Potato seed tubers are imported annually to Israel from northern Europe. Although the seed is registered as certified, a survey carried out over a 9-year period indicated that most lots were affected by latent or active bacterial and fungal infections. Latent infection byErwinia carotovora subsp.atroseptica, the causal agent of blackleg, at a level of 103 cells/g peel, was present in 30% of the lots in most years. Black scurf caused byRhizoctonia solani was present in 20–70% of the imported lots, with a moderate to high level of infection in all years except 1985. In contrast, although many lots were affected by powdery scab, common scab, and Fusarium dry rot in most years, disease incidence within lots was generally low. The gangrene pathogen (Phoma exigua) was rarely detected. The survey findings are of marked importance, due to the extensive use of soil fumigation in Israeli agriculture.  相似文献

Phosphites (Phi) are alkali metal salts of phosphorous acid, with the ability to protect plants against different pathogens. In this research, the effect of Phi applied to potato plants on severity of three important potato diseases in Argentina was assessed. Seed tubers and foliage of potato cvs Shepody and Kennebec were treated with Phi to assess effects on resistance against Phytophthora infestans, Fusarium solani and Rhizoctonia solani. Protection resulting from Phi treatment in seed tubers was high against P. infestans, intermediate against F. solani, and low against R. solani. In addition, seed tubers treated with calcium or potassium phosphites (CaPhi and KPhi, respectively) at 1% of commercial product emerged earlier than untreated ones. When Phi were foliarly applied two or four times at different doses, high levels of protection against P. infestans were achieved in both cultivars. Higher protection was observed in Kennebec when CaPhi was applied, while in Shepody this was true for KPhi. Expression of β-1,3-glucanases was induced at different times after treatment but no correlation between β-1,3-glucanases expression and foliar protection level was found. On the other hand, Phi positive protection effects did not produce negative effects in plant growth. Leaves from CaPhi-treated plants showed a darker green colour than leaves from control plants; also an increase in Rubisco protein and a delay in crop senescence was observed.  相似文献

Inoculation of seed potatoes with the mycoparasiteVerticillium biguttatum, isolate M73 (combined withGliocladium roseum in 1981, either alone or mixed with isolate M180 plus antibiotics-producing isolates ofAzotobacter chroococcum in 1982) repeatedly proved successful in reducingRhizoctonia solani on stolons and stems. In field experiments, this ultimately led to a reduced formation of sclerotia on new tubers, particularly in neutral sandy loam and clay loam soils. In 1981 inoculation with antagonists led, when compared with no inoculation, to average reductions of 22 and 42% for the harvest from clean, and 15 and 26% for the harvest from infected seed tubers grown on slightly acid sandy soils and on neutral loam soils, respectively. The harvest from clean, inoculated seed tubers had the lowest sclerotium index. In 1982 inoculation of seed tubers planted in slightly acid sandy soils gave reductions of the sclerotium index of up to 22%. In the neutral marine loam soils considerable reductions were often achieved, viz., in slightly infected loams 51–68% and in rather heavily infected ones 4–43%. Chemical disinfection of seed tubers proved effective only in loam soils that were slightly infested withR. solani. In both years inoculation of seed tubers with antagonists led to significantly lower sclerotium indices of the harvest (p=0.1% in 1981; p=5% in 1982). V. biguttatum was present more frequently and in greater densities on stems and stolons of plants from inoculated than from non-inoculated seed tubers. The latter were colonized by wildV. biguttatum strains from the soil, apparently less effective antagonists.Early in the season, the soil temperature was too low for growth ofV. biguttatum. Suspected Dickeya sp. strains were obtained from potato plants and tubers collected from commercial plots. The disease was observed on crops of various cultivars grown from seed tubers imported from the Netherlands during the spring seasons of 2004–2006, with disease incidence of 2–30% (10% in average). In addition to typical wilting symptoms on the foliage, in cases of severe infection, progeny tubers were rotten in the soil. Six strains were characterised by biochemical, serological and PCR-amplification. All tests verified the strains as Dickeya sp. The rep-PCR and the biochemical assays showed that the strains isolated from blackleg diseased plants in Israel were very similar, if not identical to strains isolated from Dutch seed potatoes, suggesting that the infection in Israel originated from the Dutch seed. The strains were distantly related to D. dianthicola strains, typically found in potatoes in Western Europe, and were similar to biovar 3 D. dadanti or D. zeae. This is the first time that the presence of biovar 3 strains in potato in the Netherlands is described. One of the strains was used for pathogenicity assays on potato cvs Nicola and Mondial. Symptoms appeared 2 to 3 days after stem inoculation, and 7 to 10 days after soil inoculation. The control plants treated with water, or plants inoculated with Pectobacterium carotovorum, did not develop any symptoms with either method of inoculation. The identity of Dickeya sp. and P. carotovorum re-isolated from inoculated plants was confirmed by PCR and ELISA.  相似文献

