The ploidy levels of the twenty-two yam (Dioscorea cayenensis-D. rotundata complex) cultivars within germplasm Cameroon Guinea yam were determined by flow cytometry. Three different ploidy levels (4x,6x, 8x) were detected within the samples analysed. Fifteen cultivars were tetraploids, five were hexaploids, and two were octoploids. The cultivar group EKOTO showed a high level of ploidy variation with tetraploid, hexaploid and octoploid cultivars. The hexaploid nature of cultivars Dobnawo and Bilougnou supported the hypothesis that they are hybrids between cultivars of the EKOTO group and either the KPE or BAKOKAE groups. The Guinea yams, Dioscorea cayenensis Lam. and D. rotundata Poir. (D. cayenensisD. rotundata complex), represent a highly important crop, widely distributed in the humid and semi-humid tropics. The ploidy levels of 170 accessions of the core set of Guinea yams from West African countries was determined using flow cytometry with propidium iodide staining. One hundred and eight of the genotypes were found to be tetraploid, 47 were hexaploid and five were octoploid. One mixoploid individual containing tetraploid and hexaploid nuclei was also detected. A deeper analysis considering each separate taxon revealed that while for D. rotundata the majority of individuals were tetraploid, for D. cayenensis this ploidy level was not detected in any of the accessions. Also, no association between ploidy level and place of cultivation was found for the evaluated germplasm. The obtained data is highly valuable for breeding programs of Guinea yam, especially for the optimization of future hybridization experiments directed to the genetic improvement of this economically important crop.  相似文献

Summary Ploidy levels were examined in 26 accessions of Actinidia chinensis: 20 accessions were diploid (2n=2x=58) and 6 accessions were tetraploid (2n=4x=116). There was no evidence of variation in ploidy level within an accession. Our results are consistent with tetraploid A. chinensis coming from a restricted part of China.Interploid crosses within A. chinensis produced only low numbers of seedlings which were mainly triploid. Crossing hexaploid A. deliciosa with pollen of tetraploid A. chinensis produced a large family of plants and those checked were pentaploid.Counts on 83 genotypes of different ploidy levels (2x, 3x, 4x, and 5x) confirmed that the basic chromosome number in Actinidia is 29.  相似文献

Summary Investigations were conducted on the identification and distribution of yam natural pollinators, flowering pattern and hybridization of white yam (Dioscorea rotundata Poir.) at the International Institute of Tropical Agriculture (IITA), Ibadan, Nigeria, in order to provide informations for producing hybrid seeds needed for vam genetic improvement.Insects belonging to the Coleoptera (48.0%), Diptera (20.4%), Hymenoptera (20.0%), Hemiptera (5.8%), and Thysanoptera (5.8%) were caught around yam plants and presumed to be pollinators: they entered open-receptive-flowers and their presence on yam plants coincided with the duration of the flowering period.Studies on the flowering pattern revealed that: (i) Male and female clones differed in their periods of flower initiation as well as the flowering duration; (ii) Seed production and conventional breeding methods are feasible if staggered planting, water and soil fertility could be managed to ensure that flowering periods of males and females nick and also extend flowering.Artificial pollinations within white yam and between white yam and yellow yam (D. cayenensis Lam) using the camel hair brush method were successful, but resulted in a relatively low fruit set (31.8% maximum). This method was more effective than natural pollination (19.2%) and can thus supplement it for the production of hybrid seeds for introducing genetic variation in yam breeding populations.  相似文献

