Of 32 bulls showing inflammatory conditions in the pelvic genital organs, 25 were examined post mortem (24 showing vesiculitis with or without ampullitis and one showing ampullitis alone). In ten cases (40%) the following malformations or rare anatomical deviations were demonstrated by careful gross-anatomical dissection: Segmental aplasia of the Wolffian duct: 1; segmental hypoplasia of the Wolffian duct: 3; asymmetric implantation of the two ampullae: 3; accessory vesicular glands: 1; uterus masculinus persistens: 2. These results corroborate earlier observations that a congenital defect may play a certain r?le in the pathogenesis of seminal vesiculitis, most probably by acting as a locus minoris resistentiae in or around colliculus seminalis. On account of the above findings, and considering the prognosis for the condition and the lack of effective treatment, it can in general be recommended, that diseased bulls should be slaughtered as soon as the diagnosis seminal vesiculitis is final.  相似文献

The accessory sex glands of 521 slaughtered bulls were examined for lesions; normal anatomical observations were also made.
The average weights and measurements (length and width) of the seminal vesicles and bulbourethral glands of 110 bulls were 34.1 g and 9.1 × 2.5 cm, and 4.6 g and 2.9 × 1.9 cm, respectively. Weights of both glands were correlated to age, but bulbourethral gland weight was more closely correlated to bodyweight than age. The average length and diameter of the ampulla was 11.1 and 1.0 cm, respectively.
Seminal vesiculitis and ampullitis were found in 9.0% and 3.8% of bulls respectively. The incidence of both conditions was highest in old bulls. Mesonephric duct anomalies (observed in 1.7% of bulls) and other congenital and inflammatory lesions of the accessory sex glands were also recorded.
Macroscopically normal accessory sex glands of 86 bulls were studied histologically and a high percentage of all glands showed inflammatory changes.
Attempts to isolate viral agents, Mycoplasma, and Chlamydia from accessory sex glands were unsuccessful. Corynebacterium pyogenes was isolated from two cases of seminal vesiculitis. IBR-IPV virus, Campylobacter fetus subsp. venerealis and Tritrichomonas foetus were frequently isolated from the prepuce of bulls, many of which also had positive serological titres to Leptospira pomona and Leptospira hardjo.
No one factor capable of causing bull infertility was consistently identified, and the incidence figures for observed conditions of the accessory sex glands were generally comparable to those found in bull populations elsewhere.  相似文献

A strain of M. bovigenitalium was isolated from semen of a bull (K) with chronic seminal vesiculitis. Using this strain a vesiculitis of the same type as found in bull K, characterized by simultaneous acute and chronic lesions, was induced experimentally in 2 bulls by direct inoculation into the vesicular glands. In the acute phase a marked infiltration of eosinophils was found in the interstitial tissues and alveoli whereas in the chronic phase fibrosis, lymphoid and epithelial hyperplasia were seen. Degeneration of vascular walls and connective tissue was common.On inoculation into the testis of 3 bulls a chronic epididymitis and ampullitis were produced. The histological changes were of the same type as found in the vesicular glands of the 3 bulls with vesiculitis.By indirect hemagglutination a specific and significant increase in serum antibody could be demonstrated. As the titers were low and the maximum titers were reached early, it will probably not often be possible to make an etiological diagnosis on the basis of serological evidence.A comparative experiment employing the type strain (PG 11) of M. bovigenitalium was performed. A rise in antibody titer was seen, but neither clinical nor histological changes could be demonstrated.  相似文献

