Methanol extract of sawdust of sugi (Cryptomeria japonica) heartwood was fractionated with toluene and n-hexane to give solvent-soluble and solvent-insoluble fractions. The n-hexane-soluble fraction showed the most inhibition activity among the fractions against phytopathogenic microorganisms, namely Fusarium oxysporum, Phytophthora capsici, Pythium splendens, and Ralstonia solanacearum. Sandaracopimarinol and ferruginol, isolated from the n-hexane-soluble fraction, showed moderate antifungal activity against the three fungi and strong antibacterial activity against R. solanacearum. The content of sandaracopimarinol (7.07 g/kg based on the dried sawdust) in the heartwood was about twice that of ferruginol. Sandaracopimarinol and ferruginol strongly inhibited the growth of Gram-positive bacteria but did not show inhibitory action against Gram-negative bacteria except for R. solanacearum. The antibacterial effect of sandaracopimarinol was first found in the present study and was stronger than that of ferruginol.  相似文献

Taiwania (Taiwania cryptomerioides Hayata) is a native and plantation species in Taiwan, and its heartwood has demonstrated excellent decay resistance. Brown root rot fungus Phellinus noxius has caused severe damage to more than 100 tree species in Taiwan. The aims of this study were to investigate the antifungal activities of Taiwania heartwood extracts against P. noxius and to identify the antifungal compounds. Antifungal activities were examined using the agar dilution method (a direct contact assay) and the inverted lid method (a vapor contact assay). Active compounds were purified using liquid–liquid partition, open-column chromatography and HPLC. Among four soluble fractions (n-hexane, ethyl acetate, n-butanol and water soluble fractions), results showed that n-hexane soluble fraction (HSF) can significantly inhibit the growth of P. noxius via the agar dilution method. Moreover, active compounds present in HSF were obtained using antifungal assay-guided fractionation. Results showed that ferruginol, T-cadinol, α-cadinol and T-muurolol were found to exhibit excellent antifungal activities against P. noxius, and their IC50 values were 16.9, 25.8, 33.8 and 50.6 μg/mL, respectively. According to the current findings, phytochemicals (ferruginol, T-cadinol, α-cadinol and T-muurolol) of Taiwania heartwood had the potential to be used as environmentally benign antifungal agents against P. noxius in place of synthetic or inorganic fungicides.  相似文献

Calocedrus formosana Florin (Cupressaceae) is an endemic tree to Taiwan. To evaluate the antioxidant activity of the plant extracts from heartwood, bark, and leaf of C. formosana, assays for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and superoxide anion scavenging activities, as well as prevention of DNA strand cleavage were performed in this study. Similar IC50 values against the DPPH radical were found for the heartwood and bark extracts at approximately 23µg/ml. Moreover, the heartwood extract exhibited the highest inhibitory activity against superoxide radicals among the test samples; a 2.3-fold lower value of IC50 for superoxide radical inhibition was found in the heartwood extract relative to that of (+)-catechin. Much less effect on inhibition of DPPH and superoxide radicals was found from the leaf extract of C. formosana. More than 70% of superoxide radicals were inhibited in the presence of 10µg/ml heartwood extract, whereas only 15% inhibition was obtained from the leaf extract. The heartwood extract, at a dose of approximately 0.5mg/ml, apparently completely prevented the X174 supercoiled DNA cleavage induced by ultraviolet photolysis of H2O2, as judged by agarose gel electrophoresis. This report also suggests that the antioxidant activities of the plant extracts of C. formosana are in good correlation with their phenolic contents.  相似文献

研究侧柏心材不同部位及正己烷和二氯甲烷依次提取的心材活性成分对白腐菌、褐腐菌、黑曲霉菌的抑菌作用,并采用GC/MS联用技术对提取物中的活性成分进行鉴定和分析.结果表明:侧柏心材从髓心向边材根据年轮所划分的3个部位对3种真菌都具有较好的抑菌效果,并且靠近边材的2部分抑菌效果更明显些.气质联机技术对正己烷和二氯甲烷提取物分析鉴定表明:侧柏心材正己烷提取物中有14种化合物,大部分化合物属于萜烯类物质及其衍生物,最高含量化合物为罗汉柏烯(25.564%)和雪松醇(19.743%);侧柏心材二氯甲烷提取物中鉴定出24种化合物,最高含量化合物也为罗汉柏烯(15.429%).侧柏心材正己烷提取物对褐腐菌、白腐菌和黑曲霉菌第14 d时的抑菌圈直径分别为10.0,9.3和8.3 mm,大于二氯甲烷提取物抑菌圈直径(0,2.7和1.3mm),表明正己烷提取物对3种真菌的抑制生长作用明显优于二氯甲烷提取物的抑菌作用.  相似文献

