In this study, full-length tilapia transferrin (OnTF) isolated from liver cDNA of Nile tilapia (Oreochromis niloticus) was found to have an open reading frame of 2,091-bp encoding 696 amino acid residues. Two additional amino acids: Gly369 and Gly370 were observed compared with the reported Nile tilapia transferrin protein sequence. Pre-mature protein has a predicted molecular weight of 78.2 kDa, while mature protein is 73.28 kDa in size. Comparative sequence analysis with transferrin from other species revealed two major putative iron-binding domains designated as the N-lobe and the C-lobe in accordance with the transferrin protein characteristics. The predicted tertiary structure of tilapia transferrin confirmed the presence of iron and anion-binding sites on both lobes that are conserved among transferrins from other species. Quantitative real-time PCR analysis showed significantly higher expression of tilapia transferrin gene in liver than in other tissues (p < 0.05). Transferrin expression in tilapia experimentally infected with 106 and 108 colony-forming units mL?1 of Streptococcus agalactiae was significantly upregulated at 24 and 12 h post-infection (hpi), respectively, and decreased afterward. Iron-deficiency in serum of bacterially infected fish was detected at 48 and 24 hpi, respectively. The expression pattern of the transferrin gene and the iron levels of infected tilapia in this study were consistent with the function of transferrin in innate immunity.  相似文献

To understand the detoxification and bioactivation mechanisms for organic contaminants, it is essential to identify the cytochrome P450 (CYP) complement. Therefore, this study aimed to clone a partial cDNA sequence of the novel CYP1D1 gene from the fish Oreochromis niloticus and examine whether intraperitoneal injection of benzo-a-pyrene (BaP), a potent AHR agonist, is capable of inducing CYP1D1 mRNA expression in different tilapia fish tissues. The cloned nucleotide sequence consisted of 713 bp representing a portion of the tilapia CYP1D1 cDNA ORF, encoding 237 amino acids. Amino acid sequence comparison of O. niloticus CYP1D1 with the sequences of CYP1D1 from other species showed that this gene shared the highest identity of 81% with Fundulus heteroclitus CYP1D1. Furthermore, analysis of the percent identities shared by the deduced amino acid sequence of O. niloticus CYP1D1 with the sequences of CYP1 from other species revealed that the highest identities were shared with fish CYP1As. Real-time PCR results revealed that the highest expression level of CYP1D1 mRNA was found in muscles, followed by gills, liver, and intestine, while there was no detectable expression recorded in bile acid. These results indicate that tilapia CYP1D1 plays an important role in the metabolism of xenobiotics, expanding our knowledge regarding the diversity of CYP1 genes in this important model fish species.

A 45-day feeding trial was carried out to evaluate the use of shrimp protein hydrolysate (SPH) in diets for Oreochromis niloticus, L. SPH was included in isonitrogenous diets replacing fish meal protein by 0, 5, 10, and 20% and offered to Nile tilapia juveniles (1.7 ± 0.4 g) stocked in 40-L glass aquaria. The inclusion of SPH produced no significant differences (P ≥ 0.05) in final weight, survival, weight gain, average daily gain, specific growth rate, feed conversion ratio, protein efficiency ratio, or apparent net protein utilization. The inclusion of SPH Nile tilapia diets significantly affected (P < 0.05) the final fish body composition. Protein and ash contents decreased and fat content increased slightly with the increase in SPH. This study has demonstrated that SPH is a promising protein feedstuff and could account for as much as 6% of Nile tilapia diets with no adverse effects on growth and nutrient utilization.  相似文献

Despite a worldwide boom in tilapia aquaculture, South Africa has failed to follow suit, resulting in a small, very much undeveloped local industry. Much of the justification for this reality lies on the inadequacy of the species used and the stringent legislation preventing the use of the Nile tilapia, Oreochromis niloticus. With momentum gained in recent years, the local industry lobbying for the legalization of the O. niloticus has made great progress and its use in certain systems has recently been facilitated. This study was conducted to compare the growth performance between the indigenous Mozambique tilapia, Oreochromis mossambicus, another African alternative gaining momentum in Zambia, the Three Spot tilapia, Oreochromis andersonii, and the globally reputable O. niloticus. The study was conducted in a biofloc technology (BFT) system, showing great potential as an environmentally friendly technology. The trial was run for 10 weeks with 60 fish/tank stocked into four repeats tanks per species. Fish were sampled on six occasions with a regression fitted to the mass data. Oreochromis niloticus showed a significantly higher growth rate with an average daily gain of 0.693 ± 0.018 g/day and the lowest feed conversion ratio (FCR) of 1.01 ± 0.05, followed by O. mossambicus with average daily gain of 0.405 ± 0.025 g/day and FCR of 2.24 ± 0.16, and then O. andersonii with an average daily gain of 0.185 ± 0.025 g/day and an FCR of 2.53 ± 0.28, respectively. From this study, O. niloticus therefore appears to be the most adequate species for use in BFT systems in South Africa.  相似文献

