SUMMARY A field population of Trichostrongylus colubriformis was divided into 4 lines for exposure to selection in the laboratory. The first line was selected with 50 mg/kg thiabendazole, the second with 4 mg/kg morantel tartrate, the third with 50 mg/kg thiabendazole followed by 4 mg/kg morantel tartrate and the fourth line was not selected for drug resistance. Following at least 9 generations of selection there was no difference in LD50 or LD95 between the unselected and single selected strains of worms. The strain selected by50both thiabendazole and morantel tartrate had a significantly higher LD50 against thiabendazole, morantel tartrate and levamisole than did the other three strains. The single selected strains had LD95's of 172, 21.5 and 2.3 mg/kg for thiabendazole, morantel tartrate and levamisole respectively, compared with corresponding values of 111, 17.3 and 2.4 in the unselected strain and 124, 15.5 and 3.0 in the double selected strain. The estimated efficiency of the recommended dose of each anthelmintic against the unselected field strain was:—thiabendazole (44 mg/kg) 50% efficient, morantel tartrate (8.8 mg/kg) 76% efficient and levamisole (7.0 mg/kg) 99.9% efficient.  相似文献

The paper describes a method for detecting levamisole and morantel tartrate resistance by determining the percentage of paralysed third stage larvae in serial dilutions of anthelmintic. Levamisole resistantOstertagia spp. had a smaller proportion of larvae undergoing tonic paralysis in either levamisole or morantel tartrate than did a non-resistant strain. Field strains of Trichostrongylus colubriformis and Ostertagia circumcincta, designated PF4 and PF5 respectively, were recovered from a farm on which the sole use of levamisole over a preceding 12 year period led to the development of anthelmintic resistance. The results of field observations and preliminary critical trials in both Merino and crossbred sheep showed that both species have varying degrees of resistance to three major anthelmintics; levamisole, morantel tartrate and thiabendazole. Mean worm count reductions for adult T colubriformis (PF4) for therapeutic doses of morantel tartrate, thiabendazole and levamisole in crossbreds were 45.7 per cent, 97.3 per cent and zero respectively, and for Merinos 80.7 per cent, 88.3 per cent and 92.0 per cent respectively. Against O circumcincta the corresponding reductions for crossbreds were 51.4 percent, 95.4 per cent and 20.3 per cent and for Merinos, 52.5 per cent, 73.1 per cent and 29.8 per cent. There was no statistically significant difference in the responses of both parasite species to either levamisole or morantel. This result suggests that resistance to the two chemically unrelated drugs may be co-inherited.  相似文献

Multiple resistance to benzimidazoles (fenbendazole, albendazole and mebendazole) in a strain of Haemonchus contortus in sheep was detected on a farm where fenbendazole resistance had already been identified. Following a faecal egg count reduction test, this was confirmed by both critical and controlled anthelmintic tests. Different groups of sheep infected naturally or given an experimental infection with the fenbendazole-resistant strain were treated with the recommended doses of various anthelmintics. Compared to the control group, percentage reductions in faecal egg counts of sheep treated with fenbendazole, albendazole, mebendazole, levamisole and morantel varied between 56% and 81% and worm counts between 71% and 86%. The results indicate the presence of multiple anthelmintic resistance in this strain of H. contortus on this farm. Sheep treated with ivermectin and closantel showed 100% reductions in faecal egg and worm counts, suggesting high efficacy of these drugs against the population of H. contortus on this farm.  相似文献

Levamisole, morantel, fenbendazole, or ivermectin was administered at 2-week intervals from May 1 through Sept 14, 1985, to breeding ewes (20 ewes/drug) infected with various gastrointestinal nematodes. All ewes had fewer gastrointestinal nematode eggs per gram of feces (epg) after 2 treatments, compared with pretreatment epg counts. Ewes administered ivermectin continued to have a low mean epg (0 to 3) throughout the study. The mean epg counts of ewes treated with levamisole increased from 3 to 483 during the study. This increase was similar to that of ewes treated with morantel (7 to 485 epg). The mean epg count of fenbendazole-treated ewes increased from 4 to 192 during the study. By the end of the study, the mean epg counts when expressed as a percentage of the pretreatment epg counts were 4% (ivermectin), 249% (fenbendazole), 627% (levamisole), and 630% (morantel). With the exception of the ivermectin-treated ewes, the epg count increased almost linearly in the ewes after the 2nd anthelmintic treatment. These data indicate that the gastrointestinal nematodes (including Haemonchus contortus) may have developed more resistance to levamisole and morantel than to fenbendazole. On the basis of the epg counts, resistance to ivermectin did not develop during the 4.5-month treatment period. The percentage of ewes shedding eggs after 2, 4, and 6 anthelmintic treatments was lowest for ewes treated with ivermectin (20%) and was similar (40%) for ewes treated with 1 of the other 3 anthelmintics. The occurrence of anthelmintic resistance in Trochostrongylus colubriformis and resistance to morantel are reported for the first time in New Zealand. In two controlled anthelmintic trials with experimentally infected sheep, morantel citrate at either 8.8 mg/kg or 10 mg/kg had no effect on worm burden. An efficacy of >99% was demonstrated for mebendazole at 12.5 mg/kg against this strain.  相似文献

