The effect of two arbuscular mycorrhizal (AM) fungi, Glomus fasciculatum and G. macrocarpum on shoot and root dry weights and nutrient content of Cassia siamea in a semi-arid wasteland soil was evaluated. Under nursery conditions, mycorrhizal inoculation improved growth of seedlings. Root and shoot dry weights were higher in mycorrhizal than non-mycorrhizal plants. The concentration of P, K, Cu, Zn and Na was significantly higher in AM inoculated seedlings than in non-inoculated seedlings. Mycorrhization led to decrease in alkalinity of the rhizosphere soil from pH 8.5 to 7.4. Under nursery conditions, the degree of mycorrhizal dependency increased with age of C. siamea seedling. On transplantation to the field, the survival rate of mycorrhizal seedlings (75%–90%) was higher than that of non-mycorrhizal seedlings (40%). Besides better survival rate, AM inoculation improved the growth performance of seedlings in terms of height and stem diameter. Among the two AM fungi used, the efficiency of Glomus macrocarpum was higher than that of G. fasciculatum under both nursery and field conditions.This revised version was published online in May 2005 with corrected page numbers.  相似文献

Some Acacia mangium Willd. plantations in Asia grow poorly due to low soil fertility and the absence of compatible mycorrhizal fungi. This legume tree can be colonized by arbuscular mycorrhizal (AM) and ectomycorrhizal (ECM) fungi but inoculation is not routinely practiced. This study aimed to compare the effectiveness of AM fungi and four isolates of the ECM fungus Pisolithus tinctorius (Pers.) Coker and Couch (Pt) in promoting growth of A. mangium seedlings under glasshouse conditions. AM inoculants were: Glomus etunicatum, G. fasciculatum, G. macrocarpum and Gigaspora margarita and mixed species extracted from rhizosphere soil of a Populus stand in Suwon (AMKFRI), and Carex (AMM6) and Populus (AMM7) growing in mine tailings in Korea. Pisolithus isolates were from Philippines (PtPhil) and Korea (PtKFRI, PtMKACC, PtKACC). Generally, ECM fungi promoted height and diameter growth of A. mangium more than the AM inoculants. The Korean Pisolithus increased plant dry weight by 122–145%, mixed AM inoculants by 61–97%, and Glomus and Gigaspora by 45–72% over the control. PtKACC gave the highest root colonization and promoted the highest growth and concentration of most nutrients. Mycorrhizal root colonization was positively correlated with plant dry weight, Na, Fe and Cu concentrations and N, P, K, Ca, Na, Fe and Cu contents. In conclusion, the results provide strong evidence for benefits of mycorrhizal inoculation on A. mangium seedlings under glasshouse conditions. The Korean Pisolithus isolates (particularly PtKACC), and two AM fungi (AMKFRI and AMM6) are potential mycorrhizal inoculants but their effectiveness and persistence should be determined on degraded lands in tropical countries where A. mangium is being planted for rehabilitation.  相似文献

Inoculation with edible fungi bestows an added value on mycorrhized plants since production of mushrooms as a secondary crop can be an economically-valuable resource in forestry management. In order to establish a protocol for controlled mycorrhization of Pinus halepensis with the edible fungus Lactarius deliciosus, several factors such as fertilisation (35, 60 or 120 mg total N and 13.5, 27 or 54 mg total P), potting substrate (sphagnum peat or sphagnum peat/vermiculite) and fungal inoculum (mycelial slurry produced in a bioreactor, alginate beads or peat/vermiculite) were assessed. The most effective inoculum was mycelial slurry at a dose of 10 ml/plant. The two potting substrates assessed were compatible with mycorrhiza formation. The greatest number of mycorrhized seedlings was obtained with a moderate level of N (35 mg/plant) or P fertilisation (27 mg/plant). Inoculation did not produce a consistent growth effect on P. halepensis, but seedlings met the quality requirements, making them suitable for planting out.  相似文献

Ophiostoma species were isolated from bark beetles and Abies mariesii, A. veitchii and A. homolepis attacked by the beetles in Nikko, Tochigi, central Honshu, Japan. One to two Ophiostoma species were frequently isolated from each species of bark beetle. Ophiostoma subalpinum was the most common associate of Cryphalus montanus. Ophiostoma sp. B as well as O. subalpinum was a common fungus associated with Polygraphus proximus. Ophiostoma europhioides was isolated from Dryocoetes hectographus and D. autographus as one of the common associates. Ophiostoma sp. J and Ophiostoma sp. S were frequently isolated from D. autographus and D. striatus, respectively. These fungi seem to have specific relationships with particular bark beetles. Ophiostoma sp. B, Ophiostoma sp. J and Ophiostoma sp. S have unique morphological characteristics and appear to be new species. Five trees of A. veitchii, approximately 43 years old, were inoculated with five Ophiostoma species to assess the relative virulence of the fungi. Ophiostoma subalpinum, Ophiostoma sp. B, and O. europhioides had relatively higher virulence than the other species studied.  相似文献

