Antiserum was raised against pooled mycelial suspensions from five isolates (designated Pf 1, Pf 2, Pf 3, Pf 10 and Pf 11) representing five physiologic races of Phytophthora fragariae. In enzyme-linked immunosorbent assay (ELISA), this antiserum detected homologous soluble antigens at protein concentrations as low as 2 ng/ml.
Fungal antigens could also be detected in extracts of strawberry plants infected with P. fragariae. Root extracts prepared from the alpine strawberry Fragaria vesca and F. ananassa cv. Cambridge Favourite infected with any of the five isolates studied produced strong reactions in ELISA. In F. vesca , ELISA-positive material was detectable 6-8 days after inoculation before macroscopic symptoms appeared. The cultivar Red Gauntlet, which is resistant to Pf 1, 2 and 3 but susceptible to Pf 10 and 11, reflected this differential response in ELISA; the absorbance produced by extracts of plants infected with virulent isolates was significantly higher than that obtained with the corresponding extracts of plants inoculated with a virulent isolates. The recently introduced cultivars Hapil, Ostara and Providence were found to be susceptible to all isolates in this study: the corresponding root extracts were also positive in ELISA.
The antiserum also detected P cactorum infections. Nevertheless, the ELISA test described should prove valuable in screening certified strawberry stocks.  相似文献

The strawberry red core disease caused by Phytophthora fragariae var. fragariae has until recently not been reported from Norway. During 1995 and 1996, samples from 323 Norwegian strawberry fields were tested in a root-tip bait test for possible detection of red core. Symptoms of the disease developed in test plants cultivated in growth medium containing root tips from 21 fields at eight locations in four different counties. Disease symptoms included wilting of plants, root rot, red steles in the roots, and oogonia and oospores identical in size and appearance to those of P.f. fragariae developing along the root steles. In V8 juice agar, the mycelium was slow-growing, thick, fluffy and aerial. Sporangia were large, mainly inversely pyriform, and without papillae. Optimum temperature for mycelial growth was 21°C, and the fungus ceased to grow at 25°C. In infested fields, the most severe attacks were found in low, often poorly drained areas. The means by which red core may have been introduced and spread in the country are discussed, together with preventive action to avoid further dissemination.  相似文献

Phytophthora cactorum strains isolated from necrotic stem lesions on Betula pendula seedlings or from Fragaria ananassa plants suffering from crown rot were pathogenic to their host plants. Only isolates from birch caused clear lesions on non-wounded bark of birch. P. cactorum isolates from birch were not detrimental to strawberry. Random Amplified Polymorphic DNA (RAPD) analysis revealed variation within P. cactorum, isolates from silver birch having different banding patterns than those from strawberry. UPGMA analysis clustered isolates from silver birch and strawberry plants into separate groups. The data show that the recent outbreak in Finland of P. cactorum in birch could not be caused by the import of strawberry plants affected by crown rot.  相似文献

 1993~1995年,对侵染新疆主要农作物的疫霉菌进行了较全面的调查研究,共从9个地区的28种作物上采集表现根腐、根颈腐、果腐症状(包括病土)的病样1531个,结果从其中17种作物上分离到452个疫霉菌株。根据形态特征、生理生化特性、致病性和菌体可溶性蛋白电泳测定,鉴定为7个种:辣椒疫霉(Phytophthora capsici Leon.),恶疫霉[P.cactorum(Leb.et Cohn) Schroeter],掘氏疫霉(P.drechsleri Tucker),菸草疫霉(P.nicotianae van Breda de Haan),苎麻疫霉(P.boehm eriaeSaw.),柑桔褐腐疫霉[P.citrophthora (Sm.et Sm.) Leonian],隐地疫霉(P.cryptogea Pethyb.etLaff.)。这些疫菌是造成新疆茄果类、瓜类、棉花、苹果、梨、枸杞、草霉、红花、白术等幼苗及成株大量死亡及烂果的主要病原,在农业生产中具有十分重要的意义。  相似文献

Phytophthora fragariae Hickman, which causes strawberry red stele and raspberry root rot, is a quarantine organism for which specific and sensitive detection methods are required to test the health of planting material. Sequences of the internal transcribed spacer regions of the ribosomal gene repeat (rDNA) were used to develop primers for P. fragariae in a nested Polymerase Chain Reaction (PCR). The fungus was readily detected in infected but symptomless roots by nested, but not single-round, PCR. It was also detected in infested water samples obtained from the Dutch General Inspection Service by nested PCR. Detection of PCR products was at least 10-fold more sensitive by PCR-ELISA than by conventional visualisation on agarose gels.  相似文献