Greater variability in starch properties is found in lower ploidy wheats than in commercial hexaploid wheats. This paper reports on the starch properties and variability in granule bound starch synthase (GBSS) loci of 17 diploid (Aegilops tauschii) and 12 tetraploid (durums) potential progenitors of wheat, compared with 29 synthetic hexaploid wheats produced from such progenitors. Starch properties examined were granule size distribution, swelling power, amylose content, gelatinisation and amylose-lipid dissociation properties. A PCR screening method was able to detect the presence or absence of each of the three GBSS genes. It also detected polymorphisms in eight diploids and nine hexaploids, all displaying the same 25 bases deletion in the D genome allele of GBSS. Two tetraploids and five hexaploids were null 4A for GBSS. There was little difference in the amylose contents and amylose-lipid dissociation peak temperatures of the synthetic hexaploids and the lower ploidy wheats. The synthetic hexaploids showed intermediate swelling power values with the durums giving the highest swelling powers. The durums also had higher B granule contents than the A. tauschii accessions, but not as high as the synthetics. However, the A. tauschii samples gave the highest gelatinisation peak temperatures. The presence of the null 4A mutation was positively correlated with swelling power, amylose content and DSC measurements. The new smaller D genome allele of GBSS was associated with slightly higher swelling power. These results confirm the value of wheat progenitor lines as sources of new starch properties for hexaploid wheat. Summary To produce hexaploid (or other polyploid) hybrids, diploid or tetraploid Hordeum vulgare was crossed with hexaploid or octoploid H. bulbosum, and perennial triploid hybrids between the two species were treated with colchicine. The crosses did not yield viable plants: seedset was low, the seed aborted and embryo culture was unsuccessful. The colchicine treatments geve rise to plants in which hexaploid chromosome numbers were observed. At the hexaploid level chromosomal instability occurred, resulting in chromosome elimination.The colchicine-treated triploid hybrids showed in the first years after the treatment better fertility after open flowering than untreated plants, but the level of fertility remained very low. The offspring consisted of haploid, diploid and approximately triploid plants like H. vulgare, tetraploid and approximately tetraploid plants like H. bulbosum, and plants with hybrid morphology and unstable chromosome number, which were highly sterile. Thus the crossing barrier between H. vulgare and H. bulbosum could not be broken down at higher ploidy level.  相似文献

Paspalum glaucescens belongs to the informal group Plicatula, reproductively characterized by the dominance of tetraploid apomitic lines in most of its populations, with rare diploid, sexual counterparts. The species shows high phenotypic variation. Twenty nine Southern Brazilian accessions were cytologically and morphologically analysed. Most of the accessions were tetraploid (2n = 4x = 40). Meiotic study of three tetraploids showed their irregular behaviour. Eight accessions presented the diploid level (2n = 2x = 20).This can be considered a very high frequency of diploids in a member of the Plicatula group. While the tetraploids are concentrated at the highest elevations, diploids were detected mostly in the lowlands. Average stomatal sizes were quite distinct on different ploidy levels, being larger in tetraploids. In the polyploid species Dioscorea alata, earlier studies have shown a prevalence of higher ploidy levels (2n = 60 and 80) restricted among female clones. Cytological screening of a number of rare flowering collections maintained at the CTCRI revealed the occurrence of hexaploids (2n = 60) among male clones for the first time. The hexaploid males were sparse flowering, showing irregular meiosis leading to very high pollen sterility. On the basis of the diploid chromosome number 2n = 20 in the Old World Dioscorea, the trivalent forming 2n = 60 male is considered as an autoallohexaploid.  相似文献

