Fourteen developed microsatellite markers were characterized for their use in genotyping and diversity studies of olive varieties. After optimisation of microsatellite assay and allele sizing, ninety-six alleles were found in nineteen varieties, with an average of 6.8 alleles per locus. The characteristics of the microsatellite markers were used to identify markers that can be reliably applied for variety genotyping. Such features were the generation of complex banding patterns supported by underlying allele sequences, `short allele dominance', an unstable repeat structure and a low number of alleles. AFLP analysis was performed on the same set of olive varieties using eight primer pair combinations. The genetic relationships among nineteen olive varieties were compared on the basis of microsatellite and AFLP polymorphisms. Genetic distances between all pairwise combinations of the varieties were calculated using Jaccard's coefficient of similarity and dendrograms were constructed by the UPGMA method. The results of clustering analysis with both molecular systems showed the common genetic background of Tuscan varieties, and genetic divergence within Slovene olive germplasm. Slovenian varieties ‘Buga’, ‘Štorta’ and ‘Samo’ might represent regionally selected olives, while ‘Zelenjak’ and ‘Črnica’ are probably derived from the Central Italian region. The predominant local ‘Istrska belica’ was introduced to Slovenia independently from the other regional varieties and showed the lowest genetic similarity with the other regional varieties. At present, testing for distinctness, uniformity and stability (DUS) of crop varieties relies on a set of morphological characters. These characters suffer fromthe limitations of number, interaction with the environment in which the variety grows and subjectivity in decision-making. The potential of DNA-based markers such as sequence tagged microsatellite site (STMS), for establishing DUS merits investigation. In the present study, a set of 55 mapped STMS markers, selected from 12 linkage groups of rice genome, was used to examine distinctness of 23 aromatic rice genotypes including the commercially important Basmati varieties. Forty-one of these markers (74.5%) showed polymorphism between the varieties. The number of alleles per locus ranged from 2–4 with an average of 2.3. The polymorphism information content (PIC) of the markers varied from 0.083 to 0.665 with an average of 0.338. All the varieties could be differentiated from each other at a low probability (0.07×10-13) of identical match by chance. The marker-based clustering of the varieties corresponded with the known phenotypic classification, thereby providing confidence in the distinctness established by the mapped STMS markers. The utility of these markers to study uniformity and stability was analysed using a commercially important crossbred Basmati rice variety Pusa Basmati 1(IET-10364) that contributes about 40–50% of Basmati rice export from India. Genotyping of twenty individual plants, grown from the nucleus, breeder, foundation, certified and farmer's saved seed samples using all the 55 markers revealed no variation among the plants. These observations suggested that the set of mapped markers employed in this study could be further used for establishing distinctness of aromatic rice varieties and for studying DUS of the important commercial variety Pusa Basmati 1. Recently a major gene determining non-specific adult plant disease resistance against stripe rust (Puccinia striiformis) designated Yrns-B1 was mapped in wheat Triticum aestivum L. by using a cross between ‘Lgst. 79-74’ (resistant) and ‘Winzi’ (susceptible). Linkage to five Gatersleben wheat microsatellite (GWM) markers was discovered, previously mapped on chromosome arm 3BS. In the present study this map was improved by the incorporation of four additional GWM markers. QTL-analysis revealed high LOD values for the resistance at all nine loci, whereas the largest LOD (20.76) was found for the newly mapped marker Xgwm1329. Microsatellite analysis and resistance tests of a collection of old German/UK wheat varieties, including probable ancestors of ‘Lgst.79-74’ were carried out. A high coincidence of non-specific adult plant disease resistance against stripe rust and the presence of ‘Lgst. 79-74’ allele (117 bp) of the marker Xgwm533 was observed among the varieties tested. Linkage during the inheritance of both the resistance and the 117 bp allele of Xgwm533 was demonstrated. The probable origin of Yrns-B1 is discussed. Carriers of this resistance gene were grown on large areas since more than 100 years. To estimate the capability of Xgwm533 as a diagnostic marker for non-specific adult plant disease resistance against stripe rust, microsatellite analysis and resistance tests of a collection of Russian spring wheat varieties were performed. The 117 bp allele of Xgwm533 was found in about 35% of the Russian cultivars analysed, however, none of them possessed the expected disease resistance. Thus, the utilisation of Xgwm533 as diagnostic marker seems to be restricted to certain genepools.  相似文献

