either the immobilization of aflatoxin B1 in the gastrointestinal tract or the modification of hepatic metabolism of aflatoxin B1. Here we studied the influence of oltipraz and a second dithiolthione, (1,2) dithiolo (4,3-c)-1,2-dithiole-3,6 dithione (DDD) on bovine hepatic aflatoxin B1 biotransformation. Oltipraz inhibited aflatoxin B1 metabolism as no aflatoxin M1 and no aflatoxin B1-dihydrodiol, the second metabolite found in bovine hepatocytes, was formed. DDD did not significantly inhibit aflatoxin B1 metabolism. It could be demonstrated that the inhibition of aflatoxin B1 metabolism was due to the inhibition of several cytochrome P450 enzyme activities by oltipraz. In contrast, DDD inhibited only ethoxyresorufin O-deethylation activity. These findings suggest a high efficacy of oltipraz in inhibiting aflatoxin M1 contamination of milk from dairy cows exposed to aflatoxin B1 contaminated feeds.  相似文献

Selenium may be related to the hepatic metabolism of the coumarin compounds aflatoxin B1 and warfarin. Selenium evidently increased the pharmacologic activity of warfarin, probably due to a displacement of warfarin from albumin by selenium, the close relationship among selenium, vitamin E, and sulphur-containing groups (eg, glutathione), or the antioxidant effect of selenium. A diet containing selenium in a concentration of 2.5 mg/kg of feed was protective against the toxic effects of both coumarins in pigs given 4 daily oral doses of 0.2 mg/kg of body weight. Selenium, as glutathione peroxidase, at least in part, protects the hepatic cells against the toxic effects of aflatoxin B1 and warfarin. The protection was demonstrated by alteration of clinical responses and hematologic (prothrombin times), electrophoretic, and clinical chemistry values. It also was demonstrated that selenium at 2.5 mg/kg of feed does not produce toxic effects; however, dietary selenium at a concentration of 5 mg/kg (and in the presence of both toxic agents) was toxic for young pigs within the 3-week experimental period. 本文通过测定肉仔鸡生长性能、血清指标及器官指数,探讨葡甘露聚糖对黄曲霉毒素B1(AFB1)的吸附作用。选用180只体重接近的8日龄肉仔鸡随机分成3组,分别为基础日粮组、基础日粮 0.1mg/kgAFB1组、基础日粮 0.1mg/kgAFB1 0.1%葡甘露聚糖组,试验期21d。试验结果表明:(1)基础日粮中加入0.1mg/kgAFB1,肉仔鸡的体重、日增重、日采食量显著下降,料重比升高。血清中总蛋白、白蛋白含量显著下降,谷草转氨酶、谷丙转氨酶活性显著升高。肝脏、肾脏、心脏和脾脏肿胀,胸腺、法氏囊萎缩;(2)0.1mg/kgAFB1日粮中加入0.1%葡甘露聚糖,明显改善肉仔鸡的生长性能,以上相关指标基本恢复到正常水平。由此可知,葡甘露聚糖减轻或基本消除了AFB对肉仔鸡生长性能、组织器官的不良影响。  相似文献