We investigated the role of tetrapartite associations between an arbuscular mycorrhizal (AM) fungus (Glomus geosporum), phosphate solubilizing bacteria (Paenibacillus polymyxa), Frankia and Casuarina equisetifolia on growth, nutrient acquisition, nutrient utilization and seedling quality of C. equisetifolia. Seedlings of C. equisetifolia were grown in an Alfisol soil and inoculated with G. geosporum, P. polymyxa and Frankia either individually or in combinations. Inoculation of bioinoculants stimulated seedling growth, the efficiency of nutrient uptake and improved seedling quality. However, microbial inoculation generally reduced the efficiency of nutrient utilization in dry matter production (nutrient use efficiency). Inoculation of P. polymyxa or Frankia increased the extent of AM colonization, which resulted in the accumulation of the nutrients. Seedlings inoculated with Frankia and G. geosporum had more, and heavier nodules compared to seedlings inoculated with Frankia alone. Dual inoculation of microbes was more effective than individual inoculations. The growth response of seedlings to inoculation involving all the microbes was greater than the response to either individual or dual inoculations. The results of this study showed that the tetrapartite association could improve the growth, nutrient acquisition and seedling quality of C. equisetifolia under tropical nursery conditions.  相似文献

We conducted a study to find out if arbuscular mycorrhizal (AM) fungi (Acaulospora scrobiculata, Scutellospora calospora) and phosphate solubilizing bacteria (PSB, Paenibacillus polymyxa) inoculation either individually or in combinations can improve Acacia auriculiformis seedling growth, uptake of nutrients and quality in a phosphorus deficient tropical Alfisol. The seedlings were assessed for various growth and nutrient uptake parameters after 60 days of treatment. Inoculation with P. polymyxa stimulated mycorrhizal formation. Seedling height, stem girth, taproot length, number of leaves and leaf area, plant dry matter production, nodulation, and nodular dry weight were significantly higher for seedlings that were either dual inoculated or triple inoculated compared to individual inoculation of AM fungi or PSB, and uninoculated seedlings. Dual and triple application of AM fungi and PSB also significantly improved the nutrient contents of shoots and roots and nutrient uptake efficiencies. The calculated seedling quality indexes of the AM fungi and PSB inoculated seedling were 25–208% higher than uninoculated seedlings. These findings show that A. auriculiformis seedlings when dual inoculated or triple inoculated performed better than seedlings inoculated with the microbes individually and compared with uninoculated control seedlings. We conclude that bioinoculation is important for the production of high-quality A. auriculiformis seedlings in tree nurseries for planting in nutrient deficient soils.  相似文献

Three VA-mycorrhizal fungi; Glomus occultum, Glomus aggregatum (local isolates) and G. mosseae (strain from Bangalore, India) were inoculated to assess their effect on growth of Acacia mangium in lateritic soil. All inoculations enhanced growth with respect to shoot height, root diameter, leaf area, chlorophyll content and biomass of A. mangium significantly compared to uninoculated control seedlings. G. occultum proved most efficient among the three. The mycorrhizal dependency factor indicated that the growth of A. mangium was 57% dependent on G. occultum, 47% on G. mosseae and 46% on Glomus aggregatum.  相似文献

Effects of mycorrhizal association on improving tolerance of host plant under stress environments have received attentions in recent years. In this paper, six isolates of AMF (arbuscular mycorrhizal fungus) were inoculated to Casuarina equisetifolia seedlings under glasshouse conditions to investigate the effects of AMF on growth and drought tolerance of host plants. All the six isolates which belong to Glomus showed high mycorrhizal colonization (88.5–96.0%) with C. equisetifolia seedlings. Seedlings were subjected to drought stress without watering for 7 days and survival of the seedlings inoculated with Glomus caledonium Gc90068, G. versiforme Gv9004 and G. caledonium Gc90036 increased by 36.6, 23.3 and 16.6%, respectively compared with uninoculated seedlings. Limited influence of AMF on seedling height growth was found, but the effects of AMF on total biomass increment were very significant; the increment ranged from 25.7 to 118.9% compared with uninoculated treatment, and it was noted that AMF exerted more influences on root biomass than shoot biomass. Based on the changes in physiological and biochemical parameters among different treatments caused by drought stress, it was concluded that AM (arbuscular mycorrhizal) associations improve the drought tolerance of C. equisetifolia seedlings by means of some physiological and biochemical responses, such as lowering permeability of plasma membrane and MDA (malondialdehyde) contents, enhancing concentrations of P nutrition, soluble sugar, soluble protein and activities of POX (Peroxidase) of C. equisetifolia seedlings.  相似文献