MVSF‐12 belonging to megalocytivirus type 3 was isolated from cultured starry flounder; Platichthys stellatus, at the moribund or subclinical stage with low mortalities in Korea. Of 20 apparently healthy fish in the farms, 17 were also confirmed in nested polymerase chain reaction to be infected by megalocytivirus. When starry flounder; rock bream, Oplegnathus fasciatus; and olive flounder, Paralichthys olivaceus, were artificially infected by MVSF‐12 or iridovirus sachun‐1 (IVS‐1, megalocytivirus type 1), starry flounder and olive flounder showed no mortality until Day 24, without any clinical signs including enlarged spleen, while rock bream showed 100% mortality by IVS‐1 infection within 11 d but no mortality by MVSF‐12. Although it was not pathogenic, MVSF‐12 in infected fish at Day 24 was viable when successfully cultured in vitro using primary rock bream embryo cells and produced a cytopathic effect (CPE) with the viral copy numbers between 1.76 × 107 and 5.23 × 107/mL of culture supernatant. In conclusion, this study demonstrates the low pathogenicity of MVSF‐12 and low susceptibility of starry flounder and olive flounder to both MVSF‐12 and IVS‐1. Indeed, MVSF‐12 at the subclinical stage could be replicated with CPE in vitro, indicating a possibility to induce pathogenic effects and mortality under adverse environment or physiologic conditions.  相似文献

IGF-I is a mitogenic polypeptide that is an important regulator of growth in fish. The potential of IGF-I mRNA abundance as a rapid growth indicator in the Nile tilapia, Oreochromis niloticus, was evaluated. Hepatic IGF-I cDNA was isolated and partially cloned. The partial sequence having 539 bases encodes for the signal peptide, mature protein and a portion of the E domain. The deduced 68 amino acid sequence for mature IGF-I showed 84–90% and 77–79% sequence identity with fish and mammalian counterparts, respectively. The deduced amino acid sequence for domains B and A was most conserved (93–97%) relative to other fishes. A sensitive TaqMan real time qRT-PCR assay for O. niloticus was developed based on the mature IGF-I peptide for measures of hepatic IGF-I mRNA levels. Hepatic IGF-I mRNA levels were found to be significantly correlated with growth rate of fish reared under different feeding regimes and temperature conditions. Higher feed consumption and water temperature produced faster-growing fish and increased hepatic IGF-I mRNA expression. These findings suggest that hepatic-derived IGF-I plays a key role in controlling growth in O. niloticus and indicates that IGF-I mRNA quantification could prove useful for the rapid assessment of growth rate in this species.  相似文献

Production of sterile triploid red tilapia [Oreochromis mossambicus (Mozambique tilapia); Peters, 1852 × Oreochromis niloticus (Nile tilapia); Linnaeus, 1758] is an effective strategy to overcome their prolific breeding. Optimal conditions for cold-shock induction of triploidy in red tilapia were investigated by experimentally examining two variables: appropriate temperature of the shock and duration of shock treatment. A constant time after insemination of 4 min was used to determine the best combination of temperature (6, 7, 8, 9, 11, 13, 15 °C) with different durations of shock (10, 20, 30, 40, 50 min) with resultant ploidy level verified karyotypically. Shock duration for 30 min at a temperature of 9 °C was found most effective in producing maximum triploidy (98.7 %) with higher rates of hatching (63.2 %) and survival up to yolk-sac stage (75.8 %). The chromosome count confirmed that triploid percentages were higher when cold shock was used for longer durations at each temperature; however, hatching rates were generally decreased. The maximum triploid yield (82.1 %) obtained was higher than the yield obtained using heat shock (72.7 %) in red tilapia previously. The application of the results of this study has the potential to greatly improve the production of triploid red tilapia in commercial aquaculture.  相似文献

