Phosphoglucoisomerase (PGI) was studied to establish the genetic system of the isozymes in section Cepa of Allium. The results of isozyme analyses using 15 cultivars and clones in two cultivated species A. fistulosum and A. cepa, eight strains in four wild species A. altaicum, A. galanthum, A. oschaninii and A. vavilovii and a total of 10 hybrids between A. fistulosum and A. cepa and between the cultivated and wild species revealed that: 1) PGI was a dimeric enzyme; 2) the two cultivated and four wild species had a common gene locus Pgi-1 where six alleles were identified; and 3) A. fistulosum had Pgi-1 F-1 and Pgi-1 F-2, A. cepa had Pgi-1 A-1 and Pgi-1 A-2, A. altaicum had Pgi-1 F-1, A. galanthum and A. vavilovii had Pgi-1 A-1, and A. oschaninii had Pgi-1 A-1, Pgi-1O-1 and Pgi-1 O-1. The intra- and inter-specific polymorphisms of PGI isozymes are expected to be valuable tools for the genetics and practical utilization of the wild species in section Cepa.  相似文献

Summary On the combined evidence from morphology, ecology, and cytogenetics, seven species are recognized in the genus Avena L. 1. A. clauda Dur., 2. A. ventricosa Bal. 3. A. longiglumis Dur, 4. A. strigosa Schreb., 5. A. magna Murphy et Terr., 6. A. murphyi Ladiz. and 7. A. sativa L. The first three species are wild diploids. The fourth is a diploid-tetraploid complex of wild forms, weeds and cultivars. The fifth and sixth are wild tetraploids and the seventh constitutes a hexaploid aggregate of wild forms, weeds and cultivated forms. A short morphological and ecological characterization is given to each species.The cytogenetic, ecological and morphological evidence available on the evolutionary divergence and on the genetic affinities between the seven species is reviewed and discussed. The conclusion in drawn that in Avena there are two independent developments of polyploidy: 1. strigosa tetraploids, where the diploid background is well known; 2. the tetraploid-hexaploid of A. magna, A. murphyi and A. sativa (series Denticulatae). Here the first two species apparently represent the general tetraploid background from which hexaploid A. sativa evolved. It is also argued that it is impossible to explain the morphology, ecology and cytology of series Denticulatae in terms of the known diploid species of Avena, and a yet missing hypothetical diploid ancestor was extrapolated.  相似文献

Summary Pollen germination and pollen tube growth after self- and intra- and interspecific pollinations of annual [M. scutellata (L.) Mill., M. disciformis DC., and M. rigidula (L.) All.] and perennial (M. sativa L.) Medicago species were studied using a Carl Zeiss microscope with a fluorescence filter attachment.Self-pollination of self-fertile annual and intraspecific pollination of cross-fertile perennials were characterized by high pollen germination and rapid normal pollen tube growth. In contrast pollen germination percentages were very low and many pollen tube growth abnormalities occurred following interspecific pollination and self-pollination of self-sterile plants. The time period from pollination to fertilization for interspecific crosses was about double that following self-pollination of the annual species. However, fertilization occurred frequently following interspecific pollinations. Limitation in mitochondrial genome diversity of leek, revealed by restriction fragment length polymorphism (RFLP) analyses with mitochondrial gene probes, prevent a cytoplasmic male sterility (CMS) system in elite populations. However, mitochondrial genome diversity was detected in Allium ampeloprasum L. wild accession and landraces, as well as in pearl onion. Within this plant material, nine mitotypes were distinguished and could be used in order to broaden the genetic basis of leek. A chimeric mitochondrial gene configuration is usable as a marker for the sterility inducing cytoplasms (S1) in chives (Allium schoenoprasum L.) and in onion (Allium cepa L.) for (S) and (T) cytoplasm. This chimeric mitochondrial gene configuration is also present in the subgenus Allium, revealed by polymerase chain reaction (PCR). However, only a faint amplicon was observed in a few accessions investigated herein, suggesting that this fragment might be present to a lesser level in mitochondrial DNA, as a sublimon.  相似文献