This study evaluated oxalic acid accumulation and bioremediation of chromated copper arsenate (CCA)-treated wood by three brown-rot fungi Fomitopsis palustris, Coniophora puteana, and Laetiporus sulphureus. The fungi were first cultivated in a fermentation broth to accumulate oxalic acid. Bioremediation of CCA-treated wood was then carried out by leaching of heavy metals with oxalic acid over a 10-day fermentation period. Higher amounts of oxalic acid were produced by F. palustris and L. sulphureus compared with C. puteana. After 10-day fermentation, oxalic acid accumulation reached 4.2 g/l and 3.2 g/l for these fungi, respectively. Fomitopsis palustris and L. sulphureus exposed to CCA-treated sawdust for 10 days showed a decrease in arsenic of 100% and 85%, respectively; however, C. puteana remediation removed only 18% arsenic from CCA-treated sawdust. Likewise, chromium removal in F. palustris and L. sulphureus remediation processes was higher than those for C. puteana. This was attributed to low oxalic acid accumulation. These results suggest that F. palustris and L. sulphureus remediation processes can remove inorganic metal compounds via oxalic acid production by increasing the acidity of the substrate and increasing the solubility of the metals.An erratum to this article can be found at  相似文献

Trees of the family Dipterocarpaceae are the dominant trees in Southeast Asian tropical forests where they play an important ecological role and are also important commercially. An experiment was conducted to determine the effect of ectomycorrhizal fungi on the growth of dipterocarp species in peat soils. Seedlings of Shorea pinanga were inoculated with spores of two ectomycorrhizal fungi, Pisolithus arhizus and Scleroderma sp. were grown in pots containing sterilized peat soil for 7 months. The percentage of ectomycorrhizal colonization on S. pinanga exceeded 86%. Colonization of S. pinanga roots by ectomycorrhizal fungi resulted in increased shoot height, stem diameter, number of leaves, and shoot fresh and dry weight. Survival rates of S. pinanga were greater for inoculated seedlings than control seedlings. These results suggest that inoculation of ectomycorrhizal fungi can improve the early growth of S. pinanga grown in tropical forests and that this technique will accelerate the rehabilitation of degraded dipterocarp forests.  相似文献

孟加拉东南部土壤中的砷含量很高,不仅威胁人的健康,而且对土壤也有破坏作用。云南石梓(Gmelina arborea)在孟加拉是个快速生长的树种,也是含砷土壤中的潜力树种。研究评价了含砷试验土中丛枝菌根真菌对云南石梓(Gmelina arborea)生长的影响。播种前,四种不同浓度的砷(10mg·kg-1、25mg·kg-1、50mg·kg-1和100mg·kg-1)被加入到试验土中。记录生长参数,如,植物的根、苗鲜重、干重、冠幅径、根长和苗高、根瘤菌和孢子菌群落。菌根植株较非菌根植株生长好。与其它含砷量高的土壤中植株的生长情况相比,在含砷量为10-mg·kg-1的土壤中,菌根植株和菌根生长效果最佳,菌根植株生物量最高。随着砷浓度的增加,种苗生长,根瘤菌和孢子菌群落均明显降低p0.05)。与非菌根植株比较,菌根植株高生长增加了40%,生物量增加了2.4倍。研究表明,根瘤菌接种可以减少有害土壤中的云南石梓(Gmelina arborea)的初生长的影响。  相似文献

Present study evaluated growth of Populus deltoides G48 and Toona ciliata over a period of 6 months, in nursery soil amended with 10% fly ash (v/v), 5% distillery waste (v/v), 20% farmyard manure (v/v) and microbial consortium of Pseudomonas striata and Azotobacter sp. @ 30 ml/pot in different combinations leading to 12 different treatments with 16 replicates in completely randomized block design. Biometric parameters such as plant height, collar diameter and total dry biomass were analyzed which indicated that the treatment (T8) comprising of fly ash @ 10% (v/v), farmyard manure @ 20% (v/v) and microbial consortium @ 30 ml/pot promoted growth of P. deltoides. The results indicated that combined addition of fly ash, farm yard manure and microbial inoculants can be used as a good potting mixture for improving survival rates and plant growth in forestry nurseries.  相似文献