In order to find new utilization method of woody wastes, we examined the bioactivities of extracts from branch heartwood of Chamaecyparis obtusa (hinoki) and compared to extracts from trunk heartwood. The bioactivities examined were antifungal activities against four fungi (Trametes versicolor, Fomitopsis palustris, Trichoderma virens, Rhizopus oryzae), and bioassay with brine shrimp (Artemia salina) which shows any allelopathic activities not measurable with fungi. Antifungal activities were observed in the hexane and ethyl acetate extracts of branch and trunk heartwood. The hexane and ethyl acetate extracts of branch and hexane extract of trunk showed strong lethality against brine shrimp. The yields of the active extracts of branch were much more than that of trunk. The identified compounds in the active extracts of branch were germacra-1-(10),5-dien-4??-ol, t-cadinol, t-muurolol, hinokiresinol, and hinokinin. Hinokiresinol and t-muurolol showed strong antifungal activities. Hinokiresinol showed bioactivities against T. virens, R. oryzae and brine shrimp. Germacra-1-(10),5-dien-4??-ol was lethal to brine shrimp. Germacra-1-(10),5-dien-4??-ol and hinokiresinol were minor components in trunk heartwood, but major components in branch. These qualitative and quantitative results suggest that the branch heartwood could be a valuable chemical resource because it contains large amounts of antifungal and allelopathic compounds.  相似文献

The aim of this work was to isolate a lectin from Myracrodruon urundeuva heartwood and to evaluate its antimicrobial activity against bacteria and fungi that attack plants, including woods. The lectin was isolated from heartwood through affinity chromatography on a chitin column monitored by hemagglutination assay. The lectin inhibited Gram-negative and Gram-positive bacteria and was more effective than antifungal Cercobin in growth inhibition of phytopathogenic fungi. The detected antimicrobial activity reveals the possible role of the lectin in the resistance of M. urundeuva heartwood against deteriorative biological agents. The M. urundeuva lectin is the first bioactive peptide found in heartwood, probably stored as a chemical protection against biodegradation.  相似文献

以长白落叶松心材、边材及树皮为原料,分别用乙醚、氯仿、丙酮、甲醇和水5种溶剂对其进行提取,采用滤纸片法研究了不同溶剂提取物分别对白腐菌和褐腐菌的抑菌性能,发现心材的甲醇提取物和丙酮提取物对白腐菌有较好的抑制效果,树皮的水提取物对2种菌都有较好的抑制效果.并研究了这两种提取物在室内对木材的防腐性能,借助扫描电子显微镜(SEM)照片观察了木材腐朽后菌丝生长的情况,与常用木材防腐剂酸性铬酸铜(ACC)进行了对比,结果显示:心材的甲醇提取物对白腐菌的抑制效果较好;而树皮的水提取物对褐腐菌抑制效果较好.  相似文献

The effect of selective removal of extractives on termite or decay resistance was assessed with matched samples of Thuja plicata Donn ex D. Don and Chamaecyparis nootkatensis (D.Don) Spach heartwood. Samples were extracted using a variety of solvents and then exposed to the subterranean termite Coptotermes formosanus Shiraki in a no-choice feeding test or to the brown-rot fungus Postia placenta (Fr.) M. Larsen & Lombard in a soil bottle test. At the same time, the effect of naturally occurring variations in heartwood extractives on termite or decay resistance was evaluated by testing samples from the inner and outer heartwood of five trees of each species against C. formosanus and P. placenta and analyzing matched wood samples for their extractive content. The results suggest that the methanol-soluble extractives in T. plicata and C. nootkatensis play an important role in heartwood resistance to attack by C. formosanus and P. placenta. Total methanol-soluble extractive content of the heartwood was positively correlated with both termite and decay resistance; however, there was much unexplained variation and levels of individual extractive components were only weakly correlated with one another. Further studies are under way to develop a better understanding of the relationships between individual extractive levels and performance.  相似文献

