Forty gnotobiotic pigs from six litters were exposed orally to Escherichia coli 083:K·:NM at 69 to 148 hours of age, while 17 pigs from the same litters served as unexposed controls. Clinical signs of infection included fever, anorexia, diarrhea, lameness, and reluctance to move.

Eighty-four percent of the exposed pigs in four litters died, while only 13% in two litters died. Gross and microscopic lesions included serofibrinous to fibrinopurulent polyserositis in 96% of the exposed pigs in four litters and 33% of the exposed pigs in two litters. A few pigs had gross and/or microscopic lesions of arthritis. Escherichia coli was routinely isolated from the serous and synovial cavities of infected pigs.

Anti-hog cholera serum administered orally as a colostrum substitute gave partial protection against E. coli infection.

A cross-sectional study on five organized pig farms was conducted to assess the faecal carriage of ESBL and blaNDM carbapenemase-producing E. coli in piglets and pig farmworkers. Faecal samples from piglets (n = 155) and pig farmworkers (n = 21) were processed for isolation and characterization of E. coli. A total of 124 E. coli isolates from piglets and 21 E. coli isolates pig farmworkers were recovered and screening for ESBL production showed that 44.4 % (55/124) of the isolates from piglets and 42.9 % (9/21) of the isolates from farmworkers were ESBL positive. The ESBL positive isolates from piglets and farmworkers harbored blaCTX-M and also co-harbored other beta-lactams, sulphonamide, quinolone and tetracycline resistance genes. Diarrhoeic (50%, 49/98) and crossbred piglets (52.7%, 39/74) harbored a significantly higher number of ESBL producing isolates than non-diarrhoeic (23.1 %, 6/26) and purebred piglets (32%, 16/50) (p < 0.05). Piglets and pig farmworkers harbored nine and two carbapenem-resistant isolates, respectively. Interestingly, two isolates from piglets and one isolate from farmworkers harbored the blaNDM gene. The blaNDM positive E. coli isolated from piglets and farmworkers of the same farm revealed similar antibacterial resistance patterns, resistant genes, sequence (ST-167) and plasmid type (IncX3). In India, carbapenems are not used in food animal treatment, hence carbapenem resistant E. coli in piglets possibly originated from the human contact or common environment and is of public health importance.  相似文献

A retrospective study (1980-1989) was conducted to describe the clinical, pathological, and bacteriological findings in 55 cases of hemorrhagic gastroenteritis (HGE) caused by Escherichia coli in piglets. The condition occurred in weaned and suckling piglets and was associated with several serogroups of E. coli. Most of the isolates of E. coli possessed the adhesin F4 (K88) and were hemolytic. Only a few of the isolates of E. coli tested produced verotoxins. Clinical signs and pathological findings noted in these cases were compatible with shock.  相似文献

The objective of this study was to determine the associations among Haemophilus parasuis, Mycoplasma hyorhinis, and porcine reproductive and respiratory syndrome (PRRS) virus (EU-field strain) infections in 95 pigs with polyserositis. A significant association between H. parasuis and M. hyorhinis was identified. H. parasuis and M. hyorhinis were significantly more often detected in PRRS virus positive pigs.  相似文献

During a survey of preweaning mortality 538 piglets which died between birth and weaning were autopsied. The results of laboratory examinations permitted a division of the findings into a number of syndromes which were considered to be associated with the immediate cause of death. The non-infectious conditions such as trauma, starvation and suffocation were the most common. Seventy-eight piglets were included in the trauma group because of depressed cranial bone fractures or visceral rupture leading to haemorrhage into the serous cavities. An absence of alimentary tract contents was detected in each of the 92 cases of starvation, fatty metamorphosis of the liver was found in 28 while the hepatocyte cytoplasm of the remainder stained uniformally. Cyanosis and visceral congestion and haemorrhage were the main features observed in the 159 deaths ascribed to suffocation. Bacterial septicaemias and infections with enterotoxic strains of E. coli were the most prevalent infectious conditions. E. coli and beta haemolytic streptococci were the most common causes of septicaemia, being isolated from 36 and 25 cases, respectively. The post-mortem findings in these cases were non-specific except for fibrinous polyserositis which was observed in 11 of the carcasses from which E. coli was recovered. Twenty-six piglets yielded E. coli in heavy pure culture from the upper small intestine and the most common serogroup involved was 08. Encephalomyocarditis virus infection was associated with the deaths of 19 piglets. Pathologically, it was characterised by myocardial degeneration and a variable influx of mononuclear inflammatory cells. No significant lesions were found in 41 piglets.  相似文献