The aim of this study is to evaluate the effects of total replacement of fish oil (FO) with pork lard (PL), soybean oil (SO), or tea seed oil (TO) on growth and fatty acid compositions of hybrid tilapia (Oreochromis niloticus × O. aureus). Four diets were formulated to contain 4.5 % of each of the different lipid sources. Each diet was assigned to triplicate groups of 15 hybrid tilapia with the same initial body weight (4.6 ± 0.5 g). After a 10-week feeding, the FO, SO, and TO groups had similar final body weight. The mean hepatosomatic index in the FO group was significantly lower than the other groups (2.5 vs 2.9–3.0; P < 0.05). Whole-body lipid levels were highest in the TO group (5.2 ± 0.4 %) and lowest in the FO group (4.3 ± 0.5 %). Total n-3 polyunsaturated fatty acids (PUFA) concentrations were greater in the SO and FO groups (5.5 ± 0.1 and 7.1 ± 0.2, respectively) than in the PL and TO groups (1.7 ± 0.1 and 1.4 ± 0.1, respectively). The FO group had a 2- to 4-fold reduction in the n-6/n-3 PUFA ratio relative to the other groups. The concentrations of both docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were significantly (P < 0.05) higher in the FO group than in the other groups. Our data demonstrate that total replacement of dietary FO with SO or TO has little effects on growth of hybrid tilapia but reduces the contents of health-benefiting DHA and EPA in fish body.  相似文献

Genetically improved farmed tilapia (GIFT) (Oreochromis niloticus) is an important aquaculture species. Lipoprotein lipase (LPL) is considered as a key enzyme in lipid metabolism and deposition. The present study was conducted to investigate the nutritional regulation of LPL in GIFT. We cloned and characterized the LPL gene from GIFT. Finally, we determined the effects of dietary lipid levels and refeeding on hepatic LPL gene expression in GIFT. The LPL gene of GIFT (Oreochromis niloticus) (O.nLPL) was 2,298 bp in length and encoded 515 amino acids. Sequence analysis showed that O.nLPL shared 57.3–87.9 % identity with LPLs from other piscine species. To study LPL expression patterns, juveniles GIFT were fed diets containing 3.7, 7.7 or 16.6 % crude lipid for 90 days and the expression of hepatic O.nLPL was examined using real-time PCR. The abundance of hepatic LPL mRNA increased with increasing dietary lipid. The expression of O.nLPL mRNA in the 16.6 % dietary lipid group was significantly higher than that of the 3.7 % lipid group (P < 0.05). The expression of O.nLPL was increased in GIFT following a 48-h fast and decreased 12 h after refeeding. Hepatic LPL mRNA returned to fasting levels 48 h after refeeding. In summary, high dietary lipid induced expression of liver O.nLPL, and expression of liver LPL is regulated by fasting and refeeding.  相似文献

Lionfish are representative venomous fish, having venomous glandular tissues in dorsal, pelvic and anal spines. Some properties and primary structures of proteinaceous toxins from the venoms of three species of lionfish, Pterois antennata, Pterois lunulata and Pterois volitans, have so far been clarified. Our recent survey established the presence of hyaluronidase, presumably a toxin-spreading factor, in the venoms of P. antennata and P. volitans. This prompted us to examine enzymatic properties and primary structures of lionfish hyaluronidases. The hyaluronidases of P. antennata and P. volitans were shown to be optimally active at pH 6.6, 37 °C and 0.1 M NaCl and specifically active against hyaluronan. These enzymatic properties are almost the same as those of stonefish hyaluronidases. The primary structures (483 amino acid residues) of the lionfish hyaluronidases were elucidated by a cDNA cloning strategy using degenerate primers designed from the reported amino acid sequences of the stonefish hyaluronidases. Both lionfish hyaluronidases share as high as 99.6 % of sequence identity with each other and also considerably high identities (72–77 %) with the stonefish hyaluronidases but rather low identities (25–40 %) with other hyaluronidases from mammals and venomous animals. In consistent with this, phylogenetic tree analysis revealed that the lionfish hyaluronidases, together with the stonefish hyaluronidases, form a cluster independently of other hyaluronidases. Nevertheless, the lionfish hyaluronidases as well as the stonefish hyaluronidases almost maintain structural features (active site, glyco_hydro_56 domain and cysteine location) observed in other hyaluronidases.  相似文献

