ABSTRACT The effects of solitary and concurrent infection by Pratylenchus pene-trans and Verticillium dahliae on gas exchange of Russet Burbank potato (Solanum tuberosum) were studied in growth chamber experiments. Treatments were P. penetrans at low, medium, and high density; V. dahliae alone at one initial density; the combination of the nematode at these three densities and V. dahliae; and a noninfested control. Gas exchange parameters of leaf cohorts of different ages in the different treatments were repeatedly measured with a Li-Cor LI-6200 portable photosynthesis system. At 45 days after planting, joint infection significantly reduced net photosynthesis, stomatal conductance, and transpiration of 1- to 25-day-old leaf cohorts. Intercellular CO(2) levels were significantly increased by co-infection, especially in older leaves. The synergistic effect of co-infection on gas exchange parameters was greater in the oldest cohort than in the youngest cohort. No consistent effects on leaf gas exchange parameters were observed in plants infected by the nematode or the fungus alone. The relationship between the assimilation rate and stomatal conductance remained linear regardless of solitary or concomitant infection, indicating that stomatal factors are primarily responsible for regulating photosynthesis. The significant reduction of gas exchange in leaves of co-infected plants without reduction in intercellular CO(2) concentrations suggests that nonstomatal factors also play a role when both organisms are present.  相似文献

ABSTRACT The interactive effects of concomitant infection by the nematode Pratylenchus penetrans and the fungus Verticillium dahliae on symptom expression in Russet Burbank potato was studied in growth chamber experiments. Treatments were P. penetrans at three initial densities, V. dahliae at one inoculum density, the combination of the nematode at these three densities and the fungus, and a noninfested control. Gas exchange was measured nondestructively in leaf cohorts of different ages, one to three times weekly, with a LI-COR portable photosynthesis system. The single-pathogen treatments had no effect on assimilation or transpiration rates, but joint infection had a significant impact. In concomitant infection, photosynthesis was impaired more than transpiration, so estimates of leaf health were based on carbon assimilation rates only. Reductions in assimilation rate were apparent before the onset of visual symptoms. Assimilation rates decreased as much as 44% in the top, and newest, leaves of concomitantly infected plants, compared to rates in control plants. Even so, the health of newly produced leaves did not become progressively worse through time. With light use efficiency less than 0.20 mol of CO(2) fixed per mol of photosynthetically active radiation used as the criterion for disease incidence, disease progressed acropetally from the oldest to the youngest leaves. In plants infected with P. penetrans (0.8 nematodes per cm(3) of soil) in combination with V. dahliae, all leaves in cohorts 1 and 2 were symptomatic by 45 days after planting, and leaves in cohorts 3 to 6 became symptomatic at weekly intervals thereafter. For the control and single-pathogen treatments, the first time that light use efficiency fell below 0.20 in all leaves in cohort 1 was 71 days after planting. Concomitant infection reduced leaf life span by about 3 weeks. Both visual and physiological symptom expression were invariant to differences in initial nematode inoculum densities ranging from 0.8 to 2.5 nematodes per cm(3) of soil in one experiment and from 1.3 to 4.1 nematodes per cm(3) of soil in a second experiment.  相似文献

ABSTRACT Phytophthora capsici and Verticillium dahliae are two mycelial microorganisms associated with wilt symptoms on chile pepper (Capsicum annuum). Both pathogens occur in the same field and can infect a single plant. This study examined the nature of the co-occurrence of P. capsici and V. dahliae. Chile pepper plants were inoculated with each pathogen separately or with both pathogens concomitantly or sequentially. In concomitant inoculations, plants were inoculated with a mixture of zoospores of P. capsici and conidia of V. dahliae. In sequential inoculations, plants were inoculated with zoospores of P. capsici 4 days prior to inoculation with conidia of V. dahliae, or plants were inoculated with conidia of V. dahliae 4 days prior to inoculation with zoospores of P. capsici. Stem necrosis and leaf wilting were visible 3 to 4 days earlier in plants inoculated with both P. capsici and V. dahliae than in plants inoculated with P. capsici alone. Stem necrosis and generalized plant wilting were observed in plants inoculated with P. capsici alone, and stem necrosis, generalized plant wilting, and vascular discoloration were observed in plants inoculated with both P. capsici and V. dahliae by 21 days after inoculation. These symptoms were not observed in control plants or plants inoculated with V. dahliae alone. The frequency of recovery of V. dahliae from stems was approximately 85 to 140% higher across inoculum levels when plants were inoculated with both P. capsici and V. dahliae than when plants were inoculated by V. dahliae alone. Similarly, the frequency of recovery of V. dahliae from roots was approximately 13 to 40% higher across inoculum levels when plants were inoculated with both P. capsici and V. dahliae than when plants were inoculated by V. dahliae alone. There was no apparent antagonism between the two pathogens when they were paired on growth media. In general, when P. capsici and V. dahliae were paired on growth media, mycelial growth of each pathogen grown alone was not significantly different from mycelial growth when the pathogens were paired. Results suggest that wilt development is hastened by the presence of both P. capsici and V. dahliae in the same plants. The presence of P. capsici and V. dahliae in the same inoculum court enhanced infection and colonization of chile pepper by V. dahliae.  相似文献