A commercial housed flock with an annual occurrence of pneumonia was investigated. The organism most commonly isolated from the respiratory tract of lambs up to 6 months old was Mycoplasma ovipneumoniae. Mycoplasma arginini, Mycoplasma conjunctivae, Acholeplasma laidlawii, ureaplasmas and Pasteurella haemolytica biotype A were also isolated: viruses were not isolated, but infection with parainfluenza virus type 3 (P13) was indicated by serology. Colostrum derived antibodies to M. ovipneumoniae and M. arginini declined to minimum levels by 50 days. The development of active immunity to mycoplasmas and P13 virus was associated with an increased incidence of clinical respiratory disease. Histopathological examination of the lungs from 34 lambs showed that 15 had lesions of a proliferative exudative (P.E.) pneumonia, a further 11 showed lymphoid hyperplasia sometimes associated with interstitial thickening, and eight showed no significant pathological changes. Isolations of M. ovipneumoniae were highest from animals with P.E. pneumonia, while M. arginini did not appear to be associated with any specific lung changes. P. haemolytica biotype A was isolated from all cases of P.E. pneumonia. M. ovipneumoniae, M. arginini and P. haemolytica were also isolated from the lower respiratory tract of a proportion of 31 ewes examined post-mortem, but P.E. pneumonia was not observed in these animals.  相似文献

The influence of the virulence of M. gallisepticum (Mg) was studied in multiple infections of chickens involving Mg, Newcastle disease virus (NDV) and E. coli. Separate groups of 3-week-old chickens were inoculated supra-conjuctivally with a virulent and an avirulent strain of Mg alone and in combination with the La Sota strain of NDV, the 01 serotype of E. coli, and both NDV and E. coli. In addition, chickens were inoculated with NDV alone, E. coli alone, and with NDV and E. coli; one group was left uninfected.Clinical signs, lesions, recovery of the pathogens and the serological response were observed for all groups for 3 weeks after infection and for those involving Mg alone and Mg together with NDV for 18 weeks.No clinical signs were seen in any of the birds; in each infected group some showed mild lesions of the trachea and air sacs without any marked difference among the groups.Although the numbers of birds examined were small, the virulent strain of Mg was more readily recovered than the avirulent, from the respiratory tract, in the first 3 weeks following multiple infections. However, the virulence of Mg had no influence on the recovery of the other pathogens.For the first 6 weeks after infection there was a direct relationship between the virulence of the Mg and the proportion of birds with agglutinins to the Mg rapid serum agglutination (RSA) test in single or multiple infections; multiple infections enhanced the antibody response to both virulent and avirulent mycoplasma. For NDV, multiple infections, particularly involving E. coli, enhanced the peak titre of haemagglutination-inhibition antibodies and accelerated their appearance; the virulence of the Mg had no apparent effect on this.Neither the mycoplasma nor NDV were detected in the trachea by immunofluorescence.  相似文献