Summary A partial male sterility system in the soybean (Glycine max (L.) Merr.) germplasm population AP6(SI)CI was found to be controlled monogenically by a recessive allele, msp. Observations of msp msp plants in different environments suggested that environmental conditions significantly affect expressivity of the msp allele with respect to male sterility. We obtained no experimental evidence of cytoplasmic effects on msp expression. The orf138 gene, which is specific to Ogura male-sterile cytoplasm, was analysed in mitochondrial DNA (mtDNA) of the wild radish, Raphanus raphanistrum, by polymerase chain reaction (PCR), Southern hybridization and sequencing. The effect of R. raphanistrum cytoplasm on the expression of male sterility was also examined in progeny with R. sativus. A PCR-aided assay and Southern hybridization revealed that three out of six strains analysed included plants with orf138. The sequence of wild type orf138 was same as that of Ogura, except for one or two nucleotide substitutions. Southern hybridization showed a novel mtDNA configuration in R. raphanistrum, in addition to the normal and Ogura types identical to those in R. sativus. Among interspecific hybrids, all the F1 had normal pollen fertility. In the F2 progeny between female wild plants having orf138 and the maintainer of Ogura male sterility, male-sterile plants were segregated, fitting the ratio of 3 fertile: 1 sterile plant. R. raphanistrum has cytoplasm that induces male sterility in radishes, and contains a dominant fertility restorer gene.  相似文献

Summary The F1 hybrids from crosses of 59 accessions of wild and cultivated Triticum types including amphidiploids T. boeoticum-Ae. squarrosa, T. timopheevi-Ae. squarrosa, T. timopheevi-T. monococcum, T. boeoticum (4n), T. macha, and T. Zhukovskyi with T. durum Sel. 56-1 and/or T. aestivum were examined for male sterility and chromosome pairing at metaphase I of meiosis in pollen mother cells. Those hybrids which produced male-sterile F1's were recurrently backrossed with pollen from T. durum or T. aestivum to study segregation for male sterility and/or to confirm cytoplasmic male sterility.All T. timopheevi and T. araraticum accessions and several T. dicoccoides types, including T. dicoccoides var. nudiglumis from the Turkey-Iran-Iraq area, had male sterility inducing cytoplasm. The chromosome pairing in the F1 hybrids indicated that all tetraploid Triticum accessions with male sterility inducing cytoplasm had genome AAGG. T. dicoccoides Körn types from the Turkey-Iran-Iraq area had genomes AABB and did not have male sterility inducing cytoplasm. Therefore, T. dicoccoides Körn and the T. timopheevi complex differ from each other cytoplasmically and cytogenetically and occur sympatrically in the Turkey-Iran-Iraq area.Possibly, the cytoplasm of the emmers was not derived from the putative diploid progenitors, T. boeoticum, Ae. speltoides, or Ae. bicornis as indicated by their nucleo-cytoplasmic and cytogenetic relationships with the tetraploid Triticum species. The cytoplasmic differences among Ae. speltoides, T. araraticum and T. timopheevi are of a relatively smaller magnitude than the cytoplasmic differences among T. timopheevi, T. boeoticum, and the emmers. Summary Barriers to interspecific hybridization in Trifolium were investigated by manipulation of mentor pollen treatments, ploidy levels, and compatibility and male sterility systems. Crosses involving the addition of mentor pollen produced fewer seeds and hybrids than crosses involving normal pollination. Lower seed set with mentor pollen was deduced to result from the use of less viable pollen, approximately half the pollen having been killed by alcohol. Pollinations at the diploid level resulted in more hybrids than at the tetraploid level, perhaps because genes for male sterility produced higher female sterility in the tetraploids. The self-compatible stock produced more seeds, mostly selfs, than the self-incompatible stock, but produced more hybrids only in one cross, T. pratense L. × T. diffusum Ehrh. The use of male-sterile female parents reduced selfing but produced fewer hybrids than male-fertile female parents. Techniques of this study were designed to affect prefertilization barriers, but the lack of effect may indicate that postfertilization barriers in Trifolium are of greater importance.Journal Article No. 98-3-208 of Kentucky Agricultural Experiment Station. Summary Investigations of variable expression of msp partial male sterility in soybeans (Glycine max (L.) Merr.) showed that higher temperatures promote male fertility in msp homozygotes and showed that infectious agents are not intrinsic to the sterility system. Exchange grafts failed to modify fertility levels of msp msp rootstocks, Msp Msp scions, and their self-progeny. Tests for soybean mosaic virus and tobacco ringspot virus were negative in partially male-sterile plants, in control fertile plants, and in self-progeny of grafted plants. Growth-chamber experiments and field observations manifested that male fertility of msp msp plants is higher in hot environments than in cooler ones. The unexpected aberrant ratios of fertile to partially male-sterile plants observed in 1977 (Stelly & Palmer, 1980) are explainable on the basis of msp temperature sensitivity.Our observations suggest that homogeneous msp msp populations may be increased in hot environments.Research Geneticist, SEA-AR, USDA, Iowa State University, Ames, Iowa 50011, USA.  相似文献

