1. The influence of in-feed lactoferrin (Lf) on bird production, intestinal microbiota, mucosal immune system and gut microarchitecture was assessed in male Cobb 500 broiler chickens.

2. Birds were given one of four diets from day of hatch: Control (basal diet with no additives), ZnB (basal diet?+?50?mg/kg zinc bacitracin), Lf 250?mg/kg (basal diet?+?250?mg/kg Lf) and Lf 500?mg/kg (basal diet?+?500?mg/kg Lf); n?=?24 birds/treatment. An apparent metabolisable energy study was performed between d 25-32.

3. Lf did not affect growth rate or feed conversion in the period 0-21?d of age, nor performance or energy metabolism during the 7?d metabolism experiment which commenced at 25?d of age.

4. The profiles of caecal microbial communities were significantly different in birds given ZnB compared with birds given a diet with no additives, or supplemented with 250?mg/kg Lf. Birds given 250?mg/kg Lf also had a different microbial profile compared with birds given 500?mg/kg Lf.

5. In comparison to control birds, Lf treated birds showed some differences in the T cell proportions in caecal tonsil and spleen. No differences in ileal villus height, crypt depth or goblet cell proportions were observed amongst dietary treatments.

6. Whilst Lf had little effect on the measured parameters, the use of an integrated approach to study the influence of novel feed additives may facilitate a greater understanding of the relationships between nutrition, gut health and bird performance.  相似文献

1. The gut microflora of chicks fed on a purified diet containing 300 g lactose plus 300 g starch/kg was compared with that of control birds receiving a diet containing 600 g starch/kg.

2. In 14‐d‐old conventional chicks, lactose in the diet decreased the incidence of lactobacilli and clostridia in the caecal contents, although when present in lactose‐fed chicks the counts of lactobacilli exceeded those of control chicks.

3. High counts of Proteus sp. were present in the caeca of control birds but they were completely suppressed in conventional birds fed on the lactose diet. In vitro tests showed that this inhibition was partially due to Escher‐ichia coli and Streptococcus faecalis.

4. The growth of Lactobacillus acidophilus was inhibited by lactose when gnotobiotic chicks were monoassociated but not when polyassociated. The protective effect was shown in vitro to be due to L. salivarius.

5. The pH was markedly lowered in the caecum of conventional and polyassociated chicks receiving dietary lactose. Of the strains used in gnotobiotic experiments E. coli, S. faecalis and L. salivarius produced the lowest pH values in the caeca.  相似文献

1. The clinical severity, histological changes, indicators of gut leakiness and inflammatory cytokine profiles were studied in chickens with dextran sulphate sodium (DSS)-induced intestinal inflammation.

2. The experimental groups (1.25%, 1.5% and 2.5% DSS) showed clinical signs, such as loose stools and weight loss, which increased with additional treatment days and, as expected, the effects of DSS-induced intestinal inflammation were time and dose-dependent.

3. After 10 d, histological manifestations were evident, including goblet cell depletion, mucus layer loss, significantly shorter villi and a thinner total ileal mucosa.

4. The d(-)-lactate value, which was used as a gut leakiness indicator, was significantly increased in the 2.5% DSS group.

5. Expression of the inflammatory cytokines interleukin-1Beta, tumour necrosis factor alpha and interleukin-10 in the serum significantly increased with DSS treatment.

6. This study indicates that the experimental intestinal inflammation induced by DSS is an ideal model to study the pathogenic mechanisms of intestinal inflammation in chickens and to test the efficacy of therapies.  相似文献

1. The present study investigated the effects of encapsulated benzoic acid (BA) supplementation in broiler feed on performance and gastrointestinal microbiota.

2. Eighty broilers were randomly divided into two groups. Birds in the control group were fed on maize–soybean-based diets. Birds in the treatment group were provided the same diet supplemented with 2 g/kg BA encapsulated in a vegetable oil matrix.

3. At the end of the trial (d 35), pH, bacterial composition and metabolites were determined in the crop, jejunum, ileum and caecum.

