Enteric redmouth disease (ERM) is a serious septicemic bacterial disease of salmonid fish species. It is caused by Yersinia ruckeri, a Gram-negative rod-shaped enterobacterium. It has a wide host range, broad geographical distribution, and causes significant economic losses in the fish aquaculture industry. The disease gets its name from the subcutaneous hemorrhages, it can cause at the corners of the mouth and in gums and tongue. Other clinical signs include exophthalmia, darkening of the skin, splenomegaly and inflammation of the lower intestine with accumulation of thick yellow fluid. The bacterium enters the fish via the secondary gill lamellae and from there it spreads to the blood and internal organs. Y. ruckeri can be detected by conventional biochemical, serological and molecular methods. Its genome is 3.7 Mb with 3406–3530 coding sequences. Several important virulence factors of Y. ruckeri have been discovered, including haemolyin YhlA and metalloprotease Yrp1. Both non-specific and specific immune responses of fish during the course of Y. ruckeri infection have been well characterized. Several methods of vaccination have been developed for controlling both biotype 1 and biotype 2 Y. ruckeri strains in fish. This review summarizes the current state of knowledge regarding enteric redmouth disease and Y. ruckeri: diagnosis, genome, virulence factors, interaction with the host immune responses, and the development of vaccines against this pathogen.  相似文献

The systemic and respiratory local immune response induced by the intradermal administration of a commercial inactivated Mycoplasma hyopneumoniae whole-cell vaccine (Porcilis® MHYO ID ONCE – MSD AH) in comparison with two commercial vaccines administered via the intramuscular route and a negative control (adjuvant only) was investigated. Forty conventional M. hyopneumoniae-free pigs were randomly assigned to four groups (ten animals each): Group A = intradermal administration of the test vaccine by using the needle-less IDAL® vaccinator at a dose of 0.2 ml; Group B = intramuscular administration of a commercially available vaccine (vaccine B); Group C = intramuscular administration of the adjuvant only (2 ml of X-solve adjuvant); Group D = intramuscular administration of a commercially available vaccine (vaccine D). Pigs were vaccinated at 28 days of age. Blood and bronchoalveolar lavage (BAL) fluid samples were collected at vaccination (blood only), 4 and 8 weeks post-vaccination. Serum and BAL fluid were tested for the presence of antibodies by ELISA test. Peripheral blood monomorphonuclear cells (PBMC) were isolated to quantify the number of IFN-γ secreting cells by ELISpot. Moreover, cytokine gene expression from the BAL fluid was performed. Total antibodies against M. hyopneumoniae and specific IgG were detected in serum of intradermally and intramuscularly (vaccine B only) vaccinated pigs at 4 and 8 weeks post-vaccination. M. hyopneumoniae specific IgA were detected in BAL fluid from vaccinated animals (Groups A and B) but not from controls and animals vaccinated with the bacterin D (p < 0.05). Significantly higher gene expression of IL-10 was observed in the BAL fluid at week 8 post-vaccination in the intradermally vaccinated pigs (p < 0.05). The results support that the intradermal administration of an adjuvanted bacterin induces both systemic and mucosal immune responses. Moreover, the intramuscularly administered commercial vaccines each had a different ability to stimulate the immune response both systemically and locally.  相似文献