Candidatus Liberibacter solanacearum’ was recently described as the causal agent of potato zebra chip disease. This pathogen occurs in North America, New Zealand, and Northern Europe on various crops, and may spread to other potato growing regions. Observation on ‘Ca. L. solanacearum’‐infected tomato and potato plants propagated in growth chambers over 5 years indicated that tomato plants (cvs Moneymaker and Roma) can be a latent carrier of ‘Ca. L. solanacearum’. Tomato plants graft‐inoculated with scions from latently infected tomato plants remained symptomless, but tested positive in a species specific PCR assay. ‘Ca. L. solanacearum’ was consistently detected in the top, middle and bottom portion of the symptomless tomato plants, including stem, petiole, midrib, vein, flowers and fruits. In tomato fruits, ‘Ca. L. solanacearum’ was evenly distributed in the tissues at the peduncle and style ends, as well as in the pericarp, and columella placenta tissues. This is the first report that ‘Ca. L. solanacearum’ is present in a plant reproductive organ. In contrast, potato plants (cvs. Jemseg, Atlantic, Shepody, Frontier Russet, Russet Burbank, Red Pontiac, and Russet Norkotah) grafted with scions from the same latently infected tomato plants resulted in typical symptoms of purple top, leaf scorch, and other disease symptoms in plants and brown discoloration in the vascular ring and medullary rays in tubers.  相似文献

Verticillium wilt of oilseed rape ( Brassica napus ) is caused primarily by Verticillium longisporum and has become a serious problem in northern Europe. In order to evaluate whether V. longisporum and V. dahliae differ in their interaction with oilseed rape, phenotypical and molecular assessments were made. Oilseed rape plants for fungal assessments were inoculated with V. longisporum and V. dahliae via root-dipping and samples were taken from roots, stems, leaves, flowers, pods and seeds during plant development. The infection by V. longisporum was found to start mainly in lateral roots and root-hairs, followed by colonization of the xylem vessels and extensive spread in stems and leaves, whereas V. dahliae infected the main roots and remained in the region below the cotyledon node of the plants. Re-isolation studies, together with PCR analysis of samples taken from early growth stages through to fully ripe plants, showed that the onset of flowering was a critical phase for V . longisporum to colonize plants. No seeds infected with V. longisporum were found. Mycelial growth from V. dahliae but not V. longisporum was significantly reduced on media containing tissue from a low glucosinolate B. napus genotype compared with growth on media containing tissue from a high glucosinolate cultivar. The results of this study suggest that V. longisporum favours B. napus as host and that the transition from the vegetative to the generative phase is of importance for the spread of the fungus in oilseed rape plants.  相似文献

