Applications of metabolomics techniques along with chemometrics provide an understanding in the relationship between metabolome of green tea and its quality. A coupled of ultra-performance liquid chromatography with time-of-flight mass spectrometry (UPLC/TOF MS) allowed a high-throughput and comprehensive analysis with minimal sample preparation. Using this technique, a wide range of metabolites were investigated. Data analysis was rapid, considering that the fingerprinting technique was performed. A set of green tea samples from 2006 tea contest of the Kansai area was analyzed to prove usefulness of the developed technique. Green tea with different qualities were discriminated through principal component analysis (PCA). Consequently, projection to latent structure by means of partial least-squares (PLS) was performed to create a constructive quality-predictive model by means of metabolic fingerprinting. Beside epigallocatechin, other predominant catechins, including epigallocatechin gallate and epicatechin gallate, detected in green tea were found to be significant biomarkers to the high quality of Japanese green tea (Sencha).  相似文献

Application of aroma extract dilution analysis using the volatile fraction of a Japanese green tea (Sen-cha) sample resulted in the detection of 36 odor-active peaks with flavor dilution (FD) factors between 10 and 5000. Thirty-six potent odorants were identified from 36 odor-active peaks by gas chromatography/mass spectrometry (GC/MS) and/or the multidimensional GC/MS (MDGC/MS) system. Among these components, 4-methoxy-2-methyl-2-butanethiol (meaty), (Z)-1, 5-octadien-3-one (metallic), 4-mercapto-4-methyl-2-pentanone (meaty), (E,E)-2,4-decadienal (fatty), beta-damascone (honey-like), beta-damascenone (honey-like), (Z)-methyl jasmonate (floral), and indole (animal-like) showed the highest FD factors. Therefore, these odorants were the most important components of the Japanese green tea odor. In addition, 4-methoxy-2-methyl-2-butanethiol, 4-mercapto-4-methyl-2-pentanone, methional, 2-ethyl-3, 5-dimethylpyrazine, (Z)-4-decenal, beta-damascone, maltol, 5-octanolide, 2-methoxy-4-vinylphenol, and 2-aminoacetophenone were newly identified compounds in the green tea.  相似文献

This study presents new sample preparation and analytical procedures for the quantification of pesticides on processed tea leaves. The new method includes tea extraction and dispersive solid phase extraction (d-SPE) to prepare gas chromatography (GC) and ultrahigh-performance liquid chromatography (UHPLC)-ready samples, providing a fast and cost-effective solution for time-sensitive industrial analysis to fulfill regulatory requirements. Both GC-negative chemical ionization mass spectrometry (GC-NCI-MS) and UHPLC-tandem mass spectrometry (UHPLC-MS/MS) were employed to produce highly sensitive and reproducible data. Excellent limits of detection (typically below 1 μg/kg for GC and 10 μg/kg for UHPLC), wide linearity ranges, and good recoveries (mostly >70%) were achieved on the selected pesticides. Twenty-seven tea samples purchased from local grocery stores were analyzed using the newly developed methods. Among the pesticides analyzed, endosulfan sulfate and kelthane were the most frequently detected by GC-NCI-MS and imidacloprid and acetamiprid by UHPLC-MS/MS in these teas. The samples were found to be relatively clean, with <1 mg/kg of total pesticide residues. The organic-labeled teas were significantly cleaner than nonorganic ones. The cost per gram of tea did not correlate with pesticide residue levels detected.  相似文献

The odorants in Chinese jasmine green tea scented with jasmine flowers (Jasminum sambac) were separated from the infusion by adsorption to Porapak Q resin. Among the 66 compounds identified by GC and GC/MS, linalool (floral), methyl anthranilate (grape-like), 4-hexanolide (sweet), 4-nonanolide (sweet), (E)-2-hexenyl hexanoate (green), and 4-hydroxy-2,5-dimethyl-3(2H)-furanone (sweet) were extracted as potent odorants by an aroma extract dilution analysis and sensory analysis. The enantiomeric ratios of linalool in jasmine tea and Jasminum sambac were determined by a chiral analysis for the first time in this study: 81.6% ee and 100% ee for the (R)-(-)-configuration, respectively. The jasmine tea flavor could be closely duplicated by a model mixture containing these six compounds on the basis of a sensory analysis. The omission of methyl anthranilate and the replacement of (R)-(-)-linalool by (S)-(+)-linalool led to great changes in the odor of the model. These two compounds were determined to be the key odorants of the jasmine tea flavor.  相似文献

