Forty-eight Duroc-cross gilts (40 kg initial BW) were fed a control or a linseed diet containing 60 g of whole crushed linseed/kg. Both diets were supplemented with 150 mg of vitamin E/kg. Eight pigs from each dietary treatment were slaughtered at 20, 60, or 100 d after the start of the experiment. There was no effect (P > 0.05) of diet on growth, carcass characteristics, or foreloin tissue composition. Feeding the linseed diet increased (P < 0.05) the content of n-3 PUFA in plasma, muscle, and adipose tissue, but docosahexaenoic acid was not (P > 0.05) altered by diet. The proportions of n-3 PUFA were highest (P < 0.01) in pigs fed the linseed-diet for 60 d, regardless of tissue (plasma, muscle, or adipose tissue) or lipid (neutral lipids and phospholipids) class. The linseed diet produced a PUFA:saturated fatty acid ratio > or = 0.4 in all groups and tissues, which is close to the recommended value for the entire diet of humans, as well as a robust decrease in the n-6:n-3 ratio. The decrease (P < 0.01) in the percentage of oleic acid in adipose tissue of pigs fed the linseed diet for 60 d could be attributed to a 40% decrease (P < 0.001) in stearoyl-CoA-desaturase activity. Diet did not (P > 0.05) affect the activities of acetyl-CoA-carboxylase, malic enzyme, or glucose-6-phosphate-dehydrogenase in any tissues. Muscle vitamin E content was decreased (P < 0.001) 30% in pigs fed crushed linseed for 60 d, whereas lower (P < 0.001) concentrations of skatole in pork fat were observed in linseed-fed pigs at all slaughter times. Inclusion of linseed (flaxseed) in swine diets is a valid method of improving the nutritional value of pork without deleteriously affecting organoleptic characteristics, oxidation, or color stability.

The Western diet is characterized by a high intake of SFA relative to PUFA, and the consumption of n-3 PUFA is decreased relative to n-6 PUFA. Therefore, there has been much interest in recent years in ways to manipulate the fatty acid composition of meat. The objective of this work was to determine the effects of dietary extruded linseed, an n-3 PUFA source, on growth performance, carcass composition, meat quality, and oxidative stability of pigs slaughtered at 111.0 (+/-4.8) kg of BW and 160.0 (+/-9.2) kg of BW. The association of these factors with BW at slaughter was also examined. Forty barrows, 78.1 (+/-1.75) kg of initial BW, were fed a control diet (2.5% sunflower oil) or a linseed diet containing 5% of whole extruded linseed. Both diets contained 170 mg of vitamin E and 250 mug of selenium. Eight pigs from each dietary treatment were slaughtered at 110 kg of BW and the others at 160 kg of BW. There was no dietary effect (P > 0.05) on growth, carcass characteristics, meat quality, or the activity of malic enzyme in LM and backfat. Inclusion of linseed increased (P < 0.05) n-3 PUFA content in both LM and backfat and decreased the n-6:n-3 PUFA ratio from 12 to 4.5 in LM, and from 11 to 3 in backfat. Liveweight at slaughter significantly influenced carcass characteristics, meat quality, total lipid and oxidative stability of LM, malic enzyme activity in adipose tissue, and fatty acid content of LM and backfat. This study shows that the inclusion of linseed in swine diets may improve the fatty acid profile of pork without deleteriously affecting oxidation or color stability. Thus, such a feeding practice may improve human health based on the n-6:n-3 PUFA ratio recommended for the human diet.

The effects of five different dietary fat supplements on fatty acid composition and oxidative stability of subcutaneous and kidney fat were evaluated in 36 Brown Swiss bulls and compared to a low fat diet in a monofactorial design. The following fat supplements were provided as additional fat at 30 g per kg feed dry matter: crystalline rumen-protected fat, coconut oil, and three types of crushed whole oilseeds (rapeseed, sunflower seed and linseed). Adipose tissues reflected differences (P < 0.05) in dietary fatty acid composition although to a lower extent. Using protected fat, which contained elevated levels of trans fatty acids, and sunflower seed, containing a high proportion of linoleic acid, significantly increased C18:1 trans fatty acid proportion in the adipose tissues. The use of sunflower seed increased conjugated linoleic acid. The oilseeds resulted in lower amounts of C16:0 in favour of C18:0. Except for linseed, all fat supplemented groups improved oxidative stability of adipose tissues as compared with control. This was explained by lower proportions of unsaturated fatty acids in adipose tissue (protected fat), by elevated alpha-tocopherol contents (rapeseed, sunflower seed) or by a combination of both (coconut oil). Fat colour remained unaffected by treatments. Compared to other fat supplements oilseeds, especially sunflower seed and rapeseed, can therefore be recommended to be fed to bulls in order to increase the proportions of C18 unsaturated fatty acids in adipose tissues and to maintain or improve oxidative stability.