During the fall of 1966 several strains of Mycoplasma were isolated from semen samples of bulls. A strain, “K”, isolated from a bull with vesiculitis was selected for a series of experiments and proved to be pathogenic for the mammary gland of cows and for the genital tract of bulls (Ernø 1967, Blom & Εrnø 1967). The “K” strain has now been examined serologically by the author, and it must be considered as belonging to the species M. bovigenitalium as determined by complement fixation and indirect haemagglutination tests. In this study, by using the polymerase chain reaction (PCR) diagnosis for the detection and identification of Mycoplasma, we investigated mycoplasmas contaminating the semen of yearling bulls affected by seminal vesiculitis. The bulls presented neither subclinical nor clinical contagious bovine pleuropneumonia signs and the complement fixation test for specific antibodies was negative. Furthermore, we have investigated mycoplasmas isolated from semen of healthy breeding bulls of several breeds and origins, which routinely underwent breeding soundness examinations and presented no clinical signs of seminal vesiculitis. We were able to demonstrate mycoplasma infection in all tested samples by i) growth on mycoplasma-specific media and ii) a PCR-based method using a mycoplasma-specific MGSO/GPO1 primer set to amplify the 16S fragment rDNA. In addition, the identification of Mycoplasma species was made by PCR using the MSC1/MSC2 primer set that specifically amplifies M. mycoides subsp. mycoides SC or the MM450/MM451 primer set followed by AsnI digestion analysis in order to identify M. mycoides subsp. mycoides LC. The data presented herein clearly show that M. mycoides subsp. mycoides SC infection was associated with seminal vesiculitis while M. mycoides subsp. mycoides LC was only found in bull semen from healthy control animals. Our findings confirm that the M. mycoides subsp. mycoides SC is shed in the sperm making the ejaculate a valuable biological sample for the isolation of these bacteria from serologically negative animals. Although the pathogenic role of M. bovigenitalium in bull seminal vesiculitis has been established, our clinical findings, semen characteristics, microbiological and bacterial genomic analysis strongly suggest that M. mycoides subsp. mycoides SC may contribute to induce vesicular adenitis in the bull.  相似文献

Twenty-two buffalo bulls suffering from three different types of infertility were slaughtered and used for this study. Except for the reproductive system, no signs of localised or generalised disease were observed. Microbiological investigations were negative for brucellosis, vibriosis, mycoplasma and other non-specific microorganisms. Nine bulls with type 1 infertility had low bodyweights and underdevelopment of testes, accessory sex glands and endocrine glands. This picture suggests a total dysfunction of the pituitary-growth-gonadal axis. One bull of this type also showed bilateral epididymitis. Four out of 11 bulls with type 2 infertility had low bodyweights and most suffered from underdevelopment of testes, accessory sex glands and endocrine glands. Six bulls of this type had lesions of either epididymitis or orchitis or both. Two of these animals showed adhesions of periorchitis. One also showed seminal vesiculitis. In two bulls with type 3 infertility, bodyweights, reproductive organs and endocrine glands were normal. In later life, they yielded poor quality semen. Semen samples collected a few months before slaughter from nine bulls with type 2 and type 3 infertility were of poor quality and had higher percentages of abnormal spermatozoa in most cases.  相似文献

Antibody responses in serum and semen, and immunoglobulin containing cell (ICC) populations in the genitalia of bulls were compared after inoculating Brucella abortus strain 19 into the seminal vesicles of two bulls (ISV route) and into testes in two other bulls (IT route). Bulls seroconverted as early as 1 week post-infection (PI). Peak serum titres as determined by the serum agglutination test (SAT), complement fixation test (CFT) and ELISA occurred at PI weeks 3, 4 and 5 respectively. Highest titres were in IT inoculated bulls. Seminal antibodies against B. arbotus S19 were demonstrated from 2 weeks PI by both the SAT and the Rose Bengal Test (RBT) and highest titres occurred at PI weeks 3 and 4. Examination of immunoglobulins (Ig) in semen, however, revealed no significant differences of Ig isotypes between infected and control animals at any examination time. When bulls were killed at 7 weeks PI, quantitation of ICC in genital sections stained by the peroxidase-anti-peroxidase method revealed an overwhelming predominance of IgG containing cells in inflamed organs. In all cases IgG1- and IgG2-containing cells were prevalent, and present in approximately equal numbers. IgA-containing cells were second in prevalence in inflamed tissues while IgM cells were always in low percentage. High prevalence of ICC in infected genitalia, associated with elevated specific seminal antibodies but not with increased seminal Ig indicates that most Ig remains localised in tissues and is not transferred into genital secretions.  相似文献

