Species of Staphylococcus isolated from animal infections   总被引:3,自引:0,他引:3  
One hundred randomly selected clinical strains of staphylococci were identified by species using a commercial micromethod system. Eight species of staphylococci were identified. Staphylococcus intermedius was the most frequent (n = 74) species identified and accounted for 70/74 (94.6%) of the coagulase-positive strains and 70/79 (88.6%) of the total isolates from dogs. Other species identified, in order of their frequency, included S. epidermidis (8), S. aureus (7), S. simulans (4), S. sciuri (2), S. xylosus (2), S. hyicus (2) and S. saprophyticus (1). These results show that at least 8 different species of staphylococci can be recovered from animal infections and that coagulase-positive species such as S. intermedius may be more common than S. aureus. The relative significance of these other species in animal infections needs to be assessed.  相似文献

Specimens obtained from pyogenic skin lesions of 210 dogs were culturally examined for staphylococci. A total of 215 isolates of staphylococci were biotyped, using the biochemical tests contained in a commercial staphylococcal identification system. Of 201 coagulase-positive isolates, 197 were identified as Staphylococcus intermedius, 3 as S aureus, and 1 as S hyicus. Of 14 coagulase-negative isolates, 5 were identified as S epidermidis, 5 as S xylosus, 3 as S simulans, and 1 as S hominis. Antimicrobial susceptibility tests were done on all staphylococcal isolates, using the standard disk-diffusion method. Staphylococcus intermedius isolates were susceptible to cephalothin, methicillin, and gentamicin. Resistance to ampicillin, penicillin G, and tetracycline was frequent. Antibiotic resistance was not associated with the depth of skin infection. Resistance to ampicillin, penicillin, tetracycline, and trimethoprim-sulfamethoxazole was not associated with previous antibiotic use. Increased resistance to chloramphenicol, clindamycin, and erythromycin was associated with previous antibiotic therapy. Antimicrobial susceptibilities of the other Staphylococcus species isolated are reported, but the small numbers of these species precluded making meaningful comparison with S intermedius.  相似文献

One hundred and twenty eight strains of Staphylococcus from lesions, mostly of the skin, in horses were identified and compared with 29 strains isolated from the healthy skin. The pathogenic species Staphylococcus aureus, S. intermedius and S. hyicus were found almost exclusively in lesions. Other species such as S. xylosus and S. sciuri were more frequently found on the healthy skin than in lesions. The S. aureus strains formed a very heterogeneous collection. Many of these strains were staphylokinase positive and rapidly coagulated bovine plasma. Such strains are rarely found in other animals and man.  相似文献

In this study, bacterial cultures were collected from five sites on each of 50 healthy cats and 48 cats with inflammatory skin disease (ISD), to determine prevalence of carriage and relative frequency of methicillin resistance in coagulase-positive staphylococci and Staphylococcus schleiferi ssp. schleiferi. Latex agglutination testing for penicillin-binding protein 2a (PBP2a) and pulsed field gel electrophoresis (PFGE) were performed on all methicillin-resistant (MR) isolates. Polymerase chain reaction (PCR) for the mecA gene was performed on MR S. intermedius and S. schleiferi isolates. Staphylococcal chromosomal cassette (SCCmec) typing was performed on all MR S. aureus isolates. Coagulase-positive staphylococci and S. schleiferi ssp. schleiferi were isolated from 24 of 48 cats with ISD: Staphylococcus aureus (14 of 24, 58%), Staphylococcus intermedius (11 of 24, 46%), Staphylococcus schleiferi ssp. schleiferi (1 of 24, 4%), and Staphylococcus hyicus (1 of 24, 4%). Prevalence of MR was 7% for S. aureus, 0% for S. intermedius, 100% for S. schleiferi ssp. schleiferi, and 0% for S. hyicus. Coagulase-positive staphylococci were isolated from 17 of 50 healthy cats: S. aureus (10 of 17, 59%), S. intermedius (11 of 17, 65%), and S. schleiferi ssp. coagulans (1 of 17, 6%). Prevalence of MR was 20% for S. aureus, 18% for S. intermedius, and 0% for S. schleiferi ssp. coagulans. All MR isolates were positive for PBP2a via latex agglutination. Methicillin-resistant S. intermedius and S. schleiferi ssp. schleiferi isolates were also positive for the mecA gene via PCR. Methicillin-resistant S. aureus isolates were identified as SCCmec type II. Results of PFGE indicated heterogeneity among isolates. There was no significant difference in staphylococcal isolation or methicillin resistance between study groups. While present, MR coagulase-positive staphylococci are significantly less common in these study populations.  相似文献