The aim of the study was to investigate the effects of dietary linseed (rich in n-3 PUFA) on expression of inflammation-related genes and on growth performance of growing-finishing barrows. Two isoenergetic and isonitrogenous diets were formulated, one as the basal diet and the other containing 10% linseed. Twenty-four Landrace x Yorkshire barrows weighing 35 +/- 3.7 kg were randomly assigned to 1 of 4 treatment groups, with 6 pigs per group. During the entire experimental period of 90 d, these 4 groups of pigs were first fed the basal diet and then fed the linseed diet for 0, 30, 60, and 90 d before slaughter, respectively. Pig growth; messenger RNA (mRNA) expression of peroxisome proliferator-activated receptor-gamma (PPARgamma), IL-1beta0, IL-6, and tumor necrosis factor-alpha (TNF-alpha); and plasma concentrations of the 3 proinflammatory cytokines were measured and analyzed. Average daily feed intake did not differ among treatment groups (P > 0.05), but ADG (P < 0.05) and G:F (P < 0.01) responded quadratically to the duration of linseed diet feeding, and pigs in the 60-d treatment group had the greatest ADG and G:F. The mRNA expression of PPARgamma in loin muscle and spleen increased linearly (P < 0.01) with the duration of linseed diet feeding, whereas its expression in adipose tissue was not affected (P = 0.095). Tumor necrosis factor-alpha and IL-6 mRNA expression in muscle, adipose, and spleen, as well as serum concentration of TNF-alpha, decreased linearly (P < 0.01) with the duration of linseed diet feeding. Peroxisome proliferator-activated receptor-gamma mRNA abundance was negatively correlated with IL-1beta, IL-6, and TNF-alpha mRNA abundance both in muscle (R(2) = 0.63, P < 0.001) and in spleen (R(2) = 0.69, P < 0.001), and PPARgamma mRNA expression in spleen (R(2) = 0.59, P < 0.01) and muscle (R(2) = 0.52, P < 0.05) was negatively correlated with serum TNF-alpha concentration. There were also significant quadratic relations between ADG and expression of PPARgamma (P < 0.05) and splenic TNF-alpha (P < 0.05). These data suggest that intake of n-3 PUFA from the linseed diet led to significant decreases in the expression of proinflammatory cytokine genes, which may stimulate growth in growing-finishing barrows, at least in part, through a PPARgamma-dependent mechanism.

Fifty Suffolk-crossbred wether lambs, with an initial live weight of 29 +/- 2.1 kg, were allocated to one of five concentrate-based diets formulated to have a similar fatty acid content (60 g/kg DM), but containing either linseed oil (high in 18:3n-3); fish oil (high in 20:5n-3 and 22:6n-3); protected linseed and soybean (PLS; high in 18:2n-6 and 18:3n-3); fish oil and marine algae (fish/algae; high in 20:5n-3 and 22:6n-3); or PLS and algae (PLS/algae; high in 18:3n-3 and 22:6n-3). Lambs were slaughtered when they reached 40 kg. Growth performance and intake were similar (P > 0.35) among treatments. By contrast, gain:feed was higher (P < 0.05) in lambs fed the fish oil compared with the linseed oil or PLS/algae diets. Total fatty acid concentration (mg/100 g) in the neutral lipid of the longissimus muscle was not affected by treatment (P > 0.87) but was least (P < 0.05) in the phospholipid fraction in lambs fed the linseed oil diet. Lambs fed either diet containing marine algae contained the highest (P < 0.05) percentage of 22:6n-3 in the phospholipid (mean of 5.2%), 2.8-fold higher than in sheep fed the fish oil diet. In lambs fed the fish/algae diet, the percentage of 20:5n-3 was highest (P < 0.05), contributing some 8.7, 0.8, and 0.5% of the total fatty acids in the muscle phospholipid, neutral lipids, and adipose tissue, respectively. The percentage of 18:3n-3 in the phospholipid fraction of the LM was highest (P < 0.05) in lambs fed the linseed oil diet (6.9%), a value double that of sheep fed the PLS diet. By contrast, lambs fed the PLS diet had twice the percentage of 18:3n-3 in the muscle neutral lipids (3.8%) than those offered the linseed oil diet, and 5.5-fold greater than lambs fed the fish/algae treatment (P < 0.05), an effect that was similar in the adipose tissue. The percentage of 18:2n-6 was highest (P < 0.05) in lambs fed the PLS diet, where it contributed 33.7, 10.1, and 11.2% in the muscle phospholipid, neutral lipids, and adipose tissue, respectively. The highest (P < 0.05) muscle PUFA-to-saturated fatty acid (P:S) ratio was obtained in lambs fed the PLS diet (0.57), followed by the PLS/algae diet (0.46), and those fed the fish oil or linseed oil diets had the lowest ratios (0.19 and 0.26, respectively). The favorable P:S ratio of lambs fed the PLS/algae diet, in conjunction with the increased levels of 20:5n-3 and 22:6n-3, enhanced the nutritional qualities of lamb to more closely resemble what is recommended for the human diet.