Summary Available evidences drawn from biosystematics, evolutionary biology, biogeography, archaeology, history, anthropology, paleo-geology and paleo-meteorology are pooled to reconstruct the series of events that led to the cosmopolitan cultivation of the Asian cultivated rice (O. sativa) and the regionalized planting of the African cultigen (O. glaberrima) in West Africa. The genus Oryza originated in the Gondwanaland continents and, following the fracture of the supercontinent, became widely distributed in the humid tropics of Africa, South America, South and Southeast Asia, and Oceania. The two cultivated species have had a common progenitor in the distant past. Parallel and independent evolutionary processes occurred in Africa and in Asia, following the sequence of: wild perennialwild annualcultivated annual. The weed races also contributed to the differentiation of the cultivated annuals. The corresponding members of the above series are O. longistaminata Chev. et Roehr., O. barthii A. Chev., O. glaberrima Steud., and the stapfii forms of O. glaberrima in Africa; O. rufipogon Griff., O. nivara Sharma et Shastry, O. sativa L., and the spontanea forms of O. sativa in Asia.The differentiation and diversification of the annuals in South Asia were accelerated by marked climatic changes following the last glacial age, dispersal of plants over latitude or altitude, human selection, and manipulation of the cultural environment.Cultivation of rice began in many parts of South and Southeast Asia, probably first in Ancient India. Cultural techniques such as puddling and transplanting were first developed in north and central China and later transmitted to Southeast Asia. Wetland culture preceded dryland culture in China, but in hilly areas of Southeast Asia, dryland cultivation is older than lowland culture. The planting method progressed from shifting cultivation to direct sowing in permanent fields, then to transplanting in bunded fields.Widespread dispersal of the Asian cultigen led to the formation of three eco-geographic races (Indica. Sinica or Japonica, and Javanica) and distinct cultural types in monsoon Asia (upland, lowland, and deep water). Varietal types changed readily within the span of a millenium, largely due to cultivators' preferences, socio-religious traditions, and population pressure. Genetic differentiation developed parallel to the ecologic diversification process.The African cultigen developed later than the Asian cultigen and has undergone less diversification. The wild races in South America and Oceania retain their primitive features mainly due to lack of cultivation pressure or dispersal.Both the African and Asian rices are still undergoing evolutionary changes at habitats where the wild, weed, and cultivated races co-exist.  相似文献

Summary Cytological studies of wild diploid Arachis species in the same section of the genus (sect. Arachis) as the cultivated peanut A. hypogaea L. show, with one exception, a karyotype characterized by the presence of 9 pairs of larger chromosomes and one pair of small (A) chromosomes. The exceptional species A. batozocoi Krap. et Greg. has a more uniform karyotype. Interspecific hybrids between diploid species of similar karyotype have moderate to high pollen stainability, those involving A. batizocoi have zero pollen stainability and a very irregular PMC meiosis. Such infertile hybrids are the most likely to produce fertile, stable amphidiploids on doubling the chromosome complement. It is suggested that the cultivated peanut could have originated from such a sterile interspecific hybrid and on morphological and phytogeographic grounds the most likely genome donors are A. cardenasii (nomen nudum) and A. batizocoi of the species within section Arachis, which have been collected up to the present time.Paper number 5560 of the Journal Series of the North Carolina Agricultural Experiment Station, Raleigh, NC 27650  相似文献