A 1207 bp cDNA fragment (PsG6PDH) was amplified by RT-PCR from cold-induced total RNA of the freezing-tolerant P. Suaveolens, using primers based on the highly conserved region of published plant glucose-6-phosphate dehydrogenase (G6PDH) genes. The sequence analysis showed that PsG6PDH coding region had 1 101 bp and encoded 367 predicted amino acid residues. Moreover, the nucleotide sequence of PsG6PDH showed 83%, 82%, 79%, 79% and 78% identity, and the derived amino acid sequence shared 44.2%, 44.7%, 42.0%, 40.5% and 43.9% identity with those of the Solanum tuberosum, Nicotiana tabacum, Triticum aestivum, Oryza sativa and Arabidopsis thaliana, respectively. The results show that PsG6PDH is a new member of G6PDH gene family and belongs to the cytosolic G6PDH gene. This is the first report on cloning of the G6PDH gene from woody plants.  相似文献

Pinus densiflora f.umbraculifera, commonly known as utsukushimatsu, is a distinctively shaped form of Japanese red pine whose growth is restricted to a forest stand in Shiga Prefecture, Japan. The inheritance mode of morphological characteristics of utsukushimatsu was studied to preserve the genetic resource of this pine. As previously reported, F1 trees grown from open-pollinated seeds harvested from trees inhabiting the native stand showed two phenotypes: one resembling utsukushimatsu, which produces multiple trunks, and the other resembling normalP. densiflora, which produces one or a few trunks. In the present study, controlled pollination was carried out using F1 and normalP. densiflora trees. Segregation ratios of the two phenotypes observed in the F2 population showed that the morphological characteristics of utsukushimatsu are inherited recessively. This suggests that the mutation of one gene or a few closely linked genes controls the morphological characteristics of utsukushimatsu. Since multiple trunk formation of utsukushimatsu might be related to a loss of lateral bud inhibition, it follows that a simple gene mutation breaks apical dominance inP. densiflora.  相似文献

This paper investigates the distribution of arbuscular mycorrhizal fungi (AMF) spores and AMF colonization in a field study in southeastern Brazil. Response to AMF and rhizobial inoculation was studied in monocultures of Plathymenia reticulata and mixed plantations with both Tabebuia heptaphylla and Eucalyptus camaldulensis in a sandy soil during two consecutive years. P. reticulata height and diameter and mycorrhizal colonization and AMF diversity were measured in dry and rainy periods. The inoculated treatment of E. camaldulensis, T. heptaphylla and P. reticulata mixed plants showed higher height and diameter growth of P. reticulata used as well as increased root colonization and AMF spore numbers. Spore populations were found to belong to five genera: Acaulospora, Entrophospora, Glomus, Gigaspora and Scutellospora, with Glomus dominating. Agroforestry practices including use of leguminous tree P. reticulata effectively maintained AMF spore numbers in soils and high AMF colonization levels compared with monocultures, proving an efficient system for productivity and sustainability.  相似文献

Spatial pattern of recruitment is an important factor influencing population dynamics of plant communities. The spatial pattern is determined by seed dispersal and by the spatial variability of germination and initial survival. In the process of forest expansion following farmland abandonment, mid- and late-successional species are often dispersed in pioneer forests by birds. This could result in an aggregated spatial pattern of seeds that could influence the dynamics of these species in mixed pioneer forests. In the sub-Mediterranean area, mid- and late-successional species such as Quercus pubescens (downy oak) and Fagus sylvatica (European beech) are expected to replace pioneer Pinus species. Using a point sampling method we demonstrated that beech and oak seedlings (height <2 m) have a clumped distribution in the understorey of pine. This could result from an aggregated dispersal by jays (dispersal effect) or from preferential recruitment in particular habitats (habitat effect). To test these hypotheses we proposed a statistical analysis of spatial patterns of regeneration of beech and oak. Ground cover variables (i.e. cover by rock outcrops, herbs, box shrubs, mosses or pine) did not differ significantly around seedlings as compared with random sample plots. Likewise, clumped seedlings had growth similar to isolated seedlings, thus refuting the hypothesis of preferential location in the most favourable microsites. Aggregated dispersal seems to be involved in the process of regeneration. Since beech and oak seedlings have contrasting ecological demands, we discuss the implication of this pattern for the replacement dynamics of pine by these species.  相似文献