4. Growth performance variables and pH were not significantly different.

5. BA concentration decreased rapidly in the proximal gut. However, the treatment diet showed higher BA in the crop, jejunum, ileum and caecum.

6. Total lactate in the crop and D-lactate in the jejunum was higher in the BA treated group. Caecal total and branched chain fatty acids were decreased due to the treatment.

7. Lactobacilli populations were significantly altered by BA supplementation. A trend for increased lactobacilli was observed in the crop, while it became significant in the jejunum and ileum. Lactobacillus species responded differently to the treatment. Four of 5 measured Lactobacillus species, particularly in the ileum, followed the course observed for total lactobacilli; only Lactobacillus salivarius was not modified.

8. Correlation analysis showed that BA modified the intestinal microbiota. Lactobacilli correlated negatively to all studied clostridial clusters and enterobacteria. Clostridial clusters IV and XIVa were significantly increased in the jejunum, whereas only clostridial cluster XIVa was increased in the caecum.

9. Encapsulated BA modified the intestinal microbiota which can lead to the conclusion, that the main beneficial mode of action of BA in the gut appears to be the enhancement of lactic acid bacteria, which in turn may act as a vanguard against pathogens.  相似文献

1. Particle size reductions of fibre-rich materials alter structure, functional and digestive properties. To determine the effects of using fibre as an additive in Japanese quail rations on performance and gut physiology, a trial using micronised wheat fibre (MWF) at levels of 0.0, 5, 10 and 15 g/kg in feed was conducted.

2. Growth rate and feed efficiency were significantly improved when diets contained MWF while feed intake was not affected by levels of the fibre. As MWF content increased, the relative weight of gizzard and gastrointestinal tract (GIT) significantly increased whereas liver relative weight significantly decreased.

3. MWF inclusion significantly increased relative length of gut segments, villi height, villus thickness, the villi height to crypt depth proportion in jejunum and ileum and the number of goblet cells in different parts of intestine.

4. Tibia weight, length and ash content were increased linearly with rising MWF inclusion. Litter moisture was affected by MWF inclusions in a quadratic manner. The colony forming unit (CFU/g) of Streptococci spp. in ileal digesta was decreased with increasing MWF inclusion levels in the diet.

5. In conclusion, MWF can be used as a feed additive in quail diets and its inclusion in feed resulted in better performance, beneficial changes in intestinal microbial counts and improvements in small intestine morphology.  相似文献

1. The mechanism of the anti‐nutritive activities of soluble non‐starch polysaccharides (NSPs) in broiler diets was investigated with emphasis on the inter‐relationship between viscosity and fermentation along the gut. Isolated soluble NSP were added to a control diet to effect high gut viscosity, and in vivo depolymerisation of the NSP was achieved using a commercial glycanase.

2. Addition of soluble NSPs significantly (P<0.01) increased gut viscosity, reduced the AME of the diet and depressed the growth and FCE of the birds. Enzyme supplementation of the NSP‐enriched diet reversed the adverse effects, increasing (P< 0.01) weight gain, FCE and AME. Comparisons of the viscosities (mPa) in birds fed on the NSP‐enriched diet and the same diet supplemented with enzyme were respectively: 11–9 v. 2–3 in the duodenum; 78–3 v. 4–4 in the jejunum and 409–3 v. 10–8 in the ileum.

3. Caecal volatile fatty acid concentration was markedly (P<0.01) elevated by enzyme supplementation, whereas ileal fermentation was inhibited.

4. Microscopic examination revealed that, among birds fed on the NSP‐enriched diet, there had been extensive small intestinal fermentation, which was eliminated by the enzyme supplementation.

5. Addition of a synthetic antibiotic (Amoxil) had no beneficial effects.

6. The current study demonstrated that increased fermentation occurs in the small intestine when a large amount of viscous NSPs is present in the diet and this is detrimental to the performance and well‐being of poultry.  相似文献

1. The effects of microbial phytase on the performance and nutrient utilisation in broilers fed on phosphorus-adequate starter diets were examined in this study. The effect of phytase on the apparent ileal digestibility of fatty acids was of particular interest. Two grain types (maize and wheat) and two inclusion concentrations of a phytase enzyme from Escherichia coli expressed in Schizosaccaromyces pombe (0 or 500 phytase units (FTU)/kg of feed) were evaluated in a 2 × 2 factorial arrangement of treatments.

