Age-related NADH oxidase (arNOX) is a cell surface protein shed into the circulation and other body fluids, which generates superoxide. The activity increases with age in human tissues and body fluids (serum, saliva, and perspiration) and is a potential source of age-related oxidative damage. We measured arNOX activity in the coelomic fluid of sea urchin species with different life spans. Coelomic fluid of long-lived sea urchin species Strongylocentrotus purpuratus and Strongylocentrotus franciscanus exhibited low levels of arNOX compared to the short-lived urchin species Lytechinus variegatus. arNOX activity was positively correlated with animal size in L. variegatus, whereas with S. purpuratus and S. franciscanus, arNOX activity and animal size were inversely correlated. The inverse correlation of arNOX activity with life span and decreased levels of arNOX with age in the long-lived species is consistent with a contribution of reduced arNOX activity to slower aging.  相似文献

Due to their marked antiinflammatory effect, synthetic corticosteroids are used to mask illness, especially lameness in horses. The detection of these drugs in equine body fluids requires accurate methods, particularly where misuse of corticosteroids is suspected. Gas chromatography/mass spectrometry (GC/MS) is well established as a reliable technique for the identification of drugs in biological fluids. Using GC/MS, we determined dexamethasone levels in horse urine and serum after intravenous application of a therapeutic dose. Dexamethasone was detectable, in serum for up to six hours, and in urine for up to 32 hours, after its administration. These findings indicate that serum measurements are unreliable for the detection of corticosteroid abuse, and demonstrate urine to be a more suitable body fluid for investigation. Nevertheless, it should be emphasized that, regardless of the technique employed, the clinical effects of dexamethasone last longer than 32 hours; thus, failure to detect dexamethasone does not disprove corticosteroid abuse.  相似文献

All evidence to date suggest that sperm motility is the primary determinant of fertilization success in externally fertilizing fish species. Ovarian fluid, which comprises 10–30% of the total egg volume in salmonids, enhances sperm motility with respect to swimming speed, trajectory and the duration of movement. It was recently demonstrated that there is individual variability in sperm motility enhancing potential of ovarian fluid of particular females. In the present study we examined the effect of particular ovarian fluids collected from 31 females on the sperm motility parameters of one male of rainbow trout (Oncorhynchus mykiss) using computer-assisted sperm analysis (CASA). During our experiment we also monitored the pH of ovarian fluid. We found that particular fluids differed in the ability to activate spermatozoa; sperm remained immotile in four fluids and exhibited 50–100% motility in 27 samples. The percentage of motile sperm, velocity and duration of movement positively correlated with ovarian fluid pH (r2 = 0.34–0.62). These data strongly suggest that the pH of the ovarian fluid is the primary determinant of sperm motility in rainbow trout under natural conditions of fertilization.  相似文献

ABSTRACT:   The ability of salmon growth hormone (sGH) to accelerate the somatic growth of juvenile abalone Haliotis discus hannai by immersion was examined. After immersion for one hour into a sGH-rich solution at a concentration of 30 mg/L, the sGH immunoreactivity in the body fluid of abalone was maximal after one day, and levels were still detectable at two days. No immunoreactivity was observed in the control group. Following immersion for one hour at one or two-week intervals for 120 weeks into the sGH-rich solution at 30 mg/L, the sGH-immersed abalone exhibited a significant increase in shell length and body weight. On the other hand, abalone immersed into a salmon prolactin (sPRL)-rich solution at 30 mg/L for one hour showed a lower increase in both shell length and body weight than the sGH-immersed abalone. No increases in shell length and body weight were observed in either the bovine serum albumin-immersed abalone or untreated controls. These results provide evidence that sGH can be transported from ambient water into the circulatory system of abalone, and can subsequently improve the somatic growth of juveniles.  相似文献

The protein composition of seminal fluid, blood serum, sperm plasma membrane and flagellum of rainbow trout were analysed by SDS-polyacrylamide gel electrophoresis. Immunological identity between proteins of the 2 fluids and sperm components was studied using crossed immunoelectrophoresis, rocket immunoelectrophoresis and immunoblotting. Results indicate that many seminal proteins are antigenically-related to serum proteins, proteins of sperm origin are present in seminal fluid in varying amounts, depending on the animals and sampling time, and several serum-like seminal proteins are bound to spermatozoa. Lipoproteins were isolated from seminal fluid (mean level: 33 μg/ml) and characterized. They were identified as being HDL-like lipoproteins. A possible physiological role is proposed for these seminal lipoproteins.  相似文献