In twenty fattened pigs with papular dermatitis (PD) and in seventeen pigs with no skin lesions (K),--the pigs had the live weight from 95 to 105 kg and came from a D. large fattening house situated in a region with extensive mining (lignite extraction)--we analyzed the skin, muscle, kidneys and liver for the contents of chemical elements (zinc, copper, manganese, lead, cadmium, mercury, chromium) and aflatoxin B1. In pancreas we analyzed only the content of aflatoxin B1. In comparison with the K group, the pigs with PD had in the skin the higher contents of zinc (PD 15.7 +/- 7.1 mg per kg, K 10.1 +/- 1.3 mg per kg, P less than 0.05) and aflatoxin B1 (PD 3.18 +/- 1.53 micrograms per kg, K 1.56 +/- 0.49 microgram per kg, P less than 0.05); in the muscle the higher contents of zinc (PD 30.2 +/- 7.6 mg per kg, K 23.8 +/- 5.4 mg per kg, P less than 0.05), of lead (PD 0.48 +/- 0.31 mg per kg, K 0.25 +/- 0.28 mg per kg, P less than 0.05) and copper (PD 3.19 +/- 3.11 mg per kg, K 0.98 +/- 0.41 mg per kg, P less than 0.01); in the kidneys the higher contents of lead (PD 0.96 +/- 0.75 mg per kg, K 0.26 +/- 0.34 mg per kg, P less than 0.01), cadmium (PD 0.38 +/- 0.14 mg per kg, K 0.26 +/- 0.20 mg per kg, P less than 0.05), manganese (PD 1.14 +/- 0.04 mg per kg, K 0.94 +/- 0.42 mg per kg, P less than 0.05).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This research compared the toxic effects of aflatoxin B1 and monocrotaline, the active principle of Crotalaria spectabilis, and the additive effect between aflatoxin B1 and monocrotaline in turkey poults. It was of interest whether selenium fed at dosage levels of 0.1, 5, or 10 micrograms/g of feed would protect against the toxic effect of aflatoxin and/or monocrotaline, and whether the toxicants would result in detectable residues in poult tissues. A total of 180 healthy 1-day-old male turkey poults was assigned at random to 12 treatment groups (15 birds/group). Body and liver weight losses, and low serum concentrations in total protein (TP), albumin (A), alpha-globulin (alpha G), and beta-globulin (beta G), as well as high values in gamma-globulin (gamma G), were produced in the groups fed crotalaria. Pathologic changes were induced by monocrotaline with no protection afforded by the added selenium. Low values in TP, A, alpha G, and beta G and in body and liver weights were observed in groups given the combination of aflatoxin plus crotalaria. Gross lesions were associated with an additive toxic effect and a lack of protective effect of selenium against this combination. However, higher values in TP, A, alpha G, and beta G, and liver weights in groups fed aflatoxin B1 plus selenium indicated that selenium had a protective effect against aflatoxin toxicity. Residues of aflatoxin B1 and aflatoxin M1 were found in the kidneys of poults fed aflatoxin B1; also, dehydroretronecine (the metabolite of monocrotaline) was detected in livers of poults fed Crotalaria spectabilis seeds.  相似文献

Twenty-one pigs weighing approximately 18 kg were placed in 7 groups of 3 and given diets containing respectively aflatoxin B1 alone at 0.375 and 0.0750 mg/kg, ochratoxin A alone at 1 and 2 mg/kg, 0.375 mg/kg of aflatoxin B1 plus 1 mg/kg of ochratoxin A and 0.750 mg/kg aflatoxin B1 and 2 mg/kg of ochratoxin A. The remaining group served as untreated control. At the respective dose levels, pigs receiving similar doses of ochratoxin A alone or in combination with aflatoxin B1, were similarly affected, the clinical effects of aflatoxin having been mostly obscured by those due to ochratoxin A. Mild degenerative hepatic changes typical of aflatoxicosis were observed in pigs fed this toxin alone or in combination with ochratoxin A. In kidneys of pigs fed diet containing 1 and 2 mg of ochratoxin A alone changes included interstitial fibrosis of the vortex and dystrophy and degeneration of the tubular epithelium. Similar lesions but less pronounced fibrosis were found in kidneys of pigs receiving both toxins. The respective lower dose levels of mycotoxins selected were judged to be about the no-effect levels for each dosed separately under the conditions of the trial. Such levels have been found not infrequently on mould affected grain and stock foods. The result highlights the difficulties that may be experienced in the recognition of such multimycotoxicoses as they are likely to occur in the field and indicate the need for toxicological analysis as well as pathological investigation in establishing a diagnosis.  相似文献

Twelve pigs were subjected to model trials to study the effect of an aflatoxin-contaminated pig-fattening diet, administered for 26 days. Aflatoxin B1 constituted 55% of the aflatoxins tested. The aflatoxin concentrations used in the trials were 30 and 300 micrograms per 1 kg of feed. No clinical effect of aflatoxin was observed in the pigs. Aflatoxins were found to have adverse effects on proteosynthesis. A decrease in the weight gains of the pigs was accompanied by a decrease in the total plasma protein and, in the group with the higher aflatoxin dose (300 micrograms per 1 kg of feed), also by an increase in the total bilirubin concentration in the blood serum. Perilobular steatosis, dissociation of the liver trabeculae, and more pronounced activation of the reticuloendothelial system were demonstrated by the histological examination of liver tissue. An increase in the content of hydroxyproline was recorded in the muscle of the experimental pigs. It is documented by the results that the aflatoxin dose of 30 micrograms per 1 kg of feed (the recommended maximum admissible concentration for the Czech Socialist Republic) is high enough to reduce the performance of pigs and the biological value of their meat.  相似文献

旨在建立检测饲料中黄曲霉毒素B1、B2、G1、G2含量的高效液相色谱分析方法。样品用甲醇溶液提取,利用免疫亲和柱净化,以C18反相色谱柱分离,光化学衍生器衍生后应用荧光检测器检测。结果表明:4种黄曲霉毒素在0.5~20ng/mL范围内呈现良好的线性关系,回收率为71.6%~89.0%。采用该方法检测5种饲料样品中的黄曲霉毒素总量,检出率范围在74.0%~86.7%,检出值的范围为0.5~4.3μg/kg。建立的方法精密度高、重复性好,为监测饲料中黄曲霉毒素含量水平提供了有效的分析方法。  相似文献