A study was designed to compare the efficacy of NexGard® and Bravecto™, 2 recently introduced oral ectoparasiticides containing isoxazolines, against fleas (Ctenocephalides felis) on dogs. Twenty-four healthy dogs, weighing 9.2 kg to 28.6 kg, were included in this parallel group design, randomized, and controlled efficacy study. On Day −1, the 24 dogs were allocated to 3 study groups: untreated control; Nexgard® treated and Bravecto™ treated. The treatments were administered on Days 0, 28 and 56 for Nexgard® (labelled for monthly administration), and once on Day 0 for Bravecto™ (labelled for a 12 week use). Flea infestations were performed weekly with 100 adult unfed C. felis on each dog from Days 42 to 84. Fleas were counted and re-applied at 6 and 12 h post-infestation and removed and counted 24 h post-infestation. The arithmetic mean flea count for the untreated group ranged from 62.9 to 77.6 at 24 h post-infestation, indicating vigorous flea challenges on all assessment days. Both the Nexgard® and Bravecto™ treated groups had statistically significantly (p < 0.05) less fleas compared to the untreated group on all assessment time points and days. Significantly fewer fleas were recorded for NexGard® treated dogs compared to Bravecto™ treated dogs at 6 h post-infestation on Day 56, 63, 70, 77 and 84 and at 12 h post-infestation on Days 70 and 84. No statistically significant (p < 0.05) differences were recorded between the treated groups at 24 h post-infestation. Efficacies recorded 6 h post-infestation for Nexgard® ranged from 62.8% (Day 49) to 97.3% (Day 56), and efficacies ranged from 94.1% (Day 49) to 100% (Days 42, 56, 70 and 84) at 12 h post-infestation. Efficacies recorded for Bravecto™ ranged from 45.1% (Day 84) to 97.8% (Day 42) at 6 h post-infestation, and from 64.7% (Day 84) to 100% (Days 42 and 56) at 12 h post-infestation. Efficacies observed at 24 h were 100% for both products during the study except 99.6% on Day 84 for Bravecto™.  相似文献

Less than 20% of the domestic dogs on tribal lands in the United States are vaccinated against rabies. One method to increase vaccination rates may be the distribution of oral rabies vaccines (ORVs). ONRAB® (Artemis Technologies, Inc., Ontario, Canada) is the primary ORV used in Canada to vaccinate striped skunks and raccoons. To investigate the potential use of ONRAB® ORV baits to vaccinate feral domestic dogs against rabies on tribal lands and beyond, we performed a flavor preference study. A total of 7 bait flavors (bacon, cheese, dog food, hazelnut, sugar-vanilla, peanut butter, and sardine) were offered in pairs to 13 domestic dogs. Each dog was offered all possible combinations of bait pairs over a period of 10 days, with each bait offered 6 times. The proportion of times each bait was consumed first by individual dogs was calculated and comparisons among dogs were conducted using the MIXED procedure in SAS (SAS Institute, Cary, NC). Pairwise comparisons between baits were performed using “contrast” statements with sugar-vanilla flavor as the default for comparison. Type 3 tests of fixed effects showed a significant treatment effect (F6,72 = 9.74, P < 0.0001). Sugar-vanilla was selected first during 14% of the offerings and exhibited the least preference among all bait types (F1,72 = 22.46, P < 0.0001). Dog food was selected first 56% of the time, and more frequently than all other bait types (F1,72 = 13.09, P = 0.0005).  相似文献

A study was undertaken to compare the virulence and serum killing resistance properties of Atlantic salmon and rainbow trout Yersinia ruckeri isolates. Five isolates, covering heat-stable O-antigen O1, O2 and O5 serotypes, were tested for virulence towards fry and juveniles of both species by experimental bath challenge. The sensitivity of 15 diverse isolates to non-immune salmon and rainbow trout serum was also examined. All five isolates caused significant mortality in salmon fry. Serotype O1 isolate 06059 caused the highest mortality in salmon (74% and 70% in fry and juveniles, respectively). Isolate 06041, a typical ERM-causing serotype O1 UK rainbow trout strain, caused mortalities in both rainbow trout and salmon. None of the salmon isolates caused any mortalities in 150–250 g rainbow trout, and only serotype O2 isolate 06060 caused any significant mortality (10%) in rainbow trout fry. Disease progression and severity was affected by water temperature. Mortality in salmon caused by the isolates 06059 and 05094 was much higher at 16 °C (74% and 33%, respectively) than at 12 °C (30 and 4% respectively). Virulent rainbow trout isolates were generally resistant to sera from both species, whereas salmon isolates varied in their serum sensitivity. Convalescent serum from salmon and rainbow trout that had been infected by serotype O1 isolates mediated effective classical pathway complement killing of serotype O1 and O5 isolates that were resistant to normal sera. Overall, strains recovered from infected salmon possess a wider range of phenotypic properties (relative virulence, O serotype and possession of serum-resistance factors), compared to ERM-causing rainbow trout isolates.  相似文献