Effects of zeranol on scrotal circumference, serving ability, semen characteristics, and postmortem measurements of the genital organs were determined in beef bulls from 9 to 20 months of age. Group 1 (n = 5) served as a nonimplanted control group. Group 2A (n = 5) was implanted with 36 mg of zeranol at birth and at 3 and 6 months of age. Group 2B (n = 5) was implanted with 36 mg of zeranol every 3 months from birth through 18 months of age. Scrotal circumference was adversely affected by zeranol in groups 2A and 2B, but values approached those of group 1 with increasing age. Serving ability was also affected adversely but tended to recover with increasing age. Semen quality was low in groups 2A and 2B and did not improve with increasing age. There was no difference in testicular weight, vesicular gland weight, and penis length among groups when bulls were slaughtered at 20 months of age. Epididymal weight was greater in group-2B bulls and was most likely a consequence of epididymal lesions. Histologic examination of the genital organs revealed that zeranol induced adenomyosis and sperm granulomas in the caudae epididymidis and markedly altered the structure of the sexual accessory glands of bulls in groups 2A and 2B. Alterations in the vesicular glands were characterized by reduced alveolar development and an increase in connective tissue. Low epithelium associated with focal areas of squamous metaplasia were common in the prostate of groups 2A and 2B bulls. Lesions in the bulbourethral glands were characterized by low glandular epithelium, focal areas of squamous metaplasia, cystic collecting ducts, and an increase in connective tissue. Groups 2A and 2B had more abnormal seminiferous tubules than did group 1. Lesions in groups 2A and 2B may have been direct effects of zeranol or may have resulted from reduced testosterone secretion.  相似文献

The genitalia from 34 bulls were examined for the presence of Brucella lesions using histological, bacteriological and serological methods. Tissue fluids extracted from the various genital organs and the serums were examined using the serum agglutination test (SAT), the Rose Bengal plate agglutination test (RBPT), the complement fixation test (CFT) and the indirect haemagglutination test (IHLT) for the presence of Brucella antibody. Titres to one or more of these tests were found in 17 bulls and B. abortus was cultured from 5 of these. The testing of tissue fluids from genital organs did not increase the number of serologically positive animals.  相似文献

Bull semen is commonly contaminated with mycoplasmas. To determine the source of contamination, semen and the genital tracts of 45 artificial insemination bulls were cultured for these organisms. The results indicate that mycoplasmas colonize the prepuce and the distal part of the urethra. Only rarely were they found in the ampullae or seminal vesicles. In 92% of the bulls with contaminated semen the same Mycoplasma species or Ureaplasma diversum was isolated from the prepuce and urethral orifice as was found in the semen. This suggests that the prepuce and distal urethra is the source of contamination. Colonization of the genital tracts with Mycoplasmas or U. diversum was not associated with histological changes.  相似文献

The objectives of this work were to determine the site of persistence of lumpy skin disease virus (LSDV) in bulls shedding the virus in semen for a period longer than 28 days, to determine if the virus is present in all fractions of semen and to study lesions that developed in the genital tract. Six serologically negative postpubertal bulls were experimentally infected with a virulent field isolate of LSDV. The polymerase chain reaction (PCR) was performed on sheath washes, vesicular fluid, supernatant and cell‐rich fractions of semen from day 10 to day 26 postinfection (p.i.). Bulls that were positive by PCR on the whole semen sample collected on day 28 p.i. were slaughtered and tissue samples from their genital tracts submitted for histopathological evaluation, immunoperoxidase staining, virus isolation and PCR. Two of the bulls developed severe lumpy skin disease (LSD) and were found to be shedding viral DNA in their semen on day 28 p.i. Viral DNA was identified in all semen fractions from all bulls, but mostly from the cell‐rich fraction and from the severely affected bulls. The PCR assay was positive on postmortem samples of testes and epididymides from the two severely affected bulls. Virus could be recovered from the testes of these two bulls and from the epididymis of one of them. Immunoperoxidase staining was positive for LSDV staining in sections of testes and epididymides exhibiting necrosis. This study suggests that the testis and epididymis are sites of persistence of LSDV in bulls shedding virus in semen for prolonged periods and revealed that viral DNA is present in all fractions of the ejaculate.  相似文献