Methods for the isolation and quantification of infectious pancreatic necrosis virus (IPNV) from ovarian and seminal fluids of Atlantic salmon are described. Both have utility for the non-lethal detection of IPNV in mature broodstock and for research into vertical transmission. Two experiments are described to check the efficiency of an elution method for the removal of IPNV from milt. The isolation rate for ovarian fluid of females was generally higher than that for seminal fluid of males from the same populations. In IPNV milt mixing experiments up to 99.98% of available IPNV adsorbed to Atlantic salmon spermatozoa and 20-100% of virus eluted using a variety of procedures. Titration of virus from naturally infected milt can be useful in estimating the relative vertical transmission risk from male broodstock.  相似文献

The Antarctic snailfish, Paraliparis devriesi (Liparididae), occupies an epibenthic habitat at a depth of 500–650 m in the subzero waters of McMurdo Sound, Antarctica. This species has watery (97%) gelatinous subdermal extracellular matrix (SECM) comprising a mean of 33.8% of the body weight, the largest known proportion of any adult fish. The protein concentration of the SECM was found to be 6–7 mg ml–1 (0.6–0.7% w/v). Separation of the polypeptides of the SECM by SDS-PAGE revealed 11 polypeptides ranging in relative molecular mass (Mr) from 67,000 to 13,000, with other unresolved polypeptides of less than 13,000. The isoelectric points of these proteins ranged from 4.85 to 8.05. Partial N-terminal amino acid sequence data were obtained for four of the major SECM polypeptides. The N-terminal amino acid sequences of three of these were not identical to or homologous with any other known sequences, whereas the N-terminal sequence of one polypeptide (Mr 51,000) was identical to partial sequence from the apolipoprotein A-I precursor of Atlantic salmon (Salmo salar). Although not isolated from either SECM or serum, melting point-freezing point behavior of body fluids suggest that Paraliparis possess modest amounts of a noncolligative antifreeze compound. Since relatively small amounts of antifreeze are present in the serum and even less in the SECM, freezing avoidance results from the combined effects of antifreeze and the elevated osmolality of body fluids. There are no special adaptations to prevent freezing in the superficially located high water content SECM.  相似文献

为了精确且批量测定草鱼肠系膜脂肪(肠脂)沉积量,避免传统人工刮取称量方法耗时费力、量化粗糙等问题,本研究利用脂溶性染料油红O可特异性着色脂肪的特性,探索开发出一种便捷定量草鱼肠脂含量的新方法。测试发现,麻醉解剖200尾平均体质量约80 g的草鱼,取出整个内脏团简单处理并塞入PIT个体识别标记后,样品集中进行多聚甲醛固定、无水乙醇脱水、油红O染液定染,可在维持样品组织完整的基础上,实现肠脂组织的特异性、均一化批量染色处理。各染色样品再分别通过无水乙醇溶剂完全萃取,萃取液吸光度测定,并依据绘制的标准曲线(y = 0.0276x + 0.0403, R2 = 0.9997),即可精确获得个体肠脂沉积的相对含量,是以萃取出的油红O质量来表示。对比发现,样品肠脂组织染色-萃取量化结果与传统刮取称量数据保持了较高的相关性(n = 20,r = 0.80)。进一步的统计分析显示,对于同塘养殖体质量变异系数8.93%的草鱼群体(n = 200),萃取测定的肠脂沉积量变异系数达到24.49%,预示该性状具有丰富变异特征及遗传改良潜力。相关与聚类分析显示,肠脂沉积量与内脏质量相关性最高(r = 0.60),并且聚为一类,符合二者同属脏器关联指标的预期。多元线性回归分析显示,利用简单易测的形态指标只能解释肠脂沉积量的少量变异(R2 = 0.20),表明基于表型性状的拟合回归方程进行间接预测的效果不佳,直接测定是该性状精准量化的有效途径。本研究为草鱼体脂性状改良提供了一种性状精确测定方法。  相似文献

本文应用高等哺乳动物血清补体B因子溶血检测技术检测仿刺参个体体内补体B因子。通过溶血法检测了健康仿刺参以及人工诱导化皮仿刺参体腔液补体B因子活性。结果显示仿刺参体腔液有补体B因子存在,且含补体B因子仿刺参体腔液溶血活性明显高于去补体B因子体腔液的溶血活性。实验对仿刺参体腔液进行加热处理结果显示其补体B因子是对热不稳定的分子,此特性与高等哺乳动物B因子特征相同。仿刺参体内存在补体B因子,它与脊椎动物补体B因子性质相似,在免疫旁路途径中起重要作用。  相似文献