SUMMARY Following the apparent failure of levamisole to control infections of Haemonchus contortus in sheep at Lawes in south eastern Queensland, a strain of this parasite was isolated at the Animal Research Institute, Yeerongpilly. This strain was used to infect sheep at Yeerongpilly and the Merrindale Research Station, Victoria where four experiments to classify the resistance pattern of the parasite were carried out. Resistance to thiabendazole was first suspected in 1969, and these experiments confirmed that resistance to this drug was still present. They also showed that a strong degree of resistance had been developed to both levamisole and morantel tartrate. Other benzimidazole anthelmintics and also the organophosphorus compound naphthalophos were only moderately effective against the original isolate but rafoxanide, nitroxynil and phenothiazine were almost 100% effective. Other highly effective chemicals were disophenol and closantel. After passaging the strain for four generations with both levamisole and albendazole, resistance to both naphthalophos and the newer benzimidazole anthelmintics increased dramatically. This is the first report of a field strain of H. contortus exhibiting resistance to benzimidazole, non-benzimidazole and organophosphorus anthelmintics.  相似文献

A strain of Haemonchus contortus from the Pietermaritzburg district of Natal was found to be resistant to levamisole (geometric mean efficacy 76.5%), morantel (41.9%), the benzimidazoles (oxfendazole: 33.7%) and rafoxanide (82.0%), but apparently fully susceptible to closantel and disophenol. In the case of ivermectin, a mean of 5.2% of the H. contortus was not removed at a dosage of 200 micrograms kg-1 live mass. A second strain of H. contortus, from Amsterdam in the south-eastern Transvaal, showed reduced susceptibility to levamisole (80.8%) and morantel (46.2%), the only 2 drugs tested. This is apparently the first report of resistance to the levamisole/morantel group of anthelmintics in sheep in South Africa.  相似文献

Two field trials, one with suckling Merino ewe lambs and the other with yearling Dohne Merino rams, are described. In these the anthelmintic efficacy of febantel (a benzimidazole), ivermectin, levamisole and morantel are compared, using the 1st stage larval reduction test. The mean natural log (+1 for zero values) of the post treatment larval counts of the treated groups was compared with that of the untreated controls and the percentage reduction used to assess anthelmintic efficacy. Febantel was only 87.4% effective against Teladorsagia in suckling lambs but the other anthelmintics were more than 99% effective against this genus. Efficacy against Haemonchus and Trichostrongylus ranged from 93.2%-100% for all 4 compounds. In the rams all compounds were 100% effective against Trichostrongylus, with the exception of morantel which was only 87.5% effective. None of the compounds were effective against Teladorsagia, particularly morantel, animals treated with which having more larvae than the controls. The interpretation of anthelmintic efficacy; the advantages of the first stage larval reduction test, compared with the faecal egg count reduction test; and the importance of incubating cultures at 30 degrees C for 24 h, in order to harvest first stage larvae, are discussed.  相似文献

Two strains of Oesophagostomum spp., consisting of both O. quadrispinulatum and O. dentatum, were subjected to a controlled in vivo assay for resistance to levamisole and pyrantel by comparison with susceptible isolates. One strain (LEM) was recently isolated from a commercial herd, where sows showed high numbers of strongyle eggs in faeces within 2 weeks of farrowing and following treatment with levamisole at the manufacturer's recommended dose rate 1 week before farrowing. Levamisole had been used as the sole anthelmintic for treatment for at least 7 years on this farm. Treatment with pyrantel in this herd also indicated cross-resistance to this drug. A mixed population of O. quadrispinulatum and O. dentatum of this strain was subjected to controlled in vivo assays. Faecal egg count reduction (FECR) was found to be -573.3% (P greater than 0.05) and worm count reductions (WCR) of O. quadrispinulatum and O. dentatum were estimated as 44.5% (P greater than 0.05) and 96.4% (P less than 0.001), respectively. Treatment with pyrantel showed that cross-resistance existed to this drug, with FECR of 10.4% (P greater than 0.05) and WCR of 64.5% (P greater than 0.05) and 90.7% (P less than 0.05) for O. quadrispinulatum and O. dentatum, respectively. Another strain (VJ) was isolated from another commercial pig herd, which was dosed with pyrantel citrate four times a year for at least 8 years. This strain showed resistance to pyrantel, with FECR of 43.8% (P greater than 0.05) and WCR of 65.9% (P greater than 0.05) and 49.4% (P greater than 0.05) for O. quadrispinulatum and O. dentatum, respectively. However, both species were susceptible to levamisole. Our results suggested that selection with levamisole gave rise to levamisole resistance and automatically conferred resistance to pyrantel, whereas selection with pyrantel only resulted in resistance to this drug alone. These findings are discussed in relation to the location of the two species of Oesophagostomum in the large intestine of pigs and the mode of action of this class of anthelmintics.  相似文献