In the period from January 1983 to December 1985 we examined thirty-five samples of commercial feed mixtures for pigs and thirty samples of pigsty dust deposition from large pig-houses in a region with extensive mining (lignite extraction) situated in the Hodonín district. In comparison with the contents of contaminants in the feed mixtures over the period from January 1983 (1/83) to June 1984 (6/84), the zinc and lindane concentrations increased to 213.3% (P less than 0.01) and 133.3%, respectively, from July 1984 (7/84) to December 1985 (12/85). The concentrations of copper, manganese, lead, cadmium, mercury (P less than 0.05), DDT (P less than 0.01) and aflatoxin B1 in the feed mixtures decreased. The highest permissible amount of contaminants in the feed mixtures was exceeded in the 1/83-6/84 period in cadmium (limit value 0.3 mg per kg) and mercury (limit value 0.1 mg per kg), in the 7/84-12/85 period only in zinc (limit value 250 mg per kg). In comparison with the values of contaminants in pigsty dust deposition recorded from January 1983 to June 1984, the lead concentration increased to 555.5% in the 7/84-12/85 period, lindane concentration rose to 569.0% (P less than 0.01), zinc concentration to 220.5% (P less than 0.01), the copper, manganese, cadmium, mercury concentrations (P less than 0.05) decreased, and the concentrations of DDT, aflatoxin B1 and polychlorinated biphenyls also dropped. It is desirable to evaluate in regular intervals the spectrum of contaminants both in feed mixtures and in pigsty dust deposition and to monitor other potential sources of contaminants in the pig-houses.  相似文献

Effects of aflatoxin on bovine rumen motility were determined by radiotelemetric techniques. Aflatoxin altered amplitude and/or frequency of rumen contractions in steers given dosages of 0.2, 0.4, 0.6, or 0.8 mg of aflatoxin/kg of body weight. Effects of aflatoxin on rumen motility were dose dependent. An increase in elimination time of aflatoxin from rumen contents was observed in steers given the aflatoxin dosages of 0.4 to 0.8 mg/kg. The increase in elimination time of this toxin facilitates diagnostic capabilities for detecting bovine aflatoxicosis by obtaining rumen contents for analysis for aflatoxin. Aflatoxin M1 was detected in rumen contents from steers at 2 hours after aflatoxin was administered. Thus, intraruminal metabolism of aflatoxin B1 to M1 may occur.  相似文献

The two possible pathways contaminating milk and milk products with mycotoxins are either the secretory or post-secretory route. The latter is of only little importance due to cooling conditions in production and storage. A secretory contamination can only occur with such mycotoxins, which undergo no complete degradation through their passage into the milk. From the mycotoxins, present in cow's feed; virtually only aflatoxin B1 yields a milkborne metabolite, the aflatoxin M1. The carry over rate is low (2 +/- 1%), but can be enhanced by polyhalogenated biphenyls, also present in the forage. Under normal conditions, however, this enhancement will not be measurable due to low equimolar concentrations of both reactants. The aflatoxin M1 content in herd's bulk milk depends exclusively on the content of the precursor aflatoxin B1 in the ration of the cow and is with less than 10 ng/kg fairly low at present in the Federal Republic of Germany. A careful supervision of the imported feed ingredients for mixed feed, however, will ensure to keep those batches out of dairy cow feeding which exceed a certain level of aflatoxin. The legal threshold is 10 micrograms/kg, being even too high to ensure a milk containing less than 10 ng/kg under high energy feeding conditions. The discussed thresholds for aflatoxin M1 in milk are 50 and 10 ng/kg resp., the latter value is scheduled for milk used in infant nutrition. To keep this low concentration the intake of aflatoxin B1 must be less than 2 micrograms/kg of the daily ration.  相似文献