2. Supplemental phytase improved the weight gain and feed per gain, but had no effect on the feed intake of young broilers receiving phosphorus-adequate diets.

3. Phytase supplementation improved the apparent ileal digestibility of nitrogen and phosphorus in both diet types. Phytase supplementation tended to improve the apparent ileal digestible energy in wheat-based diets, but had no effect on the apparent metabolisable energy in both diet types.

4. Supplementation of phytase increased the apparent ileal digestibility of fat, with similar effects for the different fatty acids measured. Increments on ileal fat digestibility due to phytase were not dependent on the type of diet.

5. Dietary supplementation of microbial phytase enhanced not only the digestibility of phosphorus, but also that of nitrogen and fat, exhibiting increased ileal digestibility for all fatty acids in P-adequate maize- and wheat-based diets.  相似文献

1. Five dietary treatments were used in a 49 d broiler trial to assess the effect of xylanase on performance, nutrient digestibility and thermal profiles of the caeca and head. Treatments included an industry-standard control diet and four further diets where xylanase was introduced with or without a metabolisable energy density dilution either from day one or the introduction was delayed until d 28.

2. The addition of xylanase with no associated energy dilution from day one resulted in the most consistent beneficial effects on performance, with significant improvements in weight gain compared with the industry-standard to d 28 and at d 49. Addition of xylanase from d 28 (with no energy dilution) was the second most successful strategy and resulted in a significant improvement in feed conversion ratio (FCR) from d 29 to 49 and overall.

3. Addition of xylanase improved ileal digestible energy values at d 28 by around 0.35 MJ/kg and ileal nitrogen digestibility coefficients by around 3%. On d 49 xylanase improved ileal digestible energy values by around 0.9 MJ/kg and ileal nitrogen digestibility coefficients by around 4.6%.

4. Thermal imaging of the head and caeca of three birds per replicate on d 49 revealed a significant increase in caecal surface temperature following xylanase addition with no effect on head temperature profile. These increases were particularly large (around 1.4ºC, or 3.9%) when xylanase was added from day one with no corresponding energy dilution in feed formulation.

5. It can be concluded that supplemental xylanase is effective in improving performance and nutrient digestibility in broilers given wheat-based diets. The correlation between the magnitude of this effect and the increased temperature in the caeca presents additional evidence that the hind-gut microflora may play an important, if yet unquantified, role in the outworking of these mechanisms.  相似文献

1. The influences of genotype, age and sex on droppings digestibility coefficients of a compound food were studied using male and female broiler chickens of three different genotypes at 2, 4 and 6 weeks of age.

2. Because the traditional method of determination of droppings digestibility coefficients of nitrogen may lead to systematic errors in estimating the feeding value of foodstuffs, a method is proposed to determine the ileal digestibility coefficients. The ileal method is compared with the droppings method for a mixed food and for two foodstuffs: wheat and solvent‐extracted soyabean meal.

3. Birds selected for efficient food conversion showed distinctly higher digestibility coefficients for all nutrients than birds selected for high growth potential or birds from a commercial strain.

4. The influence of age on digestibility coefficients was not consistent.

5. Female birds showed digestibility coefficients which were, in general, 3% higher than those of male chickens.

6. Interactions between genotype and sex and between genotype and age for energy metabolisability were the only interactions observed for digestibility measurements.

7. The method of determination influenced the amino acid digestibility coefficients of the mixed food and the relative feeding values of wheat and soyabean meal.

8. It is important to use well defined animals (genotype, sex, age) in evaluating foodstuffs.

9. The preferred method for determination of digestibility coefficients of nitrogen and amino acids is based on ileal sampling, although the differences in amino acid digestibility coefficients were small between methods.  相似文献

1. A 2 × 2 factorial design was used to test the hypothesis that impaired intestinal starch digestibility is attributable to rapid passage of digesta from the gizzard to the intestine, and that, compared to steam pelleting, increasing the availability of starch through extrusion cooking may alleviate the potential negative effect of rapid digesta flow on starch utilisation.