Shortening age at puberty of crossbred breeding bull is an important issue in the tropics. This study aimed at selecting crossbred bulls at earliest possible age with bigger scrotum and potential for donating quality semen. One hundred and 31 pre-joining crossbred bulls of Central Artificial Insemination Laboratory, Saver, Dhaka were examined. The bulls being trained by seeing semen collection from mature bulls were allowed ejaculation into the artificial vagina at homosexual mount during a 20 min time at three occasions, every three months. Eighty one of 131 bulls produced at least one ejaculate during the study and their mean +/- SD age and scrotal circumference (SC) were 20.3 +/- 4.7 months and 28.2 +/-2.7 cm, respectively. Bulls' body weight, body condition score (BCS) and SC influenced the attainment of their puberty (p < 0.05). Bull's body weight had positive effects on scrotal circumference and ejaculate volume (p < 0.05). Scrotal circumference positively influenced the percentages of normal spermatozoa (p < 0.05). Scrotal skin-fold thickness negatively influenced the proportion of spermatozoa with normal head (p < 0.05). Based on age at first ejaculate and SC, 29.6% bulls (n = 24) were selected by cluster analysis. Selected bulls had mean +/- SD age 17.9 +/- 2.2 months, body weight 287.3 +/-48.6 kg, SC 30.5 +/- 1.5 cm, ejaculate volume 3.4 +/- 1.3 ml, sperm motility 50.8 +/- 17.2%, total spermatozoa per ejaculate 2541.9 +/- 1699.2 million and normal spermatozoa 81.5 +/-14.5%. The selected pubertal bull group was different from the unselected pubertal bulls at MANOVA (p < 0.0001). About 30% of pubertal crossbred bulls can be selected with shorter age and larger scrotum at puberty under conditions prevailed in Bangladesh.  相似文献

Simmental and Hereford bulls were individually fed varying levels of the same diet to determine the effects of energy intake after weaning, degree of fatness, and short-term weight change on reproductive characteristics of yearling beef bulls. For 200 d (ending in May), 29 Simmentals were fed an average of 14.6, 19.2 or 23.8 Mcal and 27 Herefords were fed 13.4, 17.5 or 22.2 Mcal metabolizable energy per bull daily. Bulls then were adjusted to a roughage diet for 10 d before grazing brome pasture for 38 d (ending in June). Energy level did not affect motility or sperm morphology of semen collected with an artificial vagina in May and June. High energy levels were not detrimental to bull performance during a 30-min serving-capacity test in May or June. Backfat thickness was not related to semen characteristics or serving capacity. Amount of weight lost from May to June did not affect the semen quality or serving capacity of Herefords. Those Simmental bulls that had a more positive weight change from May to June had a more favorable change in semen quality from May to June (P less than .05) due to lower semen quality in May (P less than .05). The high level of energy was not detrimental to semen characteristics or serving capacity. Some of the Simmental bulls may have been underfed for maximum semen quality at the beginning of the pasture period. Within the normal range of energy fed to beef bulls from weaning to the beginning of the breeding season as yearlings, it may be more likely to underfeed breeds of large mature size than to overfeed British breeds.  相似文献

Paratuberculosis, caused by Mycobacterium avium subsp. paratuberculosis, has particular importance in cattle due to the resulting chronic diarrhoea, weight loss, decreased production, infertility and eventual death. While faecal oral route of infection is generally recognised, reports about semen-derived infection are rare. The objective of this work was to assess whether M.a. paratuberculosis may disseminate from the gastrointestinal tract to reproductive organs, and compare this event between naturally infected bull-calves and breeding bulls. Ten bull-calves, aged 6-28 weeks and four breeding bulls were tested by serology, faecal and tissue culture, IS900 PCR and RFLP. In seven bull-calves M.a. paratuberculosis was isolated predominantly from mesenteric lymph nodes (75%); isolates from mucosa of the intestine constituted 25%. In three breeding bulls, M.a. paratuberculosis was isolated both from intestinal mucosa and mesenteric lymph nodes. Head and mediastinal lymph nodes, liver, spleen and semen of bull no. 1 (Holstein-Friesian); testes and epididymis of bull no. 2 (Piemonte); testes, epididymides and seminal vesicle of bull No. 3 (Hereford); and seminal vesicle of bull No. 4 (Simmental) tested positive by culture. Hot-start PCR revealed M.a. paratuberculosis in semen, seminal vesicle and intestinal tissue where culture isolation was difficult. Isolates from bull-calves and breeding bulls were of RFLP types B-C9 and B-C1, respectively. Bull-calves born in infected herd can be sources of infection when later used for natural mating or artificial insemination. Sub-clinically infected bulls release M.a. paratuberculosis into semen, consequently infecting the uterine environment of cows.  相似文献