Mycoplasma hyopneumoniae, the causative agent of porcine enzootic pneumonia, is present in swine herds worldwide. However, there is little information on strains infecting herds in Canada. A total of 160 swine lungs with lesions suggestive of enzootic pneumonia originating from 48 different farms were recovered from two slaughterhouses and submitted for gross pathology. The pneumonic lesion scores ranged from 2% to 84%.Eighty nine percent of the lungs (143/160) were positive for M. hyopneumoniae by real-time PCR whereas 10% (16/160) and 8.8% (14/160) were positive by PCR for M. hyorhinis and M. flocculare, respectively. By culture, only 6% of the samples were positive for M. hyopneumoniae (10/160). Among the selected M. hyopneumoniae-positive lungs (n = 25), 9 lungs were co-infected with M. hyorhinis, 9 lungs with PCV2, 2 lungs with PRRSV, 12 lungs with S. suis and 10 lungs with P. multocida. MLVA and PCR-RFLP clustering of M. hyopneumoniae revealed that analyzed strains were distributed among three and five clusters respectively, regardless of severity of lesions, indicating that no cluster is associated with virulence. However, strains missing a specific MLVA locus showed significantly less severe lesions and lower numbers of bacteria. MLVA and PCR-RFLP analyses also showed a high diversity among field isolates of M. hyopneumoniae with a greater homogeneity within the same herd. Laboratory surveillance data from the Animal Health Laboratory, University of Guelph, on the etiological diagnoses of neonatal diarrhea in piglets were analyzed to determine the relative importance and trends of different enteric pathogens in Ontario. A total of 237 cases, including live and dead 1- to 7-day-old piglets, were submitted for diagnosis of gastrointestinal illness between 2001 and 2010. The combined frequencies for cases of gastrointestinal illness involving Escherichia coli, Clostridium perfringens type A, rotavirus, and Clostridium difficile, either as single pathogens or a complex of pathogens, accounted for 56% of the total cases. In a total of 33% of cases of gastrointestinal illness, an etiological agent was not identified. The frequency of cases diagnosed with enterotoxigenic E. coli was decreased from 2007. Cases submitted in 2010 were more likely to be diagnosed with C. perfringens type A compared to cases submitted in 2002 to 2007 (P < 0.05). There was a significant trend for cases submitted in the winter to be diagnosed with C. perfringens type A, enterotoxigenic E. coli, rotavirus, and Cystoisospora suis (formerly Isospora suis) (P < 0.05). Enterotoxigenic E. coli was less likely diagnosed if C. difficile, C. perfringens, or rotavirus were detected (P < 0.05). Younger piglets were more likely to be diagnosed with C. perfringens type A (P < 0.05) and C. difficile (P < 0.05) than older piglets. This study shows that E. coli, C. perfringens type A, rotavirus, and C. difficile are enteric pathogens of concern for Ontario swine farrowing operations and further research is required to understand the reasons for the cases that are not diagnosed.  相似文献

Cellulitis caused by Escherichia coli in broilers results in substantial losses to the broiler industry in North America and Europe due to condemnations at slaughter. The objective of this study was to identify cellulitis in broilers in Sri Lanka and to characterize the E. coli from cellulitis and other colibacillosis lesions. Twenty-four farms from the low- and mid-country were selected and bacterial isolations were obtained from 241 birds. Two hundred and ninety-one gross lesions were observed in these 241 birds and 162 E. coli isolates were obtained. Cellulitis was observed in 21% of the birds. Twenty-one per cent of the birds had multiple lesions due to E. coli. The frequency of detection of other disease syndromes was 162 (67%) birds with pericarditis, 26 (11%) airsacculitis, 24 (10%) hepatitis, 12 (5%) perihepatitis, and 16 (7%) polyserositis (a combination of pericarditis, perihepatitis and airsacculitis). Serogroups O78, O2, O85 and O88 were distributed among the 32% of typable E. coli and 81% of isolates were assigned to three biotypes. Forty-four per cent of the E. coli isolates produced aerobactin and 88% demonstrated resistance to the bactericidal effect of normal chicken serum. The majority of the E. coli isolates were resistant to the antibiotics commonly used in poultry. All the E. coli isolates were non-haemolytic and 25% of the isolates produced K1 capsule. This study demonstrated the presence of cellulitis in Sri Lanka and this report describes some of the phenotypic characteristics of the E. coli isolates.  相似文献

1. The aim of this study was to determine the most likely time interval after infection with influenza virus H9N2 for co-infection with Escherichia coli to cause colibacillosis, the importance of lung load of E. coli and the involvement of respiratory phagocytes.

2. Specific pathogen free chickens were inoculated intranasally with 106EID50 of influenza virus or uninfected. After specified time intervals, 107 CFU E. coli or phosphate-buffered saline was inoculated. The presence of lesions, the number of respiratory phagocytes in the respiratory lavage fluid and the E. coli load in the lung were determined after different time intervals.

3. Compared with the number of lesions in chickens receiving only E. coli inoculation, the number lesions in co-infected chickens were increased at 0- and 3-d time intervals, but reduced in the groups at 6- and 9-d intervals between co-infection.

4. At 1–3 d after E. coli inoculation, the number of lesions chickens was correlated with the number of respiratory phagocytes harvested and related to the E. coli load in the lungs at 5 d.