2. Thus, 7-d-old-broiler chickens were distributed to 48 cages and given a wheat-based (WB) pelleted diet containing either coarse oat hulls (OH-Pel) or fine cellulose (Cel-Pel) until d 19 to stimulate divergent development of the gizzard. Thereafter, both groups were further subdivided and challenged with a WB diet containing cellulose in either pelleted (Cel-Pel) or extruded (Cel-Ext) form on d 20 and 22. Either excreta or intestinal contents were collected at time intervals after feeding and analysed for marker and starch.

3. OH-Pel increased gizzard size and holding capacity. No excessively high starch levels (maximum 25 g/kg) were detected in the excreta. However, 8 h feed-deprived birds given Cel-Pel and challenged with Cel-Pel exhibited higher starch excretion and showed large individual variation during the first 135 min of collection.

4. Contrary to the OH-Pel group, more digesta and starch passed to the jejunum at 1 and 2 h and ileum at 2 and 3 h after feeding for birds given Cel-Pel, resulting in lower jejunal and ileal starch digestibility.

5. Increased starch gelatinisation through extrusion processing significantly improved starch digestibility regardless of gizzard function. However, at 1, 2 and 3 h after feeding, more digesta was retained in the foregut of birds given Cel-Ext.

6. The current data showed that starch degradation rate is associated with the flow of digesta which is linked to gizzard development, and that enzymatic hydrolysis of intact starch granules may be limited with more rapid feed passage through the gut.  相似文献

1. Three experiments were carried out to study the effects of two experimental yeast cell wall (YCW) supplements, one from the yeast extract industry and the other from the brewery industry, added to maize or wheat based-diets, on performance and intestinal parameters of broiler chickens (Ross 308).

2. In the first and second experiments, a completely randomised block design with 4 experimental treatments was used: T-1) Negative control, no additives T-2) Positive control, avilamycin group (10 mg/kg feed), T-3) Yeast extract-YCW (500 mg/kg), and T-4) Brewery-YCW (500 mg/kg feed). There were 6 replicates of 20 (experiment 1) and 22 (experiment 2) chicks per treatment.

3. In experiment 1 (wheat based diets), yeast extract-YCW increased BW and daily feed intake (42 d). The effects were comparable to those of avilamycin. In experiment 2 (maize based diet), avilamycin, yeast extract-YCW and brewery-YCW treatments improved the feed conversion ratio with respect to the negative control group (0 to 14 d).

4. At 24 d, in both experiments, the ileal nutrient digestibility and ileal bacterial counts were not affected by any experimental treatment. In maize diets, lower intestinal viscosity was obtained with avilamycin, yeast extract-YCW and brewery-YCW than with the negative control. In wheat diets, yeast extract-YCW and brewery-YCW reduced intestinal viscosity.

5. A third experiment was conducted to study the effect of yeast extract-YCW on animal performance, intestinal mucosa morphology and intestinal viscosity. A 2 × 2 factorial arrangement of treatments was used; one factor was the dietary yeast extract-YCW supplementation (0 or 500 mg/kg feed) and the other the cereal in the diet (maize or wheat).

6. At 43 d, the heaviest BW was in chickens fed on yeast extract-YCW compared to those given the negative control. At 22 d, yeast extract-YCW increased villus height, mucus thickness and number of goblet cells with respect to negative control.

7. Results of these experiments suggest that supplementation of yeast extract-YCW to broiler chicken diets increased animal performance by favouring intestinal mucosal development.  相似文献

1. An attempt has been made to modify the composition of the gut flora of broiler chickens by dietary means in order to determine any concurrent changes in meat flavour.

2. Birds given a diet containing an increasing proportion of whole wheat supplemented with fresh green vegetable matter (special diet) were compared with conventionally‐fed controls. At 12 weeks of age those receiving the special diet had 10 to 100‐fold higher counts of Escherichia colt and faecal streptococci in the duodenum and lower small intestine and showed differences in the intestinal incidence of species and biotypes of lactobacilli.