为了对不同产地的晒青毛茶进行鉴别,研究了晒青毛茶高效液相色谱指纹图谱的不同识别方法。运用相关系数、夹角余弦和重叠率3种方法分别计算指纹图谱的相似度,以数字化指纹图谱为基础,对样品进行了系统聚类,分析了18个茶叶样品的主成分,并以前2个主成分作二维散点图。结果表明,3种方法均能准确地体现指纹图谱的相似程度,使毛茶与绿茶得到良好的分离;茶叶内含成分复杂,主成分较多,7个主成分累计贡献率为88.61%,系统聚类和二维排序散布图能够区分晒青毛茶和绿茶,具有简便、直观的特点。4种识别模式均能较好地对指纹图谱进行识别,为普洱茶原料鉴别和质量控制提供新的试验依据。  相似文献

A rapid and easy determination method of green tea's quality was developed by using Fourier transform near-infrared (FT-NIR) reflectance spectroscopy and metabolomics techniques. The method is applied to an online measurement and an online prediction of green tea's quality. FT-NIR was employed to measure green tea metabolites' alteration affected by green tea varieties and manufacturing processes. A set of ranked green tea samples from a Japanese commercial tea contest was analyzed to create a reliable quality-prediction model. As multivariate analyses, principal component analysis (PCA) and partial least-squares projections to latent structures (PLS) were used. It was indicated that the wavenumber region from 5500 to 5200 cm(-1) had high correlation with the quality of the tea. In this study, a reliable quality-prediction model of green tea has been achieved.  相似文献

In recent years the consumption of tea and herbal infusions has increased. These hot drinks are consumed as daily drinks as well as for medicinal purposes. All tea varieties (white, yellow, green, oolong, black and puerh) originate from the leaves of the tea plant, Camellia sinensis. All extracts made of plant or herbal materials which do not contain Camellia sinensis are referred as herbal infusions or tisanes. During processing and manufacturing fungal contamination of the plant materials is possible, enabling contamination of these products with mycotoxins. In this study a multimycotoxin UPLC-MS/MS method was developed and validated for the analysis of the raw tea and herbal infusion materials as well as for their drinkable products. The samples were analyzed by ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), with a mobile phase consisting of variable mixtures of water and methanol with 0.3% formic acid. The limits of detection for the different mycotoxins varied between 2.1 μg/kg and 121 μg/kg for raw materials and between 0.4 μg/L and 46 μg/L for drinkable products. Afterward 91 different tea and herbal infusion samples were analyzed. Only in one sample, Ceylon melange, 76 μg/kg fumonisin B(1) was detected. No mycotoxins were detected in the drinkable products.  相似文献

An innovative technique for green tea's quality determination was developed by means of metabolomics. Gas-chromatography coupled with time-of-flight mass spectrometry and multivariate data analysis was employed to evaluate the quality of green tea. Alteration of green tea varieties and manufacturing processes effects a variation in green tea metabolites, which leads to a classification of the green tea's grade. Therefore, metabolic fingerprinting of green tea samples of different qualities was studied. A set of ranked green tea samples from a Japanese commercial tea contest was analyzed with the aim of creating a reliable quality-prediction model. Several multivariate algorithms were performed. Among those, the partial least-squares projections to latent structures (PLS) analysis with the spectral filtering technique, orthogonal signal correction (OCS), was found to be the most practical approach. In addition, metabolites that play an important role in green tea's grade classification were identified.  相似文献