The aim of the study was to investigate the effect of duration of feeding linseed (rich in n-3 PUFA) on peroxisome proliferator-activated receptor γ (PPARγ) and tumor necrosis factor (TNF-α) gene expression, and muscle mass of growing–finishing barrows. Two isoenergetic and isonitrogenous diets were formulated, and one of which was the basal diet and another one was the linseed diet including linseed at the level of 10%. Twenty-four Landrace × Yorkshire barrows weighing 35 ± 3.7 kg were randomly assigned to four treatments with six individuals per treatment. Pigs in treatment 1 (T1) fed the control diet throughout the experimental period, while pigs in T2, T3 and T4 fed the control diet except for 30, 60, and 90 d prior to slaughter when the linseed diet were fed. The experiment was conducted for 90 days. The longissimus muscle mass and each muscle mass in the hind leg were weighted. PPARγ and TNF-α mRNA expression levels in muscle, spleen and adipose tissue, and plasma concentrations of TNF-α data were measured and analyzed. The results showed that the longissimus muscle mass, quadriceps femoris muscle mass and semitendinosus muscle mass increased linearly (< 0.01) as prolonged the time of feeding linseed diet. The expression of PPARγ in longissimus muscle and spleen increased (< 0.01) linearly as prolonged the time of feeding linseed diet, while the expression of PPARγ in adipose tissue were not affected (= 0.095). Duration of linseed addition linearly decreased (< 0.01) TNF-α gene expression levels in the longissimus dorsi muscle, adipose and spleen, and serum concentration of TNF-α as well. The expression levels of PPARγ negatively correlated with the expression of TNF-α in muscle (R2 = 0.70, < 0.001) and spleen (R2R2 = 0.77, < 0.001) respectively. Likewise, PPARγ expression level in spleen (R2R2 = 0.59, < 0.01) or muscle (R2R2 = 0.52, < 0.05) negative correlated with serum TNF-α concentration, while there were significant quadratic relation between muscular PPARγ (R2R2 = 0.80, < 0.01) or muscular TNF-α (R2R= 0.87, < 0.01) expression and the longissimus dorsi muscle mass. These data suggested that duration of feeding linseed diet lead to a linear decrease of TNF-α gene expression, which may increase the muscle mass in growing–finishing barrows, at least in part, through a PPARγ-dependent mechanism.

Sixty-one finishing pigs (53.4 kg) were fed a control diet (containing soybean meal) or diets containing 20% intact canola (IC) or 20% ground canola (GC) for 8 wk. Diets were not isocaloric. Daily gain and feed efficiency were not affected by dietary treatment, but pigs fed GC ate less than pigs fed either IC or the control diet. Carcass measurements, obtained on 43 of the pigs, were not affected by diet. For 27 pigs, fatty acid composition of perirenal adipose tissue (PRF), subcutaneous adipose tissue (SCF), and longissimus muscle (LDM) was analyzed. Nine pigs (three per treatment) were randomly selected for fatty acid composition analysis of intramuscular adipose tissue (IMF) and for cholesterol analysis of several tissues. Pigs fed canola had greater (P less than .05) proportions of mono- and polyunsaturated fatty acids and less (P less than .05) saturated fatty acids in PRF and SCF. The differences were more pronounced for PRF than for SCF. In the LDM, pigs fed canola tended to have elevated levels of unsaturated fatty acids at the expense of the saturated fatty acids, but this effect was significant for linolenic acid only. The fatty acid composition of IMF was not affected by diet (P greater than .05). Diet did not alter the cholesterol content of the tissues, but cholesterol in IMF was higher (P less than .05) than in PRF, SCF, and LDM. In conclusion, 20% IC or GC did not alter growth performance or carcass characteristics of pigs. Feeding of canola increased the degree of unsaturation of PRF and SCF, but it had less effect on IMF and LDM.

