The effects of alternative corn wet‐milling (intermittent milling and dynamic steeping (IMDS), gaseous SO2 and alkali wet‐milling) and dry grind ethanol (quick germ and quick fiber with chemicals) production technologies were evaluated on the yield and phytosterol composition (ferulate phytosterol esters, free phytosterols, and fatty acyl phytosterol esters) of corn germ and fiber oil and compared with the conventional wet‐milling process. Small but statistically significant effects were observed on the yield and composition of corn germ and fiber oil with these alternative milling technologies. The results showed that the germ and fiber fractions from two of the alternative wet‐milling technologies (the gaseous SO2 and the IMDS) had, for almost all of the individual phytosterol compounds, either comparable or signficantly higher yields compared with the conventional wet‐milling process. Also, both of the modified dry grind ethanol processes (the quick germ and quick fiber) with chemicals (SO2 and lactic acid) can be used as a new source of corn germ and fiber and can produce oils with high yields of phytosterols. The alkali wet‐milling process showed significantly lower yields of phytosterols compounds in germ but showed significantly higher yield of free phytosterols, fatty acyl phytosterol esters and total phytosterols in the fiber fraction.  相似文献

Corn fiber gum (CFG) is a hemicellulose (arabinoxylan)-enriched fraction obtained by the extraction of corn bran/fiber using a proprietary alkaline hydrogen peroxide process. When purified CFG prepared by this process was hydrolyzed with more concentrated base (1.5 N methanolic KOH at 70 degrees C for 1 hour), considerable amounts of hydroxycinnamic acids (up to 0.015% of mainly ferulic acid) and lipids (up to 0.43%) were released. The released phenolic acids and lipids were identified and quantified using high-performance liquid chromatography (HPLC) with detection by both UV and evaporative light-scattering detection (ELSD). During the wet milling of corn, two types of corn fiber are produced: coarse fiber, which is primarily from pericarp, and fine fiber, which is from the endosperm. The total phenolic acid content in CFGs purified from coarse corn fiber (pericarp fiber) is comparatively higher than that purified from fine corn fiber (endosperm fiber). It was also determined that the purified CFG samples contained significant amounts of strongly associated proteins, from 2 to 5% by weight. The presence of these phenolic acids, lipids, and proteins strongly associated or bound to CFG may contribute to its excellent ability to emulsify oil-in-water emulsions.  相似文献

The conventional corn wet‐milling process requires a long steeping time and has environmental and health concerns from the use of SO2. A recently proposed two‐stage enzymatic milling procedure with the first stage of water soaking and coarse grinding of corn and the second stage of incubating with enzymes has been shown to reduce the soaking time and possibly eliminate the need for SO2 addition. This current work explored the applications of protease and high‐intensity ultrasound in the second stage of the two‐stage enzymatic milling for corn starch isolation to further shorten the process time without use SO2. of The starch yield from sonication alone was 55.2–67.8% (starch db) as compared with 53.4% of the water‐only control with stirring for 1 hr and 71.1% of the conventional control with SO2 and lactic acid steeping for 48 hr. Protease digestion alone for 2 hr was not effective (45.8–63.9% yield) in isolating corn starch, but the starch recovery was increased to 61.2–76.1% when protease was combined with sonication. The preferred combination was neutral protease digestion for 2 hr followed by sonication at 75% amplitude for 30 min. The results demonstrated that combinations of high‐intensity ultrasound and neutral protease could replace SO2 and shorten the steeping time in the enzymatic wet‐milling process for corn starch isolation.  相似文献