Vaccine delivery using microneedle (MN) patches is an easy, safe and painless alternative to traditional needle injections. In this study, we examined whether MN patches can enhance the efficacy of a Streptococcus suis serotype 2 (S. suis 2) vaccine in a mouse model. Results showed that MNs can reach 200–250 μm into the skin, a depth beneficial for targeted delivery of antigens to antigen-presenting cells in the epidermis and dermis. Vaccination with prime-boost of MN induced higher levels of IgG2a antibody titer, T cell proliferation, and TH1 cytokines (IFN-γ and IL-12) as compared to intramuscular (IM) injection. In addition, single dose MN vaccination better protected mice against lethal challenge than IM vaccination. MN vaccination also conferred long-term IgG2a antibody against S. suis 2 bacteria presence for up to 7 months. Taken together, these data showed that vaccine delivery by MNs results in superior immune response and protection rate when compared to IM injections.  相似文献

Ancylostoma ceylanicum is a common zoonotic hookworm of dogs and cats throughout Asia and has also been reported to occur within the Australasian region. The aim of this study to was to determine the efficacy of a spot-on formulation containing emodepside and praziquantel (Profender®, Bayer) and praziquantel and pyrantel oral tablets (Drontal® for Cats, Bayer) against experimental A. ceylanicum infections in cats.Twenty-four kittens were each subcutaneously injected with 100 infective third-stage larvae of A. ceylanicum. Kittens were stratified by egg count and randomly allocated equally into control and two treatment groups. The first group were treated with emodepside 2.1%/praziquantel 8.6% (Profender®, Bayer) at the recommended label dose. The second group was treated with 80 mg pyrantel and 20 mg praziquantel (Drontal® for Cats, Bayer) at the recommended label dose. The kittens in the control group were not treated. Egg counts were performed daily until the end of the study period and compared for the treated and control groups. No eggs were detected in the treated group of kittens within 4 days of treatment and faecal samples from this group remained negative throughout the rest of the study, resulting in a treatment efficacy (egg reduction) of 100% (P < 0.0001). The egg counts remained high (993 ± 666 epg) in the untreated control group for the rest of the study period. This study demonstrated that both combination products containing topical emodepside/praziquantel (Profender®, Bayer) and praziquantel/pyrantel oral tablets (Drontal® for Cats, Bayer) given at the recommended dose is highly effective against infection with A. ceylanicum in cats.  相似文献

The aim of the present study was to determine the serological response of heifers after vaccination with two inactivated bovine viral diarrhoea virus (BVDV) vaccines by means of various ELISA tests. Three dairy farms were selected from the Galicia region of Spain. In each herd, a batch of heifers to be vaccinated for the first time was selected and followed for 15 months. Heifers from farm 1 (n = 25) were vaccinated with Vaccine A, whereas heifers from farm 2 (n = 16) were vaccinated with Vaccine B. Heifers from farm 3 (n = 17), where no BVDV vaccines were used, acted as controls. Blood samples were analyzed periodically for BVDV antibodies, using five commercial ELISAs, based on BVDV p80 antigen or whole virus.At the end of the study, none of the animals vaccinated with Vaccine A seroconverted according to p80 antibody status, whereas up to 80% tested positive by ELISA against whole virus antigen. For the animals vaccinated with Vaccine B, 2/16 animals seroconverted according to p80 antibody ELISAs, whereas all had seroconverted according to the ELISA against whole virus antigen. In most cases, based on the use of ELISAs to detect specific antibodies against the p80 protein, at 15 months post-vaccination with inactivated BVDV vaccines the responses did not seem to interfere with detection of antibody to BVDV infection. However, the finding of a small proportion of vaccinated animals seropositive against BVDV p80 antigen suggests that antibodies that interfere with diagnosis of BVDV infection within the herd could exist, even when using p80 ELISAs.  相似文献