3. Evaluation of cooked breast‐meat samples by a trained sensory panel showed a significant difference in flavour (P < 0.01) between conventional and special‐diet birds which varied from perceptually very slight to slight. The main terms used in distinguishing special‐diet birds from conventional controls included ‘richer’, ‘meatier’ and ‘sweeter’.

4. Storage of plucked, uneviscerated birds at 4 °C for 8 d maintained the flavour difference between the dietary groups but the effect of storage on flavour was greater in both groups of birds than the influence of diet alone. The likely influence on flavour of autolytic changes occurring in the muscle during storage of the birds is discussed.  相似文献

1.?This research was conducted to determine the impact of diet supplementation with yeast cell walls and Yucca schidigera extract on the growth performance, antibody titres, and intestinal tissue histology of layer chicks.

4.?Overall, feed consumption did not differ between the control and YCW, YE, YCW?+?YE groups during the 60?d study period. Feed efficiency was better in the YE and YCW?+?YE groups than in the control group between d 1-60.

5.?During the 60?d evaluation period, live weight gain, and final live weight were higher in YE and YCW?+?YE groups than in the control group.

6.?Antibody titres against infectious bronchitis and infectious bursal disease did not differ among the 4 treatments, but those for Newcastle disease were higher in the YE?+?YCW groups than in the control, YCW and YE groups on d 45.

7.?There were differences in intestinal histomorphometry between the 4 treatments. The height of the jejunal and ileal villi was greater in the YE and YCW?+?YE groups than in the control and YCW groups

1. The effects of an indigestible soluble polysaccharide (carboxy methyl cellulose: CMC) on broiler performance (body weight gain, food and water intake) and on chyme characteristics (moisture content, viscosity, pH, osmolality and retention time) in broilers were studied.

2. In semi‐synthetic diets 0, 5.0 and 10.0 g/kg of cellulose was replaced by CMC on weight basis. These diets were fed to male broilers from 3 to 5 weeks of age.

3. When 10.0 g/kg CMC was included in the diet, food intake and body weight gain were reduced, compared to the 0, and 5.0 g/kg CMC diets. Food:gain ratio and water intake were increased at each CMC concentration. The significant quadratic response showed an increased response per 5.0 g/kg dietary CMC at the higher CMC concentration.

4. The viscosity in the supernatant of the chyme was linearly increased in all intestinal segments by CMC. A quadratic increase was observed in the crop. In the lower ileum, differences between the 5.0 and 10.0 g/kg CMC diets were not significant.

5. CMC increased the mean retention time of chromium in the duodenum and in the upper jejunum, and reduced the maximal rate of marker excretion. The transit time (first appearance of the marker in the excreta), however, was significantly reduced at the highest CMC concentration.

6. The osmolality of the intestinal fluid decreased less as the chyme moved from the duodenum into the lower ileum, with increasing concentrations of CMC.

7. The ileal pH was reduced linearly by the CMC content of the diet.

8. Based on a higher moisture content of the chyme in the CMC‐fed birds, and the higher jejunal and ileal osmolarities in those birds, it was concluded that the efficiency of both digestion and absorption was reduced by CMC inclusion in broiler diets.  相似文献

1. The effects of Aspergillus oryzae- and Bacillus subtilis-based direct-fed microbials (DFM) were investigated on the performance, ileal nutrient transport and intestinal integrity of broiler chickens, raised under experimental conditions, with increased intestinal microbial challenge.

2. The first study was a 3 × 2 factorial experiment, with 3 dietary treatments (control (CON), CON + DFM and CON + antibiotic growth promoter) with and without challenge. Chicks were fed experimental diets from 1 to 28 d, while the challenge was provided by vaccinating with 10 times the normal dose of commercial coccidial vaccine on d 9. In a second experiment, two groups of 1 d-old broilers, housed on built-up litter (uncleaned from two previous flocks), were fed the same CON and CON + DFM diets from 1 to 21 d.