A modified dry‐grind corn process has been developed that allows recovery of both pericarp and endosperm fibers as coproducts at the front end of the process before fermentation. The modified process is called enzymatic milling (E‐Mill) dry‐grind process. In a conventional dry‐grind corn process, only the starch component of the corn kernel is converted into ethanol. Additional ethanol can be produced from corn if the fiber component can also be converted into ethanol. In this study, pericarp and endosperm fibers recovered in the E‐Mill dry‐grind process were evaluated as a potential ethanol feedstock. Both fractions were tested for fermentability and potential ethanol yield. Total ethanol yield recovered from corn by fermenting starch, pericarp, and endosperm fibers was also determined. Results show that endosperm fiber produced 20.5% more ethanol than pericarp fiber on a g/100 g of fiber basis. Total ethanol yield obtained by fermenting starch and both fiber fractions was 0.370 L/kg compared with ethanol yield of 0.334 L/kg obtained by fermenting starch alone.  相似文献

To better understand the role of lactic acid (LA) in corn wet‐milling, steeping studies were performed on different yellow dent corn hybrids using four different solutions containing LA, sulfur dioxide (SO2), a combination of LA and SO2, or no added chemicals. Although there was variation in protein solubilization among the hybrids, protein release was consistently higher when LA was included in the steepwater than when it was excluded (both with and without SO2). Several groups have reported that starch recoveries are improved when steepwater contains LA. To explore the relationship between protein solubilization and starch yield as effected by LA, several yellow dent hybrids were steeped in 0.20% SO2 and 0.50% LA‐0.20% SO2 solutions and milled to recover starch by a 100‐g laboratory corn wet‐milling procedure. In all instances, both starch yields and protein solubilization were enhanced in solutions containing LA. These results support the hypothesis that direct dissolution of the endosperm protein matrix by LA contributes to the improved starch recoveries.  相似文献

Starch yield was significantly affected by all three main unit operations in alkali wet‐milling (debranning, roller milling, and steeping). The conditions for the three unit operations were studied using a single hybrid. Studies on debranning showed that optimal separation between pericarp and corn endosperm was obtained when corn was soaked in a 1.5–2% NaOH solution at 85°C for 5 min. Passing debranned corn through smooth roller mill once or twice did not affect the product yields, but passing the corn through the roller mill three times decreased the germ yield because of a large amount of broken germ. A 62% higher processing rate could be achieved when passing corn through the mill twice than by passing it through the mill once. The gap should be set at 2.0 mm when passing corn through the mill once, and it should be set at 3.5 mm for the first pass and 2.0 mm for the second pass when passing corn through the mill twice. Starch yield was more sensitive to NaOH concentration and steep temperature than to steep time. The highest starch yield was obtained when steeping corn in 0.5% NaOH for 1 hr at 45°C.  相似文献

The phenolic acid composition and concentration of four manually separated fractions (pericarp, aleurone layer, germ, and endosperm fractions) as well as whole grains of yellow corn, wheat, barley, and oats were analyzed by HPLC‐MS/MS following microwave‐assisted alkaline aqueous extraction. Phenolic acid compositions in whole grains and their fractions were similar, with minor differences among the grain fractions. Significant differences (P < 0.05), however, were observed in phenolic acid concentrations among cereal types, within cereal varieties, and among grain fractions, with yellow corn exhibiting the highest values. The concentrations of p‐coumaric and syringic acid in the pericarp were 10‐ to 15‐fold and 6‐ to 10‐fold higher, respectively, in yellow corn than in wheat, barley, and oats. In the aleurone layer, sinapic and vanillic acids in yellow corn were about 8‐ and 30‐fold more than in wheat. The germ fraction of wheat had 1.8 times more syringic acid than yellow corn germ. Grain fractions, excluding endosperm, had enhanced levels of phenolic acids compared with whole grain. Sinapic acid was more concentrated in the pericarp and germ of wheat, whereas isoferulic acid was concentrated in the germ of purple barley. Syringic and vanillic acids were concentrated in the pericarp and sinapic acid in the aleurone layer of yellow corn. These findings are important in understanding the composition and distribution of phenolic acids, and they act as a guide in identification of grain fractions for use as food ingredients. In addition, yellow corn fractions (aleurone and pericarp) may be potential alternative phenolic‐rich functional food ingredients in grain‐based food products.  相似文献

