The lower results in cryopreservation of in vitro‐produced (IVP) sheep embryos, when compared to the in vivo derived, limits its use. Four groups of blastocyst (BL) were evaluated: fresh IVP (n = 3), fresh in vivo derived (n = 3), warmed IVP cryopreserved in open pulled straws (OPS, n = 3) and warmed in vivo derived cryopreserved in OPS (n = 3). Ultrastructural observation of processed fresh embryos showed a reduced number of microvilli and mitochondria in the IVP ones, as well as a lower number of mature mitochondria, that can be associated with deficient metabolism in IVP embryos, possibly involved in the lower resistance to cryopreservation. Both in vivo‐derived and IVP embryos had a large number of vesicles, with light and dense content. In embryos vitrified by OPS, major changes were observed mainly in IVP embryos with small changes in grade 2 (fair) and high changes in grade 3 (bad) semithin scoring. The main changes associated with cryopreservation included disruption of cellular membranes and poor intracellular preservation, with loss of microvilli and the presence of cellular debris. In conclusion, ultrastructural evaluation of IVP blastocysts cryopreserved in OPS was herein described for the first time, reporting more severe cellular damage in these embryos when compared to those produced in vivo. This is probably associated with a lower cryotolerance that can be related to their lipid content and metabolism.  相似文献

The IGF system is related to embryo quality. We aim to determine the effect of the heat stress on the mRNA expression of IGF1 and IGF2, IGFR1 and IGFR2, IGFBP2 and IGFBP4, and PAPPA in in vitro production (IVP) blastocysts from Nelore and Holstein after ovum pick up (OPU) to better understand the differences between these breeds. Oocytes from four Nelore and seven Holstein were collected in six OPU sessions. Following in vitro maturation and fertilization using six Nelore or Holstein sires, embryos were divided into control (cultured at 39°C) and heat stress (HS; exposed to 41°C for 9 h). Blastocysts were submitted to RNA extraction. The IGF1 expression was higher in blastocysts under HS in both breeds, and the expression of IGFBP2 and IGFBP4 was higher in Holstein blastocysts under HS. The high PAPPA expression and the low expression of IGFBP2 and IGFBP4 are associated with a more efficient degradation of IGFBPs, which results in greater IGF bioavailability in Nelore blastocysts and may contribute to the superior HS tolerance in Nelore, when compared to Holstein.  相似文献

The present study evaluated the effects of genetic backgrounds on the developmental competence and thermotolerance of bovine in vitro‐produced (IVP) embryos. First, Holstein (Hol) and Japanese Black (JB) oocytes were fertilized with sperm from Hol, JB and a thermotolerant breed (Brahman), and in vitro development was evaluated when the embryos were exposed to heat shock on Day 2 (Day 0 = day of fertilization). Sperm genetic backgrounds affected the developmental competence in controls (P < 0.05). Second, the effect of sperm pre‐incubation for 4 h on subsequent in vitro fertilization was assessed using different sperm genetic backgrounds. The pre‐incubation of sperm did not decrease the embryonic development regardless of the breed of the sperm. A milder heat shock (40.0°C) effect on parthenotes (Hol and JB) and IVP embryos were evaluated. JB parthenotes showed developmental arrest after Day 4, and the rate of development to the blastocyst stage decreased by heat shock, but not in Hol parthenotes. Heat shock decreased developmental competence after cleavage of IVP embryos regardless of genetic background. The thermotolerance of IVP embryos would be controlled by both maternal and paternal factors but genetic involvement was still unclear. Further evaluation is needed to reveal the genetic contribution to thermotolerance.  相似文献