By analysing a number (20–38) of individuals from selected varieties of wheat and tomato, we have been able to assess intra-varietal uniformity at certain micro satellite (simple sequence repeat, SSR) loci. In total, 45 varieties of wheat were analysed at between 7–9 different SSR loci, and 10 varieties of tomato were analysed at six loci. The results showed that there was variation both between varieties and between microsatellites in the degree of non-uniformity observed, and it was possible to identify a number of different probable sources of non-uniformity. Twenty-four of the wheat varieties and nine of the tomato varieties were sufficiently uniform to meet the standards currently applied for distinctness, uniformity and stability (DUS) testing using phenotypic characteristics. The level of random amplified polymorphic DNA (RAPD) and microsatellite variation present in four ecotypes and two varieties of alfalfa (lucerne) from Italian and Egyptian germplasm sources was evaluated. A sample of 100 plants from 10 populations was analysed by means of 41 RAPD markers and 37 simple sequence repeat (SSR) markers. Both molecular approaches revealed a high degree of genetic diversity within each of the cultivated populations and enabled each of the plants considered to be uniquely fingerprinted. The genetic relationships among plants and populations were analysed by computing AMOVA (analysis of molecular variance) and FST analyses. RAPDs were able to separate the Italian populations from the Egyptian variety. SSRs allowed strong separation of the four Italian alfalfa ecotypes. It was concluded that RAPD and microsatellites could be useful and powerful tools for assessing genetic variation and genetic relationships in tetraploid alfalfa.  相似文献

Differentiation of cultivars with simple sequence repeat (SSR) markers is a very useful technique for the true-to-type characterization of cultivars and clarification of parent-offspring relationships. We developed an SSR marker set for cultivar identification comprising 15 markers that were screened from 46 previously published SSRs. This marker set could be used for apple varieties including Malus × domestica and/or other Malus species. These SSRs successfully characterized 95 apples, including the leading and major founding cultivars used worldwide for modern apple breeding. Therefore, this marker set could be applied to almost all apple cultivars. We also analyzed the parent-offspring relationships of 69 cultivars by considering allele transmissions. This analysis revealed the true parentage of the following seven cultivars: ‘Kizashi’, ‘Chinatsu’, ‘Honey Queen’, ‘Haruka’, ‘Seirin’, ‘Ozenokurenai’, and Morioka #48. This analysis also revealed a parentage discrepancy for ‘Hacnine’. From the parent-offspring analysis, two microsatellite mutation events at alleles inherited from pollen parents were observed.  相似文献