Enzymatic milling (E‐Milling) is a process that could potentially replace the sulfur dioxide procedure currently used in all commercial wet‐milling facilities. E‐Milling incorporates the use of a short water soaking step (≤6 hr), a coarse grind, and the use of a protease to release the starch granules from the corn endosperm. E‐Milling does not require sulfur dioxide to obtain starch yields equivalent to conventional wet milling; however, the important antimicrobial effects of sulfur dioxide are not duplicated by the enzymatic process. The use of low levels of sulfur dioxide (sufficient for antimicrobial activity) is being proposed as an easily implemented means of microbial control during E‐Milling. To assess the effectiveness of E‐Milling under these conditions, fraction yields for milling experiments adding sulfur dioxide with and without added enzyme were compared with fraction yields from conventional 24‐hr steeping with 2,000 ppm SO2 and 0.55% lactic acid. Because adding enzyme and SO2 can both improve product yields and compositions independently, it was necessary to use a reduced level of enzyme (much less than necessary to generate “product quality” material) to observe differences in terms of product yields. The results show significant differences in starch, fiber, total gluten, and insoluble gluten recoveries between samples milled with SO2 and enzyme compared with those at the same SO2 level without enzyme addition. No significant differences were observed for soakwater or germ yields regardless of the SO2 level used. The yield benefits from adding both enzyme and SO2 are clearly shown over the addition of each individually, for all coproduct yields with the exception of the yields for germ.  相似文献

New corn fractionation technologies that produce higher value coproducts from dry‐grind processing have been developed. Wet fractionation technologies involve a short soaking of corn followed by milling to recover germ and pericarp fiber in an aqueous medium before fermentation of degermed defibered slurry. In dry fractionation technologies, a dry degerm defiber (3D) process (similar to conventional corn dry‐milling) is used to separate germ and pericarp fiber before fermentation of the endosperm fraction. The effect of dry and wet fractionation technologies on the fermentation rates and ethanol yields were studied and compared with the conventional dry‐grind process. The wet process had the highest fermentation rate. The endosperm fraction obtained from 3D process had lowest fermentation rate and highest residual sugars at the end of fermentation. Strategies to improve the fermentation characteristics of endosperm fraction from 3D process were evaluated using two saccharification and fermentation processes. The endosperm fraction obtained from 3D process was liquefied by enzymatic hydrolysis and fermented using either separate saccharification (SS) and fermentation or simultaneous saccharification and fermentation (SSF). Corn germ soak water and B‐vitamins were added during fermentation to study the effect of micronutrient addition. Ethanol and sugar profiles were measured using HPLC. The endosperm fraction fermented using SSF produced higher ethanol yields than SS. Addition of B‐vitamins and germ soak water during SSF improved fermentation of 3D process and resulted in 2.6 and 2.3% (v/v) higher ethanol concentrations and fermentation rates compared with 3D process treatment with no addition of micronutrients.  相似文献

The objective was to describe a laboratory‐scale dry‐milling procedure that used single‐stage tempering and determine the effect of hybrid on yields and fraction compositions in milled corn. Samples of 11 commercially available hybrids were processed through a laboratory dry‐milling procedure that used 1 kg samples of corn to produce milling fractions of large grits, small grits, fines, germ, and pericarp. Compositions of milling fractions (protein, neutral detergent fiber, ash, and crude fat) were determined. The procedure used a single‐stage tempering step that increased corn moisture from 15 to 23.5% wb during an 18‐min tempering period. Germ were separated from endosperm particles using a roller mill followed by screening over a sieve with 1.68‐mm openings. Coefficients of variability were small, indicating acceptable repeatability. Overall yield means were 39.2, 25.3, 13.8, 78.2, 14.3, and 6.8 g/100 g (db) for large grits, small grits, fines, total endosperm, germ, and pericarp, respectively. There were effects due to hybrid (P < 0.05) on fraction yields and compositions of milling fractions. Correlations (r) among endosperm fractions (large grits, small grits, and fines) ranged from 0.54 to |–0.92|. Correlations among endosperm fractions and germ and pericarp were <0.68. The developed dry‐milling method estimated milling yields among hybrids with low standard deviations relative to the means and should be a useful tool for research and industry in measuring dry‐milling characteristics.  相似文献

