Several Fusarium species occurring worldwide on maize as causal agents of ear rot, are capable of producing mycotoxins in infected kernels, some of which have a notable impact on human and animal health. The main groups of Fusarium toxins commonly found are: trichothecenes, zearalenones, fumonisins, and moniliformin. In addition, beauvericin and fusaproliferin have been found in Fusarium-infected maize ears. Zearalenone and deoxynivalenol are commonly found in maize red ear rot, which is essentially caused by species of the Discolour section, particularly F. graminearum. Moreover, nivalenol and fusarenone-X were often found associated with the occasional occurrence of F. cerealis, and diacetoxyscirpenol and T-2 toxin with the occurrence of F. poae and F. sporotrichioides, respectively. In addition, the occurrence of F. avenaceum and F. subglutinans usually led to the accumulation of moniliformin. In maize pink ear rot, which is mainly caused by F. verticillioides, there is increasing evidence of the wide occurrence of fumonisin B1. This carcinogenic toxin is usually found in association with moniliformin, beauvericin, and fusaproliferin, both in central Europe due to the co-occurrence of F. subglutinans, and in southern Europe where the spread of F. verticillioides is reinforced by the widespread presence of F. proliferatum capable of producing fumonisin B1, moniliformin, beauvericin, and fusaproliferin.  相似文献

This fluorescence and immunoelectron microscopic study showed that β-1,3-D-glucan accumulated only in leaves of a susceptible cultivar of Japanese pear after treatment with a host-specific toxin, AK-toxin I, from Alternate, alternata Japanese pear pathotype. The positive fluorescent reaction of callose was detected only in aniline blue fluorochrome-stained sections from toxin-treated leaves of the susceptible cultivar: positive sites were observed on cell walls of leaf cells. The sites of callose deposition were probably consistent spatially with modified sites on the plasma membrane that were observed only in the toxin-treated leaves of the susceptible cultivar. The toxin-induced modifications, identified as damage to the plasma membrane, were characterized by invagination of the plasmalemma specifically at plasmodesmata and as the concomitant accumulation of extracellular polysaccharides at the invaginated sites. A positive reaction to anti-β-1,3-D-glucan antibody was detected at the polysaccharides, Golgi vesicles, and trans-Golgi network (TGN) of toxin-treated leaves of the susceptible cultivar, but not at Golgi vesicles and TGN of water-treated ones. The cis-, medial and trans-Golgi stacks of toxin-treated leaves of the susceptible cultivar were negative for the antibody. The results showed that the polysaccharides, Golgi vesicles and TGN contained abundant β-1,3-D-glucan and that the glucan was transported from the Golgi apparatus via Golgi vesicles to the modified sites in cells of toxin-treated leaves of the susceptible cultivar. The extent of mycotoxin-induced responses in living roots of two maize (Zea mays L.) cultivars differing in their susceptibility to Fusarium was studied. Application of mycotoxin zearalenone (ZEN) or its derivates α-zearalenol (α-ZEL) and β-zearalenol (β-ZEL) caused a rapid depolarization of plasma membrane potential (EM). The extent of EM depolarization was dependent on the type of mycotoxin that have been used and showed concentration dependence. Interestingly, ZEN, but not its derivatives α-ZEL and β-ZEL, significantly decreased respiration of maize root cells. Electrolyte leakage increased with the duration of toxins treatment and was significantly higher in susceptible cv. Pavla than in resistant cv. Lucia. A strong superoxide dismutase insensitive nitro blue tetrazolium (NBT) reduction activity was identified in the root tips of control plants. This activity was rapidly inhibited by mycotoxin application in the meristematic/distal transition zones of roots in both cultivars examined. The level of recovery was a function of the mycotoxin concentration. Moreover, mycotoxin treatment also resulted in the onset and the progression of root cell death which was dependent on both, the type and concentration of mycotoxin.  相似文献