A collection of 241 isolates of Rhizoctonia solani obtained from potato plants grown in different areas in France was characterized for anastomosis grouping, symptomatology on tubers of different cultivars and sensitivity to three fungicides. Most isolates collected belonged to (anastomosis groups (AGs)) AG 3, but 2% and 4% of the isolates were AG 5 and AG 2-1. AG 3 and AG 2-1 isolates were mostly obtained from sclerotia on tubers, but all AG 5, some AG 3 and some AG 2-1 isolates were recovered from superficial tuber alterations, like deformations, corky or scabby lesions. Sclerotia were formed on tubers produced by healthy stem cuttings grown in soil artificially infested with AG 3, but not on tubers grown in soil infested with either AG 5 or AG 2-1. No variation in susceptibility to sclerotial formation was observed among five potato cultivars. In all cases, a large proportion of tubers showed superficial corky lesions, often associated with deformations. The proportion of tubers with lesions and deformations was highest in soil infested with AG 2-1 and significantly lower on cv. Samba in all treatments. All isolates were highly sensitive to flutolanil, iprodione and pencycuron, except the AG 5 isolates, moderately sensitive to pencycuron. These results show that, although AG 3 is the most common R. solani group on potato in France, AG 5 and AG 2-1 may be present. Isolates differed for pathogenicity. In vitro sensitivity to fungicides varied among AGs.  相似文献

A total of 109 samples of potato seed tubers imported to Jordan from France, Netherlands and Denmark during the 2007/2008 growing seasons were surveyed for 10 different diseases caused by fungi and fungus‐like organisms to determine the prevalence of different tuberborne diseases and to evaluate the efficacy of the current visual examination procedures in detecting different potato diseases. The results demonstrated that most potato seed tuber lots imported to Jordan were infected with one or more of the following potato pests: Colletotrichum coccodes (black dot) (66.0%), Rhizoctonia solani (black scurf) (42.4%), Helminthosporium solani (silver scurf) (92.6%), and Spongospora subterranea (powdery scab) (13.8%). Just over a quarter (25.7%) of all lots examined in this study were found to exceed the acceptance limits for one or more potato diseases according to the Jordanian standards. This study suggests that the current visual examination procedure is not adequate to detect all tuberborne diseases of potato and to estimate their incidence and severity precisely.  相似文献