Three fibrous corn wet-milling fractions, coarse fiber, fine fiber, and spent flake, were isolated. More highly valued uses are sought for these milling products, which are generally directed into the corn gluten feed product stream. Coarse fiber was further dissected into pericarp and aleurone layers. An alkaline hydrogen peroxide process was used to efficiently extract corn fiber gum (CFG) from each of the materials. CFG is a hemicellulose B arabinoxylan which also contains low levels of D,L-galactose and D-glucuronic acid. CFG yield information was obtained from each source, as well as structural information in terms of degrees of branching of the beta-D-xylopyranose backbone with alpha-L-arabinofuranosyl moieties. There were significant differences in degree of branching among the CFGs from the various fractions. A novel capillary electrophoresis procedure was developed to measure these differences. Solution viscosity differences among the CFGs were also observed.  相似文献

We recently reported that corn fiber oil contains high levels of three potential cholesterol-lowering phytosterol components: ferulate-phytosterol esters (FPE) (3–6 wt%), free phytosterols (1–2 wt%), and phytosterol-fatty acyl esters (7–9 wt%). A previous study also indicated that corn bran oil contained less phytosterol components than corn fiber oil. The current study was undertaken to attempt to confirm this preliminary observation using more defined conditions. Accordingly, oil was extracted from corn fiber and corn bran prepared under controlled laboratory conditions, using the same sample of corn hybrid kernels for each, and using recognized bench-scale wet-milling, and dry-milling procedures, respectively. After extraction, the chemical composition of the phytosterol components in the oil were measured. This study confirmed our previous observation—that FPE levels were higher in corn fiber oil than in corn bran oil. During industrial wet-milling, almost all of the FPE are recovered in the fiber fraction (which contains both fine and coarse fiber). During laboratory-scale wet-milling, ≈60–70% of the FPE are recovered in the coarse fiber (pericarp) and 30–40% are recovered in the fine fiber. During laboratory-scale dry-milling, <20% of the FPE are recovered in the bran (pericarp), and the rest in the grits. The recoveries of the other two phytosterol components (free phytosterols and phytosterol-fatty acyl esters) revealed a more complex distribution, with significant levels found in several of the dry- and wet-milled products.  相似文献

The stabilities of orange oil emulsions stabilized with various concentrations of two different types of corn fiber gum (CFG-1 and 2) isolated from coarse (pericarp) and fine (endosperm) fiber from corn wet milling have been studied. The emulsion stabilities in all these studies increased with increasing gum concentration up to a gum-to-oil ratio of 0.05, and after that it either levels off or changes very slightly. These results indicate that only 0.25% of CFG is required to make stable emulsion containing 5% orange oil under the experimental conditions used in this study. At this CFG concentration, CFG-2 from each fiber source was found to be a superior emulsifier relative to the corresponding CFG-1 from each source in a 10-day emulsion stability study at room temperature. The emulsion stability was also investigated by confocal laser scanning microscopy measurement, and it was found that CFG-1 and 2 from both coarse and fine fiber made stable emulsions with an average particle size of less than 1 mum for 10 days at room temperature. Sugar composition analysis of CFGs from both sources indicated that they were typical galactoglucuronoarabinoxylans containing mainly 55-59% xylose, 29-36% arabinose, and 4-6% galactose as neutral sugars and 3-5% glucuronic acid. Methylation analysis revealed a highly branched structure of all CFGs, in which only 16-25% of the 1--> 4-linked xylose residues were not substituted at O-2 and/or O-3. Arabinose is present both as a terminal residue and at branch points.  相似文献