Attempts were made to identify 900 species of staphylococci or micrococci recovered from samples of bovine milk examined for mastitis pathogens. The presence and identity of haemolysins was recorded together with results of disc diffusion antibiotic sensitivity tests. The occurrence of clinical mastitis was also noted and somatic cell counts (SCC) were performed on milk samples which were normal in appearance. Eight hundred and thirty-one coagulase positive staphylococci were obtained, of which 810 were S. aureus and 21 were S. intermedius. Of 65 coagulase negative staphylococci the species of 19 could not be determined by the identification systems used. The remainder were identified as S. hyicus sub sp. hyicus (1), S. hyicus sub sp. chromogenes (19), S. haemolyticus (17), S. hominis (3), S. epidermidis (4), S. capitis (1) and either S. hominis or S. warneri (1). Four other isolates could not clearly be assigned to the genus Staphylococcus or Micrococcus and were designated irregular strains. No micrococci were identified. The presence of alpha, beta, or delta haemolysins occurring singly or in various combinations was identified in 98.3% of coagulase positive staphylococci and in 60% of coagulase negative staphylococci. Epsilon haemolysin was detected in 47.6% of the coagulase negative staphylococci and in 9.5% of S. intermedius. All staphylococci were sensitive to tetracycline (30 microg), novobiocin (1.6 microg), nafcillin (30 microg), methicillin (10 microg) and cephalothin (30 microg) and variable numbers of each species were sensitive to penicillin (2 iu) and streptomycin (10 microg). One non-identified species of coagulase negative staphylococcus was sensitive to erythromycin (0.4 microg) the remaining staphylococci were resistant. Each of the four irregular strains was sensitive to erythromycin and novobiocin. Clinical mastitis was associated with 30.6% of coagulase positive staphylococci, 15.3% of coagulase negative staphylococci, and two of the four irregular strains (50%). Subclinical mastitis as determined by SCC of 500 x 10(3) or greater was associated with 92.7% of coagulase positive and 37.5% of coagulase negative staphylococci.  相似文献

The development of a PCR assay based on the 16S ribosomal RNA gene (rDNA) sequence was carried out for the identification of Staphylococcus intermedius. Sixty-six strains of S. intermedius, 70 of Staphylococcus aureus and 2 of Staphylococcus hyicus were examined for the assay. The 16S rDNA, of which the PCR target fragment makes up 901 bp corresponding to the sequence data of the gene, was detected in all strains of S. intermedius, but it was not detected in any strains of either S. aureus or S. hyicus. These results suggest that the PCR allows a simple and precise identification of S. intermedius.  相似文献

Swab specimens obtained from skin lesions of 45 cats were cultured bacteriologically for staphylococci. Thirty-two staphylococcal isolates were recovered from 30 cats and were biotyped, using biochemical tests contained in a staphylococcal identification system. Of 23 isolates considered coagulase-positive, 16 were identified as Staphylococcus aureus, 5 as S intermedius, and 2 as S hyicus. Of 9 isolates considered coagulase-negative, 6 were identified as S simulans, 2 as S epidermidis, and 1 as S xylosus. Antimicrobial susceptibility tests were done on all staphylococcal isolates, using a disk-diffusion method. Staphylococcal isolates were susceptible to clavulanic acid-amoxicillin, cloxacillin, cephalothin, chloramphenicol, gentamicin, erythromycin, and trimethoprim-sulfamethoxazole. Resistance to penicillin G, ampicillin, and tetracycline was frequent.  相似文献

Three rapid agglutination assays for the identification of Staphylococcus aureus Monostaph (Bionor A/S, Skien, Norway), Staphyslide-Test (BioMerieux, Lyon, France) and Staph-Rapid-Test (Roche, Basel, Switzerland), were compared. A total of 104 Gram-positive, catalase positive cocci were tested: Nineteen Staphylococcus reference strains comprising 15 spp. (4 strains were coagulase positive), and 7 Micrococcus reference strains comprising 4 spp.; 22 food isolates comprising 13 S. aureus, 8 coagulase positive Staphylococcus spp., and 1 Micrococcus sp.; 56 animal isolates comprising 11 S. aureus, 9 S. hyicus subsp. hyicus, 2 S. intermedius, 15 coagulase positive and 19 coagulase negative Staphylococcus spp. Totally 54 strains were coagulase positive. Considering agglutination of a coagulase positive strain as a correct identification, Monostaph, Staph-Rapid-Test, and Staphyslide-Test correctly identified 52 (96.3%), 47 (87.0%) and 48 (89.0%) of the coagulase positive staphylococci, respectively. Monostaph, Staph-Rapid-Test and Staphyslide-Test showed 1 (2.0%), 4 (8.0%) and 4 (8.0%) false positive reactions respectively. Monostaph, Staph-Rapid-Test and Staphyslide-Test gave 0 (0.0%), 6 (5.8%) and 7 (6.7%) non-interpretable reactions, respectively. Monostaph may be a good alternative to the tube-coagulase test for rapid and reliable identification of coagulase positive staphylococci from both food and veterinary sources. However, false negative reactions may occur with coagulase positive strains of S. hyicus subsp. hyicus and S. intermedius.  相似文献