The aim of this study was to determine whether vitrification is an effective method when used for Japanese Black Cattle (Wagyu) in vivo‐derived embryos, collected following a superovulation treatment and embryo transfer (MOET) programme. In vivo‐derived morula and blastocysts collected on day 7 after artificial insemination, were vitrified using a modified droplet vitrification (MDV) procedure and subsequently warmed for transfer (ET) into synchronized recipients. Fresh embryos, and embryos cryopreserved using a standardized slow freezing procedure (direct thaw/direct transfer, DT) served as ET controls. Two different follicle‐stimulating hormone (FSH) sources, Folltropin® Canada (FSH BAH, 24 donors) and a brand prepared by the Chinese Academy of Science (FSH CAS, 16 donors), were compared in a series of superovulation outcomes following well‐established FSH administration protocols. Following data analysis, the total number of ovulations recorded at the time of embryo flushing (10.5 vs 8.5; p = 0.28) and the total number of transferable embryos (6.2 vs 5.1; p = 0.52) were similar between the two FSH sources. ET for MDV (39.7%, n = 78), DT (35.2%, n = 71) and fresh controls (47.1%, n = 34) resulted in similar pregnancy rates (p > 0.05). When MDV was used, a higher pregnancy rate (42.6%) resulted from the transfer of vitrified morulae, when compared to the DT counterparts (24.3%), (p = 0.05). Transfer of vitrified morulae resulted also in higher pregnancy rate, when compared to the transfer of vitrified blastocysts (42.6% vs. 29.4%; p < 0.05). Transfer of DT blastocysts resulted in higher pregnancy rate than morulae, similarly cryopreserved (47.1% vs. 24.3%, p < 0.05). In conclusion, MDV is an effective alternative methodology for cryopreservation of in vivo‐derived embryos. This study gives also indication that, compared to vitrified blastocysts, MDV of morula stage embryos results in higher pregnancy rates following warming and transfer into synchronized recipients.  相似文献

Interest in indicus–taurus cattle has been increasing, as these animals are likely to present the best characteristics of Zebu and European bovine breeds. The aim of this study was to compare the embryo production of indicus–taurus donors with high vs low antral follicle counts obtained by ovum pickup/in vitro production (OPU/IVP) and superovulation (SOV)/embryo collection. Braford females at weaning age (3/8 Nelore × 5/8 Hereford, n = 137, 9 ± 1 month old) were subjected to six serial ovarian ultrasonographs and were assigned to two groups according to the number of antral follicles ≥3 mm as follows: G‐High antral follicular count (AFC, n = 20, mean ≥40 follicles) and G‐Low AFC (n = 20, mean ≤10 follicles). When the females (n = 40) reached 24 months of age, they were subjected to both OPU/IVP and SOV/embryo collection. The average number of follicles remained highly stable throughout all of the ultrasound evaluations (range 0.90–0.92). The mean number of COCs recovered (36.90 ± 13.68 vs 5.80 ± 3.40) was higher (p < 0.05) for females with high AFC, resulting in higher (p < 0.05) numbers of total embryos among females with high vs low AFC (6.10 ± 4.51 vs 0.55 ± 0.83). The mean number of embryos per collection was also higher (p < 0.05) for G‐High vs G‐Low (6.95 ± 5.34 vs 1.9 ± 2.13). We conclude that a single ultrasound performed at pre‐pubertal ages to count antral follicles can be used as a predictor of embryo production following IVP and SOV/embryo collection in indicus–taurus females.  相似文献

The present study was conducted to evaluate the possibility of using in vitro‐produced parthenogenetic (PA) embryos for co‐transfer with morulae that had been collected in vivo and cryopreserved. The proportion of PA blastocysts (20.5%) was higher than that of their in vitro fertilization (IVF) counterparts (16.6%). Although there were no differences in morphology or diameter between the two groups, the number of cells in early PA blastocysts after in vitro culture for 6 days was lower than for IVF blastocysts (25.7 and 30.4 cells, respectively), and the number in recovered PA blastocysts was also smaller than that in recovered IVF blastocysts (37.4 and 50.2 cells, respectively). When 10 morulae warmed after vitrification were co‐transferred with 10 PA blastocysts (total 20 embryos) to the uterus of five recipients, the rates of pregnancy and farrowing did not differ, but the average period until spontaneous abortion tended to be longer relative to the control (when 20 morulae were transferred). These data suggest that in vitro‐produced PA embryos offer the possibility of assisted pregnancy for cryopreserved embryos; further experiments will be needed to confirm the beneficial effect of this approach on piglet production.  相似文献