The addition of six acids (organic and inorganic) and four sulfite compounds (including gaseous SO2) during the conventional corn wet‐milling steeping process of two yellow dent corn hybrids were evaluated for the effect on corn fiber yield, corn fiber oil yield, and the composition of three phytosterol compounds (ferulate phytosterol esters [FPE], free phytosterols [St], and phytosterol fatty acyl esters [St:E]) in the corn fiber oil. No significant effect of different sulfite compounds and acids were observed on corn fiber yields. However, a significant effect was observed on corn fiber oil yield and the composition of corn fiber oil for phytosterol compounds. Three of the sulfite compounds (including gaseous SO2) caused very little effect on the levels of phytosterol compounds compared with the control sample (corn steeped with sodium metabisulfite and lactic acid). However, for one hybrid, ammonium sulfite gave a significantly higher yield of FPE and St:E and had no effect on the yield of St. For the other hybrid, it gave a significantly higher yield of FPE and had no effect on the yield of St and St:E compared with the control sample. This indicates that the effect of these sulfite compounds on yields of these phytosterol compounds in corn fiber oil is probably hybrid‐dependent. No significant effect of acids was observed on corn fiber yields, but significant effects were observed on corn fiber oil yields and yields of phytosterol compounds in the corn fiber oil. The effect also seems to be hybrid‐dependent because different acids affected the two hybrids differently. Overall, it seems that weak acids have a positive effect on increasing the individual phytosterol compounds in the corn fiber. When comparing the effect of experimental acids and sulfites on the two hybrids, acids have a more positive effect than sulfites in increasing the yield of phytosterol compounds in corn fiber oil.  相似文献

To eliminate the diffusion barriers associated with enzyme addition during conventional steeping, we have developed a two‐stage milling procedure to evaluate the effects of enzyme addition on corn wet milling. The current study compares the effects of the addition of commercially available enzyme preparations during conventional steeping to their comparable addition in the two‐stage procedure. Results are presented in terms of yields of fiber, starch, germ, and gluten. The results demonstrate that the application of enzymes to the normal steeping step of wet milling is not an effective means of decreasing the steeping time or sulfur dioxide usage. Only when specific enzymes are added to the hydrated ground corn, using the modified two‐stage procedure, are enzymes effective in decreasing the steeping time and sulfur dioxide requirements. The overall steeping time with the two‐stage modified procedure ranges from 6 to 8 hr, representing a 67–83% reduction over the conventional process. The modified process greatly decreases, and possibly eliminates, the need for sulfur dioxide addition, while producing starch yields and quality equivalent to that from the conventional process.  相似文献

Different corn types were used to compare ethanol production from the conventional dry‐grind process to wet or dry fractionation processes. High oil, dent corn with high starch extractability, dent corn with low starch extractability and waxy corn were selected. In the conventional process, corn was ground using a hammer mill; water was added to produce slurry which was fermented. In the wet fractionation process, corn was soaked in water; germ and pericarp fiber were removed before fermentation. In the dry fractionation process, corn was tempered, degerminated, and passed through a roller mill. Germ and pericarp fiber were separated from the endosperm. Due to removal of germ and pericarp fiber in the fractionation methods, more corn was used in the wet (10%) and dry (15%) fractionation processes than in the conventional process. Water was added to endosperm and the resulting slurry was fermented. Oil, protein, and residual starch in germ were analyzed. Pericarp fiber was analyzed for residual starch and neutral detergent fiber (NDF) content. Analysis of variance and Fisher's least significant difference test were used to compare means of final ethanol concentrations as well as germ and pericarp fiber yields. The wet fractionation process had the highest final ethanol concentrations (15.7% v/v) compared with dry fractionation (15.0% v/v) and conventional process (14.1% v/v). Higher ethanol concentrations were observed in fractionation processes compared to the conventional process due to higher fermentable substrate per batch available as a result of germ and pericarp fiber removal. Germ and pericarp yields were 7.47 and 6.03% for the wet fractionation process and 7.19 and 6.22% for the dry fractionation process, respectively. Germ obtained from the wet fractionation process had higher oil content (34% db) compared with the dry fractionation method (11% db). Residual starch content in the germ fraction was 16% for wet fractionation and 44% for dry fractionation. Residual starch in the pericarp fiber fraction was lower for the wet fractionation process (19.9%) compared with dry fractionation (23.7%).  相似文献