Summary Approximately 1000 seedlings from 20 combinations crossed in 1979, 1980 and 1981 (Theiler-Hedtrich 1985a) were tested for several characters: fruit set (yield), fruit size, fruit colour, formation of abscission layer and bleeding after fruit removal from fruit stalks, bacterial canker resistance, flowering and harvesting time. From progeny of crosses with Stella as pollinator, 56% (Vittoria × Stella) and 46% (Schüttler × Stella) of the seedlings were self-compatible, of which 14 were high yielding with good fruit size and quality. From the data recorded it can be concluded:fruit set is a recessive character; only 5 to 20% of very good yielding seedlings were obtained in different progeny, even if the parental plants were both very good croppers.Fruit juice and skin colour was in most progenies ‘black’ even if they were from combinations with ‘white’ varieties, e.g., Merton Glory or Schüttler. Only from the combination Schiittler (‘white’) × Stella (‘black’), 50% of the seedlings were ‘white’; Stella therefore is heterozygous for the character of fruit juice and skin colour.Fruit size is evenly distributed in progeny with respect to the fruit size of their parent plants.Abscission layer formation and non-bleeding is a genetically complex character. In combinations where both parent plants formed fruits with complete abscission layers and which were not bleeding after fruit removal from the stalk, this character was inherited only to 50% (Vittoria × Schüttler) or 85% (Vittoria × Frühe von der Weid) in the progeny. For the genetical control of this character further studies are necessary.Bacterial canker susceptibility was evenly distributed in seedlings from all combinations even if the highly resistant cv. Vittoria was used as one parent plant, thereby not confirming the expected results of a higher proportion of resistant seedlings from combinations with Vittoria.Flowering and harvest time of the seedlings from different combinations was within the range of the parent plants. Only in the combination of Vittoria × Stella (mid-to late-ripening season) one seedling out of 99 was found to form ripe fruits two weeks earlier than the parental plants. Eighty-two varieties of rice from different regions in Thailand were selected to explore the Waxy (Wx)gene diversity and indica-japonica differentiation of chloroplast DNA. A comparison of the 5 splice site in the first intron was made between glutinous and nonglutinous rice. It revealed that non-glutinous with low-amylose content and glutinous rice were characterized as the Wxb allele based on the G-to-T base substitution, whereas non-glutinous rice with intermediate and high amylose carried the Wxa allele. Four Wx microsatellite alleles, (CT)n repeat, (n = 16,17,18 and 19) were found in glutinous rice. In contrast, non-glutinous rice showed five Wx microsatellite alleles (n = 11, 16, 17, 18 and 19). The (CT)17 allele was prominent allele in Thai population, while the (CT)11 allele was found only in intermediate and high amylose rice varieties from southern Thailand. Almost all of upland rice grown by various ethnic groups in northern Thailand were characterized as japonica type based on their having the PstI-12 fragment in their cpDNA, whereas most of rainfed lowland varieties from other regions of Thailand were indica. This exploration of DNA-based genetic markers is important, as it enhances our ability to describe and manipulate sources of genetic variation for rice breeding programs.  相似文献

Microsatellites are valuable tools as molecular markers in plant breeding. To establish genetic linkage maps or for population studies, information about the occurrence and usability of microsatellite markers in different species is necessary. Sequences of spinach Spinacia oleracea from computer databases were therefore searched for the presence of microsatellites. Sixty simple sequence repeats were found in 237 spinach sequences with a total of 349.4 kb DNA. After removing duplicated sequences, 50 different microsatellites with various motifs remained. Differences between nuclear and chloroplast DNA were not in the number of microsatellites but in their type and length. Chloroplast sequences from spinach contain only short strings of A and AT repeats, whereas nuclear sequences show a wider variety of motifs. Flanking primers for polymerase chain reaction (PCR) analysis were designed for 13 of these microsatellites and tested with two different varieties of spinach. Twelve primer pairs gave amplification products and seven of these showed polymorphisms in the variety ‘Wiremona’ but only one in the variety ‘Monatol’. These markers may be used for linkage analysis or population studies in spinach.  相似文献

为了探索InDel分子标记技术在鉴定甜菜品种一致性中的可行性,本研究以8个甜菜品种为试材,每个品种取24个单株,分别提取基因组DNA,利用9对InDel引物对每个甜菜品种的24个单株分别进行InDel-PCR扩增,根据扩增条带分析每个甜菜品种的一致性分布特征。结果表明8个甜菜品种所有位点的平均一致性比率在67.7%~88.1%之间。单一引物在不同品种中表现出的一致性比率具有一定差异。例如,利用ND286引物在品种985和品种1123中检测出的一致性比率为100%,但是在其他品种中检测出的一致性比率分布在46%~63%之间。不同引物在同一品种中表现出的一致性比率可能相同,但其区分出的不一致植株可能相同也可能不同。比如,在平均一致性最好的品种1015中,ND31和ND71引物在24个植株中检测出的一致性比率相同,但ND31引物区分出的差异植株为13、14和15号植株,而ND71引物区分出的差异植株为8、15和21号植株。因此利用单个位点引物对甜菜品种一致性进行鉴定不具有普遍性和科学性,应该综合多个位点的表现进行一致性鉴定,且现阶段甜菜品种一致性鉴定的标准不宜设置过高。  相似文献