1. This study was conducted to evaluate the protective effect of natural sodium bentonite (NaB) in the prevention of toxic effects of aflatoxins. Five hundred and twenty-eight 1-d-old Ross male broiler chickens were housed in pens (22 chickens per pen) for 42 d. There were 3 inclusion rates of NaB (0, 2.5, and 5 g/kg) and 2 of aflatoxins (0 and 3 mg/kg food). Each treatment had 4 replicates of 22 chickens. 2. All chickens treated with aflatoxin and without bentonite were adversely affected. NaB treatment at 5.0 g/kg improved body weights at 42 d of age by 31.3%, increased food intake by 23.8% and improved productive efficiency by 40.1%. Weights of liver, heart, pancreas and crop and biochemical variables were not affected by dietary NaB. However, serum phosphorous concentration was reduced by 30% compared with chickens that received aflatoxin. 3. NaB caused no adverse effects on chickens that did not receive aflatoxin. 4. It is concluded that NaB at pH 7.9 partially neutralises the effects of aflatoxins on broiler chickens when included at 5.0 g/kg in the diet.  相似文献

Aflatoxins are potent hepatotoxic and carcinogenic secondary metabolites produced by toxigenic fungi. The present study investigated the protective effect of methanolic leaf extracts of Monanthotaxis caffra (MLEMC) against aflatoxin B1-induced toxicity in male Sprague-Dawley rats. The rats were randomly divided into 6 groups of 8 animals each. Five groups were administered orally for seven days with three different concentrations of MLEMC (100 mg/kg, 200 mg/kg and 300 mg/kg), curcumin (10 mg/kg) or vehicle (25% propylene glycol). The following day, these groups were administered 1 mg/kg b.w. of aflatoxin B1 (AFB1). The experiment was terminated three days after administration of AFB1. Group 6 represented untreated healthy control. Serum aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, creatinine and liver histopathology were evaluated. Methanolic leaf extracts of M. caffra decreased the levels of aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase and creatinine in the sera of rats as compared with the AFB1 intoxicated group. Co-administration of MLEMC improved the histological characteristics of the hepatocytes in contrast to the AFB1 treated group, which had mild to severe hepatocellular injuries including bile duct proliferation, bile duct hyperplasia, lymphoplasmacytic infiltrate and fibrosis. Extracts of M. caffra were beneficial in mitigating the hepatotoxic effects of AFB1 in rats by reducing the levels of liver enzymes and preventing hepatic injury.  相似文献

When aflatoxin-contaminated grain is consumed by dairy cows, aflatoxin M1 is excreted in the milk. Sixteen neonatal male Holstein calves were given milk which had been collected from cows given 5 to 6 mg of aflatoxin B1 each day. The calves were examined for possible detrimental effects of the mycotoxin at pseudophysiologic concentrations. Calves were allotted to 1 of 4 groups given different milk dietary aflatoxin M1 concentrations: group 1--given 0 microgram of aflatoxin M1/L (undetectable); group 2--given 0.5 microgram/L; group 3--given 1 microgram/L; and group 4--given 2 micrograms/L. Whole milk equal to 8% of body weight was fed daily and adjusted each week to maintain this ratio. Water and a 15% crude protein complete calf starter ration were offered ad libitum for the 6-week feeding study. Weekly blood samples were collected via jugular venipuncture and analyzed for serum alkaline phosphatase and aspartate aminotransferase activities. Daily means for milk dry matter intake (in kg) and complete ration intake (in kg) for the calf groups were as follows: 0.46 and 0.36 for group 1; 0.46 and 0.25 for group 2; 0.42 and 0.18 for group 3; and 0.49 and 0.40 for group 4. Significant differences in complete ration and total dry matter intake were noted. The average daily gains (in kg) and gains in height at withers (in cm) were 0.39 and 4.1 for group 1; 0.36 and 4.0 for group 2; 0.29 and 5.7 for group 3; and 0.42 and 5.1 for group 4.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Embryotoxic effects of 25 mycotoxins were investigated in two-, three- and four-day chick embryos; the results were evaluated on the eighth day of development. The embryotoxicity ranged from 0.0001 to 0.1 microgram per embryo in T-2 toxin, aflatoxin B1, G1, B2 and M1, cytochalasin E, ochratoxin A and PR-toxin; from 0.1 to 1.0 microgram per embryo in sterigmatocystin, aflatoxin G2, vomitoxin (4deoxynivalenol), patulin, rubratoxin B, secalonic acid D, mycophenolic acid, and from 1.0 to 100 micrograms per embryo in penicillic acid, cyclopiazonic acid, tenuazonic acid, citrinine, brevianamide A, zearalenone, fusaric acid, griseofulvin, kojic acid and 8-methoxypsolaren. Acute cardiotoxic effects were observed in PR-toxin, patulin, rubratoxin B, penicillic acid, citrinine and zearalenone. Teratogenic effects with a spectrum of different embryonal malformations occurred in T-2 toxin, ochratoxin A, PR-toxin, patulin, secalonic acid D, mycophenolic acid and citrinine. The embryotoxic effects demonstrated in chick embryos correlated with the well-know literary data on mammals. Considering the different chemical composition and biological effects of mycotoxins, we suppose that the embryotoxicity test of chick embryos will also be suitable for testing other biologically active substances in the environment.  相似文献