 Sugi green logs with red or black heartwood were smoke-heated, and the changes in the color of the heartwood after ultraviolet (UV) (λ = 365 nm) radiation exposure were then observed. After UV radiation exposure, the redness and yellowness increased in both the red and black heartwoods, whereas the brightness decreased. In the black heartwood, the resulting color turned from yellowish white to reddish brown. Reddening in black heartwood after exposure to a combination of smoke heating and UV radiation is thought to be due to a decrease in brightness and an increase in both redness and yellowness. However, the degree of change in heartwood color by UV radiation exposure was not greatly affected by smoke-heating treatments of various lengths. When methanol extracts were fractionated and exposed to UV radiation, the yellowness increased in the n-hexane-soluble portion and the redness increased in the acetone-soluble fractions from the n-hexane-insoluble portion. These results suggest that the n-hexane-soluble fraction contains the substances that allow heartwood color to change to yellow after UV radiation exposure, and the acetone-soluble-fraction from the n-hexane-insoluble portion contains the substances that allow it to change to red. Robinia pseudoacacia L. heartwood is characterized by a very high natural durability. However, a significant difference was observed between the mature and juvenile heartwood, the latter presenting less durability against fungi decay, which could be attributed to lower extractive content. In order to elucidate this idea, extractives from mature and juvenile heartwoods of black locust trees were investigated. Results showed that extractive and phenolic contents were higher in mature than in juvenile heartwoods. The identification of phenolic compounds by UPLC–DAD–MS/MS revealed, for the first time, the presence of resveratrol and piceatannol. These two stilbenes as well as the flavonoid dihydrorobinetin were present at the highest level in mature heartwood, and as they are known antifungals, they could account for the great durability of mature heartwood. The stilbenes were detected in significant amounts particularly in mature heartwood where piceatannol reached a level tenfold higher than that reported for Japanese knotweed roots, the primary natural source of these stilbenes, whereas resveratrol level was comparable with reported values. As resveratrol and piceatannol receive increasing demand for nutraceutical, cosmetic and, possibly, pharmaceutical purposes, due to their beneficial health effects, this study underlines the use of R. pseudoacacia as a promising sustainable and economical source of resveratrol and piceatannol.  相似文献

    Two novel biphenyl dimers, caesappanin A (1) and B (2), were isolated from the ethanol extract of the heartwood of Caesalpinia sappan. Their structures were elucidated on the basis of spectroscopic data including IR, HRESIMS, 1D and 2D NMR. Preliminary bioassays showed that compound 1 possessed potent cytotoxic effects against 4 cell lines including HCT-8, BGC-823, A549 and A2780 with IC50 between 1.67 μM and 4.88 μM.  相似文献

    The variation in extractives content in sapwood and heartwood was investigated among 12 trees in each of four commercial plantations of Eucalyptus globulus in central Portugal. The study was carried out at the 15% height level and extractions used successively dichloromethane, ethanol and water. At all sites, heartwood had significantly more extractives than sapwood, on average 3.8 and 2.4%, respectively. Most extractives consisted of ethanol soluble material (on average 52% of total extractives). Among the sites, there was a statistically significant difference in the content of extractives but the most important source of variation was the within-tree variation between sapwood and heartwood. Differences in the content of extractives were also observed among trees. A strong relation between extractives content and heartwood proportion was found. The potential loss of pulp yield and problems associated with accumulation of extractives are directly related to the heartwood proportion in the eucalypt stems. Forest management should take into account heartwood development and selection for minimising heartwood extractives.  相似文献

    The aim of this study is to characterise the properties of juvenile and mature heartwood of black locust (Robinia pseudoacacia L.). Content, composition and the subcellular distribution of heartwood extractives were studied in 14 old-growth trees from forest sites in Germany and Hungary as well as in 16 younger trees of four clone types. Heartwood extractives (methanol and acetone extraction) were analysed by HPLC-chromatography. UV microspectrophotometry was used to topochemically localise the extractives in the cell walls. The natural durability of the juvenile and mature heartwood was analysed according to the European standard EN 350-1. Growth as well as chemical analyses showed that, based on extractives content, the formation of juvenile wood in black locust is restricted to the first 10–20 years of cambial growth. In mature heartwood, high contents of phenolic compounds and flavonoids were present, localised in high concentrations in the cell walls and cell lumen of axial parenchyma and vessels. In juvenile wood, the content of these extractives is significantly lower. Juvenile wood had a correspondingly lower resistance to decay by Coniophora puteana (brown rot fungus) and Coriolus versicolor (white rot fungus) than mature heartwood.  相似文献

    The essential oils of eighteen Egyptian plants were extracted by hydrodistillation and their chemical compositions were analyzed by GC-MS. The antifungal activity of the isolated oils was evaluated against two wood decay fungi Hexagonia apiaria and Ganoderma lucidum in vitro. The essential oil of Artemisia monosperma showed the highest inhibitory effect against H. apiaria (EC50 = 31 mg L?1) and G. lucidum (EC50 = 53 mg L?1). The results of in vitro tests indicated that the essential oils of Cupressus sempervirens, Citrus limon, Thuja occidentalis, Schinus molle, A. monosperma and Pelargonium graveolens were the most potent inhibitors against both fungi. These six oils caused significant reduction of wood mass loss of Scots pine sapwood after 6 weeks of fungal exposure. The oil of C. limon revealed the highest reduction of wood mass loss caused by H. apiaria, while A. monosperma oil displayed the highest reduction of wood loss caused by G. lucidum. These results support the potential use of essential oils for wood protection against decay fungi.  相似文献