Cauliflower (Brassica oleracea var. botrytis subvar. cauliflora) is susceptible to wilt caused by Verticillium dahliae but broccoli (B. oleracea var. italica subvar. cyamosa) is not. Infection of broccoli and cauliflower by a green fluorescent protein-expressing isolate of V. dahliae was examined using epifluorescence and confocal laser-scanning microscopy to follow infection and colonization in relation to plant phenology. Plant glucosinolate, phenolic, and lignin contents were also assayed at 0, 4, 14, and 28 days postinoculation. V. dahliae consistently infected and colonized the vascular tissues of all cauliflower plants regardless of age at inoculation, with the pathogen ultimately appearing in the developing seed; however, colonization decreased with plant age. In broccoli, V. dahliae infected and colonized root and stem xylem tissues of plants inoculated at 1, 2, or 3 weeks postemergence. However, V. dahliae colonized only the root xylem and the epidermal and cortical tissues of broccoli plants inoculated at 4, 5, and 6 weeks postemergence. The frequency of reisolation of V. dahliae from the stems (4 to 22%) and roots (10 to 40%) of mature broccoli plants was lower than for cauliflower stems (25 to 64%) and roots (31 to 71%). The mean level of aliphatic glucosinolates in broccoli roots was 6.18 times higher than in the shoots and did not vary with age, whereas it was 3.65 times higher in cauliflower shoots than in the roots and there was a proportional increase with age. Indole glucosinolate content was identical in both cauliflower and broccoli, and both indole and aromatic glucosinolates did not vary with plant age in either crop. Qualitative differences in characterized glucosinolates were observed between broccoli and cauliflower but no differences were observed between inoculated and noninoculated plants for either broccoli or cauliflower. However, the phenolic and lignin contents were significantly higher in broccoli following inoculation than in noninoculated broccoli or inoculated cauliflower plants. The increased resistance of broccoli to V. dahliae infection was related to the increase in phenolic and lignin contents. Significant differential accumulation of glucosinolates associated with plant phenology may also contribute to the resistant and susceptible reactions of broccoli and cauliflower, respectively, against V. dahliae.  相似文献

Experiments carried out in agar culture showed that the potato cultivar Pentland Javelin is relatively resistant to infection by Verticillium dahliae because it has a multilayered cortex of thick-walled cells, each producing lignituber appositions to invading hyphae. Its thick-walled xylem vessels are difficult to penetrate, its large vessel lumens difficult to obstruct, and its ability to produce tyloses limits the spread of the fungus, By comparison. Maris Anchor roots have fewer cortical layers and very small xylem vessels which are more easily blocked by hyphae. Maris Peer is intermediate in both the number of cortical cell layers and the size of xylem vessels. Globodera pallida juveniles assist V. dahliae to evade the natural defences of the root by opening an invasion channel for the fungus. In Maris Anchor and Maris Peer, but not in Pentland Javelin, the nematode provokes a widespread hypersensitive response; the resulting cell wall lignification impedes the growth of those hyphae which invade several days after the nematode. Hyphae grow well in syncytia and this enhances the probability of their penetrating xylem vessels. Although V. dahliae is generally held to induce symptoms through xylem blockage, the phloem colonization which was noted may affect the growth of both plant and nematode through its effect on assimilate movement towards root lips and into syncytia.  相似文献

We used cover crops with demonstrated efficacy against Verticillium dahliae and Pratylenchus penetrans in combination with the biocidal practice of solarization to determine the importance of targeting both organisms for managing potato early dying, an issue relevant to the search for alternatives to soil fumigation. Two experiments were conducted in commercial fields using a split-plot design with cover crop treatments of rapeseed, marigold, forage pearl millet, sorghum-sudangrass, and corn as the main plot factor and solarization as the subplot factor. Cover crops were grown and solarization applied in year one, followed by potato in year two. The main effect of solarization was significant for reduced inoculum levels of both organisms in year two and increased tuber yields. The main effect of cover crop was also significant with lower population densities of P. penetrans following the marigold and millet treatments and of V. dahliae following rape and sorghum-sudangrass. The cover crop treatments influenced yield in only one of the experiments in the absence of solarization. The combinatorial effect of cover crops and solarization resulted in a wide range of pathogen population densities. Mean soil inoculum levels were negatively related to yield for V. dahliae in experiment 1, and for P. penetrans and the P. penetrans × V. dahliae interaction in both experiments.  相似文献

ABSTRACT Potato early dying (PED), also known as Verticillium wilt, caused by Verticillium dahliae, is a seasonal yield-limiting disease of potato worldwide, and PED-resistant cultivars currently represent only a small percentage of potato production. In this study, we developed a real-time quantitative polymerase chain reaction (Q-PCR) approach to detect and quantify V. dahliae. The efficiency of the designed primer pair VertBt-F/VertBt-R, derived from the sequence of the beta-tubulin gene, was greater than 95% in monoplex Q-PCR and duplex (using Plexor technology) procedures with primers PotAct-F/PotAct-R, obtained from the sequence of the actin gene, designed for potato. As few as 148 fg of V. dahliae DNA were detected and quantified, which is equivalent to five nuclei. Q-PCR detected V. dahliae in naturally infected air-dried potato stems and fresh stems of inoculated plants. Spearman correlations indicated a high correlation (upward of 80%) between V. dahliae quantifications using Q-PCR and the currently used plating assays. Moreover, Q-PCR substantially reduced the variability compared with that observed in the plating assay, and allowed for the detection of V. dahliae in 10% of stem samples found to be pathogen free on the culture medium. The described Q-PCR approach should provide breeders with a more sensitive and less variable alternative to time-consuming plating assays to distinguish response of breeding lines to colonization by V. dahliae.  相似文献