A randomized, blinded, negative controlled study was conducted to determine whether treatment with afoxolaner (NexGard®, Merial, Inc.) would prevent the transmission of Borrelia burgdorferi to dogs by wild caught Ixodes scapularis ticks. Twenty healthy dogs were randomly assigned to two groups of ten dogs each. Ten dogs were treated orally on Day 0 at a dose near the minimum recommended dose of afoxolaner of 2.5 mg/kg (actual doses 2.5–3.1 mg/kg) and ten control dogs were not treated. On Day 28, each dog was infested with approximately 50 adult unfed wild caught I. scapularis that had a 67% B. burgdorferi infection rate (determined by polymerase chain reaction). On Day 33, live ticks were counted and removed. No ticks were found on treated dogs while control dogs had an average of 21.4 ticks. To detect infection, the B. burgdorferi-specific C6 antibody SNAP® 4Dx® test (IDEXX) was performed on serum collected before infestation (all dogs seronegative on Days -6 and 27) and on Days 48, 63, 77 and 92. The ten treated dogs remained seronegative through the end of the study (Day 92), while nine out of the ten control dogs were infected, as demonstrated by their seroconversion to being positive for the presence of the B. burgdorferi-specific C6 antibody starting on Day 48. In this study, all dogs treated with NexGard® 28 days prior to challenge with wild caught I. scapularis ticks were protected from B. burgdorferi infection, while nine out of the ten untreated control dogs were infected.  相似文献

This study evaluated immune cell populations in pigs following weaning and vaccination for Mycoplasma hyopneumoniae. Piglets (n = 24) were weaned (day 0) at 16 (±1) days of age, and randomly assigned to the vaccination group (n = 16) or control group (n = 8). Complete blood cell counts, flow cytometry and serology were completed for blood samples collected on days 0 (within hours of weaning), 3, 7, 14, 30 and 60. The M. hyopneumoniae S:P ratios (sample optical density: positive control optical density) were negative in the vaccination group until days 30 and 60, when the S:P ratios were 1.3 and 1.0, respectively. Control animals remained serologically negative. The percentage of CD4+ T cells was less (P < 0.01) in control pigs than vaccinated pigs at day 3. In contrast, numbers of CD8+ and CD4+CD8+ T cells were greater (P < 0.01) in control pigs than in vaccinated pigs at days 3 and 7. After day 7, few differences in immune cell types were evident between the groups. Differences in lymphocyte populations could not be solely attributed to vaccination, due at least in part, to the confounding influence of weaning. It was difficult to distinguish the influence of vaccination from the impact of weaning on peripheral immune cell populations.  相似文献

To develop colloidal gold immunochromatographic strips for the direct detection of the Streptococcus suis serotype 2 antigen, colloidal gold was prepared by reduction of a gold salt with sodium citrate and coupled with polyclonal antibody against S. suis serotype 2. The optimal concentrations of the capture antibody and the coating antibody were determined to be 22 μg/mL and 2.0 mg/mL, respectively, and that of the blocking buffer was determined to be 1.5% bovine serum albumin. Different serotypes of S. suis and other related bacteria were used to evaluate the sensitivity, specificity, and stability of the immunochromatographic strips. The detection sensitivity was found to be as high as 106 CFU/mL. There was no cross-reaction of the antibodies with other serotypes of S. suis (except with SS1/2, which shares some common sugar residues or antigenic determinants with serotype 2) and other related bacteria. In conclusion, we developed colloidal gold immunochromatographic strips that had high sensitivity and specificity. This method proved to be feasible, convenient, rapid, and effective for detecting S. suis serotype 2.  相似文献

Currently there are no effective vaccines for the control of bovine neosporosis. During the last years several subunit vaccines based on immunodominant antigens and other proteins involved in adhesion, invasion and intracellular proliferation of Neospora caninum have been evaluated as targets for vaccine development in experimental mouse infection models. Among them, the rhoptry antigen NcROP2 and the immunodominant NcGRA7 protein have been assessed with varying results. Recent studies have shown that another rhoptry component, NcROP40, and NcNTPase, a putative dense granule antigen, exhibit higher expression levels in tachyzoites of virulent N. caninum isolates, suggesting that these could be potential vaccine candidates to limit the effects of infection. In the present work, the safety and efficacy of these recombinant antigens formulated in Quil-A adjuvant as monovalent vaccines or pair-wise combinations (rNcROP40 + rNcROP2 and rNcGRA7 + rNcNTPase) were evaluated in a pregnant mouse model of neosporosis. All the vaccine formulations elicited a specific immune response against their respective native proteins after immunization. Mice vaccinated with rNcROP40 and rNcROP2 alone or in combination produced the highest levels of IFN-γ and exhibited low parasite burdens and low IgG antibody levels after the challenge. In addition, most of the vaccine formulations were able to increase the median survival time in the offspring. However, pup survival only ensued in the groups vaccinated with rNcROP40 + rNcROP2 (16.2%) and rNcROP2 (6.3%). Interestingly, vertical transmission was not observed in those survivor pups immunized with rNcROP40 + rNcROP2, as shown by PCR analyses. These results show a partial protection against N. caninum infection after vaccination with rNcROP40 + rNcROP2, suggesting a synergistic effect of the two recombinant rhoptry antigens.  相似文献