Conventional methods for the selection of papaya pure lines are time-consuming. Procedures involving the use of molecular markers and the indirect selection of homozygous plants can reduce this time considerably. The objective of this study was to evaluate the informativeness of a microsatellite marker set when used in marker-assisted selection (MAS) for the development of new papaya lines. Eighty-three lines originating from two segregating F3 populations and from papaya germplasm were used for the molecular analysis of 27 microsatellite primers. Twenty polymorphic microsatellite primers were identified, allowing the identification of 86 alleles, with an average of 3.18 alleles per primer. The observed heterozygosity values were low for both the markers (0.00–0.29) and the individual lines (0.00–0.35). The inbreeding coefficient (f) ranged from 0.634 to 1.00. Eleven lines with f = 1.00 and 18 lines with f varying from 0.953 to 0.961 were identified. In addition, papaya lines showed high genetic diversity, which will certainly contribute to the development of new varieties. Our results show that the use of microsatellites in MAS is a quick and effective procedure for the development of papaya lines.  相似文献

The elite and popular cultivars of Cymbopogon martinii were examined for genomic and expressed molecular diversity through RAPD, enzyme and SDS-PAGE protein polymorphisms. The allelic score at each locus of the enzymes as well as presence and absence profiling in RAPDs, overall occurrence of band types etc. were subjected to computation of gene diversity, expected heterozygosity, allele number per locus, and similarity matrix. These, in turn, provide inputs to derive primary account of allelic variability, genetic bases of the cultivated germplasm, putative need for gene/trait introgression from the wild or geographically diverse habitat etc. in elite selections. ‘PRC1’ possessed highest number of unique bands based on RAPD polymorphism. In variety ‘IW31245E’, diaphorase and glutamate oxaloacetate transaminase isozymes generated two unique bands as dia-III 2 and got-II 4. ‘RRL(B)77’ exhibited three unique bands; one produced by esterase as allele est-II 1 and two by malic enzyme (me-III 1,3). Only one unique band was generated by malic enzyme in variety ‘Trishna’. But sofia had three unique bands, two contributed by diaphorase (dia-II 3 and dia-II 4 and one by glutamate oxaloacetate transaminase (got-II 2). SDS-PAGE analysis revealed presence of unique polypeptide fragments (97.7 kDa to 31.6 kDa) in varieties ‘IW31245E’, ‘RRL(B)77’, ‘Tripta’, ‘Trishna’, ‘PRC1’ and var. sofia, generated as a diagnostic marker. In general, molecular distinctions associated with var. motia and var. sofia have been clearly noticed in C. martinii. Researchers interested in evaluating watermelon (Citrullus lanatus(Thunb.) Matsum. & Nakai) cultivars for yield often use multiple-row plots to simulate the monoculture system growers use or ingle-row plots to save on land, labor, and seeds. An important question is whether there is significant interaction of border rows with center rows when diverse cultivars are planted in adjacent rows. Based on recommendations from watermelon researchers in the U.S., ‘Charleston Gray’, ‘Crimson Sweet’, and ‘Sugar Baby’ were chosen to represent long, medium, and short-vined cultivars, respectively. Cultivars were planted in three-row plots with all nine combinations of the three represented in border and center rows. The experiment was a randomized complete block with the nine border by center plot combinations, two locations (Kinston, Clinton), and three replications at each location. Vine length was measured during the season, and fruit from four harvests were graded (marketable and cull), counted and weighed. Results showed that ‘Charleston Gray’ had the longest vines, followed by ‘Crimson Sweet’ and ‘SugarBaby’. In the analysis of variance, the largest effects (F ratio) on yield were from cultivar, location, and the interaction of the two. The smallest effects were border row and the interaction of center with border row. Center by border interactions were significant (5%level) in some cases, but were usually small and did not involve change in rank. Therefore, researchers interested in running trials with many cultivars and small seed quantities can obtain good data using single-row plots. However, there is a small (but significant) interaction of center with border rows in some cases, so testing at the final stage should be with trials having multiple-row plots or with cultivars