The effect of n-3 fatty acid-enriched diets (in the form of 0.5% linseed oil with either 1.5% sunflower oil or 1.5% olive oil) and alpha-tocopheryl acetate supplementation (200 mg/kg feed) on lipid oxidation (thiobarbituric acid-reactive substances, TBARS) and cholesterol oxide products (COPS) in cooked pork was investigated. Longissimus muscle was studied. Meat from pigs fed 0.5% linseed oil-enriched diets had a higher proportion of n-3 fatty acid than meat from pigs in other dietary groups in neutral (P < 0.0001) and polar lipids (P < 0.0001), and a 20% reduction in the n-6:n-3 ratio was observed. Alpha-tocopheryl acetate supplementation increased (P < 0.05) monounsaturated fatty acids in polar lipids and increased (P = 0.0001) alpha-tocopherol levels in muscle. Alpha-tocopherol concentration in muscle was affected by dietary fat (P < 0.05). Groups receiving diets enriched with sunflower oil had significantly higher alpha-tocopherol levels (P < 0.05) in muscle than those groups receiving olive oil-enriched diets. Numbers of TBARS were significantly (P < 0.05) lower in the group fed supplemental olive oil than in those fed sunflower oil. Dietary linseed oil increased (P < 0.05) lipid oxidation principally at the initial period of storage in cooked pork. Overall, dietary alpha-tocopheryl acetate supplementation significantly increased (P < 0.001) lipid stability and decreased (P < 0.05) total COP production across the dietary groups. Alpha-tocopherol was a more effective antioxidant for decreasing TBARS values in cooked meat when adding sunflower oil to the diets instead of olive oil.

The intake of linoleic acid (LA) and alpha-linolenic acid (ALA) in relation to average daily weight gain (ADG) was studied in growing-finishing pigs kept on smallholdings in Central Vietnam. Groups of three piglets each were assigned randomly to 12 farms, where they were fed on local feedstuffs according to the farmer's choice but were given a restricted amount of dry matter according to a preset feeding regimen. On arrival at the farms, the pigs weighed 10.4 +/- 0.9 kg (mean +/- SD, n = 36) and at 130 days of age they weighed 45.4 +/- 9.6 kg. Dietary LA concentration ranged from 1.34 to 2.41 g/MJ metabolizable energy (ME) and ALA from 0.06 to 0.33 g/MJ ME. On a farm level, dietary LA and ALA concentrations were significantly correlated with their concentrations in adipose tissue, both correlation coefficients being 0.63. Dietary protein concentration and protein:energy ratio were significantly correlated with ADG, the correlation coefficients being 0.68 and 0.64. For individual piglets there were significant correlations between either LA or ALA in adipose tissue and ADG, the correlation coefficients being 0.37 and 0.45. Dietary protein concentration or protein:energy ratio was correlated with the dietary contents of LA and ALA. It is uncertain whether LA and ALA intake had a causal relationship with ADG. Since dietary LA levels were above the LA requirement, LA intake may not have limited growth. It is suggested tentatively that, through enhanced disease resistance, supplementation with ALA of the diets on the farms studied might have a positive influence on ADG.