A total of 304Rhizoctonia solani isolates and 60 binucleateRhizoctonia-like fungi were recovered from stems and tubers of infected potato plants over a 2-yr period in northeast Turkey.R. solani isolates were identified to 11 anastomosis groups (AGs): AG-1 (0.66%), AG-2-1 (5.6%), AG-2-2 (0.99%), AG-3 (83.9%), AG-5 (4.6%), AG-6 (0.66%), AG-8 (1.32%), AG-9 (0.33%), AG-10 (1.32%), AG-12 (0.33%), and AG-13 (0.33%). In the greenhouse tests, most of the AG-3 isolates were significantly more virulent than isolates belonging to other AGs on potato cv. Batum. Isolates of other anastomosis groups differed in their virulence. Results indicated that AG-3 is an important pathogen on potatoes grown in the study area. Passalora blight of anise, caused byPassalora malkoffii (Bubák) U. Braun, is an important disease of anise in Turkey. The disease affects all the aboveground parts of plants including flower clusters. Infected seeds have dark, linear stromata. Detection of the pathogen on seeds was studied by the blotter method, agar method, washing test and sowing infected seeds in disease-free soils. The pathogen was recovered only by the washing test and to a limited extent by water agar + seed decoction agar. Sixteen of 24 seed samples from diseased regions were found to be infected. The pathogen was not detected by any other methods. However, several indigenous fungi,e.g. Alternaria alternata, were isolated, which may have prevented the growth of the pathogen. Seed washings of infected seed samples had typical spores of the pathogen up to 106 conidia per gram of seed. Transmission of the pathogen was shown by sowing infected seeds in disease-free field soils in two locations where anise had not been grown previously. Azoxystrobin, chlorothalonil + carbendazim and flutriafol seed treatments at 0.04 g a.i. kg−1 seed, 1.0 g + 4.5 g a.i. kg−1 seed and 0.015 g a.i. kg−1 seed reduced the disease by 92.5%, 89.6% and 36.2% in 2002 and by 78.9%, 75.8% and 41.2% in 2003, respectively. Three foliar applications of axosystrobin, chlorothalonil + carbendazim and flutriafol at the rates of 187.5 g a.i. ha−1, 1500 g + 6750 g a.i. ha−1 and 31.3 g a.i ha−1 reduced disease incidence by 92.5%, 86.0% and 96.8% in 2002 and by 97.5%, 90.8% and 97.0% in 2003, respectively. http://www.phytoparasitica.org posting May 17, 2005.  相似文献

Infection with potato virus S Andean (PVSA) and ordinary (PVSo) strains was found in potato breeder's selection No. 8163-511 imported from West Germany; the two PVS strains were differentiated by their reactions on Chenopodium quinoa Tests on potato leaf samples using enzyme-linked immunosorbent assay followed by inoculation to C quinoa were subsequently used to detect PVSA and PVSo in a large-scale survey of imported and domestic potato material. PVSA was detected in breeders' selections and cultivars imported from the Netherlands and West Germany, but not in domestic certified seed potato stocks or farmers' once-grown stocks. PVSo was found in both imported and domestic certified stocks, but infection was commoner in the imported ones. When plants of C. quinoa, C. amaranticolor, C. murale and Nicotiana debneyi were inoculated with four isolates of PVSA, one induced mild symptoms while the reactions of the others ranged from moderate to severe. When plants of different potato cultivars were inoculated with three isolates, the plants were mostly infected without symptoms. However, when tubers from some were grown on, the progeny plants of most of the different combinations of cultivar and isolate of PVSA developed one or more of the following symptoms: vein deepening, rugosity, interveinal chlorosis, premature senescence and early loss of lower leaves.  相似文献