Antiserum (anti-PfM) raised against mycelial suspensions of Phytophthora fragariae isolates reacted strongly with antigens from several Phytophthora species. Some cross-reactions with antigens from Pythium species were decreased by fractionating on an affinity column of Sepharose 4B bound to extracts of Fragaria vesca roots infected with P. fragariae. The affinity-purified anti-PfM retained its high cross-reactivity with the various Phytophthora species tested. It also detected infection of raspberry and strawberry roots by some Phytophthora species. This antiserum could, therefore, prove useful as a broad-spectrum Phytophthora-detecting antiserum.
Anti-PfM could not be made specific for P. fragariae because it was raised against components shown to be antigenically similar in all Phytophthora species tested. However, immunoblotting with the affinity-purified anti-PfM produced distinct patterns for P. fragariae, P. erythroseptica and P. cactorum: three serotypes were identified for the latter species. This antiserum might therefore prove useful in classifying Phytophthora species.  相似文献

A bioassay was used to monitor the release of inoculum in drainage water from strawberry plants inoculated with zoospores of Phytophthora fragariae var. fragariae. The fungus was detected in drainage water from plants that had been held at temperatures between 2 and 20 C. but not from plants held at 26°C. The lag phase before secondary inoculum was first released, the maximum and total amounts of inoculum released, and the length of time over which inoculum was released were all greater at the lower temperature regimes, especially those below 10 C. The results were consistent with observations on the effect of temperature on zoospore production from agar discs and on zoospore motility: more zoospores were produced at lower temperatures and they remained motile for longer. From this it is concluded that the inoculum detected consists mainly of motile zoospores. In most experiments with standardized suspensions c. 10-15 were sufficient to initiate infection of the plants in the bioassay. In general, more inoculum was produced by host genotype/fungal isolate combinations in which there were marked root rot symptoms than in combinations in which the host was resistant.  相似文献

A survey for the possible occurrence of red-core disease ( Phytophthora fragariae var. fragariae ) was carried out on 250 strawberry production sites in Finland. The fields were inspected visually. A total of 1080 samples of strawberry roots were taken in spring and autumn 1995 and examined visually, microscopically and by isolation in the laboratory. Root-tip bait-plant tests were performed in the glasshouse to look for the latent presence of the fungus. No red-core disease was detected in any of the inspected fields or in the examined samples.  相似文献

ABSTRACT Marginal chlorosis has affected strawberry production in France for about 15 years. A phloem-restricted uncultured bacterium, "Candidatus Phlomobacter fragariae," is associated with the disease. A large-scale survey for marginal chlorosis in French strawberry production fields and nurseries by polymerase chain reaction amplification of "Ca. P. fragariae" 16S rDNA revealed that symptoms of marginal chlorosis were not always induced by "Ca. P. fragariae" and that the stolbur phytoplasma could induce identical symptoms. "Ca. P. fragariae" was found to be predominant in strawberry production fields, whereas the stolbur phytoplasma was predominantly detected in nurseries. Two transmission periods of the disease, one in spring and the other from late summer to early fall, were evident. Cixius wagneri planthoppers captured on infected strawberry plants were demonstrated to be efficient vectors of "Ca. P. fragariae." The involvement in natural disease spread of the whitefly Trialeurodes vaporariorum, previously shown to acquire and multiply "Ca. P. fragariae" under greenhouse conditions, remains uncertain.  相似文献

Validated protocols for DNA purification and PCR amplification are reported for detection of Phytophthora cactorum in diseased strawberry plants. To remove PCR inhibitors, necrotic strawberry tissues were soaked in 5% alconox solution for >12 h before DNA extraction, and the extracted genomic DNA was embedded in an agarose gel chamber and subjected to electrophoresis. The purified DNA was amplified reliably by PCR. Nested PCR was used to detect a portion of the rRNA gene of P. cactorum in samples. In the first round of PCR, primers ITS1 and ITS4 amplified fragments of varying sizes from total genomic DNA from diseased strawberry plants. In the second round of PCR, a 1:25 dilution of the first-round PCR products was used as template with two P. cactorum- specific primer pairs (BPhycacL87FRG and BPhycacR87RRG, which amplified a 340-bp fragment and a 480-bp fragment from the rRNA gene; and BPhycacL89FRG and BPhycacR176RRG, which amplified a 431-bp fragment). Validation tests using culture-based isolations as a standard for comparison indicated that the DNA purification and PCR primers and amplification protocols were reliable and specifically amplified a portion of the rRNA gene of P. cactorum from necrotic root, crown and petiole tissues of strawberry naturally infected by the pathogen.  相似文献