5. These results suggest that the lesions caused by E. coli in chickens were increased within a 0–3 d interval following H9N2 virus inoculation and that this effect is related to the number of respiratory phagocytes.  相似文献

A blocking ELISA was developed by using a monoclonal antibody (4082-05-344-18) which specifically detected an epitope on the Mycoplasma hyppneumoniae 40 kDa membrane protein without cross-reacting with M flocculare or M hyorhinis. The results obtained with sera from specific pathogen-free pigs inoculated with M flocculare or M hyorhinis confirmed the specificity of the assay. An immunoblotting procedure was used to characterise the antibody response of pigs experimentally infected with M hyopneumoniae. Antibodies to the 40 kDa antigen were detected two weeks after infection and remained as major markers for at least 20 weeks. Cross-reacting antibodies to this antigen were not detected in convalescent sera from piglets infected with M flocculare or M hyorhinis. Sera from experimentally infected pigs were compared by means of the blocking ELISA and an indirect ELISA. The kinetics of ELISA antibodies after experimental inoculation were also studied. The detection of antibody was rather more stable for a longer time with the blocking ELISA than with the indirect ELISA. In an evaluation of more than 1000 sera from the field there was excellent agreement between the two methods.  相似文献

Escherichia coli was isolated from the feces of 122 piglets with diarrhea on 55 farms in Korea. The virulence genes of each isolate were characterized by polymerase chain reaction (PCR). Of the 562 isolates, 191 carried 1 or more of the virulence genes tested for in this study. Of the 191 isolates, 114 (60%) carried 1 or more of the genes for enterotoxigenic E. coli (ETEC) fimbriae F4, F5, F6, F18, and F41 and ETEC toxins LT, STa, and STb, 57 (30%) carried 1 or more of the genes for the Shiga-toxin-producing E. coli (STEC) toxins Stx1, Stx2, and Stx2e, and 21% and 37% carried the gene for enteropathogenic E. coli intimin and for enteroaggregative E. coli toxin, respectively. Collectively, our results indicate that other pathotypes of E. coli as well as ETEC can be strongly associated with diarrhea in piglets. In addition, detection of the genes for Stx1 and Stx2 indicates that pigs are reservoirs of human pathogenic STEC.  相似文献

This study aimed at evaluate the presence and to study characteristics of Escherichia coli in the respiratory system microbiota of healthy broilers. Trachea, air sacs, and lungs of 20 broilers were analyzed at 21 days of age, reared in experimental conditions, without receiving antimicrobials. E. coli strains were isolated and identified using conventional bacteriology through morphological and biochemical characterization. The production of bacteriocin-like substances, the presence of virulence-associated genes (VAGs) of APEC (Avian Pathogenic Escherichia coli) predictors, and the antimicrobial susceptibility were evaluated. E. coli was found in 85 % of the animals (17/20), in the trachea, air sacs or lungs; and it was not found in 15 % of the animals (3/20). A total of 34 isolates were recovered, 13 from the air sacs, 13 from the lungs, and 8 from the trachea, which showed no production of bacteriocin-like substances nor virulence genes associated with APEC. Most isolates, 59 % (20/34), showed resistance to at least one of the tested antimicrobials, and six multiresistant strains were identified. The results demonstrated that strains of E. coli were commensal of the respiratory microbiota, and that they did not present pathogenicity to the host, since there were no clinical signs of disease, macroscopic lesions in the organs of the evaluated broilers, production of bacteriocin-like substances, nor virulence-associated genes considered as predictors of APEC in bacteria. These strains of E. coli were mostly susceptible to antimicrobials. However, the occurrence of multidrug-resistant strains suggests that these animals can act as reservoirs of resistant to antimicrobials E. coli.  相似文献

An optimized protocol was developed for the simultaneous detection and differentiation of Haemophilus parasuis, Streptococcus suis, and Mycoplasma hyorhinis in formalin-fixed, paraffin-embedded (FFPE) tissues with multiplex nested polymerase chain reaction (PCR). This method also determines the prevalence of these bacteria in pigs with polyserositis. DNA extraction with a combination of a commercial reagent and proteinase K resulted in more frequent detection of the pathogens than DNA extraction with proteinase K alone. Among FFPE tissue samples from 312 cases of polyserositis in which at least 1 bacterial species was detected, multiplex nested PCR detected H. parasuis in 239 (77%), S. suis in 124 (40%), and M. hyorhinis in 40 (13%). The disease was caused by a single pathogen in 224 (72%) of the cases and multiple pathogens in 88 (28%). Among the pigs positive for H. parasuis, S. suis, and M. hyorhinis by multiplex nested PCR, the pathogen was isolated from only 11%, 35%, and 28%, respectively. Therefore, the PCR protocol developed in this study is a useful diagnostic method when samples are negative after isolation methods and even for samples in which only 1 pathogen was isolated.  相似文献