Data from surveys of winter wheat fields in the period 1974–1986 and of seed lots in the period 1962–1986 and identifications of diseases on plant samples were compiled to describe the occurrence of snow mould (Monographella nivalis) andFusarium spp. On average,M. nivalis dominated overFusarium spp. The complex ofFusarium spp. constituted mainly ofF. culmorum, followed byF. avenaceum andF. graminearum. M. nivalis was dominant in May on stem-bases and in July on leaves and leaf sheaths. On seedsM. nivalis predominated only in years with low temperatures in July and August.Average brown footrot infection in the field was 4% tillers in May and 5% culms in July. Brown footrot intensity in July was high in cropping seasons with high precipitation in October and with low temperatures in October, November and December. In July during the early eighties, an average of 8% of leaves and 6% of flag leaf sheaths were infected byM. nivalis. Average ear blight incidence was 1.2% glumes infected. Seed contamination by these pathogens averaged 16% in the years 1962–1986. The contamination was high in years with high precipitation in June, July and August. Plug seedlings, widely used in cabbage cultivation in Japan, are often infected by seed-borne pathogens, especially the serious pathogen Alternaria brassicicola. Because information on seed infestation is scant in Japan, we investigated fungal infestation in commercial batches of cabbage seeds produced between 1984 and 2001. A total of 123 lots were divided into six groups by production period (1984–1989, 1994–1998, and 2001) and by use or nonuse of fungicide. One hundred seeds from each lot were incubated separately on agar at 25°C to isolate the predominant fungus. Alternaria brassicicola was isolated most frequently, 0%–94% of the seeds depending on seed lot or 6%–21% of the seeds grouped by production period and fungicide treatment. Thus, the pathogen was isolated even from seeds refrigerated for 17 years. Alternaria brassicicola accounted for 57%–95% of all isolated fungi by the group and was higher on older or fungicide-treated seeds. Seeds that were not treated with fungicide in lots grouped by production districts in western Japan were infested with A. brassicicola at a rate of over 12%, higher than that in the eastern region (<4%). Infestation was higher in the warmer areas of Japan. Eighty-five isolates, other than A. brassicicola, produced spots on cabbage cotyledons, although they were not isolated as frequently: less than 5% of seeds by group separated by production period and fungicide treatment. Most of these isolates were Alternaria alternata. This is the first report on the frequency of fungal infestation of commercial cabbage seeds in Japan.  相似文献

The transmission of Stagonospora nodorum from four naturally infected winter wheat seedlots was quantified in controlled environment germination chambers at 9, 13, 17, 21, and 25 °C. Seedlings were harvested when the second leaf began to emerge. Coleoptiles and first seedling leaves were examined for the presence of lesions caused by S. nodorum. First leaves were incubated on Bannon's medium for 2 weeks, after which they were examined for pycnidia of S. nodorum. Transmission to the coleoptile occurred at all temperatures, but decreased from 100% to 72% as temperature increased from 9 to 25 °C. Transmission to the first leaf was less, dropping from 37% to 2% as temperature increased from 9 to 25 °C. At least 44% of infected first leaves were symptomless at all temperatures, with 96% of infected leaves showing no symptoms at 25 °C. Transmission to seedling leaves occurred over a broad temperature range. Under the high densities at which wheat is sown, a significant number of infected seedlings per unit area may originate from relatively low initial seed infection levels and transmission efficiencies.  相似文献

Dickeya strains isolated in Israel in 2006–2010 were characterized by dnaX sequence analysis, pulsed‐field gel electrophoresis (PFGE), biochemical assays and pectolytic activity, and found to be homogeneous: most of them could be classified as ‘Dickeya solani’. Of the 34 strains isolated from imported seed tubers or potato plants grown from imported seed, 32 were typed as ‘D. solani’ and only two were characterized as Dickeya dianthicola. Biovar typing indicated that all ‘D. solani’ strains were biovar 3. ‘Dickeya solani’ strains were most closely related to Dickeya dadantii subsp. dieffenbachiae according to PFGE and dnaX analyses and both species exhibited high pectolytic activity. Expression levels of two putative virulence genes, pelL (encoding a pectic enzyme) and dspE (encoding a type III effector) were significantly induced in ‘D. solani’ strains isolated from potato plants or tubers grown in hot climates such as the Negev region in Israel, compared to those isolated from seed tubers imported from the Netherlands, France or Germany. Results of this study support the hypothesis that ‘D. solani’ strains isolated in Israel are also clonal; however, they appear to be more virulent than strains isolated in Europe.  相似文献