不同茶树品种(系)的绿茶滋味分析及评价模型构建   总被引:1,自引:3,他引:1  
为客观准确地评价不同茶树品种(系)绿茶的滋味品质,该研究系统分析了25个茶树品种(系)绿茶的主要滋味成分含量及其Dot值,利用主成分分析法对不同茶树品种(系)绿茶滋味品质进行综合评价并建立滋味品质评价模型。结果表明:儿茶素、咖啡碱是不同茶树品种(系)绿茶苦涩味强度差异的主要原因,其中表没食子儿茶素没食子酸酯(Epigallocatechingallate,EGCG)是所测样品中涩味的主要贡献物质,EGCG和咖啡碱为所测样品中苦味的主要贡献物质。氨基酸是不同茶树品种(系)绿茶鲜味、甜味差异主要因素,谷氨酸是所测样品中鲜味的主要贡献物质。主成分分析表明:前5个主成分的累计方差贡献率为79.018%,并以前5个主成分的线性回归方程和贡献率构建了滋味品质评价模型,模型评价结果与感官审评结果较为相似,存在极显著相关性(P0.01),相关系数为0.791;利用模型中各主成分的得分,能够较好的区分所测样品的苦、涩、鲜、爽等滋味特征。因此,该研究所建模型能够较好的评价所测样品的滋味品质及其主要滋味特征的差异,为各茶树品种(系)的推广应用提供理论依据,也为不同茶树品种(系)绿茶滋味品质的科学评价提供新的思路和方法。  相似文献

Components of green tea ( Camellia sinensis) have been of considerable interest in recent years because of their potential utility as pharmaceutical agents, particularly for their antioxidant and anticarcinogenic activity. Responding to the increasing scientific validation of numerous health benefits of tea, a comprehensive approach was adopted to carry out analysis for the quality assessment of flavonoids in tea samples of different origins. For this purpose, extraction, separation, and mass spectrometric parameters were optimized. Extraction methods evaluated include reflux extraction, a modified accelerated solvent extraction (ASE), namely, Aquasolv extraction, and microwave-assisted extraction (MAE) using different percentages of solvents. Separation was performed by a specifically developed reversed phase high-performance liquid chromatography (RP-HPLC) method using different C18 and C8 stationary phases. Optimization of extraction techniques clearly proved the performance of MAE, which delivered highest yields in a very short time. Additionally, the comparison with Aquasolv extraction provided new insights, as variations in quantified amounts of target compounds between the extracts could be explained on the basis of thermal degradation and epimerization phenomena. Especially the epimerization phenomenon for catechin/epicatechin oligomers, that is, of procyanidins P 2 and P 3, was observed for the first time. Finally, an optimized extraction and separation system was used for qualitative and quantitative investigations of compounds from different green tea samples from Ceylon (cultivated under biologically controlled conditions), Japan, India, and China as well as from one black tea sample from India.  相似文献

Quantitative methods are detailed for determination of ethyl carbamate in distilled alcoholic beverages by capillary gas chromatography with flame ionization detection (GC/FID) and by packed-column gas chromatography/mass spectrometry (GC/MS) using selected ion monitoring. Five g samples of distillate of known ethanol concentration are diluted with water to 25% ethanol (v/v), washed with petroleum ether, and extracted with dichloromethane prior to GC/FID or GC/MS analysis. As necessary, sample extracts that exhibit GC/FID interference are passed through alumina for additional cleanup. When internal standards (tert-butyl carbamate and n-butyl carbamate for GC/FID, or ethyl 13C-15N-carbamate for GC/MS) were used for quantitation, the limit of detection for ethyl carbamate was in the range of 5-25 ppb. Coefficients of variation ranged from 3.5 to 6.0% for GC/FID determinations, and from 1.4 to 3.2% for GC/MS. Correlation between methods for 22 random distillate samples ranging in concentration from approximately 40 to 800 ppb gave a correlation coefficient (r) of 0.996.  相似文献