Abstract. Hypoosmoregulatory capacity was examined in anadromous Arctic char, Salvelinus alpinus (L.), caught whilst migrating towards the sea in the river Å-elva, northern Norway (69°04′N and 17°00′E). Metacercariae of the marine digenean Cryptocotyle lingua (Creplin) were found on 47% of the fish investigated, demonstrating that they had been in brackish or sea water on at least one previous occasion. The fish were randomly divided into three groups, and exposed to sea water (35 ppt salinity at 5–6°C) for 7 days- one group immediately after capture, and the other two groups after 16 days of acclimation in fresh water and brackish water (15–17ppt), respectively. Blood plasma osmolality, Na+ and Mg2+ levels showed only a small and transient rise during seawater exposure, and there were only minor changes in muscle water content, irrespective of prior treatment. A slight, but insignificant, reduction in hypoosmoregulatory capacity appeared to occur in the groups retained in fresh water or brackish water for 16 days prior to seawater exposure. The results did not indicate that the fish show any improvement of hypoosmoregulatory capacity following a period of acclimation in brackish water. The hypoosmoregulatory capacity of seaward-migrating Arctic char is well developed before they enter the sea, and may be comparable to that seen in salmonids which undergo a parr-smolt transformation. There was a negative correlation between blood plasma electrolyte levels and both body length and weight, indicating that hypoosmoregulatory capacity was affected by the size of the fish. The effects of body size upon the development of hypoosmoregulatory capacity are discussed with respect to estuarine residence and the constraints placed upon the survival of small fish in full-strength sea water.  相似文献

胰多肽(pancreation polypeptide,PP)是胰脏胰多肽细胞分泌的一种多肽类激素。为了研究胰多肽对肉鸭生产性能和消化机能的影响,实验通过从肉鸭小肠中提取PP粗品,并对其进行纯度鉴定,之后添加到肉鸭体内,产生动物模型,以进行动物实验。具体方案如下:100只肉鸭被随机分为5组,每组20只。Ⅰ组为对照组,每天饲饮自来水。Ⅱ、Ⅲ组分别饲饮剂量为0.58mg/d的PP粗品及剂量为0.29mg/d的PP纯品。Ⅳ、Ⅴ组分别于每周颈部皮下注射0.24mg/只的PP粗品及0.12mg/只PP纯品。饲养称重,宰杀后取小肠,对其进行处理后用紫外吸收法进行消化酶活力测定。结果表明:PP能促进肉鸭机体生长发育,促进体重增长,对肉鸭生产性能产生影响;对肉鸭小肠蛋白酶、淀粉酶和脂肪酶的活力有影响,从而影响消化机能。  相似文献

传统疫苗是基于抗原刺激机体产生特异性抗体的原理,它们大多数激发机体的体液免疫,很难启动细胞免疫。而核酸疫苗是将外源基因与真核质粒重组后直接导入细胞内,使外源基因在宿主细胞内表达合成保护性抗原蛋白。这是模拟病毒自然感染提呈过程,这既能产生细胞免疫,又能产生体液免疫。本文就核酸疫苗的构成、特点、影响免疫效果的因素、研究进展及其存在的问题和展望作一综述。  相似文献

Certain physiological responses of two groups of rainbow trout acclimated at 10 and 17°C to a sudden change in salinity were investigated in order to determine the most appropriate temperature for direct transfer in salt water. Plasma osmolarities of fish acclimated at 17°C increased at a much faster rate following transfer and approached a much higher value than that reached by the fish acclimated at 10°C. The same was apparent for body tissue dehydration. Haematocrit values, the number of red blood cells, as well as red cell volume showed no significant difference between the two groups of fish for most of the experimental period. The larger increases in plasma osmotic pressure and tissue dehydration observed at 17°C are discussed in connection with the physiological adjustments accompanying higher temperature acclimation.  相似文献

The aims of this study were to establish an accurate size measurement method and a nutritional condition evaluation method of Holothuria scabra (Jaeger). Although 0.5% KCl and 0.05% MgSO4 did not induce anaesthesia, 2% menthol–ethanol for 20 min was found to be effective and harmless. The anaesthetization significantly reduced the coefficient of variation of the mean body length and weight by 68% and 43% respectively. During starvation, body size and weight decreased concomitantly, resulting in an unchanged condition factor (body weight/volume), suggesting that the condition factor cannot be used as an index of nutritional condition. Protein, cholesterol and carbohydrate concentrations in the body fluid were analysed to study the relationship with starvation. As the protein and cholesterol concentrations initially increased and then decreased during the starvation period, it is difficult to use them as an index of nutritional condition. The carbohydrate concentration showed a gradual one‐fold increase during 10 days of starvation, and it may be used as a proxy for nutritional condition; however, further physiological studies are needed. Body fluid density and volume relative to body size gradually increased and decreased, respectively, during starvation. These methods may be used to correctly monitor the conditions of H. scabra in studies for aquaculture and stock enhancement techniques.  相似文献