Mycorrhizal and non-mycorrhizal holm oak (Quercus ilex L.) seedlings inoculated with black truffle (Tuber melanosporum) were grown under nursery conditions and subjected to drought hardening for 4 months in autumn and winter followed by irrigation for 10 days. Leaf water potential and stomatal conductance were monitored during the 4 months of drought. When the test was completed (March), measurements were made for each treatment (inoculated or non-inoculated), and watering regime (watered and water-stressed). Pressure–volume curves, osmotic potential at full turgor, osmotic potential at zero turgor and the tissue modulus of elasticity near full turgor were calculated. Mycorrhizal colonization and growth, and the content of the main mineral nutrients N, P, K, Ca and Mg were measured. Water stress affected plant growth, caused an elastic adjustment of the plant tissues, and decreased the P and K content, and inoculation improved the nitrogen content. Drought acclimation apparently achieved the goal of improving the drought tolerance of holm oak seedlings, without depressing ectomycorrhizal root colonization by T. melanosporum. Currently, massive dieback of Fraxinus excelsior is observed in countries of eastern, northern and central Europe, and the reasons for it are unclear. The aims of the present work were (a) to study fungal communities in declining F. excelsior crowns; (b) to clarify role of fungi in the decline. Shoots from symptomatic crowns were collected in four localities in central Sweden, and distributed into the following categories: (a) visually healthy; (b) initial necroses; (c) advanced necroses; (c) dead tops. The most frequently isolated fungi were Gibberella avenacea, Alternaria alternata, Epicoccum nigrum, Botryosphaeria stevensii, Valsa sp., Lewia sp., Aureobasidium pullulans and Phomopsis sp., and these taxa were consistently found in shoots of all four symptomatic categories. Forty-eight taxa of other fungi were isolated, and fungal diversity was not exhausted by the sampling effort. The same taxa of fungi were dominant in F. excelsior shoots of different symptomatic categories, and moderate to high similarity of fungal communities was observed in shoots despite the symptoms. Forty-four isolates from 24 fungal taxa were used for artificial inoculations of 277 1-year-old F. excelsior seedlings in bare root nursery. After 2 years, only four fungi caused symptomatic necroses of bark and cambium: A. alternata, E. nigrum, Chalara fraxinea and Phomopsis sp. The most pathogenic was C. fraxinea, inducing symptoms on 50% of inoculated trees, while three other fungi caused necroses on 3–17% of inoculated trees. Infection biology of C. fraxinea and environmental factors determining susceptibility of F. excelsior to decline deserve future investigations.  相似文献

Regeneration of stands of valuable tropical hardwood tree species for sustainable harvest requires production of seedlings with high probabilities of survival. One way to enhance the vigor of plants for outplanting is pre-colonization of roots by arbuscular mycorrhizal (AM) fungi. We pursued the strategy that the most promising AM fungus candidates for inoculation would be those associated with the tree of interest in the field. AM fungus communities were assessed in five plantations of Tectona grandis Linn.f. A total of 18 AM fungal morphotypes were found, representing four families: Glomeraceae (49.6%), Acaulosporaceae (24.9%), Claroideoglomeraceae (20.8%), and Gigasporaceae (4.8%). AM fungus spore density was negatively correlated with soil organic carbon. Some of these AM fungi, plus Rhizophagus irregularis, were established in pot culture and in vitro with transformed carrot roots, and subsequently used to inoculate micropropagated plantlets of T. grandis. Tectona grandis plantlets inoculated in vitro were successfully colonized by all AM fungi studied. Plants inoculated with Funneliformis mosseae were taller than uninoculated plants. Tectona grandis plantlets inoculated with the AM fungus Claroideoglomus etunicatum PBT03 were taller than uninoculated controls in ex vitro experiments. This study provides early insight for the targeted use of the AM symbiosis in production of important tree species in future greenhouse studies and reforestation.  相似文献

The problem of salinized soils has become one of the most serious constraints to agricultural and forest productivity. With the purpose of enhancing salt stress tolerance of Populus tomentosa, we transformed this tree species with spermidine synthase (SPDS) genes derived from an apple by an Agrobacterium-mediated method. Four transgenic clones were confu'med by PCR and Southern blot analysis. As well, the expression of introduced SPDS genes was analyzed by real-time quantitative PCR.  相似文献
