以番鸭呼肠孤病毒MW9710分离株原核表达蛋白pGEX-4T-1-σC为靶分子,利用噬菌体随机七肽库进行亲和筛选,经过4轮亲和筛选后,对整个被选噬菌体集合单链DNA进行测序。结果:噬菌体展示序列为CATCCTATTCATCCGCGTCAT,相应的短肽序列为HPFYSCY,该短肽序列与MDRVσC氨基酸序列N端一处-P-YS--的氨基酸片段相似,推论MDRVσC的模拟抗原表位可能是由该不连续氨基酸片段所构成的构象表位。通过Genbank的蛋白质检索,发现该构象表位也存在于前B细胞克隆增强因子的多肽序列中。结果表明,噬菌体展示随机肽库技术是一种用于研究抗原表位结构的有效方法。  相似文献

肽是分子结构介于氨基酸和蛋白质之间的一类化合物,含2或3个氨基酸残基的为小肽。小肽可能存在3种转运系统。反刍动物吸收肽的主要部位是瓣胃。小肽本身的理化性质、动物生理状态、日粮蛋白和采食水平等影响小肽的吸收。小肽可避免氨基酸间的吸收竞争,促进氨基酸的吸收,加速蛋白质的合成,促进肠道黏膜结构和功能发育,刺激消化酶的分泌和活性的提高,改善提高生产性能。对小肽的吸收机制及营养功能进行了综述。  相似文献

【目的】苹果着色是一个重要的质量性状,构建苹果磷酸化蛋白质组并进行鉴定与分析,了解在苹果着色过程中的蛋白质磷酸化,为进一步阐明苹果着色机理提供参考。【方法】以着色‘嘎拉’和非着色‘金冠’两个品种苹果果皮为试材提取总蛋白,用TiO2富集磷酸化肽段,对富集到的磷酸化肽段进行高效液相色谱分离,利用质谱鉴定磷酸化肽段,采用Mascot2.2和Proteome Discoverer1.4(thermo)软件进行蛋白质查库鉴定,使用的数据库为Uniprot_Maloideae.fasta。对鉴定到的磷酸化肽段和蛋白质进行分析,找出着色与非着色苹果品种中有差异的磷酸化蛋白质及其修饰位点。【结果】在着色苹果品种中鉴定到1 323个蛋白质、3 339个肽段和225个磷酸化肽段,发现磷酸化蛋白质约200-225种;在非着色苹果品种中鉴定到569个蛋白质、1 152个肽段和128个磷酸化肽段,发现磷酸化蛋白质约117-128种。比较分析表明,43个磷酸化肽段仅在非着色苹果品种中发现,139个磷酸化肽段仅在着色苹果品种中发现,在着色苹果品种中有更多的蛋白质发生了磷酸化,其中质膜H+-ATPase在两个苏氨酸位点发生磷酸化,过敏原蛋白和两个蛋白激酶在丝氨酸位点发生磷酸化,3个转录因子、1个钾转运蛋白和1个受体激酶都发生磷酸化。在两个苹果品种鉴定出的蛋白质中,发生蛋白质磷酸化的位点主要是丝氨酸,其次是苏氨酸,在酪氨酸位点发生蛋白质磷酸化很少。蛋白质WD40和花青苷合成途径中的相关酶蛋白C4H、4CL、CHS、CHI、F3′H、FLS、LDOX和UFGT在着色过程中未发生磷酸化。在着色和非着色苹果品种中都存在MYBR转录因子,并且都发生了蛋白质磷酸化。【结论】在着色苹果品种中产生了一些特有的磷酸化蛋白质,其中包括已知与着色相关的质膜H+-ATPase,苹果着色可能受到蛋白质磷酸化的影响。  相似文献

【目的】证实蓝细菌Anabaenasp.PCC 7120复制酶中分离的DnaE内含肽的自我剪接活性。【方法】分别将AnabaenaPCC 7120中分离的2个dnaE基因的部分序列克隆到表达载体上,获得重组表达质粒,经超量表达和纯化后,免疫家兔获得相应的抗体;同时,两段分离的dnaE序列被克隆到同一表达载体,构建双重组表达质粒,两段序列具有各自的ORF,但由同一启动子调控。双重组表达质粒经诱导之后,利用以上获得的抗体对含有内含肽的DnaE蛋白在大肠杆菌表达体系中的自我剪接反应进行检测。【结果】在双重组表达质粒的表达产物中,抗体不仅可以分别与对应的蛋白质反应,而且还可以同时识别1条新条带,该条带的表观分子量与成熟DnaE的理论值大致相符。【结论】蓝细菌分离的DnaE内含肽在大肠杆菌表达体系中可以进行自我剪接;本研究中获得的抗体具有较好的特异性,无明显背景干扰,而且效价高,抗体可以稀释1 000倍以上使用,也可以用于相关方面的研究。  相似文献