Air quality, respiratory disease, and growth rate were followed in four different farrowing and nursery systems. Ammonia levels varied with ambient air temperature, but were within normally accepted levels (25 ppm). These levels of ammonia did not appear to affect the health or performance of the pigs raised in these units. Hydrogen sulfide levels were consistently low. Counts of bacterial colony forming particles (BCFP) varied and the organisms identified were predominantly micrococci. Bordetella bronchiseptica was isolated from nasal cavities of pigs from 3 out of 4 farms. Three of the farms did not have evidence of atrophic rhinitis; pigs farrowed in the last quarter of the test year on one farm from which B. bronchiseptica was isolated developed lesions of atrophic rhinitis. The B. bronchiseptica isolates from the 3 farms were virulent for gnotobiotic piglets. Groups of pigs for slaughter inspection from one farm had lungs with 11–28% pneumonic lesions; these lesions were not typical of mycoplasmal pneumonia.  相似文献

Mycoplasma suis (M. suis), a hemotrophic pathogen of pigs, causes economic losses in swine production throughout the world. Inorganic pyrophosphatase (ppa) is a very important gene in M. suis. The ppa gene of M. suis was synthesized by PCR-based accurate synthesis (PAS) and overlapextension PCR, inserted into vector pMD18-T, and then subcloned to the prokaryotic expression vector pET28c.The recombinant plasmid pET28c_ppa was transformed to E. coli BL21 for expression under induction of isopropyl thiogalactoside. The expressed product was identified by SDS–PAGE and Western blot, which suggested that the recombinant protein has good antigenicity. Piglets were immunised with purified recombinant protein, and specific antibodies to the recombinant protein were detected in piglet serum. The results show that the ppa gene can be efficiently expressed in E. coli and that the expressed recombinant protein can elicit a specific serum antibody response in piglets. PAS and overlap-extension PCR were first used to synthesize the ppa of M. suis. They provide simple, rapid, reliable and relatively inexpensive methods to synthesize, clone, and express genes. The experiment conducted in this paper will enable future research into the role and function of the ppa gene.  相似文献

A cross-sectional survey on the prevalence of diarrhoea in pre-weaning piglets, and on management and feeding systems under farm conditions was carried out in Thua Thien Hue Province. Faecal samples were collected from 63 piglets without, and 90 piglets with diarrhoea to determine the occurrence of Salmonella, Escherichia coli (E. coli) and different E. coli antigens (K88, K99 and 987P). The prevalence of diarrhoea was higher in the rainy season than in the dry season (33% vs 18%) and the results indicated differences in prevalence between areas. Salmonella and E. coli were found to the same extent in faeces from piglets without and with diarrhoea. All E. coli antigens were isolated from piglets without and with diarrhoea. However, the frequency was much higher in piglets suffering from diarrhoea. In piglets with diarrhoea antigen K88 was found in 26% and 20% of the samples, antigen K99 in 37% and 24% of the samples, and antigen 987P in 31% and 32% of the samples collected in the dry and rainy seasons, respectively. Nutrient supply for sows and for piglets was low in comparison with feeding standards, which may be a contributory factor to the high incidence of diarrhoea in piglets. Thus, the nutrition of sows as well as piglets could be important components in the aetiology of the disease and needs to be studied further.  相似文献

Mycoplasma (M.) hyorhinis and M. hyosynoviae are pathogens known to cause disease in pigs post-weaning. Due to their fastidious nature, there is increased need for culture-independent diagnostic platforms to detect these microorganisms. Therefore, this study was performed to develop and optimize quantitative real-time PCR (qPCR) assays to rapidly detect M. hyorhinis and M. hyosynoviae in pen-based oral fluids as well as nasal and tonsillar fluids as proxies for samples used in swine herd surveillance. Two methods of genomic DNA extraction, automated versus manual, were used to compare diagnostic test performance. A wean-to-finish longitudinal study was also carried out to demonstrate the reproducibility of using pen-based oral fluids. Overall, pen-based oral and tonsillar fluids were more likely to be positive for both types of bacteria whereas only M. hyorhinis was detected in nasal fluids. DNA extraction protocols were shown to significantly influence test result. Although the initial detection time somewhat differed, both organisms were repeatedly detected in the longitudinal study. Overall, this study evaluated two qPCR methods for rapid and specific detection of either mycoplasma. Results from the present investigation can serve as a foundation for future studies to determine the prevalence of the two microorganisms, environmental load, and effectiveness of veterinary interventions for infection control.  相似文献