Plants have developed mechanisms to resist secondary infection upon inoculation with a necrotizing pathogen, chemical treatment as well as treatment with some non-pathogenic microorganisms such as rhizosphere bacteria. This phenomenon has been variously described as induced systemic resistance (ISR) or systemic acquired resistance. In the present study, the chemical benzo(1,2,3)thiadiazole-7-carbothioic acid-S-methyl ester (BTH, acibenzolar-S-methyl), and the rhizobacteriaPseudomonas aeruginosa KMPCH andP. fluorescens WCS417 were tested for their ability to induce resistance toColletotrichum lindemuthianum in susceptible and moderately resistant bean plants (Phaseolus vulgaris L.). BTH induced local and systemic resistance when bean leaves were immersed in 10−3 to 10−7 M BTH 3 days before the challenge inoculation. At a high concentration (10−3 M), BTH induced resistance of the same order as resistance induced by the pathogenC. lindemuthianum, although at this high concentration BTH appeared to be phytotoxic. Soil and seed treatment with 1 mg kg−1 BTH protected beans against anthracnose. BTH-mediated induced resistance was effective in susceptible and moderately resistant plants.P. aeruginosa KMPCH induced resistance in bean againstC. lindemuthianum only in a moderately resistant interaction. 为明确玉米对多堆柄锈菌Puccinia polysora侵染后病理反应的细胞学特征,利用扫描和透射电镜技术分析了玉米自交系与多堆柄锈菌互作中二者的细胞变化过程。多堆柄锈菌对玉米的侵染主要以直接穿透叶片表皮侵入为主,少量可从气孔和细胞间隙侵入。接种后,病菌夏孢子在感病自交系叶片上快速并大量萌发,在叶表生长蔓延并侵入表皮组织细胞,7 d后形成夏孢子堆;在抗病自交系上,病菌萌发、菌丝生长均受到明显抑制,少量入侵的病菌也由于寄主细胞死亡而导致菌丝和夏孢子干瘪死亡。侵染早期在感病寄主细胞间隙出现菌丝并穿透细胞壁,在胞内产生分枝菌丝,此时寄主细胞结构正常;随着菌丝进一步扩展,叶绿体等结构发生紊乱,被侵染细胞逐渐死亡。在抗病自交系上,接菌24 h后寄主即出现过敏性坏死反应,侵入位点与周围细胞快速坏死,抑制菌丝生长蔓延;叶绿体中清晰可见深色颗粒状物质;72 h后细胞壁外侧产生大量致密的深色结晶体,应为与抗病反应相关的酚类物质。表明抗多堆柄锈菌的玉米材料可能存在2种抗病途径,即寄主与病菌互作中由分子识别引起的免疫反应和病菌侵入后的系统防卫反应。  相似文献

The rapid generation of reactive oxygen species (ROS), called the oxidative burst, is one of the earliest host responses to pathogen infection or elicitor treatments. Therefore, we looked for the induction of ROS generation in Japanese pear leaves by the host-specific toxin, AK-toxin I using a cytochemical method for detecting H2O2. A small amount of non-specific generation of H2O2 was found in the cell walls in toxin- and water-treated susceptible and resistant leaves. Thus, the generation of H2O2 at cell walls appears to be caused by wounding stress during sampling. Specific generation of ROS, however, was found only in the membrane fragments and extended desmotubules characteristic of modified sites of the plasma membrane in the toxin-treated susceptible leaves. In addition, generation of H2O2 at plasma membranes was observed with higher frequency in toxin-treated susceptible leaves. This result indicates that the H2O2 generation was associated with damaged sites in the plasmalemma after toxin treatment and perhaps with the formation of membrane fragments from altered portions of the invaginated plasma membrane. The development of new maize hybrids with resistance to Fusarium infection is an effective means of minimizing the risk of mycotoxin contamination. Several maize hybrids have been investigated for Fusarium ear rot and accumulation of fumonisin B1 (FB1), fumonisin B2 (FB2), beauvericin (BEA) and fusaproliferin (FP) after artificial inoculation in the field with toxigenic strains of Fusarium verticillioides and Fusarium proliferatum. The year of inoculation had a significant influence on the disease severity and mycotoxin accumulation in maize kernels. Of all the hybrids tested, only Mona exhibited resistance to ear rot caused by F. verticillioides and produced low levels of fumonisins during three years of experiments. In Fusarium-damaged kernels (FDK), fumonisin B1, fumonisin B2, beauvericin and fusaproliferin were detected at concentrations much higher (up to 10–20 times) than in healthy-looking kernels (HLK). Animal and human exposure to these mycotoxins can be drastically reduced by removing mouldy and visibly damaged kernels from the commodity.  相似文献