The bulbourethral glands of 323 Bos indicus or B. indicus crossbred bulls more than 1 1/2 years old were examined in an abattoir study. Bulbourethral adenitis was diagnosed grossly and confirmed by histological examination in 4 (1.2%). Unilateral chronic interstitial inflammation was seen in 2 cases; one of these was associated with a degenerative-type seminal vesiculitis. In the others adenitis was bilateral; in one case it was associated with a concretion and foreign (plant) material in the principal duct of the left bulbourethral gland; in the other bilateral case, numerous calculi were present and microscopically, a chronic active and diffuse inflammation was observed. Chemical analysis of the calculi showed calcium oxalate and tricalcium phosphate to be the most important components. Corynebacterium spp was isolated from the lesion with multiple calculi but attempts to isolate Chlamydia spp, Mycoplasma spp and Brucella abortus from the 4 adenitis cases were unsuccessful. Congenital abnormalities such as glandular fusion (2.2%) or unilateral aplasia (0.6%) were also observed. Cysts were the most common finding (19.2%), and duct dilation was frequent (7.1%). The significance of these findings in relation to fertility is considered.  相似文献

OBJECTIVE: To evaluate age and scrotal circumference as predictors of semen quality in young beef bulls. DESIGN: Cohort study. ANIMALS: 1,173 beef bulls < 15 months old. PROCEDURE: During initial breeding soundness examination, variables for bulls producing > or = 70% morphologically normal spermatozoa were compared with those for bulls producing < 70% morphologically normal spermatozoa. Mean and 95% confidence interval for age and scrotal circumference were constructed to detect differences between groups of bulls over all observations and within the 5 most common breeds. For these 5 breeds, chi 2 analysis was used to evaluate differences in the proportion of bulls that had values less than the population mean for scrotal circumference, age, and percentage of morphologically normal spermatozoa. Multivariate regression was used to quantify variation in the proportion of morphologically normal spermatozoa that could be explained by age and scrotal circumference. RESULTS: Mean (+/- SD) age and scrotal circumference differed significantly for bulls that produced > or = 70% morphologically normal spermatozoa (12.7 +/- 1.1 months and 35.6 +/- 2.7 cm) and bulls that produced < 70% morphologically normal spermatozoa (12.1 +/- 1.2 months and 34.8 +/- 3.3 cm). The proportion of bulls younger than mean age at first examination and the proportion producing > or = 70% morphologically normal spermatozoa differed among breeds. Age and scrotal circumference explained only 11% of the variation in semen quality. CLINICAL IMPLICATIONS: Among young beef bulls, those that were older and had larger testes were more likely to produce > or = 70% morphologically normal spermatozoa. Age and scrotal circumference were not sufficient predictors of semen quality.  相似文献

Chlamydiae cause abortion and reproductive disorders in sows. Although organisms can infect the male genital tract, little is known about the disease situation in boars. Hence, we examined the prevalence of chlamydial infection in semen and genital tracts of boars. Samples collected from Swiss boars (group A: n=42), and boars from Germany (group B: n=39) were examined by bacteriology, LPS-ELISA, immunohistochemistry (IHC) and polymerase chain reaction (PCR). The latter methodology involved use of three PCR assays including 16Sig rDNA, IGS-S (intergenic spacer 16S/23S-Short) and IGS-L (intergenic spacer 16S/23S-Long) PCR for comparison methods. PCR sensitivity and the presence of potential PCR inhibitors were determined by spiking semen with Chlamydophila (Cp.) abortus DNA. Detection limits of the 16Sig and IGS-S PCR were 10 templates, while the IGS-L PCR was less sensitive (100 templates). Of 25 semen samples that were collected from group A, one semen sample was positive for Cp. psittaci and two were positive for Chlamydia-like organisms by 16Sig PCR. Screening of sera from Swiss boars revealed three animals with positive reactions in the LPS-ELISA, although we failed to detect chlamydiae within organs of these or sera-negative animals by IHC or IGS-S PCR. In group B, 10 ejaculates were positive for Chlamydia (C.) suis and two were positive for Chlamydia-like organisms by 16S PCR. The identification of DNA from Chlamydia-like organisms in semen from both groups of boars was surprising and a role for these bacteria in reproductive diseases requires further assessment. In conclusion, the prevalence of chlamydial infection was low in group A animals indicating that venereal transmission may not be significant for Chlamydia-associated reproductive diseases in pigs, although rare cases may occur.  相似文献