Summary The world collection of cultivated safflower, C. tinctorius L., containing about 2000 lines, was screened for resistance to one of its severest insect pests, the safflower fly, Acanthophilus helianthi R. Only 3 lines with low infestation levels were found but they cannot be considered reliable sources of resistance. Nine wild Carthamus species were also tested. Several species contained fly-free accessions, among them two species which are closely related to cultivated safflower, viz C. flavescens Willd. and C. palaestinus Eig. Breeding for earliness which will lead to more escapes, appears the best approach towards minimizing fly damage in the Mediterranean area.  相似文献

In this article a survey is given on sugar cane breeding, as it was performed in Java during a period of about fifty years. When in the eighties of the nineteenth century sugar cane was heavily affected by the sereh disease it was Soltwedel, the first director of the Sugar Experiment Station Midden Java, who tried to obtain sereh resistant cane varieties by species hybridization, until his early death intervened. The first species hybrids were obtained in 1893 by Wakker, who crossed noble sugar cane, Saccharum officinarum, with Kassoer, considered by him as a wild species. In later years it appeared from morphological investigations by Jeswiet (1916) and from cytological investigations by Bremer (1921) that Kassoer is to be considered as a spontaneous hybrid between S. officinarum and S. spontaneum, the wild glagah. In 1895 Kobus imported the Indian sugar cane Chunnee in Java. Chunnee, not belonging to S. officinarum, was crossed with noble sugar cane (for the first time) in 1897. From this cross many clones were obtained which appeared to be resistant against the sereh disease but were highly susceptible to mosaic. The continued crossing between noble sugar cane and Kassoer, however, was very successful. Many clones were obtained, which as commercial varieties, showed a high degree of resistance against sereh disease and mosaic and moreover gave a much higher sugar production than the susceptible varieties of noble sugar cane.The following terms are introduced: first, second and third nobilisation of the wild S. spontaneum. Kassoer cane itself is a product of the first nobilisation, the direct cross between noble sugar cane and wild cane. The back cross between Kassoer and noble sugar cane is called the second nobilisation. When cane varieties belonging to the second nobilisation are crossed again with noble sugar cane, the third nobilisation of S. spontaneum is said to take place. The well-known sugar cane variety 2878 P.O.J. belongs to the third nobilisation.In all clones investigated Saccharum officinarum has 2n=80 chromosomes. Within S. spontaneum types occur which in chromosome number vary from 2n=48 to 2n=128. The Java glagah has 2n=112 chromosomes.Clones of the first glagah nobilisation S. officinarum (n=40) x S. spontaneum (n=56) did not have 2n=40+56 chromosomes, but 2n=40+40+56=136 chromosomes.In relation to the taxonomy of Saccharum many other cytological details are given. Within S. officinarum the basic chromosome number x=10. About S. spontaneum opinions are divided. The author supposes that x=6,8 and 10 are basic numbers of S. spontaneum.  相似文献

Seed regeneration is one of the most important tasks in genebank work. To regenerate germplasm of allogamous, entomophilous plant species in isolation facilities, insects are often used as pollen vectors. The objective of this study was to determine the effect of pollinator species, pollinator densities, pollinator combinations, and flower preferences on the seed quantity and quality of Daucus carota, Allium cepa, and Brassica rapa. The isozyme phosphoglucoisomerase (PGI) served as genetic marker in determining outcrossing rates and pollen dispersal distances. Field plots and isolation cages without insects were used as controls. The pollinators studied consisted of the following insect species from German native fauna: red mason bees (Osmia rufa L.), hoverflies (Eristalis tenax L.), and blowflies (Calliphora sp. and Lucilia sp.). In D. carota and B. rapa, higher seed yields were achieved in isolation cages than in field plots, but in A. cepa lower seed yields were obtained in the isolation cages. Although O. rufa showed no liking for A. cepa, unintentional pollination was effected by this insect in isolation cages. Lower insect densities were necessary for O. rufa than for the various fly species.  相似文献