 Reactive oxygen species (ROS) generation was examined in the interaction of Alternaria alternata Japanese pear pathotype and host plants using three methods: nitro blue tetrazolium (NBT) method for microscopic detection of O2 , diaminobenzidine (DAB) methods for microscopic detection of H2O2, and cerium chloride methods for ultrastructural detection of H2O2. ROS generation was detected by NBT and DAB methods at appressoria on leaves of susceptible cultivars and heat-shocked leaves of resistant cultivars but not in leaves of resistant cultivars. Ultrastructural detection by the cerium chloride method identified ROS generation at cell walls of appressoria and penetration pegs in susceptible, resistant leaves and heat-shocked leaves. These differences in the ultrastructural and microscopic data in resistant areas were due to the restriction of ROS generation in limited areas, the side facing the plant surface, of appressoria and penetration pegs. Therefore, ROS generation was apparently induced regardless of the resistance or susceptibility of the cultivar with the difference being in the volumes generated. After evaluating the pathological role of ROS generation in fungal structures, such generation was found to be associated with early penetration of cell walls in pear plants. Additionally, ROS generation in plants was also found in degrading pectin layers near infected hyphae and in plasma membrane modification sites in susceptible leaves but not in resistant leaves. ROS generation in susceptible leaves might be accompanied with plasma membrane damage, although the role of ROS generation in the pectin layers is not clear. Conidia of Cochliobolus carbonum secrete a toxin (HC-toxin) during appressorium formation on maize leaves. Plasma desorption mass spectrometry revealed that approximately 70 ng of toxin per 106 conidia were secreted during the first 16 h of morphogenesis. Growth of the fungus was monitored microscopically. Extensive fungal growth occurred in the susceptible interaction by 24 h. In spite of a substantial amount of HC-toxin, the fungus failed to become established in the resistant host even after 36 h. Results suggest that the resistance conditioned by Hm1, which encodes a toxin reductase, causes inactivation of the toxin early in the interaction.  相似文献

Leaf blight of garlic (Allium sativum) is a severe disease in garlic-growing regions. SS-toxin is a newly described non-proteinaceous toxin produced by the phytopathogenic fungus, Stemphylium solani, the cause of garlic leaf blight. In this study, the effects of SS-toxin on the H+-ATPase activity and standard redox activity in the plasma membranes isolated by aqueous polymer two-phase partitioning from garlic seedling leaves were studied in vitro. The H+-ATPase activity, NADH oxidation rate and Fe(CN)63− reduction rate of the plasma membranes isolated from susceptible and resistant cultivars were all inhibited in a dose-dependent manner. Our results suggest that, under in vitro conditions, the plasma membrane H+-ATPase and standard redox system can be both the cellular targets of SS-toxin.  相似文献

Absorption, translocation and metabolism of 14C-pyridate were compared in tolerant maize. moderately susceptible Polygonum lapathifolium and susceptible Chenopodium album. Foliar absorption was limited in all species, but comparatively higher penetration levels were observed in younger leaves of dicotyledonous species. The absorbed radioactivity was not very mobile and translocation appeared mainly sym-plastic. Herbicide selectivity could not be explained on the basis of absorption and transport. Chenopodium and P. lapathifolium degraded pyridate and formed unstable water-soluble conjugates that easily released a phytotoxic metabolite. By contrast, more stable unidentified water-soluble metabolites were found in maize. That metabolic difference could explain the selectivity of pyridate.  相似文献

为阐明入侵害虫草地贪夜蛾Spodoptera frugiperda对我国不同农作物的适应性,利用年龄-阶段两性种群生命表技术研究取食玉米粒以及玉米、花生、棉花、大豆、高粱和谷子叶片等7种不同食物对草地贪夜蛾生长发育与繁殖的影响。结果表明,不同食物显著影响草地贪夜蛾种群。整个成虫前期从长到短依次为取食棉花叶>取食大豆叶>取食高粱叶>取食花生叶>取食谷子叶>取食玉米叶>取食玉米粒,取食玉米粒要比取食棉花叶的时间缩短27.95 d。雌雄成虫寿命均以取食玉米粒和高粱叶的最长,以取食玉米叶的较短,且取食棉花叶的雄成虫寿命仅有3.00 d。产卵量以取食玉米粒的最多,为619.27粒,是取食其他食物的6.00倍~61.25倍。取食玉米叶的草地贪夜蛾初孵幼虫个体完成幼虫、蛹和成虫阶段的概率均较高,分别为95.38%、78.46%和78.46%,而取食棉花叶的最低,分别为37.29%、20.34%和20.34%。草地贪夜蛾取食玉米粒的净增值率、内禀增长率、周限增长率均最高,分别为105.59、0.12 d-1、1.13 d-1,平均世代周期最短,为36.91 d,而取...  相似文献