Noniodinated subunits of thyroglobulin can aggregate, but iodination of the aggregate is required for its stabilization (maturation). Rat-thyroid slices incorporate amino acids into subunits, but cannot form mature thyroglobulin from the newly synthesized subunits. This defect leads to an accumulation of 16S and 12S proteins, although the preexisting thyroglobulin is 19S. Accumulation of 16S and 12S proteins can be produced in rat thyroids by the administration to the animals of a thiocarbamide derivative, methimazole. Upon withdrawal of methimazole, iodination of the 16S and 12S proteins proceeds, and 19S protein appears.  相似文献

N-glycosylation of eukaryotic proteins helps them fold and traverse the cellular secretory pathway and can increase their stability, although the molecular basis for stabilization is poorly understood. Glycosylation of proteins at na?ve sites (ones that normally are not glycosylated) could be useful for therapeutic and research applications but currently results in unpredictable changes to protein stability. We show that placing a phenylalanine residue two or three positions before a glycosylated asparagine in distinct reverse turns facilitates stabilizing interactions between the aromatic side chain and the first N-acetylglucosamine of the glycan. Glycosylating this portable structural module, an enhanced aromatic sequon, in three different proteins stabilizes their native states by -0.7 to -2.0 kilocalories per mole and increases cellular glycosylation efficiency.  相似文献

The crystal structure of the double-stranded DNA bacteriophage HK97 mature empty capsid was determined at 3.6 angstrom resolution. The 660 angstrom diameter icosahedral particle contains 420 subunits with a new fold. The final capsid maturation step is an autocatalytic reaction that creates 420 isopeptide bonds between proteins. Each subunit is joined to two of its neighbors by ligation of the side-chain lysine 169 to asparagine 356. This generates 12 pentameric and 60 hexameric rings of covalently joined subunits that loop through each other, creating protein chainmail: topologically linked protein catenanes arranged with icosahedral symmetry. Catenanes have not been previously observed in proteins and provide a stabilization mechanism for the very thin HK97 capsid.  相似文献

During 1–7-day stages of ontogenesis (for example, in winter rye), biochemical changes in the aerial part of perennial grasses with adaptive Triticum-type proteins in the process of entering into the state of anabiosis were studied. It was shown that, at this point, biomass of the aerial part of plant was much higher than of the root system, and it accumulated raffinose, sucrose, glucose, and half-cystine. Synthesis of raffinose and accumulation of half-cystine continued until the stabilization of negative temperatures; as for sucrose and glucose, it ceased upon reaching the average temperature of the physiological zero, then the content of half-cystine and raffinose increased sharply, and the content of sucrose and glucose decreased at the same time. In the aerial parts of plants, fructose and phenylalanine were found, and their content increased upon reaching freezing temperatures. It was concluded that the polymerization of water-soluble proteins occurred in perennial grasses in a state of anabiosis. These proteins, along with raffinose, serve as cryoprotectants; sucrose and glucose are substrates for respiration; fructose is a substrate for respiration and protective metabolite; and phenylalanine is a substrate for the future synthesis of antioxidants.  相似文献

The beak of the Humboldt squid Dosidicus gigas represents one of the hardest and stiffest wholly organic materials known. As it is deeply embedded within the soft buccal envelope, the manner in which impact forces are transmitted between beak and envelope is a matter of considerable scientific interest. Here, we show that the hydrated beak exhibits a large stiffness gradient, spanning two orders of magnitude from the tip to the base. This gradient is correlated with a chemical gradient involving mixtures of chitin, water, and His-rich proteins that contain 3,4-dihydroxyphenyl-L-alanine (dopa) and undergo extensive stabilization by histidyl-dopa cross-link formation. These findings may serve as a foundation for identifying design principles for attaching mechanically mismatched materials in engineering and biological applications.  相似文献

Green fluorescent protein (GFP) and its derivatives have transformed the use and analysis of proteins for diverse applications. Like proteins, RNA has complex roles in cellular function and is increasingly used for various applications, but a comparable approach for fluorescently tagging RNA is lacking. Here, we describe the generation of RNA aptamers that bind fluorophores resembling the fluorophore in GFP. These RNA-fluorophore complexes create a palette that spans the visible spectrum. An RNA-fluorophore complex, termed Spinach, resembles enhanced GFP and emits a green fluorescence comparable in brightness with fluorescent proteins. Spinach is markedly resistant to photobleaching, and Spinach fusion RNAs can be imaged in living cells. These RNA mimics of GFP provide an approach for genetic encoding of fluorescent RNAs.  相似文献

针对欠驱动机器人Acrobot,提出一种基于线性矩阵不等式的鲁棒镇定控制方法。通过将Acrobot在垂直向上不稳定平衡点附近的第一杆角速度看作一种不确定性,得到Acrobot的不确定模型,在此基础上设计一种基于线性矩阵不等式的鲁棒镇定状态反馈控制律,实现Acrobot较大范围的平衡控制。仿真和对比结果验证了方法的有效性和优越性。  相似文献