The pathogenicity of five species of Phytophthora to English walnut was studied in a greenhouse experiment. Phytophthora cinnamomi was the most aggressive species, causing severe root rot and seedling mortality. The other species tested, P. cambivora , P. citricola , P. cactorum and P. cryptogea , did not induce visible crown symptoms on seedlings 2 months after inoculation. Some strains of P. cambivora and P. cactorum also caused taproot damage to seedlings. All except one of the tested isolates caused significant necrosis of fine roots and a significant reduction of root weight compared with noninoculated seedlings. Reduction of above-ground plant development was not statistically significant. While P. cinnamomi is well known as an aggressive primary pathogen of English walnut, the other species of Phytophthora may act as predisposing factors to walnut decline, affecting root system development and increasing host vulnerability to environmental stress.  相似文献

Gnomonia fragariae has been occasionally listed among the fungi associated with diseased strawberry plants. However its pathogenicity has not been established. During the investigation on strawberry decline in Latvia and Sweden, a fungus was repeatedly recovered from discoloured root and crown tissues of severely stunted plants. Attempts to induce sporulation of the isolates grown on several agar media resulted in the formation of mature ascomata only on potato carrot agar and oatmeal agar. On morphological grounds and comparisons with reference herbarium specimens these isolates were identified as Gnomonia fragariae. The pathogenicity of the fungus was evaluated initially in the detached leaf assay and subsequently in three bioassays on strawberry plants. All the bioassays showed that G. fragariae was pathogenic on strawberry and capable of causing severe root rot and petiole blight. The symptoms that developed in the greenhouse experiments closely resembled those observed in the fields. The fungus did not cause rapid plant death but growth and development of inoculated strawberry plants was severely affected. To our knowledge this is the first time when pathogenicity of G. fragariae as a root rot pathogen has been clearly established. Our study shows that G. fragariae is one of the serious pathogens involved in the root rot complex of strawberry in Latvia and Sweden.  相似文献

In four neighbouring regions of southern Italy, Basilicata, Campania, Apulia and Calabria, pepper and zucchini plants showing Phytophthora blight symptoms, tomato plants with either late blight or buckeye rot symptoms, plants of strawberry showing crown rot symptoms and declining clementine trees with root and fruit rot were examined for Phytophthora infections by means of polymerase chain reaction (PCR) assays, using primers directed to nuclear ribosomal DNA (rDNA) repeat sequences. All diseased plants and trees examined tested positive. The detected fungal-like organisms were differentiated and characterized on the basis of primer specificity as well as through extensive restriction fragment length polymorphism (RFLP) and sequence analysis of PCR-amplified rDNA. Phytophthora capsici was identified in diseased pepper and zucchini plants, P. infestans was identified in tomato with late blight symptoms whereas buckeye rot-affected tomatoes and diseased strawberry plants proved to be infected by P. nicotianae and P. cactorum, respectively. Declining clementine trees were infected with P. citrophthora and P. nicotianae in about the same proportion. Also, thirty-one pure culture-maintained isolates of Phytophthora which had previously been identified in southern Italy by traditional methods but were never examined molecularly, were examined by RFLP and sequence analysis of PCR-amplified nuclear rDNA. Among these, an isolate from gerbera which had previously been identified by traditional methods only at genus level, was assigned to P. tentaculata. For the remaining pure culture-maintained isolates examined, the molecular identification data obtained corresponded with those delineated by traditional methods. Most of the diseases examined were already known to occur in southern Italy but the pathogens were molecularly detected and fully characterized at nuclear rDNA repeat level only from other geographic areas, very often outside Italy. A new disease to southern Italy was the Phytophthora blight of zucchini. This is also the first report on the presence and molecular identification of P. tentaculata from Italy.  相似文献

Phytophthora syringae, P. drechsleri, P. cactarutn, P. cambiuora and P. megasperma were isolated from the roots of red raspberry plants affected by severe root and crown rot with associated cane death. Phytophthora megasperma occurred most frequently and consisted of two types of isolates which differed in colony morphology, growth rates, and oogonial, oospore, sporangial and zoospore size, and pathogenicity to a range of plants. One type with large oospores was typical of P. megasperma var. megasperma , and was non-pathogenic to red raspberries, while the other with smaller oospores and which grew more slowly in culture than the first, was highly pathogenic, producing symptoms similar to those observed in the field. Highly pathogenic isolates from Germany and the USA were of this type.
All red and black raspberry cultivars tested were susceptible to the pathogenic type, although North American cultivars were generally less affected than British ones. Inoculated plants had reduced shoot and root weights, stem lesions and wilted and yellowed leaves. The blackberry × raspberry hybrid Tayberry and its blackberry parent were immune.
Phytophthora drechsleri, P. cactorum and P. cambivora produced small to moderate amounts of root rot on red raspberry, and P. cambivora also caused slight symptoms on shoots.  相似文献