Feedlot traits, carcass traits and distribution of commercial cuts of crossbred intact male progeny (n = 556) from young and mature Hereford, Red Poll, Hereford X Red Poll, Red Poll X Hereford, Angus X Hereford, Angus X Charolais, Brahman X Hereford and Brahman X Angus dams were evaluated. First-calf heifers were bred to Red Angus bulls; Santa Gertrudis sires were used for each cow's second and third breeding seasons. Calves from these young dams were slaughtered at 13 mo. Calves of mature dams were all sired by Limousin bulls and slaughtered at 12 mo. Dam breed was a major source of variation in most bull traits. Progeny of Brahman-cross dams were inferior (P less than .01) in daily gain, final weight, carcass weight and in edible cuts/day of age compared with progeny from Bos taurus dams. Intact male progeny of Angus X Charolais dams ranked highest in longissimus area, cutability, and edible cuts/day of age. The range of dam breed means in percentage of steak, roast, bone-in cuts (chuck short ribs and back ribs), short plate and thin cuts, and lean trim was just over 1%. Greater variation among dam breeds existed in fat measurements. Analyses in which Hereford-Red Poll diallel data for young dams and mature dams were combined showed positive maternal heterosis for dressing percentage (P less than .05), carcass weight (P less than .05), carcass weight/day of age (P less than .05), estimated carcass fat (P less than .05), fat thickness (P less than .01) and marbling score (P less than .01). Reciprocal effects were inconsequential. Results illustrate the importance of dam breed-type effects in formulating breeding strategies for commercial beef herds.  相似文献

Pregnancy‐associated glycoproteins (PAGs) are produced by mono‐ and binucleate trophoblast cells in the ruminant placenta. PAG appears in maternal blood and, from approximately 4 weeks after fertilization onward, may serve as a reliable means of diagnosing pregnancy. A range of factors are said to affect plasma PAG concentrations, such as number and sex of foetus, mass of calf and placenta, level of milk production and genetic constitution. In this study, PAG pregnancy profiles of a dual‐purpose (Simmental) and two beef breeds (Uckermark and Aubrac) are compared with the profile of the specialized dairy breed Holstein–Friesian. Holstein–Friesian cows were sampled weekly; the levels of the other breeds were presented at 3‐week intervals. The overall significant breed difference (p = 0.013) was founded on deviations during the initial 3 weeks of pregnancy and from 23 weeks onward. During the period critical for the detection of pregnancy, between four and 22 weeks, agreement between PAG levels of various breeds was close (p > 0.05). No significant effect of body mass of cow or calf (relative to mass of dam) was detected. These findings imply that the PAG pregnancy test may be executed uniformly irrespective of breed or type of cow, affirming the suitability of the test as a valuable asset for the cattle industry.  相似文献

Summary Data on first lactation performance of 751 Holstein×zebu and Jersey×zebu crossbred heifers of various grades were collected under rural field conditions in Maharashtra. The heifers were the progeny of seven Danish Holstein (Sortbroget Dansk Malkekvæg) sires, 33 homebred Holstein sires, six Danish Jersey sires and 35 homebred Jersey sires. The least-squares analysis showed that the location and genetic group within the two classes of crossbreds significantly affected the milk yield. In pooled data the 7/8 Holstein crossbreds produced by homebred sires were significantly superior to other genetic groups of either Holstein or Jersey inheritance. The milk yield increased with increasing level of homebred Holstein inheritance and with Jersey inheritance. Danish Holstein sires were the best to produce the halfbred Holstein crosses. The differences in the genetic groups accounted for 20% of the phenotypic variance in 305-day milk yield and for 2% of the variance of the lactation length in the crossbred population. It is inferred that the optimum level of exotic breed inheritance in Maharashtra would be above 50% and that the genetic group of the dams needs to be considered when evaluating the performance of a sire's progeny.

Resumen Se colectaron datos sobre la primera lactancia de 751 novillonas Holstein×Cebu y Jersey×Cebu bajo condiciones de campo en Maharashtra. Las novillonas pertenecian a la progenie de siete Holstein Daneses (sortbroget Danks Malkerase) sires, 33 sires Holstein locales, seis sires Jersey Daneses y 35 sires Jersey locales. El análisis de cuadrados mínimos mostró, que la localización y grupo genético dentro de dos clases de cruzamientos, afectó significativamente la producción de leche. Los datos globales mostraron, que las Holstein 7/8 provenientes de sires locales, fueron significativamente superiores a otros grupos genéticos, tanto Holstein como Jersey. La producción de leche se incrementó paralelamente a la herencia Holstein de sires locales, asi como de sires Jersey locales. Los sires Holstein Daneses fueron los mejores para producir los cruces Holstein. Las diferencias entre los grupos genéticos influyeron en un 20% de la variación fenotípica en 305 días de lactancia y en 2% de la variación del período de lactación en la población cruzada. Se infiere, que el nivel óptimo de herencia de razas exóticas en Maharashtra sería superior al 50% y que el grupo genético de las madres reproductoras tiene que ser evaluado conjuntamente con el de los machos.