在眉茶拼配过程中,为了客观定量的评价试拼小样与标准样之间的相似性,该研究提出了一种基于深度度量学习的相似性评价方法,采用7种等级的眉茶标准样作为训练集,并在标准样中加入不同含量半成品茶构建具有不同相似性的测试集。采集茶样的高光谱数据并获取光谱特征与图像特征,分别以光谱数据、图像数据、图谱融合数据3种数据类型作为模型的输入。为了构建距离特征空间,该研究提出了基于三元组损失的深度特征提取网络,并设计了Center Anchor Triplet Loss损失函数,通过样本在特征空间的距离,表征相似程度,达到定性判断相似性和定量度量相似度的目的。结果表明:图谱融合数据结合Center Anchor Triplet Loss的方法精度最高,相似性判断准确率为98.89%,相似度度量准确率为100%。该研究采用未经训练的独立样本评价模型,可以获得较好的结果,说明算法具有较好的泛化能力。研究结果为眉茶的相似性评价提供了理论依据。  相似文献

Protonated water molecules generated by an ambient corona discharge were directed to impact tea leaves for desorption/ionization at atmospheric pressure. Thus, a novel method based on surface desorption chemical ionization mass spectrometry (DAPCI-MS) has been developed for rapid analysis of tea products without any sample pretreatment. Under the optimized experimental conditions, DAPCI MS spectra of various tea samples are recorded rapidly, and the resulting mass spectra are chemical fingerprints that characterize the tea samples. On the basis of the mass spectral fingerprints, 40 tea samples including green tea, oolong tea, and jasmine tea were successfully differentiated by principal component analysis (PCA) of the mass spectral raw data. The PCA results were also validated with cluster analysis and supervised PCA analysis. The alteration of signal intensity caused by rough surfaces of tea leaves did not cause failure in the separation of the tea products. The experimental findings show that DAPCI-MS creates ions of both volatile and nonvolatile compounds in tea products at atmospheric pressure, providing a practical and convenient tool for high-throughput differentiation of tea products.  相似文献

茶叶含水率是影响茶叶加工品质的一项重要指标。为了实现茶叶加工中含水率的快速检测,该文提出了一种应用漫反射光谱技术的绿茶初制过程中含水率无损检测方法。采用波长范围在325~1 075 nm 的可见-短波近红外光谱仪,对炒青绿茶在8个加工工序中随机抽取的568个茶叶样本进行漫反射光谱扫描,光谱扫描后立即测量样本的含水率。对于得到的光谱数据,采用小波变换降低其信息维度并提取小波系数,比较小波低频系数对于光谱特征信息的提取能力,结果显示,小波低频系数能够有效提取原始光谱数据中的特征信息。采用3种回归算法:偏最小二乘回归、神经网络和支持向量机分别建立含水率的测量模型。比较发现支持向量机回归模型的结果最优,建模相关系数为0.9985,预测相关系数为0.9875。研究结果表明,漫反射光谱可以用于绿茶含水率的无损、快速检测,小波变换是一种有效的光谱特征提取算法,而且支持向量机回归算法具有高精度和强泛化能力,可广泛用于回归分析。  相似文献

Phenolic acids are aromatic secondary plant metabolites, widely spread throughout the plant kingdom. Existing analytical methods for phenolic acids originated from interest in their biological roles as secondary metabolites and from their roles in food quality and their organoleptic properties. Recent interest in phenolic acids stems from their potential protective role, through ingestion of fruits and vegetables, against oxidative damage diseases (coronary heart disease, stroke, and cancers). High performance liquid chromatography (HPLC) as well as gas chromatography (GC) are the two separation techniques reviewed. Extraction from plant matrixes and cleavage reactions through hydrolysis (acidic, basic, and enzymatic) are discussed as are the derivatization reagents used in sample preparation for GC. Detection systems discussed include UV-Vis spectroscopy, mass spectrometry, electrochemical, and fluorometric detection. The most common tandem techniques are HPLC/UV and GC/MS, yet LC/MS is becoming more common. The masses and MS fragmentation patterns of phenolic acids are discussed and tabulated as are the UV absorption maxima.  相似文献