Potato common scab caused by the actinobacterium Streptomyces scabiei is characterized by the formation of corky lesions on tubers that reduce their marketability. Management of common scab is very complex and often ineffective under various environmental conditions. Using potato varieties that are more resistant to common scab remains one of the most efficient strategies to control this disease. However, very little is known about the factors associated with resistance to common scab. Somaclone RB9 regenerated from thaxtomin A-habituated potato Russet Burbank calli produced tubers more resistant to common scab than the original variety. Comparison of the RB9 tuber proteome with that of Russet Burbank using label-free quantitative proteomic analysis revealed changes in the accumulation of defence-related proteins from the patatin and lipoxygenase (LOX) families, which are involved in the metabolism of lipids, and of two miraculins of the Kunitz-type protease inhibitors family. The implication of LOX during common scab infection was studied using synchronized minitubers developed from leaf-bud cuttings. S. scabiei infection stimulated the accumulation of LOX in both Russet Burbank and RB9 minitubers, but this accumulation was intensified in RB9 minitubers. Infection also increased LOX activity in Russet Burbank and RB9 minitubers. However, LOX activity measured in noninfected RB9 minitubers was similar to that of infected Russet Burbank minitubers, indicating endogenous activation of LOX activity in RB9 minitubers. We discuss how increased LOX abundance and activity in the somaclone RB9 may contribute to improving tuber defence against common scab.  相似文献

 由大丽轮枝菌引起的棉花黄萎病是一种难以防治的土传病害,西兰花残体还对其有一定的防治作用,防效为62.82%。为解析西兰花残体对棉花黄萎病的防治机制,本研究通过土壤杆菌转化的方法构建了大丽轮枝菌的绿色荧光标记菌株,采用共聚焦显微镜观察标记菌株在添加西兰花残体营养基质中和空白对照营养基质种植的棉花体内的侵染和扩展情况。结果表明构建的标记菌株gfp-wx-1与野生菌株wx-1的生物学特性无显著性差异。同时发现大丽轮枝菌在添加西兰花残体营养基质中棉花体内扩展较慢,具体表现为:在空白营养基质种植的棉花体内,大丽轮枝菌在接种第2 d时就可侵染根尖表层及根内部,第3 d到达茎部维管束,第5 d叶片可观察到少量病原菌,第7 d第一片子叶呈现大量病原菌,后期迅速扩展,叶片出现黄萎病斑;而在营养基质中添加西兰花残体种植的棉花体内,大丽轮枝菌在接种第2 d时就可侵染根尖表层,第3 d侵染根尖内部,第5 d侵染茎部维管束,直到第7 d第一片子叶才出现少量病原菌,后期扩展慢,叶片病斑较少。本研究通过荧光定量PCR方法检测了大丽轮枝菌在两个处理棉花根部定殖情况,结果表明西兰花残体能够显著降低大丽轮枝菌在棉花根部定殖的量。该研究初步明确了西兰花残体对棉花黄萎病的防治机制,为棉花黄萎病的绿色防治提供了一条新的途径。  相似文献

Interactions between lettuce and a green fluorescent protein (GFP)-expressing, race 1 isolate of Verticillium dahliae, were studied to determine infection and colonization of lettuce cultivars resistant and susceptible to Verticillium wilt. The roots of lettuce seedlings were inoculated with a conidial suspension of the GFP-expressing isolate. Colonization was studied with the aid of laser scanning confocal and epi-fluorescence microscopes. Few differences in the initial infection and colonization of lateral roots were observed between resistant and susceptible cultivars. Hyphal colonies formed on root tips and within the root elongation zones by 5 days, leading to the colonization of cortical tissues and penetration of vascular elements regardless of the lettuce cultivar by 2 weeks. By 8 to 10 weeks after inoculation, vascular discoloration developed within the taproot and crown regions of susceptible cultivars well in advance of V. dahliae colonization. Actual foliar wilt coincided with the colonization of the taproot and crown areas and the eruption of mycelia into surrounding cortical tissues. Advance colonization of stems, pedicels, and inflorescence, including developing capitula and mature achenes was observed. Seedborne infection was limited to the maternal tissues of the achene, including the pappus, pericarp, integument, and endosperm; but the embryo was never compromised. Resistant lettuce cultivars remained free of disease symptoms. Furthermore, V. dahliae colonization never progressed beyond infected lateral roots of resistant cultivars. Results indicated that resistance in lettuce may lie with the plant's ability to shed infected lateral roots or to inhibit the systemic progress of the fungus through vascular tissues into the taproot.  相似文献