为研究饲料中添加硫辛酸对蛋鸡开产阶段血液抗氧化及免疫功能的影响,本试验选用22周龄的雪山鸡288只,随机分为3组,每组设6个重复,每个重复16只鸡,分别饲喂不同浓度(0、300、500 mg/kg)的硫辛酸日粮。进行为期6周的饲养试验。结果表明:试验组血清中谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和免疫球蛋白M含量显著提高(P<0.05),血清总抗氧化能力(T-AOC)、免疫球蛋白G(IgG)和干扰素-γ(INF-γ)含量有升高趋势,但各组间差异不显著(P>0.05),试验2组白细胞介素-2(IL-2)、白细胞介素-4(IL-4)和γ-谷氨酰基转移酶(GGT)含量显著降低(P<0.05),肿瘤坏死因子-α(THF-α)、肌酐(CRE)和尿酸(UA)含量有降低趋势,各组间差异不显著(P>0.05)。由此可见,饲料中添加硫辛酸可以提高动物机体的健康水平。  相似文献

Five steers given 1 dose of partially purified aflatoxin at concentrations sufficient to provide 0.2 mg to 0.8 mg of aflatoxin B1 equivalents/kg of body weight were compared with 4 steers given 14 daily doses of 0.25 mg of aflatoxin B1 equivalents/kg of body weight for complement activity and bacteriostasis. Complement activity was measured by hemolysis in gel, and bacteriostatic activity, by growth inhibition of Escherichia coli in liquid medium. In the single-dose group, complement activity and bacteriostatic activities decreased by 57 hours after dosing, and both returned to near base line by 168 hours. In the daily dose group, only bacteriostatic activity decreased, and the decrease persisted 2 weeks after the last dose of aflatoxin was given. Apparently, aflatoxin affects both complement-dependent and independent serum bacteriostatic activity.  相似文献

In order to facilitate a practical large-scale introduction of the radioimmunochemical control of aflatoxin B1 contamination in cereals and feeds a new method of the preparation of sample extracts was verified. In comparison with the procedure used until now when water-nonmiscible chloroform must be evaporated from a chloroform extract and the solids must be dissolved in acetone, while using the tested method the sample is extracted by methyl alcohol or acetone and the extract is diluted by a buffer before the radioimmunoanalysis. This new method of sample extract preparation displays aflatoxin B1 recovery comparable to that of the older method (83-90% for concentrations of 2-20 micrograms per kg), the worse theoretical detection limit (1 microgram per kg vs. 0.2 microgram per kg) but it prevents the result distortion by interfering substances and it makes the whole analytical procedure simpler and expeditious. Radioimmunochemically aflatoxin was quantified by means of commercially produced analytical kits developed in one institute at Kosice.  相似文献

The efficacy of melatonin co-administration on aflatoxicosis in chicks was investigated. Ross PM3 breed chicks were divided into groups of 10 and given conventional feed. One of the groups was kept as a control (C), and the others were given 150 ppb aflatoxin (AF1), 300 ppb aflatoxin (AF2), 150 ppb aflatoxin plus 10 mg/kg/bwt melatonin (AF1+M), 300 ppb aflatoxin plus 10 mg/kg/bwt melatonin (AF2+M), 10 mg/kg/bwt melatonin (M), and 1% ethanol (E). After 21 day-treatment period, the chicks were sacrificed, liver and kidney tissues were collected, processed for immuno-histochemical staining, in situ TUNEL method, and biochemical analyses. Vacuolar degeneration, necrosis, bile duct hyperplasia in liver, and mild tubular degeneration in kidney were detected in AF groups. Pathological changes were markedly reduced in AF+M groups, and a microscopic view similar to group C was observed. Increased immunoreactivity against inducible nitric oxide synthase (iNOS) and nitrotyrosine was detected in AF groups compared to weak immunoreactivity in group C. Immunoreactivity in AF+M groups was markedly reduced compared to AF groups and was similar to group C in liver and kidney. Many apoptotic cells were detected in the livers of AF groups, whereas there were no apoptotic cells in AF+M groups. While reduced glutathione (GSH) levels in liver and kidney of AF groups were greatly reduced, malondialdehyde (MDA) levels increased. With melatonin co-administration, the levels of GSH and MDA approached to the values of group C. These results indicated that nitrosative tissue degeneration caused by aflatoxin could be greatly reduced by melatonin supplementation in chicks.  相似文献