Distribution of staphylococcal species on clinically healthy cats   总被引:5,自引:0,他引:5  
Among 827 isolates derived from 113 clinically healthy cats, 12 species of staphylococci were identified. Staphylococci were isolated from each cat and from 54.9% of the anatomic sites evaluated. A mode of 6 (range = 2 to 11) of the 11 anatomic sites evaluated per cat yielded staphylococci. A mode of 8 (range = 2 to 12) isolates were found per cat. Staphylococcus simulans was the most isolated (43.9% of total) coagulase-negative species. Moreover, S simulans was the most isolated species from each of the 11 sites evaluated and, except for the mouth and haircoat, comprised greater than 50% of the isolates from each site. Staphylococcus intermedius was the most isolated (13.5% of the total) coagulase-positive species. Three other species (S epidermidis, S xylosus, and S aureus) comprised 32.2% of the isolates, and 7 species (S haemolyticus, S hominis, S hyicus, S capitis, S warneri, and S saprophyticus) comprised 10.4% of the isolates. Six species (S intermedius [96 of 112 isolates], S haemolyticus [20 of 22], S sciuri [17 of 18], S warneri [10 of 13], S hyicus [10 of 10], and S capitis [7 of 8]) were isolated primarily from household cats. Only 1 species, S xylosus (75 of 87), was isolated primarily from cattery cats. Haircoat specimens (n = 452) yielded 508 isolates (61.4% of the total) distributed among all 12 staphylococcal species and included greater than 50% of the isolates of all species other than S simulans and S sciuri. A more heterogeneous population of staphylococci was isolated from household cats than was isolated from cattery cats.  相似文献

Surface hydrophobicity of 90 Staphylococcus intermedius and 55 S hyicus isolates was evaluated using the hexadecane adherence assay and the ammonium sulphate salt aggregation test. A strongly positive hydrocarbon adherence in the hexadecane adherence assay was demonstrated in 11 per cent of the S intermedius isolates and 7 per cent of the S hyicus isolates. Bacterial aggregation in 1.6 M, or less, ammonium sulphate was observed in 28 per cent of the S intermedius isolates and 37 per cent of the S hyicus isolates. There was no statistical correlation between the two assays. The adherence of both bacterial species to hexadecane was eliminated when the cells were first treated with pronase and trypsin, while it was mildly enhanced by prior heat treatment (60 degrees C and 95 degrees C for up to three hours). In contrast, aggregation of S intermedius in ammonium sulphate was not influenced by trypsin pretreatment, and aggregation of both bacterial species was diminished, or eliminated, with pronase or prior 95 degrees C heat treatment. Surface hydrophobicity, as measured in both assays, appeared to have no relationship with growth patterns in serum soft agar or production of slime. Similarly, the presence or absence of substantial surface receptor activity to fibrinogen, fibronectin or IgG did not appear to be related to surface hydrophobicity.  相似文献

Antimicrobial resistance of feline staphylococci in south-eastern England   总被引:1,自引:0,他引:1  
Staphylococcus aureus is reported as the predominant feline staphylococcal pathogen. There is concern that cats may transfer resistant staphylococci to humans. In this study, staphylococci were obtained from skin and mucosae of 20 domestic cats, 9 with lesions, and 10 healthy feral cats. Species were identified by DNase and API ID32 Staph tests. Of 187 isolates, 21.4% were coagulase-positive and predominately from lesional cats; 90% of these were Staphylococcus intermedius . Coagulase-negative species were isolated equally in all three groups. All isolates were susceptible to coamoxiclav, cephalexin and bacitracin. Twenty-two, including 18 coagulase-negative isolates, showed some resistance to cotrimoxazole, lincomycin, enrofloxacin or oxytetracycline. Two isolates were resistant to more than one antibiotic. More resistant isolates were obtained from feral cats ( P < 0.01). The results suggest that S. intermedius is the principal coagulase-positive species. Antibiotic resistance is generally low amongst feline staphylococci. Higher resistance amongst feral cats suggests exposure to environmental antibiotics.  相似文献