A real-time PCR assay was designed to quantify seed-borne infection of Pyrenophora graminea in barley (Hordeum vulgare). Conventional tests such as the freezing blotter method cannot distinguish P. graminea from the closely related P. teres. The seed infection threshold for P. graminea is lower than the one for P. teres and is therefore applied for both species although P. graminea may be absent. This results in unnecessary rejections of seed lots. PCR primers and a TaqMan probe were designed to target a P. graminea-specific DNA sequence. The potential of the real-time PCR assay for quantifying seed-borne infection of P. graminea was investigated by examining seed lots harvested from P. graminea-infected fields. The major part (84%) of the variation in the amount of P. graminea DNA measured by real-time PCR could be attributed to variation between seed lots while only about 8% was due to variation within seed lots. DNA quantities of P. graminea were positively correlated with seed infection incidence detected by the freezing blotter method as well as with the infection incidence of plants examined in the greenhouse. Both correlations were highly significant (P < 0.001) but the DNA quantities accounted only for 59% (R 2 = 0.59) and 56% (R 2 = 0.56), respectively, of the variation in the results obtained by the two conventional methods. Seed lots of varieties resistant to P. graminea contained considerable amounts of P. graminea DNA but showed no or only few leaf symptoms in the greenhouse test suggesting that the recommended seed infection thresholds could be raised for resistant varieties.  相似文献

Potato mop‐top virus (PMTV), the cause of spraing in potato tubers, is transmitted by Spongospora subterranea, the cause of powdery scab, and by planting infected seed tubers. This study was undertaken to determine the relative importance of these sources of infection in seed potato production in Scotland. The transmission of PMTV from tested seed tubers to daughter plants was examined over 2 years and six cultivars. The development of foliar symptoms varied with year and cultivar. Infection of daughter tubers derived from PMTV‐infected seed tubers was more prevalent on plants affected by foliar symptoms than those without symptoms. The rate of transmission of PMTV from infected seed tubers to daughter tubers ranged from 18 to 54%. Transmission was affected by cultivar and by origin of seed tubers used for a cultivar, but not by a cultivar's sensitivity to PMTV infection. The incidence of PMTV in daughter tubers of cv. Cara grown from seed potatoes from one source (common origin) by more than 25 seed producers was examined over two successive generations. The incidence of PMTV in daughter tubers was not correlated with that in the seed tubers but appeared to be strongly associated with soil inoculum. The incidence of PMTV was correlated with powdery scab in those crops in which both were present. There was some evidence from soil tests conducted in 2006 using a tomato bait plant and real‐time RT‐PCR that planting PMTV‐infected seed potatoes could increase the risk of introducing the virus into land not infested by PMTV.  相似文献

An indexing system for detectingTomato mosaic virus (ToMV) in commercial tomato seed lots is described. Factors associated with the procedure were analyzed and the following standard two-step working scheme is proposed: (i) mass screening by ELISA for the presence of the virus; (ii) evaluation of virus infectivity within the infested seed lots. A threshold of 10 ng ml−1 was determined for detection of purified ToMV by either ELISA or plant inoculation.Nicotiana tabacum cv. Xanthi NN was found to be a highly sensitive local lesion assay plant for the detection of ToMV. Seed transmission ofPyrenophora tritici-repentis, a common foliar pathogen of wheat, was investigated in soft white winter wheat cv. Frankenmuth and found to be non-systemic; the emerging coleoptile was infected externally by hyphal growth from the infected pericarp. Hyphae from the infected coleoptile then infected the first and second seedling leaves as they emerged. Coleoptile symptoms ranged from tiny brown streaks or spots to large, brown necrotic areas accompanied by cracking and distortion of the coleoptile. Small brown spots sometimes occurred on the first and rarely on the second seedling leaves, often accompanied by leaf distortion. Pseudothecial initials of the fungus were present within or on the seed remnants. Seed transmission efficiency was as high as 92%in vitro and 60% in potting soil outdoors. Seed infection did not affect germinationin vitro, but slightly reduced emergence in potting soil. Seedling weight and height were reduced significantly. In potting soil, seed infection also resulted in delayed plant growth and increased tan spot severity at later stages of plant development. Under controlled conditions, seed transmission efficiency and incidence of pseudothecia on seed were negatively correlated with seed germination temperature in the range of 9 to 21 °C, whereas frequency of recovery of the fungus from symptomatic coleoptiles and leaves was positively correlated with seed germination temperature.These results suggest that infected seed may serve as a source of inoculum for tan spot epidemics and for dispersal of strains of the fungus to new areas.  相似文献