Performance Des Croisements de Taurins (Bos Taurus) Et de Zebu En Elevage Villageois
Résumé Les données concernant les performances de première lactation de 751 génisses croisées Holstein×Zebu et Jersiais×Zebu, de différents degrés ont été recueillies sur le terrain dans des conditions villageoises dans le Maharashtra. Les génisses étaient les descendantes de sept taureaux Holstein danois (sortbroget Dansk Malkerace), de 33 taureaux Holstein indigènes, de 6 taureaux Jersiais danois et de 35 taureaux Jersiais indigènes. L'analyse des moindres carrés a montré que le lieu et le groupe génétique à l'intérieur des deux classes de métis avaient une influence significative sur le rendement laitier. Dans les fichiers de données, les métisses 7/8 Holstein engendrées par des taureaux indigènes étaient très supérieures aux autres groupes génétiques d'ascendance Holstein ou Jersiaise. La hausse de la production de lait était parallèle à la hausse de l'héritage de sang Holstein indigène et à celle de l'héritage de sang Jersiais. Les taureaux Holstein danois étaient les meilleurs pour la production de demi-sangs Holstein. Les différences dans les groupes génétiques étaient responsables de 20 p. 100 de la variance phenotypique de la production laitière de 305 jours et de 2 p. 100 de la variance de la durée de lactation dans la population métisse. Il en découle que le niveau optimal de sang importé à Maharashtra se situerait au dessus de 50 p. 100 et que le groupe génétique des mètres doit être pris en considération quand on évaluée les performances de la descendance d'un taureau.

In Bos taurus cattle, antimullerian hormone (AMH) has been demonstrated to have a high degree of correlation with ovarian antral follicle count and the number of healthy follicles and oocytes. To document the correlation between the plasma concentration of AMH and follicular number in Bos indicus and Bos taurus heifers, Nelore (Bos indicus, n = 16) and Holstein heifers (Bos taurus, n = 16) had their ovarian follicular waves synchronized. After synchronization, ovarian antral follicular population (AFP) was evaluated three times at 60‐day (d) intervals (T‐120 d, 120 days before plasma AMH determination; T‐60 d, 60 days before; and T0, at the time of plasma AMH determination). The plasma AMH concentration was positively correlated with the number of ovarian follicles on the day of the follicular wave emergence in Bos indicus (Nelore) and Bos taurus (Holstein) heifers at each evaluation time (p < 0.05). The AFP was higher in Bos indicus (Nelore) than in Bos taurus (Holstein) heifers (p < 0.05). Similarly, the AMH concentration was higher in Bos indicus (Nelore) than in Bos taurus (Holstein) heifers (p < 0.0001). When heifers were classified as to present high or low AFP according to the mean of the AFP within each genetic group, high‐AFP heifers presented a greater (p < 0.0001) AMH concentration than low‐AFP heifers, regardless of the genetic group. In conclusion, the AFP is positively correlated with plasma AMH concentration in both Bos indicus (Nelore) and Bos taurus (Holstein) heifers. Furthermore, Bos indicus (Nelore) heifers presented both greater plasma AMH concentrations and AFP than Bos taurus (Holstein) heifers.  相似文献

The objective of the research was to evaluate the efficiency of the spermatogenesis through the morphology of the testicular parenchyma in bulls of different zebu breeds. We used testicular fragments from bull of the breeds Nelore (n = 10), Polled Nelore (n = 6), Gyr (n = 5), Guzerat (n = 5) and Tabapuã (n = 5). The tissue was perfused with Karnovsky solution, included in glycol methacrylate and stained with toluidine blue–sodium borate 1%. Animals of the Nelore breed presented higher population of primary spermatocyte in pre‐leptotene/leptotene (38.30) and in pachytene (38.14) and round spermatids (113.30), higher yield of spermatogonia mitosis (21.2) and higher daily spermatic production per gram of testicular parenchyma (32.8 × 106) than those from breeds Gyr, Guzerat and Tabapuã and higher general yield of spermatogenesis (62.4) than breeds Gyr and Tabapuã. There was no significant difference in any of the evaluated parameters between breeds Nelore and Polled Nelore. The rate of Sertoli cells did not vary between the studied breeds. Apparently, the genetic selection applied to the breeds has been improving the yield in the spermatogenic process by decreasing cellular loss, although it did not increase the support capacity of the Sertoli cells.  相似文献