Temporal study of staphylococcal species on healthy dogs   总被引:5,自引:0,他引:5  
During a 1-year period, specimens were obtained monthly from 5 hair coat and 7 mucous membrane sites of 11 healthy dogs. Among 804 isolates of staphylococci, 13 species were identified. Staphylococcus intermedius was the most frequently isolated (40.2% of total isolates) coagulase-positive species, and S xylosus was the most frequently isolated (17.3%) coagulase-negative species. Moreover, S intermedius was the most frequently isolated species from the 12 sites evaluated and was isolated persistently from 8 of the 9 dogs that completed the 1-year study. On the basis of a commercial identification system, 14 profile numbers were identified for isolates of S intermedius. However, 2 profile numbers accounted for a majority (70.9%) of the isolates. Specific S intermedius biotypes identified on the basis of hemolysis, coagulase production, beta-lactamase activity, and antimicrobial susceptibility patterns were found repeatedly in 3 dogs. Seemingly, S intermedius was a resident of the normal bacterial microflora of these dogs; however, the inability to isolate S intermedius from 1 dog during the study year indicated that not all dogs harbor S intermedius as a resident microorganism.  相似文献

A study was made of the staphylococcal population on the skin and on the nasal mucosa and in the milk of 133 healthy goats. Of a total of 346 strains isolated and characterised as belonging to the genus Staphylococcus, 74 (21.4%) were coagulase-positive (68 S. aureus and 6 S. hyicus), and 272 (78.6%) coagulase-negative. The novobiocin-sensitive species S. haemolyticus (23.5%), S. warneri (16.5%), S. epidermidis (11.8%), S. chromogenes (8.5%), S. caprae (6.6%) and S. hyicus (2.6%), and the novobiocin-resistant species S. xylosus (8.5%), S. sciuri (7.4%), S. saprophyticus (4.8%), S. cohnii (2.2%), S. lentus (1.1%), S. equorum (1.1%) and S. kloosii (1.1%) were identified. Twelve (4.4%) of coagulase-negative strains remained unidentified. Strains isolated in the skin of the udder and teats of the 133 goats were mainly novobiocin-sensitive coagulase-negative staphylococci, the most prevalent species being S. haemolyticus, S. warneri and S. epidermidis. Staphylococci indicative of subclinical infection were determined in the milk of 47 (35.3%) of the 133 goats sampled.  相似文献

Using the Streptococcus equi decapsulation test, 879 strains of S. aureus, 212 strains of S. intermedius and 214 coagulase-negative staphylococci were examined for hyaluronidase production; six of the coagulase-negative staphylococci belonged to S. hyicus subsp. hyicus. Positive hyaluronidase production was recorded in all strains of S. aureus and in the strains of S. hyicus subsp. hyicus. No hyaluronidase was produced by 208 coagulase-negative staphylococci and the strains of S. intermedius. The decapsulation test is recommended as a reliable method of the differentiation of S. aureus and S. intermedius.  相似文献

Staphylococcus intermedius has been the predominant coagulase-positive Staphylococcus isolated from canine skin and mucosae and the most commonly reported staphylococcal pathogen in small animal practice for the last 35 years. Although microbiological tests have historically indicated variability in biochemical characteristics amongst S. intermedius isolates from animals, an acceptable level of diagnostic accuracy for clinical purposes was readily achievable with routine phenotypic testing. However, three recent developments have changed our understanding of the term "S. intermedius" and have challenged veterinary bacteriologists to ensure correct species identification of pathogenic staphylococci from small animals. First, the increasing recognition of meticillin-resistant Staphylococcus aureus in small animal practice and its human health implications demand accurate species identification. Secondly, the application of molecular techniques to analysis of staphylococcal isolates has led to a revised taxonomy and canine isolates of S. intermedius being re-named S. pseudintermedius. Thirdly, the recent, rapid emergence of meticillin- and multi-drug-resistant strains of Staphylococcus pseudintermedius (MRSP) has become a major therapeutic challenge in veterinary practice worldwide, including the UK. This article discusses the background of the recent taxonomic changes within the genus Staphylococcus and reviews the key features of MRSP and its implications for day-to-day laboratory diagnosis and small animal practice.  相似文献