The in vitro activities of thiabendazole, levamisole, pyrantel, morantel and ivermectin against Oesophagostomum spp., the nodular worm of pigs, were determined and compared. The study was carried out using isolates of O. dentatum and O. quadrispinulatum, which had been defined in vivo. Infective larvae were exposed to the anthelmintics for 24 h and then placed in a micromotility meter. All the treatments significantly reduced the motility of the ensheathed L3 larvae, but the micromotility meter was not able to differentiate between anthelmintic resistant and anthelmintic susceptible isolates.  相似文献

Following the failure of anthelmintic treatment to control an outbreak of trichostrongylosis in sheep, multiple resistance to levamisole and oxfendazole was confirmed in field strains of Trichostrongylus colubriformis at the CSIRO Pastoral Research Laboratory, Armidale. Resistance in Trichostrongylus spp to levamisole was also confirmed on an adjoining farm. From the results of an experiment where lambs were treated at the recommended dose rate with one or both anthelmintics, it was estimated that 32% of T. colubriformis were resistant to levamisole, 19% to oxfendazole, and 12% to both drugs. Simultaneous administration of levamisole and oxfendazole resulted in an additive anthelmintic effect. Naphthalophos (36.6 to 51.2 mg/kg) was 93% efficient against the multiple resistant strain. The similar histories of anthelmintic usage at the CSIRO Pastoral Research Laboratory and on the northern tablelands of New South Wales generally, suggest that multiple anthelmintic resistance in T. colubriformis may soon emerge as a problem on the northern tablelands.  相似文献

A strain of Trichostrongylus colubriformis from Nottingham Road, in Natal, was found to be solidly resistant to levamisole and morantel at the recommended dose levels. Untreated control sheep in fact harboured fewer worms at slaughter than either of the 2 treated groups. In contrast, the benzimidazoles and ivermectin were more than 99.9% effective against this worm strain. The possible implications of escalating resistance to anthelmintics in the gastrointestinal nematodes of sheep are discussed.  相似文献

A survey was undertaken to assess the prevalence of anthelmintic resistance in sheep nematode populations on 40 commercial farms distributed throughout the central tablelands of New South Wales. Representatives of the 2 major groups of broad spectrum anthelmintics with different modes of action (thiabendazole and levamisole) were used at the manufacturer's recommended dose rates. Efficacy was assessed on the basis of the reduction in faecal strongyle egg counts 7 days after treatment. An efficacy of less than 90% using both anthelmintics was obtained on 4 farms. Thiabendazole had an efficacy of less than 90% on a further 21 farms and levamisole had an efficacy of less than 90% on an additional 4 farms. There was no evidence of anthelmintic resistance on 8 farms, while the remaining 3 had insignificant parasite burdens. Based on larval cultures from faeces, Ostertagia and Trichostrongylus were the most significant species in resistant populations. Haemonchus burdens were sporadic and levels of resistance relatively low. Nematodirus burdens were widespread but no evidence of resistance was detected.  相似文献

In order to study the reliability of the larval paralysis test as an in vitro assay for the detection of resistance of nematodes to levamisole and morantel tartrate, the influence of different parameters was evaluated using resistant and susceptible Ostertagia ostertagi strains. The operator, the sample (10% of the larvae present in the suspension), the incubation time (24, 48 or 72 h), the incubation temperature (20 or 25 degrees C) and the observation period of the larvae (5 or 15 s) had no statistically significant influence on the test results. Statistical differences were obtained only when L3 larvae of different ages (1 or 2 months) were used. Reversibility of the paralysis did not occur when concentrations of levamisole of less than or equal to 200 micrograms ml-1 or morantel tartrate of less than 2000 micrograms ml-1 were used. The reproducibility of the test was fairly good, with a mean standard deviation of 21.3% for the LC50 values. Morantel resistance in a strain of O. ostertagi was not confirmed as such by the larval paralysis test. The resistance factor was less than or equal to 1, in spite of an efficacy of morantel of 77.4% as shown in vivo.  相似文献

A population of Ostertagia circumcincta from sheep has been found which, in field and laboratory trials, showed a lack of response to the recommended dose (44 mg/kg) of thiabendazole, suggesting that a degree of resistance exists to this anthelmintic. Preliminary experiments indicated a less than expected response to levamisole. This was not confirmed in a laboratory trial and therefore resistance to this anthelmintic has not been shown conclusively.  相似文献

Changes in anthelmintic resistance in nematode parasites were monitored in sheep grazing on 2 separate farms, but with the same anthelmintic treatment program, over 16 years. High levels of benzimidazole resistance emerged in Ostertagia and Trichostrongylus spp populations on both farms following 9 years of continuous use of this class of drug. Subsequently, variations in the levels of resistance occurred for the same species between farms and between species on the same farm. A change to levamisole for 2 years resulted in a significant reversion towards benzimidazole susceptibility, but a concomitant rise in levamisole resistance, in Ostertagia on one farm. However, benzimidazole resistance increased rapidly following the re-introduction of oxfendazole into the anthelmintic treatment program. Results from both farms illustrate the pitfalls of using one anthelmintic class for an extended period and provide indirect support for the alternation of anthelmintic classes at approximately yearly intervals.  相似文献