The present assay attempts to evaluate the feasibility of using embryo transfer in small community farmers by in vivo study and by modelling the results obtained. From the total of 59 donor cows, 62.7% responded to treatment, with a significant difference (p = 0.002) in the percentage of the response between breeds, being 90.5% (19/21) in Holstein and 47.4% (18/38) in Brahman. A total of 283 embryos were graded as transferable, while 141 as non‐transferable, without difference in the percentage of transferable embryo by breed (p = 0.18). The mean of transferable embryos graded as class I and II was not different between Holstein and Brahman (p = 0.96 and p = 0.92, respectively); besides, no differences were observed in the other grades (non‐transferable). The highest difference in costs, regardless of its quality by breed, was seen in the lower levels of probable fertility of the embryo transferred, even reaching several hundred dollars. When modelling the expected costs for embryo produced and transferred, values can reach nearly $2000.00 when the probable fertility is only 10%. However, when the probable fertility was 60%, embryo cost was close to $300.00. This technology seems to be viable on average or high‐scale systems, having a superovulatory response between 60 and 80% with 4–6 transferrable embryos. Yet, in small‐scale farming, due to the reduced number of donors and/or recipients, the costs surpass the economical feasibility of the technique.  相似文献

Dairy cattle management lacks consideration of fetal breed, the effect of which on fetal growth and nutrition are unclear. We investigated blood parameters in 12 late-pregnant Holstein heifers with similar (Holstein, n = 5) or different (Japanese Black [n = 4] or F1 cross [n = 3]; Holstein × Japanese Black) fetus breeds and in their umbilical cords and calves. Samples were obtained from dams 1 week before calving (−1 week) and immediately after calving, from the umbilical vein at calving, and from calves immediately after birth. Dams with beef fetuses had higher serum glucose levels (−1 week; p < .05) than those with Holstein fetuses. Plasma total amino acid, total essential amino acid, total nonessential amino acid, and other amino acid concentrations were lower in the umbilical veins of dams with calves of the beef breeds than in those of the Holstein breeds (p < .05). Furthermore, serum glucose and plasma amino acid levels were lower in the beef calves than in the Holstein calves (p < .05). Overall, nutrient supply from dams to beef fetuses was lower than that to Holstein fetuses. Our findings may facilitate feeding management of dairy cattle pregnant with beef breeds for appropriate fetal growth and nutrition.  相似文献

This study investigated the feasibility of applying fixed‐time (cryopreserved) embryo transfer in ewes. Embryos (n = 106) were non‐surgically recovered from superovulated donors (n = 39) on day 6–7 after oestrus. Straws containing one or two embryos (morulae and/or blastocysts) subjected to either slow freezing (SF, n = 62) or vitrification (VT, n = 44) were randomly used within fixed‐time embryo transfer on Day 8.5. Recipient ewes were nulliparous (n = 58) bearing corpora lutea after synchronous oestrous induction protocol. The pregnancy rate was higher (p = .03) in SF (39.4%) than VT (16.9%) and survival rate tended (p = .08) to be higher in SF than in VT (25.8% vs. 15.9%). Lambing rates were similar (p = .13) between SF (20.9%) and VT (15.9%). Embryos recovered by non‐surgical route after cervical dilation treatment and later cryopreserved by either slow freezing or vitrification produced reasonable pregnancy rates after FTET.  相似文献

This study was performed to evaluate plasma concentrations of anti‐Mullerian hormone (AMH) and the ovarian antral follicle population (AFP) in different genetic groups. Cyclic heifers (13 Bubalus bubalis [Murrah]; 15 Bos taurus [Holstein] and 10 Bos indicus [Gyr]) were maintained under the same management and were synchronized with two doses of 150 μg IM d‐cloprostenol administered 14 days apart. After the second d‐cloprostenol treatment, heifers had their ovaries scanned daily by ultrasound to define the day of ovulation. On the same day, the AFP was determined and a plasma sample was collected to measure AMH. Murrah heifers had less AFP (25.6 ± 2.1 follicles; p = 0.01) and plasma AMH concentration (0.18 ± 0.03 ng/ml; p < 0.001) than Gyr (60.0 ± 12.2 follicles and 0.60 ± 0.12 ng/ml of AMH); however, data were similar when compared to Holstein (35.9 ± 6.8 follicles and 0.24 ± 0.06 ng/ml of AMH) heifers. Regardless of genetic background, there was a positive relationship between the AFP and plasmatic AMH concentration (Murrah [r = 0.62; p < 0.01], Holstein [r = 0.66; p < 0.001] and Gyr [r = 0.88; p < 0.001]). Also, when heifers were classified according to high‐ or low‐AMH concentration based on the average within each genetic group, high‐AMH heifers had greater (p < 0.0001) AFP than low‐AMH heifers. In conclusion, both Murrah and Holstein heifers presented lower plasma AMH concentration and AFP when compared to Gyr.  相似文献