Fifteen isolates of binucleate Rhizoctonia fungi (BNR) were studied as potential biocontrol agents for protection of potato from Rhizoctonia canker in artificially infested greenhouse soil and potato fields naturally infested with Rhizoctonia solani (AG-3). Eight of the BNR reduced incidence and severity of Rhizoctonia stem canker in greenhouse experiments by an average of 78 and 85%, respectively. In a field naturally infested with R. solani, selected isolates of BNR and the fungicide Tops 2.5D (thiophanate-methyl) were equally protective of potato from Rhizoctonia stem canker. BNR isolates gave protection of potato from Rhizoctonia stolon canker similar to PCNB and superior to Tops 2.5D. Cultivars Atlantic, Irish Cobbler, Kennebec, Norchip, Russet Burbank, and Superior were protected equally from Rhizoctonia stem canker by selected isolates of BNR under field conditions. Isolates of BNR show potential as biocontrol agents for protection of potato from Rhizoctonia canker.  相似文献

大丽轮枝菌是引起棉花、马铃薯等重要作物黄萎病的土传病原真菌,其分泌的胞外蛋白是侵染寄主的重要毒力因子,因此研究胞外蛋白与寄主的相互作用具有重要意义。本研究旨在构建高效的棉花酵母双杂交cDNA文库,并通过已知大丽轮枝菌效应子VCR1互作蛋白的筛选来评价构建文库的质量。分别采用大丽轮枝菌及茉莉酸甲酯、水杨酸和乙烯处理海岛棉‘海7124’以诱导其抗病基因表达,混合RNA样品构建酵母双杂交cDNA文库,文库滴度达1.18×108 cfu/mL,检测重组率100%,插入片段在300~2000 bp之间。以效应子VCR1为诱饵筛选获得27个潜在互作蛋白,并进一步确证了1个与VCR1互作的靶标蛋白。本研究构建的酵母双杂交文库将为筛选与大丽轮枝菌效应子互作的蛋白及后续互作机理研究提供重要的研究基础。  相似文献

ABSTRACT Digestive cysteine proteinases have been isolated from plant-parasitic nematodes as well as coleopteran and hemipteran insects. Phytocystatins, inhibitors of cysteine proteinases, are found in a number of plants where they may play a role in defense against pathogens and pests. The cDNAs of the phytocystatins from rice, oryzacystatin I (OC-I) and oryzacystatin II (OC-II), were expressed in alfalfa (Medicago sativa) plants under the control of the potato protease inhibitor II (PinII) promoter and the plants were evaluated for resistance to the root-lesion nematode (Pratylenchus penetrans). A PinII-beta-glucuronidase (GUS) gene was introduced into alfalfa to determine the pattern of gene expression from this promoter. Constitutive GUS expression was observed in leaf and root vascular tissue, and in some plants, expression was observed in leaf mesophyll cells. Mechanical wounding of leaves increased GUS expression approximately twofold over 24 h. Inoculation with root-lesion nematodes resulted in localized GUS expression. Populations of root-lesion nematodes in alfalfa roots from one line containing the PinII::OC-I transgene and one line containing the PinII::OC-II transgene were reduced 29 and 32%, respectively, compared with a transgenic control line. These results suggest that oryzacystatins have the potential to confer increased resistance to the root-lesion nematode in alfalfa.  相似文献