The present study was designed to examine the potential for inducing ovulation in starry flounder (Platichthys stellatus) using gonadotropin-releasing hormone analog (GnRHa) and human chorionic gonadotropin (hCG) to assess whether starry flounder are differentially responsive to GnRHa and hCG. Female starry flounder were injected or implanted with different doses of hCG or GnRHa pellets to examine their ovulation-inducing potential and effects on plasma levels of testosterone (T), 17β-estradiol (E2), and 17,20β-dihydroxy-4-pregnen-3-one (17,20βP). Blood samples were collected for up to 10 or 25 days post-injection or post-implantation in two separate experiments designed to mimic the early and middle stages of spawning, respectively. Fish treated with the GnRHa pellets (100 µg) showed a significant increase in the total number of stripped eggs relative to the controls. GnRHa administration had no effect on the floating rate or fertilization rate of ovulated eggs in the both experiments, whereas hCG injection affected both of these rates. Plasma T levels were not significantly different between the exogenous hormone-treated and control fish. In contrast, the plasma E2 level was elevated in those fish treated with GnRHa, regardless of injection or implantation, and was accompanied by increased numbers of stripped eggs in both experiments. Treatment with GnRHa resulted in higher 17,20βP levels compared to the controls, and there was a positive relationship between elevated plasma 17,20βP and an increase in ovulated eggs in response to GnRHa treatment. The implantation of starry flounder with GnRHa-containing pellets was effective at inducing sustained ovulation compared to hCG treatment.  相似文献

测定了养殖星斑川鲽肌肉中的蛋白质、脂类和灰分等生化组分,并计算了其比能值,分析了肌肉中构成蛋白质的16种常见氨基酸和主要脂肪酸含量。结果表明,星斑川鲽属高蛋白、低脂肪鱼类;肌肉氨基酸含量同其他鲆鲽鱼类相比,属中上等水平;必需氨基酸和呈味氨基酸的含量较高。肌肉蛋白中氨基酸的支/芳值为2.33。各类脂肪酸中,油酸(18:In)含量最高,其次为亚油酸(18:2n6)和棕榈酸(16:0)。星斑川鲽富含EPA和DHA,营养丰富且平衡良好,是值得推荐的优良养殖品种和优质食用鱼类。  相似文献

Major fish bacterial diseases in Korea are edwardsiellosis, streptococcosis, and vibriosis. Among vibrionaceae, Listonella anguillarum, Vibrio harveyi, V. ichthyoenteri, and Photobacterium damselae were identified as causative organisms of vibriosis in flounder. In this study, we developed a multiplex PCR method using the RNA polymerase β subunit (rpoB) gene, known as a housekeeping gene for identification of Vibrio spp. causing vibriosis in flounder. Three pairs of PCR primers were designed based on the rpoB sequence of three species, V. harveyi, V. ichthyoenteri, and P. damselae. The PCR assay, using a mixture of six primers, yielded amplicons of 601, 434, and 533 bp in V. harveyi, V. ichthyoenteri, and P. damselae. None of the untargeted species yielded an amplicon. The detection limits for pure culture in kidney were 2.5 × 104 cfu/g kidney for V. harveyi, 2.5 × 105 cfu/g kidney for V. ichthyoenteri, and 2.5 × 106 cfu/g kidney for P. damselae. From the colonies on TCBS agar plates of different samples, 632 Vibrio spp. isolated from aquacultured flounder between 2004 and 2010 were identified by the multiplex PCR method. As a result, 265 strains (41.9 %) were V. ichthyoenteri; 115 strains (18.2 %) were V. harveyi and 72 strains (11.4 %) were P. damselae.  相似文献