grouped by vine length. Additional research is needed to determine the effect of cultivars having extreme plant types, for example dwarf cultivars in bordering rows with long-vined cultivars. Genetic capacity for green plant regeneration in anther culture were mapped in a population comprising 50 doubled haploid lines from a cross between two wheat varieties ‘Ciano’ and ‘Walter’ with widely different capacity for green plant regeneration. Bulked segregant analysis with AFLP markers and composite interval mapping detected four QTLs for green plant percentage on chromosomes 2AL (QGpp.kvl-2A), 2BL (QGpp.kvl-2B.1 and QGpp.kvl-2B.2) and 5BL (QGpp.kvl-5B).The three QTLs detected on chromosome 2AL and 2BL all derived their alleles favouring green plant formation from the responsive parent ‘Ciano’.The remaining QTL on chromosome 5BL had the allele favouring green plants from the low responding parent ‘Walter’. In a multiple regression analysis the four QTLs could explain a total of 80% of the genotypic variation for green plant percentage. None of the chromosomal regions with QTLs for green plant percentage showed significant influence on either embryo formation or regeneration frequencies from the anther culture. The three major QTLs located on group two chromosomes were fixed in a second DH population derived from two parents ‘Ciano’ and ‘Benoist’,both with high capacity to produce green plants. A QTL explaining31.5% of the genetic variation for green plant formation were detected on chromosome 5BL in this cross as well. Twenty-eight Egyptian cotton (Gossypium barbadense L.) genotypes (varieties and hybrids) were used for analysis of genetic diversity using DNA based markers (ISSR, SSR, and EST) and to study varietal development of cotton. The ISSR markers gave the highest percentage of polymorphic bands as well as polymorphic information content compared with the other molecular markers (i.e. EST and SSR markers). Using clustering analysis, no general clustering according to the pedigree history of the genotypes was observed. Using principal coordinate analysis (PCOORDA), cotton genotypes were separated by the first three principal coordinates (PC1, PC2, and PC3) accounting for 11.5, 8.6, and 7.2% of the total genetic variance, respectively. The cotton genotypes were distributed into three parts based on the first PC, each part containing a group of varieties having a common ancestor. ‘Giza 12’ variety was the common ancestor for the varieties included in the first part and ‘Ashmouni’ variety was the common ancestor for the varieties included in the second part, while both ‘Sakha 3’ and ‘Sakha 4’ varieties were common ancestors for the varieties included in the third part. The results of the PCOORDA also showed better resolution of the genetic diversity than cluster analysis especially in the illustration of the varietal development of cotton. That means that principal coordinate analysis can be strongly used either alone or in combination with cluster analysis to discuss both genetic diversity and varietal development in the cotton genotypes.  相似文献

Components of seed yield were estimated for each set of 100 plants derived from six varieties of Lolium perenne. Significant differences between the varieties were found for all components of seed yield, but the range was broader within each variety. An indirect selection of seed yield using the character seeds per spike produced a selection response similar to that obtained by direct selection on high seed yield per plant. A direct and indirect selection of characters estimated in small, drilled plots of topcross off-springs (F1) was less successful than the selection based on the single plants. Spearman rank correlation coefficients between the single plants and their offsprings did not show significant agreement for seed yield. But altogether the results indicated that the increase of seed yield/area was about 6 % when 10 % of the best plants were selected.  相似文献

A novel photoperiod response gene, designated Ppd-B2, was mapped to wheat chromosome arm 7BS, using a set of lines carrying various segments of 7BS from the early flowering breeding line ‘F26-70 7B’ in a background of the variety ‘Favorit’. The gene was 4.4 cM distal of the microsatellite locus Xgwm0537 and 20.7 cM proximal to Xgwm0255. In contrast to the well-characterized Ppd-1 genes, which require short days for expression, Ppd-B2 was detected when plants were exposed to a long photoperiod. The accelerated flowering produced by Ppd-B2 was correlated with increased grain protein content.  相似文献