This study investigated the effect of modifying the n-6:n-3 fatty acid ratio (FAR) of diets using linseed, soybean, and cottonseed oils on apparent digestibility, ruminal fermentation characteristics, growth performance, key circulating hormones, and the fatty acid profile of ruminal digesta, liver, and fore-shank muscle of growing lambs fed a high concentrate diet. Forty individually housed Katadhin Dorper lambs (average of 20.0 kg of BW) were fed Bermudagrass hay in ad libitum amounts and concentrates at 3.7% of BW daily. The concentrate contained 68.9% corn, 23.8% soybean meal, 3.3% limestone, and 4.0% oil supplements (DM basis). The treatments consisted of dietary n-6:n-3 FAR of 2.3:1, 8.8:1, 12.8:1, and 15.6:1. After feeding for 35 d in metabolism crates, lambs were slaughtered 15 h after feeding, and samples of ruminal digesta, blood, liver, and foreshank tissue were collected. Increasing dietary n-6:n-3 FAR did not affect the intake of DM nor the apparent digestibility of DM, ether extract, NDF, or ADF, but did increase apparent digestibility of CP (linear, P < 0.05). Concentrations of ruminal butyrate increased linearly (P < 0.05) with increasing dietary n-6:n-3 FAR, whereas the valerate concentration decreased linearly (P < 0.001). Concentrations of plasma insulin and IGF-I were not affected by dietary n-6:n-3 FAR. Concentrations of C18:3n-3 increased linearly (P < 0.001), whereas that of C18:2n-6 decreased linearly (P < 0.001) in ruminal digesta with decreasing dietary n-6:n-3 FAR. Concentrations of transisomers of fatty acids in ruminal digesta did not change. Proportions of C18:0 in liver and foreshank muscle were unchanged by diet. The proportion of trans11 C18:1 and cis-9 trans11 CLA decreased (P < 0.05) in liver but increased (P < 0.05) in foreshank muscle as dietary n-6:n-3 FAR decreased. Proportions of all measured n-3 fatty acids were greater in liver when diets contained more C18:3n-3 from linseed oil. By decreasing the dietary n-6:n-3 FAR, the proportions of n-6 fatty acids in foreshank muscle decreased dramatically; specifically, C18:2n-6 decreased linearly (P < 0.001) from 28.0 to 16.5% and C20:4n-6 decreased linearly (P < 0.001) from 14.7 to 8.6%. Although feeding a diet that contained more n-3 fatty acids increased the n-3 fatty acid concentration of muscle, the ratio of PUFA to SFA was decreased.

The effect of porcine somatotropin (pST) on the lipid profiles of adipose tissue and muscle was investigated. Sixteen crossbred barrows were injected daily with either 3 mg of pST or a placebo. After slaughter, total lipid and fatty acid composition of raw subcutaneous (SC) adipose and intermuscular (IM) adipose tissue and longissimus muscle were determined. The SC adipose tissue from pST-treated pigs had a 7.5% decrease in total lipid content; specific fatty acids 16:0, 18:0, and 18:1(n-9)c decreased most. The IM fat from pST-treated pigs had lower levels of 16:0 and 20:0. There was no effect of pST treatment on the lipid profile of the longissimus muscle. The data suggest that pST treatment produces small but significant changes in the saturated fatty acid content of adipose tissue in pigs.

The effect of peak lactation on the activities of a number of enzymes of glucose and lipid metabolism of perirenal and subcutaneous adipose tissue, skeletal muscle, liver, kidney cortex and mammary parenchyma of sheep are described. Enzymes studied included hexokinase (glucose utilization), pyruvate carboxylase (gluconeogenesis), pyruvate dehydrogenase (glucose oxidation and production of acetyl CoA for fatty acid synthesis), acetyl CoA carboxylase (fatty acid synthesis) and glycerol-3-phosphate acyltransferase (fatty acid esterification). Major changes that were found include a decrease in activities of enzymes of fatty acid synthesis and esterification in adipose tissues, decreased activity of pyruvate dehydrogenase in muscle and adipose tissues and increased pyruvate carboxylase; there was no change in activities of enzyme of fatty acid esterification in liver. Activities of hexokinase, acetyl CoA carboxylase and glycerol-3-phosphate acyltransferase have been estimated per tissue; this shows the quantitative importance of limiting glucose utilization by muscle and of suppression of fatty acid synthesis in adipose tissue for efficient partitioning of nutrients for milk production.