In greenhouse experiments factors which are involved in the stimulation of sclerotia formation byRhizoctonia solani Kühn on potato tubers after haulm destruction were investigated. Cutting off the shoot stimulated the production of sclerotia as much as chemical haulm destruction. This was also observed when potato tubers were grown away from the roots in a separate compartment filled with steamed perlite. Fewer sclerotia were produced when roots were cut in addition to haulm destruction compared to haulm destruction alone. Cutting off the roots alone often stimulated sclerotia production. The data indicate that sclerotia production was directly affected by the tuber and probably due to physiological changes in the tuber caused by elimination of the shoot.Samenvatting In kasproeven werd onderzocht welke factoren betrokken zijn bij de stimulering van de produktie van sclerotiën op aardappel doorRhizoctonia solani Kühn als gevolg van loofvernietiging. Doodspuiten plus doorsnijden van de wortels resulteerde in minder lakschurft dan doodspuiten alleen. Wanneer alleen de wortels werden doorgesneden resulteerde dat vaak in meer lakschurft. Routine laboratory testing of 710 bean seed lots from various origins forPseudomonas syringae pv. phaseolicola (Psp) with immunofluorescence microscopy (IF) showed that 27.5% of the seed lots (five subsamples of 1000 seeds tested per sample) contained two or more IF-positive cells in a total of 500 microscope fields (magnification 500×). Simultaneously performed dilution-platings of IF-positive subsamples on King's medium B confirmed presence of Psp for one-third of these IF-positive seed lots. The grey area of disagreement between both laboratory tests was studied by comparison of test data and by field trials.The number of IF-positive cells per subsample was positively correlated with isolation and identification of Psp (R=0.85). The detection level of IF was ca. 102 Psp cells per ml of undiluted subsample extract. The detection level of Psp by isolation on King's medium B was variable, being inversely related with the saprophyte to Psp ratio. The high sensitivity of IF was in part due to high percentages of dormant or dead IF-positive cells in the sample extract. Field trials over two years with 10 000 seeds per seed lot, showed disease incidence for 9 of the 22 seed lots. Of ten IF-positive lots with five positive subsamples per sample, nine were positive in the field test plot (the negative lot gave primary infection spots of Psp when used for commercial growing). By isolation, seven of these ten IF-positive lots were positive. Of the five IF-positive lots with two or less positive subsamples, isolation and field trial were both negative. Based on data on seed transmission from literature, field incidence was unlikely for these five samples in a 10 000 seeds field trial. All seven IF-negative lots were negative in the field trial. Eremothecium ashbyi and E. coryli were present in the maxillary and mandibular stylet pouches of the contaminated heads of the true bug Riptortus pedestris collected during a field survey, 2007. Ascospores of E. ashbyi and E. coryli were taken up in the stylet pouch when R. pedestris fed on soybean seeds infected with the respective fungus. In the case of E. ashbyi, the ascospores had subsequently germinated and were recognized as large masses of mycelia in the stylet pouch. In contrast, masses of E. coryli including buds of irregular size and shape were recognized about 3 days after. These results proved that these yeasts are taken up and become lodged in the stylet pouch when the insect feeds on infected host plants. In stylet sheaths stained with erythrosine, E. ashbyi was found as fragments of mycelia and E. coryli was found as small buds. These observations yield evidence to confirm that E. ashbyi and E. coryli are transmitted as fragments of mycelia and as small buds, respectively, after the insect has fed on infected host plants.  相似文献