Despite the importance of vegetative reproduction in annual tiller replacement, little is known about the patterns and timing of tiller recruitment from the bud bank, especially regarding fire return intervals and seasons of fire. We examined aboveground plant density, temporal patterns of tiller production, and belowground bud bank dynamics for Bouteloua gracilis (Willd ex. Kunth) Lag. ex Griffiths), Pascopyrum smithii (Rydb.) A. Löve, and Hesperostipa comata (Trin. & Rupr.) Barkworth following summer, fall, and spring prescribed fires at 2-yr, 3-yr, and 6-yr fire return intervals, and their interactions. Fire treatments were initiated in 2006, and buds were assessed July 2011 through July 2013. Density and number of reproductive B. gracilis tillers increased in 2013 following drought during 2012, unlike H. comata, which decreased reproductive tiller production. Irrespective of fire treatments, B. gracilis produced the most buds (8 ? 10 buds ? tiller? 1) and H. comata produced the least (2 ? 3 buds ? tiller? 1), with P. smithii producing an intermediate amount (6 ? 8 buds ? tiller? 1). Immediate B. gracilis and P. smithii bud mortality did not occur for all season and fire return interval treatments. However, H. comata bud mortality increased immediately following summer and fall prescribed fires. Three-yr fire return intervals increased active buds throughout the 2013 winter and growing season for B. gracilis and P. smithii relative to control plots and 2- and 6-yr fire return intervals. Fire stimulated bud activity of B. gracilis and P. smithii relative to nonburned plots. The aboveground and belowground response of H. comata indicated meristem limitations following fire treatments, illustrating greater vulnerability to fire for that species than B. gracilis and P. smithii.  相似文献

This study quantified herbaceous biomass responses to increases in honey mesquite (Prosopis glandulosa Torr.) cover on two soils from 1995 to 2001 in north central Texas. Vegetation was sampled randomly with levels of mesquite ranging from 0% to 100%. With no mesquite covering the silt loam soils of bottomland sites, peak herbaceous biomass averaged (±SE) 3 300 ± 210 kg · ha−1 vs. 2 560 ± 190 kg · ha−1 on clay loam soils of upland sites (P = 0.001). A linear decline of 14 ± 2.5 kg · ha−1 in herbaceous biomass occurred for each percent increase in mesquite cover (P = 0.001). The slope of this decline was similar between soils (P = 0.135). Herbaceous biomass with increasing mesquite cover varied between years (P = 0.001) as did the slope of decline (P = 0.001). Warm-season herbaceous biomass decreased linearly with increasing mesquite cover averaging a 73 ± 15% reduction at 100% mesquite cover (P = 0.001) compared to 0% mesquite cover. Cool-season herbaceous biomass was similar between soils with no mesquite, 1 070 ± 144 kg · ha−1 for silt loam vs. 930 ± 140 kg · ha−1 for clay loam soils, but averaged 340 ± 174 kg · ha−1 more on silt loam than on clay loam soils at 100% mesquite cover (P = 0.004). Multiple regression analysis indicated that each centimeter of precipitation received from the previous October through the current September produced herbaceous biomass of 51 kg · ha−1 on silt loam and 41 kg · ha−1 on clay loam soils. Herbaceous biomass decreased proportionally with increasing mesquite cover up to 29 kg · ha−1 at 100% mesquite cover for each centimeter of precipitation received from January through September. Increasing mesquite cover reduces livestock forage productivity and intensifies drought effects by increasing annual herbaceous biomass variability. From a forage production perspective there is little advantage to having mesquite present.  相似文献