A system for scoring the virulence of isolates of Phytophthora fragariae based on a scale of root rot from 0 ( no symptoms ) to 5 (76-100% roots roiled) on a series of strawberry cultivars is described. Thirty-two single-zoospore isolates from one field site were compared by subjecting their root rot scores to cluster analysis and this grouped them into two major clusters equivalent to physiologic races B66–3 and B66-11, Different sub-clusters of isolates of race B66-11 produced different degrees of rotting on the same hosts. Apart from differences in virulence between the sub-clusters there was some evidence for differences in aggressiveness between isolates within sub-clusters.
Increasing inoculum concentration by over 300-fold increased rotting by c . 25% but did not alter the rankings of different isolate/host combinations. Repeated passage of isolates through cultivars of differing susceptibilities did not affect their pathogenicity.  相似文献

Xanthomonas fragariae , the causal agent of angular leaf spot on strawberry, is a quarantine organism in strawberry propagation material in the European Union. For the reliable screening of planting material for latent infections, a real-time PCR assay based on Taqman® chemistry for the detection of X. fragariae was developed. Primers and probe sequences were based on a DNA fragment amplified by a previously reported X. fragariae -specific technique. The sequence of this genomic fragment had no significant similarity with any published GenBank sequence. Specificity of the designed assay was tested with an extended range of X. fragariae collection strains and isolates, with other Xanthomonas spp. and with unidentified bacterial isolates from strawberry plants. A nested PCR, which until now was the reference method for sensitive detection in planta , cross-reacted with the reference strain of Xanthomonas campestris pv. campestris . In combination with an elaborated DNA extraction procedure, the Taqman® PCR enabled reliable detection down to 300 colony forming units in a 100 mg strawberry leaf sample. The assay offers a new tool for epidemiological research and for sanitary control of plant material with low level or latent infections of X. fragariae .  相似文献

The mycelial growth of Phytophthora cactorum on corn meal agar was completely inhibited by metalaxyl or mancozeb at 50 or 100 μg ml?1, but not by fosetyl-aluminium at the same concentrations. Soil drenches with metalaxyl, followed by metalaxyl+mancozeb, around the base of naturally infected trees under orchard conditions, prevented the growth of P. cactorum in the infected bark of the tree. Soil drenchs with metalaxyl, followed by metalaxyl+mancozeb, also prevented the infection of apple trees with P. cactorum under orchard conditions. The trunk circumference and spur length were not significantly affected by any of the fungicide treatments in all the trials over 2 years, except in one of the four orchards used in the study. Soil drench with metalaxyl alone, or followed by metalaxyl+mancozeb, may provide an effective control method for crown rot of apple trees.  相似文献

A PCR-based 'molecular tool box', based on a region of the ras-related protein gene Ypt 1, was developed for the identification of 15 Phytophthora species that damage forests and trees: P. cactorum , P. cambivora , P. cinnamomi , P. citricola , P. europaea , P. inundata , P. lateralis , P. megasperma , P. nemorosa , P. kernoviae , P. pseudosyringae , P. psychrophila , P. quercina , P. ramorum and P. ilicis . Most primers proved highly specific in blast analyses and in tests with DNA from 72 isolates of 35 species of Phytophthora and nine species representative of Pythium . Exceptions were primers designed for P. cactorum and P. ilicis , which cross-reacted with P. idaei and P. nemorosa , respectively. Amplification with Phytophthora -genus-specific primers before amplification with the various species-specific primers (nested PCR) increased the sensitivity of detection over amplification with species-specific primers only: detection limits ranged between 100 and 10 pg target DNA µ L−1 in the latter, compared with 100 fg µ L−1 in nested PCR. Using existing methods for rapid extraction and purification of DNA, single-round amplification was appropriate for detection of target Phytophthora species in leaves, but nested PCR was required for soil and water samples. The quarantine pathogens P. ramorum and P. kernoviae were detected in a number of naturally infected leaves collected in England and Wales, whereas P. citricola was commonest in water and soil samples from natural Scottish ecosystems.  相似文献