The effect of dietary n-3 fatty acids on the fatty acid composition and lipid peroxidation of different tissues in pigs were studied. 20 castrated male pigs were included in this investigation, one half was fed daily a diet containing 1.3 g n-3 fatty acids/kg diet (control) and 10 pigs were fed a diet containing 14 g n-3 fatty acids/kg diet (n-3 diet) at the growing-finishing period. The intake of dietary n-3 fatty acids increased the concentration of these fatty acids in backfat, and the neutral and polar fractions of skeletal muscle and heart homogenates. The polar fraction showed an increased relative concentration of n-3 fatty acids in comparison to control, while the n-6 fatty acid content was reduced. In heart homogenates there was an enlargement of n-3 fatty acids both in polar lipids and in neutral lipids whilst n-6 fatty acids were decreased. Feeding n-3 fatty acid enriched diet had no influence on meat quality parameters drip loss, meat colour or pH value. The lipid peroxidation (measured as malondialdehyde equivalents) was in the order liver > heart > skeletal muscle with higher values in the n-3 group. However, by stimulation of oxidation by Fe2+/ascorbate for 3 hours the order of oxidative products in the n-3 group was muscle > liver > heart, whereas in the control group the order was liver > heart = muscle. Summarized, feeding a highly n-3 fatty acid enriched diet caused an incorporation of these fatty acids and increased the susceptibility to peroxidation in all investigated tissues.

Our objective was to determine the effect of oil supplementation of pasture fed, beef cattle on the fatty acids, particularly CLA and PUFA, of muscle and s.c. adipose tissue. Forty-five Charolais crossbred heifers were blocked on BW and randomly assigned to 1 of 3 dietary regimens in a randomized complete block design (n = 15). The 3 treatments were: unsupplemented grazing (GO), restricted grazing plus a sunflower oil-enriched ration (SO), or restricted grazing plus a linseed oil-enriched ration (LO). Heifers were fed the experimental diets for approximately 158 d. Samples of LM muscle and s.c. adipose tissue were taken postmortem, the muscle fat was separated into neutral lipid and polar lipid (no separation was performed on the s.c. adipose tissue), and the fatty acid profile was determined by GLC. No effect of dietary treatment on carcass weight or total fatty acid concentration (mean 2,571 mg/100 g of muscle) in muscle fat was detected. Heifers offered SO had a greater (P < 0.001) proportion of CLA and C18:1trans-11 (1.90 and 9.35 vs. 1.35 and 6.89 g/100 g of fatty acids, respectively) in neutral lipid of muscle fat compared with those offered LO, which had a greater proportion of CLA and C18:1trans-11 than heifers offered GO (0.78 and 3.37 g/100 g of fatty acids, respectively). Similar effects were observed in the polar lipid and s.c. lipid. The PUFA:SFA ratio was greater in muscle fat and s.c. adipose tissue from supplemented heifers than in those offered GO (P < 0.001). Compared with LO, the PUFA:SFA ratio was greater (P < 0.05) in muscle fat of heifers offered SO, but there was no difference between SO and LO for this ratio in s.c. adipose tissue. The n-6:n-3 PUFA ratio was similar in muscle and s.c. adipose tissue for GO and LO, but it was greater (P < 0.05) for SO. It is concluded that supplementation of pasture-fed cattle with plant oil-enriched concentrates resulted in an increase in beef fat of some fatty acids considered to be of benefit to human health. Concentrates enriched with sunflower oil were more effective in increasing the CLA concentration, whereas linseed oil-enriched concentrates resulted in a more favorable n-6:n-3 PUFA ratio. The relevance to human health of the associated increase in C18:1trans-11 merits investigation.

  1. The objective of the present study was to determine effect of pomegranate seed oil (PSO) and linseed oil (LO) on haematological parameters, serum lipid profile and liver enzymes as well as fatty acids profile of adipose tissue in broilers.

  2. Broilers (n = 400) were fed on diets containing graded PSO levels (0.0%, 0.5%, 1.0%, 1.5%) with or without 2% LO. After 6 weeks of feeding, 6 male broilers from each group were slaughtered and abdominal fat, liver and blood samples were collected.

  3. Mixtures of pomegranate seed oil (0.5%, 1%) with linseed oil increased white blood cell level in broilers. Total cholesterol was elevated after LO supplementation whereas administration of PSO (1.5%) significantly decreased this parameter. PSO administration caused c9,t11 conjugated linoleic acid (CLA) concentration-dependent deposition in adipose tissue. By LO addition α-linolenic acid (ALA) content was enhanced, decreasing the n-6/n-3 ratio. PSO and ALA also affected oleic acid proportion in adipose tissue. Neither pomegranate seed oil nor linseed oil had any effect on liver parameters.