Redberry juniper (Juniperus pinchotii Sudworth) is an invasive, evergreen tree that is rapidly expanding throughout western and central Texas. Goats will consume some juniper on rangelands; however, intake is limited. The objective of our research was to determine how the age and body condition of goats influence their consumption of juniper and an artificial feed containing 4 monoterpenes. Two separate experiments were conducted. Experiment 1 examined the intake of redberry juniper foliage and used 39 goats either young (2 yr) or mature (> 6 yr). One-half of each age group was fed appropriate basal rations to reach either a high (HBC) or low body condition (LBC). Goats in LBC ate more (P < 0.01, 8.6 g · kg−1 body weight [BW] ± 0.7 SE) juniper than those in HBC (2.3 g · kg−1 BW ± 0.3 SE), and young animals consumed more (P < 0.05, 7.2 g · kg−1 BW ± 0.7 SE) juniper than mature goats (3.9 g · kg−1 BW ± 0.5 SE) across body condition treatments. In experiment 2, 36 goats, either young (2 yr) or mature (> 6 yr) and in either HBC or LBC, were offered a synthetic ration treated with 20.8 g · kg−1 of 4 monoterpenes found in redberry juniper. Goats in LBC ate more (P < 0.01, 25.3 g · kg−1 BW ± 1.0 SE) of the terpene-treated feed than those in HBC (17.5 g · kg−1 BW ± 0.7 SE), and young animals ate more (P < 0.05, 22.5 g · kg−1 BW ± 0.8 SE) than mature goats (20.3 g · kg−1 BW ± 0.8 SE) across body condition treatments. Total intake as a proportion of body weight was also affected by body condition. Age and body condition are important factors that influence intake of chemically defended plants. A better understanding of how these attributes affect diet selection will aid livestock producers in improving grazing management.  相似文献

Objective

To compare airway management during induction of anaesthesia, spontaneous ventilation (SV) and controlled mechanical ventilation (CMV), using an endotracheal tube (ETT), laryngeal mask (LM), rabbit-specific supraglottic airway device (v-gel) or facemask (FM).

Study design

Prospective randomized crossover experiment.

Animals

Ten New Zealand White rabbits.

Methods

After premedication, rabbits were randomly allocated to four groups: 1) ETT; 2) LM; 3) v-gel or 4) FM. The required dose of propofol, duration and number of attempts to place an airway device and leakage during SV and CMV at different peak inspiratory pressures (6, 10, 12, 14 and 16 cmH2O) were recorded. Computed tomography (CT) of the head, neck and abdomen were performed before and after CMV.

Results

Significantly less propofol and time [2.0 ± 0.5 mg kg?1, 82 ± 34 seconds, p < 0.001] were needed to place the FM compared to the three other groups [v-gel 5.1 ± 2.1 mg kg?1, 302 ± 124 seconds; LM 4.8 ± 1.2 mg kg?1, 275 ± 89 seconds; ETT 5.5 ± 1.4 mg kg?1, 315 ± 147 seconds]. A leak > 25% of the tidal volume occurred at the lowest pressure in FM [median (range), 6 (6–8) cmH2O], which was significantly lower than with v-gel [16 (6–no leak at 16) cmH2O], LM [>16 (6–no leak at 16)] or ETT [>16 (no leak at 16) cmH2O] (p < 0.001). On CT images, the height and width of the larynx were significantly smaller with v-gel in comparison to FM and LM (p = 0.004). A significant increase in the amount of gas in the stomach (p = 0.007), but not gastric volume, was detected in FM and LM.

Conclusions and clinical relevance

The v-gel is a practical alternative to LM and ETT for airway management and CMV, but can compress the larynx. The FM is easily placed, but significant leakage occurs during CMV.  相似文献