Fourier-transform mid-infrared (FTIR) spectroscopy was investigated as a method to quantify the relative wheat grain tissue proportion in milling fractions. Spectra were acquired with a FTIR spectrometer equipped with an attenuated total reflectance device on ground samples, and the relative tissue proportion was determined according to the biochemical marker methodology as the reference method. Partial least-squares models were developed independently to predict the amount of outer pericarp, aleurone layer, starchy endosperm, and an intermediate layer (made up of inner pericarp plus seed coat plus nucellar epidermis). Good quality of prediction was obtained regardless of the target tissue. The standard errors of prediction obtained for the outer pericarp, intermediate layer, aleurone layer, and starchy endosperm quantification were, respectively, 3.4, 1.3, 3.4, and 4.6%.  相似文献

A new process was developed to recover corn fiber from the mash before fermentation in dry-grind ethanol production. In this process, corn is soaked in water (no chemicals) for a short period of time and then degermed using conventional degermination mills. In the remaining slurry, corn coarse fiber is floated by increasing the density of the slurry and then separated using density differences. The fiber recovered is called quick fiber to distinguish it from the conventional wet-milled fiber. This study evaluated the percent of quick fiber recovery for a normal yellow dent and high oil corn hybrid. The quick fiber was analyzed for levels of corn fiber oil, levels of ferulate phytosterol esters (FPE) and other valuable phytosterol components in the oil and compared with conventional wet-milled corn coarse and fine fiber samples. Fiber samples were also analyzed and compared for yields of potentially valuable corn fiber gum (CFG, hemicellulose B). Comparisons were made between the quick fiber samples obtained with and without chemicals in the soakwater. An average quick fiber yield of 6–7% was recovered from the two hybrids and represented 46–60% of the total fiber (fine and coarse) that could be recovered by wet-milling these hybrids. Adding steep chemicals (SO2 and lactic acid) to the soakwater increased the quick fiber yields, percent of FPE recoveries, and total percent of phytosterol components to levels either comparable to (for the dent corn hybrid) or higher than (for the high oil corn hybrid) those recovered from the total conventional wet-milled fiber samples. CFG yields in the quick fiber samples were comparable to those from the wet-milled fiber samples. CFG yields in the quick fiber samples were not significantly affected by the addition of chemicals (SO2 and lactic acid) to the soakwater.  相似文献

The structure of the aleurone layer was considered for many years as a potential factor influencing wheat milling efficiency. Eight durum wheat samples of different milling values, including distinct cultivars and harvesting conditions, were employed to investigate the structural characteristics of the aleurone layer through image analysis of kernel sections. Particular attention was paid to tissue thickness and structural irregularity of its interface with the starchy endosperm. Wheat cultivar, agricultural conditions, and location of measurement within the grain had an influence similar to both thickness and irregularity of the aleurone layer. Conversely, grain weight and morphology showed no effect on these parameters. Statistical investigation demonstrated no correlation between structural characteristics and wheat milling behavior. However, the negative correlation between the extraction rate of semolina and starch content in the bran fraction, which was used as an indicator of the endosperm‐aleurone dissociation extent, demonstrated the relevance of the tissue adhesion on milling efficiency.  相似文献