Dairy bulls, 322 Ayrshires (Ay) and 85 Friesians (Fr), were studied at the age of 11 months. Of the bulls, 286 Ay-bulls and 80 Fr-bulls produced semen of acceptable quality for use in A.I. Scrotal circumference, tonometer measure, scrotal fold thickness, 1-year body weight and testicular palpation were used to predict unsuitable bulls for A.I. Non-return rate was used as a measure of fertility. Scrotal fold thickness and 1-year weight had no significant correlation with fertility or semen quality. Scrotal circumference had a significant positive correlation with fertility. Tonometer ratio had a significant negative correlation with fertility. Testicular palpation was the best basis for predicting bulls with poor semen quality in this study. Twelve bulls were recorded as having testicles of different sizes, 1 testicle being more than 20% bigger than the other. Only 2 of these 12 bulls produced semen of acceptable quality. One of these 2 bulls was, after slaughter, diagnosed as having a hereditary testicle disease. Friesians were shown to have significantly higher fertility than Ayshires.  相似文献

Venereal infection of bulls with bovine herpesvirus type 1.2 (BHV-1.2) may result in acute balanoposthitis followed by the establishment of latent infection, presumably in dorsal root nerve ganglia. We herein report the characterization of the acute and latent infection of young bulls with a Brazilian BHV-1.2 isolate and the investigation of neural and non-neural sites in which viral DNA persists during latent infection, i.e. 110 days after inoculation and 50 days after experimental reactivation. Intrapreputial inoculation of BHV-1.2 isolate SV-56/90 (10(6.5)pfu per animal) resulted in severe balanoposthitis, characterized by redness of the penis and preputial mucosa, coalescent vesicles and fibrinous exsudate in all four infected bulls. Virus shedding was detected in preputial secretions and semen up to days 14 and 13 pi, respectively. Dexamethasone administration at day 60 pi led to reactivation of the infection in all animals, resulting in virus shedding in preputial secretions and/or in semen. At day 50 post-reactivation (pr), the animals were euthanized and regional tissues were collected for PCR and virus isolation. Viral DNA was consistently detected in the dorsal root ganglia of nerves genito-femoral (4/4) and obturator (4/4); frequently in the pudendal (3/4), sciatic (3/4) and rectal caudal nerve ganglia (2/3). In addition, viral DNA was detected in the pelvic sympathetic plexus of one bull and in regional lymph nodes (deep inguinal (2/4); sacral (1/4); medial iliac (1/4)) of two bulls. No infectious virus could be recovered from homogenates of DNA positive tissues, indicating the absence of actively replicating virus. These results demonstrate that BHV-1.2 DNA may persist in several sacral nerve ganglia and in regional lymph nodes as well during latent infection, i.e. 50 days after experimental reactivation. These findings may help in understanding the pathogenesis of acute and latent genital infection by BHV-1.2.  相似文献

Contents 1. 206 bulls, 13–24 months old, were tested from the A. I. centre (Spotted Mountain Cattle and German Brown Cattle). The measurements of the testes, the semen quality on the basis of 10 ejaculates of each bull, and the fertility quotient by means of the 60 to 90-Days-Non-Return results of a three months utilization (about 800 to 1000 first inseminations) were determined beside a general clinical investigation of the sexual organs. 2. From the tested bulls 12% showed a rotation of the testes about the longitudinal axis, 4,35% comparatively small testes, 2,9% an asymmetry of the testes, and 3,4% an underdevelopment of the cauda epididymidis. 3. A decrease of the semen quality and of the fertility quotient was determined in bulls with small testes, with asymmetrical testes and with an underdevelopment of the cauda epididymidis. The rotation of the testes had no influence on the fertility and quality of the sperm. 4.