Angus, Boran and Red Poll sires were mated to Ankole, Boran and small East African Zebu (Zebu) females to produce the cows characterized in this study, which produced calves by Friesian, Brown Swiss, Simmental, Boran and Red Poll sires. Individual traits included cow parturition weight, cow weight at weaning, cow mean weight and cow weight change from parturition to weaning; maternal traits included calf crop born percentage, preweaning viability percentage, overall viability percentage, birth weight, weaning weight, weight at 12, 18 and 24 mo and calf weight weaned per cow exposed to breeding (productivity index). Cows by Angus and Red Poll sires significantly exceeded cows by Boran sires (all cows in this comparison had Ankole and Zebu dams) in weaning weight of progeny and in cow weight at parturition and at weaning. Cows by Red Poll sires significantly exceeded cows by Boran sires in birth weight of progeny. Progeny of cows by Boran sires significantly exceeded progeny of cows by Angus and Red Poll sires in 24-mo weight. Cows by Angus and Red Poll sires did not differ (P greater than .05) in any of the traits analyzed. Cows with Ankole and Boran dams significantly exceeded cows with Zebu dams (all cows in this comparison had Angus and Red Poll sires) in progeny weight at birth, 18 mo and 24 mo and in cow weight at parturition and at weaning. Cows with Boran dams significantly exceeded cows with Zebu dams in weaning and 12-mo weight of progeny and significantly exceeded cows with Ankole dams in weaning weight of progeny. The significantly heavier weaning weight and significantly lighter 24-mo weight of the 3/4 Bos taurus progeny of cows with Angus and Red Poll sires relative to the 1/2 Bos taurus progeny of cows with Boran sires suggest that cattle that are 1/2 Bos taurus breed composition have greater general adaptation than cattle with 3/4 Bos taurus breed composition in the postweaning nutritive and climatic environment under which this experiment was conducted. Significantly heavier weights of Angus and Red Poll crossbred cows relative to Boran crossbred cows (all breeds crossed on Ankole and Zebu cows) indicate that the two Bos taurus breeds exceed the Boran (Bos indicus) breed in additive direct genetic effects for size when they have general adaptation to the environment.  相似文献

This study was carried out to compare the post‐thaw cryosurvival rate and the level of apoptosis in vitro produced zona‐free cloned buffalo blastocysts subjected to slow freezing or vitrification in open‐pulled straws (OPS). Zona‐free cloned embryos produced by handmade cloning were divided into two groups and were cryopreserved either by slow freezing or by vitrification in OPS. Cryosurvival of blastocysts was determined by their re‐expansion rate following post‐thaw culture for 22–24 h. The post‐thaw re‐expansion rate was significantly (p < 0.05) higher following vitrification in OPS (71.2 ± 2.3%) compared with that after slow freezing (41.6 ± 4.8%). For examining embryo quality, the level of apoptosis in day 8 frozen‐thawed blastocysts was determined by TUNEL staining. The total cell number was not significantly different among the control non‐cryopreserved cloned embryos (422.6 ± 67.8) and those cryopreserved by slow freezing (376.4 ± 29.3) or vitrification in OPS (422.8 ± 36.2). However, the apoptotic index, which was similar for embryos subjected to slow freezing (14.8 ± 2.0) or OPS vitrification (13.3 ± 1.8), was significantly (p < 0.05) higher than that for the control non‐cryopreserved cloned embryos (3.4 ± 0.6). In conclusion, the results of this study demonstrate that vitrification in OPS is better than slow freezing for the cryopreservation of zona‐free cloned buffalo blastocysts because it offers a much higher cryosurvival rate.  相似文献