Polyploid induction represents a useful tool for breeders of floral crops as larger flowers, longer flowering period and deeper colors can be achieved through chromosome doubling. This study aimed at testing the efficiency of colchicine and oryzalin in inducing polyploidy in three Rhododendroncultivars grown in vitro. The chemicals were used in two concentrations with 24 h and 48 h treatment durations. The survival of the plants was better in colchicine than in oryzalin solutions. The higher concentration of both chemical skilled more plantlets. The treatment duration in oryzalin did not affect the survival, but 48 h in colchicine was more destructive than 24 h. The low survival rate may not be a disadvantage, if the treatment induces desired ploidy. The ploidy levels were screened with flow cytometry. Oryzalin was more efficient than cochicine in inducing polyploidy, the treatment duration and the concentration did not have significant effects as main factors. The biggest proportion of solid tetraploids (18.2% of the survived plants) was obtained from the 24 h treatment in 0.005% oryzalin. Immediately after the treatment the polyploids grew very slowly, whereas most of the unaffected diploids were vigorous from the very beginning. More mixoploids than solid tetraploids were obtained in all treatments. Most of the mixoploids retained their chimerism, one third shifted todiploidy and one single plant to tetraploidy. Summary Flow cytometry and stomata characteristics were used for screening ploidy levels in a large population of in vitro induced autopolyploids of the Musa acuminata breeding clone SH-3362. Culturing shoot tips in liquid medium stipplemented both with 5.0 mM colchicine for 48 hours or 30 M oryzalin (3,5-dinitro-N4,N-dipropylsulphate) for seven days, both in combination with 2% (v/v) DMSO, resulted in a high (23.1% and 29.1%) frequency of non-chimeric tetraploids in the fourth vegetative generation. Although mixoploidy persisted in subsequent cycles of vegetative propagation, tetraploids as identified by flow cytometry remained solid non-chimeric during two more cycles. These autotetraploids were propagated for field testing. A rough pre-selection of regenerated V4 plants based on their stomata characteristics resulted in a population in which only 56.2% of the plants were solid tetraploids. Landraces of tetraploid and hexaploid wheat from the Northern Negev in Israel were evaluated over two years for their grain quality attributes. Twenty-one populations of tetraploid wheat (Triticum durum, represented by 56 accessions) and 8 populations of hexaploid wheat (T. aestivum and T. compactum, represented by 13 accessions) were compared with 3 and 4 modern improved Israeli cultivars of tetraploid and hexaploid reheat, respectively. This comparison allowed to estimate the progress made in the improvement of gram quality m present-day cultivars. Except for grain protein content in the hexaploids, significant variation was revealed among landraces in kernel weight, protein content (tetraploids), sedimentation, mixograph score and carotin content. The best modern cultivars were comparable to the best landraces in kernel weight and carotin content (tetraploids), indicating that modern wheat breeding maximized kernel weight and carotin content, as compared with the tested landraces. Gram protein content and/or quality was not maximized in modern cultivars and its improvement was deemed possible by introgression from the best landraces in this respect.  相似文献

Difficulties in obtaining new breeding lines of Brachiaria (Trin.) Griseb., an important forage grass in Brazil, are mostly related to differences in ploidy among the accessions, and to apomixis, an asexual mode of reproduction. Usually, sexual accessions are diploid while apomicts are polyploid. Induced tetraploids of Brachiaria brizantha (A. Rich.) Stapf have been successfully obtained and this paper presents the results of a study of their reproductive modes and fertility. Despite frequent meiotic aberrations during microspore development, the induced tetraploids produced viable pollen and produced progeny after controlled self-pollination. Similarly to the original diploid sexual progenitor, embryo sacs of the Polygonum type with confirmed meiotic origin were present in the induced tetraploids suggesting chromosome doubling did not alter the reproductive mode. The embryo sac of the Polygonum type was also observed in progenies obtained after self and open pollination. Nevertheless, embryo sacs of the Polygonum and the Panicum types within the same ovule were observed in some progenies obtained after open pollination, probably having resulted from hybridization with tetraploid, apomictic plants. Indeed, the compatibility of the progeny with tetraploid, apomictic B. brizantha was confirmed by the formation of mature caryopses after controlled pollination. Evidence is presented that the induced tetraploids and their progeny are sexual plants and that they are compatible with natural tetraploid B. brizantha. The induced tetraploids will be useful for analyses of apomictic inheritance as well as in the development of sexual tetraploid lines in Brachiaria breeding programs.  相似文献

Summary A number of methods of producing synthetic octoploids from Fragaria species of lower levels of ploidy are described. Six synthetic octoploids were produced from various combinations involving one hexaploid species, two tetraploid species and four diploid species. Three of the synthetic octoploids are male fertile and were used successfully as male parents in crosses with octoploid strawberry cultivars.  相似文献