This study investigated the efficacy of the aqueous fraction obtained after fractionating silage of tilapia (Oreochromis niloticus) and cobia (Rachycentron canadum) in supporting the growth of Escherichia coli and Staphylococcus aureus. The silages were prepared using combinations of citric, formic, and propionic acids. The aqueous fractions, used as test peptones, contained lower levels of total protein. The concentrations of 18 amino acids in all the samples were observed to be lower than those in the commercial peptone. Glutamic acid, lysine, glycine, and aspartic acid were present in higher concentrations than other amino acids, for both types of silage. Biomass production from E. coli culture ranged from 38.4 to 65.9 mg 100 mL?1 for all the tested treatments, while that for S. aureus was from 26.3 to 53.7 mg 100 mL?1. This indicated that products from fish silage were effective for bacterial growth in terms of biomass, by providing the main sources of nitrogen and carbon to facilitate their growth. The tested silages yielded similar efficiency to the commercial peptone. The findings revealed that it is feasible for the fish processing industry to incorporate freeze-dried by-products obtained after fractioning waste silage from the processing of tilapia and cobia.  相似文献

The objective of this study was to investigate the dietary effect of vitamin C in amelioration some of studied alterations induced by fenitrothion in Nile tilapia (Oreochromis niloticus). Nile tilapia was exposed to sub-lethal concentration of fenitrothion 0.04 mg/l (96 h LC50 value was 0.8 mg/l), and basal diet was supplemented with two different dose of vitamin C (500 and 1200 mg/kg B wt/day) for 30 days. Vitamin C supplemented groups showed significant decrease in plasma cortisol and glucose level, enzymes activity of liver and gills (catalase, glutathione-S-transferase and superoxide dismutase) and % of tail DNA damage compared to exposed group. Moreover, fish revealed significant increase in total plasma protein, albumin, globulin and A/G ratio. High dose of vitamin C dietary supplementation (1200 mg/kg B wt/day) returns these parameters to its normal levels with no significant difference compared to non exposed control group. These results indicated that incorporation of high dose of vitamin C (1200 mg) in aqua feed for 30 days could be potentially less expensive and effective in reducing the alterations induced by fenitrothion in Nile tilapia.  相似文献

Francisella noatunensis orientalis is a bacterium that causes emerging bacteriosis in Nile tilapia (Oreochromis niloticus) in many parts of the world, including Brazil. It is a non-motile, Gram-negative, strictly aerobic, facultative intracellular coccobacillus. This species of bacteria is responsible for low to high mortality in fish farms, causing economic losses for fish farmers. This study aimed to detect the presence of F. noatunensis orientalis using qPCR (real-time polymerase chain reaction) and to describe lesions caused by the bacterium in O. niloticus in Brazilian aquaculture. For this purpose, 360 fish from six fish farms (30 per farm) were sampled at two time points (n = 180 per sampling). Necropsies and histopathology were performed for lesion observation, in addition to qPCR and sequencing for detection and identification of Francisella species. Environmental data were collected using a multiparameter sonde YSI EXO2. All measured limnological variables were within the optimum range for cultivation of Nile tilapia. The major lesions present were melanization of the skin, splenomegaly, granulomas, and inflammatory cell responses. The prevalence of francisellosis varied from 0 to 86.66% between time periods and fish farms analyzed, and an outbreak was observed during the second sampling period. This study describes the prevalence of francisellosis in O. niloticus and reports that the lesions found are not exclusively associated with this bacterial disease.  相似文献

Tilapia, Oreochromis niloticus, is among the fish species with high potential for aquaculture in intensive farming, and Brazil is among the largest producers worldwide. Some of the amino acid requirements in practical diets for tilapia are still unknown. Thus, this study determined the dietary arginine requirements for Nile tilapia juveniles based on growth performance, hematological and biochemical responses, and muscle growth. Three hundred Nile tilapia juveniles (2.95 ± 0.79 g) were distributed into 20–500 L fiberglass aquaria and fed five extruded isoproteic (28% crude protein) and isoenergetic (3160 kcal/kg) diets formulated to contain 0.95, 1.10, 1.25, 1.40, and 1.55% arginine. Based on the quadratic regression analysis, the best results in weight gain, feed conversion, protein efficiency ratio, and protein retention were estimated in fish fed diets containing 1.36, 1.34, 1.36, and 1.37% arginine, respectively. The best amino acid body retention values were estimated in fish fed diets containing 1.31–1.37% arginine. Muscle growth occurred mainly by hyperplasia in fish fed 0.95% arginine, whereas reduction in the hyperplasia time and signs of hypertrophy occurred in fish fed 1.10–1.55% arginine diets. It was concluded that a diet with 1.36% of arginine (with 1.53% lysine in diet) meets the requirements of Nile tilapia juveniles.  相似文献