Embryo transfer using cryopreserved B. indicus embryos results in low pregnancy rates. The low viability of B. indicus embryos is likely a result of their low freezability relative to embryos from other species, notably B. taurus. Freezability is probably related to the quantity of lipid droplets in the embryonic cells. However, the mechanism underlying this association is unknown. Using multiphoton and transmission electron microscopy, the proportion, volume, morphology and composition of lipid droplets in fresh and cryopreserved B. indicus and B. taurus embryos were studied. B. indicus embryos have more droplets than B. taurus and a greater percentage of lipid droplets in clusters (34% vs. 24%; < 0.05). Also, B. indicus individual droplets are of greater volume than those of B. taurus (47 μm3 vs. 34 μm3; < 0.05). After freezing, the percentage of clusters decreased in B. indicus (11% vs. 5%; < 0.05), while in B. taurus they increased (2% vs. 7%; < 0.05). Freezing influenced the lipid droplet morphology, increasing the number of cracked droplets in B. indicus embryos (33% fresh vs. 62% frozen; p < 0.05), but not in B. taurus embryos (35% fresh vs. 34% frozen; ns). Regarding composition, saturated lipid droplets prevail in B. indicus embryos, contrary to B. taurus embryos. We observed differences in lipid droplet organization, proportion, volume, morphology and composition between B. indicus and B. taurus embryos which are altered in opposite directions following freezing.  相似文献

Increasing use of fixed‐time artificial insemination (FTAI) in beef cattle production has presented an opportunity for the use of fresh or chilled semen as an alternative to standard cryopreserved semen. The objective of this study was to examine in vitro sperm function and pregnancy rate of electroejaculated semen, chilled and stored for 48 hr, compared to conventionally cryopreserved semen with an optimized FTAI protocol in Brahman cattle. Semen from three Brahman bulls was collected, and aliquots were extended in either chilled (at 5°C) or frozen (LN2) in a Tris‐egg yolk extender base with 2.4% or 7.0% glycerol, respectively. Semen samples were assessed 48 hr after collection or post‐thaw and warming, for sperm motility, in vitro sperm function and fertilizing ability, and used in a FTAI programme. The overall pregnancy rates was significantly different (< .01) after FTAI with frozen (n = 173; 53.2%) and chilled semen (n = 174; 31.6%). In contrast, the in vitro sperm assessment showed that the chilled semen had significantly faster motility (< .05), a higher proportion of progressively motile spermatozoa (< .05), with significantly higher proportions of acrosome intact, viable spermatozoa (< .01). This study showed that reasonable pregnancy rates in Brahman cattle can be achieved using FTAI with chilled semen collected using electroejaculation and stored for up to 48 hr. However, improvements in semen extenders are required in consideration of semen collection method to improve the longevity of sperm fertilizing ability to significantly increase FTAI output using chilled storage of bull semen.  相似文献

Records of birth weight (BW), weaning weight (WW) and condition score (CS) from 1,467 Brahman and Brahman X Angus crossbred calves from Brahman and crossbred Brahman sires and Brahman, crossbred Brahman and Angus dams were collected at the Subtropical Agricultural Research Station, Brooksville, Florida, from 1971 to 1982. Best linear unbiased estimates (BLUE) of Brahman sire and dam group additive genetic effects (as deviations from Angus) and Brahman X Angus dam and calf group nonadditive (intralocus) genetic effects (as deviations from intralocus group genetic effects in the parental breeds) were obtained. Linear combinations of these were used to compute direct and maternal Brahman additive and Brahman X Angus nonadditive (intralocus) group genetic effects. The respective BLUE of these four effects were 5.99 +/- 2.08, -5.70 +/- 1.91, .52 +/- 1.81 and 2.85 +/- .72 kg for BW; 9.60 +/- 10.29, 8.76 +/- 9.47, 9.47 +/- 8.96 and 20.95 +/- 3.56 kg for WW; and -1.10 +/- .55, 1.64 +/- .50, 1.47 +/- .47 and .05 +/- .19 units for CS. Linear combinations of the BLUE of sire, dam and calf group genetic effects can be used to predict the genetic worth of crossbred groups composed of any combination of Brahman and Angus breeding. Nonadditive maternal group genetic effects were the most important factor for BW and WW, whereas nonadditive direct group genetic effects were the most important for CS.  相似文献