Summary The first stage in the domestication of the protein-rich tetraploid oats Avena magna and A. murphyi has been achieved by introducing seed non-shattering from the cultivated hexaploid oats A. sativa. A great variation in the protein content was encountered in the tetraploid domesticated types and plants having up to 27 31% protein were selected. The potential of these newly domesticated oats in increasing protein yields is briefly diseussed: also the significance of the tetraploids A magna and A. murphyi in improving the protein content of the hexaploid cultivars is stressed.  相似文献

Summary Bahiagrass (Paspalum notatum Flugge) is a polymorphic species indigenous to South America which has become naturalized in the southeastern U.S. The most common form in the United States is Pensacola bahiagrass,P. notatum var.saurae Parodi., which is a valuable forage. Pensacola is a sexual diploid, while most other bahiagrasses are apomictic tetraploids. Pensacola bahiagrass is considered to have greater heat tolerance (based on anin vitro heat tolerance test) than a number of otherPaspalum species, but has less leaf epicuticular wax (a drought avoidance mechanism) than other species. Both heat tolerance and leaf epicuticular wax are desirable characteristics for species grown where periodic drought occurs. We measured both characteristics over two years in a collection of 23 bahiagrass accessions, many of which had been collected in South America near the center of origin of the species. The collection included various ploidy levels. For both years, no accessions ranked statistically lower in damage in the heat tolerance test than Pensacola, although eight had significantly higher damage. Two entries in addition to Pensacola had very low damage in the heat tolerance test. Pensacola was high in leaf wax, with 16 accessions rated significantly lower in wax. The accession having the lowest wax content was a triploid, which also exhibited considerable leaf death in the field in response to drought and heat. The diploid entries tended to be higher in leaf wax than the other ploidy levels. This study has identified additional bahiagrass germplasm which may be of use in a breeding program.  相似文献

Resistance to stripe rust (caused by Puccinia striiformis Westend.) of 34 Triticum turgidum L. var.durum, 278 T. tauschii, and 267 synthetic hexaploid wheats (T. turgidum x T. tauschii) was evaluated at the seedling stage in the greenhouse and at the adult-plant stage at two field locations. Mexican pathotype 14E14 was used in all studies. Seedling resistance, expressed as low infection type, was present in all three species. One hundred and twenty-eight (46%) accessions of T. tauschii, 8 (23%) of T. turgidum and 31 (12%) of synthetic hexaploid wheats were highly resistant as seedlings. In the field tests, resistance was evaluated by estimating area under disease progress curve (AUDPC). Synthetic hexaploid wheats showed a wide range of variability for disease responses in both greenhouse and field tests, indicating the presence of a number of genes for resistance. In general, genotypes with seedling resistance were also found to be resistant as adult plants. Genotypes, which were susceptible or intermediate as seedlings but resistant as adult plants, were present in both T. turgidum and the synthetic hexaploids. Resistances from either T. turgidum or T. tauschii or both were identified in the synthetic hexaploids in this study. These new sources of resistance could be incorporated into cultivated hexaploid wheats to increase the existing gene pool of resistance to stripe rust.  相似文献

Summary Adventitious shoots were obtained from the diploid watermelon [Citrullus lanatus (Thunb.) Matsum. & Nakai] cultivars Dixielee, Jubilee II, Mickylee, Minilee, and Royal Sweet by culturing excised cotyledons on shoot regeneration medium for six weeks. Tetraploid and diploid regenerants were identified by counting the number of chloroplasts per guard cell pair from leaves of regenerated plants. Cross fertilization of putative tetraploids with diploid pollinators and the production of triploid seed confirmed the efficacy of this approach. The mean number of chloroplasts for tetraploid regenerants was 19.1 whereas diploids averaged 11.2. These values were similar to tetraploid and diploid plants from seed. Ovary diameter, petal, and anther diameter of male flowers, and leaf length by width ratio were also good indicators of plant ploidy. Progeny obtained from self-fertile tetraploids of Mickylee were crossed with various diploid pollinators to produce triploid hybrid seed. All triploid plants from tissue culture-derived tetraploids produced fruit comparable in quality to fruit produced by currently-available triploid hybrids, demonstrating that in vitro tetraploid induction can be used to produce high quality tetraploid plants for use in triploid hybrid seed production.  相似文献