    Acacia confusa is an indigenous plantation species in Taiwan, and its heartwood has excellent decay resistance capability. The aim of this study is to investigate the resistance mechanisms of heartwood extracts from A. confusa against Laetiporus sulphureus (brown-rot). Results from wood block decay resistance tests indicate that aqueous crude extract and its three soluble fractions can significantly decrease the weight loss of wood blocks caused by L. sulphureus. However, none of the extracts can inhibit the growth of L. sulphureus on agar plate test. The performance of decay resistance from three soluble fractions is consistent with their antioxidant activities (DPPH free radical scavenging activity, reducing power and ferrous ion chelating ability). 3,4-Dihydroxybenzoic acid (3,4-DHBA), the compound present in three soluble fractions, exhibited excellent antioxidant activities and was proven to have excellent decay resistance ability against L. sulphureus. These findings suggest that 3,4-DHBA may be responsible for the inhibition of hydroxyl radical formation from Fenton reaction caused by L. sulphureus. Thus, 3,4-DHBA has good potential to be used as a novel and environmentally benign wood preservative.  相似文献

    Infection of heartwood by decay fungi (heartrot) is a concern for growers of Acacia mangium for solid‐wood products as the incidence can be high in some regions of Indonesia. Variation of heartrot incidence for different provenances of A. mangium was determined using two field trials in Sumatra, Indonesia. In a Riau Province trial of 21 provenances, the effect of provenance was statistically significant for natural heartrot incidence, which ranged from 1.6% to 27.2%. In a smaller trial using artificial inoculation in South Sumatra, heartwood infection incidence ranged from 39.4% to 70.8% across six provenances and both wound type and provenance were statistically significant factors. There was also significant variation in sapwood infection length related to provenance. Wood extractives (yield, total phenols, protein‐precipitable tannin and 2,3‐trans‐3,4′,7,8‐tetrahydroxyflavanone) were quantified from a subsample of trees for each trial. However, no significant differences in extractive concentration were detectable according to provenance and evidence for a relationship between heartwood extractives and heartrot incidence was generally poor. While further studies need to be completed to establish the basis for heartrot incidence, results from these trials allow for recommendations on provenance selection to reduce heartrot incidence and provide information for further genetic selection programmes.  相似文献

    Ethanolic extract from the fruits of Pimpinella anisoides, an aromatic plant and a spice, exhibited activity against AChE and BChE, with IC50 values of 227.5 and 362.1 μg/ml, respectively. The most abundant constituents of the extract were trans-anethole, (+)-limonene and (+)-sabinene. trans-Anethole exhibited the highest activity against AChE and BChE with IC50 values of 134.7 and 209.6 μg/ml, respectively. The bicyclic monoterpene (+)-sabinene exhibited a promising activity against AChE (IC50 of 176.5 μg/ml) and BChE (IC50 of 218.6 μg/ml).  相似文献

    Phenolic extractives in the trunk of Toxicodendron vernicifluum (syn. Rhus verniciflua) were investigated. Seventeen compounds, gallic acid, protocatechuic acid, (?)-fisetinidol-4β-ol, (?)-fisetinidol-4α-ol, 2-benzyl-2,6,3′,4′-tetrahydroxycoumaran-3-one, (?)-fustin, 1,2,3,6-Tetra-O-galloyl-β-d-glucose, (?)-epifustin, (+)-taxifolin, 1,2,3,4,6-penta-O-galloyl-β-d-glucose, (?)-garbanzol, (?)-fustin-3-O-gallate, (?)-epifustin-3-O-gallate, fisetin, sulfuretin, quercetin and butein, were identified from the heartwood extractives. It was found that only (+)-taxifolin which had 5,7-dihydroxy A-ring possessed a 3R configuration although other flavonoids which had 7-hydroxy A-ring possessed a 3S configuration. Quantitative analysis revealed that the total phenolic contents were much higher in the heartwood (5–7 wt%) than in the sapwood and bark (1–2 wt%), and (?)-fustin was the most abundant extractive in the heartwood (1.4–2.4 wt%). For the radial distribution of phenolic extractives, it was generally found that their content was lowest in the sapwood, increased to the highest in the outer heartwood, and then decreased in the inner heartwood.  相似文献