Summary Populations of wild barley, Hordeum spontaneum, in Israel, originating from diverse habitats, and tested earlier for allozyme (Nevo et al., 1979a) and disease resistance polymorphisms (Moseman et al., 1984; Nevo et al., 1984a), were compared and contrasted for performance in agronomically important phenotypic traits. The traits compared involved 10 variables comparing germination, earliness, numerical and weight variables of biomass and yield. The field experiments were conducted in 1980, 1981 and 1982 in two relatively standardized and contrasting environments: mesic (Mount Carmel, Haifa) and xeric Avedat farm, and Sede Boqer (both in the northen Negev desert). The experimental design involved 26 population quadrangles at Avedat farm (1980), and rows of randomized plants of 11 populations in both Haifa and Sede Boqer (1981 and 1982).The results indicate that the characters studied are partly genetically determined. Striking genetic variation was found between and within populations in each site, whereas remarkable environmental variation including genetic environmental interaction was found between the mesic and the xeric sites, as well as between populations and years. We conclude that natural populations of wild barley in Israel vary not only in genetic polymorphisms of allozymes and disease resistance, but also in quantitative traits of agronomie importance. These traits are economically significant and should be conserved and utilized in barley crop improvement.  相似文献

Summary Studies of intermediate wild-cultivated plants suggested that the Indica-Japonica differentiation of cultivars of Oryza sativa L. has taken place with the domestication of wild plants. The wild progenitor, which is considered to be the Asian form of O. perennis Moench, shows no trace of differentiation into two such types as the Indica and Japonica. To elucidate the potentiality of the wild progenitor to evolve the two types when domesticated, the selfed progenies from an Indica × wild and a japonica × wild cross were examined with regard to three characters known to be useful for distinguishing between the two types. The data showed that Japonica-like plants were derived from the Indica × wild cross and Indica-like plants from the Japonica × wild cross. The Indica-Japonica differentiation was considered to have resulted from selection of incipient domesticates having such potentiality under different climatic conditions; the dynamics was discussed.  相似文献

Polyacrylamide gel electrophoresis (PAGE) was used to study the polymorphism of esterases in Allium cepa L. and A. fistulosum L. Two varieties of each species, an A. fistulosum × A. cepa interspecific F1 hybrid, and (A. fistulosum × A. cepa) hybrid derivatives were analyzed for determination of banding patterns upon staining with α- and β-napthyl acetate substrates of esterase. Complex band patterns were observed. In total, 10 bands were detected between A. cepa and A. fistulosum — five inA. cepa, six in A. fistulosum with only one band common to both species. With the exception of one band unique to A. fistulosum which appeared only when stained with α-substrate, extracts of both A. cepa and A. fistulosum leaf tissue exhibited the same bands when stained with both α- and β-substrates. Bands stained with the different systems are distinguished by color: α-substrate always appeared black, while bands stained to β-substrate are always red. Esterase bands were assigned into 5 presumptive loci of four zones of activity with according to the migration distance of the bands from the front, color of each band upon staining with α- and β-substrates, and segregation observed in crosses and hybrid derivatives. Esterase enzymes detected in this study appear to be monomeric. Polymorphisms were identified between A. cepa and A. fistulosum by esterase banding patterns. Esterase enzymes provide an additional marker in monitoring introgression of foreign germplasm in interspecific onion breeding. Summary Atylosia scarabaeoides Benth. and A. platycarpa Benth., close relatives of the species Cajanus cajan (L.) Millsp., were screened for photoperiodic response. Four photoperiods ranging from 12 h 45 min to 19 h were studied in three environments. A. scarabaeoides flowered freely only in the first photoperiod. A. platycarpa not only flowered early (39 to 63 days after planting) in all four photoperiods, but also exhibited a relatively constant vegetative phase up to 16 h of illumination. Cajanus cajan, in which most cultivars-if not all-exhibit a response to photoperiod, was successfully hybridized with the two Atylosia species. It is suggested that genes for earliness and insensitivity to day-length could be transferred from A. platycarpa to C. cajan.  相似文献

Summary A high concentration of gibberellic acid (GA3, 2%) was found to act as a gametocide for the common onion, Allium cepa L., when sprayed in the beginning of the bolting process.This technique is suggested as a possible substitute for hand emasculation, since it would greatly facilitate interspecific and intervarietal crosses, recurrent backcrosses and the selection of good B lines.  相似文献