Using potato, eggplant and thorn apple as test plants, the relationship between soil inoculum density and plant infection was studied as a basis for the development of a quantitative bioassay of Verticillium dahliae. A linear relationship was demonstrated (P < 0.05) between soil inoculum density and population density on roots for all three test plants and for soil inoculum density and population density in sap extracted from stems for eggplant. Correlation coefficients were higher with densities on or in roots (R2 varying from 0.45 to 0.99) than with densities in stems (R2 varying from 0.04 to 0.26). With eggplant, population densities on/in root and in sap extracted from stems were significantly correlated at 20 and 25°C with Pearson's correlation coefficients of 0.41 and 0.53, respectively. For potato, root colonization was higher at 15 than at 20°C, whereas the reverse applied to eggplant. Stems of potato were less colonized than stems of eggplant. The pathozone sensu Gilligan (1985) was calculated to be <300 µm, indicating that infection was caused by microsclerotia which were located close to the roots. To assess the density of V. dahliae in plant tissue pipetting infested plant sap on solidified ethanol agar medium without salts yielded higher densities than using pectate medium or mixing sap with molten agar. A bioassay for determining effects of (a)biotic factors on development of V. dahliae in the plant is recommended with eggplants as a test plant, grown in soil infested with 300 single, viable microsclerotia g-1 soil at a matric potential of –6.2 kPa, and incubated at 20°C for 8 weeks.  相似文献

ABSTRACT Broccoli is resistant to Verticillium dahliae infection and does not express wilt symptoms. Incorporation of broccoli residues reduces soil populations of V. dahliae. The effects of broccoli residue were tested on the colonization of roots by V. dahliae, plant growth response, and disease incidence of both broccoli and cauliflower in soils with different levels of V. dahliae inoculum and with or without fresh broccoli residue amendments. The three soils included a low-Verticillium soil, a high-Verticillium soil, and a broccoli-rotation soil (soil from a field after two broccoli crops) with an average of 13, 38, and below-detectable levels of microsclerotia per g of soil, respectively. ABSTRACT Six potato cultivars were grown with or without the addition of Verticillium dahliae inoculum and were watered at 50, 75, or 100% estimated consumptive use. The applied water x cultivar interaction was significant (P = 0.009 and P = 0.001 for 1996 and 1997, respectively) for the relative area under the senescence progress curve (RAUSPC). With a decrease in water, there was an increase in RAUSPC. A significant interaction of inoculum density x cultivar also was found, based on RAUSPC (P = 0.0194 and P = 0.0033 for 1996 and 1997, respectively). In V. dahliae-infested plots, 'Katahdin' and 'Ranger Russet' were resistant to Verticillium wilt. Population size of V. dahliae in stem apices was significantly lower in 'Katahdin' in both 1996 and 1997 (P = 0.0001) and in 'Ranger Russet' in 1997 (P = 0.0001) than in the other cultivars. 'Russet Burbank' and 'Shepody' had large apical stem populations of V. dahliae and higher RAUSPC values associated with both V. dahliae inoculum and decreased amount of applied water. Marketable tuber yield was unaffected by V. dahliae in both years. Cultivar resistance to Verticillium wilt was related to cultivar tolerance to moisture deficit stress. Results suggest that moisture deficit stress response has the potential to be a useful tool in protocols for screening potato for Verticillium resistance.  相似文献

ABSTRACT Controlled environments were used to study the relationship between the root-knot nematode (Meloidogyne incognita) and Thielaviopsis basicola on cotton. Temperature treatments were continuous 20, 24, and 28 degrees C or two cyclic linear regimes with ranges of 14 to 32 or 18 to 28 degrees C over 24 h. Cotton seeds were planted in fumigated soil infested with T. basicola, M. incognita, or both. After 42 days, pathogen effects on plant growth and pathogen development were evaluated. Histology was conducted on roots collected 14, 28, and 42 days after planting in the continuous 24 degrees C treatment. Reductions in plant height-to-node ratio and total fresh weight were observed for soils infested with both pathogens compared with the control or with soils infested with either pathogen, except for M. incognita-infested soil at 28 degrees C. T. basicola reduced root galling and reproduction of the nematode at all temperatures. Vascular discoloration caused by T. basicola was greater in the presence of M. incognita compared with that by T. basicola alone. At 2 and 4 weeks, histological studies showed that plants grown in all T. basicola-infested soils contained chlamydospore chains on the root surface and in cortical cells. The fungus was not observed inside the vascular cylinder. Roots from 4-week-old plants from soils infested with T. basicola and M. incognita showed fungal sporulation in vascular tissue and localized necrosis of vascular tissue adjacent to the nematodes. At 6 weeks, plants grown in soil infested with T. basicola alone exhibited no remaining cortical tissue and no evidence of vascular colonization by the fungus. Six-week-old plants grown in T. basicola + M. incognita-infested soils exhibited extensive vascular necrosis and sporulation within vascular tissue. These studies suggest that coinfection expands the temperature ranges at which the pathogens are able to cause plant damage. Further, M. incognita greatly increases the access of T. basicola to vascular tissue.  相似文献