为了初步探讨西瓜抗蚜性的营养与次生代谢物质防御效应,采用分光光度法测定并分析了几种生化物质与西瓜抗蚜性的相关性。结果表明,抗性品种黑皮、绿美人、黑美人和惠兰的游离氨基酸、脯氨酸和可溶性氮含量分别为2.22~2.27、0.23~0.26和1.88~3.03 mg/g,显著低于感性品种金美人、小玲、甜美人和花绿的3.24~3.83、0.67~0.87和5.26~5.82 mg/g;抗性品种的糖/氮比为5.38~7.94,显著高于感性品种;抗、感品种间可溶性糖和维生素C(Vc)含量无显著差异;瓜蚜为害4 d后的抗性品种叶组织中的总酚和单宁酸含量显著增加,感性品种显著降低;丙二醛含量增加幅度显著高于抗性品种。研究表明,西瓜品种抗蚜性与糖/氮比及总酚、单宁酸含量均呈显著正相关,与可溶性氮、游离氨基酸、脯氨酸及丙二醛含量均呈显著负相关,但与可溶性糖和Vc含量无显著相关性。  相似文献

The occurrence of Fusarium species of Liseola section and related toxins was investigated for two years (1996 and 1998) on maize ear rot samples collected in the most important areas for maize growing in Slovakia. The species most frequently isolated was F. verticillioides, followed by F. proliferatum in 1996 and F. subglutinans in 1998. Most of the strains belonged to mating populations A, D, and E of the teleomorph Gibberella fujikuroi. Fusarium graminearum was also frequently recovered in both the years of investigations. Toxin analysis of maize ears showed that most of the samples (21 out of 22) were contaminated with at least one toxin. In particular, the concentration of fumonisin B1, and fumonisin 2 was up to 26.9 and 5.1gg-1, respectively in 1996, and up to 12.1 and 6.3gg-1, respectively in 1998. Beauvericin was detected only in one sample in 1996. Seven samples in 1996 were contaminated by fusaproliferin up to 8.2gg-1, but just traces of the toxin were found in one sample in 1998. All 29 strains of F. verticillioides, two of three strains of F. proliferatum and none of eight F. subglutinans strains isolated from samples produced fumonisin B1 in culture on whole maize kernels (0.1–5646 and 940–1200ugg-1, respectively). Two strains of F. subglutinans and two of F. proliferatum produced beauvericin (up to 65 and 70gg-1, respectively). Ten strains of F. verticillioides produced beauvericin: 9 strains produced a low amount (up to 3gg-1), while only one of them produced a high level of toxin (375gg-1). Fusaproliferin was produced by two F. proliferatum strains (220 and 370gg-1), by seven F. subglutinans (20–1335gg-1) and by three F. verticillioides (10–35gg-1). This is the first report on fusaproliferin production by F. verticillioides, although at low level.  相似文献

A concentration of atrazine of 0?1 mm 1?1 in the nutrient solution resulted in complete inhibition of photosynthesis in intact leaves of susceptible biotypes of Amaranthus retroflexus L., Polygonum lapathifolium L., Chenopodium album L., Solanum nigrum L., Poa annua L. and Stellaria media (L.) Vill. within a few hours of treatment, whereas the inhibition of the resistant biotypes by the same concentration varied from small to moderate. In contrast, diuron (10 or 20 μm 1?1) produced only minor differences between resistant and susceptible biotypes. The influence of some other herbicides on photosynthesis of these resistant biotypes was also smaller than that on the susceptible biotypes. This cross-resistance was evaluated with the resistance factor for intact leaves of Brassica napus L., A. retroflexus, and S. nigrum. This factor is equal to the ratio of the herbicide concentration in leaves of the resistant biotype to that in leaves of the susceptible biotype with inhibition to half-maximum rate of photosynthesis. This concentration in the leaves was calculated from the concentration in the nutrient solution, and the total transpiration divided by leaf area from the beginning of the herbicide treatment until the moment of half-maximum of photosynthesis. The resistance factors for intact leaves of A. retroflexus, S. nigrum and B. napus were 26–30 for atraton, 3–7 for metamitron, 2–9 for bromacil, 3–5 for monolinuron, 1 for diuron and < 1 for bentazone. For isolated chloroplasts much higher values have been reported. The reason for this discrepancy is not clear. A somewhat higher resistance factor (around 50–60) was derived after infiltration of detached leaves of these species with atraton solutions.  相似文献