Summary Wheat pentaploids were produced by hybridizing a high kernel weight (1000 grain wt=56 g), high protein (25.4%) line of wild tetraploid wheat (Triticum turgidum dicoccoides) as male parent, with the three hexaploids (T. aestivum) — normal Chinese Spring and its two homoeologous pairing mutants, ph 1b and ph 2. The pentaploids were crossed as female parents to the two commercial hexaploid cultivars Warigal and Barkaee and 42-chromosome stable plants selected from the F1 of the pentaploid x hexaploid crosses.Mean protein content of certain F3 lines from all six pentaploid x hexaploid crosses was significantly higher than Chinese Spring and the respective commercial hexaploid parent (p<0.005) indicating high protein had been transferred from the tetraploid to the hexaploid level.Kernel weight amongst certain F3 lines of the three pentaploids x Barkaee was significantly (p<0.0005) higher than either Chinese Spring or Barkaee, indicating the transfer also of high kernel weight from the tetraploid to the hexaploid level. However kernel weight was not significantly increased over Warigal in any F3 lines of its crosses with the three pentaploids.High levels of homoeologous chromosome pairing in the ph-mutant pentaploids, plus evidence for significant modification of the composition of high-molecular weight (HMW) glutenin subunits of grain protein in certain F3 derivatives of the ph-mutant pentaploid x hexaploid, crosses indicates that the ph-mutant-derived lines may possess novel (intergenome) genetic recombination, at least for high protein, and possibly kernel weight.  相似文献

Summary Pollen grain size in many genera positively correlated with chromosome number. In this study, oat (Avena) pollen grain size was examined in diploids (2×=14, one species, for accessions), tetraploids (4×=28, five species, 20 accessions), hexaploids (6×=42, one species, eight cultivars), and in 10 octoploid (8×=56) accessions. Mature anthers ready to dehisce pollen were sampled from one to six plants per accession, and pollen grains were squeezed out of the anther with tweezers. Oat pollen grains are slightly elliptical, and the length of the major axis was found to be highly correlated with the minor axis width (r=0.94**). Pollen grain length, 39.3 m for diploids, 41.3 m for tetraploids, 47.0 m for hexaploids, and 48.8 m for octoploids, was positively correlated (r=0.86**) with ploidy level. No genomic or species effects appeared to influence this trait. Only tetraploidAvena vaviloviana accession PI 412767 produced two distinct class sizes of pollen grains, 99% normal (43.0 m) and 1% large (52.7 m).  相似文献

Summary Changes in incidence of apomixis were determined at different intervals of the flowering period in a highly sexual diploid cytotype of Paspalum cromyorrhizon Trin., a natural tetraploid cytotype of the same species, and in the triploid and tetraploid intraspecific hybrids that were produced by pollinating the 2x cytotype with pollen of the tetraploid. Reproductive behaviour was determined from observations of embryo sacs in mature ovules. Gametophytic apomixis in this species in characterized by aposporous embryo sacs which differ from normal sacs by their number, orientation inside the ovule, their dimensions and shape, and the absence of antipodal cells.The potential for apomictic reproduction increased in relation to the ploidy level, suggesting a gene dosage effect in the incidence of apomixis. In the three ploidy levels, the highest incidence of apomixis was observed when the plants reached the maximum number of flowering heads. These findings suggest that the same environmental conditions that favour flowering should also be responsible for increasing the incidence of apomixis. An additional experiment under controlled conditions indicated that the light regime should be one of the factors that govern the amount of apomictic reproduction. Thus, polyploidy and apomixis should be of special significance in the dispersion and evolution of this grass species. The 4x facultative apomictic cytotype is far more frequent in nature than the highly sexual 2x. Moreover, most of the seeds produced in a flowering season should arise asexually. So, apomixis rapidly increases the number of plants bearing the successful maternal genotype, and sexual reproduction becomes important in adverse environmental conditions.  相似文献