Teosinte comprises different Zea species (Zea mays, Zea diploperennis, Zea perennis, Zea luxurians) that can be crossed with cultivated maize (Z. mays ssp. mays). Nine microsatellites from maize were applied to different teosinte species in order to evaluate their usefulness in markerbased exploitation of these genetic resources. The same microsatellites were tested with rye, barley, and sorghum as potential molecular markers for these species. Almost all microsatellite × teosinte combinations yielded polymerase chain reaction (PCR) fragments in the range of cultivated maize. Using an F2 population of a cross between maize inbred A188 and an individual of Zea mays ssp. mexicana, amplification products for maize and teosinte originated from the same genomic location for each of nine microsatellites investigated. PCR fragments of reduced intensity were generally obtained by applying maize microsatellites to rye, barley and sorghum. Polymorphisms among accessions within teosinte (sub)species occurred frequently. In contrast, no polymorphisms were obtained within rye, barley, and sorghum. Hence, application of maize microsatellites to teosinte for fingerprinting or marker-assisted introgression of genomic regions from teosinte into cultivated maize appears promising.  相似文献

The development of soybean varieties that lack the β-conglycinin α′ subunit is an attractive goal because the β-conglycinin α′ subunit exerts a negative influence on nutrition and tofu gelation, and is also a major allergen. We sought to develop a co-dominant DNA marker for the β-conglycinin α′ subunit (Cgy-1) gene for use in marker-assisted selection (MAS). We identified a deleted sequence responsible for the null allele of Cgy-1 in a soybean variety lacking the β-conglycinin α′ subunit known as ‘Keburi’. The deletion spanned 12,998 bp and included Cgy-1 and its incomplete tandem duplicate on chromosome 10. Based on the Cgy-1 sequence from both the Williams 82 soybean reference sequence and the Keburi variety, a set of three allele-specific primers was designed for multiplex PCR assay. These primers enabled allelic discrimination by the sizes of PCR products. This amplified two distinct DNA fragments of 913 and 649 bp in Williams 82 and Keburi, respectively. The practicality of the developed co-dominant marker for Cgy-1 was also confirmed by amplification in five other soybean varieties including three wild types and two mutants. The heterozygosity of the F1 plants at the Cgy-1 locus was ascertained using our novel co-dominant marker. This PCR-based co-dominant marker is capable of detecting the presence or absence of β-conglycinin α′ subunit for soybean marker assisted breeding system.  相似文献

Rice blast resistance gene ‘Pi-z’ present in rice genotypes, Zenith and Fukunishiki, represents a potential source of blast resistance for the north-western Himalayan region of India. We tested the reliability of microsatellite markers linked to Pi-z for assessing blast resistance phenotype in crosses of commercial importance. A new set of microsatellite markers linked to Pi-z was also developed by exploiting the publicly available marker and genomic resources of rice. Of the three previously reported markers for Pi-z, only MRG5836 was suitable for the marker assisted selection of Pi-z. Among the 17 microsatellites selected from the putative region of Pi-z locus, two, RM8225 and RM8226 cosegregated with MRG5836 and were located at distance of 1.2–4.5 cM from the gene. A new microsatellite marker ‘SSR236’ was developed from the (CT)16 repeat of PAC clone P0502B12, which exhibited closer linkage (0.6–1.2 cM) to Pi-z. Survey of the allelic diversity at the loci of the Pi-z linked microsatellite markers revealed that the Fukunishiki and Zenith type alleles were not present in majority of the local indica rice genotypes. As these markers are polymorphic between the Pi-z donors and a great majority of local indica rices tested, they can be used as a selection tool in rice breeding programs aimed at improving the blast resistance of local rices.  相似文献