In this study, the efficacy period of an intraperitoneal vaccination and effect of a booster shot of vaccine against herpesviral haematopoietic necrosis (HVHN) in goldfish Carassius auratus were investigated. Cell culture supernatant of cyprinid herpesvirus 2 (CyHV-2), causative agent of HVHN, propagated in goldfish fin (GFF) cells was inactivated with formalin (0.1%, v/v) for 2 days at 4 °C. Three groups of the variety Ryukin were individually intraperitoneally injected with the vaccine and each group was separately maintained in replicate tanks. After 4 weeks (Vaccinated-4w-1 and 2) and 8 weeks (Vaccinated-8w-1 and 2) from the first vaccination, the fish were CyHV-2-challenged by the immersion route (10 TCID50 l−1). In addition, the other vaccinated group of fish were injected with a booster vaccine 4 weeks after the first vaccination as the Vaccinated-booster groups, then the fish of these groups were CyHV-2-challenged by the immersion route (10 TCID50 l−1) after 8 weeks from the first vaccination. The mean of the relative percentage survival (RPS) values of the Vaccinated-4w and 8w groups showed 42.5% and 57.6%, respectively. In addition, the mean RPS value of Vaccinated-booster groups showed 63.6%. Statistical analysis showed significantly higher survival rates in all the vaccinated groups than those of the respective negative control groups using Fisher's exact test. Moreover, the survival rates of vaccinated-booster groups were significantly higher (p = 0.036) compared with the respective control groups by Student's t test. The present study shows the efficacy period of the vaccine is at least 8 weeks and a booster shot showed a tendency to enhance the protection against HVHN in goldfish.  相似文献

Denileukin Diftitox (ONTAK®, DAB389 IL-2) is a recombinant DNA-derived fusion protein depleting cells that express high-affinity IL-2 receptor. Important cell targets are CD4+CD25+Foxp3+ regulatory T cells (Treg). Elimination of immunosuppressive Treg by Denileukin Diftitox may provide a way to modulate immune tolerance following stem cell transplantation. Here, we combined Treg depletion with a vaccination approach to induce donor-specific immune reactions. To investigate this approach we chose the mixed chimerism canine stem cell transplantation model which represents a high state of tolerance between two hematopoietic systems. The aim was therefore to induce a graft versus hematopoiesis effect thereby converting mixed to full donor chimerism. Dog leukocyte antigen identical siblings that had developed a stable mixed chimerism after non-myeloablative stem cell transplantation received a single dose of Denileukin Diftitox (18 μg/kg, i.v.) followed by several cell-lysate vaccinations. Host peripheral blood mononuclear cell lysates combined with CpG-ODN, and Montanide® ISA 51 were locally applied. In vitro studies demonstrated that canine Treg are a target of Denileukin Diftitox. The suppression of T-cell proliferation by Treg was abolished by addition of Denileukin Diftitox (10 nM). An increase of proliferation of median 300% (range: 200%–425%) was observed. No change in donor chimerism was observed after administration of Denileukin Diftitox and vaccination. This study highlights that application of Denileukin Diftitox resulted in a depletion of Treg followed by an increase of immune response in vitro. This effect could not be confirmed in vivo even if the immune system was stimulated by vaccinations.  相似文献

In this study the disposition kinetics and plasma availability of moxifloxacin in Muscovy ducks after single intravenous (i.v.), intramuscular (i.m.) and oral (p.o.) administrations of 5 mg kg?1 b.wt. were investigated. The concentrations of moxifloxacin in the plasma were measured using high-performance liquid chromatography (HPLC) with fluorescence detection on samples collected at frequent intervals after drug administration. Following intravenous injection, the decline in plasma drug concentration was bi-exponential with half-lives of (t1/2α) 0.22 ± 0.10 h and (t1/2β) 2.49 ± 0.26 h for distribution and elimination phases, respectively. The volume of distribution at steady-state (Vdss) was 1.02 ± 0.14 l kg?1 and the total body clearance (Cltot) was 0.32 ± 0.11 l kg?1 h?1, respectively. After intramuscular and oral administration of moxifloxacin at the same dose the peak plasma concentrations (Cmax) were 2.38 ± 0.43 and 2.11 ± 0.36 μg ml?1 and were obtained at 1.47 ± 0.26 and 1.83 ± 0.16 h (Tmax), respectively, the elimination half-lives (T1/2el) were 3.14 ± 0.42 and 2.63 ± 0.44 h, respectively, and AUC0–24 were 15.87 ± 2.35 and 14.52 ± 2.37 μg ml?1 h?1, respectively. The systemic bioavailabilities were 96.36 ± 11.54% and 86.79 ± 12.64%, respectively. In vitro plasma protein binding percent was 32%. We concluded that moxifloxacin might be clinically interesting alternative for the treatment of most sensitive bacterial infections in Muscovy ducks.  相似文献