Comparisons were made among rapid latex agglutination tests and conventional biochemical tests used to identify Streptococcus agalactiae and Staphylococcus aureus. Ninety-eight streptococci and 149 staphylococci isolated from bulk tank milk were tested. Sensitivity and specificity for the latex agglutination test used for identification of Str agalactiae were 97.6 and 98.2%, respectively. Sensitivity and specificity for the latex agglutination test used for identification of S aureus were 90.2 and 67.5%, respectively. Of 25 staphylococci considered false-positive by the latex agglutination test, 14 (56%) were considered tube coagulase-positive. Fifteen staphylococci considered false-positive by latex agglutination test had biotypes representative of S hyicus of S xylosus.  相似文献

The Staph-Zym system was evaluated as a means for identifying cultures of Staphylococcus hyicus isolated from pigs and bovines and cultures of Staphylococcus intermedius isolated from canines. The selected cultures had been identified by conventional methods. The Staph-Zym system correctly identified all 52 S. hyicus and all 33 S. intermedius. It is concluded that the Staph-Zym system is a practical and reliable test for identifying mostly animal pathogenic S. hyicus and S. intermedius, and might possibly be useful for veterinary microbiologists.  相似文献

3 commercially available tests STAPHYSLIDE (api bio Mérieux), STAPHYLASE (Oxoid) and STAPHAUREX-D (Welcome) for identification of Staphylococcus aureus were evaluated by 110 coagulase-positive S. aureus strains and 40 coagulase-negative Staphylococcus strains, isolated from various materials of different animal species. STAPHYSLIDE showed a sensitivity of 53.6% and a specificity of 97.5%, STAPHAUREX-D 56.4% respectively 85% and STAPHYLASE 32.7% respectively 100%. The possible reasons for the weak results of these 3 tests well established in bacteriology of human medicine by use in veterinary bacteriology are discussed.  相似文献

A simplified scheme for the subdivision of coagulase-positive staphylococci and another simplified scheme for the diagnostics of coagulase-negative species were worked out. On the basis of the production of staphylokinase, coagulation of human and bovine plasma, acetoin production from glucose, and growth on agar with crystal violet, it is possible to identify S. aureus with its biovars A, B, C1, C2, D, as well as S. intermedius. The coagulase-negative species can be diagnosed according to their sensitivity to novobiocin, nitrate reduction, fermentation of maltose, sucrose, salicin, xylose, trehalose, mannitol and mannose, and haemolytic activity. The proposed diagnostic schemes were verified with success on the collection strains and on the 1305 staphylococci strains isolated largely from the bovine mammary gland, from dogs, man and domestic fowl. In S. aureus strains a close correlation was demonstrated between their biotype characteristics and the host species. A similar correlation was determined for S. intermedius. As to the coagulase-negative species, S. epidermidis, S. hominis and S. haemolyticus were diagnosed most frequently. Both schemes represent a reliable, prompt and technically simple method of the diagnostics of the Staphylococcus microorganisms.  相似文献

The aim of the study was to isolate gram-positive cocci from cows with mastitis and to determine their resistance to beta-lactamic antibiotics. Eight hundred and nine strains were isolated and identified as staphylococci (n=516), streptococci (n=199) and enterococci (n=94) from sub-clinical and clinical cases of bovine mastitis in Lithuania. The most common causative agents of udder disease included: S. epidermidis (n=176), S. aureus (n=176), S. agalactiae (n=134), S. hyicus (136) and E. hirae (n=68). Isolates were analysed for antimicrobial resistance to penicillin, ampicillin, amoxicillin, cephalothin, cephalexin, amoxicillin + clavulanate. The susceptibility patterns were analysed using the agar disk diffusion method. S. aureus showed the highest level of resistance to amoxicillin (81.3%), penicillin (76.7%) and ampicillin (78.4%). The corresponding values for CNS strains were 59.7%, 59.7% and 50.6% against penicillin, ampicillin and amoxicillin respectively. Streptococci were the most frequently resistant to amoxicillin (29.3%), and enterococci to penicillin (27%), amoxicillin (27.5%) and amoxicillin/clavulanic acid (23.8%). The resistance of all tested mastitis pathogens to aminopenicillins and penicillin highly correlated (r=0.83). Compared with other antibiotics, amoxicillin and clavulanic acid combination tended to be more effective (p<0.05) against all tested bacteria in vitro. However, S. aureus, in 38.1% of cases, was resistant to this combination of antimicrobials. This study demonstrates that S. epidermidis, S. aureus, S. hyicus, S. agalactiae and E. hirae remain the most frequent mastitis causative agents on Lithuanian cattle farms. The highest resistance in vitro to penicillins was demonstrated by S. aureus, S. hyicus and S. intermedius. Resistance to cephalosporins remains low, irrespective of bacterial species of gram-positive cocci.  相似文献