This study determined whether cutaneous antibodies were present in the excised skin of channel catfish, Ictalurus punctatus Rafinesque, immune to Ichthyophthirius multifiliis Fouquet (Ich). Theronts were immobilized on or near the excised skin from immune fish. The survival of immobilized theronts was significantly reduced after exposure for 8 h to the culture of excised skin from immune fish. Culture fluids from excised skin of immune fish immobilized theronts with a peak in the immobilization titre at 24 h post-exposure. Immobility of theronts in the culture fluid from immune skin was removed after immunoabsorption with theronts. Indirect immunofluorescent staining of theronts treated with culture fluid from excised skin of immune fish revealed strong and uniform fluorescence on the cilia and cell surface of theronts. Western blot analysis of the culture fluid from immune fish revealed a 70-kDa band which corresponded to the molecular weight of catfish immunoglobulin heavy chain. The results of this study show that cutaneous antibodies to Ich theronts were present in and released from the excised skin from fish immune to Ich. Immobilization and killing of the theronts are two characteristics of the antibody response that appear to prevent the successful invasion of theronts into excised skin.  相似文献

The impairment of fluid, electrolyte and acid-base status which is observed in calves and juvenile cattle as a result of ileus and subileus is not only dependent on form and site of the obstruction, but is also influenced considerably by the duration of the condition and any coexisting disease. This publication reviews the most important blood, urine and ruminal fluid parameters in such patients. The diagnostic significance of the laboratory values under discussion is not so great with obstructions to the caudal part of the digestive tract. However, markedly raised ruminal fluid chloride levels, combined with hypochloraemia and a metabolic acidosis, are indicative of an "abomasoruminal reflux syndrome" resulting from an obstruction in the region of the abomasum or cranial small intestine.  相似文献

Measurement of blood osmolarity of pre-metamorphic turbot larvae demonstrated that hypoosmoregulation is well established in larvae 6 days post-hatch (121 degree-days) and older. Blood osmolarity of 121–420 degree-day larvae reared in 100% seawater was significantly greater than blood osmolarity of larvae reared in 50% seawater. Hypoosmoregulation involved drinking, but instantaneous drinking rates of 340 degree-day larvae reared in 100% seawater were only slightly more than those of similarly aged larvae reared in 50% seawater. Adaptation to environmental salinity involved changes in gut water absorption; 65–74% water absorption occurred in larvae reared in seawater compared to 30–35% water absorption in larvae reared in 50% seawater. Gastrointestinal water absorption occurred prior to the rectum. In seawater this occurred alongside a decrease in gut fluid osmolarity but desalting was apparently less significant than in adult fish. Absolute water absorption by the gut of 340 degree-day larvae reared in seawater was about 2-fold that of larvae held in 50% seawater, while the osmotic gradient between internal body fluids and environmental media differed by 4-fold, which implies changes the in water permeability of skin and/or developing gills.  相似文献

Radial immunodiffusion was performed with body fluids from a variety of marine invertebrates to detect the presence of any vertebrate-type immunoglobulin, viz., IgG, IgA and IgM, or antigenically similar substances. Selenastrum, Chlorella and the Cyanophyta groups, important components of the phytoplankton in the natural habitat of Anodonta cygnea in Mira Lagoon, were used separately as experimental diet for Anodonta cygnea. To assess the effects of the different diets, several organic and inorganic compounds of the haemolymph and extrapallial fluid were measured. Protein content was significantly reduced with experimental Non Feed, and carbohydrate content with Non Feed, Selenastrum, Chlorella , and Cyanophyta diets compared to the control in the lagoon. Lipid contents in body fluids were similar among the different diet groups, with the exception of the Chlorella diet, which showed a higher value. In all diets, the inorganic elements, magnesium and chloride, presented a non-significant variation profile. The concentration of sodium and potassium was significantly affected in Selenastrum and Chlorella diets, observing in the Non Feed diet a minimum value for potassium. The non-significant variation of osmolality suggests the presence of a specific osmotic regulatory mechanism independent of the diet. A particular regulation mechanism seems to exist for both calcium and manganese ion inducing similar profiles for all experimental diets with higher contents in Selenastrum and Cyanophyta diets, respectively. The higher calcium ion concentration appears associated with lower protein-carbohydrate contents, probably due to weak calcium aggregation and deposition mechanism, except for the control and Mixture diets. In general, the concentrations of organic and inorganic compounds in the body fluids of Mixture-fed A. cygnea , were more similar to those found in the control diet group. This suggests that an equal mixture of the three algae, Selenastrum, Chlorella , and Cyanophyta, is more appropriate diet to provide the necessary nutrients for a normal maintenance of A. cygnea in the wild.  相似文献