A method has been developed for the determination of ethanol in canned salmon using automated headspace sampling in conjunction with analysis by gas chromatography. The thermal process for the commercial sterilization of canned salmon is shown to provide an effective extraction of the ethanol so that the fluid removed from the can may be used as the analytical sample with minimal preparation prior to analysis. Ethanol content is measured directly, without the need for an internal standard, by either GC/MS or GC/FID. The headspace autoanalyzer allows for a rapid determination of ethanol with greater reproducibility than could be obtained with manual injection systems. The GC/MS technique can also provide an advantage in that simultaneous single ion monitoring of the two major ethanol ions provides additional protection from interferences. To assess the applicability of this technique to other substrates, Atlantic sea scallop meats were also successfully analyzed by this technique.  相似文献

A set of 191 green teas from different countries was collected and analyzed by (1)H NMR. It was proposed to establish if the teas could be discriminated according to the country of origin or with respect to quality. Both principal component analysis (PCA) and cluster analysis were applied to the data. Some separation of Chinese and non-Chinese teas was observed. The present results did not allow allocation of samples to individual countries, but cluster analysis suggested that it might be possible with an augmented sample set. The PCA did show a separation between the Longjing type (highest quality Chinese tea) and most other Chinese teas and indicated some metabolites that could be responsible for the difference. Longjing teas showed higher levels of theanine, gallic acid, caffeine, epigallocatechin gallate, and epicatechin gallate and lower levels of epigallocatechin when compared with other teas. These compounds have been mentioned previously in connection with quality, but it was also shown that higher levels of theogallin (5-galloyl quinic acid), theobromine, 2-O-(beta-l-arabinopyranosyl)-myo-inositol and some minor sugar-containing compounds were found in Longjing teas while higher levels of fatty acids and sucrose were found in the other teas. These new markers could prove to be useful for the authentication of bulk tea.  相似文献

自动化加工生产线改善机采绿茶理化品质研究   总被引:2,自引:1,他引:1  
以茶树机采鲜叶为原料,选配、改装和组建了茶叶加工自动化生产线,并跟踪加工工序中茶坯理化成分的动态变化,以同一原料所制的传统炒青工艺为对照,比较新设备和新工艺对机采卷曲形绿茶色泽和品质成分的影响。结果发现:新的生产线在杀青、二青、做形和提香等关键工序中,集成应用了电磁加热耦合热风、热管余热回收和流化床干燥技术,提高了茶叶的滋味和色泽品质,所制卷曲形绿茶的感官得分比传统炒青绿茶高2分,酚氨比相对较低(P0.05),干茶的色相值、茶汤、叶底的亮度和色相值相对更好(P0.01);相比传统单机加工设备,该生产线可日均生产绿茶1 750 kg,产能提高了37.5%,能耗成本为4.6~4.8元/kg干茶;热效率提高100%,用工成本减少50%。机采卷曲形绿茶自动化生产线扩大了茶叶加工产能,减轻了劳动强度,提高了卷曲形绿茶品质,为实际生产提供了指导。  相似文献

Classification of tea quality is now mainly performed according to the sensory results by professional tea tasters. However, this evaluation method is inconsistent in differentiating their qualities. A combination of a (1)H NMR technique and a multivariate analysis was introduced to the quality evaluation of green tea by means of a metabolomic technique. A broad range of metabolites were detected by (1)H NMR spectrometry. The principal component analysis (PCA) was used to reduce the complexity of the (1)H NMR spectra data set and provided the quality discrimination result. It offered an extensive clue for classification and quality assessment without any prepurification method. A set of green teas from a Japanese tea contest were analyzed by (1)H NMR to classify the quality with respect to that judged by tea tasters and to conceive a quality prediction model. Metabolic profiling and fingerprinting of (1)H NMR spectra of green teas with different quality were studied. PCA showed a separation between the high- and the low-quality green teas. The taste marker compounds contributing to the discrimination of tea quality were identified. Reliable prediction models were obtained by the partial least-squares projection to latent structure (PLS) analysis together with a preprocessing filter of both orthogonal signal correction (OSC) and a combination between OSC and wavelet transform algorithms.  相似文献