Vulculic acid(2-乙酰基-3,4-二羟基-5-甲氧苯基-乙酸)是从空心莲子草(Alternanthera philoxeroides)潜在生防真菌莲子草假隔链格孢(Nimbya alternantherae)培养滤液中分离获得的致病毒素.研究表明,该毒素为非寄主专化毒素,能够引致空心莲子草叶片细胞膜透性升高,导致电解质渗漏,同时降低叶片细胞内过氧化物酶(POD)和过氧化氢酶(CAT)的活性,使细胞防御酶系统发生紊乱,还能够造成空心莲子草叶片细胞质壁分离、质膜消解,叶绿体片层紊乱、膜消解[1,2].  相似文献

为评估转cry1Ab/cry2Aj、cry1Ab/vip3DA玉米对棉铃虫Helicoverpa armigera(Hübner)、甜菜夜蛾Spodoptera exigua(Hübner)和斜纹夜蛾Prodenia litura(Fabricius)的抗虫性,在室内测定了3个转cry1Ab/cry2Aj玉米品系和1个转cry1Ab/vip3DA玉米品系对3种害虫幼虫存活和生长发育的影响,研究了该系列Bt玉米不同组织器官对害虫的杀虫活性和控制效果。结果显示,棉铃虫初孵幼虫取食各品系Bt玉米叶片96h后死亡率为87.50%~90.00%,取食花丝和雌穗的幼虫96h后几乎全部死亡;甜菜夜蛾初孵幼虫取食各品系Bt玉米叶片、花丝和雌穗168h后死亡率为22.50%~68.33%,幼虫的生长发育受到明显抑制,体重抑制率达85.00%~95.00%;斜纹夜蛾初孵幼虫取食各品系Bt玉米叶片、花丝和雌穗96 h后死亡率显著高于非转基因亲本对照,168h后幼虫死亡率达90.00%以上。研究表明,转cry1Ab/cry2Aj和cry1Ab/vip3DA玉米品系对棉铃虫和斜纹夜蛾的初孵幼虫表现出较好的抗性,可以作为转多基因抗虫玉米育种的备选材料。  相似文献

为明确新入侵农业害虫草地贪夜蛾Spodoptera frugiperda对药用植物金银花Lonicera japonica的潜在为害风险,采用室内饲养观察法比较取食玉米Zea mays和金银花叶片后草地贪夜蛾的种群特征,并采用室内生测法测定取食2种植物幼虫体内消化酶和解毒酶的活性。结果显示,草地贪夜蛾在金银花上能完成完整的世代发育。取食金银花叶片的草地贪夜蛾幼虫发育历期为24.08 d,显著长于取食玉米的幼虫发育历期(18.47 d);取食金银花叶片的幼虫存活率为63.67%,较取食玉米叶片幼虫的存活率78.00%明显降低;取食金银花叶片的草地贪夜蛾雌、雄蛹重分别为197.98 mg和198.78 mg,显著大于取食玉米叶片时;取食金银花和玉米叶片的草地贪夜蛾成虫的繁殖力与寿命均无显著差异。取食金银花叶片的草地贪夜蛾5龄幼虫中肠中的淀粉酶、脂肪酶、胰蛋白酶、谷胱甘肽S-转移酶和细胞色素P450酶的活性均较取食玉米叶片时显著降低,但羧酸酯酶活性较取食玉米叶片时显著升高。表明草地贪夜蛾在金银花上能较好地生长发育和繁殖,这对金银花的安全生产构成潜在威胁。  相似文献

The rate of photosynthesis in intact leaves attached to plants of an atrazine-resistant cultivar of Brassica napus L. was compared with that in leaves of an atrazine-susceptible, rather isogenic cultivar, in a number of irradiance series. The resistant cultivar had been obtained from the susceptible cultivar by crossing the resistant biotype of Brassica campestris L. with the susceptible cultivar of B. napus, and using the fourth back-cross for comparison. At a natural CO2 concentration (360 cm3 m?3) the rate of photosynthesis of leaves of the atrazine-resistant cultivar was significantly (17–21%) less than that of leaves of the susceptible cultivar, over the whole range of irradiances up to 250 W m?2 At the enriched CO2 concentration of 1500 cm?3 m?3 this reduction in rate amounted to 9–12%.  相似文献