3. The challenge in the first experiment reduced performance, but no differences were observed among dietary treatments from 8 to 28 d. The challenge reduced the ileal epithelial flux for D-glucose, L-lysine, DL-methionine and phosphorus on d 21. Epithelial flux for D-glucose, L-lysine and DL-methionine were increased by DFM. Ileal trans-epithelial electrical resistance (TER) was increased in challenged broilers fed DFM, although this was not observed in unchallenged birds as indicated by a significant interaction.

4. Ileal mucin mRNA expression and colon TER were increased, and colon endotoxin permeability was reduced by DFM on d 21 in the second experiment.

5. It was concluded that the addition of DFM in the diet improved the intestinal integrity of broiler chickens raised under experimental conditions designed to provide increased intestinal microbial challenge.  相似文献

1. The efficacy of supplementation of a low-phosphorus (low-P) maize–soyabean meal diet for broiler chickens with two different microbial (fungal and bacterial) phytases was examined.

2. Broiler chickens received a low-P maize–soyabean meal diet containing either no phytase or one of the two phytases included at one and two times the manufacturer's recommended inclusion level for 21 d.

3. Titanium dioxide was included in the diets as an indigestible marker. Excreta were collected quantitatively from d 18 to 21, and at the end of the study the birds were killed and ileal digesta and leg bone samples collected.

4. No differences were observed for body weight gain and feed intake or apparent metabolisable energy (AME) among all dietary treatment groups.

5. Dietary phytase supplementation improved the apparent retention of Ca, Na and Cu and ileal phytate P absorption from 32% to 44% across inclusion levels.

6. Bone mineral density (BMD) was improved for both phytases across inclusion levels by, on average, 9% for the tibia and 13% for the femur.

7. Dietary phytase supplementation of the low-P diet improved apparent ileal digestibility of serine, glycine, valine, isoleucine, tyrosine, histidine, lysine and arginine.

8. When the results from the present study were combined with the results from other similar published studies and analysed statistically, factors such as dietary P and Ca concentration, as well as bird breed and age, rather than the type or activity of microbial phytase, had the greatest impact on the extent to which dietary supplementation improved P and Ca retention.

9. More work is required to explain the interrelationships between the multiple factors influencing the efficacy of phytase on the availability of dietary minerals.  相似文献

1. The behaviour and reactivities of visceral receptors to different mechanical and chemical stimuli in single unit preparations from the caudal mesenteric nerve of adult ducks were studied to examine the physiological roles of such receptors.

2. In total, 83 single unit activities were recorded from the caudal mesenteric nerve.

3. Receptors were grouped according to their locations in different layers of the rectum as (i) over the branching points of the blood vessel or in the serosal layer, (ii) in the muscle layer or (iii) in the mucous membrane of the rectum.

4. All the receptors, both spontaneous and non‐spontaneous, responded to mechanical probing. Receptors of the muscle layer responded to distension also and showed a slowly‐adapting nature. Most of the receptors of the other two groups were of the rapidly‐adapting type.

5. Conduction velocity ranged from 0.76 to 34.6 m/s.

6. Sympathetic afferent fibres from the hind gut ascend along the intestinal nerve and join the caudal mesenteric nerve via the colonic ganglion.  相似文献

1. An experiment was conducted to study the effect of two different diets on zootechnical performance and the major bacterial groups in association with the host mucosa and dispersed in the lumen contents of the small intestine of broiler chickens.

2. The two experimental diets were maize or sorghum-based. In addition to the total bacteria, bacterial groups belonging to the Enterobacteriaceae (Enterococcus and Lactobacillus) were quantified by real-time PCR.

3. There were no differences in body weight gain and feed intake, but feed conversion ratio increased for sorghum-fed broilers at 21 and 42 d of age.

4. The Enterococcus group decreased in all gut segments from 7 to 42 d, while the Lactobacillus group increased in both ecosystems. In the ileal mucosa, the enterobacterial counts decreased from 7 to 42 d in the maize-based diet, but remained stable in the sorghum-based diet.