  4. Pomegranate seed oil had no negative effects on broiler health status and can be considered as a functional poultry meat component.

We conducted 3 independent experiments to demonstrate functional G-coupled protein receptor 43 (GPR43) and GPR120 in bovine intramuscular (i.m.) and subcutaneous (s.c.) adipose tissues. We hypothesized that media volatile fatty acids and long-chain fatty acids would affect cAMP-activated protein kinase-alpha (AMPKα) protein expression and cAMP concentrations differently in i.m. and s.c. adipose tissue. Experiment 1: oleic acid (18:1n-9) decreased phosphorylated AMPKα protein (p-AMPKα) and the p-AMPKα/AMPKα protein ratio in i.m. preadipocytes, increased the p-AMPKα/AMPKα protein ratio in bovine satellite cells, and had no effect in s.c. preadipocytes. Experment 2: ex vivo explants from the 5th to 8th longissimus thoracic rib muscle section of Angus crossbred steers were cultured 48 hr in media containing 0.25 µM ciglitizone, 5 mM glucose, and 5 mM acetate, in the absence or the presence of 100 µM oleic acid. Oleic acid increased acetate incorporation into fatty acids and GPR43 gene expression in i.m. adipose tissue (P < 0.05), but oleic acid had no effect on fatty acid synthesis or GPR43 expression in s.c. adipose tissue. Experiment 3: fresh s.c. and i.m. adipose tissue from the 5th to 8th longissimus thoracic rib muscle section of Angus crossbred steers was transferred immediately to 6-well culture plates containing 3 mL of KHB/Hepes/5 mM glucose. Samples were preincubated with 0.5 mM theophylline plus 10 μM forskolin for 30 min, after which increasing concentrations of acetate or propionate (0, 10−3, 10−2.3, and 10−3 M) in the absence or the presence of 100 μM oleic acid or 100 µM palmitic acid (16:0) were added to the incubation media. Acetate had no effect on forskolin-stimulated cAMP production in s.c. adipose tissue but decreased cAMP in i.m. adipose tissue (P < 0.05); this indicates a functional GPR43 receptor in i.m. adipose tissue. The combination of 10−2 M acetate and oleic acid decrease cAMP production in s.c. adipose tissue, consistent with GPR120 receptor activity, but oleic acid and palmitic acid attenuated the depression of cAMP production caused by acetate in i.m. adipose tissue. Palmitic acid depressed cAMP production in s.c. adipose tissue, and increased cAMP production in i.m. adipose tissue (P < 0.05). Propionate had no effect on cAMP production in s.c. or i.m. adipose tissue. These results provide evidence for functional GPR43 receptors in i.m. adipose tissue and GPR120 receptors in s.c. adipose tissue, both of which would suppress lipolysis.

OBJECTIVE: To examine effects of dietary protein quality (casein [CA] vs corn gluten [CG]) and dietary lipids (corn oil [CO] vs oil blend [OB] rich in long-chain polyunsaturated fatty acids [LCPUFAs]) on fatty acid composition in liver and adipose tissue after weight loss in overweight cats. ANIMALS: 24 ovariohysterectomized adult cats. PROCEDURE: Cats were allowed ad libitum access to a high-quality diet until they weighed 30% more than their ideal body weight. Cats were then randomly assigned to 1 of 4 weight-reduction diets (6 cats/diet) and were fed 25% of maintenance energy requirements per day. Diets consisted of CG-CO, CA-CO, CG-OB, and CA-OB, respectively, and were fed until cats lost weight and returned to their original lean body mass. Liver biopsy specimens and samples of perirenal, subcutaneous, and abdominal fat were obtained and analyzed for fatty acid content. RESULTS: Following weight loss, fatty acid composition of the liver and adipose tissue was primarily affected by protein quality in that cats fed CA had significantly higher percentages of 20:4(n-6) and 22:6(n-3) fatty acids than those fed CG. Cats fed the CG-CO diet had the lowest concentrations of LCPUFAs, suggesting that dietary lipids and protein quality each influence fatty acid composition in tissues. CONCLUSIONS AND CLINICAL RELEVANCE: These data provide direct evidence that dietary protein quality alters fatty acid composition of tissues during weight loss in cats. The fatty acid patterns observed suggest that protein quality may alter fatty acid composition through modulation of desaturase activity.
